- Description: In this research, the objective was to investigate under the chosen liquefaction conditions whether co-liquefaction of coal and microalgae biomass as agglomerates does possess a distinct advantage over the individual liquefaction of microalgae and coal. This was initially done by preparing dry micro algal biomass, followed by the preparation of coal fines and finally coal-algae as agglomerates. The agglomerates were prepared by continuously adsorbing wet concentrated microalgae slurry/solution onto the dry coal fines in order to prepare different ratios of coal-algae agglomerate(s). Ultimate analysis of the starting material (coal fines and the microalgae) as well the agglomerates then followed. According to research, wet microalgae biomass has the potential of acting as a hydrogen donor. By preparing the agglomerates, the rationale was to promote hydrogen supply from the micro algal biomass that has high hydrogen content to the coal with lower hydrogen content. The resulting product was then expected to have fairly higher hydrogen content. However, this was not observed or evident from the elemental analysis results. The results (elemental composition) showed no difference between the agglomerates and the coal. Based on the observed ultimate analysis results, liquefaction experiments were conducted to determine whether hydrogen transfer and the expected synergistic effect between micro algal biomass and coal would occur. The crude oil product was recovered and simulated distillation technique was used for the characterization of the crude oil product. The results we in agreement that higher yields can be achieved during co-liquefaction as compared to the individual liquefaction. A possible synergistic effect that was investigated in this research existed. Liquefaction results provided evidence of higher yields recovery from the agglomerates compared to those of coal fines and microalgae biomass. Although there was higher recovery on the agglomerates, there appears to be a downward trend (decrease) in product recovery with higher loadings of micro algal biomass on to the coal. This means that higher loading of micro algal biomass tend to suppress liquefaction of coal, thus resulting in lower product recovery. While lower loadings tend to effectively facilitate the liquefaction of coal, thus proving that the proposed synergistic effect between the two does exist. Overall product recovery (yields) of the agglomerates is higher compared to the individual recovery of coal fines and microalgae biomass. This was followed by mass balancing to determine the overall conversion. Higher conversions were achieved on the agglomerates compared to coal. This could mean that microalgae have the potential to facilitate the liquefaction of coal and improve its conversion. The observed trend results in improved conversion as well as higher oil yields (simulated distillation results). Overall, the mass balancing provided insightful information regarding the coal-microalgae interaction based on the conversion, and this further corresponds or supports the simulated distillation results. This research paper provides evidence of the synergistic effect that exists between micro algal biomass and coal.

- Description: The cyanobacterially synthesized non-canonical amino acid β-N-methylamino-ʟ-alanine (BMAA) has been proposed to be a causative agent in the development of sporadic neurodegenerative diseases. This neurotoxin bioaccumulates and biomagnifies with increasing trophic levels in ecosystems by associating with proteins. It has been suggested that these associations with host proteins also serve as a reservoir from where BMAA is slowly released with normal protein catabolism, resulting in a continuous low level exposure. However, the nature of these associations remains poorly defined. The widely accepted hypothesis regarding the nature of these associations is that BMAA associates with proteins through primary incorporation into proteins with specific replacement of serine. In addition to excitotoxicity, BMAA misincorporation has been proposed as a potential mechanism of toxicity because of its link to protein tangle diseases. Interactions between BMAA and proteins that are not the result of misincorporation, have also been observed. However, the nature of these non-primary associations has not been investigated. This study focussed on establishing whether BMAA is misincorporated into host proteins with consequent toxicity, and on elucidating the nature of the BMAA-protein associations not linked to primary incorporation. In comparative studies between BMAA and canavanine, an arginine analogue known to misincorporate, exposure to BMAA did not result in any toxicity in prokaryotes or in an undifferentiated eukaryotic mammalian cell line, in contrast to what was observed upon canavanine exposure. Differentiation of the cell line with nerve growth factor to express glutamate receptors resulted in marked toxicity upon BMAA exposure, highlighting excitoxicity as the main mechanism of BMAA toxicity. Furthermore, it was demonstrated that BMAA interacts with free amino acids and proteins in the absence of de novo protein synthesis, causing enzyme inhibition and protein misfolding. It was concluded that BMAA does not interact with proteins through primary incorporation and that the observed associations are the result of an interaction between BMAA and amino acid side chains to form covalent bonds.

Investigating the use of social networking via mobile phone as an extension tool in small-scale (emerging) agriculture in selected farming communities in the Cacadu District

- Description: We live in the era in which the internet is now available on the majority of mobile phones at a very cheaper rate. This advancement in technology has created a boom in the use of mobile phone social networking as a primary communication tool - not only for individuals but also commonly used by professionals in most fields. The trends and growing usage of social networking via mobile phone indicate a potentially effective new platform for increasing production especially in agricultural sectors. In this age of information technology, farming communities can be empowered with the latest information and knowledge through mobile phone social networking to enhance agricultural development. The study investigates the use of mobile phone social networking as an extension tool in small-scale (emerging) farmers in selected farming communities in the Cacadu District Municipality of the Eastern Cape of South Africa. Specifically, the study investigates the use of available mobile phone social networks; what they are used for; and the importance and benefits of social networking both generally and to the farming communities. The study further identifies problems inhibiting the use of mobile phone social networking. Other channels through which the small-scale (emerging) farmers acquire agricultural information, aside from via mobile phone, and the type of agricultural information these farmers acquire, are also investigated. This study focuses on the use - and not the testing or adoption of - mobile phone social networking in small-scale (emerging) agriculture. In the exercise to investigate the use of social networking apps via mobile phone, data was collected from 40 small-scale (emerging) farmers using a purposive sampling method. This study used a survey design, exploratory and descriptive research methodologies. Data was captured by administering a questionnaire through personal face-to-face interviews. Descriptive statistics such as simple frequency tables, percentages and bar graphs were used. From the Chi-square testing, it was found that agricultural knowledge levels, internet access and size of production farm land influence the use of social networking by the small-scale (emerging) farmers. However, the result of this study showed that gender, age, other income and educational level of the small-scale (emerging) farmers are not statistically significant determinants of social networking use. Cronbach’s alpha coefficient was found to be 0.95. To establish the strength of associations of the variables, Cramer’s V ranging between 0.04 and 0.9 (signifying relationships from negligible to very strong association) was used. The findings showed that different social networks are used by the farming communities, such as WhatsApp, Facebook, and Blackberry messenger, MXit, Twitter and YouTube. The various uses of these social networks by the farmers are discussed. The benefits and problems inhibiting their use are also outlined. Other sources of acquiring agricultural information by the farmers aside from social networking via mobile phone are examined. The result of the study shows that [agricultural extension officers, other farmers, farmers’ discussion groups, friends and family,] are major sources of agricultural information to the small-scale (emerging) farmers while other sources are television, radio, print media and result demonstrations. The study suggests that to enhance mobile phone social networking to ensure that timely and effective agricultural information is readily accessible to the small-scale (emerging) farmers. These include agricultural extension officers making optimal use of social networking via mobile phone, due to the strong interaction and trust between them and the farmers. This interaction can be instrumental in the effective use of this technology as a resourceful medium for accessing agricultural information to enhance productivity. Proper awareness and understanding of the potential and benefits of mobile social networking by the farmers will motivate the small-scale (emerging) farmers to use the technology for effective agricultural purposes. Also, ensuring the availability of agricultural information to small-scale (emerging) farmers by setting up on-line pages or websites to discuss matters of interest and educate and update farmers on agricultural issues should be encouraged. This may attract the younger generation and the youth to participate fully in agricultural activities - thereby enhancing agricultural development.

Long term effects of temperature and humidity on lignocellulosic fibres and composites

- Description: The study deals with the chemical and flame retardant (FR) treatment of woven flax fabric and preparation and characterization of flax reinforced phenolic composites. Sheets of flax fabric were subjected to chemical treatments using NaOH and silane coupling agents. A phosphate-based flame retardant (DAP) was applied to decrease the flammability of the flax fabric. The effect of the chemical treatments and FR treatments on the thermal and flammability properties of the fabric and composites was investigated using thermogravimetric analysis (TGA), vertical flame resistance test and cone calorimeter. The mechanical properties of the flax fabric and composites, before and after environmental ageing, were investigated. Ageing studies were carried out by exposing the samples in an environmental chamber at specified conditions for two weeks. TGA results showed that the treatment of the fabric with FR shifts the decomposition temperature to lower level and increases the char residue. Vertical flame resistance testing showed that FR treatment of the flax fabric improved the flammability properties. There was no after flame and afterglow observed for FR treated flax fabric due to self-extinguishment after removal of the flame. Flax Fabric without FR burned completely and there was no result for the char length due to the complete destruction of the fabric. The FR treatment, however, seemed to have had a negative effect on the tensile strength of the flax fabric. This was further intensified upon exposure of FR treated flax fabric to high temperatures during ageing studies which drastically reduced the tensile strength by more than 90%, and the flax fabric were found to be brittle and darker in visual appearance. TGA results of flax/phenolic composites showed that for composites containing chemically treated and FR treated flax fabric the decomposition temperatures shifted to lower temperatures; however there was no significant difference in the amount of char residue. Untreated flax/phenolic composites exhibited the lowest char residue. Cone calorimeter results showed that the peak heat release rate (PHRR), smoke production rate (SPR) and carbon dioxide (CO2) emission rate was reduced for the flax/phenolic composite produced using FR treated flax fabric. The tensile strength of these composites was reduced while there was an increase in modulus value. Exposure of the FR treated composites to high temperatures further reduced the tensile strength and increased the E-modulus. Both FR treated and untreated composites changed in colour and the FR treated composites were found to be brittle after exposure to high temperatures.

On the characterisation of diffused light and optical elements in high concentrator photovoltaic modules

- Description: High Concentrated Photovoltaics (H-CPV) promise a more efficient, higher power output than traditional photovoltaic modules. This is achieved by concentrating sunlight onto a small triple junction (CTJ) InGaP/InGaAs/Ge cell (ranging from 3.14 mm2 to 1 cm2) by using precision optical systems. These systems utilise non-imaging optics to concentrate and distribute the incident solar flux uniformly onto the CTJ device receiver to achieve maximum performance and power output from an H-CPV module. However, the performance of the device can be reduced due to the partial or complete absorption of a range of wavelengths present in the solar spectrum by the optical materials that are used for concentration. An investigation to determine the current density topographies of each subcell in a CTJ cell by multiple raster scans of an optical fibre receiver of a spectrometer in the plane of the aperture of the secondary’s optical element was conducted. Results showed that the physical properties of the optical elements’ material absorbed different amounts of the spectral content with respect to the subcell photosensitive wavelength regions. The facet properties of the primary optical Fresnel lens showed that the more rounded the Fresnel facets were, the lower the concentration of sunlight incident onto the CTJ cell. The increase in facet numbers showed an increase in scattering of the incident sunlight and chromatic aberrations. Chromatic aberration created by the refractive optics showed a variation in the amount of concentration on each individual subcell as well as the difference in intensity profiles across for the different subcells. Based on these results and the development of new multi-junction devices by industry, the performance of a four and six-junction device with the optical materials was investigated by simulations. The simulations showed that the careful integration of an additional subcell in a multi-junction device could rectify current mismatch between the subcells in the device. Based on the simulations, the best performing multi-junction cell was identified as the four-junction device that showed a cell and module efficiency under operation of 42.5 % and 35.5 %, respectively. Additionally, based on the performance results observed from the H-CPV module, the development of an HCPV module that would attempt to harness the incident tracked diffuse sunlight available to a concentrator photovoltaic (CPV) module for additional energy yield was undertaken. The part of the study comprised of measurements of the solar source, design of a prototype Hybrid High Concentrator Photovoltaic (HH-CPV) module. Results showed that power generation from the H-CPV system was highly dependent on the DNI levels and fluctuates greatly with variation in the DNI. The irradiance levels within the diffuse regions of the H-CPV module showed that the baseplate and vertical sides had an average irradiance range of 140-450 and 50-225 W.m-2, respectively. Irradiance topographic raster scans revealed that the baseplate and vertical sides had a relatively uniform intensity distribution and was identified as favourable sites for diffuse cell population. Simulations of various PV technologies showed the most suitable technology for the placement within the cavity of the HH-CPV module. The developed HH-CPV module was finalized with the utilization of CIS modules to harness the diffuse irradiance. During a 3 month power monitoring of the HH-CPV system, it was determined that the major power generation for the HH-CPV module come from the CPV component, while the CIS modules showed a minor power contribution. The total energy yield for the monitoring period was 45.99, 3.89 and 1.76 kW.h for the CPV, four-vertical sides and baseplate components, respectively. The increase in energy yield of the HH-CPV module when compared to the standard H-CPV module was determined to be 12.35 % for the monitoring period. - Description: High Concentrated Photovoltaics (H-CPV) promise a more efficient, higher power output than traditional photovoltaic modules. This is achieved by concentrating sunlight onto a small triple junction (CTJ) InGaP/InGaAs/Ge cell (ranging from 3.14 mm2 to 1 cm2) by using precision optical systems. These systems utilise non-imaging optics to concentrate and distribute the incident solar flux uniformly onto the CTJ device receiver to achieve maximum performance and power output from an H-CPV module. However, the performance of the device can be reduced due to the partial or complete absorption of a range of wavelengths present in the solar spectrum by the optical materials that are used for concentration. An investigation to determine the current density topographies of each subcell in a CTJ cell by multiple raster scans of an optical fibre receiver of a spectrometer in the plane of the aperture of the secondary’s optical element was conducted. Results showed that the physical properties of the optical elements’ material absorbed different amounts of the spectral content with respect to the subcell photosensitive wavelength regions. The facet properties of the primary optical Fresnel lens showed that the more rounded the Fresnel facets were, the lower the concentration of sunlight incident onto the CTJ cell. The increase in facet numbers showed an increase in scattering of the incident sunlight and chromatic aberrations. Chromatic aberration created by the refractive optics showed a variation in the amount of concentration on each individual subcell as well as the difference in intensity profiles across for the different subcells. Based on these results and the development of new multi-junction devices by industry, the performance of a four and six-junction device with the optical materials was investigated by simulations. The simulations showed that the careful integration of an additional subcell in a multi-junction device could rectify current mismatch between the subcells in the device. Based on the simulations, the best performing multi-junction cell was identified as the four-junction device that showed a cell and module efficiency under operation of 42.5 % and 35.5 %, respectively. Additionally, based on the performance results observed from the H-CPV module, the development of an HCPV module that would attempt to harness the incident tracked diffuse sunlight available to a concentrator photovoltaic (CPV) module for additional energy yield was undertaken. The part of the study comprised of measurements of the solar source, design of a prototype Hybrid High Concentrator Photovoltaic (HH-CPV) module. Results showed that power generation from the H-CPV system was highly dependent on the DNI levels and fluctuates greatly with variation in the DNI. The irradiance levels within the diffuse regions of the H-CPV module showed that the baseplate and vertical sides had an average irradiance range of 140-450 and 50-225 W.m-2, respectively. Irradiance topographic raster scans revealed that the baseplate and vertical sides had a relatively uniform intensity distribution and was identified as favourable sites for diffuse cell population. Simulations of various PV technologies showed the most suitable technology for the placement within the cavity of the HH-CPV module. The developed HH-CPV module was finalized with the utilization of CIS modules to harness the diffuse irradiance. The LBIC maps shows performance degrading defects present in the bulk and the surface of the solar cells as a function of spatial distribution. These localised defects acts as trapping mechanism for the charge carriers and therefore limits recombination within the solar cell and thus decreasing the performance of the solar cell device. The resulting I-V characteristics obtained from the LBIC system were used to determine the performance parameters using the two algorithms. The resultant effect of these parameters on the performance of the solar cells was observed. The overall results showed that LBIC is a useful tool for identifying and characterising defects in solar cells.

The LBIC is a non destructive characterisation technique that uses a focused light beam to raster scan a solar cell surface as the photo-generated current is recorded as a function of position to generate a photo-response map. The technique was used to obtain photoresponse maps for a mc-Si, Back contact Back junction (BC-BJ) silicon solar cell and the InGaP/InGaAs/Ge concentrating triple junction (CTJ) solar cell from which various local current reducing defects were mapped. A reflection signal detector was incorporated into the LBIC measurement system to enable us distinguish between optical blockages on the cell surface and current reducing defects within the solar cell devices. By dynamically biasing the solar cell devices, the electrical activity of the identified defects was investigated and also point-by-point current-voltage (I-V) characteristics were obtained. An interval division algorithm was applied to the measured point-by-point I-V characteristics to extract device and performance parameters from which device and performance parameter uniformity of the devices were mapped. Dark and full cell solar illumination I-V characteristics were also measured to extract device parameters. Analysis of extracted parameters revealed differences between extracted dark and illuminated device parameters which was attributed to departure from the superposition principle due to non-linearity of the semiconductor device equations with respect to carrier concentration. An investigation into the effect of illumination intensity on the I-V parameters of a spot illuminated BC-BJ Si solar cell showed a linear increase and a logarithmic increase of the short circuit current and open circuit voltage respectively with intensity while the series resistance decreased with intensity, which was attributed to increase in conductivity of the active layer. The ideality factor and saturation current were observed to increase while the shunt resistance initially increased before decreasing at higher intensity levels. Under monochromatic illumination, the photo-response of the BC-BJ Si cell was higher at 785nm than at 445nm due to low absorption coefficient of Si for longer wavelength radiations, resulting in carrier generation within the bulk, where there is a higher probability of carriers being collected at the p-n junction before they recombine. Under solar illumination, as the spectral content was altered using long pass colour filters with cut off wavelengths of 610nm and 1000nm, the performance parameters were observed to decrease and this was mainly due to decrease in intensity. For the CTJ solar cell, however, blocking of radiations below 610nm resulted in current mismatch that severely degraded the short circuit current (Isc). The current mismatch affected the extracted device and performance parameters. With a 1000nm long pass filter, a dark I-V was obtained since only the bottom Ge subcell was activated.

Perspectives of stakeholders on engagement around benefits and use of the Wilderness and Swartvlei lakes

The Wilderness and Swartvlei lake systems, which form an integral part of the Wilderness Section of the Garden Route National Park (GRNP), were chosen as the study area. The park is juxtaposed with urban and other land uses making it one of the most integrated urban conservation areas in South Africa. The park is an open-access park with only enclosed areas being the camping and chalet areas that borders onto the Touw River Estuary. A major contribution of this research was that it classified stakeholder groups into ten meta-identities (associations, businesses, charity organisations, conservancies, government departments, informal groupings, learning/educational institutions, multiple stakeholder projects, spiritual groupings and sports clubs) and that through an iterative research approach it stimulated dialogue between individuals across the various meta-identities. Engagement is a way of allowing stakeholders to develop a sense of ownership through the decision-making process. This could also lead to a higher level of trust and cooperation. The main insights drawn from this research were that, history is important; engagement is characterised by an on-going blame-game (at least in part as a result of this particular history); meta-identities share some values, but differ in how they see the world; there is a concern over social issues (widespread concern, but few mechanisms to address the issue); management agencies are stewards of the feedbacks between social and ecological systems (responsible for regulating flows of benefits), but in open-access systems cannot do so on their own; all meta-identities are keen to contribute and this can, with appropriate facilitation, be harnessed towards collective action. Stakeholders associated with all ten meta-identities identified provisioning and cultural services as a benefit derived from the lakes. No regulating or supporting services were identified as benefits. This could indicate a gap in awareness of the importance of these services. A category that emerged from this study is the importance of employment opportunities linked to the management of natural resources. The study showed that engagement does not occur around the benefits that can be derived from the lakes, but rather around the issues that have a direct or indirect influence on the ecosystem services and therefore the suite of benefits that would be available for sharing. The issues could be divided into two broad themes; social issues and developmental pressures. The social issues pose an indirect threat to the lakes while the developmental pressures pose a direct threat. As the mandates across agencies differ, with the municipalities concerned with the social issues and SANParks with the pressures, the importance of communication and cooperative governance was highlighted.

Present state of macrophytes and responses to management scenarios at the St. Lucia and Mfolozi estuaries

- Description: The St. Lucia Estuary, the largest estuary in South Africa, has been subject to many natural (a decade long drought) and anthropogenic impacts. A particular mouth manipulation activity, the artificial separation of the Mfolozi River and the St Lucia Estuary in 1952, was done to stop the perceived “silting up” of the estuary, but resulted in a decrease in freshwater supply. The changes in inflows (both fresh and marine) are controlled by management decisions and affect other system parameters such as salinity, water level and turbidity, which influence the distribution of biota. Therefore knowledge on the physico-chemical environment and eco-physiological tolerances of macrophytes will lead to informed future management decisions. The first of the three objectives carried out for this study determined the present state and distribution of the macrophytes of the St. Lucia and Mfolozi estuaries. The macrophyte habitats mapped in 2008 and 2013 were the submerged macrophytes, reeds and sedges, mangroves, grass and shrubs, salt marsh (succulent) and swamp forest. Results indicated that low salinity in the lakes and high water level in 2013 caused die-back or expansion of particular habitats. Submerged macrophytes, in particular Stuckenia pectinata, grows well in water with salinity <15 ppt, therefore this habitat increased by 412 ha (96%) in cover since 2008. Salt marsh decreased by 553 ha (57%) due to inundation. Since 2008 the reeds and sedges increased by 390 ha (in North Lake and the Narrows) due to the salinity decrease. In the Narrows the mangroves decreased by 28 ha (9%) in area cover. This was due to the drought that persisted for so many years, which caused low water levels and non-tidal conditions. The second objective was to determine the present state / health of the mangroves at four sites along the Narrows by assessing sediment condition and population structure of the trees. These results were compared to those obtained in 2010. The total density of Avicennia marina increased since 2010, however this was due to the large increase in seedlings at Site 1, the back channel site. The highest sediment salinity (26 ppt) and porewater salinity (29 ppt) was recorded for this site and these results show that this back channel site was strongly influenced by the open Mfolozi Estuary (increase in marine waters) and tidal conditions at the time of sampling in 2013. The total density of Bruguiera gymnorrhiza decreased, but an increase in adults was recorded at Site 2, the freshwater site. The soil collected from the Bruguiera quadrats was fresher and drier than that of the Avicennia quadrats of Site 2. Lack of seedlings (of both species) was due to the dense stands of Acrostichum aureum (mangrove fern) and Phragmites australis (common reed) and a thick, impenetrable mat of Avicennia pneumatophores. At Sites 3 and 4, the drier sites (where sediment moisture contents were the lowest for all sites at 43 and 42% respectively), seedling and sapling density was low, but adults increased in density since 2010. Recruitment and survival were impacted by the harsh environmental conditions that prevailed prior to 2013 (low water level and non-tidal influence), but adults survived. The results of the test that determined the percentage of aerenchyma of the pneumatophores indicated that waterlogging stress did not affect the aerenchyma of the pneumatophores. However the period of inundation was probably not significant enough to have affected the production of aerenchyma. The third objective formed part of an ongoing study by the Global Environmental Facility (GEF) on the feasibility of linking the Mfolozi River back to the St Lucia Estuary and the responses of these systems to different management scenarios: 1) the “do nothing” scenario; 2) maintain separate Mfolozi and St Lucia mouths representing an open mouth condition; and 3) actively facilitate a single mouth (therefore linking the Mfolozi and St Lucia mouths). Data gathered on the eco-physiological tolerances of the dominant macrophyte species was used to predict the response of the different habitats to these various management scenarios and the results indicated that the best management scenario would be to actively facilitate a single mouth (Scenario 3) as the estuary habitats would increase significantly because of preferred tidal and saline conditions, as this would represent more natural conditions. The results of the study will provide input to recommendations for future “adaptive management” strategies for the Global Environmental Facility (GEF) Project.

Salt marsh decreased by 553 ha (57%) due to inundation. Since 2008 the reeds and sedges increased by 390 ha (in North Lake and the Narrows) due to the salinity decrease. In the Narrows the mangroves decreased by 28 ha (9%) in area cover. This was due to the drought that persisted for so many years, which caused low water levels and non-tidal conditions. The second objective was to determine the present state / health of the mangroves at four sites along the Narrows by assessing sediment condition and population structure of the trees. These results were compared to those obtained in 2010. The total density of Avicennia marina increased since 2010, however this was due to the large increase in seedlings at Site 1, the back channel site. The highest sediment salinity (26 ppt) and porewater salinity (29 ppt) was recorded for this site and these results show that this back channel site was strongly influenced by the open Mfolozi Estuary (increase in marine waters) and tidal conditions at the time of sampling in 2013. The total density of Bruguiera gymnorrhiza decreased, but an increase in adults was recorded at Site 2, the freshwater site. The soil collected from the Bruguiera quadrats was fresher and drier than that of the Avicennia quadrats of Site 2. Lack of seedlings (of both species) was due to the dense stands of Acrostichum aureum (mangrove fern) and Phragmites australis (common reed) and a thick, impenetrable mat of Avicennia pneumatophores. At Sites 3 and 4, the drier sites (where sediment moisture contents were the lowest for all sites at 43 and 42% respectively), seedling and sapling density was low, but adults increased in density since 2010. Recruitment and survival were impacted by the harsh environmental conditions that prevailed prior to 2013 (low water level and non-tidal influence), but adults survived. The results of the test that determined the percentage of aerenchyma of the pneumatophores indicated that waterlogging stress did not affect the aerenchyma of the pneumatophores. However the period of inundation was probably not significant enough to have affected the production of aerenchyma. The third objective formed part of an ongoing study by the Global Environmental Facility (GEF) on the feasibility of linking the Mfolozi River back to the St Lucia Estuary and the responses of these systems to different management scenarios: 1) the “do nothing” scenario; 2) maintain separate Mfolozi and St Lucia mouths representing an open mouth condition; and 3) actively facilitate a single mouth (therefore linking the Mfolozi and St Lucia mouths). Data gathered on the eco-physiological tolerances of the dominant macrophyte species was used to predict the response of the different habitats to these various management scenarios and the results indicated that the best management scenario would be to actively facilitate a single mouth (Scenario 3) as the estuary habitats would increase significantly because of preferred tidal and saline conditions, as this would represent more natural conditions. The results of the study will provide input to recommendations for future “adaptive management” strategies for the Global Environmental Facility (GEF) Project.

- Description: In the recent years, the shale gas discourse has become central to discussions about future energy supply in South Africa. In particular, the Permian black shales of the Lower Ecca Group formations in the Karoo Basin are considered potential source rocks for shale gas. The research presented in this thesis advances the understanding of the shale gas potential of mainly the Prince Albert, Whitehill and Tierberg/Collingham Formations. These shale sequences were sampled from eight deep boreholes spread across the main Karoo Basin and geochemically analysed at the GFZ - Helmholtz Centre Potsdam, Germany. Three key questions guided the study, these are: (i) what is the lithology of the sequence; (ii) where in the basin do the shale sequences attain maximum thickness at optimum depth i.e. beneath 1000-1500m; and (iii) and their shale characteristics. To evaluate these, borehole core logging, petrology and organic geochemistry were used extensively. Petrology involved the use of thin section, scanned electron and transmission electron microscopy for mineralogy as well as the identification of sedimentary features, organic matter and nano-scale porosity. These were coupled with standard organic geochemistry techniques such as Rock Eval. analysis, open pyrolysis gas chromatography and thermovaporisation to quantify the free gas, total organic carbon (TOC), present-day gas generative potential and kerogen type. The results show that the Whitehill Formation, away from the CFB and not intruded by dolerite, has the most potential for shale gas. Microscopic studies of this pyritic black shale reveal the occurrence of porous amorphous matter, indicating thermal maturity within the gas generation zone (i.e. > 1.1 percent Ro, 120ºC). The TOC content is consistently high within the Whitehill (exceeding industry requirement of 2 percent), attaining maximum of 7.3 percent. The highest yields of free and desorbed gas, especially methane, were emitted within this formation (S1 and nC1 peaks); mostly within its dolomitic units. In addition, dissolution porosity within dolomite units of the Whitehill Formation was identified as the predominant type of porosity. Thus, it is deduced that the dolomitic units of Whitehill Formation potentially contain the greatest volumes of free gas. HI values attain maximum of 25 mg HC/g TOC, whereas the OI values 26 mg CO2/g TOC. Such low HI and OI values are typically attributed to the dominance of Type IV kerogen, and consistent with overmaturity. Open pyrolysis (GC) show the main the chemical compound of the organic matter to be m-p-xylene, consistent with a mix of Type III, Type I/II and Type IV kerogen. Lithologically, the Whitehill Formation is composed of ~ 35 quartz, 13 percent feldspar, 26 percent illite and ~ 23 percent dolomite with variable amounts of pyrite. The dominance of quartz is directly proportional to the brittleness of the rock. Thus it can be deduced that the Whitehill Formation is relatively brittle and therefore fraccable. Burial trends indicate increasing depth (from ground level) to the top of the Whitehill Formation towards the south and south-eastern portion of the basin. It is in the southern region where thicknesses of this black shale exceeding 50m occur at depths more than 1500m; 1000m beneath fresh water aquifers. It therefore concluded that Whitehill Formation in the southern portion of Karoo Basin, but away from the thermo-tectonic overprint of the Cape Orogeny, is the most probable shale gas reservoir in South Africa.

- Description: In South Africa, the limited extent of indigenous forests accelerated the development of fast-growing introduced species plantations, on which the country is now heavily reliant for its fibre and timber products. However, the plantation forestry industry is challenged with limited freshwater resources, the need for sustainable management of introduced tree species in commercial forest production systems, and a diversity of plant ecological production factors such as soils that have to be manipulated for the purposes of increasing production capacities. Additionally, plantations are established in the limited high rainfall regions of the country and the industry’s water use has been regulated since 1972. Conversely, natural forests also provide valuable goods, however, their slow growth rates have restricted their development as commercial tree production systems. In this regard the forestry industry is continuously seeking to provide for the country’s timber and fibre needs while ensuring the provision of other ecosystem services from tree production systems. Recent developments in the forestry industry have therefore been focused on water use efficiency in current and alternative tree production systems. There is also widespread unsubstantiated belief that indigenous tree species are efficient users of water. Against this background this study sought to explore the single tree water use and water use efficiencies of introduced commercial plantation species (Pinus radiata) and that of important indigenous species (Ilex mitis, Ocotea bullata and Podocarpus latifolius) for timber and fibre production in the southern Cape region of South Africa. Single tree water use measurements were carried out for a year using the heat pulse velocity method. Growth measurements for utilisable stem wood were done on a quarterly basis for the specimen trees and growth increments over the year were determined. Single tree water use efficiency was a calculated as a function of grams of stem wood gained per litre of water transpired. Measurements of daily weather conditions and soil water content were concurrently taken during the year. The relationships between the variables that express daily climatic conditions, soil water content and daily volumes of transpired water were explored. Empirical models for the prediction of daily transpiration as a function of climate and soil water content were then developed using multiple linear regression analysis. Pinus radiata trees exhibited higher totals of volumetric transpiration than the indigenous species. The water use patterns of Pinus radiata showed higher peaks of maximum and minimum daily and seasonal water use while the indigenous species showed temperate patterns of water use throughout the year. Podocarpus latifolius attained the lowest water use efficiency in the year (0.49g/L) while Pinus radiata, Ocotea bullata and Ilex mitis had comparable water use efficiency values ranging between 1g/L to 2.50g/L. Significant positive correlations existed between climatic variables and daily sapflow volumes of the tree specimens. Daily total solar radiation showed the highest positive correlation with daily transpiration and the average value of the correlation coefficient for solar radiation and water use for all tree species was 0.70 (p < 0.001). Empirical models for predicting daily water use integrated the variables expressing plant available moisture and energy. The study enhances our understanding of single tree water use, water use efficiency, the drivers of transpiration and the applicability of such studies in developing rapid estimation techniques for water use in current and alternative South African tree production systems.

Spatial and temporal variability in water quality characteristics of the Swartkops Estuary

- Description: Water quality characteristics of the heavily urbanised and industrialised Swartkops River catchment in the Eastern Cape has been the focus of several studies since the 1970s. Overloaded and poorly maintained wastewater treatment works (WWTW), polluted stormwater runoff and solid waste have had a negative impact on the water quality status of the Swartkops River and estuary. Past studies have revealed that a distinct relationship exists between land use activities and the water quality of the Swartkops Estuary, which in turn has raised concerns pertaining to the ecological, economic, recreational, and cultural value of the estuaryThe Swartkops Estuary has a Present Ecological State (PES) of Category D(1) and a Recommended Ecological Category (REC)(2) of a C (Van Niekerk et al., 2014). It is known that effective management of anthropogenic impacts on coastal systems requires a thorough understanding of the system’s biological responses to wastewater discharges and to hydrologic changes. For this reason, the objective of this study was to determine the current water quality status of the Swartkops Estuary, and to gain greater insight into factors controlling eutrophication. This was important as outdated water quality information was used in the Swartkops Integrated Environmental Management Plan (Enviro-Fish Africa, 2011) due to a lack of current data. To determine the current water quality status of the Swartkops Estuary the present study investigated spatial and temporal variability in physico-chemical parameters, nutrients, phytoplankton biomass and community composition, faecal bacteria, and “where possible” related this to historical water quality data. In general, points of entry into the estuary were investigated for their impact on nutrient enrichment and the bacteriological status of the estuary. Water quality surveys were completed in September 2012, November 2012, February 2013, May 2013 and August 2013The present study found evidence to suggest that water is not flushed as efficiently from the estuary as was previously the case, and that the natural hydrology of the estuary has been modified. These changes appear to be the effect of large volumes of wastewater discharges from the wastewater treatment works (WWTW), which has led to the additional stresses of increased vertical stratification and reduced vertical mixing. A build-up of dissolved inorganic nutrients has given rise to persistent eutrophic conditions and phytoplankton blooms occurring from the middle reaches to the tidal limit of the estuary. These findings were associated with a generally well oxygenated estuary; however, bottom water hypoxic conditions were recorded in the upper reaches of the estuary on two occasions and were generally associated with bloom.forming flagellates. Elevated concentrations of inorganic nutrients stimulated phytoplankton to attain high biomass ranging from 0 to 248 g l-1 (31.8 6.56 g l-1). All nutrients displayed positive linear gradients from the mouth to the tidal limit, showed significant (p < 0.05) temporal and spatial variability, and were significantly (p < 0.05) correlated with phytoplankton biomass. Phytoplankton blooms (> 10 000 cells ml-1) of several different groups were recorded from the middle reaches of the estuary to the tidal limit. Diatoms were the dominant group during increased freshwater inflow (at mean daily flow rate of 2.14 m3 s-1) and low DIP levels, whereas flagellates were generally the dominant group during reduced flow and under higher nutrient levels. Although the different tidal stages had no effect on phytoplankton biomass per se, it did support co-existance between phytoplankton groups. This was noted during the spring ebb tide in September 2012 (i.e. flagellates, diatoms and dinoflagellate) and in February 2013 (i.e. dinoflagellates, diatoms and chlorophytes). Phytoplankton blooms have become persistent in the middle to upper reaches of the estuary where chlorophyll-a was > 20 µg l-1 and cell density exceeded 10 000 cells ml-1; a situation not reported in previous studies. The Motherwell Canal was and still is the main source of nitrogen (generally in the form of ammonium) to the estuary, whereas the Swartkops River is still the primary source of phosphorus to the estuary. Since the stormwater canal services the large residential area of Motherwell where leaks in the sewer system, the dumping of night soil buckets, and faulty pumps are often reported, polluted discharges from the Motherwell Canal can enter the canal at any given point. In contrast to the canal, DIP loading from the Swartkops River to the estuary generally occurred under conditions of low flow, whereas nitrogen showed no apparent relationship. Faecal bacteria originating from the Motherwell Canal had the most profound effect on the bacteriological status in the middle reaches of the estuary, whereas the Swartkops River had an intermediate effect due to bacteria die-offs occurring between the point of release from the WWTW to the riverine reaches and the tidal limit of the estuary. Nevertheless, Escherichia coli and enterococci levels are still high, especially in the summer months rendering the estuary unsafe for recreation during this season. Historical data on trace metals in the water column were limited and thus observations from the present study could not be concluded with much confidence. However, preliminary data suggest that levels of copper, zinc, iron and cadmium have increased by at least 90% in the estuary, at the tidal limit of the estuary and in the Markman and Motherwell canals. High inputs of nutrients, trace metals and faecal bacteria to the estuary from land-use activities indicate the necessity for remedial actions with the main objective being to conserve and protect the estuary’s recreational, ecological and economic functions.

- Description: Water quality characteristics of the heavily urbanised and industrialised Swartkops River catchment in the Eastern Cape has been the focus of several studies since the 1970s. Overloaded and poorly maintained wastewater treatment works (WWTW), polluted stormwater runoff and solid waste have had a negative impact on the water quality status of the Swartkops River and estuary. Past studies have revealed that a distinct relationship exists between land use activities and the water quality of the Swartkops Estuary, which in turn has raised concerns pertaining to the ecological, economic, recreational, and cultural value of the estuaryThe Swartkops Estuary has a Present Ecological State (PES) of Category D(1) and a Recommended Ecological Category (REC)(2) of a C (Van Niekerk et al., 2014). It is known that effective management of anthropogenic impacts on coastal systems requires a thorough understanding of the system’s biological responses to wastewater discharges and to hydrologic changes. For this reason, the objective of this study was to determine the current water quality status of the Swartkops Estuary, and to gain greater insight into factors controlling eutrophication. This was important as outdated water quality information was used in the Swartkops Integrated Environmental Management Plan (Enviro-Fish Africa, 2011) due to a lack of current data. To determine the current water quality status of the Swartkops Estuary the present study investigated spatial and temporal variability in physico-chemical parameters, nutrients, phytoplankton biomass and community composition, faecal bacteria, and “where possible” related this to historical water quality data. In general, points of entry into the estuary were investigated for their impact on nutrient enrichment and the bacteriological status of the estuary. Water quality surveys were completed in September 2012, November 2012, February 2013, May 2013 and August 2013The present study found evidence to suggest that water is not flushed as efficiently from the estuary as was previously the case, and that the natural hydrology of the estuary has been modified. These changes appear to be the effect of large volumes of wastewater discharges from the wastewater treatment works (WWTW), which has led to the additional stresses of increased vertical stratification and reduced vertical mixing. A build-up of dissolved inorganic nutrients has given rise to persistent eutrophic conditions and phytoplankton blooms occurring from the middle reaches to the tidal limit of the estuary. These findings were associated with a generally well oxygenated estuary; however, bottom water hypoxic conditions were recorded in the upper reaches of the estuary on two occasions and were generally associated with bloom.forming flagellates. Elevated concentrations of inorganic nutrients stimulated phytoplankton to attain high biomass ranging from 0 to 248 g l-1 (31.8 6.56 g l-1). All nutrients displayed positive linear gradients from the mouth to the tidal limit, showed significant (p < 0.05) temporal and spatial variability, and were significantly (p < 0.05) correlated with phytoplankton biomass. Phytoplankton blooms (> 10 000 cells ml-1) of several different groups were recorded from the middle reaches of the estuary to the tidal limit. Diatoms were the dominant group during increased freshwater inflow (at mean daily flow rate of 2.14 m3 s-1) and low DIP levels, whereas flagellates were generally the dominant group during reduced flow and under higher nutrient levels. Although the different tidal stages had no effect on phytoplankton biomass per se, it did support co-existance between phytoplankton groups. This was noted during the spring ebb tide in September 2012 (i.e. flagellates, diatoms and dinoflagellate) and in February 2013 (i.e. dinoflagellates, diatoms and chlorophytes). Phytoplankton blooms have become persistent in the middle to upper reaches of the estuary where chlorophyll-a was > 20 µg l-1 and cell density exceeded 10 000 cells ml-1; a situation not reported in previous studies. The Motherwell Canal was and still is the main source of nitrogen (generally in the form of ammonium) to the estuary, whereas the Swartkops River is still the primary source of phosphorus to the estuary. Since the stormwater canal services the large residential area of Motherwell where leaks in the sewer system, the dumping of night soil buckets, and faulty pumps are often reported, polluted discharges from the Motherwell Canal can enter the canal at any given point. In contrast to the canal, DIP loading from the Swartkops River to the estuary generally occurred under conditions of low flow, whereas nitrogen showed no apparent relationship. Faecal bacteria originating from the Motherwell Canal had the most profound effect on the bacteriological status in the middle reaches of the estuary, whereas the Swartkops River had an intermediate effect due to bacteria die-offs occurring between the point of release from the WWTW to the riverine reaches and the tidal limit of the estuary. Nevertheless, Escherichia coli and enterococci levels are still high, especially in the summer months rendering the estuary unsafe for recreation during this season. Historical data on trace metals in the water column were limited and thus observations from the present study could not be concluded with much confidence. However, preliminary data suggest that levels of copper, zinc, iron and cadmium have increased by at least 90% in the estuary, at the tidal limit of the estuary and in the Markman and Motherwell canals. Stratigraphic characterisation of the Collingham formation in the context of shale gas from a borehole (SFT 2) near Jansenville, Eastern Cape, South Africa

- Description: This study is an extensive lithological, petrographical, mineralogical and geochemical description of fresh Collingham Formation core samples collected from borehole SFT 2, located on the farm Slangfontein, south of Jansenville in the Eastern Cape, South Africa. The borehole, drilled to 295 m on the northerly limb of a shallow westerly plunging syncline, intersected the lower Ecca Group rocks of the Ripon, Collingham, Whitehill and Prince Albert Formations and terminated in the upper Dwyka Group. A comprehensive log and stratigraphic column were compiled for the Collingham Formation and fresh core samples were analysed using X-Ray Diffraction (“XRD”), X-Ray Fluorescence (“XRF”), mercury porosimetry, and Total Organic Carbon (“TOC”). Thin section microscopy and Scanning Electron Microscopy (“SEM”) analyses were carried out on selected samples of core from borehole SFT 2. The matrix supported, massive to laminated lithological units of the Collingham Formation are interpreted as detrital, terrigenous sediments. These sediments are composed of intercalated fine-grained, poorly sorted, non-fissile mudstone; fine- to very fine-grained, predominantly pyroclastic airfall tephra; and less common fine-grained sandstones. Sediments of the Collingham Formation are considered to be immature, composed primarily of clay and aluminosilicates. The predominance of a clay fraction and aluminosilicates in mudstone samples is indicated by elevated K2O/Al2O3 ratio values, and the relationship of Zr, Al2O3 and TiO2. The presence of glauconite within the Collingham Formation indicates deposition in a mildly alkaline, slightly reducing marine environment. Rb/K ratio values (1.9 – 2.3 x 10-3) indicate brackish to slightly marine conditions, while low Zr/Rb ratio values indicate a low hydro-energy environment, with stable bottom water conditions. Hf and Nb concentrations indicate that detrital input was greatest during the deposition of tuffaceous units; while stable mineral assemblages and a low Fe2O3/K2O ratio values indicate deposition close to the source. A variation in Si/Ca values indicate times when sediments were affected by turbidity, interspersed with times of relative quiescence. The predominance of K2O over Na2O indicates that the Collingham Formation is alkali-rich, while SiO2/Al2O3 ratio values and the relationship of Zr, Al2O3 and TiO2 indicate that sediments are immature. In the lower portion of the formation, non-sulphidic, anoxic conditions are indicated by Mn/Al, V/(V+Ni), V/Cr ratio values, the Fe-Mn- V content, and the correlation between V and TOC. The upper portion of the formation is considered dysoxic, due to the presence and distribution of pyrite framboids, which indicate a fluctuating O2 level, likely indicating deposition at the interface between anoxic and slightly more oxic conditions. V/Cr ratio values indicate that the O2 regime was lowest during the deposition of the mudstones. The Chemical Index of Alteration (“CIA”) indicates a consistent weathering regime throughout the deposition of the Collingham Formation, associated with a temperate climate on the interface between glacial and tropical conditions. Although an anoxic and low hydro-energy environment is generally favourable for hydrocarbon accumulation, the Collingham Formation contains low levels of Total Organic Carbon (well below 0.9 per cent) and low porosities (ranging from 0.35 per cent to a maximum of 2.22 per cent), both of which are characteristic of a poor source for gas accumulation. Due to the laminate nature, permeability and fracturability of the Collingham Formation, there is the potential that the formation may form a good sealing sequence to the potentially gas-rich Whitehill Formation below. The metamorphic impact related to the Cape Orogeny (± 250 Ma), and reflected in the textures of the minerals making up the sediments of the Collingham Formation, suggests the enhancement in the sealing efficiency of this formation.

- Description: The use of natural fibers as reinforcement in thermoplastics presents an interesting alternative for the production of low cost and ecologically friendly composites. One of the advantages of using natural fibres is their low specific weight, resulting in higher specific strength and stiffness when compared to glass reinforced composites. Natural fibres also present safer handling and working conditions. They are non-abrasive to mixing and can contribute to significant cost reduction. This work is divided into two phases: Phase 1 deals with developing nonwoven mats composites from flax/polypropylene (PP) and evaluating their properties. Flax/polypropylene fibres (at different weight ratios) were processed by needle-punching technique in order to form nonwoven mats. The mats were compression-molded at a temperature of 180oC to form composite materials. The mechanical, thermal and viscoelastic properties of the composites were analyzed. Composites (untreated and silane-treated) were also subjected to varying conditions of temperature and humidity and the tensile properties of the conditioned and unconditioned composites were investigated. The mechanical properties (tensile, flexural and impact) of flax/PP composites were found to increase and reach maximum values at 30 per cent fibre loading and then decrease at higher fibre content. Thermal studies revealed that the composites were stable up to 320oC and samples containing 40 per cent flax fibres were found to exhibit greater thermal stability than neat PP. The dynamic mechanical analyses of the composites showed that the incorporation of flax in the composites resulted in an increase of the storage modulus with a maximum value exhibited by composite containing 40 per cent fibre loading. Composites containing chemically modified fibres exhibited low tensile modulus after conditioning. Phase 2 is based on the investigation of the effect of nano-calcium carbonate (CaCO3) on the properties of two types of polymer matrices: recycled PP and virgin PP. In this case, composites were prepared by melt-mixing and injection molding. The mechanical and thermal properties of the composites were characterized. The tensile modulus of the nano-CaCO3 filled PP (virgin and recycled) composites were found to increase and reach maximum at 30 per cent nano-CaCO3 loading, while the tensile strength decreased with increasing filler content. Thermal studies showed that the nano-CaCO3 filled PP samples exhibited a one-step degradation pattern and are thermally stable up to 450oC. The thermal stability of the samples was found to decrease following the addition of nano-CaCO3. SEM micrographs of the tensile fractured surfaces of composites of the nano-CaCO3 filled virgin and recycled PP revealed the presence of nano-CaCO3 agglomeration.

Tetrol and derivatives: synthesis, host-guest properties and racemate resolutions

- Description: In this study, we investigated the potential of a novel chiral host compound (+)-(2R,3R)-1,1,4,4-tetraphenylbutane-1,2,3,4-tetraol (TETROL) and its derivatives for use in racemate resolution using host-guest chemistry. The parent TETROL molecule is composed of a butane chain bearing a hydroxy functionality on each carbon, and two phenyl rings on each of the terminal carbons. The two internal carbon atoms are chiral. The syntheses of TETROL and derivatives were carried out by modifying the diester of naturally-occurring optically active tartaric acid using a variety of aromatic Grignard reagents. These included phenyl, anisyl, tolyl and naphthyl groups, as well as aromatic rings bearing fluoro and trifluoromethyl substitutents. The substituents on the aromatic rings were located in the ortho, meta or para positions. The so-obtained potential host compounds were investigated for their inclusion abilities by recrystallizing them from a number of potential chiral guest compounds such as 2- and 3-methylcyclohexanone, camphor, i-propanol and 2-butanol, as well as various achiral organic compounds. Host:guest ratios were determined by means of 1H-NMR spectroscopy. Of the hosts investigated, TETROL appeared to favour a host:guest ratio of 1:1 for all included compounds. It complexed with cyclic ketones such as cyclohexanone and derivatives, and also cyclic alcohols and amines like cyclohexanol and morpholine. TETROL, however, was not able to include short chain or branched chain alcohols such as i-propanol and 2-butanol. p-AnisylTETROL showed similar inclusion abilities to TETROL but, in addition, enclathrated i-propanol and 2-butanol. p-TolylTETROL showed a preference for the inclusion of alcohols over ketones. X-Ray data of the inclusion complexes indicated that a pair of 1,3-intramolecular hydrogen bonds was a significant stabilizing factor of the geometries of all the hosts. The guest was always held in the host crystal by means of a hydrogen bond with the host, where the host functioned as the hydrogen bond donor and the guest as the acceptor. There were a number of other inter- and intra-molecular contacts that further stabilized the inclusion complexes. A surprising feature of the inclusion of 3-methylcyclohexanone, as elucidated by X-ray analysis, was that its methyl group adopted the axial orientation, the higher energy conformation for these kinds of molecules, and a 3-alkylketone effect was proposed to be one of the reasons for this observation. Thermal data was used to assess the relative thermal stabilities of the complexes, and the results compared with features of the X-ray structures, in order to determine whether thermal stability is related in some way to the nature of the guest packing in the host crystal. the case of the racemic guests, complexes obtained were analysed using chiral GC-MS. TETROL preferred the R-enantiomers of 2- and 3-methylcyclohexanone (21.7% and 16.7% e.e.). The S-enantiomer of camphor was favoured but the e.e. was low (3.8%). p-AnisylTETROL had a preference for the S-enantiomer in the case of 2- and 3-methylcyclohexanone as well as 2-butanol (44.3%, 20.4% and 1.7% e.e., respectively). p-TolylTETROL could only successfully resolve 2-butanol (23.5% e.e. in favour of the R-enantiomer). o-TolylTETROL preferred the R-enantiomers of methyl phenyl sulfoxide (29.2% e.e.) and 2-butanol (21.5% e.e.). Overall, TETROL and its derivatives exhibited the ability to resolve racemic mixtures to some extent.

TETROL, however, was not able to include short chain or branched chain alcohols such as i-propanol and 2-butanol. p-AnisylTETROL showed similar inclusion abilities to TETROL but, in addition, enclathrated i-propanol and 2-butanol. p-TolylTETROL showed a preference for the inclusion of alcohols over ketones. In general, the ortho-substituted aromatic derivatives of TETROL faired relatively poorly as hosts, with some exceptions. X-Ray data of the inclusion complexes indicated that a pair of 1,3-intramolecular hydrogen bonds was a significant stabilizing factor of the geometries of all the hosts. The guest was always held in the host crystal by means of a hydrogen bond with the host, where the host functioned as the hydrogen bond donor and the guest as the acceptor. There were a number of other inter- and intra-molecular contacts that further stabilized the inclusion complexes. A surprising feature of the inclusion of 3-methylcyclohexanone, as elucidated by X-ray analysis, was that its methyl group adopted the axial orientation, the higher energy conformation for these kinds of molecules, and a 3-alkylketone effect was proposed to be one of the reasons for this observation. Thermal data was used to assess the relative thermal stabilities of the complexes, and the results compared with features of the X-ray structures, in order to determine whether thermal stability is related in some way to the nature of the guest packing in the host crystal. the case of the racemic guests, complexes obtained were analysed using chiral GC-MS. TETROL preferred the R-enantiomers of 2- and 3-methylcyclohexanone (21.7% and 16.7% e.e.). The S-enantiomer of camphor was favoured but the e.e. was low (3.8%). p-AnisylTETROL had a preference for the S-enantiomer in the case of 2- and 3-methylcyclohexanone as well as 2-butanol (44.3%, 20.4% and 1.7% e.e., respectively). p-TolylTETROL could only successfully resolve 2-butanol (23.5% e.e. in favour of the R-enantiomer). o-TolylTETROL preferred the R-enantiomers of methyl phenyl sulfoxide (29.2% e.e.) and 2-butanol (21.5% e.e.). Overall, TETROL and its derivatives exhibited the ability to resolve racemic mixtures to some extent.

- Description: Treatments with natural products rich in anti-oxidants have attracted remarkable interest in the cosmetic and pharmaceutical industry to combat oxidative stress and reverse the effects of ageing. Rooibos (Aspalathus linearis) is a South African fynbos plant, well-known for its strong anti-oxidant capacity and use in many cosmetic products. However, little published research exists on its potential as an anti-ageing treatment. The anti-ageing properties of fermented and green rooibos were investigated using an in vitro cell culture model designed to evaluate the involvement of mitochondrial dysfunction in the age related decline in preadipocyte function. Mitochondrial DNA (mtDNA) deficient preadipocytes, ρ0 3T3-L1preadipocytes, were generated following continuous long-term exposure to sub lethal concentrations of ethidium bromide (EtBr). Depletion of the mtDNA resulted in a significantly reduced mitochondrial membrane potential, rate of proliferation in culture, as well as an increased glucose utilization and lactate production. Treatment with the green rooibos (100 μg/mL) stimulated cell growth rates for both the wildtype and mutant cell lines. MtDNA depleted cells showed arrest in the G1 phase (48.8 ± 3.34%) compared to wildtype cells (44.6 ± 1.38%), which was significantly attenuated after treatment with green rooibos for mutant (42.0 ± 0.83%) and wildtype (36.5 ± 5.80%) treated cells. The results obtained for glucose utilization and lactate production, indicated a significant increase in glucose utilization along with a concomitant increase in lactate production after treatment with both green and fermented rooibos for wildtype and mutant cell lines. A significant improvement in mitochondrial membrane potential was also later observed after treatment with green and fermented rooibos on both the wildtype and mutant cell lines. The results obtained indicate that rooibos extracts, particularly the green rooibos, exhibit effects which preserve the functional capacity of preadipocytes exposed to ageing related insults, and indicate that rooibos could cause a metabolic shift in cells redirecting carbon flow away from mitochondrial metabolism, and towards lactate production and consequently, cells become resistant to mitochondrial dysfunction.

The antifungal activity of an aqueous Tulbaghia violacea plant extract against Aspergillus flavus

- Description: Phytochemical analysis of both HEA1 and the crude plant extract showed the presence of phenolics, tannins and saponins. Saponins were the predominant secondary metabolites and were mostly abundant in the plant extract and to a lesser extent in the active compound. Steroidal saponins, tannins and phenolics were also detected in the plant extract, but only the phenolics were detected in the active compound. The results of the phytochemical analysis showed that those compounds that were not present in the active compound could be removed from the crude extract during the TLC purification process. Investigation on the mechanism of action of the crude plant extract on the sterol production by A. flavus showed that the plant extract affected ergosterol biosynthesis by causing an accumulation of oxidosqualene in the ergosterol biosynthetic pathway resulting in a decline in ergosterol production. An oscillatory response in lanosterol production was observed in the presence of the plant extract, which may be an adaptation mechanism of A. flavus to unfavourable conditions and compensation for the loss of enzyme activity which may have occurred as a result of the accumulation of oxidosqualene. The antifungal activity of the plant extract on ergosterol production by A. flavus may also be due to saponins which target the cell membrane and ergosterol production in fungi. The effect of the plant extract on the fungal cell wall of A. flavus also showed that the plant extract caused a decline in β-(1, 3) glucan production by inhibiting β-glucan synthase. The plant extract also affected the chitin synthesis pathway of A. flavus, by causing a decline in chitin production, which was due to the inhibition of chitin synthase. Investigation of chitinase production using 4MU substrates showed that the plant extract caused an accumulation of chitobioses, by activating chitobiosidases and endochitinases. A decline in N-acetylglucosaminidase activity in the presence of the plant extract was observed and this prevented the formation of N-acetylglucosamine. The accumulation of chitobiosidase and endochitinase may be as a result of autolysis that may be triggered by A. flavus as a survival mechanism in the presence of the plant extract and as a compensatory mechanism for the loss of β-glucans and chitin. The antifungal effect of the plant extract on various components of the cell wall of A. flavus, makes T. violacea aqueous plant extract an ideal chemotherapeutic agent against both human and plant pathogens of Aspergillus. The broad spectrum of antifungal activity of T. violacea against A. flavus also eliminates any chances of the fungus developing resistance towards it and would make it a candidate for use as a potential antifungal agent. Further identification and possible chemical synthesis is needed to shed light on the safety and efficacy of the active compound for further development as a chemotherapeutic agent.

- Description: The use of a handheld Raman analyser for the positive identification of raw materials in a manufacturing pharmaceutical company was evaluated using paracetamol as test raw material to evaluate whether such a system would meet Aspen’s regulatory requirements. The approach involved subjecting the chosen raw material to identification tests under a variety of conditions so as to evaluate robustness, and specificity of the system. Thus, raw material provided by different suppliers, different packages of one manufacturing batch, and raw materials subjected to different storage conditions were evaluated. Specificity was evaluated by deliberately contaminating a sample of paracetamol with either acetanilide, or 4-aminophenol, or both at varying concentration levels. The results obtained from these investigations showed that the handheld Raman analyser can correctly identify the selected raw material (paracetamol) under a wide range of conditions, but could not correctly identify the presence of the selected contaminants at lower concentration levels (< 10 – 20 mass percent). Finally, a cost-benefit analysis was carried out in which a scenario of an existing FTIR-ATR system is used for the analysis of a specific number of raw material samples per year as opposed to a scenario in which a new handheld Raman analyser has to be purchased, set up, and used for the analysis of the same number of raw material samples. This comparison showed that the handheld Raman analyser had a pay-back time of approximately 6 months and gave a return on investment of approximately the same value as the actual purchase cost.

The adjusted R2, AIC (Akaike information criterion), the BIC (Bayesian information criterion) and standard deviation of residuals were used to investigate which model is the best to predict energy demand in each province using all available information. The results show that population and household energy demand are associated with each other. The findings imply that the causality between population growth and household energy demand is bidirectional in most cases.
