Molecular markers are useful tools to understand genetic contributions in hybrid populations, especially where morphological differentiation is difficult. Nymphaea mexicana Zuccarini (Nymphaeaceae) is an invasive alien plant in South Africa that is being targeted for biological control, but hybrids with intermediate morphological traits are also present at several sites. In this study, ISSR (inter simple sequence repeats) and ITS (internal transcribed spacer) markers were used to determine which Nymphaea species are likely to be putative parents of these hybrids, and morphological characters were also investigated to determine if genetic and morphological traits matched. Two major hybrid groups were identified, with one group clustering with Nymphaea odorata Aiton and the other clustering with Nymphaea alba L. A third, smaller group clustered with Nymphaea tetragona Georgi, whereas the remaining samples clustered with pure N. mexicana from the native range. Morphological features agreed with deductions drawn from molecular data. These results allow us to focus efforts to find compatible biological control agents and better understand the complicated genetic structure of N. mexicana and Nymphaea hybrids in South Africa.

The principle intention of the study is to illustrate and shed light upon human infant development and how the newborn becomes a fully functioning member within the family. The study provides a context in which to consider parent-infant interaction beginning in utero, expanding to the birthing process, and continuing through the infant's first year. It focuses on specific themes of parent-infant interaction. The following situations are explored: the role of the mother; the mother as a container; the infant's experience of containment; the internalisation of experience; the symbolic meaning of food; dealing with distress and the development of concrete communication; the growth of a sense of ego; and, the infant's internal world. The study concludes by addressing various implications for further psychotherapy and compares the therapist-client relationship to the mother-infant relationship. The research outlines one particular psychoanalytic theoretical orientation of mental and emotional development. It is a model derived predominantly from The Developmental School Theorists and Object Relations Theorists, namely, Bowlby, Klein, Mahler and Winnicott. This model looks at the infant's earliest relationships and the processes these set up within the infant's developing mind. Infant observation, asa research method proposed by Bick and Sidoli, links method and theory, and serves as the methodological approach utilised in the present study. The electrografted spots of diazonium were performed by positioning a Pt tip at a given distance above the gold substrate and the SECM was used in a three-electrode configuration (the Pt tip serving as the microanode) in acetonitrile containing 5 mM 4-azidobenzenediazonium and 0.1 M Bu4NBF4 during 10 ms. The dimensions of the derivatized areas of the substrates were finely tuned by using different experimental conditions (tip distance above the substrate, tip diameter, presence or absence of supporting electrolyte). The use of the azido-derivated diazonium molecule and these preliminary results open the gate to important applications and developments devoted to the local micro functionalization of electrodes by thin layers that allow the implementation of the emerging and attractive interfacial click reaction.

Electrochemistry was also used to determine the blocking characteristics of the MnPc self-assembled monolayers on gold, which proved to be highly dependent on the type of substituent. This study aims to extend our current understanding of the shared genetic influences between SZ and subcortical brain volumes using data from the latest genome-wide association studies for the respective phenotypes (GWAS) and novel statistical approaches. Additionally, we will explore whether the association between schizophrenia and abnormal regional brain volumes is causal in nature. Methods: Summary statistics were obtained from the largest Psychiatric Genomic Consortium (PGC)-SZ GWAS (Ncase = 69,369, Ncontrol = 236,642) and the CHARGEENIGMA-UKBB GWAS of volumetric measures for eight subcortical brain regions (the nucleus accumbens, amygdala, brainstem, caudate nucleus, hippocampus, globus pallidus, putamen, and thalamus), and total intracranial volume (N = 30,983 - 40,380). Single nucleotide polymorphism (SNP) effect concordance analysis (SECA) was used to assess pleiotropy and concordance. Genetic correlation was assessed using linkage disequilibrium score regression (LDSR) and the pleiotropy informed conditional FDR approach was applied to identify SNPs associated with SZ conditional on their association with subcortical brain volumes. Mendelian randomization (MR) was used to test for causal association between SZ and each brain region. Results: There was evidence of global pleiotropy between SZ, and all examined subcortical brain regions. Inverse concordance between the genetic determinants of SZ and volumes of the nucleus accumbens, amygdala, brainstem, hippocampus, and thalamus was observed. Increased statistical power to detect SZ risk loci was shown when conditioning on subcortical brain volumes. There was no significant evidence for a causal effect of any of the examined brain regions on schizophrenia risk. Discussion: These data confirm the shared genetic basis of SZ and specific intracranial and subcortical brain volumes and provide evidence for negative concordance between SZ and volumes of the nucleus accumbens, amygdala, brainstem, hippocampus, and thalamus. Leveraging the genetic overlap between SZ and subcortical brain volumes has the potential to provide novel insights into the biological basis of the disorder.

Discussion: These data confirm the shared genetic basis of SZ and specific intracranial and subcortical brain volumes and provide evidence for negative concordance between SZ and volumes of the nucleus accumbens, amygdala, brainstem, hippocampus, and thalamus. Leveraging the genetic overlap between SZ and subcortical brain volumes has the potential to provide novel insights into the biological basis of the disorder.

For N. mexicana, hybrids exist in the wild and horticultural trade, but identification is difficult, so understanding the genetic structure of populations is required to ensure that potential agents are collected off plants similar to invasive populations in South Africa. ISSR (inter-simple sequence repeats) analysis was used to determine whether invasive N. mexicana populations from South Africa were genetically similar to native range populations from USA or whether they were hybrids. Results from these analyses were matched with the morphotypes of each population based on petal colour, shape, and size. The genotypes suggested by the ISSR analyses corroborated the presence of both hybrid and pure forms of N. mexicana in South Africa. Populations of N. mexicana in the invaded range that are genetically similar to native range populations are more likely to be suitable for biological control, while other populations are likely to be hybrids formed by crossing of parents from the native range or within the horticultural trade, which may present difficulties for management using biocontrol.

There is a need for tools that can screen for and improve potential research participants’ understanding of the research study at the time of recruitment. Limited empirical research has been conducted in low and middle income countries, evaluating informed consent processes in genomics research. We sought to investigate the quality of informed consent obtained in a South African psychiatric genomics study. A Xhosa language version of the University of California, San Diego Brief Assessment of Capacity to Consent Questionnaire (UBACC) was used to screen for capacity to consent and improve understanding through iterative learning in a sample of 528 Xhosa people with schizophrenia and 528 controls. We address two questions: firstly, whether research participants’ understanding of the research study improved through iterative learning; and secondly, what were predictors for better understanding of the research study at the initial screening? During screening 290 (55%) cases and 172 (33%) controls scored below the 14.5 cut-off for acceptable understanding of the research study elements, however after iterative learning only 38 (7%) cases and 13 (2.5%) controls continued to score below this cut-off. Significant variables associated with increased understanding of the consent included the psychiatric nurse recruiter conducting the consent screening, higher participant level of education, and being a control. The UBACC proved an effective tool to improve understanding of research study elements during consent, for both cases and controls. The tool holds utility for complex studies such as those involving genomics, where iterative learning can be used to make significant improvements in understanding of research study elements. The UBACC may be particularly important in groups with severe mental illness and lower education levels. Study recruiters play a significant role in managing the quality of the informed consent process.

The goals were to describe a year of activities, within the TALON environmental club at Hudson Park High School in East London, South Africa and to identify how pedagogical, contextual and relational factors influenced action competence processes amongst club members. I explored how action competence theory and praxis have helped in other school education settings with the aim of establishing better environmental practices. The IVAC model of pedagogy (Jensen & Schnack, 1997) was used as a framework to operationalise action competence processes within the TALON Club. This entailed carefully examining investigative, visioning, action and change processes unfolding through the four themes of the club: plants, animal welfare, waste issues and our community outreach programme at Bongulethu High School. Using questionnaires, interviews, observation and a journal as the main methods of data collection I was able to make five analytical statements that related to the influence of pedagogy, the influence of context and the influence of relational dynamics within the TALON Club. The pedagogical processes were affected by the TALON members wanting to avoid any activities that were ‘too much like school’. However this served to compromise the action and visioning phases within the club, resulting in superficial and poorly-informed action-taking. The influence of context served to both enable and constrain action competence processes. The club and school structures therefore shaped the form that fledgling actions took as the TALON members participated in the meetings and outings and fulfilling the traditional expectations of the school community. Relational dynamics and a desire for fun took precedence over the action competence processes. Although the Club members were motivated by a sense of moral responsibility they were unable to articulate this concept into the club activities. In addition there were the power gradients between the educators and members to consider as determinants of the extent of participatory, democratic interactions. This complex interplay of factors influenced the action competence processes in the club. Recommendations were made on how best to work with the concepts of participation and democracy towards improving the reflexivity and knowledge base of both the educators and learners. In this way action competence processes will be better supported within the club in future.

An exploration of the other and the disruption of self in schizophrenia

He argues that development of ongoing dialogues from within and without the Self constitute the Self. These parts of Self that participate in this internal dialogue are referred to by Hermans (2002) as I-positions. Healthy, dynamic internal dialogue between I-positions is argued to contribute to maintaining a unified sense of Self. The Self is also argued to be socially constructed, in so far as Others outside the Self participate in these internal dialogues and are able to influence and occupy I-positions. Research has shown that people with a diagnosis of Schizophrenia experience a unique form of Self disruption. It has been postulated that this disruption is due to disruptions in this internal dialogue. To date, seminal research has primarily focussed on the internal mechanisms and phenomenological accounts of these disruptions. However, little research has focused on the role that Others outside the Self play in these disruptions. Since the Self is also constructed as a social entity, the aim of this research is to explore in what ways the Other contributes and/or minimises this disruption in people with a diagnosis of Schizophrenia. In order to achieve this, I made use of archival data which was made up of the transcripts from semi-structured interviews previously conducted with people with a diagnosis of Schizophrenia as part of a Self and Schizophrenia study. The interviews were analysed using deductive thematic analysis, and utilised Herman’s theory of the Dialogical Self as the theoretical lens for this study. The emergent themes were organised according to those that were present before a diagnosis of Schizophrenia was given and those themes that were present post-diagnosis. The themes reflected that the Other plays a significant role in the both the maintenance of healthy dialogue post diagnosis and well as in the disruption of dialogue. This disruption was primarily observed throughthecompromising of previous I-positions and in the development of new performative and deficit I-positions that contributed to the disruption in internal dialogue.

- Description: South Africa’s history, pre- and post-democracy, has been designed by different laws which moulded the socio-economic conditions of rural communities in numerous ways. The introduction of democracy in the country created a platform for understanding different rights for many citizens; amongst those rights a right to shelter, proper sanitation, and clean water. The provision of housing by the three spheres of government, local, provincial, and national aimed at addressing these rights for all citizens of the country, which is a privilege for both urban and rural communities. Even though provision of houses for the rural poor was not the priority in the first administration of the democratic government, as time progressed, discussions and implementation of the policy on housing provision for rural communities became a strong political debate. The intensity on housing provision for rural houses as from 2010 became more resilient. The objectives of this study firstly viewed the botches of the Department of Human Settlement, Water and Sanitation and the Municipality to monitor and evaluate infrastructural challenges faced by rural communities who became beneficiaries of houses under Rural Development Programme (RDP). The issue of poor sanitation and access to clean water by rural poor even in the fourth and fifth administration was also a concern of the study. The socio-economic factors on the provision of houses for rural housing beneficiaries was also a major apprehension, and support services provided by the relevant stakeholders to rural housing beneficiaries was also a point for discussion. It is therefore imperative that the study evaluate and recommend strategies that can be put in place for improving housing provision for the rural poor and create sustainable human settlements. The Eastern Cape Province is one of the provinces that are battling with backlogs related to housing provision, high unemployment rates, low rural household incomes, and low levels of education more especially to older citizens, which can create a higher risk of crime more especially by the youth. This study, therefore, attempted to highlight cases of poor administration, since it its findings the issue of wasteful expenditure of people’s taxes when houses constructed for community beneficiaries are not occupied. In such cases, poverty becomes rife, and health-related issues tend to expand. Similarly, economic growth and social development becomes limited. This treatise was concerned with the strategies used in the provision of rural housing in South Africa, the Eastern Cape Province. The findings further identify that the legislation that was supposed to be employed by the Department of Human Settlement in the provision of rural housing was not considered as a priority, hence there were many flaws identified. The policies applied in other countries, internationally, were suggested to be used as a benchmark for improving housing provision strategies in rural Eastern Cape municipalities. Since the study had adopted a descriptive approach, meaning the use of secondary data, the issue of public participation in any designed development project in rural communities should be as an important aspect of rural development, The Batho Pele Principles as key instrumental legislation in the government should be taken into serious consideration always more especially the principle of consultation. Rigorous debates are also required to produce policies that are informed by evidence in order to build and produce a better future.

- Description: South Africa’s history, pre- and post-democracy, has been designed by different laws which moulded the socio-economic conditions of rural communities in numerous ways. The introduction of democracy in the country created a platform for understanding different rights for many citizens; amongst those rights a right to shelter, proper sanitation, and clean water. The provision of housing by the three spheres of government, local, provincial, and national aimed at addressing these rights for all citizens of the country, which is a privilege for both urban and rural communities. Even though provision of houses for the rural poor was not the priority in the first administration of the democratic government, as time progressed, discussions and implementation of the policy on housing provision for rural communities became a strong political debate. The intensity on housing provision for rural houses as from 2010 became more resilient. The objectives of this study firstly viewed the botches of the Department of Human Settlement, Water and Sanitation and the Municipality to monitor and evaluate infrastructural challenges faced by rural communities who became beneficiaries of houses under Rural Development Programme (RDP). The issue of poor sanitation and access to clean water by rural poor even in the fourth and fifth administration was also a concern of the study. The socio-economic factors on the provision of houses for rural housing beneficiaries was also a major apprehension, and support services provided by the relevant stakeholders to rural housing beneficiaries was also a point for discussion. It is therefore imperative that the study evaluate and recommend strategies that can be put in place for improving housing provision for the rural poor and create sustainable human settlements. The Eastern Cape Province is one of the provinces that are battling with backlogs related to housing provision, high unemployment rates, low rural household incomes, and low levels of education more especially to older citizens, which can create a higher risk of crime more especially by the youth. This study, therefore, attempted to highlight cases of poor administration, since it its findings the issue of wasteful expenditure of people’s taxes when houses constructed for community beneficiaries are not occupied.
