- Description: Subtropical thicket supports a large variety of indigenous browsing herbivores, such as elephants, black rhino, kudu and bushbuck, but impala historically never occurred in the Eastern Cape. The aim of this study was to determine what impacts extralimital impala (Aepyceros melampus) have on subtropical thicket. The thicket of the Shamwari Game Reserve, Eastern Cape, was used for the study. To quantify the effects of impala, feeding behaviour and diet was investigated during summer and winter. Sixteen impala were also enclosed in a camp of subtropical thicket where they would be limited to feedings on thicket. Vegetation composition and structure were monitored for a period of 12 months in the enclosure. Impala followed the expected high percentage graze and a low percentage browse in thicket in summer. However, the expected was not observed during winter: at this time grazing and browsing were equally important. Field collection of feeding behaviour data showed that impala are attracted to fringes of intact bush clumps or degraded thicket. Analysis of the impala diet confirmed this, with a significantly higher percentage graze (48 percent) than browse (22 percent) during summer. However, during winter, diet analysis indicated grazing to be much higher (36 percent) than browsing (6 percent) with the impala temporarily taking advantage of nutritious grasses still present. The enclosed portion of thicket was originally at 80 percent ecological status but it showed significant changes after addition of 16 impala. Animal trails increased in number and in width, resulting in a trampling effect. Grass cover was significantly different inside the enclosure after 12 months compared to outside. The amount of available browse of the enclosed thicket was also significantly different after addition of the impala. It is concluded that impala have a negative effect on thicket, with trampling the most significant effect. Long term monitoring of the effects of extralimital impala in different types of thicket will be required to accurately quantify their direct effects on subtropical thicket in the Eastern Cape. The main aim of this study was to determine the impacts of impala on Thicket. The study showed significant impacts on Thicket as found in Shamwari, and therefore the aim of the study was achieved.

Metabolic effects brought about by tricyclic antidepressants and the contribution of a medicinal plant in alleviating high fat diet induced insulin resistance in male wistar rats

- Description: Type II diabetes is becoming a growing problem in developed countries worldwide. The median age for diagnosis was around sixty, but recent surveys have shown that the entire age distribution curve shifting left. The incidence of type II diabetes is thought to be parallel with the growing rate of obesity associated with an unhealthy western diet. Type II diabetes is an expensive disease to manage, it is for this reason that cheaper medication needs to be investigated in the form of traditional plants, such as Sutherlandia frutescens. Prescription medication, such as tricyclic antidepressants, may also increase body weight thereby playing a role in obesity. The cause of weight gain in such cases may go unrecognized or lead to cessation of the medication with or without the practitioner’s knowledge or approval. It is therefore necessary to investigate the causative agents responsible for the excessive weight gain. Drinking water containing extracts of S. frutescens or metformin was administered to two groups of eleven insulin resistant male Wistar rats. The insulin resistant control group received water without any medication. Rats were sacrificed after 8 weeks allowing for fasting blood glucose, insulin and tissue glycogen content determination. Glucose uptake was also determined using [3H] deoxyglucose. The effect of the medication and the diet on muscle post receptor insulin signaling proteins was determined through Western blots. Liver proteomics was also performed using 2-D electrophoresis. In a separate experiment 26 male Wistar rats were exposed to strepotozotocin toxin, 7 of these rats received intravenous insulin treatment, 7 rats received S. frutescens extract and 7 rats received a combination of both medications, the remaining 5 received no treatment and served as the control. Rats were sacrificed after 6 days allowing for fasting blood glucose, insulin and tissue glycogen content determination. Two groups of 14 male Wistar rats received amitriptyline or trimipramine (common tricyclic antidepressants) in their drinking water, the control group (30 rats) received water without any medication. The rats’ weight and food consumption was monitored throughout the trial and their oxygen consumption was also determined. Rats were sacrificed after 6 weeks or 14 weeks of medicinal compliance allowing for fasting blood glucose, insulin and tissue glycogen content determination. Glucose uptake was also determined using [3H] deoxyglucose. S. frutescens treatment normalized circulating serum insulin levels and significantly increased the rate of glucose clearance. Certain post receptor insulin signaling proteins were also significantly increased relative to the insulin resistant control group. 2-D electrophoresis identified the normalization of protein levels associated with the urea cycle. S. frutescens was also able to, independently; maintain normoglycaemic levels in the strepotozotocin treated group. The tricyclic antidepressants significantly increased blood glucose levels while significantly reducing tissue glycogen levels for both sacrifice periods. Serum insulin remained unchanged while a significant increase in insulin degradation and insulin degrading enzyme levels were found for both antidepressants. S. frutescens shows promise as a low cost antidiabetic medication for future use. Although the antidepressants did not promote weight gain, the increase in blood glucose levels may be cause for concern in patients with a pre-disposition toward developing diabetes.

- Description: Photovoltaic (PV) modules based on thin film systems of CuInSe2 (CIS) and its alloys on low cost substrates are promising candidates to meet the long term efficiency, reliability and manufacturing cost goals. The attention to the CIS solar cell technology is because of the high absorption coefficient of the solar cell absorber layer. Solar cells and PV modules are conventionally assessed by measuring the currentvoltage characteristic of the device. This thesis presents an assessment procedure developed capable of assessing the device parameters with reference to I-V measurements. This thesis then characterizes the performance of the CIS based solar cells and modules in conjunction with other PV modules of different technologies such as crystalline Silicon modules by analyzing the light and dark I-V measurements of the devices. The light and dark I-V characteristics of PV devices were investigated and device parameters were extracted from the I-V data. The extraction and interpretation of these device parameters has a variety of important applications. It has been proven that the device parameters can be used for quality control during production and to provide insights into the operation of the PV devices, thereby improving the efficiency of the devices. The assessment comprises light I-V measurements at standard test conditions (STC), irradiance dependence measurements, parasitic series and shunt resistances measurements and the dark I-V measurements of the PV devices. The PV modules assessed comprise different technologies, namely, thin film based modules (CIS and a-Si) and multicrystalline Si and Edged-defined Film-fed Growth Si (EFG-Si). The dark I-V measurements results showed that the EFG-Si module has acceptable shunt (900 W) and series (0.4 W) resistances, thereby leading to the higher power output depicted from the light I-V measurements. The low quality cells of a-Si module were so low that the fill factor was the smallest (43%). In addition, the dark I-V measurements results revealed that CIS modules are less dependent to temperature at high voltages.

- Description: The need for alternative strategies for the prevention and treatment of diabetes is growing rapidly as type II diabetes is reaching epidemic status in our society. This need was the basis for the creation of this study, as it was necessary to start looking towards medicinal plants as potential antidiabetic treatment and no comprehensive in vitro model existed. In creating a model for determining the effects of alternative traditional medicines as antidiabetic potentiates, it was necessary that two metabolic pathways, namely glucose uptake and insulin secretion, which play a significant role in glucose homeostasis, be at the centre of our investigations. The objective of this project was to optimize the methodology required to screen and ultimately determine the effectiveness of the plant extracts Kankerbos and MRC2003, as antidiabetic potentiates, through observing their effects on glucose utilisation and insulin secretion. If these medicinal plants are going to make a positive contribution to the health of type II diabetic South Africans, then the determination of their efficacy is essential. The cell lines used in this study included 3T3-L1 preadipocytes, Chang liver, C2C12 muscle and INS-1 rat pancreatic cells. Each cell line represents a different in vivo organ that is known to have an influence on glucose homeostasis in our bodies, each with its own unique metabolic pathways and mechanisms of activity, thereby making each one a vital component in the study. The positive controls for the two models were insulin and metformin (glucose utilisation) and glibenclamide (insulin secretion). Insulin was shown to provide a significant increase in the amount of glucose taken up in C2C12 muscle and Chang liver cells for acute conditions. Chronic treatments with metformin provided a significant increase in glucose utilised by Chang liver cells. Glibenclamide was an effective positive control for stimulating insulin secretion by INS-1 cells under acute conditions as there was a significant increase in the amount of insulin secreted. MRC2003 did not show any significant antidiabetic activity. Sutherlandia frutescens (Kankerbos) showed biological activities comparable to some of the more recognized antidiabetic compounds throughout the study. With regards to the glucose utilisation model, Kankerbos was seen to have both acute and chronic effects in different cell lines. In the C2C12 muscle cell line, Kankerbos significantly increased glucose uptake when they were exposed to acute conditions. Kankerbos also had a significant effect on the Chang liver cells as it was observed that under both acute and chronic conditions, this plant extract induced the uptake of glucose into these cells. With respect to the insulin secretion model involving INS-1 cells, no significant effect was seen during acute exposure with Kankerbos treatment. However during chronic exposure, an increase in insulin secretion was initiated by this plant extract. Overall, the results of this study suggest that Kankerbos has a twofold mechanism of action for its glucose-lowering effects. Given that Kankerbos is widely available in South Africa, this study was valuable as it provided an indication that Kankerbos has antidiabetic activities and could possibly be used as an alternative antidiabetic medication.

- Description: The method of paired comparisons can be found all the way back to 1860, where Fechner made the first publication in this method, using it for his psychometric investigations [4]. Thurstone formalised the method by providing a mathematical background to it [9-11] and in 1927 the method’s birth took place with his psychometric publications, one being “a law of comparative judgment” [12-14]. The law of comparative judgment is a set of equations relating the proportion of times any stimulus k is judged greater on a given attribute than any other stimulus j to the scales and discriminal dispersions of the two stimuli on the psychological continuum. The amount of research done for discrete models of paired comparisons is not a lot. This study develops a new discrete model, the SL-model for paired comparisons. Paired comparisons data processing in which objects have an upper limit to their scores was also not yet developed, and making such a model is one of the aims of this report. The SLmodel is thus developed in this context; however, the model easily generalises to not necessarily having an upper limit on scores.

- Description: The South African ostrich industry is presently characterised by a drastic decline in the slaughter price of slaughter birds, due to the drop in the price of meat as well as leather. As a result, the economic viability of ostrich farming is severely hampered by these developments, and many generally accepted industry practices have to be reviewed. Among the practices under review is the possibility of slaughtering ostriches at an earlier age than the traditional slaughter age of 12 - 14 months, in order to minimise feed costs and the incidence of damage to the skin. However, little information is available on the effect of the slaughter age on the yield and quality of the economically important end-products. The effect of slaughter age on production and the economic viability of a commercially intensive ostrich production system was consequently evaluated in a completely randomised experimental design involving two batches (n= 81 and n= 94) of ostriches. The experiments took place at Kromme Rhee experimental farm of the Elsenburg Research Centre near Stellenbosch. The birds were subject to experimental treatment at the age of 4 months (n= 81, from May 2004 to March 2005) and 6 months (n= 94, from August 2004 to August 2005) respectively. The ostriches were randomly divided into 10 groups of between 16 and 20 birds per group. Standard production practices, as applied in an intensive ostrich production unit, were implemented. Clean drinking water and self-mixed grower diet (min 15 percent crude protein, 0.68 percent lysine and 11.4 MJ ME per kg feed) and finisher diet (min 14 percent crude protein, 0.55 percent lysine and 9.8 MJ ME per kg feed) were provided ad libitum to the birds. Birds were slaughtered at the ages of 8.5, 10.5, 12.5, 14.5, and 16.5 months respectively. Data on feed intake and yields of cold carcass, crust skin size, skin grade and total feathers was recorded for birds slaughtered at the respective ages. Data was analysed according to the analysis of variances. Values for meat yield, skin size yield and feed intake were predicted by regression analysis based on experimental values. Industry figures and norms were used as proxy for other production inputs and costs. A gross margin type analysis was performed to evaluate the effect of different slaughter ages on the profitability of each production system. There was a positive correlation between age and feed intake (r2= 0.40), slaughter weight (r2= 0.97), cold carcass yield (r2= 0.48), skin surface area (r2= 0.39), skin grade (r2= 0.19) and total yield of feathers (r2= 0.29). The dry skin grade showed deterioration as slaughter age was prolonged. Highly significant differences (P< 0.01) in slaughter weight, cold carcass yield, skin surface area, dry skin grade and total feathers were detected at the different slaughter ages. There were no significant differences (P> 0.05) with respect to gender for slaughter weight, skin surface area and dry skin grade within the different age groups. Males had significantly higher (P< 0.05) cold carcass yield and total feather yield than their female counterparts. From this study it can be concluded that an increase in slaughter age significantly increases feed intake, slaughter weight, cold carcass yield, and skin surface area. It is also clear that an increase in slaughter age can be detrimental to dry skin grade. The relative income contribution of the three main products (skin: meat: feathers) occurred in a ratio of 47:53:0, 52:47:1, 47:50:3, 44:51:5, 39:56:5 at the respective slaughter ages. Total income revealed an increase of 26 percent from 8.5 to 10.5 months slaughtering, an increase of 4 percent between 10.5 and 12.5 months, and an increase of 11 percent between 12.5 and 14.5 months. The income dropped by 0.3 percent between 14.5 and 16.5 months slaughtering. Total costs increased with slaughter age, particularly feed costs which showed an increase (at a decreasing rate) between the different slaughter ages. The margin above specified cost per bird was the highest for birds slaughtered at the age of 10.5 months. The margin above cost showed a steady decline as birds were slaughtered at higher ages. Slaughtering at 16.5 months revealed a negative margin. To compare the effect of differences in time span on different slaughter age regimes, margin above costs for each system over 5 years were discounted to a present value. In this study, with current feed costs and income from end products, slaughtering at 10.5 months revealed the highest present value for the margin above cost. Furthermore, slaughtering at 16.5 months revealed a negative present value on the margin above cost after 5 years. Financial viability calculations are relevant and valid for data obtained in this case study. Financial viability is however case-specific and may vary between producers, depending on their individual management practices, production systems and cost structures. Results from this study may be of significant value to managerial decision-making in the ostrich industry.

- Description: Thamnochortus insignis (Albertinia thatching reed) is a restio specie which is endemic in the narrow (20 km wide) coastal dune veld, from the Gouritz River in the east to the Breede River in the west (120 km). The area within this belt, where this reed is the dominant restio, covers 65 500 hectares. The culms of the reed is harvested as a natural product from the veld and used as thatch on roofs of houses. The traditional use has been as roofing material in Cape Dutch architecture. At this time, thatch roofs are regarded as a luxury commodity and are used in prestigious residential areas and also eco-friendly housing developments. The first section of the research project was based on a Delphi technique questionnaire completed first by 37 and secondly by 10 stakeholders in the industry. This group of growers, contract harvesters, crop agents and thatchers represented 80 percent percent of the estimated 2005 harvest and 87 percent of the estimated farm gate crop value. The thatching reed industry is the main agricultural enterprise situated in the coastal dune veld of the Southern Cape, i.e. 55 percent of total Gross Product Value generated by the survey respondents. The second section is based on an analysis of experimental harvest plots (50 m2), selected at random (4 replications per site) in the 2 x 3 different production systems, i.e. traditional harvest from natural veld, harvest from veld which had been subjected to mechanical injury ("sleep") and harvest from established orchard-type lands. Harvest data was collected at each site in terms of the following components, i.e. number of harvestable tussocks, circumference of tussocks and number of reed bundles (minimum circumference 210 mm, minimum length 1,2 m). The gross income per site was calculated on the basis of R1,80 per bundle (2006-price level). Economic analysis (Gross Margin above selected costs) indicates that plant density (reed tussocks/ha) is a critical factor, in order to offset the establishment cost of R2 100/ha in established lands, which is not incurred in the other two production systems. Economic returns from the first planted lands (2 100 and 2 900 plants/ha) averaged R7 666/ha against R8 781/ha for the mechanical-injury plants. However, at a density of 5 000 plants/ha, the projected Gross Margin increases to R15 765/ha. The use of mechanicalinjury and natural vegetation production systems both interfere with biodiversity and raise major concerns with regard to sustainability of the sensitive coastal dune fynbos. ANOVA-analysis of the data indicates a highly significant difference (p = 0,01) for all sites and production systems. Statistical analysis of averages indicates that mechanical injury treatment results in a significant increase in the number of tussocks, when compared to established and natural veld, respectively. The variance in the circumference of tussocks was greatest in natural veld (55 to 71 percent) and mechanical injury (54 to 61 percent). Tussocks harvested from established plantings reflected the least level of variance (28 to 38 percent). The larger reed tussocks in the established lands produced more bundles of marketable reed (8 200/ha) than the mechanical-injury (7 625/ha) and natural veld (3 450/ha) respectively. Establishment of T. insignis plantlets in an "orchard" system at spacings of 2 m x 1 m on previous winter cereal lands or old pastures, is shown to meet all the requirements within a sustainable production system, i.e. viability, productivity, environmental-friendly, risk management and social acceptance. Furthermore, the quality of the yield was in line with the proposed grading standard for thatching reed, i.e. minimum circumference 210 mm, minimum length 1,2 m and less than 6,5 percent grey culm content.

A commercial process development for plant food formulation using polyprotic acids from natural extracts as chelating agents

- Description: The citrus industry is one of South Africa's largest agricultural sectors in terms of export earnings with lemon fruits and juice as a trendsetter due to their high grade quality. According to growers, the Eastern Cape Province of South Africa alone produces an excess of about 10-14,000 tons of lemon juice which is presently of no economic value due to the sour taste and “bitterness”. As a result of this excess and in order to make use of the polyprotic acids naturally occurring in the lemon juice, four fertilizer nutrient mixtures are formulated, using lemon juice as base. From a conceptual scientific approach, characterization (physico-chemical and functional properties determinations) of Eureka Lemon fruit juices were undertaken, followed by smaller scale batch formulation experiments. On the basis that these lemon juice-based fertilizer mixtures are prepared following standard liquid fertilizer formulation guidelines, a field test was conducted to evaluate their potential effectiveness to influence plant growth. A growth chamber testing on tomato plants revealed high growth response (> 99.9 % certainty) potential in two of the semi-organic mixtures formulated while the organic mixture showed a relatively good growth rate as compared to the control (pure tap water). According to statistical analysis (ANOVA) comparison, two of the semi-organic mixtures performed considerably better than the two commercial samples evaluated. Potential benefits profoundly associated with these nutrient mixtures as compared to similar liquid fertilizer products on the market is that most nutrients are chelated and dissolved in solution. Also, the mixtures contain all necessary nutrients including plant growth substances required for healthier plant growth. The most important socioeconomic impact is the value addition to the technology chain in the citrus industry. The use of fluid fertilizers in significant quantities is less than twenty years old. Nevertheless, growth has been so rapid that in South Africa demand for mixed liquid fertilizer has greatly increased from 90 000 tons NPK & blended micronutrients in 1955 to more than 600 000 per annum tons today (Report 41/2003, Department of Minerals and Energy). The liquid fertilizers market is sparsely specialized with major competitors like Omnia, Kynoch and Foskor supplying more than 50 % of the market demand. Amongst the nutrient mixtures formulated, mixture one is an NPK (1-1-2) based nutrient mixture containing both secondary nutrients (0.5 % Mg & 1.0 % Ca) and seven micronutrients (0.1 % Fe, 0.05 % Cu, 0.05 % Zn, 0.05 % Mn, 0.02 % B, 0.0005 % Mo and 0.0005 % Co). The composition of this mixture offers the formula a potential to be used as a general purpose (all stages of plant growth) fertilization mixture in view of its balanced composition (containing all essential plant nutrients). Mixture two contains essentially the micronutrients and in higher concentrations (0.3 % Fe, 0.3 % Cu, 0.1 % Zn, 0.2 % Mn, 0.02 % B, 0.0005 % Mo and 0.0005 % Co) as compared to mixture one except for boron, molybdenum and cobalt. The concentration of the micronutrients contained in this mixture is adequately high which offers a potential for it to be used in supplementing nutrition in plants with critical micronutrient-deficient symptoms. Mixture three is very similar to mixture two (1.0 % Fe, 0.05 % Cu, 0.05 % Zn, 0.05 Mn, 0.05 % B, 0.0005 % Mo and 0.0005 % Co) except that the concentrations of all seven micronutrients are considerably less than those of contained in mixture two. However, the concentration of iron in this mixture is as high as 1.0 %. The mixture has a potential to be used in high iron-deficient situations. Mixture four is an organic formula with relatively low nutrient concentrations (NPK-0.02-0.02-1, 0.27 % Mg, 0.02 % Ca, 0.008 % Fe, 0.26 % Cu, 0.012 % Zn, 0.009 % Mn). Nevertheless, this mixture is appealing for organically grown crops where the use of chemicals is prohibited by standards. These lemon juice-based nutrient mixtures were further characterized and tested for stability and storability over a period of eight weeks. This study revealed no major change in the physical quality (colour, pH and “salt out” effect). The basic formulation methodology is a two-step procedure that involves filtration of the lemon juice to remove membranous materials, mixing at ambient temperature and stabilization of the nutrient mixtures. However, for the organic nutrient formula mix, filtration follows after extraction of nutrients from plant materials using the lemon juice.

- Description: In this dissertation, the PMD measurements of field and laboratory optical fibre will be discussed. In the laboratory, temperature cycling of different configurations of fibre were tested. It is shown that the environmental temperature plays a significant role in determining the behaviour of PMD. The PMD of all fibres tested showed changes in the PMD when the environmental temperature was changing, but remained constant for constant temperatures. This is explained by the structure and thermal coefficients of the materials used to make up the fibres. It is found that the temperature gradient has a large effect on the magnitude of the PMD fluctuations. Field measurements were conducted on buried and aerial cable. It is shown that the PMD of aerial cables shows a higher degree of scatter than the PMD of buried cables. The scatter seen in the PMD can be attributed to changing environmental temperature as well as movement of the cable. The PMD of the deployed fibre is in all cases found to be considerably higher than the PMD of laboratory fibre. Age of the fibre, deployment conditions and installation procedures are believed to be responsible for the higher value. Long term monitoring of the PMD is also found to be critical when testing fibres. Over- or underestimations of the average PMD are often made in the field due to only a limited number of readings being taken. Having a large test ensemble of input states of polarization is shown to increase the accuracy of the results. For aerial cables, movement of the cable has the same effect as scrambling the input SOP and hence the results will be more accurate. Buried cables on the other hand show little movement and hence only one input SOP is often tested. Scrambling the input SOP will therefore increase the accuracy of these results.

- Description: Habitat selection may be considered a behavioural consequence of an individual actively selecting where it lives or passively persisting in a certain habitat. There are a variety of approaches to assessing habitat selection, including relating densities to predefined habitats and the characteristics of these habitats, measuring the behaviour of animals within predefined habitats in order to assess the relative benefits to the animal and comparing these between habitats, and using optimality theory to allow the animal to provide measures of its preference for particular habitats or patches. Each approach provides different perspectives on an animal’s choice and use of habitat, with some approaches working more effectively with certain species or habitats than others. There have, however, been no attempts to apply all these approaches to a single species at a single site. The objective of this study was therefore to apply the three above-mentioned approaches to assess habitat use and quality to a single species, the springbok (Antidorcas marsupialis) at the Augrabies Falls National Park (AFNP), on the northern bank of the Orange River, in order to provide the basis for further work on comparing and integrating these approaches. The relative abundance of springbok in Augrabies Falls National park was used to develop a habitat suitability model for the park, and thus determine the habitat variables that influenced habitat suitability. Isodar analysis revealed information on the mechanisms underlying habitat preference. Behavioural models were developed to improve our understanding of how springbok behaviour changes in relation to the habitat, and incorporated the variables identified in the habitat suitability model. The different spatial and temporal scales influencing springbok habitat selection were determined using optimal foraging theory and giving-up densities. Springbok preferred open habitats providing high food quality. Springbok behaviour was related to the foraging and predation risk characteristics of the habitat, and springbok varied their temporal and spatial utilisation so as to minimise their risk of predation and maximise their food intake. The habitat suitability model, behaviour models and giving-up densities were compared for ease of use and applicability. Using giving-up densities to reveal habitat suitability had greater applicability and was both efficient and objective.

- Description: This study focuses on the growth and characterization of epitaxial InAs and InAs1-xSbx. Layers are grown on InAs, GaAs and GaSb substrates by metalorganic vapour phase epitaxy, using trimethylindium, trimethylantimony and arsine as precursors. The growth parameters (V/III ratio, Sb vapour phase compositions) are varied in the temperature range from 500 ºC to 700 ºC, in order to study the influence of these parameters on the structural, optical and electrical properties of the materials. The layers were assessed by X-ray diffraction, electron and optical microscopy, photoluminescence and Hall measurements. Furthermore, the influence of hydrogenation and annealing on the electrical and optical properties of GaSb was investigated. It is shown that the growth temperature and the V/III ratio play a vital role in the resulting surface morphology of homoepitaxial and heteroepitaxial InAs layers. Growth at low temperatures is found to promote three-dimensional growth in both cases, with improvements in the surface morphologies observed for higher growth temperatures. All the investigated epilayers are n-type. It is shown that the electrical properties of heteroepitaxial InAs epilayers are complicated by a competition between bulk conduction and conduction due to a surface accumulation and an interface layer. The low temperature photoluminescence spectra of homoepitaxial InAs are dominated by two transitions. These are identified as band-to-band/excitonic and donor-acceptor recombination. The incorporation efficiency of antimony (Sb) into InAs1-xSbx is dependent on the growth temperature and the V/III ratio. Under the growth conditions used in this study, the incorporation efficiency of Sb is controlled by the thermal stability of the two constituent binaries (i.e. InAs and InSb). Changes in the low temperature photoluminescence spectra are detected with increasing x. From temperature and laser power dependent measurements, the highest energy line is attributed to band-to-band/excitonic recombination, while the peak appearing approximately 15 meV below this line is assigned to donor-acceptor recombination. The origin of an additional “moving” peak observed for higher Sb mole fraction x is tentatively attributed to quasi-donor-acceptor-recombination, arising from increased impurity/defect concentrations and a higher compensation ratio in the material. However, the unusual behaviour of this peak may also be ascribed to the presence of some degree of ordering in InAsSb. The exposure of a semiconductor to a hydrogen plasma usually leads to the passivation of shallow and deep centres, thereby removing their electrical and optical activity. In this study, the passivation and thermal stability of the native acceptor in p-type GaSb is also investigated. It is shown that this acceptor can be passivated, where after improvements in the electrical and optical properties of GaSb are observed. Upon annealing the passivated samples above 300 °C, the acceptor is reactivated.

On the growth and characterisation of AIGaN alloys for optoelectronic applications

- Description: In this study the growth and characterisation of undoped and Si-doped AlxGa1-xN has been performed. The layers were grown using low-pressure metalorganic vapour phase deposition (MOCVD) on sapphire substrates. The optical and electrical properties of the AlxGa1-xN layers were studied using variable temperature Hall effect and photoluminescence measurements. AlxGa1-xN layers were grown over the entire composition range. Room temperature ultraviolet (UV) transmission measurements showed that the material quality was very good for layers with an Al content, x, of 0 _ x _ 0.5. However, the quality of layers of higher composition was seen to rapidly decrease with increasing x. The electrical and optical properties of AlxGa1-xN with x < 0.5 were also good, comparable to those reported on in literature. The study of the Si-doping of AlxGa1-xN was performed in two parts; firstly a series of Al0.23Ga0.77N samples was grown in which the doping level was increased from zero to n _ 3 × 1018 cm-3. A similar, albeit a less rigorous, study was performed for Al0.41Ga0.59N and Al0.5Ga0.5N. A second series of samples was then grown in which the doping level was kept constant, while the Al content was incrementally increased. Room temperature Hall effect measurements performed on Si-doped Al0.23Ga0.77N showed that the electron concentration did not scale linearly with the silane flow, as was the case in GaN. It was also seen that the electron mobility of the layers increased with slight Si-doping, possibly due to an improvement in the crystalline quality and/or a change in the conduction mechanism. It was also found that at higher compositions (x = 0.41 and 0.50) an increase in the doping level resulted in an increase in the mobility. Variable temperature Hall effect and photoluminescence measurements, performed on the Al0.23Ga0.77N samples, revealed a good correlation between the first PL activation energy E1 and the donor activation energy ED, prompting the conclusion that the first PL recombination channel in AlxGa1-xN is due to the delocalisation of excitons bound at neutral Si donors. Furthermore, E1 and ED were seen to decrease with n1/3, as is the case for GaN and other semiconductor materials. It was also observed that strong exciton localisation occurs in slightly Si-doped material, with the amount of localization becoming less at higher doping levels. Possible mechanisms responsible for the second PL recombination channel of activation energy E2 were also proposed. The electrical and optical properties of the second set of AlxGa1-xN samples was then studied. The PL properties of undoped AlxGa1-xN were typical of a homogeneous alloy system, with the increase in the PL FWHM and exciton localisation energies with x following the trend predicted by alloy disorder theory. The variation of the band gap energy with the Al content could not, however, be fitted over the entire composition range using a single bowing parameter. It was proposed that this was due either to an effect of the 9 7 valence band crossover, or due to exciton localisation at alloy disorder and/or impurities. As was the case for GaN and Al0.23Ga0.77N, all undoped material was highly resistive. As was mentioned earlier, the exciton localisation energies increased according to alloy disorder theory in undoped AlxGa1-xN. In the doped samples, however, a large increase in the donor localisation energy was measured for x > 0.3. The possibility that Si could become a DX-centre in AlxGa1-xN was then investigated. However, Hall effect measurements showed that the Si activation energy increased in good agreement with the model of a shallow effective mass state donor, with no sudden increase in ED being observed up to x = 0.4. It was then suggested that the increase in the E1 and E2 activation energies, as well as the exciton localisation energies, could be due to the 9 7 valence band crossover, which occurs at roughly the same composition. However, due to the scarcity of reports on the valence band structure in AlxGa1-xN no conclusions could be made at this stage as to the effect of the 9 7 valence band crossover on the PL properties of AlxGa1-xN.

- Description: South Africa is not well-endowed with indigenous forests and those that remain are known to be degraded and declining at an alarming rate. This constitutes a direct threat to the quality of life of the resource-poor rural households which are directly dependent on the resources indigenous forests offer, as well as to ecological integrity. It is also recognised that the declining tree resources, particularly the indigenous tree species of high value, are increasingly threatened by a number of growing subsistence demands. This emphasises the need to establish, cultivate and conserve high-value tree species. Community forestry is recognised as a viable option for optimising land productivity; reducing pressure on indigenous forests and woodlands; ensuring a sustainable supply of desired tree products and services; and improving the quality of life of the resource-poor rural households. The primary purpose of Community Forestry Development (CFD) in the Hlabisa District is the provision of an information service and of technical support to enhance the livelihoods of rural communities by means of tree-related resources. Community forestry mainly focuses on tree resources that form an integral part of or contribute to rural people’s livelihoods. This includes small-scale growers (woodlots), woodlands and indigenous forests. Community forestry can be defined as tree growing and managed by the people for the people. Community forestry encompasses a wide range of activities which include farm forestry, agroforestry, village planting, woodlots, woodland management and indigenous-forest management by rural people, as well as tree planting in urban and peri-urban areas. Natural forests in South Africa account for less than 1 percent of the total land area. It has been suggested that these natural forests will no longer be able to meet the demand for forest products by 2020. The KwaZulu Department of Agriculture and Forestry embarked on community forestry in 1976 (but only on an ad hoc basis) as a possible solution to this dilemma. Later, the two South African pulp and paper giants, SAPPI and Mondi, started the Project Grow and Khulanathi Schemes respectively. One of the objectives of out-grower schemes was to encourage rural communities to plant trees on their own lands for the economic, social and environmental benefits trees offer. For example, timber could be sold to forestry companies. Financial assistance was offered to these communities to encourage implementation of such schemes. This exercise was aimed at bringing economic activity, capacity- building and community empowerment to the rural areas. The growers would benefit from the financial assistance and the readily available market provided by the company, while the company would be able to satisfy its demand for timber.

- Description: Aspen Pharmacare is listed on the Johannesburg Securities Exchange South Africa (JSE) and is Africa’s largest pharmaceutical manufacturer. The company is a major supplier of branded pharmaceutical and healthcare products to the local and selected international markets. For decades, Aspen has manufactured a basket of affordable, quality, and effective products for the ethical, generic over-the-counter (OTC) and personal care markets. Aspen is also the leading supplier of generic medicines to the public sector, providing comprehensive coverage of the products on the Essential Drug List. Aspen continues to deliver on its commitment toward playing a role in social responsibility diseases such as HIV/AIDS, tuberculosis and malaria. In August 2003 Aspen developed Africa’s first generic anti-retroviral drug, namely Aspen-Stavudine. Aspen’s manufacturing facilities are based in Port Elizabeth (PE) and East London. Aspen has recently completed an Oral Solid Dosage (OSD) manufacturing facility worth approximately R150 million in PE. The Group manufactures approximately 20 tons of product daily and in excess of 400 tons of solid dosage pharmaceuticals, which equates to more than 2 billion tablets. The development of an electrochemical process for the production of para-substituted di-hydroxy benzenes

- Description: The project was concerned with the investigation of the electrochemical oxidation of various phenols, and to develop a viable reaction system for the production of the respective hydroquinones. Current production routes utilizing phenol as starting material have the limitations of using large amounts of acids, having to be stopped at low conversions and producing a mixture of the hydroquinones and catechols. Of the possible routes to the respective hydroquinones from the various phenols, the electrochemical oxidation of these phenols offers commercial and environmental advantages and hence formed the theme of the investigation. The synthetic possibilities proved to be more prevalent in a system when the electrochemical oxidation of these phenols was performed in an aqueous medium utilizing an organic co-solvent. Results obtained during this investigation made it possible to make certain predications about the mechanism taking place. This was found to depend on the anode material used for the oxidation. The results showed that the process developed for the electrochemical oxidation of these phenols, yields mainly the para-isomers of the respective hydroquinones and benzoquinones in good yields and selectivities, with fair current efficiencies and good mass balances at high conversions. For example: * Phenol (batch) 8 F: 345% current efficiency, 70% hydroquinone, 6% catechol, 9% benzoquinone, 9% phenol and 94 mass balance. * 2-Tert-butylphenol (flow) 10 F: 37% current efficiency, 65% hydroquinone, 33% benzoquinone, 2% phenol and 100% mass balance. * 2,6-Di-tert-butylphenol (flow) 11 F: 23% current efficiency, 92 % hydroquinone, 6% benzoquinone, 1% phenol and 99% mass balance. The developed electrochemical oxidation system offers the following advantages over previous and current methods: simplified isolation and extraction procedures, smaller amounts of acid usage, reasonably selective synthesis of the para-isomer and a less corrosive system all at high conversions.

- Description: Many network monitoring tools do not provide sufficiently in-depth and useful reports on network usage, particularly in the domain of application services data. The optimisation of network performance is only possible if the networks are monitored effectively. Techniques that identify patterns of network usage can assist in the successful monitoring of network performance. The main goal of this research was to propose a model to mine and visualise application services data in order to support effective network management. To demonstrate the effectiveness of the model, a prototype, called NetPatterns, was developed using data for the Integrated Tertiary Software (ITS) application service collected by a network monitoring tool on the NMMU South Campus network. Three data mining algorithms for application services data were identified for the proposed model. The data mining algorithms used are classification (decision tree), clustering (K-Means) and association (correlation). Classifying application services data serves to categorise combinations of network attributes to highlight areas of poor network performance. The clustering of network attributes serves to indicate sparse and dense regions within the application services data. Association indicates the existence of any interesting relationships between different network attributes. Three visualisation techniques were selected to visualise the results of the data mining algorithms. The visualisation techniques selected were the organisation chart, bubble chart and scatterplots. Colour and a variety of other visual cues are used to complement the selected visualisation techniques. The effectiveness and usefulness of NetPatterns was determined by means of user testing. The results of the evaluation clearly show that the participants were highly satisfied with the visualisation of network usage presented by NetPatterns. All participants successfully completed the prescribed tasks and indicated that NetPatterns is a useful tool for the analysis of network usage patterns.

- Description: Web Usage Mining (WUM) can be used to determine whether the information architecture of a web site is structured correctly. Existing WUM tools however, do not indicate which web usage mining algorithms are used or provide effective graphical visualisations of the results obtained. WUM techniques can be used to determine typical navigation patterns of the users of organisational web sites. An organisational web site can be described as a site which has a high level of content. The Computer Science & Information Systems (CS&IS) web site at the Nelson Mandela Metropolitan University (NMMU) is an example of such a web site. The process of combining WUM and information visualisation techniques in order to discover useful information about web usage patterns is called visual web mining. The goal of this research is to discuss the development of a WUM model and a prototype, called WebPatterns, which allows the user to effectively visualise web usage patterns of an organisational web site. This will facilitate determining whether the information architecture of the CS&IS web site is structured correctly. The WUM algorithms used in WebPatterns are association rule mining and sequence analysis. The purpose of association rule mining is to discover relationships between different web pages within a web site. Sequence analysis is used to determine the longest time ordered paths that satisfy a user specified minimum frequency. A radial tree layout is used in WebPatterns to visualise the static structure of the organisational web site. The structure of the web site is laid out radially, with the home page in the middle and other pages positioned in circles at various levels around it. Colour and other visual cues are used to show the results of the WUM algorithms. User testing was used to determine the effectiveness and usefulness of WebPatterns for visualising web usage patterns. The results of the user testing clearly show that the participants were highly satisfied with the visual design and information provided by WebPatterns. All the participants also indicated that they would like to use WebPatterns in the future. Analysis of the web usage patterns presented by WebPatterns was used to determine that the information architecture of the CS&IS web site can be restructured to better facilitate information retrieval. Changes to the CS&IS web site web were suggested, included placing embedded hyperlinks on the home page to the frequently accessed sections of the web site.

A comparison of programming notations for a tertiary level introductory programming course

- Description: Increasing pressure from national government to improve throughput at South African tertiary education institutions presents challenges to educators of introductory programming courses. In response, educators must adopt effective methods and strategies that encourage novice programmers to be successful in such courses. An approach that seeks to increase and maintain satisfactory throughput is the modification of the teaching model in these courses by adjusting presentation techniques. This thesis investigates the effect of integrating an experimental iconic programming notation and associated development environment with existing conventional textual technological support in the teaching model of a tertiary level introductory programming course. The investigation compares the performance achievement of novice programmers using only conventional textual technological support with that of novice programmers using the integrated iconic and conventional textual technological support. In preparation for the investigation, interpretation of existing knowledge on the behaviour of novice programmers while learning to program results in a novel framework of eight novice programmer requirements for technological support in an introductory programming course. This framework is applied in the examination of existing categories of technological support as well as in the design of new technological support for novice programmers learning to program. It thus provides information for the selection of existing and the design of new introductory programming technological support. The findings of the investigation suggest strong evidence that performance achievement of novice programmers in a tertiary level introductory programming course improves significantly with the inclusion of iconic technological support in the teaching model. The benefits are particularly evident in the portion of the novice programmer population who have been identified as being at risk of being successful in the course. Novice programmers identified as being at risk perform substantially better when using iconic technological support concurrently with conventional textual technological support than their equals who use only the latter form. Considerably more at risk novice programmers using the integrated form of technological support are in fact successful in the introductory programming course when compared with their counterparts who use conventional textual technological support only. The contributions of this thesis address deficiencies existing in current documented research. These contributions are primarily apparent in a number of distinct areas, namely: • formalisation of a novel framework of novice programmer requirements for technological support in an introductory programming course; • application of the framework as a formal evaluation technique; • application of the framework in the design of a visual iconic programming notation and development environment; • enhancement of existing empirical evidence and experimental research methodology typically applied to studies in programming; as well as • a proposal for a modified introductory programming course teaching model. The thesis has effectively applied substantial existing research on the cognitive model of the novice programmer as well as that on experimental technological support. The increase of throughput to a recommended rate of 75 percent in the tertiary level introductory programming course at the University of Port Elizabeth is attributed solely to the incorporation of iconic technological support in the teaching model of the course.

- Description: The present product development project was aimed at studying the synergism and/or antagonism amongst various known insect repellent actives with the view to formulating a multi-active repellent product with improved properties when compared to current single-active commercial products. Advanced statistical methods were used to identify synergism between individual active substances and to define a formulation as close as possible to the “ideal” formulation. Several mosquito repellent samples were prepared and sent to the South African Bureau of Standards (SABS) in Pretoria to test for their efficiency in repelling mosquitoes. From the results of the repellency tests of the various active combinations, three actives were identified that showed promising signs of synergism. These actives were then studied in further detail to determine their optimum combination. In addition, it was shown that when using a natural flavourant as promoter and incorporating a slow-release agent into formulations for aerosols and lotions, a product is obtained that gives comparable levels of efficiency to current commercial products, but at much reduced levels of active loading. Accelerated stability tests performed on the final combination of the three actives used in the final formulation showed no adverse reactions over a three-week study. These tests shall be repeated once the final application form (lotion, aerosol, etc) and product packing have been decided.

- Description: The purpose of this dissertation was to provide a review of the theory of Optimization, in particular quadratic programming, and the algorithms suitable for solving both convex and non-convex quadratic programming problems. Optimization problems arise in a wide variety of fields and many can be effectively modeled with linear equations. However, there are problems for which linear models are not sufficient thus creating a need for non-linear systems. This dissertation includes a literature study of the formal theory necessary for understanding optimization and an investigation of the algorithms available for solving a special class of the non-linear programming problem, namely the quadratic programming problem. It was not the intention of this dissertation to discuss all possible algorithms for solving the quadratic programming problem, therefore certain algorithms for convex and non-convex quadratic programming problems were selected for a detailed discussion in the dissertation. Some of the algorithms were selected arbitrarily, because limited information was available comparing the efficiency of the various algorithms. Algorithms available for solving general non-linear programming problems were also included and briefly discussed as they can be used to solve quadratic programming problems. A number of algorithms were then selected for evaluation, depending on the frequency of use in practice and the availability of software implementing these algorithms. The evaluation included a theoretical and quantitative comparison of the algorithms. The quantitative results were analyzed and discussed and it was shown that the results supported the theoretical comparison. It was also shown that it is difficult to conclude that one algorithm is better than another as the efficiency of an algorithm greatly depends on the size of the problem, the complexity of an algorithm and many other implementation issues. Optimization problems arise continuously in a wide range of fields and thus create the need for effective methods of solving them. This dissertation provides the fundamental theory necessary for the understanding of optimization problems, with particular reference to quadratic programming problems and the algorithms that solve such problems.
