- Description: Two readily synthesized Sn(IV) porphyrins (SnP, SnPH) have been prepared with and without the cationic triphenylphosphonium moiety (TPP+), which have high singlet oxygen quantum yields (ca. 0.72) and long triplet state lifetimes. The Sn(IV) porphyrin with a TPP+ moiety (SnPH) exhibits favorable photodynamic activity against MCF-7 cells with IC50 values of 2.9 μM. SnPH exhibits higher cellular uptake than SnP in MCF-7 cells. A Rhodamine 123 (Rh123) assay showed that SnPH targets mitochondria and induces apoptosis by generating reactive oxygen species. The results demonstrate that this structural modification strategy merits further in-depth study.

- Description: The synthesis and characterization of A3B-type phthalocyanines, ZnPc1–4, bearing bulky 2,6-diisopropylphenoxy-groups or chlorine atoms on isoindoline units “A” and either one or two carboxylic anchors on isoindoline unit “B” are reported. A comparison of molecular modelling with the conventional time dependent—density functional theory (TD-DFT) approach and its simplified sTD-DFT approximation provides further evidence that the latter method accurately reproduces the key trends in the spectral properties, providing colossal savings in computer time for quite large molecules. This demonstrates that it is a valuable tool for guiding the rational design of new phthalocyanines for practical applications.

- Description: In the present work, the performance of aptamer and antibody bioreceptors for the detection of the human epidermal growth factor receptor (HER2) cancer biomarker on a glassy carbon electrode is reported. The carboxylic acid group rich graphene quantum dots (GQDs) modified with gold nanoparticles and a porphyrin binuclear framework (CoP-BNF) were used to modify the glassy carbon electrode. The aptamer and antibody were both amine functionalized and attached to GQDs and CoP-BNF through an amide bond. The designed immunosensors and aptasensors in this work were characterized using electrochemical impedance spectroscopy. The aptasensors, compared to the immunosensors gave better limit of detection values. The aptasensor outperforms the immunosensor in terms of its reusability and storability, while the immunosensor could not be regenerated for subsequent experiments. The potential applicability of all sensors in this work was also investigated, by detection of HER2 in spiked human serum with acceptable results.

- Description: A method is presented for the electrochemical detection of the breast cancer biomarker human epidermal growth factor receptor 2 (HER2). A glassy carbon electrode was modified using two techniques known as sequential adsorption and electro-polymerization, and the results are compared. The highly conductive polypyrrole (PPy) is used, in the presence of sulfur/nitrogen doped graphene quantum dots (SNGQDs) and a known cobalt phthalocyanine (CoPc). The different nanomaterials were used as an immobilization platform for the HER2 specific HB5 aptamer via amide linkage. The nanomaterials were arranged in various ways on the glassy carbon electrode, to investigate the effect of the electrode interface on the operational characteristics of a biosensor. The immobilized aptamer selectively recognizes HER2 on the electrode interface, and this leads to an increased charge transfer resistance (Rct) of the electrode when using ferricyanide as the electrochemical probe. The developed immunosensors showed high sensitivity with the best detection limit of 0.00141 ng/mL. The results showed that the method is simple and sensitive enough for the determination of HER2 in serum samples with good reproducibility and accuracy.

- Description: The role of the biointerface design towards the development of an impedimetric biosensor for the electrochemical detection of human epidermal growth factor receptor 2 (HER2) is investigated. Two novel cobalt phthalocyanines: cobalt tetraphenyl acetic acid phthalocyanine and cobalt tetraphenyl propionic acid phthalocyanine are compared as signal amplifiers and immobilization platforms of the HB5 aptamer towards the electrochemical detection of HER2. In addition, the phthalocyanines are coupled with the metal based cerium oxide nanoparticles. The efficiency of each electrode modification step and the performance of the constructed aptasensors were assessed by impedance spectroscopy. The aptasensors showed very low limit of detection values (all less than 0.2 ng/mL) with high sensitivity and stability. Furthermore, the aptasensors showed very good performance even in human serum samples. Considering these results, the aptasensors demonstrate great potential for improved monitoring of human epidermal growth factor receptor 2 levels for the management of breast cancers.

- Description: Ibuprofen (IBU) is a common anti-inflammatory drug that is consumed by many individuals in the world. As such, analytical studies have detected high concentrations of the drug in many waterbodies, which poses a risk of harmful effects on the environment and public health. The hydroxyl radical technologies, a collective of techniques also known as advanced oxidation processes (AOPs), can be utilized to degrade this emerging pollutant. In this study, the photodegradation of ibuprofen using 5,10,15,20-tetrakis(4-bromophenyl) porphyrin conjugated to graphene quantum dots was investigated using a custom-built photoreactor. Three different concentrations of IBU (200, 300 and 500 μM) were utilized as initial concentrations. The pH of the IBU was varied between acidic (pH 3.0), natural (pH 5.0) and alkaline (pH 9.0) to note the effect on IBU degradation as a function of time. The Highest ФΔ was obtained for InTBrP- GDQs (ФΔ = 0.80), followed by InTBrP (ФΔ = 0.74). The photodegradation efficiency of the TBrP-GQDs and InTBrP-GQDs were determined to be 43.2 and 76.1% respectively.

- Description: Novel 5-(pyridyl)-10-15-20-tris(4-bromophenyl) porphyrin (complex 1), indium metal derivative (complex 2), and quaternized derivative (complex 3) were synthesized and conjugated to graphene quantum dots (GQDs). The conjugation of the porphyrins to GQDs was through ππ-ππ stacking. Herein, the ππ-ππ stacking approach was used to avoid covalent conjugation which might compromise the intrinsic chemical and physical properties. The photodynamic activities of the proposed nanomaterials were assessed towards Staphylococcus aureus cell obliteration. The photophysical properties of the prepared complexes were also studied prior to the application. Moreover, a decrease in fluorescence lifetimes was observed upon metalation of complex 1. As anticipated, singlet oxygen quantum yield (ΦΔ)ΦΔ) increased notably upon heavy metal (indium) insertion and upon composite formation. Antimicrobial photodynamic therapy comparative studies were done on quaternized and unquaternized indium porphyrins conjugated to GQDs. Complex 3-GQDs exhibited the highest antibacterial activities compared to other complexes, and this was attributed to the high ΦΔΦΔ which plays an imperative role in photodynamic therapy applications.

The dark toxicity and photodynamic therapy studies were conducted for 2, NGQDs and their conjugates using MCF-7 breast cancer cells. The cell viability for dark toxicity of all the compounds was above 90%, and 2-B-NGQDs showed high PDT activity at a concentration of 40 µg/mL with cell viability of 22%.

- Description: Heterogeneous photocatalysis is a promising approach for environmental remediation from contaminants including microorganisms and organic pollutants. In this work, hematite nanofibers are fabricated and modified with a novel monosubstituted Pc (4) as well as an asymmetrical tetrasubstituted Pc (5) with the aim of creating hybrid photocatalysts. The photocatalytic activities of the unmodified and phthalocyanine modified hematite nanofibers were compared based on their efficiencies in the photoinactivation of S. aureus and photooxidation of methyl orange. For both applications, the hybrid nanofibers were found to be more efficient photocatalysts than the unmodified hematite nanofibers. Comparison of the modified nanofibers (4-Fe2O3 and 5-Fe2O3) showed that they have comparable antibacterial activity while the 5-Fe2O3 nanofibers are the best for the photooxidation of methyl orange. The singlet oxygen generation efficiency, high activity, versatility, regenerability and thus reusability of the fabricated hybrid nanofibers makes them ideal candidates for real life water treatment studies.

- Description: The synthesis, photophysicochemical and photodynamic therapy (PDT) activities of benzothiazole substituted zinc phthalocyanine (Pc): 1 (asymmetrically substituted and composed of no charges), 2 (asymmetrically substituted and composed of three positive charges), and 3 (symmetrically substituted and composed of four positive charges), are presented. The triplet and singlet oxygen quantum yields were highest for complex 2 showing the importance of asymmetry and charge. The complexes are covalently and non-covalently linked to B doped detonation nanodiamonds (B@DNDs) to yield nanohybrids (B@DNDs-1, B@DNDs-2, B@DNDs-3). The presence of B@DNDs, asymmetry and positive charge resulted in improved PDT with the lowest cell viability being observed for B@DNDs-2 at 5%. The cell viability ranged from 5% to 7% for the nanohybrids compared to 19–26% for Pcs alone.

- Description: This work focused on syntheses of novel asymmetrically tetra substituted Zn phthalocyanines (ZnPc) each containing a single carboxyl group, with the other substituents bearing alkynyl (complex 1) and fluorine containing groups (complexes 2 and 3). The complexes were conjugated to cobalt tungstate nanoparticles and employed as efficient photocatalysts for degradation of tetracycline in water. ZnPc complexes and their conjugates showed good photophysical and photochemical properties behaviour with complex 1 giving higher triplet and singlet oxygen quantum yields compared to 2 and 3. Complex 1 showed higher activity towards the photodegradation of tetracycline compared to complexes 2 and 3, with higher kobs and initial rates for the former. The photocatalysis obeyed the Langmuir-Hinshelwood kinetic model.

- Description: In this study, we report for the first time on the photodynamic antimicrobial chemotherapy (PACT) of asymmetrical zinc(II) phthalocyanines and their conjugates with nickel tungstate (NiWO4), cobalt tungstate (CoWO4) and bismuth tungstate (Bi2WO6) nanoparticles. CoWO4, NiWO4 and Bi2WO6 were capped with glutathione and covalently linked to three asymmetrical zinc(II) phthalocyanines for PACT. The plate count method was used to investigate the effect of covalently linking asymmetrical zinc(II) phthalocyanines to nanoparticles under light (for PACT against Staphylococcus aureus) and in the dark. Both Pc complexes and the conjugates indicated minimum dark-toxicity. The conjugates showed greater antimicrobial activity against a gram-positive and drug resistant bacteria, Staphylococcus aureus, than Pc complexes alone.

- Description: A new Schiff base probe (QT) consisting of 8-aminoquinoline (Q) and thiophene-2-carboxaldehyde (T) moieties has been synthesized. QT undergoes chelation-enhanced fluorescence quenching when exposed to Hg2+ due to coordination by the sulfur and nitrogen atoms of QT thus forming a facile “turn-off” sensor. The formation of the chelation complex was confirmed by UV–visible absorption and emission spectral measurements, 1H NMR titration and density functional theory calculations. These studies revealed that the probe exhibits high selectivity and sensitivity towards Hg2+ in the presence of other common metal ions. A low detection limit of 23.4 nM was determined and a Job plot confirmed a 2:1 stoichiometry between QT and Hg2+. The potential utility of QT as a sensor for Hg2+ ions in human HeLa cells was determined by confocal fluorescence microscopy, and its suitability for use in the field with environmental samples was tested with Whatman filter paper strips.

- Description: In this work, the preparation of glucose encapsulating nanoliposomes was achieved using two different lipid formulations, labelled as F1 and F2. Both formulations contained phosphatidylcholine (PC), oleylamido-4-butanoic acid (OABA) and in addition, F1 had cholesterol (CHO) while F2 contained cholesteroyl hemisussinate (CHEMS). These formulations were studied for their pH sensitivity and controlled release of encapsulated glucose for indirect detection of prostate-specific antigen (PSA) using sandwich immunoassay. As a signal generator, encapsulated glucose in nanoliposomes was quantified directly using the personal glucose meter (PGM) and colorimetrically using peroxidase property of horseradish peroxidase (HRP) enzyme and Pd|PdO as nanozymes. Controlled release of the encapsulated glucose was achieved using the pH effect or Triton-X 100 as a surfactant to destabilize the liposomal structure. The F2 formulation showed maximum controlled release at acidic phosphate buffer saline (PBS, pH 5.0). The concentration of encapsulated glucose was found to be high in F2 formulation and these were applied for the indirect detection of PSA. The limit of detection (LOD) values for PSA were found to be 53 fg mL−1, 64 fg mL−1 and 10 fg mL−1 when HRP, Pd|PdO and PGM were respectively used. The detection signal was linear over a wide concentration range for PSA including the clinical range of 4–10 ng mL−1. The HRP system showed low LOD value when compared with Pd|PdO nanozymes. PGM system gave lowest LOD values owing to the sensitivity of the system towards glucose. Pd|PdO nanozyme system showed good stability over a wide temperature up to 80 °C. PGM system required less reaction time (2 min), low reagents and results were readily generated in digital format.

- Description: This work reports on the synthesis and electrochemical sensing properties of benzothiazole substituted cobalt phthalocyanine (CoPc) when π-π stacked on detonation nanodiamonds (to form CoPc-DNDs(ππ). The synthesized materials were characterized using UV–visible, mass, Fourier transform infrared, and Raman spectroscopies as well as transmission electron microscopy and dynamic light scattering. The electrochemical studies were conducted using cyclic voltammetry, electrochemical impedance spectroscopy and chronoamperometry. Hydrazine was utilized as an analyte of interest, due to its mutagenic and carcinogenic effects. Glassy carbon electrodes (GCE) were modified with DNDs, CoPc, and CoPc-DNDs(ππ) represented as GCE/DNDs, GCE/CoPc and GCE/ CoPc-DNDs(ππ). GCE was also modified sequential addition of the CoPc and DNDs onto the GCE, represented as GCE/CoPc-DNDs(seq) when CoPc is placed before DNDs on the electrode and GCE/DNDs-CoPc(seq) when DNDs are placed before CoPc, where seq represents sequential. GCE/CoPc-DNDs(ππ) electrode gave better results in terms of limit of detection (1.68 μM), sensitivity (9.59 μA.mM−1) and catalytic rate constant (1.25 × 106 M−1 s−1).

- Description: This work investigates the use of manganese phthalocyanines (MnPcs) in the electrochemical detection of nitrite. These are two Schiff base derivatives: tetrakis 4-(4-thio-thiophene-2-ylmethanamino) phthalocyaninato Mn(III)Cl (2) and tetrakis 4-(3-(pyrrolidine-1-yl)propan-1-amino)phthalocyaninato Mn(III)Cl (3) and one aldehyde: tetrakis(4-formylphenoxy)phthalocyaninato Mn(III)Cl (1). The MnPc complexes were adsorbed onto the glassy carbon electrode surface using the drop-dry method and used as electrocatalysts to detect nitrite. Nitrite oxidation peak potentials obtained using cyclic voltammetry ranged between 0.70 and 0.84 V. Detection limits as low as 0.16 µM and sensitivity values as high as 276 µMmM−1 were obtained through the use of chronoamperometry. The Schiff base MnPcs 2 and 3 exhibited better nitrite detection properties (in terms of limit of detection and sensitivity) than their aldehyde precursor (1).
