Assessment of bioflocculant production by two marine bacteria isolated from the bottom sediment of marine Algoa Bay

- Description: Bioflocculants are polymers, mostly, of microbial origin which floc out suspended particles from liquid medium. The ability of these biopolymers to remove suspended particles from solutions is termed bioflocculation, and the efficiency of flocculation activities depends on the characteristics of the flocculants. In comparison with conventionally used flocculants, bioflocculants have the advantage of being safe (no toxic effects known), biodegradable and harmlessness to the environment. The study assessed production of bioflocculant by two marine bacteria from the bottom sediment of marine environment. The 16S rDNA was used for identification, and the two bacteria species were identified as Enterococcus hirae and Bacillus thuringiensis. Factors affecting the production and activity of the bioflocculants produced by these two organisms were studied. The bacteria optimally produced bioflocculant with fructose (91.7%) and urea (91%) as sole carbon and nitrogen sources respectively. Mg2+ (87%) and Ca2+ (86%), likewise, served as best cation sources on the production of the bioflocculant at pH 5(93%). Additionally, the flocculating activity of the bioflocculant increased with the addition of Mg2+ (81%) and Na+ (81%), and the highest flocculating activity was at pH 5 of the kaolin clay. The Fourier transform infrared spectroscopy (FTIR) shows that the bioflocculant is a glycoprotein. The second bacterium (Bacillus thuringiensis) produced bioflocculant optimally when the media had mixed nitrogen sources (Urea, ammonium chloride and tryptone (67%)) and glucose (85.65%) as a sole carbon source, also Ca2+ (74.6%) was the best cation that induced the production of bioflocculant. After purification, the bioflocculant flocculated optimally in alkaline pH 12 (81%) in the presence of Mn2+ (73%) and Ca2+ (72.8%). Chemical analysis of the bioflocculant revealed it to be a polysaccharide. Both bioflocculants flocculate efficiently and can be used to replace synthetic flocculants in water treatment, wastewater, in downstream processing, and processing of food and chemicals and other industrial uses of flocculants. Challenges though (i) are to develop conditions for large scale production of the bioflocculant, (ii) to do further characterization of the both bioflocculants (iii) to assess the bioflocculants for treatments of water/wastewater, and to apply it in various industrial processes.

Assessment of the effects of stream channel incision on soil water levels, soil morphology and vegetation in a wetland in the Hogsback area, Eastern Cape, South Africa

- Description: Wetland degradation in the form of channel incisioning can significantly alter the hydrological functioning of a wetland. In this study in a small headwater wetland in the Hogsback area, Eastern Cape Province, the impact of channel incisioning on soil water levels and soil morphology was examined. A good correlation (R2 = 0.89) exists between the depth of channel incisioning and average water table depths in most of the 21 installed piezometers. In localised cases the upslope supply of water was in equilibrium with drainage from the piezometers. Although all the studied soils show hydromorphic characteristics, those continuously saturated close to the surface exhibit redox accumulations in oxygen supplying macropores, whereas gleyic colour patterns occur deeper in soils where the water table has been lowered by channel incision. The nature and occurrence of different hydromorphic soil indicators observed confirm the contribution of soil morphology as a valuable indicator of long-term average soil water conditions. The study evaluates the effects of hydrological processes on plant species composition and distribution in a Hogsback wetland. Twenty-four circular plots with a radius of 2 m were established between March and August 2013. Within each sample plot, the habitat information and species present were recorded including Braun-Blanquet cover-abundance value for each and every species present in the plot. Plant species were identified in the field and the unknown plant species were collected, pressed, oven-dried and identified at the National Herbarium, South African Biodiversity Institute (SANBI), Pretoria and Schonland Herbarium, Rhodes University, Grahamstown. A total of 39 taxa belonging to 19 families and 34 genera were recorded from the Hogsback wetland. Of the documented taxa, three were exotic and three endemic to South Africa, indicating diversity and dynamic nature of Hogsback wetland flora. Plant families with the highest number of species were: Poaceae (11 species), Asteraceae (5 species), Onagraceae and Cyperaceae with 3 taxa each. The low number of exotic plant species in Hogsback wetland (three taxa in total) indicates limited anthropogenic influence. Unique species recorded in Hogsback wetlands are three species that are endemic to South Africa, namely, Alchemilla capensis, Helichrysum rosum and Lysimachia nutans. Five main floristic associations were identified from the Hierarchical Cluster Analysis whilst Detrenched .Correspondence Analysis (DCA) identified four groups. The Canonical Correspondence Analysis (CCA) indicates that edaphic factors, particularly in an area covered with water, erosion category, organic matter content and water table depth were the most important environmental variables measured accounting for the vegetation pattern present in the Hogsback wetland.

Assessment of the quality indices and prevalence of Escherichia coli pathotypes in selected rivers of Osun state, Southwestern Nigeria

- Description: Surface waters are important freshwater sources used for domestic, industrial, agricultural and recreational activities, and the availability of good quality freshwater is indispensable for preventing water-borne diseases and improving quality of life especially in communities that lack pipe-borne water. Water samples were collected from ten rivers at different locations in Osun State, Southwestern Nigeria. A total of 12 physicochemical parameters, counts of total coliforms (TC) and Escherichia coli isolates were determined using standard analytical procedures. Confirmed Escherichia coli isolates (n=300) were assessed for the presence of 10 virulence genes (VGs) associated with Escherichia coli strains causing intestinal and extra-intestinal infections. The recovered Escherichia coli isolates were elucidated for their antibiogram profiling by disk diffusion method and the resistant isolates were further profiled for their genotypic antimicrobial resistance by polymerase chain reaction technique. The physicochemical qualities ranged as follows: pH (6.9 - 7.6), temperature (26 – 29 ºC), turbidity (2.28 – 9.46 NTU), electrical conductivity (229 – 581 μS/cm), nitrate (0.03 – 0.05 mg/L), nitrite (0.00 – 0.01 mg/L), sulphate (3.33 – 20.33 mg/L), chloride ions (7.83 – 27.33 mg/L), dissolved oxygen (4.23 – 5.57 mg/L), total dissolved solids (56 – 184 mg/L), total hardness (78 – 519 mg/L) and alkalinity (50.67 – 146.67 mg/L). Statistical analysis showed that pH, temperature, electrical conductivities, nitrates, nitrites, chloride, dissolved oxygen, total dissolved solid, total hardness and alkalinity were significantly different (P < 0.05), whereas turbidity and sulphate were not significantly different (P ˃ 0.05) from each parameter with respect to sampling sites. While the VG lt for enterotoxigenic E. coli had the highest prevalence of 45%, the enteropathogenic E. coli genes eae and bfp were detected in 6% and 4% of the isolates respectively. The VGs stx1 and stx2 specific for the enterohemorrhagic E. coli pathotypes were equally detected in 7% and 1% of the isolates respectively. Also, the VG eagg harboured by enteroaggregative pathotype and diffusely-adherent E. coli VG daaE were detected in 2% and 4% of the isolates respectively and enteroinvasive E. coli VG ipaH was not detected. In addition, the VGs papC for uropathogenic and ibeA for neonatal meningitis were frequently detected in 19% and 3% of isolates respectively. While all the isolates tested were susceptible to imipenem, meropenem, amikacin and gatilofloxacin, others were variously susceptible, and resistant as follows; ciprofloxacin (96%), kanamycin (95%), neomycin (92%), streptomycin (84%), chloramphenicol (73%), nalidixic acid (66%), nitrofuratoin (64%), gentamycin (63%), doxycycline (58%), cefepime (57%), tetracycline (49%) and cephalothin (42%). Conversely, all the isolates were resistant to sulphamethoxazole, and high levels of resistance were equally observed against amoxycillin (59%), ampicilin (57%) and cefuroxime (40%). Cefepime, cephalothin, cefuroxime, nalidixic acid, nitrofuratoin, chloramphenicol and tetracycline were not significantly different in their effect against the isolates from all locations (P > 0.05), whereas the resistance profile of the isolates against gentamycin, ciprofloxacin, sulphamethoxazole, ampicillin and amoxicillin were significantly different (P < 0.05). Amikacin, kanamycin, streptomycin, meropenem, imipenem and gatilofloxacin were statistically excluded from the analysis since all tested isolates showed total susceptibility to these antimicrobials. The multiple antibiotic resistance indexing ranged from 0.50 to 0.80 for all the sampling locations and exceeded the threshold value of 0.2. Prevalence and distributions of the 19 resistance determinants assessed were obtained as follows; [sulfonamides (sulI (8%), sulII (41%)], [beta-lactams; (ampC 22%; blaTEM, (21%), blaZ (18%),], [tetracyclines (tetA (24%), tetB (23%), tetC (18%), tetD (78%), tetK (15%), tetM, (10%)], [phenicols; (catI (37%), catII (28%), cmlA1 (19%)] and [aminoglycosides; (aacC2 (8%), aphA1 (80%), aphA2 (80%), aadA (79%) and strA (38%)]. The Pearson chi square exact test revealed many strong significant associations among ampC, blaTEM, blaZ and tetA genes with some determinants screened. In the same vein, a grand total of 366 resistance gene fingerprints were spotted across the sampling locations and among the resistant pathotypes, the modal prevalent gene prints were found among the ETEC strains in 148 (40%), being the predominant pathotype observed, followed by UPEC strains 80 (22%) while the lowest was the least occurring EAEC pathotype 14 (4%). While some physicochemical parameters exceeded prescribed standards for drinking water, some fell within. The total coliforms obtained in all the sampling sites were above the acceptable limits. Findings reveal the presence of diarrhoeagenic and non-diarrhoeagenic E. coli in the selected rivers and suggest a potential public health risk as the rivers are important resources for domestic, recreational and livelihood usage by their host communities. The multiple drug resistance indexing signifies isolates and pathotypes of high antimicrobial usage origin. An increase in the antimicrobial resistance signatures towards conventionally used antibiotics as observed in this study necessitates for safe water supply, adequate sanitation facilities and proper surveillance programs towards the monitoring of antimicrobial resistance determinants in water-bodies. Generally, results from this study indicate that the river waters are not suitable for consumption, domestic or recreational use and re-echo the importance of safeguarding the freshwater resources of Southwestern Nigeria.

Bush encroachment in the semi-arid communal grazing lands of Eastern Cape and farmer's perception of causes and livelihood impacts

- Description: The study was conducted in two communal rangelands of the Eastern Cape Province (Thorn and Mixed bush veld). The objective of the study was to investigate 1) the historical trends of bush encroachment, causes and impacts based on indigenous knowledge and perceptions of the communal people in two savannah vegetation types and 2) to determine the density and cover of woody species in the grazing areas. In each veld type, two villages were selected purposefully. A total of 120 households who own livestock were randomly selected from the four communal areas and a total of 48 elders were selected to provide information on bush encroachment and rangeland condition. For woody density and cover, grazing areas surrounding two homesteads (settlements) were selected from each communal rangeland, Two long transects radiating in the opposite direction from each homestead were established. The woody vegetation survey was conducted at 100, 200, 300, 400, 500, 600, 700, 800 and 900m distance from each homestead. In each distance, 3 (10 m x 10 m) quadrates were laid to record individual woody plant species, the height, basal stem and canopy diameters. Woody species were classified into seedlings (>0 - 1 m), saplings (>1 - 1.5 m), young shrubs (>1.5 - 2.0 m), mature shrubs (>2 - 3.0 m) and mature trees (>3.0 m). The average household size in of goat was higher than that of other livestock species. A total of 40 woody plant species were identified in all study areas. Acacia karoo, Coddia rudis and Scutia myrtina were the most dominant woody species in all study areas. Most of the woody plant species had the highest abundance in the height class >0 - 1 m. The average total woody plant density in Thorn bush and Mixed bush were 3461 and 2416 number ha-1, respectively. In conclusion both study areas were highly encroached and it was also perceived by elder from both areas that bush encroachment is a problem in the rangelands.

Characterization of some virulence and antibiotic resistance genes of Staphylococcus aureus isolated from cases of Bovine Mastitis in Nkonkobe Municipality, Eastern Cape Province, RSA

- Description: Staphylococcus aureus is one of the predominant causative agents of mastitis disease in dairy herds. Mastitis disease has a negative impact in the economic losses in the dairy sector across the globe. The aim of this study is to detect some of the virulence genes in the S. aureus isolated from 400 milk samples of subclinical and clinical mastitis dairy cows in Fort Hare dairy farm and Middle Drift dairy farm in Alice in the Eastern Cape province of South Africa. In addition antibiotic resistance pattern and antibiotic resistance genes were investigated. Gram-staining, oxidase test, catalase test and API Staph kit were preliminary biochemical tests used for the identification of S. aureus isolates. The MALDI-TOF-MS was also used for further identification. Polymerase chain reaction was performed of genes encoding antibiotic resistance as well as clumping (clfA), coagulase (coa) gene, toxic shock syndrome (tsst), exfoliative toxin A and B (eta and etb), and the gene segment encoding the immunoglobulin G binding region and X region of protein gene spa. A total of 20 (5%) S. aureus strains obtained from 400 milk samples from the two farms were subjected to 16 antibiotics for antibiotic susceptibility testing. In Middle Drift dairy farm 11 (5.5%) isolates were obtained from 200 samples and 9 (4.5%) isolates were obtained in Fort Hare dairy farm from 200 samples. A large percent of the isolates were resistant to penicillin G (60%), followed by trimethoprim (60%) and tetracycline (60%), trimethoprim-sulfamethaxazole (55%), telithroprim (55%) and doxycycline (45%). Most of the isolates were sensitive to several (50-85%) antibiotics. Of the twenty isolates tested 12 samples contained the penicillin antibiotic resistance gene (blaZ gene), 8 samples contained at least one aminoglycoside-modifying enzyme gene (AME gene); the (aac(6’)/aph(2’’) gene and no amplification occurred for aph(3’)-IIIa and ant(4’)-Ia) genes. In the case of the tetracycline antibiotic resistance gene (tetK and tetM), 2 samples contained tetM and a single sample contained tetK gene. No amplification was observed for the erythromycin antibiotic resistance genes (ermA, ermB, ermC, Mef and msrA). All the samples tested were negative for the expression of toxic syndrome gene (tsst), etb, and Immunoglobulin G binding region. However, amplification of the clumping factor was observed in 7 (35%) isolates of S. aureus, exfoliative toxin (eta) expressed 4(20%) isolates; coagulase gene (coa) yielded six DNA bands of six differences sizes from 16 (80%) isolates. A total of four different bands size were expressed for the spa X region from 12 (60%) isolates. The data obtained in this study suggests that poor hygienic practices and inadequate management practices are responsible for the increase in Staphylococcus aureus isolation. The high resistance of S. aureus to antibiotics and the distribution of virulence genes contribute in bovine mastitis in these farms may cause health problems in the community consuming raw milk purchased from these farms.

Chemical constituents and biological studies of Tagetes minuta L. and Rauvolfia caffra Sond

- Description: Consequently, specific parts or the whole plant can be used for various treatments. The aim of this study was to extract, isolate and characterize the biologically active volatile and non-volatile compounds from Tagetes minuta and Rauvolfia caffra respectively. Tagetes minuta plant was considered for extraction of essential oils using hydrodistillation method. Cis-β-ocimene (38.03%), caryophyllene oxide (18.04%), alloocimene (25.35 %), isopropyl tetradecanoate (17.02 %), cis-ocimene (38.14%) and trans-β-ocimene (37.03%) were the major components of essential oil analyzed from fresh stem, dried stem, fresh leaf, dried leaf, fresh flower and dried flower respectively. The volatile compounds were identified by Gas Chromatography-Mass Spectrometry (GC-MS) and Gas Chromatography (GC). Essential oil of dried stem leaf and flower parts of T. minuta exhibit antioxidant activity as demonstrated by the DPPH and FRAP bioassays. Crude extracts were extracted from R. caffra stem bark using sequence of solvents namely n-hexane, dichloromethane, ethyl acetate, methanol and ethanol. A white powder, a β-sitosterol (non-volatile compound) was isolated by column chromatography from ethyl acetate fractions of R. caffra stem bark and identified on various spectroscopic techniques such as FTIR and (1D and 2D) NMR. Melting point was also determined to be a sharp 129-130 ºC. DCM, EA, MetOH and EtOH fractions as well as β-Sitosterol (compound SM/01), showed antioxidant activity when tested on DPPH, FRAP, total phenolic and flavonoid bioassays. This antioxidant activity might be due to the presence of hydroxyl groups in the compound and crude fractions. On the other hand, T. minuta’s essential oil showed high antioxidant activity when evaluated on the DPPH and FRAP bioassays, which can be attributed to the presence of oxygenated monoterpenes and sesquiterpenes known to act as free radical scavenging and reducing agents. The use of R. caffra stem bark extracts against hypertension and other diseases by traditional healers could be attributed to the presence of phytochemicals (polyphenols and flavonoids) with known health benefits. Thus, it is recommended that the plant should be exploited further using modern techniques involving separation and purification of compounds that can be used for drug formulation. This study supports the use of T. minuta and R. caffra as the potential natural antioxidant source to manage various diseases including hypertension.

- Description: Cooperatives have principles and goals that regulate their day-to-day operations. Their aim is to create jobs through carrying out income generating projects. This aim is usually difficult to achieve as ‘human’ factors sometimes obscure the projects’ progress. This study therefore explored the nature of relationship that exists between the University of Fort Hare (UFH) and the cooperative members involved in the project, which in this study are referred to as, Ilima Agripark Project (IAP) members. This relationship was investigated through the three particular forms (information, consultation and participation) of community engagement, their nature, meanings, the influence they possess and how IAP/UFH members comprehend these. Guided by her research matrix and interview guide, the researcher used the qualitative and interpretive approach method to collect data. The case study made use of document analysis, key informant interviews, direct observations, face to face interviews and focus group discussions to collect data. In order to validate her data, she also made use of a tape recorder and notebook. The study’s key finding is that the three particular community engagement forms are not fully utilised by the university. The study revealed that there is lack of transparency when it comes to information dissemination hence, the IAP members feel insecure and socially excluded.

- Description: Beef constitutes an important part of many consumers’ diets. Beef is the most consumed red meat in South Africa. Consumer perceptions can affect the whole supply or value chain of beef. Recently there has been increased interest in food safety; greater concern for environmental and animal welfare issues, increased importance of eating quality and healthy food as well as the greater role of food services. The demand for beef is no longer limited to economic factors alone but to non-economic factors as well. Consumers of beef in Alice have become more concerned about meat-borne risks and personal health. This research prioritized the exploration of the beef consumers’ (i) self-perception (ii) price perception (iii) benefit perception [in the form of – (a) value perception and (b) quality perception]. The study was conducted in the Eastern Cape Province specifically in Alice town and the surrounding rural areas (Ntselamanzi, Gaga, Hillcrest, Gqumashe, and Dyamala) of the Nkonkobe municipality. Anyone who admitted to eating beef and at the point of data collection bought beef for their own consumption or family consumption was deemed as a unit of analysis. Non – probabilistic sampling techniques which included accidental and random sampling were used to collect data from 100 interviewees. A semi structured questionnaire was used to analyse the perceptions and attitudes of beef consumers. The respondents were asked to indicate their agreement or disagreement with 47 statements or items presented on either five alternatives in a Likert scale scored from 1 (strongly disagree) to 5 (strongly agree) or a six Likert scale ranging from 1 (very low) through 5 (very high) to 6 (I do not know).The data was analysed using SPSS. It was discovered that beef consumers did not consider either intrinsic or extrinsic cues in isolation when purchasing beef but rather all characteristics contribute to the final perception. The Alice consumer market is heterogeneous and consists of different races with different cultures and market segments with varying needs and preferences. The results showed that five factors were extracted from forty seven items Factor 1: Information reliance and quality indication; Factor 2: convenience; Factor 3: Traceability and animal Welfare; Factor 4: Health and safety Conscious and Factor 5: Price and Branding. Furthermore, four main consumer segments emerged as - Segment 1: Informed buyers; Segment 2: Elite buyers; Segment 3: Health & safety conscious buyers and Segment 4: Apathetic buyers. The cluster analysis shows that quality is a subjective concept that is informed by a consumer’s personal taste and preferences. Taste and preferences inform the consumer’s effective demand which in retrospect is informed by the consumer’s socioeconomic status.

Creating a safe learning environment in two schools in the Butterworth District, Eastern Cape

- Description: This study focuses on the creation of a safe learning environment at two schools in the Butterworth District of the Eastern Cape in South Africa. The two schools are Ndabankulu Senior Secondary School and Butterworth High School. The study is of a comparative design and aims to assist the communities of Ndabankulu Senior Secondary School, and Butterworth High School to improve health, safety and security issues of their children by reducing their exposure to environmental risks while attending school. An ethnographic study was used to collect data and the study employed questionnaires, document analysis and interviews as methods of collecting data. The primary aim of this study was to assess whether the school is still a safe place for learning and teaching to take place without fear and harm so that both learners and teachers can attain growth and development. Physical risks were assessed through observation and document analysis. Interviews were conducted with the senior members of staff while questionnaires were issued to eighty learners and were all returned back for analysis. The content analysis and interpretation yielded findings that suggests that hazards that may harm the development of learners at Ndabankulu S.S.S. and Butterworth High school exist. The study concludes that it is crucial that strategies and necessary interventions be set up and implemented to ensure that all stakeholders perceive school safety as an important aspect, both for the benefit of the school and that of the wider society. The barriers to school safety, specifically to both schools in this study need to be attended to. The study has presented findings that are consistent with those of other studies which emphasized that if barriers to school safety are identified, confronted and tackled, the schools could benefit positively from effects of involving everyone to create a safe learning environment. The conditions under which learners receive education in South Africa have to be closely and constantly monitored by Departmental officials. Teachers and managers need to be trained in their duties so that schools become places where learners are safe and enjoy reaching their future goals.

Determination and validation of medicinal plants used by farmers to control internal and external parasites in goats in the Eastern Cape Province, South Africa

- Description: The broad objective of the study was to determine and validate medicinal plants used by resource-limited farmers to control internal and external parasites in goats in the Eastern Cape Province, South Africa. A survey was conducted among 50 farmers and three herbalists to determine medicinal plants used to control parasites in goats. The survey revealed nine plant species belonging to eight families that were used. Among the identified plant species, Aloe ferox, Acokanthera oppositifolia and Elephantorrhiza elephantina were the plants having the highest Fidelity Level for their use, each scored 100.00 percent, followed by Albuca setosa (83.33 percent). These plants were then selected for validation studies. Gas-Chromatography-Mass-Spectrometry (GC-MS) revealed 7, 33, 26 and 32 bioactive phytochemicals in A. ferox, E. elephantina, A. oppositifolia and A. setosa, respectively. Terpenes and fatty acids were present, oxygenated terpenes being the most abundant hydrocarbons present in all the four plant species. The effect of acetone, methanol and ethanol extracts of leaves of Aloe ferox and Acokanthera oppositifolia on tick repellency and acaricidal activity were investigated on blood engorged Ambylomma hebraeum and Rhipicephalus decoloratus ticks at concentration 15, 30 and 50 percent. The 30 and 50 percent acetone extract of A. ferox and Dazzel dip had the highest acaricidal properties of 100 percent. The 50 percent methanol extract of A. oppositifolia and 50 percent acetone extract of A. ferox had the highest repellency activity of 89 percent and 85.33 percent, respectively. Results from this study revealed that the efficacy of medicinal plants used by farmers to control ticks vary with the type of solvent used for extracting the bioactive compounds. Furthermore, it revealed that Aloe ferox and A. opppositifolia plant extracts possess repellent and acaricidal activities. In a study to investigate the anthelminthic effect of crude extracts of Elephantorrhiza elephantina and Albuca setosa plants, significant anthelminthic effect on nematodes was observed in both plants. In this study, all E. elephantina and A. setosa extracts caused paralysis and mortality. Methanol was the most effective solvent in extracting bioactive compounds and methanol extract showed the best anthelminthic effects among the crude extracts investigated in both plants. The least time taken for the worms to be paralysed was 8.33 mins and 14.33mins in 100mg/ml methanol extracts of E. elephantina and A. setosa, resepectively. Methanol extract of E. elephantina and A. setosa (100mg/ml) had the highest anthelminthic activity and mortality was recorded after 18mins and 20mins, respectively. Results from this study revealed that these two plants possess anthelminthic activities. The study revealed that resource-limited farmers use medicinal plants to control internal and external parasites in goats. Gas-Chromatography-Mass-Spectrometry analysis showed that these plants contain bioactive compounds that have a potential in controlling parasites. Validation studies showed that A. ferox and A. oppositifolia possess repellent and acaricidal activities whereas A. setosa and E. elephantina possess anthelminthic activities.

- Description: The main aim of the study was to investigate the detriments associated with substance abuse among the out- of- school youth of Ntselamanzi Village in Alice. This study intended to achieve the following objectives. (i) to establish the causes of increased indulgence in substance abuse in Ntselamanzi village, Eastern Cape. (ii) to identify the various kinds of drugs abused by the society of Ntselamanzi (iii) to identify the consequences of substance abuse to the lives of the out-of-school youth. (iv) to suggest the strategies that can be used to reduce substance abuse in order to enhance the youth‟s socio-economic growth and societal bliss. Using the quantitative method ninety-eight questionnaires were administered to the out-of-school youth in Ntselamanzi village and eighty questionnaires were returned fully answered. The major findings of the study outlined that dominating factors of substance abuse in Ntselamanzi village were family drug use, increased drug accessibility, lack of recreation and peer influence. As a result drug abuse among the out of school youth has indicated that the youth end up dropping out of school, unemployed, facing health complications and committing anti-social activities in the village which causing family conflicts and rifts in the community. These all affected the out-of-school youths‟ well-being, academic future and their quality of life was lowered due to all related issues with drug abuse. It can be concluded that drug use among rural outof-school youth is detrimental to the out-of-school youths, their families destroyed and their community crippled from the anti-social activities that take place. The future of the youths‟ education is hampered; the quality of persons in the community and families is lowered. Development of high performance computing cluster for evaluation of sequence alignment algorithms

- Description: As the biological databases are increasing rapidly, there is a challenge for both Biologists and Computer Scientists to develop algorithms and databases to manage the increasing data. There are many algorithms developed to align the sequences stored in biological databases - some take time to process the data while others are inefficient to produce reasonable results. As more data is generated, and time consuming algorithms are developed to handle them, there is a need for specialized computers to handle the computations. Researchers are typically limited by the computational power of their computers. High Performance Computing (HPC) field addresses this challenge and can be used in a cost-effective manner where there is no need for expensive equipment, instead old computers can be used together to form a powerful system. This is the premise of this research, wherein the setup of a low-cost Beowulf cluster is explored, with the subsequent evaluation of its performance for processing sequent alignment algorithms. A mixed method methodology is used in this dissertation, which consists of literature study, theoretical and practise based system. This mixed method methodology also have a proof and concept where the Beowulf cluster is designed and implemented to perform the sequence alignment algorithms and also the performance test. This dissertation firstly gives an overview of sequence alignment algorithms that are already developed and also highlights their timeline. A presentation of the design and implementation of the Beowulf Cluster is highlighted and this is followed by the experiments on the baseline performance of the cluster. A detailed timeline of the sequence alignment algorithms is given and also the comparison between ClustalW-MPI and T-Coffee (Tree-based Consistency Objective Function For alignment Evaluation) algorithm is presented as part of the findings in the research study. The efficiency of the cluster was observed to be 19.8%, this percentage is unexpected because the predicted efficiency is 83.3%, which is found in the theoretical cluster calculator. The theoretical performance of the cluster showed a high performance as compared with the experimental performance, this is attributable to the slow network, which was 100Mbps, low processor speed of 2.50 GHz, and low memory of 2 Gigabytes.

- Description: Drought remains a huge challenge in Africa, particularly in Zimbabwe. The study investigated the effectiveness of drought management strategies and poverty alleviation in Zimbabwe with particular focus on Mwenezi District. In order to come up with a comprehensive investigation, the researcher grounded the study in sustainable livelihood approach and the community driven development theory. Due to drought being the number one disaster in Zimbabwe, the government and NGOs have collectively come up with strategies aimed at ameliorating the impact of drought in the area. The study employed a qualitative methodology as it provided an understanding and description of people’s personal experiences of drought. The study investigated the roles of both the government and NGOs involved in drought management in the district. NGOs and Government institutions charged with drought relief implement various programmes designed to manage drought in this area. This study’s findings were that cattle rearing, provision of food aid, provision of seed packs, supplementary feeding, among other programmes, are being implemented in the district to curb drought. Local people in the area have adopted a number of strategies so as to cope with drought effects. These strategies include growing of drought-resistant crops, livestock selling, change of farming methods, informal cross-border trading and establishing gardening plots. The Zimbabwean government has been criticised for being reactive rather than being proactive when it comes to drought management. The study further calls for improved and effective policies to tackle drought and poverty in the area. Effective monitoring and evaluation of programmes designed to manage drought should be prioritised if ever drought is to be effectively managed.

- Description: This study investigates Educators‟ and learners‟ perceptions on English first additional language speakers‟ use of English as medium of instruction in two primary schools in the East London District. The study made use of a qualitative research method which includes interviews, observation and document analysis (learners‟ books). The uncovered data is analyzed against the background of the socio-economic and historical factors that led to this medium of instruction using content analysis (CA). After the apartheid regime in 1994 and the formation of a new government in 1996, the South African Constitution declared 11 languages in an attempt to encourage multilingualism in the country with each province having its provincial language policy. In the Eastern Cape where the study is conducted the language policy recognizes Xhosa, English, and Afrikaans as the official languages with English as the medium of instruction especially at the intermediate phase. The theoretical and conceptual framework for the study is provided by studies on language policy and planning, English as an international language, and Language as a mediating tool. The findings of the study indicate the crucial role that language of instruction plays in teaching and learning. However, challenges arise when the language of instruction is not the mother tongue. The study indicates dissatisfaction in the use of EMOI as a result of inadequate proficiency in the language of instruction, school environment, lack of resources; and lack of parental support. As a result of the above, some educators resorted to code switching when teaching. Hence, the study sheds light on EFAL learner‟s perspective on both comprehension and the level of educator‟s English skills and whether there is a connection between these two aspects. - Description: The purpose of the study was to investigate the educators’ perceptions on the role of the South African Democratic Teachers’ Union (SADTU) in teacher professional development. The study also investigated the Union’s perceived challenges in contributing to teacher professionalism in South Africa, their perceived professional priorities and perceived expectations of teachers. The research genre used for this study was the interpretivism or constructivism paradigm. The qualitative methodology was used and case study employed as the research design of the study. The findings of this study were that SADTU were perceived as militant activists. Their activities were perceived as often very disruptive to education and their role appeared to be more politically focused than educational. They promoted and protected teacher labour rights and interests. However, they had no programme, nor the expertise nor the resources to respond to teachers’ professional development needs. The perceptions about them were that they had minimal or negligible contribution to teacher professional development. SADTU conducted training, but with nothing of value to teacher professional development. The unethical, unconstitutional and professionally non-developmental cadre deployment of SADTU leadership compromised the Union. Another finding was the important role of appropriate continuous professional development programmes in the improvement of the teachers’ own professional development, in the improvement of classroom practices and eventual enhanced learner performance. Inclusive to professional development are educationally good skills and values to colleagues, learners and parents both at school and out of school. Recommendations for SADTU were the: development of a clear programme on Teacher Professional Development; mobilisation of SADTU members towards restoring and enhancing the dignity of the teaching profession; engaging politically in curriculum policymaking to form inclusive professional associations; ending cadre deployment for senior management posts by SADTU leadership; establish development centres for critical maths and science, computers skills and other subjects and resuscitation of teacher colleges.

Effect of post-slaughter handling on physico-chemical and microbiological quality of red meat along the distribution chain in the Eastern Cape Province, South Africa

- Description: The broad objective of the study was to investigate the effect of post-slaughter handling in the distribution chain on red meat quality and safety. A survey was conducted among 300 consumers and 100 meat handlers in five different municipalities (Buffalo City, Nkonkobe, Ngqushwa, Lukhanje and Amahlathi) in the Eastern Cape Province of South Africa to investigate their perceptions on meat quality and safety, together with challenges faced by meat handlers during the distribution of meat from the abattoir to retailers. The microbiological profile and physico-chemical quality of red meat at different stages of the abattoir to retail outlets in the distribution chain were also determined. Swabs (n=216) and meat samples (n=450) were collected from beef, pork and mutton carcasses during the loading process of carcasses into trucks at the abattoir, when offloading carcasses at the supply points and during marketing. Physico-chemical qualities such as colour (L* - lightness, b* - redness and a* - yellowness) and meat pH measurements were taken at each point. To determine the microbiological profiles of the carcasses, four microbiological parameters were considered: Total bacteria count (general bacteria), coliform count (related to hygiene and indicator for pathogens), Escherichia coli (Gram-negative pathogen) and Staphylococcus aureus (Gram-positive pathogen). Two types of packaging (vacuum and overwrapping) were used to determine their effect on shelf-life and microbiological quality of red meat under the normal marketing conditions over a storage duration of 15 days. The results from the study showed low awareness of consumers about the pathogenic diseases which arise from meat. A strong significant association (p ˂ 0.05) between educational status and awareness on meat safety was observed. Most of the consumers perceived that quality goes beyond safety such that 35.6 percent of the respondents indicated that they did not have a problem with consuming spoiled meat, whilst the remaining 64.4 percent indicated that they would r eject spoiled meat. Although retailers indicated that they take meat safety into consideration in their shops, 92 percent of the retailers revealed that they do not perform microbial assessment of meat in their shops. A series of loading and off-loading, temperature fluctuations, environmental temperatures and ques during offloading were reported as the major challenges during transportation of carcasses from the abattoir to the supply points. The microbial counts were significantly (p<0.05) higher in samples from the commercial abattoir than in those from the communal abattoir. Escherichia coli was the predominant microbial contaminant in the samples from both abattoirs. When following the chain, total bacterial count (TBC), coliform count (CC) and the levels of E. coli contamination increased progressively between the loading and the off-loading points (5.1 to 7.9 log10 CFU/cm2; 5.0 to 5.6 log10 CFU/cm2 and 2.7 to 3.7 log10 CFU/cm2, respectively). The storage period, meat type, distance during transportation and temperature were found to have a significant impact on the microbial levels during the distribution of carcasses. Distribution stage had a significant effect (p<0.05) on some of the physico- chemical meat quality attributes and differences in the lightness (L*) and redness (a*) values between the loading, off-loading and display points were observed. Consumers perceived retailer class as one of the factors influencing meat quality, but according to the instrumental measurements retailer class did not have a significant effect on physico-chemical meat quality. However, distance and storage duration significantly (p<0.05) affected (L*) and (a*) values in the meat during distribution chain. Vacuum and overwrapping packaging significantly affected (p<0.05) the shelf life of meat. Therefore, it was concluded that post-slaughter handling during the distribution chain affects the physico- chemical, microbiological and shelf-life of meat.

Effect of Tenebrio molitor larvae as a protein source on growth performance, carcass yield and meat quality of broiler chickens

- Description: This study was conducted to assess the effect of including Tenebrio molitor larvae (T. molitor L) as a protein source in different diets on broiler performance, carcass characteristics and meat quality. A total of 144 day-old Cobb-500 chicks were randomly allocated into three treatment groups, each was allocated 16 birds and reared in 9 identical pens. Experimental diets used were as follows: T1 Control (no T. molitor L inclusion); T2 and T3 contained levels of T. molitor L at 5 percent and 10 percent of dry matter (DM) intake, respectively. Body weights (BW), average daily gain (ADG), feed intake (FI), and Feed conversion ratio (FCR) were measured for the 1st experimental chapter. For the second experimental chapter; live weights (LW) of broilers were recorded before slaughter and thereafter carcass weights (CW), meat yield (weights of breast, thigh, drumstick and wing) were recorded. The dressing percentage (DP percent) was also calculated. Breast muscles were sampled for meat pH and colour measurements. The LW of birds from T1 (0 percent T. molitor L) were significantly different (P<0.05) from both T2 (5 percent T. molitor L) and T3 (10 percent T. molitor L) which were similar to each other, with T2 exhibiting the highest live weights (2166g) and the control treatment exhibiting the lowest live weights (2018.3g). In CW, T1 was significantly different (P<0.05) from T2 while it was similar (P>0.05) to T3. The dressing percent of T1 was significantly different (P<0.05) from T2 and T3 which were similar to each other, with T2 having the highest dressing percentage (78, 2 percent) and T1 having the lowest DP percent (66 percent). The breasts in T2 were significantly higher and different (P<0.05) from both T1 and T3 which were similar to each other. The drumsticks in T3 were significantly different (P<0.05) from T1 while they were similar to T2 with values with T2 having highest values. After 45 minutes of slaughter, a significant difference (P<0.05) was observed in L⃰ among all treatments. In redness (a⃰), T3 was significantly different from T1 but was similar to T2 and all treatments in this study exhibited a darker red meat, with T3 muscles exhibiting darker red colour than the other treatments. Similar results were observed in yellowness (b⃰), where the breast muscles from T3 were more yellow than the other treatments. After 24 hours of slaughter, T2 L⃰ values were significantly different (P<0.05) from both T1 and T3 which were similar to each other. It was also found that the broiler chickens given diet with no T. molitor L inclusion (T1) had lower values of BW, FI, ADG and FCR throughout the experiment than those that were in T2 and T3 with 5 percent and 10 percent T. molitor L inclusion levels, respectively. However, it was also found that although broilers with 5 percent T. molitor L inclusion (T2) in their diet had high ABW and ADG than the broilers with 10 percent T. molitor L inclusion (T3), the T3 birds compared favourably to T2 birds as they required low feed intake to reach the same slaughter weight due to high FCR. It was, therefore, concluded that T. molitor L meal can be incorporated into the diets of broilers to produce heavy birds either at 5 or 10 percent. However, although 5 percent T. molitor L inclusion yields heavier carcasses, the 10 percent T. molitor L inclusion compared favourably to 5 percent inclusion since it required low feed intake to reach the same slaughter weights and there were slight differences on meat quality attributes between the two treatments.Thus T. molitor L at 10 percent inclusion levels was the best inclusion level to enhance broiler growth performance, carcass yield, meat yield and meat quality.

Effects of natural antioxidants and thermal treatment on quality of meat from Bonsmara and non-descripts cattle

- Description: The broad objective of this study was to determine the effects of natural antioxidants and thermal treatment on quality of meat from Bonsmara and non-descript cattle. A survey was conducted among 222 consumers in Alice (Fort Hare University community), Eastern Cape Province to determine the level of their awareness on application of antioxidants as preservatives in meat and meat products during storage. Representative samples of Muscularis longissimuss thoracis et lumborum and liver were collected from each carcass of Bonsmara (n=40) and non-descript (n=40) cattle reared on natural pasture to determine the effect of natural antioxidants and thermal treatment on their quality. The results from the survey revealed that 51.35 percent of the respondents had not heard about the use of antioxidant as preservatives in meat products. However, among the remaining respondents (48.65 percent) that were aware of antioxidant and its use as preservative, about 19 percent knew of natural antioxidants, 35 percent synthetic antioxidants while 46 percent had knowledge of both. The results further revealed that the majority of the respondents (82 percent) that had knowledge of natural antioxidants were concerned about the use of synthetic antioxidants in meat and meat products due to their health consequences. The in vitro antioxidant and antibacterial analyses of Bidens pilosa and Moringa oleifera leaf extracts revealed that they contain rich bioactive compounds. Furthermore, the addition of leaf extracts from Moringa oleifera (ML, 0.05 and 0.1 percent w/w) and Biden pilosa (BP, 0.05 and 0.1 percent w/w) to ground raw beef as a natural antioxidant were found to improve the physicochemical, oxidative stability and microbiological qualities of meat compared to the control (meat without extract) and BHT treatment (0.02 percent w/w) during 6 days storage at 40 C. Cattle breed did not have much effect on colour parameters. However, the overall pH of ground beef treated with extracts showed lower values than control and BHT treated beef. Also, ground beef samples containing extracts exhibited (P < 0.05) better colour stability, especially higher redness (a* values), than the control. The formation of TBARS in beef samples treated with extracts was significantly (P < 0.05) lower than the control and BHT treatment. The antibacterial assay of the extracts revealed an appreciable broad spectrum activity against the tested bacteria and microbial counts in ground beef samples compared to control and BHT group. Ground beef treated with plant extracts exhibited lower microbial and lactic acid bacterial counts (P < 0.05) at day 0 and 3 than control samples. Moreover, it was observed that application of sous vides thermal method did not significantly affect the fatty acids and mineral loss in beef and liver across the treatments. However, the total concentration of MUFA was lower in raw liver (20.11±2.38 - 21.08±1.23 percent) than in beef (40.22±1.90 - 42.53±1.29 percent), while total PUFA content were higher in liver (30.73±2.60 - 31.11±1.19 percent) than in beef samples (10.13±3.36 - 11.02±2.74 percent) (p < 0.05). The results also revealed that liver samples from Bonsmara and non-descript cattle had a higher percentage of intramuscular fat content of 4.67 ± 0.53 percent and 4.44 ± 0.53 percent respectively, and fat free dry matter of 27.51 ± 1.05 percent and 25.73 ± 1.05 percent, respectively, than the beef samples (p < 0.05). The concentrations of Mg (52.80±0.22 - 53.70±0.02mg/100g) and Zn (8.90±0.15 - 19.60±0.15mg/100g) were higher in beef than liver samples. The level of Ca (17.00±0.17 - 17.50 ± 0.17mg/100g) in liver was higher than that of beef samples. It was concluded that most consumers preferred the use of natural antioxidants in meat products than synthetic antioxidants due to their health risk. The effectiveness of M. oleifera and B. pilosa leaf extracts on meat quality also revealed that these plants are promising candidates as natural preservatives and their application should be considered in the meat industry. Lastly, findings from this study showed that application of the sous vide technique could protect meat products from nutritional loss during thermal treatment.

- Description: This study offers an explanation of enabling and / or constraining factors in Zimbabwe‘s 3-3-3 secondary teacher education model for Post ‗O‘ Level Science. It is a theory driven study that derives its theoretical foundation from Roy Bhaskar‘s critical realism and Margaret Archer‘s morphogenetic approach to reality. The study therefore offers explanations about structural, cultural and agential influences that facilitate and / or hinder the 3-year program for Post ‗O‘ Level Science. This was a qualitative case study of one secondary teacher education college in Zimbabwe. Qualitative data were collected through interviews, focus group discussions and document analysis. One official from the Department of Teacher Education(DTE) at the University of Zimbabwe(UZ),the Principal, the Vice Principal and 10 lecturers from the studied college,3 Heads of Science department in secondary schools,3mentors and 5 groups of 10 and 11eleven student teachers participated in the study. The DTE Handbook (2012), vision and mission statements and core values of the Ministry of Higher and Tertiary Education Science and Technology Development (MHTESTD),DTE and the studied college, syllabuses, teaching practice reports, policy documents, external examining reports, College Academic Board (CAB) minutes, admission records, mark profiles and pass lists among other relevant documents complemented interview data. As a theory driven study, structural, cultural and agential influences were found to be enabling and / or constraining the model. The acute shortage of Science teachers in secondary schools and the few Post ‗A‘ Level Science graduates led to the re-introduction of the 3- year Post ‗O‘ Level Science program in secondary teacher education colleges. The bureaucratic structures in educational institutions, the In-Out-In structure, institutional structures such as the family, the University, the studied college, secondary schools, infrastructural facilities, material and financial resources, transport facilities and utilities such as water, electricity and the internet were established as some among other structural factors affecting the 3-3-3 model. Discourses held about the teaching profession, the vision, mission and core values of the MHTESTD,DTE and the studied college, beliefs about what Science teachers should learn, knowledge, skills, attitudes and values they should acquire and how they should be taught were established as cultural factors enabling and / or constraining the 3-3-3 model. Agential influences offering causal explanation for enablers and / or constrainers of the model were established as the decision by the Principal and the CAB to re-introduce the 3- year Post ‗O‘ Level Science program, the decision by the students to enroll for the program, the recruitment of under qualified students, the use of various teaching methods, conducting staff development and mentorship workshops and failure to increase staff establishment. The study has put forth recommendations for the improvement on constraining factors in pre-service teacher education programs. With the understanding that agency has power to reinforce or transform structures and cultures, it should not be seen to be reinforcing disadvantaged structural positions and cultures; instead, after having identified structural and cultural constrainers, it should engage in communicative and meta-reflexivity to come up with the best possible solutions to the hindrances. Courses of action should then be taken accordingly.

Evaluating revenue collection and allocation challenges faced by Ndlambe Local Municipality (Eastern Cape Province)

- Description: In this project, the researcher evaluates revenue collection and allocation challenges faced by South African Municipalities in the case of NDLAMBE Local Municipality and this research project consist of five Chapters. Chapter one introduces and outlines the setting of the study and also provides the problem statement, research questions, research objectives and significance of the study. The chapter further clarifies the key concepts and terms that were used in the study, delimitation and limitations of the study. Chapter two provides a critical discussion of the concept of revenue collection and allocation, focusing on the various perspective of revenue. A source of municipal revenue and revenue collection and allocation model is explained as well as theoretical framework that guides it. Chapter three gives an account of the research design and methodology used in the study and in this study the researcher used the qualitative research approach as a data collection technique and document surveys were used because it is cheap and it saves time. The chapter also highlights the revenue collection and allocation applicable to the study and conclude by discussing the data analysis techniques that were used in the empirical study. Chapter four dealt with the data analysis of data collected presentation and discussion of the results and the researcher analyzed data using the filtering technique and content analysis. Chapter five drew conclusions and made scientific recommendations based on the research findings.The municipality is largely dependent on grants from upper spheres of government and generate less than 20 percent of current expenditures from own resources. As a whole, the revenue collection and allocation challenge of the NDLAMBE Local Municipality, given the current scenario of flows, is a reason for concern. Due to historical and political factors, local governments in South Africa differ substantially in terms of potential revenue base, but it may be that in many cases potential revenue is not exploited and that the high level of dependency on grants, high level of bad debts and lack of political will to be more self-reliant is the result of poor revenue in NDLAMBE Local Municipality.
