Hegemon and handmaiden: a neo-gramscian approach to South Africa’s position in the global political economy

- Description: Despite its assumed leadership of and investment in the African Union (AU) and the Southern African Development Community (SADC), South Africa has not developed its own hegemonic project and is consequently unable to exert significant influence over the ideas of the two institutions. Rooted in Antonio Gramsci’s absolute historicism, which locates decisions and actions at the level of the state within specific contexts, Robert Cox’s Method of Historical Structures (MHS) offers a framework that enables an approach informed by Critical Theory to analysing and empirically understanding the underlying factors of this situation. Utilising the three elements of the MHS, namely, material capabilities, ideas and institutions, and applying them to the context of international relations, it was found that South Africa’s choices have been informed by the global context of the post-USSR global arena it entered in 1990. The triumph of the neoliberal world order led by the United States framed the context of South Africa’s engagement with Africa and the world, with its choices informed by a composite of neoliberally defined global social relations of production and ideas that had already elicited global consent. South Africa’s organisation of material capabilities and resources was thus limited within the parameters of a neoliberal world order, and the predominance of institutions that facilitated the penetration of neoliberalism, regionally and globally. The global hegemonic order relies on surrogates for its functioning and South Africa’s subsequent domestic adherence to the hegemonic neoliberal order has been transferred to SADC and, to a limited extent, to the AU through the policies and institutional projects that South Africa has championed since 1994. Nonetheless, in the face of persistent inequality and poverty, internal contestation to this adherence has emanated from trade unions and civil society organisations. It was found that the continued and intensifying domestic contestation iii reinforces the lack of a domestic hegemonic project that could have been projected externally to SADC and the AU, so that South Africa, as a dominant peripheral state in the global world order, displays more characteristics of handmaiden than hegemon.

High-intensity Interval Training utilising a standardised versus customised training programme among endurance trained male cyclists

- Description: The primary aim of this study was to compare the effect of a standardised versus customised high-intensity interval training programmeon performance indicators of endurance trained male cyclists utilising a standardised method of prescribing intervals in comparison to a customised method of prescribing intervals based on training feedback. The study was exploratory and quasi-experimental in nature and utilized a quantitative approach. A quasi-experimental, pretest-postestcontrol group study design was used. A total number of 22 participants that met the inclusion criteria were included in the study by means of purposive sampling. The participants were subdivided into an experimental group (n=11) and a control group (n=11)by means of random stratification in order to ensure performance equality between the two groups. The experimental group completed a High-Intensity Interval Training (HIIT) block of 28 days (four weeks) with three weeks of interval-based sessions and one recovery week ending off the block. The programmeutilised the same “anaerobic” type intervals (10 x 30 seconds at 170% PPO with 4 minutes 30 seconds recovery) as the control group, performed twice per week for three consecutive weeks. The Lamberts and Lambert submaximal cycling test (LSCT) was used to determine whether a participant in the experimental group could start his HIIT session on a particular day.Participants were notallowed to start their HIIT session if twoor more of the set criteria weremet.If the participants did not qualify to participate in a HIIT session they were required to perform a low intensity training session in the same venue. Differences between the pre-and post-test showed statistically non-significant improvements (p>0.05) from pre-to post-test for both the experimental and control groups in respect of the physiological variables measured (gas exchange threshold, respiratory compensation point, maximal oxygen consumption). The experimental group did however show more positive, but statistically non-significantimprovements in peak power output. The findings of this study therefore showed that no significant within group nor between group improvements were achieved utilizing a standised versus customised HIIT training programme over the four-week study period. The findings do however suggest that the LSCT may have merit in prescribing HIIT sessions in trained athletesas suggested by the improvements of the experimental group’s peak power output.

- Description: This thesis critically examines the Salem commonage claim, a dispute that has shaken the hamlet of Salem to its core. On ground level it has caused racialized fault lines to reopen, while suspicion and distrust has also grown between the black Africans of the area as well. On a national level, the Constitutional Court judgement has potentially set a precedent with regards to its jurisprudential approach in determining the validity of land claims in South Africa. Its interpretation of the law was determined by the restorative justice jurisprudence enshrined in the Restitution of Land Rights Act 22 of 1994 (the Act). It based its own understanding of the history of the commonage on this jurisprudence. In a bold step towards realising the aims and purposes of the Act, the Constitutional Court found that both the black African claimants as well as the white landowners have equal rights to the land. One of the reasons why the decision of the Constitutional Court is ground-breaking is that the dispute involves a former commonage – land used for common purpose. The Constitutional Court emphasised that the Act was an “extraordinary piece of legislation” and had to be interpreted in such a way so as to address the injustices of the past. This included provisions of the Act which dealt with how oral testimonies from claimants would be dealt with. Another interesting feature was the heavy reliance by all parties on expert witnesses in the persons of eminent historians, Professors Martin Legassick and Herman Giliomee. This case gave much-needed clarification as to what the appropriate role of an expert historian witness may be in a land claim. The success or failure of land claims often depend on the weight of the evidence supplied by the expert historian witness. But the historian must also take cognisance of the fact that the evidence s/he gives is appropriate according to the scope of law. This case also dismisses the assumption that colonial instruments of land assignation are beyond reproach. These instruments which grant rights to land may also be scrutinised in a court of law, just like when oral testimony is tested for its credibility. This is important to note, especially when balancing land rights of the claimants against those of the landowners. This thesis agrees with the decision taken by the Constitutional Court in this instance. However, it also cautions that such softly-softly approaches may appear as a suitable compromise on paper, but the feeling on the ground may not be as receptive to reconciliation as what the courts would have hoped for. To the jurist, this judgement accurately encapsulates the purpose and aims of the Act. However, such a judgement may not seem satisfactory to the people of Salem. The decisions of the Salem commonage case are sure to inform the discourse of land claims in South Africa.

Interrogating the past of colonization of African narratives I could point that this is the case because African expression had always packaged in a ‘come see the Africans are dancing, singing or storytelling. Listen to their clicks.’ Writing, however, could only be executed by those Africans of white assimilation with higher social status and missionary education. Among amaXhosa, the disparity of socially lesser African arts and that of the educated has been termed the narrative of Amaqaba and Amagqobhoka. Amaqaba being those whose stories have taken longer to be documented in modern means of writing but have been enriched through years of live telling. Amagqobhoka on the other hand who easily documented their narrative after having been trained in writing have enjoined the audience of readers and access into literary space longer.

- Description: Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) is native to South America but has invaded the Afro-Eurasian supercontinent where it is currently the most devastating invasive arthropod pest of tomato. As a part of the first classical biological control programme against T. absoluta in Africa, a larval parasitoid, Dolichogenidea gelechiidivoris Marsh. (Syn.: Apanteles gelechiidivoris Marsh) (Hymenoptera: Braconidae), of T. absoluta was imported from Peru into the quarantine facility of the International Centre of Insect Physiology and Ecology, Kenya. We report on the host larval preference of D. gelechiidivoris and the host suitability, and the parasitoid’s reproductive strategy, including lifetime fecundity and egg maturation dynamics. Dolichogenidea gelechiidivoris females preferentially oviposited in early (1st and 2nd) larval instars of T. absoluta but parasitized and completed development in all four instars of the host. Host instar did not affect D. gelechiidivoris sex-ratio but females reared on the first instar had significantly fewer eggs than when reared in late larval instars (3rd and 4th). Females of the parasitoid emerged with a high mature egg load which peaked 2 d post eclosion. The females of D. gelechiidivoris survived 8.51 ± 0.65 d and produced 103 ± 8 offspring per female at 26 ± 4 °C (range: 24 to 29 °C) and 50–70% relative humidity (RH) with males present and fed honey-water (80% honey). Increasing maternal age decreased the proportion of female offspring. Under the aforementioned laboratory conditions, the Gross and Net reproductive rates were 72 and 39.5 respectively, while the mean generation time was 20 d. The potential intrinsic rate of natural increase was 0.18. This study shows that D. gelechiidivoris is a potential biological control agent of T. absoluta and should be considered for release in Kenya and across Africa following host specificity testing and risk assessments.

- Description: Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) is native to South America but has invaded the Afro-Eurasian supercontinent where it is currently the most devastating invasive arthropod pest of tomato. As a part of the first classical biological control programme against T. absoluta in Africa, a larval parasitoid, Dolichogenidea gelechiidivoris Marsh. (Syn.: Apanteles gelechiidivoris Marsh) (Hymenoptera: Braconidae), of T. absoluta was imported from Peru into the quarantine facility of the International Centre of Insect Physiology and Ecology, Kenya. We report on the host larval preference of D. gelechiidivoris and the host suitability, and the parasitoid’s reproductive strategy, including lifetime fecundity and egg maturation dynamics. Dolichogenidea gelechiidivoris females preferentially oviposited in early (1st and 2nd) larval instars of T. absoluta but parasitized and completed development in all four instars of the host. Host instar did not affect D. gelechiidivoris sex-ratio but females reared on the first instar had significantly fewer eggs than when reared in late larval instars (3rd and 4th). Females of the parasitoid emerged with a high mature egg load which peaked 2 d post eclosion. The females of D. gelechiidivoris survived 8.51 ± 0.65 d and produced 103 ± 8 offspring per female at 26 ± 4 °C (range: 24 to 29 °C) and 50–70% relative humidity (RH) with males present and fed honey-water (80% honey). Increasing maternal age decreased the proportion of female offspring. Under the aforementioned laboratory conditions, the Gross and Net reproductive rates were 72 and 39.5 respectively, while the mean generation time was 20 d. The potential intrinsic rate of natural increase was 0.18. This study shows that D. gelechiidivoris is a potential biological control agent of T. absoluta and should be considered for release in Kenya and across Africa following host specificity testing and risk assessments.

Household food insecurity and its association with psychological well-being of pregnant women in Ghana: the role of selected psychosocial factors

However, the impact of food insecurity on well-being has not received the needed attention by researchers in Sub-Saharan Africa where the state of food insecurity and its consequence is very pandemic. The objective of this study was to assess the impact of food insecurity on psychological well-being and its components among pregnant women in Northern Ghana and to assess the roles of self-esteem, locus of control, proactivity, and social support on the underlying relationship. The cross-sectional survey was adopted where 386 pregnant women were recruited to complete questionnaires on demographic, some psychological factors, psychological well-being and food insecurity. Dimensionality and Item analyses were conducted to detect complex and poor items and also to ensure unidimensionality. Regression analysis, MANOVA, Baron and Kenney Model of moderation and mediation, the multivariate ANOVA and the Two-way ANOVA were used to test the hypotheses. Results indicated that the measurement and the structural models showed a good fit with the observed data. Results showed that about 37.5% (n=144) out of the 384 pregnant women experienced severe food insecurity. Food insecurity was negatively significantly related with psychological well-being and all its components among pregnant women. Food insecurity elucidated 48.5% of the observed variance of food insecurity. Food insecurity correlated highly to personal growth (β = -.327) element of psychological well-being compared to autonomy (β = -.227), interpersonal relationship (β = -.207), environmental mastery (β = -.168), self-acceptance (β = -.117), and of purpose in life (β = -.107). Self-esteem and locus of control mediated the relationship between food insecurity and psychological well-being. Proactivity and social support also moderated the nexus between food insecurity and psychological well-being. 4 Final Submission of Thesis, Dissertation or Research Report/Project, Conference or Exam Paper Having higher income status and being employed were associated positively with higher psychological well-being. As an International Federation, SOS Children’s Village espouses four values, namely Courage, Trust, Commitment, and Accountability. The case was selected because SOS Children’s Village’s performance in Lesotho has been outstanding, due to employee commitment in the organization. The review of the literature drew upon three theories: the theory of organizational culture by Schein (2010), which shed a light on cultural embedding mechanisms, the relational leadership components of Komives, S, Lucas, N, and McMahon, T (1998), and the strategic leadership roles of Ireland and Hitt (1999). This study adopted a deductive qualitative research method, where structured interviews supplemented with documents were used to collect data. Thematic analysis and pattern matching analysed the data with the aid of a pre-developed codebook that identify theoretical codes and themes in the data. The findings confirmed that with the use of cultural embedding mechanisms, relational and strategic leadership shaped the culture of commitment. The main characteristics of the culture of the commitment of SOS Quthing are punctuality, teamwork, and individual initiative. Relational and strategic leadership behaviours helped to create a culture of commitment through the fair and equitable allocation of resources, the inclusion of employees in decision making, having an orientation program for new employees, giving rewards and recognition, holding regular meetings, supervision, utilizing the online collaboration system and the systematic development of human capital. The literature supported the study results. The study acknowledges the limitations and delimitations of the research. The study’s contribution was in identifying how leadership behaviours operate through cultural embedding mechanisms to "identify" creates a culture of commitment. In conclusion, managerial recommendations were made for SOS to strengthen their leadership behaviours further and further research recommendations.

How to do things with speeches: a critical discourse analysis of military coup texts in Nigeria

- Description: Coup speeches that usher the military into political power in Nigeria are the central focus of this thesis. There are seven coup speeches that are notable in the changing of the political course in Nigeria and in enabling the military to rule Nigeria for 30 years, establishing another alternative political construct and party (Bangura 1991). The seven coup speeches along with two others, one a colonial proclamation of conquest and the other a counter coup speech (altogether making nine) constitute the data of this thesis. The analysis done here uses Critical Discourse Analysis, based on a combination of Fairclough (1989, 2001), Fairclough and Fairclough (2012), Thompson’s (1984, 1988 and 1990) works with complementary insights by Chilton (2004), to analyze the speeches in order to understand the ideologies, perceptions and arguments of the coup makers enshrined in the texts. I also employ a concordance analytic system in corpus linguistics to sort uses of important terms and lexical items. The analysis is divided into three broad parts, namely: an analysis of representation of social actors and their action, an analysis of the processes of interpellation and then an analysis of the premises of the arguments contained in the speeches. In the concluding part, there is a discussion of the dialectical nature of the coup speeches especially in the areas of mutual influences which aids in the gradual sedimentation of the political ideology of the military. In particular, there is a longitudinal intertextual analysis across all the speeches, from the earliest to the latest, to see how a coup speech genre is created. The contribution of this work to knowledge is in terms of combining discourse analysis and social theory to illuminate some aspects of Nigeria’s socio- political crises in depth and multifariously. This work helps in understanding the nature of Nigerian autocratic democracy, subservient followership by the citizenry and the supremacy of the military elite. The work employs a novel combination of representation, argumentation, interpellation and constitutive intertextuality in understanding military discourse. It looks at speaker intention, the exploitation of interpretation or reception and the formation of subjects in general and each with its importance and social context. The work as a whole reveals that the military try to build legitimacy by way of establishing authority through rhetorical arguments in varying degrees. These arguments are laid bare, and what they discern is that charges are decidedly trumped up by the military against their opponents and constructed to suit the spin of their moments. The coup makers in some instances construct strawmen of opponents and then go ahead to attack their constructed assumptions or they charge without substance using nominalizations, metaphorical constructions and presuppositions. They apply stipulative definitions and emotionally loaded words in evaluating their actions favourably and also in the negative evaluation of the actions of the opponents. At the level of interpellational analysis, the data reveals the use of language in gradually hailing the citizens as military subjects. The role of the audience changes here i.e. from those to be convinced in rhetorical evaluation of opponents to those to be firmly controlled. The persistent hailing and positioning of the citizens as military subjects help in concretizing their subjecthood. The reaction of the people in affirmation of support to the rule of the military is crucial and it completes the interpellation process. As observed by Clark (2007, 141) “many African societies are so inured to military intervention as not to regard it as aberrant”. This inuring of the societies has to do with hegemonic ideological practices in military discourses claiming legitimacy and the right to rule. At the reception level, this shows that most of the citizens have bought into the dominant ideology and are as such interpellated by it or have adopted what Hall (2015, 125) would call the ‘dominant-hegemonic position’. Aspects of argumentation, speech acts, and deontic modals used by the coup makers help in gradually solidifying the subservient nature of the citizens to the military junta. The diachronic and intertextual nature of the analysis also reveals that the colonial proclamation of conquest in Nigeria by Lord Fredrick Lugard possibly influenced the first coup speech in 1966 in terms of structure and genre. There are traces of the colonial proclamations found in the 1966 coup speech. In substance, the military appear to copy their colonial progenitors. Historically, the military were formed as an army of colonial conquest. There is a dialectical interplay between colonial discourse and military coup speeches. The first coup speech, for its part, influences other coup speeches and they in general impact on civilian political language. The work analyzes from the minute to the global and in this bid unties the layers of assumptions, constructions and points of views that underpin an otherwise objective presentation of reality. The study also engages social theory in illuminating aspects of discourse, social practice and political action. The works of post-structuralists like Foucault, Althusser, Bourdieu, Habermas, Laclau and Mouffe, Derrida etc. are employed in shedding light on the processes of social formation in the interpellation of subjects and in the construction of a new political authority by the military regimes.

Here, we further characterized the HSP90 and FN interaction. FN bound to the M domain of HSP90 and interacted with both the open and closed HSP90 conformations; and the interaction was reduced in the presence of sodium molybdate. HSP90 interacted with the N-terminal regions of FN, which are known to be important for matrix assembly.

Human capital Development and Economic Growth: A cross country study

The primary focus of the study is on examining the contribution of human capital index components on economic growth for the period 2014 to 2017. The panel data model was employed in the study. Firstly, the hausman test was used to determine the suitable method between fixed and random effects. The fixed effects model was selected as the best panel econometric technique to be used in the study. The empirical results showed that there is a positive and statistically significant relationship between human capital index components (capacity, development, deployment and know how) and economic growth. The study also found that these components of human capital index have more impact on economic growth when they are interacted together based on theories of human capital and economic growth. Capacity and development are found to be the best contributors to economic growth for all the regions; thus, the study recommends that for economic growth to be increased, countries should invest more of their funds on capacity and development sub- indexes of human capital index. Exchange rates and inflation are found to have a negative relationship with the economic growth, while interest rates are found to have a positive and statistical relationship with the economic growth. The study also found out that there is a regional effect associated with human capital development, with European region being the best contributor to GDP. The components of human capital index also found to be important for all regions to invest on to improve economic performance. Deployment is found not to be significant is Sub-Saharan Africa. The existence of income group effect is found with developed countries performing much better compared to developing countries. The study also recommended that countries should invest more of their wealth in human capital development components such as education and health to enhance their economic growth

Hydrogeochemistry of shallow karoo basin aquifers in the border-kei region flanking the Indian Ocean, Eastern Cape, South Africa

- Description: As the potential for shale gas exploitation becomes a real possibility across parts of the Karoo in South Africa, and groundwater is one of the systems that could become heavily stressed if the shale gas industry becomes operational, it is important therefore to gain a better understanding into the hydrogeochemical systems and processes that occur within the Karoo Basin. Groundwater systems are very complex and are heavily relied upon in many areas across the Karoo that are surface water deficient. Groundwater is used domestically, for irrigation and livestock as well as in industry. A hydrogeochemical baseline investigation was undertaken in the shallow Karoo Basin aquifers (< 100 m) of the Border-Kei region with in the Eastern Cape of South Africa, which flanks the Indian Ocean. Since it has been suggested that seawater might be used as a potential fracking fluid, it is important to also establish baseline data linked to potential seawater salinisation in the coastal aquifers. This was achieved by sampling water from both 35 production and 3 non-production boreholes (by making use of a bailer and discrete interval sampler). Water from all 38 samples was analysed for major anions and cations, trace elements as well as for δ 18O and δ 2H stable isotopes. Results indicate that the majority of the groundwater in the study area falls within acceptable limits for domestic use, and although slightly saline, is exploitable for other uses as well. It was also found that dolerite dykes that have intruded into the Karoo Basin sediments have slight effects on the groundwater chemistry, but that these effects are also masked by the water-rock interactions occurring with the aquifer. It was also determined with major ions and stable isotopes that although the coastal aquifers do have saline signatures (EC > 200 mS/m) this is not due to salinisation via seawater intrusion. It is instead postulated that the salinity arises from marine aerosols that have introduced NaCl onto the soil layer, and which then leaches into the groundwater system through precipitation or irrigation. In the event of shale gas extraction through fracking or the expansion of rural and urban centres, this project serves as a preliminary hydrogeochemical baseline before any of these processes proceed within the Karoo Basin.

Ichthyofaunal and isotope changes along the rivere-stuarine continuum in a fluvially dominated Southern African coastal system

- Description: Catchment to coast and the continuum concept recognize linkages and make it imperative to take an ecosystem approach to catchment management, water allocation, fisheries management and environmental management as a whole. Fishes that utilise estuarine environments are influenced by physical and chemical conditions as well as biological interactions. Community structure, drivers and trophic linkages between species of different origin, utilising the low salinity waters along the riverestuarine continuum remain poorly understood in the lower Orange River and estuary. This body of work investigated the spatio-temporal dynamics structuring fishes in the lower Orange River Estuary Continuum (OREC) within the context of River Continuum (Vannote et al., 1980). Eight years (2004 - 2018) of seasonal sampling at 18 sites comprised of multi-species catches were analysed, including three years (2016 - 2018) exploring trophic linkages between the dominant species of marine, estuarine and freshwater fishes in the system. In total, 30 species belonging to 16 families were collected in the lower OREC. The Family Mugilidae dominated all catches. Fish species composition was comprised mostly of freshwater (47%), followed by the marine (33%) and estuarine species (20%). Numerically, the marine euryhaline Chelon richardsonii and solely estuarine Gilchristella aestuaria were the dominant species in the estuary region (site 1 - 11) with 95% and 4% contribution, respectively. Chelon richardsonii also dominated all catches (67%) upstream into the river (site 12 - 18), followed by freshwater Labeobarbus aeneus (13%) and Pseudocrenilabrus philander (8%). This highlights the point that besides the marine Chelon richardsonii, other species albeit marine, estuarine and freshwater are utilising unique areas within the OREC. Temporal differentiation in fish abundance (catch-per-unit-effort, CPUE), diversity, richness and evenness were higher in the high-flow season than the low-flow season. Mean CPUE was higher at the mouth of the estuary and declined upstream into the river, whilst species diversity (including richness and evenness) increased upstream. High catches but low species diversity observed close to the mouth of the estuary were attributed to the domination of Chelon richardsonii. However, species diversity increased upstream in the river due to the dominance of freshwater fishes coupled with several marine and estuarine species. This diversity patterns were inconsistent with the general Remane and revised model for estuaries, hence a new RiverEstuarine Continuum concept suitable for OREC was developed. With the exception of Chelon richardsonii, spatial variability in the length-frequency distribution of most fishes was observed. Physico-chemical drivers provided valuable information on adaptation limits of species movement and distribution. Fishes were structured along a salinity gradient and interaction between temperature and salinity influenced community structure and diversity patterns in the OREC. Fish species utilising the lower OREC are predominantly zoobenthivores but a few detritivores/planktivores dominate numerically. Trophic inferences from stable isotope analysis showed increased carbon isotope values (δ 13C) with salinity toward the mouth of the estuary, whilst nitrogen isotope values (δ 15N) increased upstream into the river. Ontogenetic shifts in the SI ratios of all five fish species (marine Chelon richardsonii; estuarine Gilchristella aestuaria; freshwater Labeobarbus aeneus, Pseudocrenilabrus philander and Mesobola brevianalis) were observed in the OREC. Variability in seasons and salinity gradient influenced the SI ratios of several fish species. Seasonal changes influenced the δ 13C ratios of all species, except for Labeobarbus aeneus and Pseudocrenilabrus philander whereas the δ 15N ratios of all species (except for Mesobola brevianalis) differed significantly between the high-flow and low-flow season. This can be due to their feeding on specific life-history stages, such as eggs, larvae, gametophytes, sporophytes or zoospores of prey. Stable isotope ratios revealed that Mesobola brevianalis fed at a higher trophic level than Chelon richardsonii and Gilchristella aestuaria. Mesobola brevianalis feed on planktonic crustacean and insects. Chelon richardsonii feed on a wide range of prey, with prey switching between planktonic and detritivore food resources, whilst Gilchristella aestuaria feed selectively on zooplankton but switched to filter feeding modes in turbid water. Generally, δ 15N SI ratios increase with growth, however, for Labeobarbus aeneus, it was significantly higher in small size length than in large size individuals, probably because most of the juveniles assessed were caught upstream where δ 15N increases. Juveniles of Labeobarbus aeneus mainly feed on zoobenthos whilst in larger fish phytoplankton becomes increasingly important, suggesting the possibility that zoobenthos was more 15N enriched than plant materials. The trophic niches of Mesobola brevianalis were isotopically distinguishable from Chelon richardsonii and Gilchristella aestuaria during the low-flow season, revealing that these species are probably accessing carbon sources from different food chains. During high-flow season, the isotopic niche overlapped among several species, indicating that these species are probably deriving carbon sources from the same or similar food chains. Results from Layman isotope-based metrics revealed that Chelon richardsonii, Pseudocrenilabrus philander and Labeobarbus aeneus occupied the widest isotopic niche in the lower OREC, suggesting that these species are utilising food sources which may be derived from a wide variety of carbon sources. Mesobola brevianalis had the narrowest isotopic niche, however this does not imply "a narrow range of food sources’’ for the species. Overall, the potential carbon sources for selected fish species revealed that they are supported by a variety of food sources (mainly phyto-and zooplanktonic) or their food items constituted different basal sources which enable them to coexist. This study represents the first, integrated investigation of trophic relations among fishes of different origin using δ 13C and δ15N SI analyses. The current study provides a broader understanding of fish ecology within the River-Estuarine Continuum in the lower Orange River and estuary. By filling research gaps identified in the lower OREC, this synthesis of work also supports and informs management, conservation and responsible utilisation of estuarine fishery resources, in particular within the OREC.

Identification of differentially expressed proteins involved in wheat (Triticum aestivum) resistance against Russian Wheat Aphid (Diuraphis noxia) SA2 using SWATH-MS analysis

- Description: Wheat (Triticum aestivum) is the second most-consumed cereal crop in the world, second only to maize. In South Africa it is mainly produced in three provinces; the Free State, Western Cape, and the Northen Cape. The average total land area used for wheat cultivation in these three Provinces, is 533 000 hectares, with a resultant annual production of between 1.3 to 2 million tons. The total wheat requirement in South Africa is currently 2.7 million tons according to the production of wheat guidelines published by the Department of Agriculture, Forestry and Fisheries, which is higher than the total annual production. One of the primary reasons that contribute to the lower production levels is insect and pest infestation, in particular, the Russian wheat aphid (Diurophis noxis), which can result in crop loss of up to sixty percent. Russian wheat aphids (RWA) are invasive insects that feed on the phloem of the plant through their specialised thin stylet-like mouthparts and release toxic agents contained in their saliva during feeding, which disrupt cellular functions inside the host plant. This damage leads to symptoms like; chlorosis, necrosis, wilting, stunting and curling of leaves, also known as leaf rolling. This study aimed to identify differentially expressed proteins in resistant and susceptible wheat (Triticum aestivum) cultivars during Russian wheat aphid Biotype South African 2 (RWA-SA2) infestation, in order to identify proteins involved in the wheat resistant mechanism against RWA-SA2. Two wheat cultivars SST398 (resistant to RWA-SA2) and SST356 (susceptible to RWA-SA2) were used, and a total number of 126 plants were planted and divided according to four different harvest times (Control day 0; day 5; day 7 and day 12). The wheat plants were infested with approximately 10 aphids per plant at the three-leaf stage, and placed within an insect cage inside a growth chamber (Conviron, Winnipeg, Canada) set at 24ºC, light intensity of 352 µmol.m-2.sec-1, ambient CO2 levels (410 ppm) and humidity of 60% with a 16h/8h day/night cycle. Wheat leaves were harvested at various time frames, and total protein extraction was performed. The protein samples were reduced with 10 mM dithiothreitol (DTT) and alkylated with 30 mM iodoacetamide (IAA). Sample clean-up and on-bead trypsin digest were performed on megReSyn Hilic columns, over four hours. The resultant peptides were vacuum dried and resuspended in 2% acetonitrile (ACN) before submitting them to LC-MS/MS (SWATH) for analysis. A total number of 611 proteins were differentially expressed, of which 19 were identified to be involved in the resistance response of wheat, and an additional 20 involved in biotic stress responses. This study showed that reactive oxidative species (ROS) such as hydrogen peroxide and hydroxyl radicals (OH-) play a significant role in the early stages of wheat resistance against RWA-SA2 infestation. They are involved in cell wall strengthening, activation of defence genes, involvement of phytohormone signals such as salicylic and jasmonic acid, which also mediates the systemic defences such as, systemic acquired resistance (SAR), leading to the expression of pathogen-related proteins such as (thaumatin-like proteins, oxalate oxidase, defensin, chitinase, and thionins). Although up-regulation of photosynthesis proteins such as (chlorophyll a/b binding protein, photosystem I and II, rubisco and divinyl chlorophyllide a, and 8 vinyl-reductase) were seen on both infested cultivars (susceptible and resistance), higher numbers of these classes of proteins were identified in the resistance cultivar, suggesting that they also play a critical role in resistance. In conclusion, the SWATH analysis used in this study was able to identify numerous proteins involved in the wheat resistance mechanism against RWA-SA2, most of which have not yet been reported to be involved in either biotic stress or RWA-SA2 resistance responses. Future studies are required to biochemically confirm the involvement of these enzymes and proteins the specific metabolic pathways where they are found, in the wheat resistance mechanism against Russian wheat aphid Biotype 2

- Description: Wheat (Triticum aestivum) is the second most-consumed cereal crop in the world, second only to maize. In South Africa it is mainly produced in three provinces; the Free State, Western Cape, and the Northen Cape. The average total land area used for wheat cultivation in these three Provinces, is 533 000 hectares, with a resultant annual production of between 1.3 to 2 million tons. The total wheat requirement in South Africa is currently 2.7 million tons according to the production of wheat guidelines published by the Department of Agriculture, Forestry and Fisheries, which is higher than the total annual production. One of the primary reasons that contribute to the lower production levels is insect and pest infestation, in particular, the Russian wheat aphid (Diurophis noxis), which can result in crop loss of up to sixty percent. Russian wheat aphids (RWA) are invasive insects that feed on the phloem of the plant through their specialised thin stylet-like mouthparts and release toxic agents contained in their saliva during feeding, which disrupt cellular functions inside the host plant. This damage leads to symptoms like; chlorosis, necrosis, wilting, stunting and curling of leaves, also known as leaf rolling. This study aimed to identify differentially expressed proteins in resistant and susceptible wheat (Triticum aestivum) cultivars during Russian wheat aphid Biotype South African 2 (RWA-SA2) infestation, in order to identify proteins involved in the wheat resistant mechanism against RWA-SA2. Two wheat cultivars SST398 (resistant to RWA-SA2) and SST356 (susceptible to RWA-SA2) were used, and a total number of 126 plants were planted and divided according to four different harvest times (Control day 0; day 5; day 7 and day 12). The wheat plants were infested with approximately 10 aphids per plant at the three-leaf stage, and placed within an insect cage inside a growth chamber (Conviron, Winnipeg, Canada) set at 24ºC, light intensity of 352 µmol.m-2.sec-1, ambient CO2 levels (410 ppm) and humidity of 60% with a 16h/8h day/night cycle. Wheat leaves were harvested at various time frames, and total protein extraction was performed. The protein samples were reduced with 10 mM dithiothreitol (DTT) and alkylated with 30 mM iodoacetamide (IAA). Sample clean-up and on-bead trypsin digest were performed on megReSyn Hilic columns, over four hours. The resultant peptides were vacuum dried and resuspended in 2% acetonitrile (ACN) before submitting them to LC-MS/MS (SWATH) for analysis. A total number of 611 proteins were differentially expressed, of which 19 were identified to be involved in the resistance response of wheat, and an additional 20 involved in biotic stress responses. This study showed that reactive oxidative species (ROS) such as hydrogen peroxide and hydroxyl radicals (OH-) play a significant role in the early stages of wheat resistance against RWA-SA2 infestation. They are involved in cell wall strengthening, activation of defence genes, involvement of phytohormone signals such as salicylic and jasmonic acid, which also mediates the systemic defences such as, systemic acquired resistance (SAR), leading to the expression of pathogen-related proteins such as (thaumatin-like proteins, oxalate oxidase, defensin, chitinase, and thionins). Identification of SANCDB compounds against G2019S and I2020T variants of leucine-rich repeat Kinase 2 (LRRK2) for the development of drugs against Parkinson’s Disease

It is the second neurodegenerative disease affecting 1-2% of people above the ages of 65 years old. There is a worldwide prevalence of 7 to 10 million affected people of all cultures and race. Studies have shown that mutation that causes Parkinson’s disease result in increased kinase activity. The c.6055 G > A in exon 41 is the most prevalent LRRK2 variation which causes a substitution of glycine to serine in G2019S in the highly activated loop of its MAP kinase domain. The LRRK2 G2019S variant is the most common genetic determinant of Parkinson’s disease identified to date. This work focused on building accurate 3D models of the LRRK2 kinase domain, that were used for large-scale in silico docking against South African natural compounds from the South African Natural Compounds Database (SANCDB; https://sancdb.rubi.ru.ac.za/). Molecular docking was performed to identify compounds that formed interactions with the active site of the protein and had the lowest binding energy scores. Molecular dynamics simulations showed different movements of the protein-ligand complexes and behavioural difference of the wildtype and the variants, all three structures proved to be compact. Network analysis was done to study residue interactions, contact maps, dynamic cross correlations, average BC and average L were used to study the residue interactions and general residue contribution to the functioning of the protein..
