- Description: Over several decades past, developing countries have received increased amounts of Foreign Direct Investment (FDI). This form of investment has been welcomed because of the perceived benefits attached to it. FDI is seen as an important driver of economic development for many nations. For South Africa specifically, GDP growth rates have remained less than required, unemployment rates have reached staggering levels, poverty and inequality levels are increasing and the list goes on. Considering the perceived benefits of FDI, one may argue that FDI can play a crucial role in reducing the mentioned challenges facing the nation, however, only if directed to initiatives contributing to growth and employment. The 2015 Investment Policy Framework for Sustainable Development includes an action menu promoting investment in sectors relating to the achievement of the Sustainable Development Goals (SDGs). Therefore, this study is aimed at investigating the relationship between sector FDI and sector growth in addition to investigating the effect of sector FDI on sector employment over the period 2000Q1 to 2016Q4 for six of South Africa’s economic sectors. The reason for such a study is based on the premise that developing nations such as South Africa lack sound trade and industrial policies favorable to foreign investors. This then leads to the nation failing to attract higher volumes of FDI which could be used to address structural challenges facing the country. It is therefore important to identify sectors in which FDI has resulted in growth and employment so that when policies are considered, the right FDI is targeted. A comprehensive review of existing theoretical and empirical literature showed that FDI does result in economic growth for developed and developing countries, although FDI crowds out domestic investment in the short run. Literature on the effect of FDI on employment showed diverse effects. Some studies found FDI to increase employment overall, other studies found FDI to increase employment only during periods of restructuring and some studies found FDI to result in job losses. For South African sectors, the present study finds that the financial services sector receives the highest volume of South African FDI, followed by the mining and quarrying sector and the manufacturing, however, FDI in all six sectors under study is associated with increased growth and employment. This finding suggests that the financial services sector has received increased volumes of FDI as a result of financialization of the South African economy. It is this increased FDI in the financial services sector that is directed to income redistribution from the real sector to the finance sector. This study employed econometric techniques and methods of analysis to investigate the relationship between sector FDI and sector growth, and the effect of sector FDI on sector employment. Panel cointegration tests were conducted for all six sectors included in the study to establish if long run equilibrium relationships exist among integrated variables. The Johansen-Fisher panel cointegration test revealed that there is evidence of cointegration in four of the six sectors. Since cointegration was established, the study proceeded to perform the Dumitrescu-Hurlin panel causality analysis and estimate a Panel Vector Error Correction Model (VECM). Results from the causality analysis found a unidirectional causality relationship between FDI and GDP growth, while the panel VECM found FDI to have a significant effect on growth in all sectors. The Seemingly Unrelated Regression (SUR) model employed to investigate the effect of FDI on employment found FDI to have an insignificant effect on employment in all sectors included, although the signs of the coefficients suggest that FDI is associated with increased employment and rising wages is associated with increased productivity growth. Since this study finds that FDI is associated with increased GDP growth in all six sectors under study, policy makers should devise strategies to attract FDI in sectors such as the transportation, storage and communication sector and the electricity, gas and water sector as FDI in these sectors are associated with increased growth however, they receive very low levels of FDI. There are a number of reasons for this, therefore, government institutions and policy makers should investigate the reasons for these low levels of FDI inflows into these sectors so that they can devise further strategies to address these reasons and perhaps attract higher levels of FDI into these sectors. Spillover benefits play a major role in host nations participating in FDI therefore, prior to entering into bilateral treaty agreements, policy makers should ensure that foreign investors are compelled to create jobs, offer training and qualifications etc. through their investments so that some of the SDGs can be achieved. Additionally, this study finds a positive, statistically insignificant relationship between FDI and employment. FDI may not have a significant relationship on employment due to jobless growth and capital-intensive growth rather than labor-intensive growth. Such a situation calls for government intervention. Skills shortage is a rising problem in South Africa; therefore, investors choose to employ advanced technologies rather than people. The in vitro biological activities of synthesized molecules were evaluated against Plasmodium falciparum malaria parasites (strain 3D7), Trypanosoma brucei brucei parasites (strain 427) and HeLa cells. All the compounds displayed modest or no activity at a concentration of 20 μM and percentage viability of >50 % was often observed. Except for compound 9o, none of the final compounds exhibited cytotoxic effects against HeLa cells at 20 μM.

The development of biological tools to aid in the genetic investigation of the black (Diceros bicornis) and white (Ceratotherium simum) rhinoceros mitochondrial genomes

A decline in the populations of these species has resulted due to human activities such as habitat fragmentation and poaching. This has contributed to the loss of genetic diversity amongst the black and white rhinoceros. Conservation and anti-poaching efforts are needed to help maintain genetic diversity. These efforts could be improved through the development of non-invasive techniques to examine DNA from threatened animals. The aim of this research was to develop a molecular technique which would allow for the identification of the black and white rhinoceros and to develop a molecular technique which would allow for intraspecies genetic variation to be examined. DNA extractions were performed on matched faecal and tissue samples that were collected from two regions in South Africa. Polymerase chain reaction (PCR) primer sets were designed to investigate several regions of the rhinoceros mitochondrial genome. PCR optimisation was completed for the target regions. Sequencing was conducted on all final PCR products. The cytochrome c oxidase subunit 1 (COIi) gene allowed for the rhinoceros family to be identified. This region was digested with the HindIII restriction enzyme, which allowed for the specific identification of either the black or white rhinoceros. A subsequent region of the cytochrome c oxidase subunit 1 (COIii) as well as the D-loop, hypervariable regions (HV1 and HV2), cytochrome b (cytb) and 16s rRNA regions were investigated. These regions displayed potential for establishing geographic origin for black rhinoceros samples, whereas the D-loop and HV2 show potential for the white rhinoceros. The white rhinoceros displayed sequence variation in the HV2 and cytb region, while variation was observed in the COIi and HV1 for the black rhinoceros. All investigated target regions allowed for the rhinoceros family to be identified. The COI (COIi and COIii), HV2 and cytb regions allowed for the subspecies of rhinoceros to be identified, however the D-loop was not able to identify the white rhinoceros species. The 16s rRNA and HV1 regions allowed for the correct subspecies of rhinoceros to be identified, however as the primers were only compatible for the black rhinoceros therefore a subsequent investigation is required for the white rhinoceros. The establishment of this novel PCR based technique to identify white and black rhinoceros will allow for efficient species identification in wildlife forensic cases. A biological method was established to study intraspecies variation for the white and black rhinoceros; however the investigated target regions did not yield sufficient genetic variation. The core techniques developed in this study will be valuable for future studies that wish to investigate genetic variation in mammal species.

Exploring para-thiophenols to expand the SAR of antimalarial 3-indolylethanones

- Description: According to the WHO, malaria is responsible for over half a million deaths annually especially in populations from disadvantaged settings. Although there has been a documented improvement in the mortality rates, malaria has proved to be a global emergency. Mostly affecting the poor population, this disease is perpetuating a vicious cycle of poverty in the developing world as current preventive measures are not adequate unless adopted in addition to effective treatment. However, there has been a worldwide increase in resistance to available treatment which presents a need for novel, affordable treatment. A study conducted in our laboratory identified two hit thiophenol containing compounds 2.24 and 2.25. These molecules provided initial insight into the SAR and potential pharmacophore of this class of compounds. We decided to further explore these compounds by making bioisosteric replacements to optimize the structure as we monitor the effect of these modifications on the anti-plasmodial activity. The synthetic pathway to form the target compounds of our study comprised of three steps which were initiated by the Friedel-Crafts acetylation of the indoles resulting in compounds 3.5 - 3.7. A bromination step followed which yielded the -bromo ketones (3.8 - 3.11). Some of the thiophenols (3.14 and 3.16) were not readily available in our laboratory and so were synthesized for the final synthetic step. This step involved the nucleophilic displacement of the -bromine to generate the -aryl substituted 3-indolylethanones (3.17 - 3.27). The thioethers displayed improved antimalarial activity from 2.24 and 2.25 against the chloroquine sensitive 3D7 Plasmodium falciparum strain. In addition, these compounds were non-toxic against HeLa cells which indicated this potential novel class of antimalarials is selective for the malaria parasite as hypothesized in the previous study conducted in our laboratory. In an attempt to predict the bioavailability of some of our compounds, in silico studies were conducted revealing that these compounds could be passively absorbed by the gastrointestinal tract, a positive result for bioavailability purposes. - Description: According to the WHO, malaria is responsible for over half a million deaths annually especially in populations from disadvantaged settings. Although there has been a documented improvement in the mortality rates, malaria has proved to be a global emergency. Mostly affecting the poor population, this disease is perpetuating a vicious cycle of poverty in the developing world as current preventive measures are not adequate unless adopted in addition to effective treatment. However, there has been a worldwide increase in resistance to available treatment which presents a need for novel, affordable treatment. A study conducted in our laboratory identified two hit thiophenol containing compounds 2.24 and 2.25. These molecules provided initial insight into the SAR and potential pharmacophore of this class of compounds. We decided to further explore these compounds by making bioisosteric replacements to optimize the structure as we monitor the effect of these modifications on the anti-plasmodial activity. The synthetic pathway to form the target compounds of our study comprised of three steps which were initiated by the Friedel-Crafts acetylation of the indoles resulting in compounds 3.5 - 3.7. A bromination step followed which yielded the -bromo ketones (3.8 - 3.11). Some of the thiophenols (3.14 and 3.16) were not readily available in our laboratory and so were synthesized for the final synthetic step. This step involved the nucleophilic displacement of the -bromine to generate the -aryl substituted 3-indolylethanones (3.17 - 3.27). The thioethers displayed improved antimalarial activity from 2.24 and 2.25 against the chloroquine sensitive 3D7 Plasmodium falciparum strain. In addition, these compounds were non-toxic against HeLa cells which indicated this potential novel class of antimalarials is selective for the malaria parasite as hypothesized in the previous study conducted in our laboratory. In an attempt to predict the bioavailability of some of our compounds, in silico studies were conducted revealing that these compounds could be passively absorbed by the gastrointestinal tract, a positive result for bioavailability purposes. The carbon atom bonded to the oxygen atom shifted upfield, while the carbon atom bonded to the nitrogen atom shifted downfield. Factors mentioned in the literature that has an influence on the hydrolytic stability on the heterocyclic borinate motif have been taken into consideration and verified for the systems researched in this study. The substitution at the nitrogen atom with one ethyl group caused the greatest increases in the N→B bond length (27) 1.714 Å, due to the increase in the nitrogen Lewis basicity, making it the least hydrolytically stable, these include compounds (24-26). Furthermore, DFT calculations were performed by employing Becke’s three parameter hybrid exchange functional with Lee–Yang–Parr functional (B3LYP) method using the Gaussian 16 program package (Rev. We first classify the left-invariant control affine systems (under detached feedback equivalence). We provide a complete list of normal forms, as well as classifying conditions. As a corollary to this classification, we derive controllability criteria for control affine systems on SE(1,1). Secondly, we consider quadratic Hamilton-Poisson systems on the (minus) Lie-Poisson space se(1,1)*. These systems are classified up to an affine isomorphism. Six normal forms are identified for the homogeneous case, whereas sixteen representatives (including several infinite families) are obtained for the inhomogeneous systems. Thereafter we consider the stability and integration of the normal forms obtained. For all homogeneous systems, and a subclass of inhomogeneous systems, we perform a complete stability analysis and derive explicit expressions for all integral curves. (The extremal controls of a large class of optimal control problems on SE(1,1) are linearly related to these integral curves.) Lastly, we discuss the Riemannian and sub-Riemannian problems. The (left-invariant) Riemannian and sub-Riemannian structures on SE(1,1) are classified, up to isometric group automorphisms and scaling. Explicit expressions for the geodesics of the normalised structures are found.

Although this pattern is widespread across Bantu, some variations in the ordering of these extensions have been observed in some languages (Kathupa 1991, Simango 1995, Sibanda 2004, among others), which suggests that the template is not as rigid as one might think. This study investigated the morphotactic constraints between four verbal extensions in isiXhosa, the Causative, Applicative, Reciprocal and Passive. It focused on the morphotactics of the transitivising extensions (Causative and Applicative) in the first instance, and morphotactics of the detransitivising extensions (Reciprocal and Passive) in the second instance. The study found that although the co-occurrence of causatives and applicatives is a regular feature in Bantu languages, isiXhosa has restrictions on the co-occurrence of these extensions on some verbs. The study also found that although Causative-Applicative is the expected order the language permits Applicative-Causative in certain contexts. With respect to the detransitivising extensions, the study revealed that there are limited contexts in which these extensions co-occur and, crucially, that these extensions are freely ordered in the language.

- Description: This thesis forms part of the Nelson Mandela University biomass and nanotechnology project, which aims at using agricultural waste for many applications, including biomedical applications, water treatment, and agricultural films. Pineapple leaf fiber (PALF) is one of the natural fibers with the highest cellulose content (about 80%), and they produce about 2.5–3.3% of the weight of green leaves. However, the other parts of the pineapple fruit, such as the leaves and crown, are still treated as waste due to a lack of understanding of the benefits and risks of these parts of the pineapple fruit. The transformation of PALF as a renewable and abundantly available natural residue into cellulose fibers (CFs) will reduce the waste accumulated in the atmosphere, and it will add value to the cultivation of pineapple. In the biomedical industry, CF is very promising in terms of drug delivery. Cellulose acetate is a remarkable biomaterial extensively used in biomedical applications due to its properties such as being non-toxic, non-irritant, biodegradable, heat resistant, and less hygroscopic. In this research, the isolation, functionalization, and characterization of cellulose from PALFs were conducted. The PALFs were dried and ground into small particles before being alkali treated with sodium hydroxide to remove the non-cellulosic components. Moreover, the remaining lignin in the residue was removed using sodium chlorite and acetic acid; this process is known as the bleaching process. The bleaching process resulted in white CFs, which was an indication of the successful removal of the non-cellulosic material. The obtained CFs were further functionalized using acetic acid, acetic anhydride, and sulfuric acid, and this reaction is known as the acetylation reaction. Subsequently, both treated, untreated, and functionalized CFs were characterised using the following instruments: FTIR, SEM, SPM, TGA, XRD, and XPS. In the FTIR data of raw PALFs, the following functional groups were more pronounced: C-O, C=C-O, and C=O, whereas peak C-O-C disappeared and peak CH2, C-O, were less pronounced. These FTIR results confirmed the successful removal of non-cellulosic components. The SEM and SPM images visually suggested the partial removal of hemicellulose and lignin from the raw PALFs after the chemical treatment, which are the cementing materials around the fiber bundles. The TGA studies revealed good thermal stability for the isolated CFs from PALFs. The XRD diffractograms of the PALFs showed a cellulose I crystal pattern where the peaks were located at angular positions (2θ) of around 18°, 26°, and 41°. Lastly, after the functionalization of CFs, the cellulose structure remained intact (not damaged); instead, the newly ix introduced functional groups (the acetyl group) were attached at the ends of the cellulose structure. Moreover, the remaining lignin in the residue was removed using sodium chlorite and acetic acid; this process is known as the bleaching process. The bleaching process resulted in white CFs, which was an indication of the successful removal of the non-cellulosic material. The obtained CFs were further functionalized using acetic acid, acetic anhydride, and sulfuric acid, and this reaction is known as the acetylation reaction. Subsequently, both treated, untreated, and functionalized CFs were characterised using the following instruments: FTIR, SEM, SPM, TGA, XRD, and XPS. In the FTIR data of raw PALFs, the following functional groups were more pronounced: C-O, C=C-O, and C=O, whereas peak C-O-C disappeared and peak CH2, C-O, were less pronounced. These FTIR results confirmed the successful removal of non-cellulosic components. The SEM and SPM images visually suggested the partial removal of hemicellulose and lignin from the raw PALFs after the chemical treatment, which are the cementing materials around the fiber bundles. The TGA studies revealed good thermal stability for the isolated CFs from PALFs. The XRD diffractograms of the PALFs showed a cellulose I crystal pattern where the peaks were located at angular positions (2θ) of around 18°, 26°, and 41°. Lastly, after the functionalization of CFs, the cellulose structure remained intact (not damaged); instead, the newly ix introduced functional groups (the acetyl group) were attached at the ends of the cellulose structure. - Description: Ursolic acid UA, a pentacyclic triterpenoid that is commonly found in many medicinal herbs and fruits, has been identified as a potential source of therapeutic agents because of its potent biological effects, which include its potential anticancer and antimicrobial activities. However, its limited solubility, rapid metabolism and poor bioavailability inhibit its clinical applications. Numerous UA derivatives have been prepared over the past years in an effort to mitigate the drawbacks associated with UA, as new chemical entities for the treatment of various infections. There is very little progress in the discovery of efficient UA derivatives. In this study, a class of ester and amide-linked ursolic acid-based hybrid compounds fused with selected pharmaceutical scaffolds were successfully synthesized using amidation and esterification reactions and tested for antibacterial and cytotoxicity activities. Hybridizing UA with other known pharmaceutical scaffolds has the potential of overcoming its drawbacks. FT-IR, Mass Spectroscopy, and 1H13C-NMR spectroscopy were used to confirm the structures of the synthesized hybrid compounds. Among the tested ester-linked hybrid compounds in Chapter three, compounds 3.14-3.19,3.21, 3.34, 3.31, and 3.30 demonstrated significant antibacterial activities against some tested bacteria, with MIC values of 15.625 μgml. Furthermore, the in vitro cytotoxicity of these hybrids was determined using the MTT assay against three human tumor cell lines MCF7, MDA-MB-231, and HeLa cells. Compounds 3.19 and 3.34 were found to have better cytotoxic activity when compared to ursolic acid, with IC50 values of 46.99 and 48.18 μg ml respectively. Both compounds revealed more promising docking results, presenting favourable binding interactions as well as better docking energy against the MCF 7 protein target compared to the parent compound ursolic acid. In Chapter Four, among the tested amide-linked hybrid compounds, Compounds 4.17 and 4.24 demonstrated significant antibacterial activity against the majority of bacterial strains with MIC values of 15.625 gml. Compound 4.24 exhibited a MIC value of 15.625gmL against BS, SA, PV, KO, PM, and EC. Compound 4.23 was more cytotoxic to HeLa cells than ursolic acid. Furthermore, molecular docking calculations revealed that compound 4.16 strongly binds to the protein epidermal growth factor receptor while e compounds 4.17 and 4.24 showed a strong binding affinity for the methionyl-tRNA synthetase. Thermal analysis using thermogravimetric analysis (TGA) confirmed the excellent thermal stability of synthesized tetra 4-(4- propargyloxy) phenoxy phthalocyanines. The metal complexes, CoTPrOPhOPc and MnTPrOPhOPc, exhibited better thermal stability when compared to H2TPrOPhOPc. The residual percentage weight remaining was approximately 70% for CoTPrOPhOPc and MnTPrOPhOPc and 45% for H2TPrOPhOPc after 600°C, clearly confirming the stability of the metal complexes. The MTPrOPhOPcs (where M = H2, Co and Mn) complexes exhibited excellent nonlinear optical properties with strong reverse saturable absorption (RSA), especially when 560 nm excitation laser was used. Their nonlinear optical properties followed this trend: H2TPrOPhOPc > CoTPrOPhOPc > MnTPrOPhOPc. According to the trend observed, the H2TPrOPhOPc was an excellent nonlinear optical limiter when compared to the CoTPrOPhOPc and MnTPrOPhOPc. All the investigated complexes exhibited optical limiting properties comparable to the phthalocyanine complexes reported in the literature. The MTPrOPhOPc complexes were further studied for their electrocatalytic and electroanalytical properties towards the detection of hydrogen peroxide. For the electrocatalytic studies, the synthesized complexes were immobilized onto gold electrode surfaces pre-functionalized with phenylazide (Au-PAz) monolayer. Copper (I) catalyzed alkynyl-azide cycloaddition reaction was used to covalently immobilize the MTPrOPhOPcs onto the gold electrode surfaces to form Au-PAz-MTPrOPhOPc. The MTPrOPhOPcs modified gold surfaces (Au-PAz-MTPrOPhOPc) exhibited good reproducibility and stability in various electrolyte conditions. Electrochemical and surface characterisation of the functionalised gold electrode surfaces confirmed the presence of the MTPrOPhOPcs and their electroanalysis was excellent towards electrocatalytic reduction of H2O2, with the limit of detection (LoD) and limit of quantification (LoQ) in the ^M range. The electrocatalytic reduction peaks for H2O2 were observed at -0.37 V for Au-PAz-MnTPrOPhOPc and -0.31 V for Au-PAz-CoTPrOPhOPc when Ag|AgCl pseudo-reference electrode was used. Electrochemical and surface characterisation of the functionalised gold electrode surfaces confirmed the presence of the MTPrOPhOPcs and their electroanalysis was excellent towards electrocatalytic reduction of H2O2, with the limit of detection (LoD) and limit of quantification (LoQ) in the ^M range. The electrocatalytic reduction peaks for H2O2 were observed at -0.37 V for Au-PAz-MnTPrOPhOPc and -0.31 V for Au-PAz-CoTPrOPhOPc when Ag|AgCl pseudo-reference electrode was used. The Au-PAz-MnTPrOPhOPc and Au- PAz-CoTPrOPhOPc gold electrode surfaces showed good sensitivity and reproducibility towards the electrocatalytic reduction of hydrogen peroxide in pH 7.4 phosphate buffer solution. , Thesis (MSc) -- Faculty of Science, Chemistry, 2017

Hemi-synthetic reactions were carried out on two isolated compounds (5 and 6) to afford two new derivatives, that is, cordicerol A (14) and cordicerol B (15), respectively. The chemical structures of all the compounds were established based on analysis and interpretation of spectroscopic data such as electron ionization mass spectrometry (EI–MS), high resolution electrospray ionization mass spectrometry (HR-ESI–MS), fast atom bombardment mass spectrometry (FAB–MS), one dimension and two dimension nuclear magnetic resonance (1D and 2D-NMR) spectral data as well as X-ray crystallography (XRC). Lupeol ester derivatives [Lup-20(29)-en-3-triacontanoate (3)], monoglycerol derivatives [1-(26-hydroxyhexacosanoyl)glycerol (4) and glyceryl-1 hexacosanoate (5)] were isolated for the first time from Cordia genus while sulfur allotrope [cyclooctasulfur (2)] was isolated for the first time from plant origin. Biological assays cordidepsine (1) exhibited significant anti-HIV integrase activity with IC50 = 4.65 μM; EtOAc extract of stem barks, EtOAc fraction of roots and leaves were not toxic against 3T3 cells.

Modelling African swine fever disease in domestic pigs with culling and removal of contaminated feeds

- Description: In this study, a modelling African Swine Fever disease in domestic pigs with culling of infected pigs and removal of contaminated Feeds is studied. The goal of the study is to examine if culling of infected pigs and removal of contaminated food or water on pig farms can reduce the ASF trans mission in the domestic pig population. The SEAI-B model without control strategies is studied and analysed. It is shown that the solution for the system of model equations is both bounded and positive. We have found the disease free equilibrium point, basic reproductive number (using next generation method), local stability of disease free equilibrium (using Routh-Hurwitz criterion), global stability of disease free equilibrium, endemic equilibrium point (mathematica), existence of endemic equilibrium point, local stability of endemic equilibrium point (row echelon expression with the help of Maple 2.8) and global stability of endemic equilibrium point (Lyapunov function). Furthermore, sensitivity analysis is performed on the key parameters driving ASF Virus and to de termine their relative importance and potential impact on the dynamics of ASF Virus. Numerical result shows that if we increase the contact rate between susceptible pigs and infected pigs will re sults in increase ASF disease in the population. The SEAI-B model is reformulated incorporating culling of infected pigs and removal of contaminated food or water and analysed. Firstly, we found the disease free equilibrium point, basic reproductive number (using next generation method), local stability of disease free equilibrium point, global stability of disease free equilibrium point, endemic equilibrium point and local stability of endemic equilibrium point. Numerical result shows that if we increase culling of infected pigs and removal of contaminated food or water results in increase of susceptible pigs. The dynamic behavior of the model appears to be dependent on the culling of diseased pigs and the removal of contaminated food or water, according to numerical simulation.
