- Description: Investigation and identification of potential lipids for the manufacture of efavirenz loaded solid lipid nanoparticles (SLN) and nanostructured lipid carriers (NLC) was undertaken. Polymorphic modification and characteristics of the lipids with the best solubilising potential for efavirenz was explored using Fourier Transform Infrared Spectroscopy (FT-IR), Differential Scanning Calorimetry (DSC) and Wide-angle X-ray Scattering (WAXS). Lipid screening revealed that EFV is highly soluble in solid and liquid lipids, with glyceryl monostearate (GM) and Transcutol® HP (THP) exhibiting the best solubilising potential for EFV. GM exists in a stable β-polymorphic modification prior to exposure to heat, but exists in an α-polymorphic modification following exposure to heat. However, it was established that the addition of THP to GM revealed the co-existence of the α- and β'-polymorphic modifications of the lipid. EFV (60% w/w) exists in a crystalline state in a 70:30 mixture of GM and THP. Investigation of binary mixtures of EFV/GM and GM/THP, in addition to eutectic mixtures of EFV, GM and THP using FT-IR, DSC and WAXS revealed no potential interactions between EFV and the lipids selected for the production of the nanocarriers.

- Description: The South African forestry industry covers approximately 1.3 million hectares and is dependent on exotic pine and eucalypt species. Nursery seedlings are not inoculated with ectomycorrhizal (ECM) fungi. This investigation assessed levels of naturally occurring ECM colonisation of Pinus patula seedlings from 10 different South African forestry nurseries using a grid line intersect method. Fungi from colonised roots were identified using morphological characteristics and Illumina sequencing. Colonisation of seedlings in production nurseries was low (2-21%). Morphologically, the ECM fungi Thelophora terrestris, Suillus sibiricus, and the genera Russula and Pseudotomentella were identified. Molecularly, the ECM fungi T. terrestris, Inocybe jacobi and the genus Sphaerosporella, as well as several other ECM-containing families were identified, along with many saprotrophic/ endophytic fungi belonging to genera such as Penicillium, Ramasonia and Talaromyces. As can be seen, a combination of both molecular and morphological identification techniques are needed as neither is able to give a full picture of the species present in isolation. This study reveals an initial insight into the root microbiome community associated with Pinus patula seedlings, which should be taken into account when inoculation with beneficial microbes is considered. It determined that natural ECM fungal root colonisation levels are very low throughout the South African nurseries investigated, indicating the need for ECM fungal inoculation, which can increase seedling growth, viability and resistance to pathogens.

In the presence of triethylamine, (Z)-(2-acetoxyl-1-alkenyl) phenyl-λ3 iodanes reacts with thioureas or thioamides in methanol to afford 2,4- disubstituted thiazoles in good yields. The reaction is thought to proceed by the generation of highly reactive α-λ3 iodanyl ketones through ester exchange of the β-acetoxy group with liberation of methyl acetate, followed by nucleophilic substitutions with thioureas or thioamides [7].

This study quantitatively estimated the total phenolic (TPC), total flavonoid (TFC) and total proautocyanidin (TPA) contents of extract and fractions of B. pinnatum and correlated them with its antioxidant and anticholinesterase activities. Methanolic extract of the dried leaves was partitioned into n-hexane, ethyl acetate and aqueous fractions. Total phenolic, flavonoids and proanthocyanidins content were estimated as gallic acid or quercetin equivalents. DPPH and nitric oxide scavenging activity as well as ferric reducing power assays were used to evaluate antioxidant activity, using 2,6-di-tert-butyl4-methylphenol (DDM) and ascorbic acid as standards. In vitro anticholinesterase activity was evaluated by Ellmann’s colorimetry assay with phsiostigmine (serine) and donepezil as positive control. The crude methanol extract had the highest phenolic, flavonoid and tannin content. The ethyl acetate fraction had the highest DPPH radical scavenging effect (IC50 0.004 mg/ml) while the aqueous fraction had the highest NO scavenging and ferric reducing effects with values of IC50 0.012 mg/ml and 0.007 mg/ml respectively. The ethyl acetate fraction had the best cholinesterase inhibitory effect (IC50 0.951 µg/ml AChE; 3.546 µg/ml BuChE). DPPH radical scavenging effect correlated strongly with total phenolic, flavonoids and proautocyanidins (r2 0.896, 0.651 and 0.619 respectively) while ferric reducing power showed weak correlation and NO scavenging as well as AChE inhibition had no correlation. The study shows DPPH radical scavenging could be due to the phenolic content while other class of compounds are responsible for the cholinesterase inhibition.

Design of experiments was used to optimize response while evaluating input variables such as temperature, CLA-resin ratio,stirring time and pH. CLA loading efficiency was determined spectrophotometrically and CLA release using USP Apparatus II. Differential Scanning Calorimetry (DSC), Scanning Electron Microscop (SEM), Fourier Transform Infrared (FT-IR) Spectroscopy and X-ray Diffraction (XRD) were used to confirm complex formation. A spectrophotometric method was used to assess taste evaluation. The optimum CLA-resin ratio, temperature, and stirring time were 1:4, 80 °C, 3 hours, respectively, at pH 8. Characterization techniques revealed that CLA was crystalline and the complex amorphous in nature. FT-IR spectra of resinate revealed the absence of resonance due to the tertiary amine functional group that is responsible for the bitter taste of CLA. CLA was stable in simulated salivary fluid and was released within 3 hours in gastric fluid. All CLAresin batches revealed complete taste masking. Taste analysis highlighted the improvement of taste masking properties of the resinate as the CLA to resin ratio, increased.

- Description: Current global change scenarios predict an increase in atmospheric CO2 from the current 380 ppm to a value ranging from 540 ppm to 960 ppm by the year 2100. The effects of three air CO2 levels (400, 600 and 800 ppm) on five submerged aquatic plants that utilize HCO3− were studied, using the elevated CO2 Open Top Chamber facility at Rhodes University (Grahamstown, South Africa). Plants grew in water with two different initial dissolved inorganic carbon (DIC) concentrations of 1.5 and 3.0 mM. Overall, the growth rates and biomass allocation to roots were not affected by the initial DIC and air CO2, even though differences between the species were found. - Description: Current global change scenarios predict an increase in atmospheric CO2 from the current 380 ppm to a value ranging from 540 ppm to 960 ppm by the year 2100. The effects of three air CO2 levels (400, 600 and 800 ppm) on five submerged aquatic plants that utilize HCO3− were studied, using the elevated CO2 Open Top Chamber facility at Rhodes University (Grahamstown, South Africa). Plants grew in water with two different initial dissolved inorganic carbon (DIC) concentrations of 1.5 and 3.0 mM. Overall, the growth rates and biomass allocation to roots were not affected by the initial DIC and air CO2, even though differences between the species were found. Consequently, cold susceptibility of fifth instar C. peltastica and the most cold-tolerant larval stages (fourth and fifth instar) of false codling moth, Thaumatotibia leucotreta (Meyrick) (Lepidoptera: Tortricidae), were compared in artificial diet. There was no survival of C. peltastica after 13 d at 1°C, whereas this was only so for T. leucotreta after 16 d. Consequently, it can be concluded that any cold treatment that has been proven effective against T. leucotreta would be as effective against C. peltastica. Finally, it was confirmed that the cold susceptibility of T. leucotreta in artificial diet did not overestimate the effect of cold on T. leucotreta larvae in litchis.

Successful control is dependent upon integration of area-wide sterile insect releases and other suppression methods. The aim of this work was to test pyrethroid and organophosphate-based insecticides (tau-fluvalinate and chlorpyrifos) for their residual effect on mortality of released irradiated T. leucotreta male moths. Both of these insecticides were effective in killing irradiated T. leucotreta for 7 days after application on leaves, after which degradation of the active ingredient resulted in a marked reduction in efficacy after 14 days and rendering them harmless. Mortality was found to be similar for irradiated and non-irradiated male T. leucotreta after treatment. Consequently, even though these insecticides might have an effect on moths in the field, ratios of sterile:wild moths should not be altered. Supporting field data from six sites in the Sundays River Valley over a season of sterile insect releases showed the conventional chemical crop protection programme to be as effective as an integrated pest management programme in facilitating effective control of T. leucotreta through sterile insect releases. The study also confirmed that the ratios of sterile:wild male moths in the commercial citrus orchards were not affected by the application of insecticides. These findings confirm the high potential of sterile insect releases for control of T. leucotreta in citrus.

- Description: Semiconductor nanoparticles also known as quantum dots (QDs) have been used in diverse areas of application due to their good optical and photo-physical properties. The synthesis of core–shell QDs have assisted to improve the stability of most nano-particles but the size and long term stability of such materials have been problematic. However, thiol-capped CdTe QDs have been regarded as some of the most widely synthesized nanoparticles due to their unique optical properties. The use of un-stable tellurium source such as NaHTe or highly toxic H2Te gas makes the reaction condition cumbersome. In this study, we prepared some CdTe QDs and core–shell QDs using K2TeO3 as a stable source of tellurium without inert gas protection. This was done using a co-capped bio-compatible coating such as cysteine and glutathione following a modified standard method to produce QDs in the yields of about 60 to 80%. All the synthesized materials were subsequently characterized using various characterization techniques. The systematic optimization of the reaction parameters such as reaction time, pH and mole ratio results with a drastic red shift in wavelength (546–600 nm) by the CdTe core proved that an extra material has been deposited unto the surface of the CdTe core. The 60 days stability test conducted demonstrated that the core–shell nanoparticles were quite stable. Since this reaction was performed under open air conditions and no special ligand or buffer solution was used, it may suitably be applied on an industrial scale.

The use of un-stable tellurium source such as NaHTe or highly toxic H2Te gas makes the reaction condition cumbersome. In this study, we prepared some CdTe QDs and core–shell QDs using K2TeO3 as a stable source of tellurium without inert gas protection. This was done using a co-capped bio-compatible coating such as cysteine and glutathione following a modified standard method to produce QDs in the yields of about 60 to 80%. All the synthesized materials were subsequently characterized using various characterization techniques. The systematic optimization of the reaction parameters such as reaction time, pH and mole ratio results with a drastic red shift in wavelength (546–600 nm) by the CdTe core proved that an extra material has been deposited unto the surface of the CdTe core. The 60 days stability test conducted demonstrated that the core–shell nanoparticles were quite stable. Since this reaction was performed under open air conditions and no special ligand or buffer solution was used, it may suitably be applied on an industrial scale.

Furthermore, very few studies have focused on MetS in children and adolescents in sub-Saharan populations. This study, therefore, aimed at determining the prevalence of the MetS and contributing risk in adolescents living in Mthatha, South Africa. Research design and methods Four hundred and ten adolescents (age range: 13–18 years, both sexes) were recruited into this cross-sectional study. In addition to a lifestyle questionnaire being administered, anthropometric measurements (weight, height and waist circumferences) were carried out for each participant. Fasting venous blood was collected for lipid profile and blood glucose assessments. Blood pressure was also measured. MetS was assessed using appropriate diagnostic criteria for children and adolescents. Results Complete data sets were collected from 371 participants. Females showed an elevation in the risk factors for MetS. Of the 371 participants, 40.2% were overweight/obese (47.5% females; 22.5% males). MetS was diagnosed in 3.1% female participants compared with 6.0% in male participants. Conclusions More risk factors for the MetS are seen among the overweight/obese as compared to lean adolescents. Given the fact that childhood overweight/obesity often continues into adulthood, it is important to address the causes of increased risk for MetS earlier in life to prevent the development of disease in adult life.
