Beyond mastery: jazz, gender and power in postapartheid South Africa

- Description: This dissertation studies the musical lives of seven South African women who have built highprofile national and international careers as jazz musicians. Empirically it records self-reported experiences and actions that they have identified as relevant to their success within a notoriously patriarchal field. Drawing on feminist research methods and Interpretative Phenomenological Analysis the dissertation strives to develop an empathic yet critical epistemological framework attuned to the complexities of their lived experiences as women and as jazz musicians. Theoretically it investigates the power relations at play in the myriad ways they negotiate or have been impelled to negotiate gender and patriarchy as musicians. Chapter one draws attention to the ubiquity of patriarchy in South African and international jazz culture, and highlights the research participants’ numerous successes to date as performers, educators, composers, and bandleaders. Chapter two positions this study in relation to cognate work in South African jazz studies, international feminist jazz studies and feminist phenomenology. Chapters three to five successively consider the participants’ early enculturation as musical and gendered subjects, their agentic responses to structures of patriarchy and/or race as emerging and established professionals, and the ways these experiences have found expression in some of their musical utterances as performers and composers. Despite the intentions of the government to promote diversity, social transformation, and inclusion, subject music is still weighted towards Western Art Music, perpetuating past biases of a Eurocentric model for music education. This study, which uses a mixed-method approach, seeks to create strategies to advance Jazz education in South Africa through an a/r/tographic analysis of the author’s Jazz pedagogical methods at Durban High School in KwaZulu Natal, South Africa. Key approaches include curriculum theory, currere, a/r/tography, among others. After reviewing Jazz pedagogy in the U.S.A. and South Africa as a starting point, using Pinar’s (1994) method of currere, the author demonstrates how past personal music educational processes have led to present Jazz pedagogical methods. Six original compositions were written that outline the influences that infused these methods and added to the creative output related to this research. Interviews were conducted with key stakeholders in the Jazz community to gain perspective about the current state of Jazz education, and with students to provide insight into their reactions to the Jazz stream of CAPS. This presents new information about the curriculum from a learners’ perspective. Ethical clearance for research with children was sought and received (Appendix A). Findings show that the ability of learners to recognise the value in, and identity relating to, their African culture through Jazz points to a pivotal departure from previous Eurocentric music education models. However, despite South Africa being positioned as one of the only countries offering Jazz as a subject choice to high school learners, the selection of Jazz in the subject of Music remains underutilised. With dreams in our hands: Towards transgressive knowledge-making cultures

- Description: Knowledge-making in universities is not neutral and takes different forms. This thesis critically examines the politics of knowledge to propose and present a transgressive schema for knowledge-making that is co-created with students. It emerges from teaching and learning encounters in the Humanities Extended Studies (ES) Programme at Rhodes University, where for the past decade we have experimented with different ways of knowledge-making that run counter to conventional pedagogic practices. We set up a project for the thesis that allowed us to work with knowledge in ways that are Afrocentric, and that hold and nurture our dreams. The theory and methodology of the project are explained in the first academic paper for this PhD by publication. The project and its derivatives use an African Feminist framing, and centre the ontoepistemologies of African young people who find themselves alienated and marginalised by a western bias in university curricula. Former ES student volunteers came up with topics, responded to them, reviewed each other’s work, and co-wrote two academic papers that demonstrate a praxis of African Feminist research and pedagogic principles. Two further projects practise the principles that emerge from the primary project, and together they have tested knowledge-making cultures that inspire critical thinking and creative humanity. These are explained in two further academic papers. One is co-written with the copresenter of an online inter-continental short course for PhD students on African Feminist Research Methodology. The other is single authored, and introduces the third project, a Political and International Studies third-year course on African Feminist theory. The schema for knowledge-making uses the hand, which holds our dream, as a descriptive metaphor. Each of the five fingers of the hand represents an aspect of how we have collaborated on the projects and in lecture rooms, and what this has taught us about how to nurture and inspire the dreams of young African people through transgressive knowledgemaking cultures. The development of ionic zinc(II) phthalocyanines for sono-photodynamic combination therapy of cervical and breast cancer

- Description: This study focuses on the development of the sono-photodynamic combination therapy (SPDT) activity of phthalocyanines (Pcs) on the cervical and breast cancer cell lines in vitro. The SPDT technique utilizes ultrasound in combination with light to elicit cytotoxic effects for cancer eradication. In this work, a selection of tetra-peripherally substituted Zn(II) cationic and zwitterionic Pcs were prepared. The photophysical parameters of the Pcs were determined including their fluorescence behaviours and efficiency of the triplet excited state population. The effects of the ultrasonic parameters (frequencies (MHz) and power (W.cm-2)) on the stability of the Pcs were evaluated. Four parameters were evaluated: Par I (1 MHz: 1 W.cm-2), Par II (1 MHz: 2 W.cm-2), Par III (3 MHz: 1 W.cm-2) and Par IV (3 MHz: 2 W.cm-2). The stability of the Pcs reduced with the increase in the ultrasonic power (for Par II and Par IV). The Par I showed the least degradation compared to the other parameters and was therefore used for the SPDT treatments. The sonodynamic (SDT), photodynamic (PDT) therapy activities of the Pcs were studied and compared to their SPDT efficacies. The Pcs showed reactive oxygen species generation during the SDT, PDT and SPDT treatments. For the SDT and SPDT, singlet oxygen (1O2) and hydroxyl radicals (•OH) were detected. For PDT, only the 1O2 were detected. The cell cytotoxicity studies for the Pcs showed relatively higher therapeutic efficacies for the SDT treatments compared to the PDT treatments, where the SPDT showed higher therapeutic efficacies compared to both the SDT and PDT monotreatments on both the cell lines in vitro. Overall, the combination treatments were better compared to the monotreatments. - Description: Unemployment is a long-standing and pressing socio-economic phenomenon that affects, markedly, both developed and developing countries. Although, in one way or another, many people are affected by unemployment, the reviewed literature concurs that youth unemployment is a critical component of the overall unemployment challenge. Indeed, the burden of unemployment is borne, tremendously, by young people, especially those who are Not in Education, Employment, or Training (NEETs). The International Labour Organisation (ILO) has declared youth unemployment a global crisis, with approximately 67,6 million unemployed young people worldwide in 2018. In South Africa, the youth unemployment rate escalated to 56.4 per cent in the second quarter of 2019. This disturbing level of unemployment among young people poses a serious threat—for as long as young people stay unemployed. Indeed, the cost of unemployment is too high, not only does it engender discontent among young people who are floundering in poverty and frustration, but it can also trigger or perpetuate all sorts of social, political, and economic effects. Owing to this grave and perpetual challenge of unemployment, exploring and understanding the dynamics of youth unemployment continues to be of paramount importance worldwide. Inspired by the claims of LMS theories, this study elected to explore demand-side factors by applying qualitative research methods and techniques to gather and analyse the data gathered, and to ultimately address the research objectives. In particular, non-probability sampling was used to sample 20 firms from four sectors (manufacturing, retail/hospitality, finance, transportation) in the Johannesburg (CBD) region. The respondents were all asked a series of semi-structured questions in order to yield qualitative results that would help determine the employers’ dispositions towards young people; and about the government’s strategies that address youth unemployment. The aim is to provide an in-depth, nuanced understanding of how the respondents’ dispositions influence the inclusion or exclusion of young people in the employers’ recruitment considerations. The aggregate results of the study generally indicate that—although there are innumerable factors that affect young peoples' employment prospects—young people (without skills and tertiary education or matric) are actually being employed, and are still being considered for employment by the majority of the sampled employers (predominantly from retail/hospitality firms). Only a minority of employers stated, categorically, that they do not employ young people, and do not intend to do so. Conversely, the results also reveal that the the majority of employers often recruit young people for general and entry-level jobs that are often offered on a non-standard employment basis. At face value, the employment of young people (without skills) in the majority of the sampled firms may seem to make a difference—and it probably does, for as long as young people are exposed to the world of work. However, in the 4 grand scheme of things, the kind of employment offered in these firms is also problematic, because it is, basically, transient and unsustainable. As such, the young people that occupy these jobs are most likely to find themselves back in the unemployment queues whenever the ‘part-time’ or ‘temporary’ employment contracts are terminated. These jobs are especially problematic because policies such as the National Development Plan (NDP) 2030—in line with Sustainable Development Goals—aim to reduce poverty by creating sustainable employment. Contrarily, these kinds of jobs make it hard to achieve poverty reduction, because they are inherently unsustainable and barely lift people (especially young people) from unemployment and poverty. Based on these results, it can be asserted that the proliferation of non-standard employment compromises the agenda for decent work, and it delays the already sluggish Sustainable Development goal of creating sustainable employment (among other goals). This, then, reveals that there is still a stark and long-standing need for more radical strategies that will ensure the creation of sustainable jobs—without which the unemployment crisis will be far from over. Moreover, the results further indicate that the reasons why the minority of the sampled employers do not employ young people boil down to the fact that young people usually do not have what employers look for in their potential recruits; or that young people do not have what employers believe will contribute to the growth and productivity of their firms. In essence, employers will never employ any young person (or any jobseeker in general) that will be a liability than an asset in their firms. Over and above, this thesis corroborates many of the explanations—encapsulated in the growing literature—for rising unemployment, as well as the claims that unemployment is highly pronounced among young people. - Description: Unemployment is a long-standing and pressing socio-economic phenomenon that affects, markedly, both developed and developing countries. Although, in one way or another, many people are affected by unemployment, the reviewed literature concurs that youth unemployment is a critical component of the overall unemployment challenge. Indeed, the burden of unemployment is borne, tremendously, by young people, especially those who are Not in Education, Employment, or Training (NEETs). The International Labour Organisation (ILO) has declared youth unemployment a global crisis, with approximately 67,6 million unemployed young people worldwide in 2018. In South Africa, the youth unemployment rate escalated to 56.4 per cent in the second quarter of 2019. This disturbing level of unemployment among young people poses a serious threat—for as long as young people stay unemployed. Indeed, the cost of unemployment is too high, not only does it engender discontent among young people who are floundering in poverty and frustration, but it can also trigger or perpetuate all sorts of social, political, and economic effects. - Description: This study is a socio-political aspect of Zimbabwean history. It examines the development of youth political violence starting from the late 1950s when violent forms of African political mobilisation emerged to 2018 when the first election without Robert Mugabe was held. It explores how early nationalist parties such as the Salisbury City Youth League (SCYL), Southern Rhodesia African National Congress (SRANC), National Democratic Party (NDP), Zimbabwe African People’s Union (ZAPU) and later the Zimbabwe African National Union (ZANU) mobilised and socialised youths into political violence to understand the roots of the violent political culture in Zimbabwe. This study shows that youths were an important part of the strategies of these political parties in countering the violence of the colonial state as well as mobilising mass support for the movements during the liberation struggle. It reveals that war collaborators (mujibhas and chimbwidos) were central role players in instigating political violence against innocent and defenceless people during the war. In the 1980s and 1990s, the Youth brigades and the ZANU-PF Youth League became a key constituent for state-socialist developmental goals but they were at times manipulated as a resource for political violence when Mugabe’s power was challenged. The study shows that more grotesque violence occurred in the 2000s era when the National Youth Service (NYS) was introduced and state-sanctioned vigilante groups like Chipangano in Mbare emerged in response to the rise of the Movement for Democratic Change (MDC) and diminishing of consensual power. This study argues that youth were not mere victims and perpetrators of political violence, but they were a collection of various interest sub-groups with diverse agendas and a sense of agency. Some joined violent groups for their social mobility, power, impunity and economic opportunities availed to the group members. Synthesis and evaluation of the medicinal potential of novel 4-hydroxycoumarin derivatives

The compounds were designed to contain additional pharmachophoric centres to enhance bioactivity and generate lead compounds with dualaction potential. The use of 4-hydroxycoumarin as the primary synthon enabled access to various series of 4-hydroxycoumarin conjugates, the reactive 3-position on the 4-hydroxycoumarin moiety being exploited for regioselective construction of the targeted compounds in several steps. Some of the reactants required in the construction of these compounds were specially synthesised and included propargyloxy benzaldehydes, benzyloxy benzaldehydes and 2,3-dihydroxysuccino-dihydride. Overall, eight different families of novel compounds were accessed, comprising conjugates of 4-hydroxycoumarin with bisethylidenesuccinohyrazide, trifluoroacetamide, amino, benzyloxyphenyl-iminoethyl, benzylidenehyrazinyl-thiazoyl, benzylidenehydrazonoethyl, propargyloxybenzylidenehydrazonoethyl and phenylacryloyl moieties using protocols that required minimal work-up and purification. The eighty novel compounds synthesised in the study were fully characterised using HMRS and advanced NMR techniques. Cytotoxicity, HIV-1 IN and PR inhibitory, and antitrypanosomal, antimalarial and anti-Mtb assays were conducted on the synthesised coumarin derivatives. Several compounds exhibited activity against HIV-1 IN, the most potent being a bis-ethylidenesuccinohyrazide with an IC50 value of 3.5 μM. Various compounds exhibited anti-malarial activity (% pLDH viability in the range 62-77%), anti-trypanosomal activity (the most potent with an IC50 = 0.9 μM against T.b. brucei) and a measure of anti-Mtb activity. Apart from two chalconyl derivatives, none of the synthesised compounds exhibited significant cytotoxicity. Adsorption of antibiotic-resistant bacteria and their cell-free deoxyribonucleic acid harbouring resistance genes in drinking water with metal oxides

The incidence of Antibiotic Resistant Bacteria ARB in drinking water and subsequent proliferation of Antibiotic Resistance Genes ARGs in drinking water is a concern for public health. The free DNA cassette harboring antibiotic resistance gene in drinking water has been linked with impaired public health. The ARGs allows bacteria to develop resistance towards antibiotics while ARB render antibiotics ineffective. Existing drinking water treatment technologies eg adsorption, ozonation and chlorination, have shown different levels of potency in the removal of conventional water pollutant. These technologies, which operating principles is based on oxidation or mass transfer, have been explored to gained an insight into their efficiency in the removal of ARB ARGs in water treatment. In general, methods that functions by mass transfer of the ARB ARGs is more effectively than bacteria oxidation. Consequently, adsorption technology was chosen using common metal oxide adsorbents. The adsorbents ZnOAg CeO2 and Al2O3 were synthesized via the self propagation combustion method. This method was selected because of the purity of the final product and the energy requirement. One of the challenges in the removal of ARB in drinking water is the release of ARGs. To address this concern, five different ZnOAg heterostructures were synthesized for the removal of Enterococci faecium. ZnOAg was chosen because of the bactericidal and bacteriostatic characteristics. The study revealed that the concentration of the precursors influences the microstructures of the adsorbents; however, it did not significantly affect the adsorption efficiency. The maximum adsorption capacity q34.11 CFUg was obtained for Ag1Zn3.5. The kinetic studies revealed that Ag1Zn1 and Ag1Zn2 adsorbents agreed to the pseudofirst-order kinetic equation and adsorbents Ag2Zn1 Ag3.5Zn1 and Ag1Zn3.5 agreed to the pseudo-second-order kinetic equation. Initial tap-water pH range was beneficial for the adsorption and the pH of the treated tap-water was within the WHO tap water recommendation 6.5 – 8.5 whereas the effect of ionic strength, anionic and cationic interference was insignificant in the adsorption of MDREF onto the different heterostructure. Interestingly the MDREF could retain its cell membrane integrity and resistance genes, suggesting that surface adsorption was the primary mechanism for the removal. Cerium IV oxide CeO2 was selected because of high adsorption towards phosphate, backbone base for DNA. To prevent the problem of antibiotic resistance, we have synthesized a CeO2 adsorbent that exhibit highly positive character in a wide pH range, via the simple self-propagation combustion protocol, for the removal of free DNA harboring antibiotic resistance genes. Molecular characterization of the extracted genes showed that the sizes for E. coli and inherent gyrB genes are 147 and 460 bp with a purity between 19 2.0. The XRD SEM TEM, and PZC results of the as-synthesized CeO2 showed an agglomerate of pure cubic-faced centered material and highly crystalline, with a net charge at pH 6.2. Experimental results revealed that the reaction proceeded via pseudo first-order kinetic, and it is governed by electrostatic attraction. The free- DNA solution pH electrolyte, and competing ions impacted on the adsorption process. Further experimental results showed that the as-synthesized CeO2 adsorbent has the potential to be used for the removal of free DNA harboring ARGs from tap-water even under oxic conditions. Alumina Al2O3 is an abundant adsorbent that has also shown high removal capacity towards phosphate. The highly pure synthesized Al2O3 adsorbent exhibit fluid-like behaviour under Scanning Electron Microscope SEM. The XRD pattern corresponds to αAl2O3. The adsorption kinetics was described by pseudo second orderadsorption capacity 11.7 μgg implying chemisorption, which agrees with the electrostatic force of attraction caused by opposing ions. This result was evident by the effect of different ions in the tap water. Existing drinking water treatment technologies eg adsorption, ozonation and chlorination, have shown different levels of potency in the removal of conventional water pollutant. These technologies, which operating principles is based on oxidation or mass transfer, have been explored to gained an insight into their efficiency in the removal of ARB ARGs in water treatment. In general, methods that functions by mass transfer of the ARB ARGs is more effectively than bacteria oxidation. Consequently, adsorption technology was chosen using common metal oxide adsorbents. The adsorbents ZnOAg CeO2 and Al2O3 were synthesized via the self propagation combustion method. This method was selected because of the purity of the final product and the energy requirement. One of the challenges in the removal of ARB in drinking water is the release of ARGs. To address this concern, five different ZnOAg heterostructures were synthesized for the removal of Enterococci faecium. ZnOAg was chosen because of the bactericidal and bacteriostatic characteristics. The study revealed that the concentration of the precursors influences the microstructures of the adsorbents; however, it did not significantly affect the adsorption efficiency. The maximum adsorption capacity q34.11 CFUg was obtained for Ag1Zn3.5. The kinetic studies revealed that Ag1Zn1 and Ag1Zn2 adsorbents agreed to the pseudofirst-order kinetic equation and adsorbents Ag2Zn1 Ag3.5Zn1 and Ag1Zn3.5 agreed to the pseudo-second-order kinetic equation. Initial tap-water pH range was beneficial for the adsorption and the pH of the treated tap-water was within the WHO tap water recommendation 6.5 – 8.5 whereas the effect of ionic strength, anionic and cationic interference was insignificant in the adsorption of MDREF onto the different heterostructure. Interestingly the MDREF could retain its cell membrane integrity and resistance genes, suggesting that surface adsorption was the primary mechanism for the removal. Cerium IV oxide CeO2 was selected because of high adsorption towards phosphate, backbone base for DNA. To prevent the problem of antibiotic resistance, we have synthesized a CeO2 adsorbent that exhibit highly positive character in a wide pH range, via the simple self-propagation combustion protocol, for the removal of free DNA harboring antibiotic resistance genes. Molecular characterization of the extracted genes showed that the sizes for E. coli and inherent gyrB genes are 147 and 460 bp with a purity between 19 2.0. The XRD SEM TEM, and PZC results of the as-synthesized CeO2 showed an agglomerate of pure cubic-faced centered material and highly crystalline, with a net charge at pH 6.2. Experimental results revealed that the reaction proceeded via pseudo first-order kinetic, and it is governed by electrostatic attraction. The free- DNA solution pH electrolyte, and competing ions impacted on the adsorption process. Further experimental results showed that the as-synthesized CeO2 adsorbent has the potential to be used for the removal of free DNA harboring ARGs from tap-water even under oxic conditions. Alumina Al2O3 is an abundant adsorbent that has also shown high removal capacity towards phosphate. The highly pure synthesized Al2O3 adsorbent exhibit fluid-like behaviour under Scanning Electron Microscope SEM. The XRD pattern corresponds to αAl2O3. The adsorption kinetics was described by pseudo second orderadsorption capacity 11.7 μgg implying chemisorption, which agrees with the electrostatic force of attraction caused by opposing ions. This result was evident by the effect of different ions in the tap water. SA EO-based formulations showed high antibacterial activity across all bacterial strains, followed by SA-based formulations compared to CMC-based formulations, which exhibited moderate antibacterial activity. Moreover, the prepared gels showed good cytocompatibility, promoted cell proliferation, and exhibited 80 percent wound closure on day 3 compared to the untreated group, which showed a 38 percent wound reduction in vitro. Excellent blood clotting properties were observed with CMC-based gels compared to other formulations. However, all the prepared formulations exhibited outstanding blood clotting ability compared to the control, showing that they can promote rapid blood coagulation. The occurrence of high concentrations being identified in wastewater treatment plants, rivers, etc is due to untreated effluents being discharged from households, hospitals, agriculture, and pharmaceutical industries. The application of adequate treatment techniques and material for the removal of bacteria DNA conveying ARGs from the effluents before their release to the environment cannot be overemphasized. Adsorption techniques seem to be effective due to their easy design, operation, and ability to regenerate adsorbents for use without producing toxic by-products. This concept was employed for the removal of bacteria DNA conveying ARGs from simulated aqueous solution, effluents from hospital, river and WWTPs using silver and silica metallic nanoparticles. This thesis investigated the effectiveness of metallic nanoparticles containing silver AgNPs and mesoporous silica nanoparticles MSNPs as well as magnetite Fe3O4 functionalized with 4 4hydroxyphenyl 2 262-terpyridine onto their surface, for the removal of bacteria DNA conveying antibiotic resistance genes from water samples from hospitals, river, and wastewater treatment plants WWTPs. Silver nanoparticles AgNPs of different molar concentrations 0.1M, 0.5M and 1.0 M and mesoporous silica nanoparticles MSNPs adsorbents were successfully synthesized in their original states and surface functionalization achieved by incorporating magnetite Fe3O4 and 4 4 hydroxyphenyl 2 2 6 2 terpyridine on the silver AgNPs Fe3O4 and silica MSNPs TPPY surfaces respectively. Their effectiveness as adsorbent for the removal of bacteria DNA conveying ARGs from aqueous solutions and real water/wastewater samples were investigated. The DNA uptake by the as-synthesized AgNPs and MSNPs were compared to the functionalized AgNPs Fe3O4 and MSNPsTPPY by determining the adsorbents with the highest removal efficiencies. All as synthesized and functionalized adsorbents were characterized by SEM, EDX, FTIR, XRD, UV spectroscopy and PZC before the removal process. The extraction of genomic DNA from antibiotic-resistant Enterococcus faecium and Vibrio parahaemolyticus was successfully achieved via the boiling method. Antibiotic susceptibility test was conducted using the disk diffusion method before the commencement of genomic DNA extraction. Molecular characterization via gel electrophoresis confirmed the presence of resistance genes at different base pairs. Adsorption batch experiment were investigated, and the best optimum parameters were evaluated through the influence of pH, contact time, initial DNA concentration, adsorbent dose, and competitive ions for each sorption process. The rate determining step were determined by fitting kinetic models such as Natarajan and Khalaf first order, pseudo first order, pseudo second order, Elovich model to experimental data. Also, the adsorption mechanisms determining adsorption equilibrium were investigated by fitting Freundlich, Langmuir and Sips model into the experimental data. The application of AgNPsFe3O4 nanocomposite and MSNPsTPPY for the removal of bacteria DNA demonstrated much enhancement for DNA uptake than the as-synthesized AgNPs and MSNPs materials. The incorporation of magnetite and 4 4hydroxyphenyl 2 2 6 2-terpyridine onto AgNPs and MSNPs significantly enhanced the binding affinity towards the removal the bacteria DNA via strong electrostatic attraction between the active sites on the adsorbent and the negative DNA molecules. Finally, high adsorption capacities were recorded with AgNPsFe3O4 nanocomposite and MSNPsTPPY compared to AgNPs and MSNPs with chaotropic salts. The kinetic adsorption models were mostly best fitted by the pseudo-second order and Elovich models while the adsorption equilibrium was best described by Langmuir and Sips isotherm models. MSNPs with different chaotropic salts, AgNPsFe3O4 nanocomposite and MSNPsTPPY also proved its effectiveness in DNA removal not only in the simulated aqueous solution but in three different real life water samples obtained from Cofimvaba hospital, Ndevana river and Uitenhage WWTPs. High adsorption efficiencies above 90 percent were achieved during the removal of DNA in all the three real water samples. Preparation, characterization, and in vitro evaluation of polymer-based wound dressings for the management of chronic wounds

- Description: Microbial infections are responsible for the retarded recovery process of chronic wounds. Polymer-based scaffolds possess features suitable for the treatment of chronic injuries. However, these scaffolds are commonly encapsulated with therapeutic agents to enhance their biological activities, including antibacterial efficacy. In this research, two types of polymer-based scaffolds were formulated and evaluated as effective formulations for the treatment of chronic wounds: sponges and topical gels. Sponges were formulated from cross-linking of gelatin and PEG. Ag nanoparticles and metronidazole were incorporated into the sponges to improve their antibacterial activity. Topical gels were loaded with essential oils and Ag nanoparticles and prepared from CMC and poloxamer. The prepared sponges and topical gels were evaluated using various analysis and characterization techniques. SEM/EDX, FTIR, and TGA were employed to characterize gelatin/PEG hybrid sponges followed by porosity, in vitro biodegradability, cytotoxicity, and antibacterial studies. FTIR, SEM/EDX, and TGA confirmed their physicochemical properties and successful fabrication of sponges loaded with metronidazole and Ag nanoparticles. The sponges were biodegradable, indicating their capability to induce skin regeneration. The drug release studies showed a rapid release of metronidazole 28.32-71.97 percent from the sponges over the first hour, followed by a sustained drug release. The Ag nanoparticles were released in a sustained manner, suggesting that these sponges can rapidly destroy bacteria and inhibit persisting bacterial infections as well as protect the lesion bed from further bacteria infections. The in vitro antibacterial studies of sponges displayed superior antibacterial activity against most of the Gram-negative and Gram-positive bacteria strains commonly found in chronic wound infections with a MIC value of 15.625 μg/mL. In vitro cytotoxicity experiments revealed excellent biocompatibility with a percent cell viability of more than 70 percent. The in vitro wound scratch healing assay exhibited that the sponges encapsulated with only metronidazole promoted high cell migration than the dual drug-loaded sponges and untreated cells, suggesting its potential to quicken the wound healing process. CMC/Poloxamer topical gels were also characterized by FTIR, followed by pH, viscosity, spreadability, cytotoxicity, and antibacterial studies. FTIR showed successful preparation of CMC/Poloxamer topical gels loaded with essential oils and Ag nanoparticles. - Description: Ursolic acid UA, a pentacyclic triterpenoid that is commonly found in many medicinal herbs and fruits, has been identified as a potential source of therapeutic agents because of its potent biological effects, which include its potential anticancer and antimicrobial activities. However, its limited solubility, rapid metabolism and poor bioavailability inhibit its clinical applications. Numerous UA derivatives have been prepared over the past years in an effort to mitigate the drawbacks associated with UA, as new chemical entities for the treatment of various infections. There is very little progress in the discovery of efficient UA derivatives. In this study, a class of ester and amide-linked ursolic acid-based hybrid compounds fused with selected pharmaceutical scaffolds were successfully synthesized using amidation and esterification reactions and tested for antibacterial and cytotoxicity activities. Hybridizing UA with other known pharmaceutical scaffolds has the potential of overcoming its drawbacks. FT-IR, Mass Spectroscopy, and 1H13C-NMR spectroscopy were used to confirm the structures of the synthesized hybrid compounds. Among the tested ester-linked hybrid compounds in Chapter three, compounds 3.14-3.19,3.21, 3.34, 3.31, and 3.30 demonstrated significant antibacterial activities against some tested bacteria, with MIC values of 15.625 μgml. Furthermore, the in vitro cytotoxicity of these hybrids was determined using the MTT assay against three human tumor cell lines MCF7, MDA-MB-231, and HeLa cells. Compounds 3.19 and 3.34 were found to have better cytotoxic activity when compared to ursolic acid, with IC50 values of 46.99 and 48.18 μg ml respectively. Both compounds revealed more promising docking results, presenting favourable binding interactions as well as better docking energy against the MCF 7 protein target compared to the parent compound ursolic acid. In Chapter Four, among the tested amide-linked hybrid compounds, Compounds 4.17 and 4.24 demonstrated significant antibacterial activity against the majority of bacterial strains with MIC values of 15.625 gml. Compound 4.24 exhibited a MIC value of 15.625gmL against BS, SA, PV, KO, PM, and EC. Compound 4.23 was more cytotoxic to HeLa cells than ursolic acid. Furthermore, molecular docking calculations revealed that compound 4.16 strongly binds to the protein epidermal growth factor receptor while e compounds 4.17 and 4.24 showed a strong binding affinity for the methionyl-tRNA synthetase. - Description: Ursolic acid UA, a pentacyclic triterpenoid that is commonly found in many medicinal herbs and fruits, has been identified as a potential source of therapeutic agents because of its potent biological effects, which include its potential anticancer and antimicrobial activities. However, its limited solubility, rapid metabolism and poor bioavailability inhibit its clinical applications. Numerous UA derivatives have been prepared over the past years in an effort to mitigate the drawbacks associated with UA, as new chemical entities for the treatment of various infections. There is very little progress in the discovery of efficient UA derivatives. In this study, a class of ester and amide-linked ursolic acid-based hybrid compounds fused with selected pharmaceutical scaffolds were successfully synthesized using amidation and esterification reactions and tested for antibacterial and cytotoxicity activities. Hybridizing UA with other known pharmaceutical scaffolds has the potential of overcoming its drawbacks. FT-IR, Mass Spectroscopy, and 1H13C-NMR spectroscopy were used to confirm the structures of the synthesized hybrid compounds.
