An investigation of experiences encountered by female adults living with HIV/AIDS and taking ARV treatment: the case of Ntselanzi location, Raymond Mhlaba municipality, Alice Eastern Cape

- Description: The study investigated the experiences encountered by female adults from Ntselamanzi living with Human Immunodeficiency Virus/ Acquired Immune Deficiency Syndrome (HIV/AIDS) and using Anti-retroviral (ARV) treatment. The study used a qualitative approach as well as a case study design that was descriptive explorative and explanative. The sample size or the unit of analysis consisted of 29 participants who included 12 female adults living with HIV/AIDS and having used ARVs for a minimum of three months; six nurses directly serving the people living with HIV/AIDS (PLWHA); six family members catering for the infected members within their family setting; and five key informants that included a social worker, a community leader, a health promoter, a community developer, and an HIV co-ordinator situated within the community of Ntselamanzi. A non-probability sampling method was adopted, specifically incorporating both snowball and purposive sampling techniques to select the sample. Significantly, the study was led by the following objectives: (1) To explore the experiences of female adults living with HIV/AIDS in Ntselamanzi, (2) To establish the coping mechanisms adopted by the PLWHA (3) To determine the effects of coping challenges and the adherence to ARVs. The data was collected by means of face-toface in-depth interviews in tandem with focus group discussion (FGD); and it was analysed qualitatively using content thematic analysis. The findings of the study revealed that all female adults living with HIV/AIDS and using ARVs encountered innumerable challenges that impaired their coping. The findings also indicated that malfunctioning and interruption of PLWHA coping was influenced by social factors such as unemployment, high level of poverty, low educational levels, lack of information sharing programmes and dependency on social grants. Lastly, the support to PLWHA especially by family members was highly indicated in the study’s findings. The following recommendations emanated from the study: strengthening research on coping challenges of female adults using ARVs in rural areas; empowering and training more caregivers for PLWHA by the Health Department; more involvement of social workers in helping the PLWHA; more involvement of government in the HIV/AIDS campaign in rural settings; and government facilitation and support to the PLWHA to form and belong to support groups.

Combined in silico approaches towards the identification of novel malarial cysteine protease inhibitors

- Description: Malaria an infectious disease caused by a group of parasitic organisms of the Plasmodium genus remains a severe public health problem in Africa, South America and parts of Asia. The leading causes for the persistence of malaria are the emergence of drug resistance to common antimalarial drugs, lack of effective vaccines and the inadequate control of mosquito vectors. Worryingly, accumulating evidence shows that the parasite has developed resistant to the current first-line treatment based on artemisinin. Hence, the identification and characterization of novel drug targets and drugs with unique mode of action remains an urgent priority. The successful sequencing and assembly of genomes from several Plasmodium species has opened an opportune window for the identification of new drug targets. Cysteine proteases are one of the major drug targets to be identified so far. The use of cysteine protease inhibitors coupled with gene manipulation studies has defined specific and putative roles of cysteine proteases which include hemoglobin degradation, erythrocyte rupture, immune evasion and erythrocyte invasion, steps which are central for the completion of the Plasmodium parasite life cycle. In an aim to discover potential novel antimalarials, this thesis focussed on falcipains (FPs), a group of four papain-like cysteine proteases from Plasmodium falciparum. Two of these enzymes, FP-2 and FP-3 are the major hemoglobinases and have been validated as drug targets. For the successful elimination of malaria, drugs must be safe and target both human and wild Plasmodium infective forms. Thus, an incipient aim was to identify protein homologs of these two proteases from other Plasmodium species and the host (human). From BLASTP analysis, up to 16 FP-2 and FP-3 homologs were identified (13 plasmodial proteases and 3 human cathepsins). Using in silico characterization approaches, the intra and inter group sequence, structural, phylogenetic and physicochemical differences were determined. To extend previous work (MSc student) involving docking studies on the identified proteins using known FP-2 and FP-3 inhibitors, a South African natural compound and its ZINC analogs, molecular dynamics and binding free energy studies were performed to determine the stabilities and quantification of the strength of interactions between the different protein-ligand complexes. From the results, key structural elements that regulate the binding and selectivity of non-peptidic compounds onto the different proteins were deciphered. Interaction fingerprints and energy decomposition analysis identified key residues and energetic terms that are central for effective ligand binding. This research presents novel insight essential for the structure-based molecular drug design of more potent antimalarial drugs.

Investigating how Grade 10 Physical Science teachers help learners to make sense of concepts of electromagnetism using easily accessible materials in under-resourced schools

- Description: This study sought to explore teachers’ perceptions and experiences of using easily accessible materials and how they might collaboratively develop learning and teaching support materials using easily accessible materials. The study explored three aspects. Firstly, the views, experiences and factors which influence grade 10 Physical Science teachers’ perceptions and experiences of teaching the topic of electromagnetism. Secondly, to find out what teachers can do to improve teaching and learning of electromagnetism in grade 10 Physical Science using easily accessible materials. Thirdly, to look at what enables or constrains grade 10 Physical Science teachers in under-resourced schools when dealing with electromagnetism, from using easily accessible materials. A qualitative method approach was adopted, underpinned by an interpretive paradigm but using some quantitative methods as well. Within the interpretive paradigm a case study approach was used. The study was carried out in Swakopmund and Gobabis education circuits of the Erongo and Omaheke Regions respectively. Data were collected using questionnaires, lesson observations and stimulated-recall interviews. The data obtained were validated in two ways, firstly, by triangulation from different data gathering techniques, and secondly, validation was done by member checking of the transcripts. To make meaning from the data generated, Vygotsky’s (1978) socio-cultural theory was used as a lens to analyse the data. The quantitative data generated were presented in figures, tables; whereas the qualitative data were coded inductively into descriptive texts to make meaning. It was found that teachers’s perceptions and experiences are that the topic of electromagnetism is quite challenging. Teachers reported that their learners find it difficult to comprehend phenomena associated with electromagnetism, thus making it difficult for their learners to grasp concepts associated with it. Lack of resources to do practical activities in the topic of electromagnetism was found to be a major contributing factor to teaching and learning of this topic. The use of easily accessible materials among teachers was found to be very limited. Easily accessible locally sourced materials were found to have the potential to enable grade 10 Physical Science teachers to help learners to make sense of concepts of electromagnetism in under-resourced schools. In light of the above findings, the study recommends that science teachers should make use of easily accessible materials which can minimise their dependence on standard laboratory equipment which is unaffordable anyway by most schools.

Synthesis and photophysical studies of crown ether-bodipy dyes and the fabrication of bodipy embedded fluorescent nanofibers

This thesis first explores the acid catalysed condensation reaction to produce two structurally analogous meso-substituted BODIPY dyes based on cuminaldehyde and 4-dimethylaminobenzaldehdye. In order to enhance the rate of ISC and promote the generation of reactive oxygen species bromine atoms were then attached to the BODIPY 2,6-positions. These BODIPY dyes were then embedded in a polystyrene solution and electrospun into nanofibers. The resulting nanofibers were found to be highly fluorescent, but were no longer able to generate singlet oxygen. Ion-sensitive BODIPYs were prepared from the dibrominated BODIPY dyes by employing a modified Knoevenagel condensation reaction to form a styryl bond with 4’-formylbenzo-15-crown-5 at the 3,5-position of the BODIPY core. Changes in the morphology and position of the absorption and emission spectra of these crown ether-styryl BODIPY dyes were observed in the presence of sodium ions. These results imply that crown ether-substituted BODIPY dyes could function as ion sensors.
