- Description: Stenotrophomonas maltophilia is increasingly emerging as an opportunistic pathogen of global concern. Due to its inherent resistance to several classes of antibiotics including carbapenems and its ability to acquire mobile resistance elements, treatment of infections caused by S. maltophilia is a constant challenge for clinicians. Trimethoprim-sulphamethoxazole (TMP-SMX) is the generally accepted antibiotic of choice for the treatment of infections caused by this organism, but resistance to the drug is increasingly being reported; hence, the need for alternative therapeutic options. In this study, the antimicrobial susceptibility profile of 110 commensal S. maltophilia isolates obtained from Nkonkobe municipality, Eastern Cape Province, Republic of South Africa was investigated. Twenty-one antibiotics including TMP-SMX and the newer fluoroquinolones; levofloxacin, gatifloxacin and moxifloxacin were included in the antibiotic panel. About 63.4 percent of the isolates were susceptible to TMP-SMX with a resistance rate of 28.2 percent. The fluoroquinolones were more effective with susceptibilities ranging from 76 percent to 94.7 percent. Resistance to the fluoroquinolones ranged from 1.3 percent to 2.7 percent. Levofloxacin was the most effective fluoroquinolone tested. Phenotypic dectection of extended spectrum β-lactamases (ESBLs) showed double disc synergy test (DDST) positivity in 59.5 percent of the isolates. Cefepime was the most sensitive indicator cephalosporin in the DDST with 77.3 percent of suspected ESBL-producing isolates showing cefepime-clavulanic acid synergy. Isolates exhibited nine different ESBL phenotypes, however, PCR amplification of the bla genes revealed four isolates that possessed genes belonging to the CTX-M group (CTX-M-1 and CTX-M-8 groups). ESBL genes are usually carried on mobile elements such as plasmids and transposons which may also bear genes that mediate resistance to aminoglycosides, tetracyclines, TMP-SMX and fluoroquinolones. ESBL positive isolates appeared more susceptible to the fluoroquinolones compared to TMP-SMX but there was no significant relationship between ESBL production and susceptibility to these drugs (p > 0.05). The newer fluoroquinolones are a possible alternative treatment option for S. maltophilia infections in this environment but further studies and clinical investigations are needed to determine the in vivo efficacy of these drugs.

- Description: A study to assess the potentials of some commensal bacteria that belong to Staphylococcus, Acinetobacter and Stenotrophomonas genera as reservoirs of antibiotic resistance determinants in the environment of Nkonkobe Municipality of the Eastern Cape Province, South Africa, was carried out using standard microbiological and molecular techniques. A total of 120 Staphylococcus isolates which consisted of Staphylococcus haemolyticus (30%), Staphylococcus aureus (23.3%) from pig; Staphylococcus capitis (15%) from goat; Staphylococcus heamolyticus (5%) and Staphylococcus xylosus (15%) from cattle and other Staphylococci (11%) from dead chicken and pigs were isolated. About 23.3% of these isolates were coagulase positive and 76.7% were coagulase negative. This difference in prevalence along coagulase production divide was statistically significant (p < 0.05). Eighty-six Acinetobacter species (Acinetobacter baumannii/calcoaceticus and Acinetobacter haemolyticus) were also isolated from Alice and Fort Beaufort towns samples, while 125 Stenotrophomonas maltophilia isolates were from grass root rhizosphere (96%) and soil butternut root rhizosphere (4%). Between 75-100% of the Staphylococccus species were resistant to Penicillin G, tetracycline, sulphamethaxole and nalidixic acid; about 38 % were methicillin resistant, consisting of 12.6% methicillin resistant Staphylococcus aureus (MRSA) from pig and a total of 12% vancomycin resistant were observed. Also, 12% of the isolates were erythromycin resistant while 40.2 % were resistant to the third generation cephalosporin, ceftazidime. The antibiotic resistance genes vanA, VanB, eryA, eryB, eryC were not detected in all the phenotypically resistant Staphylococccus species, but mec A gene and mph genes were detected. In the Acinetobacter species, a wide range of 30-100% resistance to penicillin G, ceftriazone, nitrofurantoin, erythromycin, and augmentin was observed. Polymerase chain reaction (PCR) revealed the presence of Tet(B) and Tet(39) genes in these species, while Tet (A), Tet(M) and Tet(H) were absent. Also, 9.3% of the Acinetobacter species showed phenotypic production of extended spectrum beta lactamases (ESBLs) while 3.5% were positive for the presence of blaCTX-M-1 genes. The Stenotrophomonas maltophilia isolates showed varying resistance to meropenem (8.9%), cefuroxime (95.6 %), ampicillin-sulbactam (53.9%), ceftazidime (10.7%), cefepime (29.3 %), minocycline (2.2%), kanamycin (56.9%), ofloxacin (2.9%), levofloxacin (1.3%), moxifloxacin (2.8%), ciprofloxacin (24.3%), gatifloxacin (1.3%), polymyxin B (2.9 %), cotrimoxazole (26.1%), trimethoprim (98.6%), aztreonam(58%) and Polymyxin B (2.9 %). The isolates exhibited significant susceptibility to the fluoroquinolones (74.3-94.7 %), polymycin (97.1%) and meropenem (88.1%). Only sul3 genes were the only sulphonamide resistance gene detected among the trimethoprim-sulphamethoxazole resistant isolates. The observed multiple antibiotic resistance indeces (MARI) of >2 for Staphylococcus species, Acinetobacter species and Stenotrophomonas maltophilia suggest that they have arisen from high-risk sources where antibiotics are in constant arbitrary use resulting in high selective pressure. The presence of tetracycline resistance genes in Acinetobacter species justifies the observed phenotypic resistance to oxytetracycline and intermediate resistance to minocycline. High phenotypic resistance and the presence of some resistance genes in Staphylococcus species is a possible threat to public health and suggests animals to be important reservoirs of antibiotic resistance determinants in the environment. Indiscriminate use of antibiotics induces this kind of antibiotic resistance and should be discouraged. Personal hygiene is encouraged as it reduces the load of Acinetobacter species contacted from the environment that may be difficult to control. Commensal Stenotrophomonas maltophilia are as important as their clinical counterparts due to their roles in opportunistic infection, antibiotic resistance and their associated genes, especially sul gene. Personal hygiene is hereby advocated especially when in contact with soil, plants and plants’ rhizospheric soil.

They are the fundamental entitlements or minimum standards to be met for individual so that they live with dignity. This study focused on discrimination of women under customary law in South Africa with reference to inheritance and succession. The study validated the findings of other researchers on the impact of cultural practices on women’s rights to inheritance and succession. In addition the findings revealed that efforts t eliminate traditional practices, should foremost come from men and from communities that hold such destructive attitudes towards women. The outcomes and recommendations of this study would assist the government and other institutions to adopt effective measures to empower women and especially educate them so that they can assert and defend their human rights

- Description: The objective of the study was to determine the avoidance-related behaviour, bleeding times and the quality of meat from sheep of different gender, breed (Dorper, Merino, and their crosses) and age groups, slaughtered at a commercial and municipal abattoir. Castrates and ewes of different age groups (1= <10months, 2= 11-12months, and 3= >12months) and breeds were used. From the commercial abattoir, 90 castrates and 110 ewes were used, while 66 castrates and 19 ewes were obtained from the municipal abattoir. Meat samples were obtained to determine meat colour, ultimate pH, temperature, cooking loss and tenderness. Correlations between bleeding times and meat quality variables were also determined. The two abattoirs differed in terms of technology and the procedures used, and this brought about different response behaviours in sheep at slaughter, bleeding times at exsanguination and the quality of meat produced. Animal-related factors (breed, gender and age classes) had no effect on the avoidance-related behaviour of sheep slaughtered at the commercial abattoir, while these factors affected those sheep slaughtered at the municipal abattoir. These factors also had an effect on the quality of meat produced at both abattoirs. Gender and weight of the animals had effects on the bleeding times at exsanguination at both abattoirs. Bleeding times did no correlate with any of the meat quality variables, but correlations among meat quality variables were observed. Avoidance-related behaviour, bleeding times, and the quality of mutton from sheep slaughtered at a commercial and a municipal abattoir was affected by breed, age, and gender.

- Description: The Kalagadi Manganese mine in the Kuruman area of the Northern Cape Province of South Africa contains reserves of Mn ore in excess of 100Mt. Mineralization in the mine lease area is restricted within the Hotazel Formation of the Voȅlwater Subgroup, belonging to the Postmasburg Group, the upper subdivision of the Transvaal Supergroup. Surface topography is characterized by flat lying, undulation with minimal faulting and the ore are slightly metarmophosed. This study investigates the general geology of the mine, lithostratigraphic subdivision and correlation of the economic Lower Manganese Orebody (LMO) of the Kalagadi Manganese Mine in order to guide mining plan and operations once the mine is fully commissioned. At the commencement of this study, Kalagadi Manganese mine was a project under exploration with no specific geology of the mine lease area and no lithostratigraphic subdivision. The study also aimed determining the extent of lithostratigraphic correlation between the LMO economic orebodies of the Kalagadi Manganese mine with that of underground Gloria and open-pit Mamatwan mines. Four methods including petrographic microscope, Scanning electron Microscope (SEM), X-ray diffraction (XRD) and X-ray fluorescence (XRF) analyses were applied mainly for the mineral identification, chemical composition and ore characterization of the Lower Manganese Orebody (LMO) at Kalagadi Manganese mine. The results of this study indicates the following: (1) Eleven textural distinct zones with economic zones restricted to the middle while the lower grade zones are confined to the top and bottom of the LMO; (2) The economic zones, comprising of Y, M, C and N subzones attain an average thickness of 10 m and are graded at an average of 40% Mn while the Mn/Fe ratio varies from 6 to 9; (3) The most economic zones are M and N subzones which are mostly characterized by oxidized ovoids and laminae, a characteristic applicable even to other zones of economic interest; (4) Braunite is the main mineral of the manganese ore and is often integrown with kutnahorite and other minerals (hematite, hausmannite, Mg-calcite, calcite, jacobsite, serpentine and garnet) which are present in variable amounts; (5) The Mg-rich calcite (Ca, Mg)CO3 is the second dominant manganese carbonate mineral and it corresponds to elevated MgO concentration and is often associated with marine environment. The occurrence of the Mgcalcite is not common in the manganese ore of this area except for the Mn-calcite, which was not determined by XRD analyses in this study; (6) MnO is the most abundant major oxide in the manganese ore while other major oxides present in decreasing order of abundance are CaO, SiO2, Fe2O3, and MgO. The oxides TiO2, Na2O, K2O, Al2O3, and Cr2O3 are depleted and are mostly 0.01wt% and 0.001wt% respectively while P2O5 concentrations are low ranging from 0.02wt% to 0.3wt%. The trace element concentrations of Ba, Zn and Sr in most borehole samples are slightly elevated ranging from 100ppm to 3.9% (36000pm) while Co, Cu, Ni, Y, As, Zr, V and La rarely exceed 50ppm. The enrichments of Cu, Zn, Ni, Co and V that are commonly associated with volcanogenic hydrothermal input in chemicals may reach up to 70ppm; (7) The mineralogical and geochemical characteristics of the manganese ore in the Kalagadi Manganese mine lease area are similar to that of Low-Grade Mamatwan-Type ore. The cyclicity (Banded Iron Formation ↔ Hematite lutite ↔ braunite lutite) and alternation of manganese and iron formation have been confirmed; and (8) The oxygen δ18O isotope values (18‰ to 22‰) indicate a slight influence of metamorphism of the manganese ore. No positive correlation exists between δ13C vs δ18O values and Mn vs δ13C values. Such observations indicate minimal action of organic carbon during manganese precipitation where the organic matter was oxidized and manganese content reduced. On the other hand, the manganese carbonates (CaO) are positively correlated with carbon isotope, this indicates diagenetic alteration and the involvement of biogenic carbonate during the formation of manganese carbonates. It is concluded that the lithostratigraphic subdivision at Kalagadi Manganese mine is best correlated physically, mineralogically and geochemically with that of Gloria mine operating in the Low Grade Mamatwan - Type ore while correlation with an open-pit Mamatwan mine is also valid.

The allelopathic potential of Arctotis Arctotoides (L.f.) O. Hoffm on some vegetables

- Description: Laboratory and greenhouse experiments were conducted to evaluate the allelopathic effects of the extracts and residue Arctotis arctotoides (L.f.) O. Hoffm on selected vegetable crops. The study aimed to address the following specific objectives to (i) examine the ultra structures of the leaf of A. arctotoides using the Scanning Electron Microscope (SEM), (ii) carry out comprehensive qualitative and quantitative phytochemical analysis of the root and shoot materials of the plant, (iii) investigate the allelopathic activities of the root and shoot aqueous extracts of A. arctotoides at concentrations of 10, 8, 6, 4 and 2 mg/ml on germination, radicle and plumule growth of cabbage, carrot, tomato and spinach, (iv) evaluate the inhibitory effects of the dried shoot residue of the plant at 10, 20 and 40 g kg-3 of soil (treatments B, C and D) and the control (treatment A) on the morphology, growth and chlorophyll pigment content of tomato and cabbage transplants at 1, 2, 3 and 4 weeks after transplanting and (v) assess the effects of the dried shoot residue of A. arctotoides on the yield, nutrient uptake by the leaves of tomato and cabbage at 4 and 12 weeks after transplanting. Finally, to analyze the residual mineral content of the soils with tomato and cabbage transplants at 12 weeks after transplanting. The the SEM revealed that anisocytic stomata and glandular trichomes (GTs) were numerous on the abaxial than the adaxial surfaces of A. arctotoides. The non glandular trichomes (NGTs) were also present on both surfaces but lesser on the abaxial. Morphologically, the GTs were peltate, uniseriate and globular head while the NGTs were cylindrical and filamentous with variable number of cells at the basal portion. The energy dispersive X-ray spectroscopy of some crystals showed that Na+ Mg2+ and Ca2+ were the major constituents of the crystal deposit found around the GTs and stomata. The results of the phytochemical composition of the root and shoot extracts of A. arctotoides confirmed the occurrence of alkaloids, flavonoids, phenolics, saponnins, tannins and triterpenes as the common constituents. In addition, cardiac glycosides and steroids were also detected in the shoot of the A. arctotoides. Quantitative estimation of the chemical constituents of the crude extracts further revealed that the alkaloid content in the root higher (0.97 percent) than the shoot (0.64 percent). The quantity of flavonoids detected in the shoot (1.02 percent) was more than that observed in the root (0.35 percent). Others (phenolics and tannins) were marginal in the two plant parts. The results of the inhibitory effects of the root and shoot aqueous extract at the varying concentrations showed that root extract at 10 mg/ml considerably reduced the germination of cabbage, carrot, tomato and spinach seeds by 84.0, 83.2, 72.8 and 37.4 percent respectively. Incubation of the shoot extract at the same concentration resulted in 100 percent inhibition of cabbage and carrot seed germination whereas those of tomato and spinach were suppressed by 91.5 and 61.2 percent respectively. The two extracts at the varying concentrations also had a significant reduction on the radicle and plumule growth of the four vegetables. Addition of the shoot residue to the soil showed massive chlorosis, necrotic lesions and wilting of tomato and cabbage leaves under treatments C and D at 2 weeks after transplanting. The number of leaves, leaf area, dry shoot and root weight of the two vegetables grown in the amended soils were also drastically reduced. The inhibition percentages due to the addition of the three concentrations of A. arctotoides dried shoot residue on the dry shoot weight at 4 weeks after transplanting were 38.6, 45.5 and 70.3. for tomato and 57.5, 73.3 and 87.5 percent for cabbage. Similarly, the declines in the dry root weight of 61.3, 82.9.4 and 83.4 percent for tomato as well as 53.1, 54.7 and 67.2 percent for cabbages were recorded for the two vegetables under treatment B, C and D during the period. The results further showed that the dry fruit yield and shoot weight of tomato under the treatments B, C and D decreased with increase in shoot residue concentrations of A. arctotoides. Relative to treatment A, no significant differences were recorded in the dry head weight of cabbage under the residue treated groups. The reductions in the fruit yield and fresh head weight caused by treatments C and D were 37.2 and 84.8 percent for tomato and 30.9 and 72.4 percent for cabbage. The findings on the mineral contents in the leaves of the two vegetables revealed significant differences in the uptake of N, Mg, Na, Cu and Fe by tomato leaves. The concentrations of N, K, Na and Zn in cabbage leaves also differed. However, the P content was relatively constant in the leaves of the two vegetables at 4 and 12 weeks after transplanting. At 12 weeks after transplanting, the Fe content in soils with tomato and cabbage treatments C and D was greatly enhanced in comparison with the other nutrients. The residual N, P and Zn detected in soils planted to cabbage were similarly equal among all the groups including the control. Thus, under the greenhouse experiment, Arctotis arctotoides (L.f) O. Hoffm has been shown to contain some phytotoxic chemical compounds in its root and shoot materials. The compounds either singly or collectively have demonstrated some inhibitory potentials on the germination, growth and yields of cabbage, carrot, tomato and spinach evaluated in this study.

- Description: Quality protein maize (QPM) has enhanced levels of two essential amino acids, lysine and tryptophan compared to normal maize. This makes QPM an important cereal crop in communities where maize is a staple crop. The main abiotic factor to QPM production is drought stress. Little information is available on the effect of drought stress on QPM. Therefore, the objectives of this study were to: (i) conduct diversity analysis of QPM inbred lines using morpho-agronomic and simple sequence repeat markers, (ii) screen available QPM inbred lines and F1 progeny for tolerance to seedling drought stress, (iii) determine the combining ability and type of gene action of QPM inbred lines for tolerance to seedling drought stress, grain yield and endosperm modification. The study was conducted in South Africa, at the University of Fort Hare. Morphological characterisation of 21 inbred lines was done using quantitative and qualitative traits. A randomised complete block design with three replicates was used for characterizing the inbred lines in the field. Genstat statistical software, version 12 (Genstat ®, 2009) was used for analysis of variance (ANOVA) and descriptive statistics. Analysis of variance was performed on all quantitative data for morphological traits. Data for qualitative traits was tabulated in their nominal classes. Traits that contributed most to the variation were days to anthesis, days to silking, anthesis-silking interval, plant height, number of kernel rows, ear length and grain yield. Cluster analysis grouped the inbred lines into three main clusters. The first cluster was characterised by tall and average yielding lines, while the second cluster showed the least anthesis-silking interval, and had the highest yield. Cluster three consisted of lines that were early maturing, but were the least yielding. Genetic distances between maize inbred lines were quantified by using 27 simple sequence repeat markers. The genetic distances between genotypes was computed using Roger’s (1972) genetic distances. Cluster analysis was then carried out using the neighbour-joining tree method using Power Marker software version 3.25. A dendrogram generated from the genetic study of the inbred lines revealed three groups that concurred with expectations based upon pedigree data. These groups were not identical to the groups generated using morpho-agronomic characterisation. Twenty one QPM inbred lines were crossed using a North Carolina design II mating scheme. These were divided into seven sets, each with three inbred lines. The three inbred lines in one set were used as females and crossed with three inbred lines in another set consisting of males. Each inbred line was used as a female in one set, and as a male in a second set. Sixty three hybrids (7 sets x 9 hybrids) were formed and evaluated in October 2011, using a 6x8 alpha-lattice incomplete block design with three replicates under glasshouse and optimum field conditions. A randomised complete block design with three replicates was used for the 21 parental inbred lines. Traits recorded for the glasshouse study were, canopy temperature, chlorophyll content, leaf roll, stem diameter, plant height, leaf number, leaf area, fresh and dry root and shoot weights. Data for the various traits for each environment, 25 percent (stress treatment) and 75 percent (non-stress) of field capacity, were subjected to analysis of variance using the unbalanced treatment design in Genstat statistical package Edition 12. Where varietal differences were found, means were separated using Tukey’s test. Genetic analyses for grain yield and agronomic traits were performed using a fixed effects model in JMP 10 following Residual Maximum Likelihood procedure (REML). From the results, inbred lines that were not previously classified into heterotic groups and drought tolerance categories were classified based on their total dry weight performance and drought susceptibility index. Inbred lines L18, L9, L8, L6 and L3, in order of their drought tolerance index were the best performers under greenhouse conditions and could be recommended for breeding new varieties that are tolerant to seedling drought stress. Evaluation of maize seedlings tolerant to drought stress under glasshouse conditions revealed that cross combination L18 x L11 was drought tolerant, while cross L20 x L7 was susceptible. Total dry weight was used as the major criteria for classifying F1 maize seedlings as being resistant or susceptible. General combining ability effects accounted for 67.43 percent of the genetic variation for total dry weight, while specific combining ability effects contributed 37.57 percent. This indicated that additive gene effects were more important than non-additive gene action in controlling this trait. In the field study (non-drought), the experimental design was a 6x8 alpha lattice incomplete block design with three replicates. On an adjacent field a randomised complete block design with three replicates was used to evaluate the parental inbred lines. The following variables were recorded: plant height, ear height, ears per plant, endosperm modification, days to silking and days to anthesis, anthesis-silking interval, number of kernels per row, number of rows per ear and grain yield. General analyses for the incomplete lattice block design and randomised complete block design for hybrid and inbred data respectively were performed using JMP 10 statistical software. Means were separated using the Tukey's test. Genetic analyses of data for grain yield and agronomic traits were conducted using a fixed effects model using REML in JMP 10. The importance of both GCA (51 percent) and SCA (49 percent) was observed for grain yield. A preponderance of GCA existed for ear height, days to anthesis, anthesis-silking interval, ears per plant and number of kernels per row, indicating that predominantly, additive gene effects controlled hybrid performance under optimum field conditions. The highest heritability was observed for days to silking (48.27 percent) suggesting that yield could be improved through selection for this trait. Under field conditions, variation in time to maturity was observed. This implies that these inbred lines can be recommended for utilisation in different agro-ecologies. Early maturing lines such as L18 can be used to introduce earliness in local cultivars, while early maturing single crosses such as L18 x L2, L5 x L9, L3 x L4 and L2 x L21 could be recommended for maize growers in drought prone areas such as the former Ciskei. Single crosses L18xL11, L16xL18, L8xL21 and L9xL6 had good tolerance to seedling drought stress. On the other hand, single crosses L18xL11 and L11xL13 had high grain yield and good endosperm modification. All these single crosses could be recommended for commercial production after evaluation across locations in the Eastern Cape Province. Alternatively they can be crossed with other superior inbreds to generate three or four way hybrids, which could then be evaluated for potential use by farmers in the Eastern Cape.

- Description: All plants are exposed to abiotic and biotic stresses and have developed intricate signalling responses to survive. They respond to cell-structure disruption caused by herbivore probing and feeding by the formation of callose. Callose is a linear homopolymer made up of β-1,3-linked glucose residues with some β-1,6-branches. Plant responses to abiotic or biotic stress share events such as phosphorylation, membrane depolarization, calcium influx and the release of reactive oxygen species such as hydrogen peroxide. These events lead to the up-regulation of several pathways leading to biosynthesis of signalling molecules such as salicylic acid, jasmonate, abscisic acid and ethylene pathways. The aim of this study was to determine the most suitable proteomic approach for identifying proteins and signalling pathways involved in cereal response to aphid infestation. An in silico approach was first evaluated in which the 5ʹ upstream regulatory region of proteins belonging to the family of callose synthases was scanned for cis-regulatory elements in order to identify which callose synthases are possibly expressed in plants during biotic or abiotic stresses. Bioinformatics tools were used in the identification of twelve Arabidopsis and ten rice callose synthase coding regions. Genome sequences for rice and Arabidopsis were scanned for the 2000 bp 5ʹ region upstream of the start codon of each callose synthase coding region. PlantCare, PLACE and Athena software were used to identify putative cis-regulatory elements present in the 2000 bp 5ʹ upstream sequences. The majority of cis-acting elements identified were involved in drought and high temperature responses and only one cis-acting element was involved in wound stress. These results therefore indicated a probable role for plant callose synthases in drought stress responses rather than in biotic stress responses. Genevestigator analysis of Arabidopsis results of micro-array experiments indicated that AtGSL10 is highly up-regulated, with AtGSL1, 3, 5, 6, 7, 8, 11 and 12 showing medium up-regulation and AtGSL2, 4 and 9 no up-regulation during aphid infestation of Arabidopsis plants, implicating a possible role for AtGSL10 in the plant response to aphid infestation. An LC/MS/MS approach was used to identify specific signalling pathways involved in wheat resistance or stress response to aphid infestation. Eight proteins were identified as being up-regulated during aphid feeding in wheat, and 11 proteins were identified as possibly involved in the wheat resistance mechanism against aphid infestation. Several proteins were also identified as constitutively expressed proteins, during normal conditions and aphid infestation. Most pathways identified with proteins up-regulated in the resistance mechanisms of TugelaDN plants, were related to energy metabolism and located in the chloroplast. Evaluation of two dimensional gel electrophoresis to identify phosphoproteins differentially regulated in wheat during aphid infestation, revealed the up-regulation of three proteins namely photosystem II oxygen-evolving complex protein 2, HVUNKNOWN from Hordeum vulgare subsp vulgare and HSKERAT9 NID from Homo sapiens. Additional 57 proteins were partially identified as involved in the stress response but due to low protein levels, the percentage of matching peptides to these proteins was below the required confidence level. Although these protein identifications were below the confidence level, it is interesting to note that several of the proteins are known stress response proteins, and therefore could serve as potential targets for future investigations. In conclusion, the down and up-regulation of wheat proteins after aphid feeding reported in this study suggest that several signalling pathways are involved in the cereal stress response to aphid feeding. In silico approaches require knowledge or identification of potential proteins whereas 2D and LC/MS can identify numerous proteins still unknown to be involved in specific stress responses. The 2D approach is also limited in that the proteins of interest may be in low abundance and therefore not detected in the gels due to the presence of high abundant proteins. Therefore the best approach to identify proteins and signalling pathways involved in the stress response of wheat to aphid infestation, is the LC/MS/MS approach, as this proved to be the most sensitive and robust, identifying the most proteins with a high degree of confidence.

- Description: Maintenance of water and solute homeostasis is a key requirement for living systems, and in vertebrates, homeostasis is in part achieved by natriuretic peptides (NP), a family of peptide hormones. A related family of peptide hormones have also been found in the plant kingdom. Plant natriuretic peptides (PNP) are a novel class of plant proteins with two closely related homologous genes (AtPNP-A and AtPNP-B) identified in the model plant Arabidopsis thaliana. AtPNP-A has been extensively studied and evidence obtained points to a role in plant water homeostasis. No research has been conducted on the function of PNP-B proteins. In this study, we focus on the bioinformatic analysis of the PNP-B gene in various plants, as well investigating whether PNP-B plays a role in water homeostasis in rice plants exposed to drought stress. Basic local alignment search tool (BLAST) queries of the ―The Gene Index‖, EST and available plant genome databases revealed the presence of the PNP-B mRNA in rice, oil seed rape, oak, leafy spurge, poplar, sugarcane, pepper, cotton, apple and maize. All the identified genome sequences contained a predicted intron/insert, which was not present in the related mRNA sequences. However, RT-PCR analysis revealed the presence of the 101 bp insert in the 976 bp amplified rice PNP-B (OsPNP-B) cDNA and therefore expression was optimized for the protein encoded by only the exon2 sequence as this contained the predicted active site region of PNP-B. OsPNP-B was shown to be translated to a protein of 14 kDa with a sequence similarity to AtPNP-B (54percent), AtPNP-A (37percent) and CjBAp12 (55percent), suggesting two possible functions for PNP-B viz water homeostasis and/or pathogenesis defence. To determine whether PNP-B is involved in water homeostasis, total protein extracted from 4 weeks old (4 leaves stage) rice plants subjected to drought treatment for a period of 24, 48, 72, 120, 168 and 240hrs were resolved by 17percent SDS-PAGE and analysed by western blot analysis. The PNP-B protein was found to be down-regulated during drought stress, implying that PNP-B may play a role in water homeostasis through the release of water from cells rather than the up-take of water as seen At-PNP-A. - Description: Lack of potable water is one of the major challenges that the world faces currently and the effects of this are mainly experienced by people in developing countries. This has therefore propelled research in advanced oxidation technologies AOTs to improve the current water treatment methods using cost effective, non toxic and efficient treatment methods. Hence, in this study the sol-gel synthesis method was used to prepare TiO2 nanoparticles that were photocatalytically active under UV and visible solar light as well as possessing antibacterial properties. Silver and nitrogen doping was carried out to extend the optical absorption of TiO2. For easy removal and reuse of the photocatalyst the nanoparticles were immobilized on chitosan and poly (vinyl-alcohol-co-ethylene) using the electrospining technique. The synthesized nanomaterials were characterized by FTIR, XRD, SEM/EDS, TEM, DRS, and TGA. FTIR and EDS analysis confirmed the formation and composition of TiO2 nanopowders for the doped and undoped nanoparticles. XRD analysis showed that the anatase phase was the dominant crystalline phase of the synthesized nanopowders. SEM and TEM respectively illustrated the distribution and size of the electrospun nanofibers and the nanoparticles of TiO2. DRS results showed that there was a significant shift in the absorption band edge and wavelength of Ag-TiO2 to 397 nm, followed by N-TiO2 at 396 nm compared to the commercial titania which was at 359 nm. The photocatalytic activities and antibacterial properties of these materials were tested on methylene blue dye and E.coli microorganism respectively. Ag-TiO2 immobilized on nanofibers of chitosan and PVAE had the highest photocatalytic activity compared to N-TiO2. Similar results were observed when the biocide properties of these materials were tested on E. coli.

A total of 432 1day old unsexed broiler chicks (Aviane 48) were randomly allocated to four dietary treatments (TRTS) in 72 cages. There were 18 cages per treatment and each cage allocated 6 chicks. Water and feed was provided at ad libitum. The feeding phases were, prestarter (0-7 Days), starter (8-18 Days), grower (19-28 Days), finisher (29-35 Days). The four TRTS contained graded levels of MOLM at 1000g/ton, 750g/ton, 500g/ton, and 0g/ton (control), respectively. The birds were slaughtered at 35 days of age. Breast muscles were sampled for meat, ultimate pH (pHu ), colour, drip loss over a 7 days shelf-life test. After each day’s test sub-samples were dipped in liquid nitrogen and kept at -180 C for thiobarbituric acid reactive substances determination. On Day1 and Day 7 extra sub-samples were also kept at -180 C for fatty acids analysis.The TRTS had no effect on average feed intake (AFI), feed conversion efficiency (FCE), and on average daily gain (ADG). Slaughter weight (SW), carcass weight (CW), dressing percentage and gizzard weight (GW) values were similar in all TRTS. Liver weight (LW), heart weight (HW), and gastro-intestinal fat (GIF) differed in all the TRTS, with treatment 2 having the highest value of HW (28.3±2.55), and LW (44.2±1.60) was the highest on treatment 4 . The pH values in all TRTS were constant from Day1 to Day5, reached peak on Day6, and then declined on Day7. Meat from broilers given treatment 1 with MOLM (1000g/ton) had the highest lightness (L*) values. The redness (a*) values were the highest in meat from treatment 2 (750g/ton MOLM). Treatments had no effect on yellowness (b*) values and on drip loss of the breasts. During storage L* values were high from Day1 to Day5 and decreased from Day6 to Day7. Drip loss increased with storage time as expected. Treatment 4 (control) had the highest proportions of poly-unsaturated fatty acids (PUFA) (30.3±1.87). Treatment 1 (1000g/ton) had the highest proportion of saturated fatty acids (SFA) (60.9±1.87). Treatment 1 (1000g/ton) had the highest proportion of SFA (60.9±4.30). Treatment 2 (750g/ton) had the highest n-6/n-3 ratio than other TRTS. Days had no effect (P>0.05) on PUFA, SFA, and n-6/n-3 ratio. Treatment 1 had a highest amount of malondialdehyde (MDA), treatment 4 had no effect (P>0.05) on MDA . Storage time had an effect (P<0.05) on MDA levels, except for on Day1 and Day7. Day2 had the highest amount of MDA (0.7±0.08). The use of MOLM as an additive in broiler diets reduced lipid oxidation in meat, and maintained the quality of the broiler meat during storage. It also did not have any adverse effects on the growth performance of broilers. Therefore, it has the potential to be used as an additive in broiler diets.

- Description: Infection with Helicobacter pylori has been associated with the pathogenesis of numerous stomach and gastroduodenal diseases that pose threats to public health. Eradicaftion of this pathogen is a global challenge due to its alarming rate of multidrug resistance. Consequently, to find an alternative treatment, the search is increasingly focused on new antimicrobial product from natural sources including honey. Honey has been used as medicine in several cultures since ancient time due to its enormous biomedical activities. Its beneficial qualities have been endorsed to its antimicrobial, antioxidant, anti-inflammatory properties added to its phytocomponents. In this study, the anti-H. pylori activity of South African honeys and their solvent extracts as well as the phytochemicals present in the two most active honeys were evaluated. Agar well diffusion test was used to investigate the antimicrobial activity of six honey varieties obtained from different locations in the country. Subsequently, the honeys were extracted with four organic solvents viz n-hexane, diethyl ether, chloroform and ethyl acetate employed in order of increasing polarity. The antibacterial activity of the different solvent extracts of each honey was evaluated by agar well diffusion; broth micro dilution and time kill assays. Different chromatographic techniques (Thin layer & column chromatography) were employed to enumerate the phytochemical constituents in the most active solvent extracts of Pure Honey (PH) and Champagne Royal Train (CRT); and were identified by gas-chromatography linked mass-spectrometry. Linalool pure compound was equally evaluated for anti-H. pylori activity in a bid to trace the antibacterial agent among the variety of compounds identified. Data were analyzed by One-way ANOVA test at 95% confidence interval. Crude honeys and their solvent extracts demonstrated potent anti-H. pylori activity with zone diameter that ranged from [16.0mm (crude) to 22.2mm (extract)] and percentage susceptibilities of test isolates between 73.3% (crude) and 93.3% (extract). The chloroform extracts of PH and CRT were most active with MIC50 in the ranges 0.01- viii 10%v/v and 0.625-10%v/v respectively, not significantly different from amoxicillin (P> 0.05); and efficient bactericidal activity (100% bacterial cells killed) at 1/2MIC and 4xMIC over different time intervals, 36-72hrs and 18-72hrs respectively. The appearance of bands on the thin layer chromatography (TLC) chromatogram spotted with the chloroform extracts of PH and CRT; and developed with hexane: ethyl acetate: acetic acid (HEA) and methanol: acetic acid: water (MAAW) solvent systems indicated the presence of compounds. Purification of the compounds contained in these extracts over silica gel column yielded numerous fractions which were evaluated for antibacterial activity and purity. PHF5 was the most active fraction with a mean MIC50 value of 1.25mg/mL. Volatile compounds belonging to different known chemical families in honey were identified in all the active fractions obtained from PH. Conversely, only four compounds were identified in the active fractions obtained from CRT hence the non volatile constituents could be of prime relevance with respect to antibacterial activity of this honey. Of novelty was the presence of thiophene and N-methyl-D3-azirdine compounds, essential precursors used for the synthesis of natural products and pharmaceuticals with vital biomedical properties. Linalool demonstrated potent inhibitory (MIC95, 0.002- 0.0313mg/mL) and bactericidal activity (0.0039-0.313mg/mL) against the test isolates. On the other hand, a significant difference was recorded (P < 0.05) in comparing the activity of linalool compound to the fractions. PH could serve as a good economic source of bioactive compounds which could be employed as template for the synthesis of novel anti-H. pylori drugs. However, further studies are needed to determine the non volatile active ingredients in PH and CRT as well as toxicological testing

Molecular detection and drug susceptibility of Mycobacterium tuberculosis complex in raw milk from a major dairy farm in the Nkonkobe region, Eastern Cape Province, South Africa

- Description: Mycobacterium tuberculosis complex (MTBC) organisms are the causative agents of tuberculosis in humans as well as animals. The study aimed to use molecular techniques for detection and drug susceptibility testing of MTBC in raw milk from cattle at a dairy farm in the Nkonkobe region of South Africa. Two hundred samples (100mL each) were collected and processed using the modified Petroff’s method. DNA was isolated using the Zymo Research bacterial DNA kit and amplified using the Seeplex® MTB Nested ACE assay. Drug susceptibility testing was performed using the Genotype® MTBDRplus assay. MTBC DNA was detected in 11 (6percent) of the samples tested. Resistance to both rifampicin and isoniazid was detected in 90.9percent of the positive samples. The most frequent rpoB mutations detected were H526Y (90percent), H526D (80percent), S531L (60percent) and D516V (20percent). No mutation was detected in the katG gene. All isoniazid resistant samples harboured mutations in the inhA gene. The most frequent (100percent) mutation conferring low level isoniazid resistance was the T8A substitution. The inhA mutations C15T, A16G and T8C were equally represented with 60percent frequency. A high prevalence of multi-drug resistance was noted in the Nkonkobe region. Therefore, the results of this study have clinico-veterinary and epidemiological significance and calls for further studies and necessary actions to delineate the situation. , Thesis (MSc) -- Faculty of Science and Agriculture, 2012

- Description: Very little is known about the spatio-temporal ecology of the yellow mongoose Cynictis penicillata, despite this small carnivore being widely distributed in the southern African subregion. Until the present study, activity patterns and daily movement distances had only been investigated over a single season, and information on home range sizes was only available from a few populations. Here, I aimed at determining the activity patterns and the space use of yellow mongooses over the different seasons of the year in the Great Fish River Reserve (Eastern Cape, South Africa). In addition, I tested some predictions related to environmental factors (variations in photoperiod, climatic conditions and food availability over the year), as well as life cycle (reproduction) and morphological and physiological characteristics (sexual dimorphism and thermoregulatory aspects associated to the possession of an elongated body). From September 2005 to August 2011, spatio-temporal data were collected through 102 continuous tracking sessions (84 during the day and 18 during the night) of 12 adult individuals (seven males and five females) fitted with motion-sensor radio-collars. Additional data were obtained through semi-continuous tracking sessions and random locations. Yellow mongooses were essentially diurnal and crepuscular. Activity levels averaged 57 percent during the day (n = 11,807 fixes) and only 11 percent at night (n = 3,623 fixes). Yellow mongooses emerged from their burrows on average (± SD) 85 ± 62 min after sunrise (range: –11 to +369 min) and returned 21 ± 84 min before sunset (range: –518 to +225 min). The duration of the daily activity period varied between 12 min and 15 h 36 min, with a mean of 9 h 28 min. Both sexes exhibited some overground activity after sunset, but this was most pronounced in males than in females. Nocturnal activity outside the activity period was generally restricted to short underground bouts of activity occurring at anytime of the night. Diurnal resting, with up to 11 bouts per activity period, occurred in 98.8 percent of activity periods. Overall, yellow mongooses were active during 68 ± 17 percent of the time spent outside the burrow. Photoperiod acted as a zeitgeber and activity was negatively affected by adverse climatic conditions (very hot or conversely low temperatures, rain, wet soil) and probably low terrestrial arthropod activity/availability. No intersexual difference in activity levels was found, although the higher body mass of males (on average 16 oercent) implies higher metabolic demands. The total home range size (MCP 100 percent ) averaged 0.55 ± 0.65 km2 (range: 0.10–2.36 km2) and the related total perimeter measured on average 2.80 ± 1.71 km (range: 1.17–7.36 km). Male home ranges were on average more than twice large than those of females, but this difference did not reach statistical significance. However, males had larger daily home ranges (0.13 ± 0.15 km2) than females (0.05 ± 0.05 km2). In addition, daily movement distances of males were longer (1.99 ± 1.07 km) than those of females (1.29 ± 0.66 km). Males also travelled at a faster speed (0.29 ± 0.13 km/h) than females (0.18 ± 0.07 km/h). Overall, sex clearly appeared to play a role in the spatial ecology of yellow mongooses, but the underlying explanation does not seem to be linked to sexual dimorphism (body mass) or reproductive activity. Whilst the role of food availability on the space use of yellow mongooses is unclear, it is likely that low temperatures negatively affect yellow mongoose movements, as they do for their activity levels.

- Description: Wild edible plants are an indispensible part of diets in some of the poor resourced rural societies. In many less resourced households a main meal comprises of a starch staple which is accompanied by a supplement of vegetables that provide the other nutritional elements as well as flavour to the meals. The vegetable relish (isishebo) thus, becomes an important component of the diet. The sustenance of many such resource poor indigenous societies over many generations has been as a result of their reliance on a readily available supply of their nutritional requirements, where edible wild plants formed the major food source. Wild edible plants provided the most important source of nutritional supply even during times of drought and famine. However, lifestyle changes have caused a serious reduction of the gathering and use of the wild edible vegetables, knowledge about them and the local culture are also lost with them. Unfortunately, others have come to regard their traditional food resource base as primitive and inferior and have come to rely on new exotic food plant whose supply is unstable and narrow compared to the wide variety of wild edible plants. Even when drought challenges play destruction with food availability, resulting in food shortages due to the reliance on cultivated crops that are poorly adapted to local conditions, communities cannot go back to the unknown food plants. However, detailed scientific investigations are needed to understand and preserve wild food uses. This study was undertaken to add to that body of knowledge. It started with an ethnobotanic survey of the use of wild food plants in 10 selected rural villages and peri-urban dwellings in the Amathole District of the Eastern Cape. These rural areas were chosen because of the belief that there are still individuals who possess the wealth of traditional knowledge within those remote areas in addition to that the villages are surrounded by indigenous vegetation from which they could be harvesting some indigenous food plants. Furthermore, the relationship between people and plants are often clearer in indigenous/traditional societies since the link between production and consumption is more direct. Besides the survey, this research employed various methods including Inductively Coupled Argon (ICP) analyses of minerals and vitamins, determination of phytochemicals and antioxidant/bio-activity that of, higher performance liquid chromatography (HPLC) determination of other vitamins, assessment of proximate and anti-nutrient compositions of raw and cooked samples of plants, scanning electron microscopy (SEM) and energy dispersive x-ray spectroscopy (EDXS) to look into the micro-morphology of leaves for details that might have a bearing in the nutrient composition of the wild edible vegetables. This was done with the aim to explore and highlight the wild-food-plant base of indigenous people, to investigate the nutritional value of the selected frequently consumed wild vegetables and to recommend ways of educating people on the value of such plants and the need to promote their continued sustainable use as well as preservation of them for the benefit of people who cannot afford a continuous nutritious meal particularly among rural communities. Twenty-five (25) indigenous wild food plants which included leafy vegetables, nuts and fruits species were identified as being arbitrarily gathered and utilised as food, of which seven wild leafy vegetable species were often mentioned as common among the 15 dwellings surveyed and most frequently consumed by the respondents. Two additional plants were also frequently used as flavoring and in their raw state respectively. Wild edible vegetables were mainly eaten boiled as ‘laxa’, mixed with maize meal as ‘imifino’, fried as relish ‘isishebo’ or raw for both snacking purposes and medicinal functions. Wild edible plant species in the Amathole District of the Eastern Cape-South Africa were documented with respect to their uses and knowledge or identification. Four indigenous wild vegetables (Chenopodium album, Solanum nigrum/nodiflorum, Amaranthus dubius and Urtica lobulata) frequently consumed by the indigenous communities in the study region were selected. All the plant species had a remarkably high content of pro-anthocyanidins (between 58.42 and 65.18 mg/g) and also exhibited high radical scavenging activity in vitro. S. nigrum and C. album were found to be rich sources of ascorbic acid, having 1.05mg/100g and 1.01mg/100g, respectively. The entire wild, traditionally edible plants contain reasonably good concentrations of riboflavin while U. lobulata was found to contain maximum amount of thiamine. All plant species were rich sources of minerals and vitamins (both water soluble and fat soluble). Appropriate cooking methods are to be used to enhance the bio-availability of vitamins and nutrients in general. Cooking generally improved the nutritive value in certain wild vegetables. However, some nutrients were drained away. For example micro-nutrient and anti-nutrient component in the ash were reduced in cooking. The EDXS analysis has shown that the indigenous, wild vegetables in the Amathole District of South Africa are excellent sources of minerals, while the mineral crystals present at the level of the mesophyll of the indigenous wild vegetables were probably mixtures of calcium oxalate, calcium sulphate and silica. Saponin, tannins, oxalates, phytates and alkaloids are present in all the indigenous wild vegetables, but in minimal safe quantities. Nutritional analysis of some wild food plants demonstrates that in many cases the nutritional quality of the plants is comparable and in some cases even superior to domesticated exotic varieties. Knowledge about gathering and use of some wild food species is common among the rural dwellers. However, most uses are known by few elderly informants only. - Description: The main objective of this study was to critically investigate the mediating role that the use of mobile technology plays in the linkage between customer satisfaction and customer loyalty in a semi-urban environment. The research problem focused on the application of mobile technology in the linkage between customer satisfaction and customer loyalty in the clothing industry in King Williams Town. In accomplishing this objective, this research study hypothesised that mobile technology plays a mediating role in the linkage between customer satisfaction and customer loyalty of customers of clothing retailers in South Africa. Multiple regression was conducted to test for the first hypothesis. The first secondary objective aimed at establishing if there is disconfirmation between customer expectations and the performance of clothing retail outlets in King Williams Town. In exploring this objective, it was hypothesised that there is negative disconfirmation between customer expectations and performance. A paired sample test was conducted to test for this hypothesis. In addition, the influence of service quality and customer satisfaction on customer loyalty was explored. To accomplish these, it was hypothesised that both service quality and customer satisfaction have an influence on customer loyalty. Pearson’s Correlation and the Chi-Square Test were applied to test the influence of service quality and Customer satisfaction on Customer loyalty. Data was analysed using the Statistical Package for Social Sciences (SPSS) statistical software. Regression analysis, chi-square tests, Paired sample tests and correlation reports were used to analyse data inferentially. Reliability and validity of the research was also tested using Cronchbach’s alpha test. The results show that mobile phones have become a basic necessity for customers and that mobile technology plays a mediating role in the linkage between customer satisfaction and customer loyalty; however, the mediation is partial. The results also show that there is positive disconfirmation between customer expectations and retailer performance; that both service quality and customer satisfaction have a positive correlation with customer loyalty and that mobile technology enhances customer loyalty. Finally, the results show that the total service experience of customers is favourable vii because the five components of service quality were rendered at a high level of service quality. The study offers several managerial recommendations. Firstly, retail managers should make strategic investments in mobile technology and use this technology to the full to reap the benefits of technologically advanced mobile communication. Secondly, managers should be creative in their use of this mobile technology to enhance customer satisfaction and, hence, customer loyalty. Thirdly, managers should prioritise the improvement of all five components of service quality in order to enhance customer loyalty. Finally, managers should provide a customer-centred service experience that exceeds customer expectations for service quality and mobile communication, one that maximises customer satisfaction and loyalty. The implementation of these recommendations could result in clothing retailers providing a customer-focused service experience in all five aspects of service quality. To successfully implement these recommendations, clothing retailers should: ensure that expectations created, could be accomplished or exceeded to build and enhance customer satisfaction; create expectations that are realistic to avoid negative disconfirmation and hence customer dissatisfaction; implement innovative and creative mobile communication strategies to build and enhance customer loyalty; and finally, improve loyalty programmes to enlarge the customer base.

- Description: This study was designed was to determine the effects of additive supplementation of Moringa oleifera leaf meal on growth performance, digestibility, digestive organ size, intestinal integrity, bone ash content and bone breaking strength, as well as meat yield and quality of broiler chickens. A total of 2400 day-old unsexed Cobb-500 broiler chicks were randomly allocated to 5 treatment groups: T1, positive control, 668 g/ton Salinomycin and 500 g/ton Albac; T2, T3 and T4 contained graded levels of MOLM at 1 percent, 3 percent and 5 percent of dry matter (DM) intake, respectively; and T5, a negative control (0 percent additives) in a complete randomized design experiment. Except for week one, FI and FCR was highest (P < 0.05) in T4 birds; while T1 birds had the highest FI in the period of 22 to 27d (P < 0.05). Throughout the production period, birds supplemented with MOLM had the highest BW (P < 0.05) than the birds fed the control diets. Feed intake (FI) and feed conversion ratio (FCR) among treatments was highest (P < 0.05) in T4 birds during the period of 8 to 14 d; and was highest (P < 0.05) for T1 birds in the period of 22 to 27d. Protein efficiency ratio (PER) and energy utilization efficiency (EEU) ratios were statistically significant among treatments (P < 0.05). However, dietary treatments had no effect (P > 0.05) on the weights of the heart, liver, spleen, or the gizzard, although the bursa for T2 birds was the lightest (P < 0.05); while gizzard erosion score was highest in T2 birds. All of the nutrients measured, except for fat, had negative intercepts that were significantly different (P < 0.05) from zero, indicating the presence of endogenous fecal losses. Tibiae length (TL) was highest in T2. The dried defatted weight (DW) was heaviest (P < 0.05) for T3 (11.20 ± 0.347) and T5 (11.08 ± 0.328). A positive correlation (r = 0.667; P < 0.01) between TW and DW was observed. There were no dietary effects on bone breaking strength (BBS), but T1 tibiae had highest resistance to breaking force (T1 > T4 > T3 > T2 > T5). Calcium was highest (P < 0.05) in T1; and lowest inT2 and T5. Phosphorus levels were lowest (P < 0.05) in T1; and highest (P < 0.05) in T5 compared. The highest Ca: P ratio was obtained in T4 (P < 0.05); while the ash percent was highest (P < 0.05) in T1. Drip loss increased as L* values increased; and a negative correlation was observed between L* and pH. On D1, C18: 0 and C22 in T2, while C15:0 was highest in T4. On D1, C20:2, C20:3n6 and C22:6n3 were highest in T2 (P < 0.05); T4 had the highest C18:3n6 (P < 0.05), while C20:2 was highest in T5 (P < 0.05). The P/S ratio on D1 was highest in T4; while n-6/n-3 was highest in T1; and n-3 was highest in T3. On D8, the n-3 was highest in T1 (P < 0.05). Results of the current study show that supplementation of M. oleifera leaf at additive levels of up to 5 percent of the bird’s DMI does have the potential to influence the bird performance without any detrimental effects on nutrient utilization, bird health, bone strength and/or meat quality, which can be concluded that MOLM enhanced the bird’s genetic potential for optimal productivity.

- Description: This study investigated the impact of intrinsic motivation and job involvement, with employee retention acting as a mediating variable. As most organisations want to retain their best employees and sustain their organisational performance, this study strives to add to the body of knowledge on motivation, job involvement and employee retention, with specific reference to selected departments within the Eastern Cape Provincial Government in the South African context. Intrinsic motivation and job involvement were used as the independent variables. Employee retention is the dependent variable, which was measured using the cumulative Michigan Organisational Assessment Two-item Questionnaire (Camman, Fichman & Klesh, 1979) and a third assessment item from Landau & Hammer (1986). A quantitative relational design was employed to investigate the degree to which intrinsic motivation and job involvement predict an employee’s intention to quit their job. The data were collected through a self administered questionnaire from a convenience sample of 160 employees within the selected government departments in the study. The empirical results obtained from the data; using the Pearson correlation coefficients, indicated significant relationships between intrinsic motivation, job involvement and employee retention. Evidence was further found that employees’ intrinsic motivation and job involvement influenced the extent of employees’ intention to quit. Furthermore, a multiple regression analysis of the data indicated that intrinsic motivation is the strongest predictor of employee retention, followed by job involvement.

- Description: Winter rotational cover crops (WRCC) are often used to boost soil fertility and plant nutrition. However, selection and use of WRCC for soil physical improvement is usually overlooked. The objective of this study was to determine the effects of WRCC on soil strength, aggregate stability and water conservation of a hardsetting soil. The soil physical properties were determined after four rotations of growing monocultures of vetch (Vicia dasycarpa cv. Max), lupin (Lupinus angustifolius cv. Tanjil) and oats (Avena sativa cv. Sederberg) and after two rotations of growing bicultures of oats (Avena sativa cv. Pallinup) and vetch (Vicia dasycarpa cv. Max) across two soil layers, 0 to 15 cm and 15 to 30 cm. The individual WRCC and a weedy fallow constituted the treatments in the monoculture study whilst in the biculture study the various combinations of WRCC namely; 90% oat plus 10% vetch (O90V10); 70% oat plus 30% vetch (O70V30) and 50% oat plus 50% vetch (O50V50) and a weedy fallow constituted the treatments. After four rotations with cover crop monocultures, oats significantly (P ≤ 0.05) reduced penetration resistance (PR) whilst vetch increased PR in both soil layers compared to the weedy fallow control. The effect of the biculture treatments was only experienced within the 15 to 30 cm depth. The treatments O50V50 and O70V30 increased the PR compared to the control. The WRCC in monoculture significantly increased the soil aggregate stability relative to the control in both soil layers. Vetch, lupin and oats resulted in a 41.7%; 20.4% and 15.7% increase in MWD in the 0 to 15 cm soil layer and 47.2%; 44.2% and 39.7% in the 15 to 30 cm depth, respectively. An increase in aggregate stability was associated with increased macro-aggregation. Under the biculture, WRCC slightly increased, non- significantly, the aggregate stability. Both hot water and dilute acid extractable polysaccharides showed no significant correlation with aggregate stability in the two studies. Oats monoculture resulted in a significant difference (P ≤ 0.05) on cumulative infiltration compared to the control. However, after 2 h vetch and lupin showed no significant difference from the control on cumulative infiltration. Oats resulted in a 7.8% increase in final infiltration rate (FIR) whilst vetch and lupin reduced FIR by 9% and 16.7% respectively, compared to the control. Bicultures of oats and vetch significantly (P ≤ 0.05) increased cumulative infiltration compared to the weedy fallow control. A similar significant increase in FIR was also observed under bicultures. The treatments O50V50; O90V10 and O70V30 resulted in a 163.3%; 113.3% and 105.4% increase in FIR respectively, compared to the control. Cover crop monocultures significantly (P ≤ 0.05) increased plant available water (PAW) compared to the weedy fallow, with vetch, oats and lupin resulting in a 28.3%; 22% and 23.9% increase respectively, in PAW. However, no significant differences were observed on PAW after two rotations with bicultures. Compared with winter weedy fallow, WRCC improved most of the soil physical properties under study, with the most suitable results expected under bicultures compared to monocultures. Under CA, selection of WRCC like oats, vetch and lupin, one should therefore take into consideration their effects on soil physical properties as a selection criterion and not biomass and fertility alone.

- Description: This is a study on the prevalence of refractive errors and patients requiring refractive services at 15 eye clinics in the Amathole, Chris Hani, Joe Gqabi and OR Tambo District Municipalities of the Eastern Cape. This is an area characterised by extreme poverty where the cost of an eye examination and prescription spectacles remains financially unobtainable for most. Optometry services are provided mainly by private optometrists who service the small proportion of the population that can afford them. Adults and children remain house bound or are labelled as dull and unproductive simply because they don’t have access to an eye examination and a pair of spectacles. Purpose The purpose of the study is to identify patients with refractive errors and those requiring refractive services at the 15 eye clinics in the Eastern Cape. Another purpose is to describe the refractive services that are available to patients attending health facilities, where the eye clinics are conducted. Lastly, the purpose is to explore the possibility of nurses providing refractive services independently or under the supervision of optometrists to supplement the lack to refracting and dispensing services. Study Method A quantitative and qualitative non-experimental descriptive design was used. Research involved the analysis of Vision Care’s eye clinic records collected from 15 eye clinics from January 2010-June 2010. Semi-structured interviews were conducted with 30 nurses working at the 15 health facilities where the eye clinics were conducted using purposive sampling. The quantitative data was analysed using excel spreadsheets and graphs and qualitative data was analysed using coding and categorizing methods. Conclusion According to Vision Care’s data of the patients assessed, 19.2 percent had a refractive error and 54 percent of the patients required refractive services. It is estimated that 71.41 percent of the patients had a refractive error according to the optometrist. Although there were some organisations active in the eradication of cataracts, there was little healthcare available in the form of refraction services. xiv Patients needed to travel an average of 63.8kms to access refraction services against the backdrop of poor roads, poverty and unemployment. 28 out of 30 nurses either ‘strongly agreed’ or ‘agreed’ that nurses could be trained to perform refractions and dispense spectacles. 29 out of 30 nurses thought that this would have a positive impact on eye care. Further research is necessary to assess the feasibility of implementing a nurse operated refractive program and whether it should be within their scope of practice to refract and dispense spectacles.
