- Description: The need for monitoring the biological impacts of instream sediments has long been recognised, yet robust and scientifically defensible tools for doing so are still in the early stages of development because of the difficulties experienced by researchers in characterising the complicated mechanisms of biological effect elicited by sediment particles. Biological monitoring is one such tool, and this paper reports on the initial stages of a study to determine the most applicable approach for measuring the effects of instream sediments on aquatic macroinvertebrates in the South African context. In this first instance, the suitability of the rapid macroinvertebrate biomonitoring tool (the South African Scoring System) was investigated by determining the extent of the correlation between concurrently measured SASS metrics and turbidity data collected for the South African River Health Programme. All three SASS metrics – SASS score, number of taxa (NOT), and average score per taxon (ASPT) – were found to be significantly negatively correlated with turbidity, although variation in the data was high. Turbidity was found to be the major driver of change in ASPT. In contrast, electrical conductivity was the major driver of SASS scores and NOT, with turbidity a close second. When combined, electrical conductivity and turbidity accounted for 80 percent (SASS score) and 75 percent (NOT) of the variation in the regression model. Consequently, SASS metrics are a crude, but reliable, indicator of the negative biological implications of excessive instream sedimentation as measured by turbidity. A number of other potential biomonitoring approaches for detecting the impacts of fine sediment exposure are identified for further investigation: spatial analyses of macroinvertebrate assemblages; and the use of structural and functional metrics.

In contrast, the biological and ecological impacts of direct LAS input to the aquatic environment, as a consequence of near-stream laundry washing, are yet to be fully realised. Consequently, this thesis posed two research questions: 1) 'What are the LAS concentrations in a small rural South African river'? and 2) 'Is the in-stream biological community negatively affected at these concentrations?' The chosen study area, the community of Balfour in the Eastern Cape Province, is like many rural areas of South Africa where inadequate provision of piped water to homesteads necessitates laundry washing alongside the nearby Balfour River. The first research question was addressed in two ways: by predicting LAS concentrations in Balfour River water by assessing detergent consumption and laundry washing behaviour of residents living alongside the river; and measuring actual in- stream LAS concentrations on different days of the week and during different seasons. Results indicated that LAS concentrations were highly variable temporally and spatially. High peak concentrations of LAS occurred infrequently and were limited to the immediate vicinity of near-stream laundry washing activity with the highest measured concentration being 342 μg.L ⁻¹ and the average 21 μg.L ⁻¹ over the sampling period. The second research question was addressed by integrating the chemical evidence, determined from the first research question, with the biological evidence of stress responses measured in macroinvertebrates collected downstream of near-stream laundry washing activity on the Balfour River. Predicted and measured LAS exposure concentrations from the Balfour River were compared to a water quality guideline for LAS (304 μg.L ⁻¹), specifically derived in this thesis. Biological stress responses were measured at different levels of organisation: two sub-cellular responses (lipid peroxidation and cholinesterase activity); three measures of macroinvertebrate tolerance to water quality impairment; five measures of community composition; three measures of community richness; and a surrogate measure of ecosystem function (functional feeding groups). Weight-of-evidence methodology was utilised to assess, integrate and interpret the chemical and biological evidence, and at its conclusion, determined no effect on the in-stream biological community of the Balfour River downstream of laundry washing activity.

Different forms of nutrients and energy move across the conceptual boundaries of habitats via organisms’ activities or physical processes such as wind or water currents, and these transfers can represent important food subsidies. Such cross-partition ecolog-ical subsidies can augment the nutritional condition, biomass and biodiversity of communities, particularly where local production (or autochthony) alone may be inadequate to support local food webs. Furthermore, organic subsidies can influ-ence population dynamics, community interactions and ecosystem processes, and can represent dominant flux inputs in ecosystem budgets. Our intention was to explore organic nutrient fluxes in relation to a primarily lotic (i.e. flowing) aquatic sys-tem at the scale of a hydrological catchment.

Water quality guidelines are an important tool in the management of these water resources, aiming to adequately balance protec-tion of aquatic ecological systems with sustainable human use needs. Jooste and Rossouw (2002) proposed guidelines or boundary values for inorganic salts to be included in the ecological Reserve. These boundary values for inorganic salts were derived as follows, acute lethality data (LC50s) from the ECOTOX database main-tained by the USEPA were projected to 336 h and the 5th percentile determined as a lethality benchmark, analogous to the Fair/Poor boundary. Similarly, the 5th percen-tile of available sublethal data was determined as the sublethality benchmark and analogous with the Natural/Good boundary value. The Good/Fair boundary was the mean value between Natural/Good and Fair/Poor values. It has been suggested however, that these guidelines might not be entirely appropriate as they were de-rived without including tolerances of South African biota. Furthermore, the accuracy for some salt boundary values have been questioned (Scherman, 2009; Scherman, 2010).

Sediment samples were also collected and analysed. In addition, a household survey was conducted to ascertain the amount of detergent used and the distribution of washing practices. The results of the survey suggested that the use of riverside locations for laundry activities was seasonal. Most washing tended to be done at home during the winter with riverside sites used more frequently during the summer months. The monitoring data showed that LAS concentrations in water were very variable. They were occasionally high in the immediate vicinity of the laundry site (up to 342 µg L− 1) but were generally very low (< 11 µg L− 1) at downstream monitoring stations, suggesting that LAS was rapidly dissipated by a combination of degradation, hydrodynamic dispersion and dilution. Concentrations in the immediate vicinity of the washing site were lower than expected on the basis of the household survey because most waste water was disposed of on the river bank rather than directly in the river. No ecological effects are expected from LAS emissions at this site.

- Description: There is a growing international trend towards the protection of freshwater resources from pollution by imposing instream guidelines and specified waste-discharge conditions. Current methods for devising freshwater quality guidelines are based on species sensitivity distributions (SSDs) that are used to identify pollutant concentrations, ensuring the protection of a modelled percentage of species (95% protection is a common goal). SSDs are derived from the toxicity test results of as many taxa as possible for each polluting substance. Waste-discharge licences can be for single substances, specified in terms of chemical concentrations, and derived in conjunction with instream guidelines; or for complex mixtures, specified in terms of toxic units. In both cases toxicity test results are the core data used. The emphasis on SSDs calls into question the species constituting the test populations. It is likely that SSDs based in part on the responses of local organisms will achieve superior site-specific ecological protection. Until the early 1990s, there were very few data on the tolerances of South African freshwater organisms. In the intervening decade, the Unilever Centre for Environmental Water Quality at Rhodes University has developed a toxicity database that, to date, records the responses of 21 South African freshwater taxa to 26 single-substance pollutants or mixtures. This is the most comprehensive database of South African toxicity responses available and has been used in the drawing up of methods and guidelines to protect water resources. This paper aims to make these data available and to describe applications of the data using selected case studies.

- Description: There is a growing international trend towards the protection of freshwater resources from pollution by imposing instream guidelines and specified waste-discharge conditions. Current methods for devising freshwater quality guidelines are based on species sensitivity distributions (SSDs) that are used to identify pollutant concentrations, ensuring the protection of a modelled percentage of species (95% protection is a common goal). SSDs are derived from the toxicity test results of as many taxa as possible for each polluting substance. Waste-discharge licences can be for single substances, specified in terms of chemical concentrations, and derived in conjunction with instream guidelines; or for complex mixtures, specified in terms of toxic units. In both cases toxicity test results are the core data used. The emphasis on SSDs calls into question the species constituting the test populations. It is likely that SSDs based in part on the responses of local organisms will achieve superior site-specific ecological protection. Until the early 1990s, there were very few data on the tolerances of South African freshwater organisms. The effect of diet and age-at-weaning on growth and survival of clownfish Amphiprion percula (Pisces : Pomacentridae)

- Description: The aim of this study was to improve the rearing of common clownfish Amphiprion percula, by weaning juveniles from a live feed to a formulated feed as early as possible, while still maintaining good growth and survival. The growth response of A. percula to a formulated dry feed was initially investigated. There was no difference in growth rate of juveniles fed a formulated feed, and the formulated feed supplemented with either Artemia or a Donax serra / Penaeus indicus combination. Amphiprion percula readily consumed the formulated feed, and the fishmeal/casein combination appeared an acceptable protein source. As the amount of protein included in a diet can have a profound effect on growth, the optimal dietary protein level for juvenile A. percula was investigated by feeding semipurified diets containing graded levels of protein, ranging from 40-65%. There was no difference in the growth rates of juveniles fed the various diets, however all diets promoted good growth with an average weight gain of 419%, and thus for the purposes of this study the diet formulation was deemed adequate. The histological study of the digestive system of larval A. percula revealed that the alimentary canal was advanced at hatching and that larvae start exogenous feeding immediately. Three days after hatch (DAH) the yolk sac is completely absorbed. In the hind-gut epithelium of 5-day-old larvae small supranuclear inclusion vacuoles appear, suggesting pinocytotic digestion, and by 7 DAH gastric glands are established in the epithelium of the stomach. Nine DAH supranuclear inclusion vacuoles appear in the epithelium of the mid-gut, indicating extracellular digestion and absorption across the lumen. As pinocytotic digestion of protein is less efficient than extracellular digestion, especially in the case of formulated feeds, it was hypothesised that the digestive system of A. percula could only effectively digest formulated feeds 9 DAH onwards. The two weaning experiments designed to test this hypothesis revealed that A. percula was able to utilise the formulated feed, without reduction in survival, from 7 DAH onwards. However, in terms of growth, the optimal time to wean juveniles from the live feed to the formulated dry feed was between 15 to 20 DAH. As A. percula accept a formulated feed and can benefit nutritionally from it, the dependence of larvae and juveniles on live feed can be reduced. This study has shown that the rearing of A. percula can been simplified and improved by weaning from 7 DAH with no reduction in survival, and from 15 to 20 DAH with no reduction in growth.
