In vitro photodynamic effect of aluminum tetrasulfophthalocyanines on melanoma skin cancer and healthy normal skin cells

- Description: Photodynamic therapy is a medical treatment that uses an inactive dye/drug and lasers as a light source to activate the dye/drug to produce a toxic form of oxygen that destroys the cancer cells. This study aimed at investigating the cytotoxic effects of different concentrations of aluminum tetrasulfophthalocyanines in its inactive and active state (laser induced) on melanoma skin cancer cells, healthy normal skin fibroblast and keratinocyte cells. Experimentally, 3 x 10(4) cells/ml were seeded in 24-well plates before treatment with different concentrations of aluminum tetrasulfophthalocyanines. After 2 h, cells were irradiated with a light dose of 4.5 J/cm(2). Post-irradiated cells were incubated for 24 h before cell viability was measured using the CellTiter-Blue Viability Assay. Results showed that aluminum tetrasulfophthalocyanines at high concentrations were cytotoxic to melanoma cells in the absence of laser activation. In the presence of laser activation of aluminum tetrasulfophthalocyanines at a concentration of 40 mu g/ml decreased cell viability of melanoma cells to 45%, fibroblasts to 78% and keratinocytes to 73%. At this photosensitizing concentration of aluminum tetrasulfophthalocyanines the efficacy of the treatment light dose 4.5 J/cm(2) and the cell death mechanism induced by photoactivated aluminum tetrasulfophthalocyanines was evaluated. A light dose of 4.5 J/cm(2) was more efficient in killing a higher number of melanoma cells and a lower number of fibroblast and keratinocyte cells than the other light doses of 2.5 J/cm(2), 7.5 J/cm(2) and 10.5 J/cm(2). Apoptosis features such as blebbing, nucleus condensation, nucleus fragmentation and the formation of apoptotic bodies were seen in the photodynamic therapy treated melanoma skin cancer cells. This in vitro photodynamic therapy study concludes that using aluminum tetrasulfophthalocyanines at a photosensitizing concentration of 40 mu g/ml in combination with a laser dose of 4.5 J/cm(2) was potentially lethal for melanoma skin cancer cells and less harmful for the normal healthy skin cells. The photocatalytic activity of the LuTPPc in the fiber was compared to that of zinc phthalocyanine (ZnPc) incorporated into the PS fiber, and the former showed higher activity. UV–Vis spectral changes of sample solutions indicated transformation of the analyte with first order kinetics and half-lives that are within one and half hours for LuTPPc/PS. Products identified from the spectral changes and gas chromatography were benzoquinone, hydroquinone and 4,4′-dihydroxydiphenol suggesting that the photodegradation of 4-chlorophenol was through both Types I and II mechanisms.

The complexes showed high singlet oxygen quantum yield values ranging from 0.27 to 0.66 in the absence of folic acid. Numerous attempts have been made by its government to define and bolster a sense of nationalism and to create a sense of cohesion; however, a shadow side of this appeal for national identity has been the rise in xenophobic violence precipitated by the steady influx of refugees into the country. The title of this article is drawn from Albert Camus’s introduction to his disarming novella of dislocation, L’Etranger (1942), and I would like to explore some of the philosophical implications of representing strangers in different ways. Drawing on works by Zygmunt Bauman, Georg Simmel and Julia Kristeva, I will consider ambivalences towards the stranger represented in Magnet Theatre’s production (2010) of Die Vreemdeling [The Stranger], and pose a few questions about our relationship with the unknown. Attempts to familiarise the constituents of various communities with aspects of each other’s strangeness is a project which has typified much South African theatre in the past; and yet this is an approach which stands in sharp contrast to the importance granted processes of defamiliarisation first proposed so succinctly by Victor Shklovsky in 1917. Instead of attempts to harness and explain the unfamiliarity of others in order to communicate diversity, a celebration of the grandeur of the stranger may provide a more enriching alternative.

The UV-Visible spectrum showed a red shift in its Q-band maximum absorption within the fiber as compared to that in solution. The triplet quantum yield in DMF was determined to be 0.51 with a lifetime of 2.7 μs and a singlet oxygen quantum yield of 0.33 with a lifetime of 19.85 μs in the same solvent. The functionalized phthalocyanine fiber could be a promising fabric material for applications such as self-disinfecting in wound dressing. A method based on the conversion of ADMA was used to estimate the singlet oxygen quantum yield of the Pc in the hybrid fiber. - Description: Cadmium tellurite quantum dots (CdTe-QDs) are linked to nickel tetraamino phthalocyanine (CdTe-QDs-NiTAPc) through an amide bond. Differential pulse voltammetry shows that that NiTAPc stabilizes the QDs against oxidative disintegration into metallic products on oxidation. Electrocatalytic oxidation of 2, 4-dichlorophenol (DCP) and pentachlorophenol (PCP) on CdTe-QDs and CdTe-QDs-NiTAPc adsorbed or electrodeposited onto a gold electrode were studied. Adsorbed CdTe-QDs-NiTAPc shows the lowest potential for DCP and PCP oxidation and it is also more stable to fouling by PCP and its oxidation products compared to adsorbed CdTe-QDs without NiTAPc. Electrodeposited CdTe-QDs or CdTe-QDs-NiTAPc show the best activity in terms of enhanced currents towards the oxidation of the chlorophenols.

- Description: Cadmium tellurite quantum dots (CdTe-QDs) are linked to nickel tetraamino phthalocyanine (CdTe-QDs-NiTAPc) through an amide bond. Differential pulse voltammetry shows that that NiTAPc stabilizes the QDs against oxidative disintegration into metallic products on oxidation. Electrocatalytic oxidation of 2, 4-dichlorophenol (DCP) and pentachlorophenol (PCP) on CdTe-QDs and CdTe-QDs-NiTAPc adsorbed or electrodeposited onto a gold electrode were studied. Adsorbed CdTe-QDs-NiTAPc shows the lowest potential for DCP and PCP oxidation and it is also more stable to fouling by PCP and its oxidation products compared to adsorbed CdTe-QDs without NiTAPc. Electrodeposited CdTe-QDs or CdTe-QDs-NiTAPc show the best activity in terms of enhanced currents towards the oxidation of the chlorophenols.

- Description: The peripherally tetra substituted zinc phthalocyanine with 1,6-hexanedithiol as substituent (THdTZnPc, 3) was synthesized and is reported for the first time in this work. The potential of this zinc complex as a suitable photosensitizer for use in photodynamic therapy was determined through the investigation of the photophysical and photochemical properties. In this work complex 3 is attached to gold nanoparticles through the terminal thiol groups of the phthalocyanine resulting in a 3-AuNP conjugate whose photophysicochemical properties are investigated. Fluorescence lifetimes were determined using time correlated single photon counting and they show an increase in the abundance of the monomeric species (τ2) for the Pc in the 3-AuNP conjugate: ∼1 and 0.71 (with respective lifetimes 2.69 ns and 2.86 ns) compared to the free complex 3 with abundances of 0.12 and 0.13 (with respective lifetimes 3.36 ns and 3.28 ns) in DMSO and DMF, respectively.

The catalytic rate constants were 4.55 × 103 M−1 s−1 and 1.79 × 104 M−1 s−1 for amitrole and diuron, respectively using chronoamperometric studies. The diffusion constants were found to be 1.52 × 10−4 cm2 s−1 and 1.91 × 10−4 cm2 s−1 for diuron and amitrole, respectively. The linear concentration range for both were from 5.0 × 10−5 to 1.0 × 10−4 M and sensitivities of 0.6603 μA/μM and 0.6641 μA/μM for amitrole and diuron, with corresponding limits of detection of 2.15 × 10−7 and 2.6 × 10−7 M using the 3δ notation, respectively.

The presence of a long chain fluorine substitituent was found to result in reduced aggregation. The singlet oxygen, photodegradation, fluorescence quantum yield, triplet quantum yield and triplet life time of the complexes in toluene were determined. The effect of fluoro-functional groups on the photophysical and photochemical parameters of the zinc(II) phthalocyanines are also reported. Fluorescence quantum yields for the complexes ranged from 0.021 to 0.041.
