- Description: The prevalence of Search Engine Poisoning in trending topics and popular search terms on the web within search engines is investigated. Search Engine Poisoning is the act of manipulating search engines in order to display search results from websites infected with malware. Research done between February and August 2012, using both manual and automated techniques, shows us how easily the criminal element manages to insert malicious content into web pages related to popular search terms within search engines. In order to provide the reader with a clear overview and understanding of the motives and the methods of the operators of Search Engine Poisoning campaigns, an in-depth review of automated and semi-automated web exploit kits is done, as well as looking into the motives for running these campaigns. Three high profile case studies are examined, and the various Search Engine Poisoning campaigns associated with these case studies are discussed in detail to the reader. From February to August 2012, data was collected from the top trending topics on Google’s search engine along with the top listed sites related to these topics, and then passed through various automated tools to discover if these results have been infiltrated by the operators of Search Engine Poisoning campaings, and the results of these automated scans are then discussed in detail. During the research period, manual searching for Search Engine Poisoning campaigns was also done, using high profile news events and popular search terms. These results are analysed in detail to determine the methods of attack, the purpose of the attack and the parties behind it

Stratigraphic characterisation of the Collingham formation in the context of shale gas from a borehole (SFT 2) near Jansenville, Eastern Cape, South Africa

- Description: This study is an extensive lithological, petrographical, mineralogical and geochemical description of fresh Collingham Formation core samples collected from borehole SFT 2, located on the farm Slangfontein, south of Jansenville in the Eastern Cape, South Africa. The borehole, drilled to 295 m on the northerly limb of a shallow westerly plunging syncline, intersected the lower Ecca Group rocks of the Ripon, Collingham, Whitehill and Prince Albert Formations and terminated in the upper Dwyka Group. A comprehensive log and stratigraphic column were compiled for the Collingham Formation and fresh core samples were analysed using X-Ray Diffraction (“XRD”), X-Ray Fluorescence (“XRF”), mercury porosimetry, and Total Organic Carbon (“TOC”). Thin section microscopy and Scanning Electron Microscopy (“SEM”) analyses were carried out on selected samples of core from borehole SFT 2. The matrix supported, massive to laminated lithological units of the Collingham Formation are interpreted as detrital, terrigenous sediments. These sediments are composed of intercalated fine-grained, poorly sorted, non-fissile mudstone; fine- to very fine-grained, predominantly pyroclastic airfall tephra; and less common fine-grained sandstones. Sediments of the Collingham Formation are considered to be immature, composed primarily of clay and aluminosilicates. The predominance of a clay fraction and aluminosilicates in mudstone samples is indicated by elevated K2O/Al2O3 ratio values, and the relationship of Zr, Al2O3 and TiO2. The presence of glauconite within the Collingham Formation indicates deposition in a mildly alkaline, slightly reducing marine environment. Rb/K ratio values (1.9 – 2.3 x 10-3) indicate brackish to slightly marine conditions, while low Zr/Rb ratio values indicate a low hydro-energy environment, with stable bottom water conditions. Hf and Nb concentrations indicate that detrital input was greatest during the deposition of tuffaceous units; while stable mineral assemblages and a low Fe2O3/K2O ratio values indicate deposition close to the source. A variation in Si/Ca values indicate times when sediments were affected by turbidity, interspersed with times of relative quiescence. The predominance of K2O over Na2O indicates that the Collingham Formation is alkali-rich, while SiO2/Al2O3 ratio values and the relationship of Zr, Al2O3 and TiO2 indicate that sediments are immature. In the lower portion of the formation, non-sulphidic, anoxic conditions are indicated by Mn/Al, V/(V+Ni), V/Cr ratio values, the Fe-Mn- V content, and the correlation between V and TOC. The upper portion of the formation is considered dysoxic, due to the presence and distribution of pyrite framboids, which indicate a fluctuating O2 level, likely indicating deposition at the interface between anoxic and slightly more oxic conditions. V/Cr ratio values indicate that the O2 regime was lowest during the deposition of the mudstones. The Chemical Index of Alteration (“CIA”) indicates a consistent weathering regime throughout the deposition of the Collingham Formation, associated with a temperate climate on the interface between glacial and tropical conditions. Although an anoxic and low hydro-energy environment is generally favourable for hydrocarbon accumulation, the Collingham Formation contains low levels of Total Organic Carbon (well below 0.9 per cent) and low porosities (ranging from 0.35 per cent to a maximum of 2.22 per cent), both of which are characteristic of a poor source for gas accumulation. Due to the laminate nature, permeability and fracturability of the Collingham Formation, there is the potential that the formation may form a good sealing sequence to the potentially gas-rich Whitehill Formation below. The metamorphic impact related to the Cape Orogeny (± 250 Ma), and reflected in the textures of the minerals making up the sediments of the Collingham Formation, suggests the enhancement in the sealing efficiency of this formation.

Cleavage of the precursor coat protein of black beetle virus strain w17 in rabbit reticulocyte lysate

- Description: Black beetle virus (BBV) is a bipartite single-stranded RNA virus belonging to the family Nodaviridae. Its host range has been found to be limited to insects. RNA 1, the larger of the two RNA molecules, with a MW of 1,15 x 10⁶ and the smaller RNA 2 with a MW of 0,46 x 10⁶, are both packaged in the same virus particle. The two RNA molecules are translated separately, with RNA 1 coding for protein A of MW 105 x 10³ and RNA 2 coding for protein α of MW 47 x 10³. Protein α is the major capsid protein precursor, which during in vivo maturation is cleaved to form the coat protein β of MW 43 x 10³, and protein γ of MW 5 x 10³. Cell-free translation of BBV (strain W17) mRNA was carried out in rabbit reticulocyte lysates. Protein α was detectable between 0 and 30 minutes after RNA addition. A protein 'β', which was found to co-electrophorese on polyacrylamide gels with authentic β and which was immunoprecipitated by anti-BBV antiserum, was detectable after 30 minutes. Results of this work show that the formation of 'β' could be prevented by the addition of RNase to the lysate, indicating that intact RNA is necessary for α to β cleavage. Arresting protein synthesis by the addition of cycloheximide to the lysate mix did not inhibit the cleavage. The formation of β could also be prevented by cooling the lysate mix to 1°C. Cleavage of α to β still occurred when RNA 2, without the presence of RNA 1, was translated. Therefore the cleavage is not dependent on a translation product of RNA 1. Sedimentation of lysate on sucrose density gradients showed that α to β cleavage was not accompanied by assembly of BBV RNA and protein lnto a viral substructure as has been shown to occur with some viruses, for example certain picornaviruses. Serial dilution of lysate containing α showed that the level of β decreased with increasing dilution, indicating that the cleavage is not mediated by autocatalysis, but by some other unknown factor. Although much work has been carried out on black beetle virus, no work has been published to date concerning α to β cleavage as an indication of assembly in rabbit reticulocyte lysates. Results of these cell-free translation experiments thus indicate that BBV coat protein precursor α, in association with its messenger RNA 2, undergoes a maturation cleavage in the lysate to produce BBV coat protein β. - Description: The spatial and temporal patterns in selected components of the ichthyofauna and piscivorous avifauna in the small temporarily open/closed Riet River Estuary located on the eastern seaboard of southern Africa was investigated monthly over the period August 2005 to July 2006. The ichthyofauna within the littoral zone of the estuary was sampled using a 5 m seine net (8 stations) while a 30 m seine net (4 stations) was employed to sample the fish in the channel. Bird counts were made along repeat transects along the length of the estuary. Total ichthyofaunal abundances and biomass ranged between 1.60 and 8.67 individuals m⁻² and 0.45 to 21.76 g wwt m⁻² within the littoral zone, and between 0.08 and 0.44 individuals m⁻² and 0.58 and 36.52 g wwt m⁻² in the channel of the estuary. The highest values were generally recorded during the summer months. Results of the numerical analysis indicated that the breaching events recorded over the study period did not lead to a common trend in the ichthyofaunal community. In the absence of a link to the marine environment, the ichthyofaunal community in the littoral zone was numerically dominated by the estuarine resident species, Gilchristella aestuaria and to a lesser extent by Glossogobius callidus, which collectively accounted for ca. 54% of the total ichthyofauna sampled. The establishment of a link to the marine environment coincided with increased numbers of marine breeding species including Atherina breviceps and Rhabdosargus holubi to total fish counts within the estuary. Hierarchical cluster analysis did not identify any spatial patterns in the community structure of the ichthyofauna in the littoral zone or channel zone of the estuary, which could likely be linked to the absence of any distinct horizontal patterns in salinity and temperature within the system. A total of thirteen piscivorous bird species was recorded over the study period. Of the recorded species, six species were wading piscivores, four species were aerial divers and the remaining three species were pursuit swimmers. There were no significant correlations between the estimates of the ichthyofaunal abundance and biomass and bird numbers evident during the study (P> 0.05 in both cases). The Reed Cormorant (Phalacrocorax africanus) was the dominant species throughout the study, with a mean of 8.25 (SD ± 7.90) individuals per count. Mean values of the Pied Kingfisher (Ceryle rudis) and Giant Kingfisher (Megaceryle maximus) were 3.42 (SD ± 1.20) and 1.17 (SD ± 0.60) individuals per count, respectively. The remaining species revealed mean values < 0.5 individuals per count. The highest bird numbers were recorded in winter reflecting the migration of large numbers of the Reed Cormorant into the system. Breaching events were associated with a decrease in total bird numbers, which was most likely due to loss of potential foraging habitat (littoral zone) for waders resulting from reduced water levels. Monthly food consumption by all piscivorous birds revealed large temporal variability, ranging from 26.35 to 140.58 kg per month. The observed variability could be linked to mouth phase and bird numbers.

- Description: The "A" -Reef occurs within the Aandenk Formation of the Central Rand Group, Witwatersrand Supergroup, in the Welkom Goldfield. It comprises the Witpan and Uitsig Reefs which are both oligomictic conglomerates, and are exploited for their gold content by Anglo American Corporation. The main Witpan channel complex is orientated in a NW-SE direction and occurs close to No.3 Shaft of Western Holdings Mine where it is currently being mined. The Witpan Reef varies in thickness from 7-220cm, with lateral facies changes controlling the thickness. Within the reef, gold is associated with degradation surfaces, carbonaceous material, increase in pebble sphericity, and channel edges. Two channel edges have yielded the best gold values on No.3 Shaft although the potential for further payable gold lies in the recognition of sieve conditions. The potential also exists for extensions of the "carbon"-bearing Uitsig channel currently being mined on President Steyn Mine. The palaeo-environment proposed for formation of the Witpan Reef is a braidplain that was partly reworked by a brief transgression. Ore evaluation using geostatistics was considered a valid technique as the dataset is sufficiently well structured. Semi-variograms in the channel and across-channel directions differ markedly. It was found that variograms of gold in cmg/t lacked sufficient structure for modelling, however, log semi-variogram modelling followed by simple log-kriging and back-transformation, proved to be the most successful method. Owing to the morphology and distribution of gold within the reef, a geologically based geostatistical valuation method is proposed. The potential for further exploration of "A" -Reef depends on a substantially higher gold price. In this event, exploration of Uitsig Reef to the southwest of the current mining area is recommended as well as a new exploration strategy for Witpan Reef.

- Description: The Event Horizon Telescope (EHT) aims to resolve the innermost emission of nearby supermassive black holes, Sgr A* and M87, on event horizon scales. This emission is predicted to be gravitationally lensed by the black hole which should produce a shadow (or silhouette) feature, a precise measurement of which is a test of gravity in the strong-field regime. This emission is also an ideal probe of the innermost accretion and jet-launch physics, offering the new insights into this data-limited observing regime. The EHT will use the technique of Very Long Baseline Interferometry (VLBI) at (sub)millimetre wavelengths, which has a diffraction limited angular resolution of order ~ 10 µ-arcsec. However, this technique suffers from unique challenges, including scattering and attenuation in the troposphere and interstellar medium; variable source structure; as well as antenna pointing errors comparable to the size of the primary beam. In this thesis, we present the meqsilhouette software package which is focused towards simulating realistic EHT data. It has the capability to simulate a time-variable source, and includes realistic descriptions of the effects of the troposphere, the interstellar medium as well as primary beams and associated antenna pointing errors. We have demonstrated through several examples simulations that these effects can limit the ability to measure the key science parameters. This simulator can be used to research calibration, parameter estimation and imaging strategies, as well as gain insight into possible systematic uncertainties.

The cultivation of Gracilaria gracilis in the Swartkops estuary, South Africa

- Description: Gracilaria gracilis (Stackhouse) M. Steentoft, L. M. Irvine & W. F. Farnham is a species of red macroalgae which occurs naturally within the Swartkops Estuary in South Africa. Bunches of the species were inserted into the lay of ropes, which were then either suspended in the water column at the Tiger Bay Marina, or staked at the surface of the sediment in the mudflats where the initial material was harvested from. The first site was used for the suspended growth trial, while the second site was for the bottom grown trial. Both of the growth trials ran for the same time period, which was approximately three weeks per season. This study examined three facets of the G. gracilis material that was grown. The growth rate and biomass were recorded in order to examine the potential for the growth of the species on longlines within the estuary. Secondly, the crude nutritional content of the species was determined in order to assess its potential for use as an abalone feed, specifically the dry and ashed weights, and the protein, carbohydrate and lipid contents. Lastly, the cultivated biomass was examined in terms of its agar gel yield and properties, which included the gel strength and the gel –melting and –forming temperatures. The growth rates for the cultivated G. gracilis were poor, and the largest positive growth rate of 1.17 ± 3.2% was found in spring, for the bottom grown trial. This season also saw the only increase in biomass above that which was seeded onto the line, and the final average weight after 22 days was 354.28 ± 68.04 g. All other growth rates were negative, and biomass declined in both growth trials across all the seasons. The suspended growth trials showed more extreme declines, and were heavily contaminated with epiphytic material, while the bottom grown trial had more gradual decrease, and it is likely that conditions associated with the culture systems were not ideal. The largest percentage of protein was found in autumn, where the bottom grown trial had a value of 88 ± 2.32%. For all the seasons, the lipid content had an average percentage of 1.80 ± 0.75% for the bottom grown trial, while the suspended trial was very similar, and had a value of 1.74 ± 0.58%. The nutritional composition of the cultivated biomass was not found to be a good abalone feed in comparison to other studies in land-based aquaculture facilities. Agar characteristics extracted in the present study showed the greatest potential for use as a valuable economic resource. The native agar extracted from the cultivated material was very close to the standards set by United States Pharmacopeia for all the characteristics, and has the potential to possibly be improved further by using an alkali treatment.

The shales, being less resistant to weathering than the quartzites below, and the Dwyka Tillite immediately above, are consequently found in valleys between these two formations. In one of these valleys Grahamstown is situated; where the Lower Dwyka Shales have been eroded by the various rivulets forming the eource of the Blaauwkrantz River. A geological map of the country round Grahamstown was prepared in eome detail, showing all the various rock formations occurring in the area, the full extent of the Lower Dwyka Shales being mapped with the Dwyka Tillite and the Witteberg Quartzite along its margins. The map includes an area of about six miles by three, with the Cathedral spire occupying an approximately central position, and is prepared on a scale of 5.65 inches to one mile. Traversing was done by means of prismatic compass and pacing.

- Description: CURRENTLY UNDER EMBARGO UNTIL THE 26/4/2019: Type 2 diabetes is a metabolic disorder characterised by inflammation, insulin resistance and the inability of pancreatic β-cells to secrete enough insulin to produce a physiological effect. Obesity and high levels of triacylglycerol’s are associated with the development of Type 2 diabetes. Adipose tissue is an active endocrine organ that secretes various protein and peptide hormones, known as adipokines, which mediate important metabolic functions. In an insulin resistant and hyperglycaemic state, levels of anti-inflammatory adipokines, adiponectin, are reduced, whereas levels of pro-inflammatory cytokines, interleukin-6 and interleukin-1β, are elevated; this results in a shift from an anti- to a pro-inflammatory state that is accompanied by dysfunction and apoptosis of the pancreatic β-cells. Cannabis sativa L. has been traditionally used as an anti-inflammatory agent in Southern Africa, specifically treating snakebites, fever and malaria. Δ9-tetrahydrocannabinol is the main psychoactive compound derived from C. sativa, whereas the other major cannabinoids, cannabinol and cannabidiol, have shown anti-inflammatory and sedative properties respectively. Marrubiin is a compound derived from the plant Leonotis leonurus L. and has been traditionally used as an anti-inflammatory and anti-diabetic agent. To determine the effects of these compounds in a hyperglycaemic state, pre- and differentiated mouse adipocytes (3T3-L1 cells) were exposed for seven and fourteen days to the following treatments: Δ9-tetrahydrocannabinol, cannabidiol, cannabinol, marrubiin, anandamide (an endogenous endocannabinoid) and cannabis extract, individually and in combination, under normal glucose and hyperglycaemic conditions. Levels of adiponectin, interleukin-6, leptin, tumour-necrosis factor-α and interleukin-1β were quantified using mouse enzyme-linked immunosorbent assay kits and Oil Red O staining was carried out to determine lipid distribution and lipid droplet characteristics. Results indicate that various cannabinoids, in combination, mediate an anti-inflammatory effect by decreasing the expression of various pro-inflammatory cytokines, which may have allowed for a shift from a pro- to an anti-inflammatory state by these compounds, and may also contribute to the reduction of lipid, which may be used as a supplementary option to current diabetic treatment regimes.

- Description: The main objective of this MSc study was to disentangle whether temperature or the presence of non-native fish was limiting the distribution of Pseudobarbus afer (Peters, 1864) in the Blindekloof stream, Eastern Cape, South Africa. The aims were to: i) describe the thermal regime of the Blindekloof stream; ii) conduct experiments to determine the preferred and critical temperatures of P. afer and; iii) use snorkel surveys to contextualise the distribution of both native and non-native fishes in the Blindekloof stream to assess whether the downstream distribution of P. afer was likely to be influenced by temperature or by the presence of non-native, predatory fishes. To describe the thermal regime, year-long temperature data from four long term monitoring sites in the Blindekloof stream were collected using Hobo temperature loggers and analysed in order to better understand the thermal profile, the thermal variation and the rate of temperature change in the stream. The warmest temperatures were recorded in late December 2015 (absolute maximum of 29.4 °C). The coolest water temperatures were recorded in early August 2015 (absolute minimum of 9.5 °C). There is both seasonal and diel variation in temperature with mean, minimum, maximum, 7 day mean, 7 day maximum and temperature ranges differing significantly between sites. With knowledge of the thermal regime of a monitored reach of the Blindekloof stream, the thermal tolerance and preference of P. afer were investigated. The thermal tolerance of P. afer was investigated using the Critical Thermal Method (CTM) which uses non-lethal endpoints (the loss of equilibrium). At low acclimatization temperatures (11.9 ± 0.7 °C), the mean CTmax of P. afer was found to be 29.9 ± 0.7 °C, while at a higher acclimatization temperature (19.9 ± 0.1 °C), the mean CTmax was 35.1 ± 0.6°C. Custom-built thermal choice tanks were used to investigate the thermal preference of P. afer in both summer and winter. The preferred median temperatures for the summer experiments ranged from 22.4 - 29.3 °C while the winter preferred median temperatures ranged from 18.5 - 23.1 °C. The thermal tolerance of P. afer was compared to the thermal regime of the stream and the results suggest that temperature is not limiting the distribution of P. afer. Snorkel surveys were used to determine the distribution of fishes in the Blindekloof stream. Analysis of the distribution data suggests that, in the absence of non-native predatory species, native fishes have the potential to inhabit pools throughout the system right to the confluence, thus predatory fishes and not temperature was limiting P. afer distributions.

At low acclimatization temperatures (11.9 ± 0.7 °C), the mean CTmax of P. afer was found to be 29.9 ± 0.7 °C, while at a higher acclimatization temperature (19.9 ± 0.1 °C), the mean CTmax was 35.1 ± 0.6°C. Custom-built thermal choice tanks were used to investigate the thermal preference of P. afer in both summer and winter. The preferred median temperatures for the summer experiments ranged from 22.4 - 29.3 °C while the winter preferred median temperatures ranged from 18.5 - 23.1 °C. The thermal tolerance of P. afer was compared to the thermal regime of the stream and the results suggest that temperature is not limiting the distribution of P. afer. Snorkel surveys were used to determine the distribution of fishes in the Blindekloof stream. Analysis of the distribution data suggests that, in the absence of non-native predatory species, native fishes have the potential to inhabit pools throughout the system right to the confluence, thus predatory fishes and not temperature was limiting P. afer distributions.

- Description: In the Upington region, there are three major- tectonic crustal provinces; namely the Kaapvaal Craton, Kheis and Namaqua tectonic provinces. The Eburnian-aged (early Proterozoic) Kheis Province developed along the western flank of the Archaean Kaapvaal Craton while the Kibaran-aged (middle Proterozoic) Namaqua Metamorphic Province, superimposed on the Eburnian-aged basement, developed to the east of the Kheis Province. The Namaqua Metamorphic Province is divided into the Gordonia and Bushmanland Subprovinces, the former being further subdivided into various tectonostratigraphic terranes. These are termed, from west to east, the Kakamas, Areachap, and Upington Terranes. The Upington Terrane includes fault bounded grabens with accompanied bimodal volcanism and sedimentation of the Wilgenhoutsdrif and Koras Groups. The Areachap Terrane consists predominantly of amphibolites generated in an island arc environment while the Kakamas Terrane is characterised by volcano-sedimentary sequences which have been extensively intruded by syn to late-tectonic predominantly I-type Keimoes Suite granitoids. The main styles of mineralisation correlate well with the various tectonostratigraphic terranes. Sedimentary exhalative massive sulphide deposits are characteristic of the Bushmanland Subprovince and are thought to be associated with the deposits at Aggeneys and Putsberg to the west of the area under investigation. These deposits are considered to have been deposited in an east-west-elongated intracontinental basin. The Kakamas Terrane is typified by granite-related mineralisation. In the eastern portion of the Kakamas Terrane, Sn-Wand base metal-bearing veins occur while pegmatites are developed in the western portion. These two styles of granite-related mineralisation is considered to reflect differing depths of formation due mainly to varying degrees of thrusting. The Areachap Terrane consists of volcanogenic massive sulphide deposits of the Besshi-type and is considered to have formed in a back-arc environment. In the Upington Terrane, the Wilgenhoutsdrif and Koras Groups consists essentially of minor Cu occurrences mainly disseminated within basalts and in structural trap sites. The possibility for sediment-hosted Cu deposits is not ruled out. More recent surface processes have led to uranium and gypsum deposits in pans, river beds and calcretes. Eburnian aged tectonic setting remains enigmatic. Kibaran-aged tectonics which best fits the metallogeny of the area under investigation is considered to be of a subduction zone from west to east formed by the collision of the Bushmanland "microcontinent" against the Kaapvaal Craton. Subduction fbrmed an island arc setting in which the massive sulphide deposits were formed in the Areachap Terrane while the Wilgenhoutsdrif Groups developed in a marginal basin. Further convergence led to collision of the two continents and underriding of the Bushmanland "microcontinent" which generated predominantly I-type granitoids represented by the Keimoes Suite. The level of emplacement of these granitoids is a reflection of the degree of foreland thrusting and produced shallower level Sn-W and base metal vein-type mineralisation closer to the suture zone and deeper level pegmatites further from the suture zone to the west. The final period of deformation is represented by northward lateral movement which created "pull apart" fault-bounded basins into which the Koras Group was deposited.

- Description: The thesis focuses on ‘green’ sample preparation techniques that utilise minimal or no organic solvents thereby producing negligible volumes of organic waste, to ensure safety both to user and the environment. Volatile compounds were extracted and pre-concentrated from Kgalagadi desert truffles (kalaharituber) by headspace solid phase microextraction (HSSPME) and supercritical fluid extraction (SFE). PHWE was employed for the extraction of amino acids and fatty acids. Subsequent analysis of volatile compounds was carried out by gas chromatography coupled with mass spectrometry. Four types of HS-SPME fibers (PDMS 100 μm, PDMS 7 μm, Polyacrylate 85 μm, CAR/DVB/PDMS 50/30 μm) were evaluated. A total of 24 volatile compounds with a molecular weight range from 110 to 354, the most prominent peak being 2-t-Butyl-2,3-dimethyl-3-buten-1-ol (C₁₀H₂₀O, MW 156) were detected after sampling with a PDMS 100 μm fiber. Less volatile compounds were detected after SFE with CO2. A total of 16 amino acids were identified while 17 fatty acids (MW from 132.12 to 367.49) were also identified. The characteristic profile of the Kgalagadi desert truffle was found to contain mainly fatty acid methyl esters and unsaturated aliphatic hydrocarbons. The most prominent compound peaks identified were; 2-t-butyl-2,3-dimethyl- 3-buten-1-ol, disulfide, ethyl benzoic acid 2-4-dhydroxy-3,6-dimethyl-methyl ester, 8,11- octadecanoic acid methyl ester, benzoic acid, 2,4-dihydroxy-3,6-dimethyl-methyl ester, isoquinoline, 1 butyl-3,4-dihydro and 3-heptanone, 6 methyl. Seasonal changes are detected in gonadotrope ultrastructure, pituitary LH levels, and plasma hormone levels, and activity pattern of LH gonadotropes generally coincide with reproductive activity in both bat species. In female M. schreibersii gonadotrope activity, pituitary LH, and plasma LH levels coincide with development, implantation and gestation. During the delayed implantation gonadotropes are inactive and and high follicular period of plasma LH levels are low, coinciding with corpus luteum inactivity and low plasma progesterone levels. Implantation coincides with increased activity of the gonadotrope activity, increased plasma LH levels, reactivation of the corpus luteum and elevated plasma progesterone levels. Life-history and stock assessment of Clarias Gariepinus in the Okavango Delta, Botswana

- Description: Large catfishes (Clarias gariepinus & Clarias ngamensis) are not exploited commercially in the Okavango Delta, Botswana. They however constitute a large proportion of the gill net fishery catch (23 %) albeit as by-catch, coming second after cichlids (70 %). Monthly experimetal gill net sampling over 8 years showed that C. gariepinus was the most abundant species in the Okavango Delta by mass making up 37 % of the catch composition by fresh weight. Despite the relatively high abundance of C. gariepinus in the Okavango Delta, its status has never been fully assessed. For this reason, this study aimed to assess the status of C. gariepinus in the Okavango Delta with the particular goal to determine if the species could support a directed commercial gill net fishery. Investigation of the effects of the flood pulse on abundance of C. gariepinus showed that variability in water levels was the major driver of population abundance for this species. Three biological variables: (1) catch per unit effort (CPUE); (2) % catch composition and (3) Shannon's diversity index (H') exhibited a negative and significant relationship with the flood index within a year. The variation in these variables was statistically significant between months. There was a weak positive relationship between water levels and the biological variables between years suggesting increase in productivity of the system at high water levels without a lag period. Age and growth were determined using sectioned otoliths. Edge analysis showed that growth zone deposition occurred during the drawdown period in summer when water temperature and day length were increasing. The maximum recorded age was 15 years. Growth of this species was best described by the linear models (y = 21.50x + 427.39 mm LT; r² = 0.35) for females and (y = 23.79x + 440.83 mm LT; r² = 0.38) for males. The von Bertalanffy growth model described growth as Lt = 10000(1- exp(-0.002(t + 18.07))) mm LT for combined sexes Lt = 10000(1- exp( -0.002(t + 18.50))) mm LT , for females and Lt =908(1-exp(-0.076(t+7.95)))mmLT for males. Age at 50 % maturity was attained in the first year of life. Spawning occurred throughout the year with an apparent peak between November and April. The mean total annual mortality rate (Z) was 0.25 per year. The mean annual natural mortality rate (M) was 0.20 per year and fishing mortality (F) was calculated at 0.05 per year. The life history strategy of C. gariepinus was in between the periodic and opportunistic strategies. Therefore management of this species should be aimed at maintaining a healthy adult population and age structure. Since a large portion of the Okavango Delta is already protected and most other parts are also inaccessible to fishers there are enough spatial refugia for the species and the status quo should be maintained. Under this scenario, there is no need for regulation of the current mesh sizes because the species is harvested after maturity and replenishment of exploited populations will always occur from other parts of the system. Per recruit analyses indicate that the current fishing mortality maintains spawner biomass at levels greater than 90 % of pristine levels. Therefore a commercial fishery may be established using mesh 93 mm to maximize yield with a rotational harvesting strategy. Close monitoring is essential to ensure that re-colonization of overexploited habitats does indeed occur. Recreational angling and commercial gill net fishing need to be separated on a spatial and temporal scale during the feeding run to minimize potential conflicts.

- Description: Large catfishes (Clarias gariepinus & Clarias ngamensis) are not exploited commercially in the Okavango Delta, Botswana. They however constitute a large proportion of the gill net fishery catch (23 %) albeit as by-catch, coming second after cichlids (70 %). Monthly experimetal gill net sampling over 8 years showed that C. gariepinus was the most abundant species in the Okavango Delta by mass making up 37 % of the catch composition by fresh weight. Despite the relatively high abundance of C. gariepinus in the Okavango Delta, its status has never been fully assessed. For this reason, this study aimed to assess the status of C. gariepinus in the Okavango Delta with the particular goal to determine if the species could support a directed commercial gill net fishery. Investigation of the effects of the flood pulse on abundance of C. gariepinus showed that variability in water levels was the major driver of population abundance for this species. Three biological variables: (1) catch per unit effort (CPUE); (2) % catch composition and (3) Shannon's diversity index (H') exhibited a negative and significant relationship with the flood index within a year. The variation in these variables was statistically significant between months. There was a weak positive relationship between water levels and the biological variables between years suggesting increase in productivity of the system at high water levels without a lag period. Age and growth were determined using sectioned otoliths. Edge analysis showed that growth zone deposition occurred during the drawdown period in summer when water temperature and day length were increasing. The maximum recorded age was 15 years. Growth of this species was best described by the linear models (y = 21.50x + 427.39 mm LT; r² = 0.35) for females and (y = 23.79x + 440.83 mm LT; r² = 0.38) for males. The global spread of mangrove species is affected by climate, with most confined to areas that are warmer and moist. At a global scale, temperature limits the distribution of mangrove forests but on a regional scale and local scale, rainfall, river flow and tidal exchange have a strong effect on the distribution and biomass of mangrove forests. Other factors that play a role in limiting growth and distribution of mangroves include accessibility of suitable habitats for growth and also conditions that are suitable for propagule dispersal. The objectives of this study were to review the distribution of mangroves in South Africa and determine the number of extreme temperature events that may limit further distribution and secondly to measure growth rates of mangrove species at Mngazana and Nahoon Estuary and the nutrient pools in the sediment of these forests. In 1982, Ward and Steinke published a list of estuaries where mangroves were present. The current study sampled the population structure, microclimate and level of expansion in two estuaries within and outside of the published range. The minimum, maximum temperature and number of extreme temperature events were measured using iButtons. Mangrove expansion will be limited by minimum temperatures (1.1 ° C) and an increase in extreme temperature events (830) (<5 ° C and 5- 10 ° C) at the latitudinal limits. Expansion of A. marina at Kwelera and Tyolomnqa Estuary was evident but sapling survival was low. Mangroves have expanded within and outside the range proposed by Ward and Steinke (1982), but are limited by physical factors, restricted by the presence of saltmarsh and other estuarine macrophytes and natural disturbance regimes. An increase in population growth was recorded over the years at both Mngazana and Nahoon Estuary. Nutrients, (ammonium, nitrates + nitrites and soluble reactive phosphorus) varied amongst sites and were related to seasonality. Nitrogen in both estuaries was available in the form of ammonium (NH4) and its concentration was generally higher (1.3-76.2 pm) than other forms of nitrogen (0.07-6.3 µm). Soluble reactive phosphorus (SRP) was higher during the wet seasons in both estuaries. An increase in porewater salinity since 2007 (41.3 practical salinity unit (PSU)) was measured at Mngazana and this is a result of freshwater abstraction and low rainfall. A. marina saplings and adults grew significantly faster at Nahoon Estuary (the distributional limit) (11.1 ± 1.1 cm year-¹) compared to Mngazana Estuary (5.3 ± 1.8 cm year-¹). Different mangrove species and forests respond differently to environmental factors and changes in mangrove distribution is expected in South Africa but changes are expected to happen slowly and opportunistically.

- Description: Internationally there is an increasing focus on involving local communities in natural resource management and monitoring. Monitoring methods which are professionally driven appear to be inadequate to deal with the monitoring of natural resource use and biodiversity conservation, globally. This is especially evident in areas such as South African rural communal land. Two community based natural resource management (CBNRM) programmes in areas which are communally governed in the Eastern Cape, South Africa, namely Nqabara and Machubeni, were used as part of this research study. This thesis identified and tested potentially simple and cost effective monitoring methods related to the utilization of the local rangelands and indigenous forests. The criteria that were tested include 1) appropriateness and effectiveness in measuring change, and 2) contribution to building adaptive capacity among local land managers through learning. The criteria were assessed using a scoring system for each monitoring method in order to evaluate their strengths and weaknesses . This was done by using both quantitative and qualitative data. Contribution to building adaptive capacity was assessed by evaluating technical capacity gained, local ecological knowledge contributed and learning by participants. This was done using qualitative data. The results show that the monitoring methods had different strengths and weaknesses in relation to the criteria, making them more appropriate for different priorities such as effectively measuring change or building adaptive capacity. It is argued that an adaptive approach is a useful component in the participatory monitoring process. An adaptive framework was developed from lessons learnt in this study for collaborative monitoring. Challenges such as low literacy levels and adequate training still need to be addressed to strengthen efforts towards participatory monitoring. Factors such as incentives, conflict and local values may negatively affect the legitimacy and sustainability of participatory monitoring and therefore also need to be addressed.
