- Description: Land use and land transformation are major threats to biodiversity. Only a small percentage of land and thus biodiversity is protected within reserves. The majority of biodiversity lies in the hands of private and communal farmers and in order to protect biodiversity they must perceive it as having some value and have the means and incentive to conserve it. This study examined two things: (i) the relationship between biodiversity and measures of ecosystem health, range condition, primary production and presence of useful plants that would be expected to be of relevant use to land users, (ii) the perceptions of farmers of vegetation states that differ in the abovementioned attributes. Within the Eastern Province Thornveld of the Smaldeel area, four different vegetation states were selected for the study, namely park-like grassland with scattered Acacia karroo (“savanna”), heavily infested Acacia karroo grassland (“acacia”), thicket-grassland mosaic (“thicket”) and heavily utilised thicketgrassland in communal lands (“communal”). The four states are a consequence of different patterns of browsing and fire, in both pre-colonial and recent times. Different ecological attributes were assessed and compared for each state and the relationships between the different attributes determined. Ecosystem health, in terms of stability or resistance to erosion, infiltration/water-holding capacity, and nutrient cycling were compared using Landscape Function Analysis. The agricultural value of the different landscapes was measured using range condition assessment techniques. Plant species richness and other measures of diversity, along with their conservation and usefulness values, were compared between states. Plant productivity and biomass were compared using satellite data. The thicket state was found to be the most functional due to the added habitat complexity provided by the vegetation. It was the most biodiverse, the most useful and contained many, but not all of the important conservation species. The communal state had high biodiversity and was be fairly resilient to heavy usage, not showing the expected signs of land degradation. The savanna state, although thought of as the optimum state for cattle production, was not significantly different from the other states in terms of agricultural potential, but had the lowest values for plant diversity, browse potential, abundance of useful plant species and biomass. The acacia state had the highest and least stable values in terms of biomass production, however it was found to contain species of conservation importance. The preference that four groups of land users, namely men and women from commercial and communal farming areas, expressed for the four vegetation states was assessed using semi-structured interviews in conjunction with A3 colour photographs. The men and women from the communal areas and the men from the commercial areas valued the thicket state highly for its farming potential. The male commercial farmers also valued the savanna state highly as they perceived it as being most productive for cattle farming. Both the thicket state and the savanna state were found aesthetically pleasing by all the user groups, in line with theories of preference for modified savannas and for familiar environments. There appears to be potential for preserving biodiversity on farmland. The farmers in this study, with their very utilitarian perspective, were found to intuitively gauge the health of the landscape and recognise biodiversity as indicating good farming land. The commercial farmers were strongly influenced by economic motives and thus attracted to the savanna state, but also recognised the opportunity for a wider variety of farming activities in the thicket state. The communal farmers have a high dependence on the land for their livelihoods and preferred the thicket state for its overall usefulness. The views of the farmers indicate that the opportunity for conservation is good, however much of the thicket state lies in the communal land, where the openaccess land use system makes managing for biodiversity difficult.

Cost-effective, post-mining environmental restoration of an open-cast phosphate mine at Langebaanweg, South Africa

- Description: Approaching the ecological rehabilitation of an open-cast phosphate mine in the West Coast of South Africa during the post-operational phase presented some challenges. The area was extensively modified during the mining operations. Soils from different layers were mixed with topsoil being covered by subsoil, overburden dumps and tailing dams being constructed resulting in extensive cross-zoned soils. Large areas of subsoil areas were exposed on the mine floor with localized and small scale salinity being evident. The modified topography as well as the complex new surface material posed a challenge in terms of identifying suitable local species that could be used to rehabilitate the post-mining environment. The mine area was heavily infested with woody alien invasive plants, such as Acacia cyclops, established in an attempt to reduce the dust and little natural vegetation cover was present. In the arid west coast environment, the four-month-long winter growing season is followed by hot and windy dry summers (Chapter 2) presenting a challenge reestablishing local vegetation in modified soils. Moreover, little was known about the local vegetation in terms of their propagation and use in stabilization techniques as an alternative to exotic vegetation such as the A. cyclops, which had been more often used in revegetation projects. A study was conducted to determine the most efficient and cost-effective methods of vegetative rehabilitation of the Chemfos site (Chapter 1). A review of the literature available at the time as well as approaches that were successfully implemented in other Western Cape rehabilitation projects such as the Du Toitskloof Pass and the Sishen–Saldanha railway line (Chapter 3), were considered. Previous studies on the west coast dunes at Blaauwberg had shown using Marram grass as a dune stabilizer was most successful. Marram grass was tested in trials alongside local grasses, such as Chaetobromus dregeanus and Ehrharta villosa, that showed potential but had not been formally evaluated. The environmental context of Chemfos (Chapter 2) as well as the Conceptual Rehabilitation Plan that considered the modified environment, soils and closure objectives of the mine, were reviewed to determine the most pressing rehabilitationrelated questions that required answers. This led to the final experimental design that was implemented mid-winter in 1996 (Chapter 5). The trials were implemented in the areas perceived to be the most difficult to rehabilitate, namely the mobile sands in the tailings dam and the exposed subsoil or mine floor areas. The use of brushwood together with specific plants and seeds appropriate for the use in either sandy soil or subsoil were evaluated in a variety of combinations and application densities to determine the most effective treatment combination at the minimum effective density. The best initial cover of the tailings dam were recorded in the Ammophila arenaria trials but the use of the local grass species Ehrharta villosa, performed better from year two onwards and was much cheaper to establish. On the subsoil, the trials where a cover of topsoil was used performed better than the combination trials. This indicated that topsoil placement on post-operational phase shaped subsoils during the mining operation to be the most desirable treatment. Initial results of the trials were used as a basis for developing the rehabilitation techniques that were rolled out across the Chemfos landscape. The approaches were refined as indications of trial responses became evident. Lessons learned were incorporated in the adaptive management approach that was followed and the rehabilitation techniques (Chapter 6) were continually re-evaluated and adjusted. This resulted in a significant step towards achieving the overall research objective of finding cost-effective approaches to rehabilitation. Components such as refinement of the seed collection and processing techniques (Chapter 7) where the post-harvest processing cost was significantly reduced by introducing specially designed drying racks. Processing techniques were adjusted to suit the different species, and a variety of mechanical processing options were explored. The scale of the Chemfos project led to the development of new techniques of manufacturing a smoke-derived germination stimulant (Chapter 8) since commercial availability of these products was very limited. The development of FireGrow assisted in the overall aim of reducing cost by increasing germination of seed in the rehabilitation sites using a very cost-effective smoke concentrate. Socio-economical aspects were considered during the implementation as well as the post-closure phases of the rehabilitation and BHPBilliton invested through the agency of the SAMANCOR Trust. This led to the development of livelihoods of the staff that remained in the area and that lived in the mine village. The demography of the population has changed over time in the Green Village as well as the skills that the inhabitants have developed. Thus, the new economic opportunities that were pursued have brought a new lease on life beyond the lifespan of the mine (Chapter 9) once the closure certificate had been issued.

- Description: Despite the many changes that have occurred in South Africa since the end of apartheid, there are still residual effects of it, as is evidenced in the disparity of living conditions between different racial groups. It is also evident that there are differences in the work tasks and working conditions of nurses working in different work environments. This project looks at how living conditions as well as working conditions interactively affect the health, subjective well-being, and work ability of nurses. Questionnaires were completed by, and interviews were conducted with nurses from Settlers Hospital and seven municipal clinics within Grahamstown (n=152). The participation rate was approximately 71%. The questionnaires included self-report, forced-choice questions regarding basic demographics of the nurses, work conditions, living conditions, subjective satisfaction levels, as well as a simplified version of the Nordic Questionnaire of Musculoskeletal Strain (Kuorinka et al., 1987), and the Work Ability Index (WAI) (Tuomi et al., 2006). The questionnaires were translated into Afrikaans and IsiXhosa. One-on-one interviews were conducted with the participants, in order to obtain a 24-hour dietary recall, an indication of physical activity levels, as well as measurements of stature, mass, waist girth and hip girth. Factor analysis was performed to identify common variance from amongst the variables, while canonical correlations examined the interaction between the sets of factors. It was found that variables relating to demographic factors, living conditions, and working conditions were closely linked to each other. Factors from each of these groups were associated with life, health, and job satisfaction, anthropometric measures, musculoskeletal strain, and WAI scores. Satisfaction levels appeared to be largely determined by socioeconomic status, while anthropometrics, WAI scores, and levels of musculoskeletal strain were associated with levels of smoking and drinking, race, age, stature, position and tenure.

Customer profiling using a service-orientated architecture

- Description: Customer profiling has recently gained much recognition in the e-commerce domain because of the benefits it is capable of bringing to online business. Customer profiling has been implemented in various systems development approaches such as in a client-server environment. Recently there has been an increase in the number of organisations adopting and implementing e-commerce systems using service-oriented architecture (SOA) principles. This research set out to determine how a customer profile can be implemented using open source SOA implementation tools, and how SOA-based customer profiles can be utilised to provide appropriate personalisation in an SOA environment. The research further endeavoured to complete a comparative study on customer profile implementation in two different architectures, namely SOA and client-server. An extensive literature review was conducted on SOA, customer profiling and e-commerce systems development. SOA enabling technologies, such as, web services, enterprise service bus (ESB) and open source Sun Java SOA implementation tools, for example, Open ESB, GlassFish application server and Netbeans IDE were analysed. A Java web services-based customer profiling system was prototyped following SOA design principles. An end-user evaluation survey was conducted using eye tracking with a sample of 30 participants. The evaluation was done on two e-commerce systems with the same interface but running on two different customer profile back-ends, SOA and client-server. The results show that participants did not experience significant difference between the two systems, however, eye tracking results showed a significant difference between the two systems. The research concluded that customer profiling using SOA offers more benefits than implementations using other architectures such as client-server. SOA component-based development proved to be easier to manage, develop, integrate and improves interoperability between different technologies. The research brought together necessary techniques and technologies that organisations can use to implement SOA. Using SOA, organisations can integrate and utilise different technologies seamlessly to achieve business goals.

Customer profiling has been implemented in various systems development approaches such as in a client-server environment. Recently there has been an increase in the number of organisations adopting and implementing e-commerce systems using service-oriented architecture (SOA) principles. This research set out to determine how a customer profile can be implemented using open source SOA implementation tools, and how SOA-based customer profiles can be utilised to provide appropriate personalisation in an SOA environment. The research further endeavoured to complete a comparative study on customer profile implementation in two different architectures, namely SOA and client-server. An extensive literature review was conducted on SOA, customer profiling and e-commerce systems development. SOA enabling technologies, such as, web services, enterprise service bus (ESB) and open source Sun Java SOA implementation tools, for example, Open ESB, GlassFish application server and Netbeans IDE were analysed. A Java web services-based customer profiling system was prototyped following SOA design principles. An end-user evaluation survey was conducted using eye tracking with a sample of 30 participants. The evaluation was done on two e-commerce systems with the same interface but running on two different customer profile back-ends, SOA and client-server. The results show that participants did not experience significant difference between the two systems, however, eye tracking results showed a significant difference between the two systems. The research concluded that customer profiling using SOA offers more benefits than implementations using other architectures such as client-server. SOA component-based development proved to be easier to manage, develop, integrate and improves interoperability between different technologies. The research brought together necessary techniques and technologies that organisations can use to implement SOA. Using SOA, organisations can integrate and utilise different technologies seamlessly to achieve business goals.

Therefore, sustained release capsule dosage forms containing 100 mg minocycline (base) were manufactured and assessed for use as sustained release oral dosage forms of minocycline. Minocycline sustained release capsules were manufactured based on matrix technologies using hydroxypropylmethyl cellulose (HPMC) and Compritol® as release retarding polymers. The rate and extent of minocycline release from the capsules was evaluated using USP Apparatus 1 and samples were analysed using a validated High Performance Liquid Chromatographic (HPLC) method with ultraviolet (UV) detection. Differences in the rate and extent of minocycline release from formulations manufactured using HPMC or Compritol® were influenced by the concentration of polymer used in the formulations. The rate and extent of minocycline release was faster and greater when low concentrations of polymer were used in formulations. The effect of different excipients on the release pattern(s) of minocycline and particularly their potential to optimise minocycline release from experimental formulations was investigated. The use of diluents such as lactose and microcrystalline cellulose (MCC) revealed that lactose facilitated minocycline release when HPMC was used as the polymer matrix. In contrast, the use of lactose as diluent resulted in slower release of minocycline from Compritol® based formulations. The addition of sodium starch glycolate to HPMC based formulations resulted in slower release of minocycline than when no sodium starch glycolate was used. Compritol® based formulations were observed to release minocycline faster following addition of sodium starch glycolate and Poloxamer 188 to experimental formulations. In vitro dissolution profiles were compared to a target or reference profile using the difference and similarity factors, ƒ1 and ƒ2 , and a one way analysis of variance (ANOVA). In addition, the mechanism of minocycline release was elucidated following fitting of dissolution data to the Korsmeyer-Peppas, Higuchi and Zero order models. Minocycline release kinetics were best described by the Korsmeyer-Peppas model and the values of the release exponent, n (italics), revealed that drug release was a result of the combined effects of minocycline diffusion through matrices and erosion of the matrices. These in vitro dissolution profiles were better fit to the Higuchi model than to the Zero order model. Two formulations that displayed a fit to the Zero order model were identified for further studies as potential dosage forms for sustained release minocycline.

Diet and population trends of warthog in the Addo Elephant National Park

- Description: The common warthog (Phacochoerus africanus) has recently been recognised as an alien invasive species in the Eastern Cape and their population is increasing rapidly within the region. This then raises a concern as it is recorded that invasive species have negative impacts on both economic and ecological aspects of biodiversity in the receiving community. There are few studies that have documented the impacts of this species in the Eastern Cape. This study therefore seeks to determine the diet and population trends of this species in the Addo Elephant National Park (AENP) Main Camp and the results obtained can be correlated with previous studies and used to quantify the impacts of this species in the Eastern Cape. Warthogs are specialized grazers, it was therefore hypothesized that they will have potential impacts on grazing resources in the AENP Main Camp. The diet of common warthogs in the AENP Main Camp was determined through microhistological analysis of faecal material of samples collected seasonally. Population trends of common warthogs were determined in relation to the effects of predation on the population growth and population age structure of these animals within the AENP Main Camp. The results were compared with the findings for common warthog population growth and population age structure of Great Fish River Nature Reserve (GFRNR), a predator free population. It was hypothesized that the population growth rate of common warthogs in the AENP Main Camp before the introduction of lions and hyaenas in the AENP would not differ from the population growth rate of common warthogs in GFRNR. Secondly, the post-lion/hyaena common warthog population in the AENP Main Camp would differ from both the AENP Main Camp pre-lion/hyaena and GFRNR common warthog populations due to the presence of large predators in the AENP Main Camp. For population age structure it was hypothesized that the two populations, AENP Main Camp and GFRNR would differ because of predators increasing mortality. The diet of common warthogs was dominated by grass (87.4 percent), with Cynodon dactylon being the dominant grass species. Common warthogs in the AENP Main Camp should be properly managed because they have potential impact on grasses. The population growth of common warthogs in the AENP Main Camp showed no effect of predation, with population growth not differing from that of GFRNR. Predation had an effect on common warthog population age structure with AENP Main Camp and GFRNR populations differing, particularly in terms of adult structure. Possible intervention involves diverting the Mfolozi River to the St Lucia Lake system. However, due to high sediment loading, water from the Mfolozi river requires considerable filtration before a link can be established. A suggested option considered in this study is to restore the existing sugarcane farmlands on the Mfolozi Floodplain (~20 800 ha) to previous wetland conditions to reinstate a sediment removal function amongst other benefits. Proposed restoration will have a direct impact on the industries currently supported by the iSimangaliso Wetland Park and the Mfolozi Floodplain (tourism, sugar, conservation). to understand a measure of such impacts, ecosystem services for both Lake St Lucia and the Mfolozi Floodplain were analysed (flood alleviation, water provision, water purification, sediment regulation, tourism, fisheries, vegetation for harvest, existence, cultural and research). Annual economic values for each ecosystem service were determined by means of valuation methods that included benefit transfer and replacement cost. Results showed a current annual minimum value of the Mfolozi Floodplain and Lake St Lucia as greater than R21 million and R1.1 billion respectively. Partial restoration of the floodplain (~6 000 ha) is expected to increase the sum of all ecosystem services values by approximately 26% for the Mfolozi Floodplain and by 23% for the St Lucia System. Full restoration (~20 800 ha) increases the total ecosystem services value by 88% and 50% for the Mfolozi Floodplain and St Lucia System respectively. Results showed that economic values for existence, fisheries, tourism and water provision increase by the greatest percentage for the St Lucia System under both restoration scenarios. Partial and full restoration of the floodplain will result in the greatest increases in economic value for the services existence, tourism, fisheries and the harvesting of vegetation on the Mfolozi Floodplain.

- Description: Epidemiological studies consistently report that muscular strains are a primary injury type in rugby union with the majority of the strains occurring to the quadricep and hamstring musculature. Recently it has been suggested that poor eccentric muscular strength is a precursor to hamstring and quadriceps strains during intermittent sports that require rapid acceleration and deceleration. Despite the high incidence of these muscle injuries in Rugby Union there has been little research into the possible mechanisms involved. Thus, the purpose of this study was to measure the physiological and perceptual responses during a simulated Rugby Union laboratory protocol and further, to identify changes in muscle recruitment patterns and muscle strength over time by comparing this protocol to a continuous, constant load protocol covering the same distance. The experimental condition (EXP) required university level players to perform 80 minutes of simulated rugby union play in a laboratory setting (on a walkway of 22m) which was compared to that of a control condition (CON) which involved subjects covering the same distance, at a constant speed of 4.2km.h-1 on a treadmill. Physiological, biophysical and perceptual responses were measured pre-, at half-time and post-protocol. Heart rate was significantly (p<0.01) greater as a result of EXP in comparison to the CON. Electromyography (EMG) of the vastus medialis was significantly (p<0.01) greater during the CON protocol. The EXP condition elicited higher iEMG activity in the hamstring musculature at all time intervals. In addition the iEMG of the semitendinosus decreased significantly (p<0.01) as a result of the EXP protocol. Peak eccentric knee extensors (EXT) (-13.19%) and flexors (FLEX) (-12.81%) torque decreased significantly during the experimental protocol. After passive half-time (236.67 + 56.27Nm (EXT) and 173.89 + 33.3NM (FLEX)) and at the end of the protocol (220.39 + 55.16Nm and 162.89 + 30.66Nm) reduced relative to pre protocol (253.89 + 54.54Nm and 186.83 + 33.3Nm). Peak eccentric knee extensors did not change during the control protocol. „Central‟ and ‟Local” Rating of Perceived Exertion values were significantly (P<0.01) greater during the EXP protocol with an increased incidence of hamstring discomfort and perceived pain (5 out of 10). The EXP protocol resulted in significantly (p<0.01) increased incidence of delayed onset muscle soreness (DOMS). In conclusion, a stop-start laboratory protocol elicited increased heart rate, negatively impacted on muscle activity of the hamstrings, decreased eccentric strength in the lower limb musculature, resulted in increased ratings of „Central‟ and „Local‟ exertion and increased pain perception and increased incidence of DOMS. Thus, a stop-start rugby specific laboratory protocol has a negative impact on performance. Due to the specificity of the protocol being designed to match the demands of competitive match play it is expected that these changes in heart rate, muscle activity and strength, particularly eccentric strength, will impact negativity on performance during rugby match play and increase the likelihood of injury

- Description: The methodology and the processes that are followed in the maintenance of government-operated district hospitals in the Eastern Cape Province of South Africa are outlined while the strategies that are employed to roll-out the maintenance plan have been investigated. The challenges in managing hospital maintenance are identified so that it can be improved and advice be given on the strategies currently being used. The objectives of the research were to determine the effectiveness of machinery maintenance in government-operated district hospitals. The study has been carried out by investigating 50 district hospitals within the Eastern Cape Province, with the aim of obtaining knowledge of practices in relation to the strategic plans and best practices in the maintenance industry. The following factors identified by the maintenance staff were found to be most important in maintenance management practices; realistic budgets, adherence to occupational health and safety regulations, engagement of qualified and skilled maintenance staff, good record management in relation to the equipment under maintenance, availability of tools and materials and keeping abreast with the latest technologies and trends of machinery advancement. The study also revealed that the majority of the hospitals use a combination of in-house and outsourced personnel during maintenance, the outsourced part being under contract from the National Department of Public Works Repair and Maintenance Plan (RAMP) projects. Service kits and recommended replacement parts for machinery are rarely stocked on the hospital premises while only minor spare parts that include replacement bulbs, electrical fittings, plumbing fittings and paint are readily available. In addition, the maintenance staff members have little or no input in maintenance budgeting, this aspect is controlled by hospital management. For maintenance to be effective, strategic planning that takes into account carefully thought-out maintenance management systems is the first step in the direction of setting out definite tangible objectives and goals. The real challenge lies in the implementation and sustainability of the maintenance management system and the monitoring thereof.

Crematogaster sp. 1 colonies were offered no choice between immature stages of the agents Hypena laceratalis Walker (Lepidoptera: Noctuidae), Falconia intermedia Distant (Hemiptera: Miridae) or Teleonemia scrupulosa Stål (Hemiptera: Tingidae) on lantana shoots. Density-dependent predation on F. intermedia and T. scrupulosa nymphs on lantana shoots was tested using Crematogaster sp. 2 colonies. In choice experiments Crematogaster sp. 2 colonies were offered F. intermedia or T. scrupulosa nymphs on potted lantana plants. Preliminary food trials confirmed that colonies foraged for protein, thereby validating results of no-choice experiments. Crematogaster sp.1 foragers removed 50% of F. intermedia nymphs, followed by 45% of H. laceratalis larvae and only 9% of T. scrupulosa nymphs. Foragers recruited most actively to H. laceratalis larvae and significantly more H. laceratalis biomass was removed than either F. intermedia or T. scrupulosa. A trade-off existed in prey size selection because larger larvae provided considerably more biomass but required forager cooperation and a longer time to subdue than did smaller prey. This increases both forager energy expense and mortality risk by other predators. This study showed that all Crematogaster sp. 1 colonies removed small (≤10mm) H. laceratalis larvae more frequently than larvae larger than 10mm. Thus, of these biological control agents, predators probably prefer small H. laceratalis larvae. Significantly more F. intermedia than T. scrupulosa nymphs were removed by Crematogaster sp. 1, while Crematogaster sp. 2 colonies removed comparable numbers of both agent species. Falconia intermedia nymphs' fast movement triggered a predatory response by these ant species. In contrast, the relatively immobile behaviour of T. scrupulosa nymphs was identified as a highly effective predator avoidance strategy. Since T. scrupulosa nymphs are unable to escape predators by moving, they appear to depend on the presence of alternative prey attracting predator attention. At high agent and/or forager density, T. scrupulosa nymphs attempted escape, but foragers identified them as prey once they moved and caught them. Predation on F. intermedia was also density dependent in that at high nymph and/or forager densities, escape routes were congested and nymphs were more easily caught. Survival of F. intermedia and T. scrupulosa nymphs in particular was low on ant-accessed shrubs in choice experiments and high on ant-excluded shrubs. It is likely that ants significantly depress F. intermedia populations in the field since besides predation, ant foragers probably interrupt F. intermedia feeding and ovipositioning. The combination of parasitism and predation on early instar larvae may explain why H. laceratalis occurs across lantana's range in South Africa but populations remain low. It is unlikely that T. scrupulosa nymphs are habitually preyed on by ant species unless they attract attention by being mobile. Although biological control of L. camara is influenced by climate and physiological defence mechanisms, this study has shown that predation by two ant species severely impacts leaf-feeding agents for L. camara. Thus, it is recommended that future selection of additional agents to control lantana should exclude leaf-feeding .

Effects of salinity on the growth and lipid production of ten species of microalgae from the Swartkops saltworks : a biodiesel perspective

- Description: Biodiesel from microalgae is a viable alternative for replacing the global demand for petro-diesel. High biomass and lipid production are key desirable characteristics needed in a species to be used for biodiesel production. It has been demonstrated in literature that the increase in salinity can increase the lipid content of microalgae, but lower the growth rate of a species. Therefore the effect that salinity has on the growth and lipid content of ten microalgal species, isolated from a warm temperate solar saltworks, was investigated. The microalgae were cultivated at a temperature of 22°C and at salinities ranging from 17 to 70 psu. It was found that growth and lipid production for all species were influenced to some degree by the salinity. Growth rates greater than 0.6 d-1 showed a decrease with higher salinity. Most (71 percent) of the growth rates that exceeded 0.6 per day were exhibited by cultures exposed to normal salinity (35 psu). This shift is a good indication that salinity inhibits/slows down growth and that the species in general prefer lower salinity conditions. Growth rates ranged from 0.17 ± 0.05 to 1.19 ± 0.17 d-1. Lipid content for the diatoms (2.78 ± 0.36 to 10.86 ± 4.59 percent DW) were lower than expected, whereas the lipid content for the green flagellates (3.10 ± 1.56 to 22.64 ± 1.19 percent DW) was on par with that reported in literature. To bring results into perspective a production model was developed to simulate a production scenario at the Swartkops Saltworks. Lipid and productivity results obtained in this study were used to estimate how much oil and biomass can be produced within the ponds of the Swartkops Saltworks. The model showed that although microalgae cultivation for biodiesel is technically feasible, at present it is not economically viable to do so.

- Description: The exotic plant Lantana camara L. has invaded the riparian areas of the Victoria Falls World Heritage Site in Livingstone, southern Zambia, threatening native plant communities which support populations of species of special concern. I trialled the mechanical control method of manual uprooting and 3 different herbicides applied through paint brushing of an imazapyr concentrate at 250g. l¯¹, spraying on cut stumps with metsulfron methyl at 600g.l¯¹, and foliar spraying on re-emergent lantana foliage with glyphosate at a dosage of 166g. l¯¹ in July 2008 in 20 100m2 treatment plots, 5 invaded control plots and 5 uninvaded controls. Follow-up treatments for re-sprouting lantana stumps and emerging seedlings were undertaken in June 2009. I measured effectiveness of the methods using adult lantana mortality in June 2009 and lantana seedling density in the different treatment plots during the follow-up exercise. The cost of the various methods and human labour applied were compared across the four treatments at initial clear and at follow-up. All treatments recorded a high adult lantana mortality rate, though there were no significant differences in lantana adult mortality amongst the treatments. Overall, uprooting had the highest adult mortality, followed by imazapyr, metsulfron and lastly glyphosate. Germination of lantana seedlings after clearing was high for all treatments but with no significant differences occurring between the treatments. Both adult lantana mortality and seedling density were however significantly different from the control. With labour included, chemical costs were far higher relative to uprooting, though uprooting costs were the highest when it came to the follow-up because of the emerging seedlings and some resprouting stumps. The effects of mechanical and chemical treatments on vegetation composition in the cleared areas were also assessed in order to detect any non-target and medium term effects of treatments. Contrary to expectation, none of the chemicals showed any significant effects on vegetation composition in the short and medium-term and no significant differences were found in plant species richness, diversity and seedling density between invaded and uninvaded plots at baseline, in October 2008 and in September 2009. In order to determine potential for unaided vegetation recovery in the riparian areas of the study site after lantana clearing, I conducted an investigation of soil seed banks and seed rain using 60 seed bank samples measuring 1800m³ collected from 30 invaded and uninvaded plots. Using the seedling emergence method, 1, 991 seedlings belonging to 66 species representing 27 families germinated from the seed bank. Sedges (Cyperaceae family) were the most abundant taxa in the seed banks from invaded areas, followed by Ageratum conyzoides, lantana, Triumfetta annua and Achyranthes aspera which also occurred in the uninvaded soil seed banks. The seed banks from uninvaded plots were dominated by the grass Oplismenus hirtellus. Overall, species richness, diversity and seedling density from seed banks in invaded areas did not differ significantly from seed bank in uninvaded areas and there was a low similarity in species composition when above ground vegetation was compared to seed banks from invaded and uninvaded areas. It would appear if natural regeneration occured from the current seed bank in disturbed areas, future vegetation would largely comprise of short lived, early successional species in the short term as the seed bank is dominated by non-native herbaceous weedy species. From the seed traps investigating seed rain, a total of 27 species numbering 623 individual seeds were found in the thirty 1m² seedtraps distributed in invaded and uninvaded areas at the five sites, over an intermittent period of three months. Lantana had the highest monthly arrival rate in the seed traps followed by Phoenix reclinata and Ricinus communis. The number of species with invasive potential found in the seed traps located in invaded areas was more than that found in seed traps under native vegetation cover by far. Considerable forest remnants still occur around the invaded sites, and these could serve as an important source for long-term natural re-establishment of native vegetation if seed availability by animals and wind dispersal continues, while the re-invasion of lantana is prevented by ongoing follow-ups and futher clearing of lantana invaded areas. It is concluded that while uprooting and other treatments are effective in the control of lantana, its successful control in the Victoria Falls World Heritage Site will require extensive clearing to keep it from reinvading infested areas after clearing as shown by the seed rain data. The high seedling density of lantana in the seed banks and in the cleared areas shows the need for ongoing follow-up in order to deplete soil stored seed banks. There is need for longer term research to establish what the exact follow-up requirements are in order to contain lantana re-infestation and create favourable micro-sites for native species to establish. It is predicted that ongoing lantana control in the cleared plots will most likely initiate long-term community recovery.

From the seed traps investigating seed rain, a total of 27 species numbering 623 individual seeds were found in the thirty 1m² seedtraps distributed in invaded and uninvaded areas at the five sites, over an intermittent period of three months. Lantana had the highest monthly arrival rate in the seed traps followed by Phoenix reclinata and Ricinus communis. The number of species with invasive potential found in the seed traps located in invaded areas was more than that found in seed traps under native vegetation cover by far. Considerable forest remnants still occur around the invaded sites, and these could serve as an important source for long-term natural re-establishment of native vegetation if seed availability by animals and wind dispersal continues, while the re-invasion of lantana is prevented by ongoing follow-ups and futher clearing of lantana invaded areas. It is concluded that while uprooting and other treatments are effective in the control of lantana, its successful control in the Victoria Falls World Heritage Site will require extensive clearing to keep it from reinvading infested areas after clearing as shown by the seed rain data. The high seedling density of lantana in the seed banks and in the cleared areas shows the need for ongoing follow-up in order to deplete soil stored seed banks. There is need for longer term research to establish what the exact follow-up requirements are in order to contain lantana re-infestation and create favourable micro-sites for native species to establish. It is predicted that ongoing lantana control in the cleared plots will most likely initiate long-term community recovery.

The process of utilising microbial hemicellulases requires an initial chemical pre-treatment step to delignify the sugar beet pulp (SBP). An alkaline pre-treatment with ‘slake lime’ (calcium hydroxide) was investigated using a 23 factorial design and the factors examined were: lime load; temperature and time. The analysed results showed the highest release of reducing sugars at the pre-treatment conditions of: 0.4 g lime / g SBP; 40°C and 36 hours. A partial characterisation of the Clostridium cellulovorans hemicellulases was carried out to verify the optimal activity conditions stated in literature. The highest release of reducing sugars was measured at pH 6.5 – 7.0 and at 45°C for arabinofuranosidase A (ArfA); at pH 5.5 and 40°C for mannanase A (ManA) and pH 5.0 – 6.0 and 45°C for xylanase A (XynA). Temperature studies showed that a complete loss of enzymatic activity occurred after 11 hours for ManA; and 84-96 hours for ArfA. XynA was still active after 120 hours. The optimised lime pre-treated SBP was subsequently degraded using various combinations and percentages of C. cellulovorans ArfA, ManA and XynA to determine the maximal release of reducing sugars. Synergistically, the highest synergy was observed at 75% ArfA and 25% ManA, with a specific activity of 2.9 μmol/min/g protein. However, the highest release of sugars was observed at 4.2 μmol/min/g protein at 100% ArfA. This study has initiated the research within South Africa on SBP and its degradation by C. cellulovorans. Preliminary studies show that SBP has the potential to be utilised as a second generation biofuel source.

The highly conserved, non-structural protein 2C has numerous roles during the virus life cycle and is essential for virus replication. Although the protein has been well studied in the case of PV, its interactions with the host cell during picornavirus infection is poorly understood. Theiler’s Encephalomyelitis virus (TMEV) is a picornavirus that infects mice, and is being used in our laboratory as a model in which to study the 2C protein. In this study, polyclonal antibodies against the TMEV 2C protein were generated and used to localise the protein in infected cells by indirect immunofluorescence. To produce antigen for immunisation purposes, the TMEV-2C protein sequence was analysed to identify hydrophilic and antigenic regions. An internal region of the 2C representing amino acid residues 31-210 was selected, expressed in bacteria and purified by nickel NTA affinity chromatography. Time course analysis of 2C (31-210) showed that the peptide was maximally expressed at 5 hours post induction. The peptide was solubilised using a mild detergent and 1.5 mg of purified antigen was used for immunisation of rabbits. Western blot analysis confirmed that the antibodies could detect both bacteriallyexpressed antigen, and virally-expressed 2C. Examination of virus-infected baby hamster kidney cells by immunofluorescence and confocal microscopy using the antiserum (anti-TMEV 2C antibodies) showed that the protein had a diffuse distribution upon early infection and at later stages it was located in a large perinuclear structure representing the viral replication complex. Furthermore, 2C localised to the Golgi apparatus as revealed by dual-label immunofluorescence using anti-TMEV 2C antibodies and wheat germ agglutinin (WGA). Furthermore, it was shown that TMEV infection results in changes in cell morphology and a redistribution of the cytoskeletal protein, β-actin. The successful production of antibodies that recognise TMEV 2C opens the way for further studies to investigate interactions between 2C and hostencoded factors.

Geology of the Kroonstad kimberlite cluster, South Africa

- Description: The Cretaceous (133Ma) Kroonstad Group II Kimberlite Cluster is located approximately 200km south west of Johannesburg on the Kaapvaal Craton. The cluster is made up of six kimberlite pipes and numerous other intrusive dike/sill bodies. Three of the pipes are analysed in this study, which includes the: Voorspoed, Lace (Crown) and Besterskraal North pipes. These pipes were emplaced at surface into the Karoo Supergroup, which is comprised of older sedimentary rocks (300-185Ma) overlain by flood basalts (185Ma). At depth the pipes have intruded the Transvaal (2100-2600Ma) and Ventersdorp (2700Ma) Supergroups, which are comprised dominantly of carbonates and various volcanic units respectively. The pipes have typical morphology of South African pipes with circular to sub-circular plan views and steep 82o pipe margins. The Voorspoed pipe is 12ha in size and is characterised by the presence of a large block of Karoo basalt approximately 6ha in size at the current land surface. This large basalt block extends to a maximum of 300m below the current land surface. The main Lace pipe is 2ha is size with a smaller (<0.5ha) satellite pipe approximately 50m to the west. No information is available on the morphology of the Besterskraal North pipe as it is sub-economic and no mining has occurred. Samples from the Besterskraal North pipe were collected from the De Beers archives. The Kroonstad Cluster has been subjected to approximately 1750m of erosion post-emplacement, which has been calculated by the analysis of the crustal xenoliths with the pipe infill. The hypabyssal kimberlite from the three pipes shows a gradational evolution in magma compositions, indicated by the mineralogy and geochemistry. The Lace pipe is the least evolved and has characteristics more similar to Group I kimberlites. The Voorspoed and Besterskraal North kimberlite are intermediately and highly evolved respectively. The gradational evolution is marked by an increase in SiO2 and Na2O contents. Furthermore the occurrence of abundant primary diopside, aegirine, sanidine, K-richterite and leucite indicates evolution of the magma. The root zones of the pipes are characterised by globular segregationary transitional kimberlite, which is interpreted to be hypabyssal and not the result of pyroclastic welding/agglutination. The hypabyssal transitional kimberlite (HKt) is characterised by incipient globular segregationary textures only and the typical tuffisitic transitional kimberlite (TKt) end member (Hetman et al. 2004) is not observed. The HKt contact with the overlying volcaniclastic kimberlite (VK) infill is sharp and not gradational. The presence of HKt in the satellite blind pipe at Lace further indicates that the distinct kimberlite rock type must be forming sub-volcanically. The HKt is distinctly different at the Voorspoed and Lace pipes, which is likely a result of differing compositions of the late stage magmatic liquid. Microlitic clinopyroxene is only observed at the Lace HKt and is interpreted to form as a result of both crustal xenolith contamination and CO2 degassing. Furthermore the HKt is intimately associated with contact breccias in the sidewall. The root zones of the Kroonstad pipes are interpreted to form through the development of a sub-volcanic embryonic pipe. The volcaniclastic kimberlite (VK) infill of the Kroonstad pipes is not typical of South African tuffisitic Class 1 kimberlite pipes. The VK at Voorspoed is characterised by numerous horizontally layered massive volcaniclastic kimberlite (MVK) units, which are interpreted to have formed in a deep open vent through primary pyroclastic deposition. MVK is the dominant rock type infilling the Voorspoed pipe, however numerous other minor units occur. Normally graded units are interpreted to form through gravitational collapse of the tuff ring. MVK units rich in Karoo basalt and/or Karoo sandstone are interpreted to form through gravitational sidewall failure deep within an open vent. Magmaclasts are interpreted to form in the HKt during the development of an embryonic pipe and therefore the term autolith or nucleated autolith may be applied. Debate on the validity of the term nucleated autolith is beyond this study and therefore the term nucleated magmaclast is used to refer to spherical magmaclasts in the VK. The emplacement of the Kroonstad pipes is particularly complex and is not similar to typical Class 1 tuffisitic kimberlites. However the initial stage of pipe emplacement is similar to typical South African kimberlites and is interpreted to be through the development of an embryonic pipe as described by Clement (1982). The vent clearing eruption is interpreted to be from the bottom up through the exsolution of juvenile volatiles and the pipe shape is controlled by the depth of the eruption (+/-2km) (Skinner, 2008). The initial embryonic pipe development and explosive eruption is similar to other South African kimberlites, however the vent is cleared and left open, which is typical of Class 2 Prairies type and Class 3 Lac de Gras type pipes. The latter vent infilling processes are similar to Class 3 kimberlites from Lac de Gras and are dominated at the current level by primary pyroclastic deposition.

Fantasy leagues, provide a platform for fans to test their hand at this selection procedure. Twenty20 cricket is a new and exciting form of cricket and has become very popular in recent years. This research focuses on bringing these concepts together by proposing a binary integer program to determine a team selection strategy for fantasy league cricket. This is done in a Twenty20 setting. The approach used in this study focuses on evaluating the effectiveness of previously developed performance measures in a fantasy league setting. Adjustments to these measures are made and new measures are proposed. These measures are then used to select a fantasy league team using a prospective approach. This is done to provide fantasy league participants with a mathematical procedure for fantasy league team selection.

- Description: The topical application of many substances, including drugs, enzymes, moisturizers and fragrances, contributes largely to the cosmetic and pharmaceutical industries. These components are often volatile in nature and dissipate in a matter of hours. When considering the different types of slow release systems, an overwhelming variety of these systems is available. Each one of the systems is unique in a way, and is designed to perform a particular function, whether it facilitates the controlled release of an active into the body via the skin surface (transdermal delivery) or whether it reduces the rate of loss of an active from the skin surface to the surrounding environment. For the purpose of this study, a previously existing fixative formulation which is believed to reduce the rate of loss of an active component to the environment, through film formation on the skin surface, was investigated. Alternative ingredients or components were incorporated together with the original fixative formulation ingredients into an experimental design which investigates the effect of each group of the components present. 18 formulations with various concentrations of the components within the groups and specified upper and lower limits for each component were formulated. The fixative properties of the formulations were analysed through the incorporation of a fixed amount of a simple fragrance molecule, 4- methoxybenzaldehyde, into each formulation and evaporation studies were conducted in an environmental room at 28±1° C over a period of 5 hours followed by gas chromatography analysis and finally data analyses using statistical methods. The most efficient fixative formulation was established using regression analysis. The fragrance compound in this formulation was found to evaporate at a rate of 0.47 g/L per hour. The least efficient fixative formulation lead to the loss of 0.78 g/L of the fragrance component per hour. From the calculated fragrance concentrations, the rate constant for each individual fixative formulation could be calculated and response surface 8 modelling by backward regression was used in order to determine how each component contributes to the rate of loss of the fragrance compound. Since the sum of the original ingredient and its alternative was constant, each of the original ingredients was coupled directly to its alternative and no conclusion could be made about the contribution of individual components. By increasing the concentration of Hydroxypropylcellulose (HPC) 100K and its alternative HPC 140K, while keeping the effects of the other components constant, a decrease in the rate of fragrance loss was observed. The same conclusion could be made when increasing the concentrations of PEG-12 Dimethicone and its alternative cetyl dimethicone (decreases the evaporation rate). An interaction took place between HPC 100K and PEG-12 dimethicone and their alternatives. The negative effect was, however, not as strong as the combined positive effect on the rate of fragrance loss of the individual components HPC and PEG-12 dimethicone. Evidence suggested that the removal of the components polyvinylpyrrolidone and its alternative, polyurethane-32 (Baycusan® C1003), would improve the effectiveness of the fixative formulation in terms of its slow release properties. A confirmation experiment established that the exclusion of these components from the fixative formulation does improve the “slow release” properties thereof. A larger, more intricate design is required to investigate the effect of each one of the individual components and where the sum of the components (original and its alternative) is not constant.

- Description: The biology of key citrus pests Thaumatotibia leucotreta Meyrick (Lepidoptera: Tortricidae), Ceratitis capitata Wiedemann (Diptera: Tephritidae) and Ceratitis rosa Karsch (Diptera: Tephritidae) includes their dropping from host plants to pupate in the soil below citrus trees. Since most EP fungi are soil-borne microorganisms, the development and formulation of alternative control strategies using these fungi as subterranean control agents, targeted at larvae and pupae in the soil, can potentially benefit existing IPM management of citrus in South Africa. Thus, a survey of occurrence of entomopathogenic fungi was undertaken on soils from citrus orchards and natural vegetation (refugia) on conventionally and organically managed farms in the Eastern Cape Province in South Africa. A method for baiting soil samples with citrus pest T. leucotreta and C. capitata larvae, as well as with the standard bait insect, Galleria mellonella Linnaeus (Lepidoptera: Pyralidae), was implemented. Sixty-two potentially useful entomopathogenic fungal isolates belonging to four genera were collected from 288 soil samples, an occurrence frequency of 21.53%. The most frequently isolated entomopathogenic fungal species was Beauveria bassiana (Balsamo) Vuillemin (15.63%), followed by Metarhizium anisopliae var. anisopliae (Metschnikoff) Sorokin (3.82%). Galleria mellonella was the most effective insect used to isolate fungal species (χ2=40.13, df=2, P≤ 0.005), with a total of 45 isolates obtained, followed by C. capitata with 11 isolates, and T. leucotreta with six isolates recovered. There was a significantly (χ2=11.65, df=1, P≤ 0.005) higher occurrence of entomopathogenic fungi in soil samples taken from refugia compared to cultivated orchards of both organically and conventionally managed farms. No significant differences were observed in the recovery of fungal isolates when soil samples from both farming systems were compared. The physiological effects and host range of 21 indigenous fungal isolates obtained in the Eastern Cape were investigated in the laboratory to establish whether these isolates could be effectively used as biological control agents against the subterranean life stages of C. rosa, C. capitata and T. leucotreta. When these pests were treated with a fungal concentration of 1 x 10⁷ conidia ml⁻¹, the percentage of T. leucotreta adults which emerged in fungal treated sand ranged from 5 to 60% (F=33.295; df=21; P=0.0001) depending on fungal isolate and the percentage of pupae with visible signs of mycosis ranged from 21 to 93% (F= 96.436; df=21; P=0.0001). Based on fungal isolates, the percentage adult survival in C. rosa and C. capitata ranged from 30 to 90% and 55 to 86% respectively. The percentage of C. rosa and C. capitata puparia with visible signs of mycosis ranged from 1 to 14% and 1 to 11% respectively. Deferred mortality due to mycosis in C. rosa and C. capitata adult flies ranged from 1 to 58% and 1 to 33% respectively, depending on fungal isolate. Entomopathogenic fungal isolates had a significantly greater effect on the adults of C. rosa and C. capitata than they did on the puparia of these two fruit fly species. Further, C. rosa and C. capitata did not differ significantly in their response to entomopathogenic fungi when adult survival or adult and pupal mycosis were considered. The relative potency of the four most virulent Beauveria isolates as well as the commercially available Beauveria bassiana product, Bb Plus® (Biological Control Products, South Africa), were compared against one another as log-probit regressions of mortality against C. rosa, C. capitata and T. leucotreta which all exhibited a dose-dependent response. Against fruit flies the estimated LC50 values of all five Beauveria isolates ranged from 5.5 x 10¹¹ to 2.8 x 10¹² conidia/ml⁻¹. There were no significant differences between the relative potencies of these five fungal isolates. When T. leucotreta was considered, isolates: G Moss R10 and G 14 2 B5 and Bb Plus® were significantly more pathogenic than G B Ar 23 B3 and FCM 10 13 L1. The estimated LC₅₀ values of the three most pathogenic isolates ranged from 6.8 x 10⁵ to 2.1 x 10⁶ conidia/ml⁻¹, while those of the least pathogenic ranged from 1.6 x 10⁷ to 3.7 x 10⁷ conidia/ml⁻¹. Thaumatotibia leucotreta final instar larvae were exposed to two conidial concentrations, at four different exposure times (12, 48, 72 and 96 hrs) and showed an exposure time-dependant relationship (F=5.43; df=3; P=0.001). At 1 x 10⁷conidia/ml⁻¹ two Beauveria isolates: G Moss R10 and G 14 2 B5 were able to elicit a response in 50% of test insects at 72 hrs (3 days) exposure. Although a limited amount of mycosis was observed in the puparia of both fruit fly species, deferred adult mortality due to mycosis was high. The increased incidence of adult mortality suggests that post emergence mycosis in adult fruit flies may play a more significant role in field suppression than the control of fruit flies at the pupal stage. The increased incidence of pupal mortality, as well as the relatively low concentrations of conidia required to elicit meaningful responses in T. leucotreta pupae may suggest that pre-emergent control of false codling moth will play a more significant role in field suppression than the control of adult life stages using indigenous isolates of entomopathogenic fungi. Various entomopathogenic fungal application techniques targeted at key insect pests within integrated pest management (IPM) systems of citrus are discussed.

Isolation of xylanolytic multi-enzyme complexes from Bacillus subtilis SJ01

- Description: Cellulose and hemicellulose account for a large portion of the world‘s plant biomass. In nature, these polysaccharides are intertwined forming complex materials that require multiple enzymes to degrade them. Multi-enzyme complexes (MECs) consist of a number of enzymes working in close proximity and synergistically to degrade complex substrates with higher efficiency than individual enzymes. The cellulosome is a cellulolytic MEC produced by anaerobic bacteria that has been studied extensively since its discovery in 1983. The aim of this study was to purify a cellulolytic and/or hemicellulolytic MEC from an aerobic bacterium of the Bacillus genus. Several bacterial isolates were identified using morphological characteristics and 16S rDNA sequencing, and screened for their ability to degrade cellulose and xylan using a MEC. The isolate that produced a high molecular weight protein fraction with the greatest ability to degrade Avicel®, carboxymethyl cellulose (CMC) and birchwood xylan was identified as Bacillus subtilis SJ01. An optimised growth medium, consisting of vitamins, trace elements, birchwood xylan (as the carbon source), and yeast and ammonium sulphate (as the nitrogen sources), increased the production of CMCase and xylanase enzymes from this bacterium. The removal of a competing bacterial strain from the culture and the inhibition of proteases also increased enzyme activities. A growth curve of B. subtilis SJ01 indicated that xylanase production was highest in early stationary growth phase and thus 84 hours was chosen as the best cell harvesting time. To purify the MECs produced by B. subtilis SJ01 size-exclusion chromatography on a Sephacryl S-400 column was used. It was concluded that (for the purposes of this study) the best method of concentrating the culture supernatant prior to loading onto Sephacryl S-400 was the use of ultrafiltration with a 50 kDa cut-off membrane. Two MECs, named C1 and C2 of 371 and 267 kDa, respectively, were purified from the culture supernatant of B. subtilis SJ01. Electrophoretic analysis revealed that these MECs consisted of 16 and 18 subunits, respectively, 4 of which degraded birchwood xylan and 5 of which degraded oat spelt xylan. The MECs degraded xylan substrates (C1: 0.24 U/mg, C2: 0.14 U/mg birchwood xylan) with higher efficiency than cellulose substrates (C1: 0.002 U/mg, C2: 0.01 U/mg CMC), and could therefore be considered xylanosomes. Interestingly, the MECs did not bind to insoluble birchwood xylan or Avicel® and did not contain glycosylated proteins, which are common features of cellulosomes. This study is, therefore, important in revealing the presence of MECs that differ from the cellulosome and that may have particular application in industries requiring high xylanase activity, such as the paper and pulp industry. The abundant genetic information available on B. subtilis means that this organism could also be used for genetic engineering of cellulolytic/hemicellulolytic MECs.

Six transects were sampled between the spring high water mark (HWST) and the bottom of the river channel in the lower estuary. Fifteen replicate samples were collected along each transect using a Van Veen type grab (211 cm2 bite) during each of three sampling sessions. Samples were sieved through a 500 μm mesh bag and the invertebrates stored in bottles for further analysis in the laboratory. Additional grab samples were collected for sediment particle size analysis and organic matter. Physical variables measured at each transect included: salinity, temperature, dissolved oxygen, depth, pH, percentage mud, organic content and turbidity. Sediment compactness was measured at all intertidal transects and additional sediment samples were collected at mid shore and high shore transects for percentage water content analysis. A total of 104 species were recorded along the intertidal-subtidal gradient in the sampling area. Species richness was higher in the subtidal zone compared to the intertidal zone and polychaetes numerically dominated the macrozoobenthic community at most transects, during all three sessions. At high shore transects the community was characterised by having fewer species, consisting mostly of brachyurans, polychaetes and gastropods. Shannon diversity index (H’) was generally higher for subtidal transects (x¯ = 2.3; range: 2.8 to 1) than for intertidal transects (x¯ = 1.4; range: 2.2 to 0.6) indicating that the distribution of individuals among species in the intertidal zone experienced greater variability. Results for Hill’s numbers followed the same trend as Shannon diversity with subtidal communities mostly consisting of abundant species followed by very abundant species. Intertidal communities generally exhibited lower numbers of abundant and very abundant species. Sedimentary characteristics played a major role in structuring benthic communities in comparison to other physico-chemical variables. Organic content and mud content of the substrate were identified as important factors influencing community patterns observed along the subtidal-intertidal gradient. In addition, sediment compactness and water content of the substrate was found to influence intertidal community structure. Subtidal community structure possibly had a greater dependence on seasonal variations in abiotic and/or biotic factors. Cluster dendrograms used in conjunction with MDS ordination mapping revealed that macrozoobenthic communities were generally distinct between high shore intertidal transects iii and subtidal transects. Most species exhibited a broad spatial distribution along the subtidal-intertidal gradient with mid and high shore transects being the exception. Most species also exhibited marked shifts in abundance and this was especially noticeable at the transition between the subtidal and intertidal zone. Two polychaete species, Prionospio sexoculata and Capitella capitata, were very abundant species and featured amongst the most numerically dominant species collected during each sampling session.
