Macrophyte phenology in a temporarily open/closed Estuary compared with a permanently open Estuary

- Description: Temporarily open/closed estuaries (TOCEs) are unpredictable environments that change in response to mouth condition, which is influenced by freshwater flooding or sea storm surges. The aim of the study was to determine whether macrophyte phenology in a TOCE was event driven rather than cyclically predictable and if it differed from permanently open estuaries (POEs). Macrophyte growth and flowering phenology in response to environmental conditions was investigated in the East Kleinemonde Estuary (TOCE) and the Kowie Estuary (POE) along the eastern coastline of South Africa. The lack of freshwater flooding due to low rainfall coupled with several overwash events resulted in a prolonged period of mouth closure from September 2008 to the end of this study period in 2010. This in turn caused the inundation of the supratidal and intertidal habitats, high water level (> 1.57 m amsl) and high salinity (30 - 42 ppt) in the TOCE. Principle Components Analysis showed that high water level and reduced sediments were the most significant environmental factors affecting macrophyte phenology. Macrophyte phenology in the POE was primarily driven by temperature, sediment redox potential and salinity. The saline high water level and reduced sediment significantly reduced macrophyte cover in all habitats in the TOCE. Macrophytes in the POE maintained high cover abundance due to seasonal re-growth compared to the TOCE where cover declined over the sampling period due to the high water level. Subsequent to water level dropping by as little as 11 - 20 cm in the TOCE, the intertidal species Sarcocornia tegetaria and Salicornia meyeriana completed their life-cycles and produced viable seeds within four and three months of germinating respectively. In contrast, the Sarcocornia hybrid and S. meyeriana in the POE took longer to complete their life-cycles, namely seven and nine months respectively, while S. tegataria did not germinate in situ but reproduced vegetatively despite producing seed. In the TOCE, the submerged species Ruppia cirrhosa and Chara vulgaris completed their life-cycles within five and three months and produced a maximum of 26 242 and 196 998 seeds m-2 respectively. Due to high water level and prolonged inundation, the reproductive periods were shorter for the intertidal and reed and sedge species in the TOCE compared to the POE. Seed output during the two reproduction periods varied between the two estuaries. Sarcocornia decumbens and S. tegetaria produced a substantially higher number of seeds in the TOCE compared to the POE, namely 0 - 102 847 versus 20 661 - 48 576 seeds m-²; and 7 001 - 45 542 versus 1 587 – 16 958 seeds m-² respectively. Seed output in the TOCE was significantly higher in S. tegetaria during the second reproduction period despite the significantly lower plant cover, which may be a function of the stressful environment in the TOCE. Seed production of S. meyeriana was significantly higher in the POE compared to the TOCE, with 264 224 - 640 292 compared with 24 050 - 27 643 seeds m-², due to higher plant cover in the POE. The research suggests that macrophyte phenology in the TOCE was significantly influenced by mouth condition. Further, macrophytes were able to demonstrate considerable phenotypic plasticity in response to changing and unfavourable environmental conditions. These data can be used in mouth management plans and freshwater requirement studies in TOCEs to ensure that macrophytes can complete their life-cycles and produce viable seeds for the safeguarding of habitat persistence and ecological processes. In impacted estuaries where artificial mouth opening is practised and the macrophytes have been severely degraded or extirpated, management should ensure that the intertidal and supratidal habitats are not inundated during peak flowering and seed production periods i.e. late spring to early autumn (November to March).

Microalgal biomass and distribution in the Mngazi and Mngazana Estuaries

- Description: The present study was undertaken in the temporarily open/closed Mngazi and permanently open Mngazana estuaries, located on the subtropical east coast of South Africa. The results from this research will assist decision makers in the freshwater management of these systems. Intertidal and subtidal benthic chlorophyll a concentrations, water column chlorophyll a, nutrients and several physico-chemical parameters were measured between June 2002 and November 2003. The objective of this study was to determine if the presence of freshwater in the estuaries had an effect on the microalgae of both estuaries. Five sites were sampled in the Mngazi Estuary and 14 sites were sampled in the Mngazana Estuary. The average water column chlorophyll a was significantly higher (p<0.05) in the Mngazana Estuary (surface 7.8 ± 0.7 μg.l-1, bottom 6.4 ± 0.7 μg.l-1) compared to the Mngazi Estuary (surface 4.9 ± 1.2 μg.l-1, bottom 7.3 ± 1.5 μg.l-1). There was no evidence of an REI (river-estuary interface) zone in areas where the water column chlorophyll a concentrations were high even during open mouth conditions in the Mngazi Estuary. The REI is that area where salinity is less than 10 ppt and is characterized by high water column productivity. Even though both systems received some freshwater during the summer periods, this was not enough to stimulate phytoplankton growth and nutrient availability seems to be the major factor limiting phytoplankton in these systems. Flagellates and diatoms were the dominant phytoplankton groups in both estuaries during the entire sampling session. The relative abundance of the different phytoplankton groups did not show differences between sites. The relative abundance of flagellates was in most cases greater than 60% and diatoms made up the remainder. The average benthic chlorophyll a was higher in the Mngazana Estuary (intertidal 24 ± 6 μg.g-1 subtidal 15.2 ± 3 μg.g-1) compared to the Mngazi Estuary (intertidal 15.3 ± 4.3 μg.g-1 subtidal 5.4 ± 1.6 μg.g-1). Regions with high benthic chlorophyll a concentrations had high sediment organic content. Sediment organic content was higher in the Mngazana Estuary (1 percent - 8 percent) compared to the Mngazi Estuary (4 percent – 6.8 percent). The sites situated on the Main Channel had on average significantly higher (p<0.05) benthic chlorophyll a biomass compared to Creek 1 and Creek 2 in the Mngazana Estuary. Peaks in benthic chlorophyll a concentrations occurred in the intertidal sediments in Creek 1 (50.4 ± 13.4 μg.g-1) and Creek 2 (57.4 ± 1.4 μg.g-1) in the Mngazana Estuary, the peaks occurred in winter during a period of low freshwater inflow into the estuary. Microphytobenthic biomass measured in the Mngazi Estuary is among the lowest values reported in the literature for temporarily open/closed estuaries. Statistical 4 analysis showed no significant difference between benthic chlorophyll a during the different mouth conditions and sampling sessions in the Mngazi Estuary. Microalgal responses in the Mngazana Estuary were similar to those observed in other permanently open marine dominated estuaries. In the temporarily open/closed Mngazi Estuary microalgal characteristics were different to that of other temporarily open/closed estuaries probably because the estuary was only sampled in the open and semi-closed state.

Microbial ecology of the Buffalo River in response to water quality changes

- Description: South Africa’s freshwater quality and quantity is declining and consequently impacting on the ecological health of these ecosystems, due to increased agricultural, urban and industrial developments. The River Health Programme (RHP) was designed for monitoring and assessing the ecological health of freshwater ecosystems in South Africa, in order to effectively manage these aquatic resources. The RHP utilises biological indicators such as in-stream biota as a structured and sensitive tool for assessing ecosystem health. Although the RHP has been widely implemented across South Africa, no attempts have been made to explore microbial ecology as a tool that could be included as one of the RHP indices. This study used selected microbial responses and water physico-chemical parameters to assess the current water quality status of the Buffalo River. This study showed that water quality impairments compounded in the urban regions of King William’s Town and Zwelitsha and also downstream of the Bridle Drift Dam. The results also showed that the lower and the upper catchments of the Buffalo River were not significantly different in terms of water physico-chemistry and microbiology, as indicated by low stress levels of an NMDS plot. Though similarities were recorded between impacted and reference sites, the results strongly showed that known impacted sites recorded the poorest water physico-chemistry, including the Yellowwoods River. However, the Laing Dam provided a buffer effect on contributions of the Yellowwoods River into the Buffalo River. Multivariate analysis showed that microbial cell counts were not influenced by water physico-chemical changes, whilst microbial activity from the water and biofilm habitats showed significant correlation levels to water physico-chemical changes. This study demonstrated that further investigations towards exploitation of microbial activity responses to water physico-chemical quality changes should be channelled towards the development of microbiological assessment index for inclusion in the RHP.

Although catsharks are an important component of the near-shore marine biodiversity in South Africa and most of the species are endemic, little is known about their movement patterns, home range and population size. With an increasing number of recreational fishers this information is crucial for their conservation. The aims of this study were threefold. Firstly, to identify and analyze existing data sources on movement patterns and population dynamics for four catshark species: pyjama (Poroderma africanum), leopard (P. pantherinum), puffadder (Haploblepharus edwarsii) and brown (H. fuscus). This highlighted a number of shortcomings with existing data sets, largely because these studies had diverse objectives and were not aimed solely at catsharks. Secondly, a dedicated study was carried out for a limited area, testing a number of methods for data collection, and where appropriate the data was analyzed to determine movement patterns and population numbers. Thirdly, the most appropriate methodology for future studies (with similar objectives) was identified, and the results of the study were used to propose a number of conservation measures. All species of catsharks exhibited strong site fidelity and limited dispersal for extended periods. A few individuals did, however, travel distances in excess of 150 km. Significant trends in temporal abundance were not observed, nevertheless, there was some evidence for higher catches from September to December. Population estimates for the study area were low, with P. africanum having the smallest population size while H. fuscus had the highest population size within the restricted study area. Limited movements, high site fidelity and small population sizes emphasize their vulnerability and suggest that catsharks would benefit from no-take marine protected areas.

- Description: Loggerhead and leatherback sea turtles nest on the beaches of the north-eastern portion of Kwazulu-Natal within the iSimangaliso Wetland Park. Loggerheads place ~60 percent of all nests within an 8 km stretch of beach, whereas leatherbacks tend to space their nests more evenly along the entire length of the monitoring area. The study aimed to determine nest site fidelity of loggerheads and leatherbacks (using four decades of nesting data housed by Ezemvelo KZN Wildlife) and the factors that influence nest site selection of both species within the 56 km of turtle monitoring area (32N to 100S) and the 5 km area of high-density loggerhead nesting (0N to 12N). The effectiveness of nest site selection was then determined through the hatching success of loggerheads over the 5km area (0N to 12N). Results showed that loggerheads show a high degree of nest site fidelity (~3 km) with nest site fidelity of individuals increasing over subsequent seasons of nesting, as well as these individuals using the same stretches of beach for nesting (the most popular area being 1N to 4N for repeat nesters). - Description: Loggerhead and leatherback sea turtles nest on the beaches of the north-eastern portion of Kwazulu-Natal within the iSimangaliso Wetland Park. Loggerheads place ~60 percent of all nests within an 8 km stretch of beach, whereas leatherbacks tend to space their nests more evenly along the entire length of the monitoring area. The study aimed to determine nest site fidelity of loggerheads and leatherbacks (using four decades of nesting data housed by Ezemvelo KZN Wildlife) and the factors that influence nest site selection of both species within the 56 km of turtle monitoring area (32N to 100S) and the 5 km area of high-density loggerhead nesting (0N to 12N). The effectiveness of nest site selection was then determined through the hatching success of loggerheads over the 5km area (0N to 12N). Results showed that loggerheads show a high degree of nest site fidelity (~3 km) with nest site fidelity of individuals increasing over subsequent seasons of nesting, as well as these individuals using the same stretches of beach for nesting (the most popular area being 1N to 4N for repeat nesters). The system is maintainable, extendable and customisable for different network architectures. The system was deployed successfully to two community networks. These complexes were synthesized and studied since many questions regarding the interaction of sulfur-donors and platinum still exist. These relate to thermodynamic and kinetic factors and their influence on anticancer action. In this dissertation the properties of novel platinum(II) complexes of a bidentate ligand having an aromatic nitrogen-donor atom in combination with a thioethereal sulfur atom capable of forming a five-membered ring with platinum(II) were studied. The general structure of the (NS)-ligands used was 2-((alkylthio)methyl)pyridine. Alkyl groups used were methyl, ethyl, propyl, benzyl and phenyl. Amine complexes of platinum have been studied extensively in the past. However, attention was given to novel aspects of substituted pyridine and imidazole ligands and their corresponding complexes. Amongst these are 2-(2-methylaminoethyl)pyridine, 1-methyl-2-methylaminoethylimidazole and 1-methyl-2-methylaminobenzylimidazole. The leaving groups included chloro, bromo and oxalato. Mononitroplatinum(IV) complexes were prepared using novel synthetic methods. Selected platinum(II) amine complexes were used as starting materials for this synthesis. Some of these compounds exhibit promising anticancer behaviour. (Trans-(R,R)-1,2-diaminocyclohexane)(oxalato)(mononitrochloro)platinum(IV) is a particularly good anticancer agent and has been patented internationally. All these complexes were characterized using mass spectrometry, chromatography, thermogravimetric analysis, kinetic aspects such as ligand exchange rates and finally their anticancer action against three different cancer cell lines was evaluated via cytotoxicity assays. Some of the compounds exhibited particularly good anticancer potential.

The peripherally tetra-substituted cationic (Q(T-2-Py)MPcs) and anionic (MTSPcs) were found to be aggregated in aqueous media, yet the octa-carboxylated (MOCPcs) counterparts were monomeric in solution. Cremophor EL (CEL) was used as a disaggregating agent, all the aggregated complexes disaggregated partially or completely in the presence of CEL. The photophysicochemical properties of aggregated complexes were investigated both in the presence of CEL and in aqueous media of pH 11 alone. Low triplet, singlet oxygen and fluorescence quantum yield were obtained in aqueous media (especially for the aggregated complexes) but a high improvement was achieved upon addition of CEL. The gallium complexes ((OH)GaOCPc and (OH)GaTSPc) showed good photophysicochemical properties with higher triplet and singlet oxygen quantum yields. For photoelectrochemistry the (MPcs) dyes were adsorbed to nanoporous ZnO, electrodeposited in the presence of eosin Y as structure directing agent (SDA) on FTO substrates by refluxing or soaking the films in a solution containing the dye of interest such that a full surface coverage was achieved. Quaternized cationic (Q(T-2-Py)MPc) and tetrasulfonated (MTSPcs) phthalocyanines formed strong aggregates when deposited on the surface of FTO/ZnO substrate leading. High external (IPCE) and internal (APCE) quantum efficiencies of up to 50.6% and 96.7% were achieved for the OTiOCPc complex. There was a lower overall cell efficiency for quaternized and tetrasulfonated metallophthalocyanines because of the strong aggregates when they were on the surface of the electrodes. Among the studied materials, OTiOCPc gave the highest overall cell efficiency of phthalocyanine electrodeposited on ZnO of so far = 0.48%.

Both MTAPcs quenched the QDs emission, with quenching constants of the order of 103–104M−1, binding constants of the order of 108-1010M-1 and the number of binding sites for the MTAPc upon the QD being 2. High energy transfer efficiencies were obtained (in some cases as high as 93%), due to the low donor to acceptor distances. The complexes [ReVOCI(had)] and [ReIVCl(had)(PPh3)](ReO4) were prepared from the reaction of trans-[ReCl3(MeCN)(PPh3)2] with N,N-bis((2-hydroxybenzyl)-2- aminoethyl)dimethylamine (H2had) in ethanol under various reaction conditions. The treatment of [ReCl3(benzil)(PPh3)] with 2-[((2- pyridinylmethyl)amino)methyl]phenol (Hham) in a 2:1 molar ratio in acetonitrile led to the isolation of the hydrogen-bonded dimer [ReOCl2(ham)]2. The distorted octahedral complex [ReOCl(hap)] [H2hap = N,N-bis(2- hydroxybenzyl)aminomethylpyridine] was prepared from the reaction of trans- [ReCl3(MeCN)(PPh3)2] with a twofold molar excess of H2hap in acetonitrile. The X-ray crystal structure analysis shows that the chloride is coordinated trans to the tripodal tertiary amino nitrogen, with a phenolate oxygen trans to the oxo oxygen.

The complexes [ReVOCI(had)] and [ReIVCl(had)(PPh3)](ReO4) were prepared from the reaction of trans-[ReCl3(MeCN)(PPh3)2] with N,N-bis((2-hydroxybenzyl)-2- aminoethyl)dimethylamine (H2had) in ethanol under various reaction conditions. The treatment of [ReCl3(benzil)(PPh3)] with 2-[((2- pyridinylmethyl)amino)methyl]phenol (Hham) in a 2:1 molar ratio in acetonitrile led to the isolation of the hydrogen-bonded dimer [ReOCl2(ham)]2. The distorted octahedral complex [ReOCl(hap)] [H2hap = N,N-bis(2- hydroxybenzyl)aminomethylpyridine] was prepared from the reaction of trans- [ReCl3(MeCN)(PPh3)2] with a twofold molar excess of H2hap in acetonitrile. The X-ray crystal structure analysis shows that the chloride is coordinated trans to the tripodal tertiary amino nitrogen, with a phenolate oxygen trans to the oxo oxygen.

It was hypothesised that these isotopic and biochemical changes were in response to increased food availability during summer. Although mangrove material appeared not to play an important role in the nutrition of these sandy beach communities, it was suggested that a high retention time of particles in the bay could enhance bacterial decay of particulate mangrove material, which could then act as fine, bacterial-enriched particulate food to the macrobenthos. Strong seasonal patterns in temperature, dissolved oxygen and total chlorophyll-a (chl-a) concentrations were evident. Total average hyperbenthic densities ranged between 0.04 and 166 ind.m-3 in the lower net and between 0.12 and 225 ind.m-3 in the upper net. Hyperbenthic biomass values ranged between 0.02 and 11.9 mg.dry weight.m-3 in the lower net and between 0.02 and 17.4 mg.dry weight.m-3 in the upper net. A spatial and temporal pattern in total densities was detected with an increase in abundance over the period of September to October 2008 particularly in the middle reaches (Stations 3 and 4). Both the lower and upper nets were numerically dominated by decapods (mainly brachyuran crab zoeae) with the exception of June and July 2008 when mysids (mainly Mesopodopsis wooldridgei) dominated, making up 72.4 ± 58.14% of the total abundance in the lower net. A redundancy analysis (RDA) indicated that 99.2% of the variance in the hyperbenthic community structure could be explained by the first two canonical axes. Axis one, which accounted for 96.8% of the total variation detected in the ordination plot was highly correlated with sedimentary organic content and to a lesser extent the chl-a concentration within the Kariega Estuary. The correlations with the second canonical axis (2.4%) were less obvious, however, salinity and seston concentration were weakly correlated with this axis. Diel variability in the hyperbenthic community structure was assessed during March 2009. Samples were collected during the day and night (n = 6 for each period) using sampling gear described above. Total average hyperbenthic densities during the day (497.9 ± 254.1 ind.m-3) were significantly higher than night-time estimates (129.9 ± 38.5 ind.m-3; p<0.05). There were no significant differences in the average dayand night-time estimates of hyperbenthic biomass (p>0.05). A hierarchical cluster analysis identified two significantly distinct groupings, designated the day and night samples. Results from the SIMPER procedure indicated that the high densities of crab zoeae recorded during the day-time accounted for the majority of the dissimilarity between the day and night groupings (44.7%). Spatio-temporal dynamics of ichthyoplankton in the Kowie estuary, South Africa

- Description: Ichthyoplankton dynamics in the permanently open Kowie Estuary, in the warm temperate region of South Africa was investigated. The composition, abundance, distribution and seasonality of larval fishes were studied over a two year period between 2004 and 2006. Additionally, tidal exchange of ichthyoplankton and the use of frontal zones in the mouth region of the estuary were also explored between 2008 and 2009. Temporal and spatial trends in occurrence of larval fishes within the estuary and associated marina were obtained from data collected seasonally using boat-based plankton netting at 14 sampling stations along the length of the estuary. A total of 11 128 larval fishes were collected, representing 23 families and 38 taxa. Clupeidae and Gobiidae were the dominant fish families, contributing 47.0 % and 24.7 % respectively to the total catch. Estuarine resident species dominated the overall catch (91 %). A notable absence of older stage larvae and early juveniles characterised the artificial channels of the marina and estuary mouth region. This was attributed to the absence of a shallow, marginal water habitat typical of successful estuarine nursery areas. Tidal exchange of larval fishes was investigated in the Kowie Estuary using a new technique. A set of drifting light traps were set repetitively on the ebb and flood tide every second night for two consecutive 14 day periods during the peak estuarine recruitment period. A total of 553 larval fishes were caught during the study, representing nine families and 26 species. Blenniidae and Clupeidae dominated the catches. Family and species occurrence changed with tide state. Species richness (d) and diversity (H’) varied with tide and was highest on flood tides. Estuary-dependent species, such as Omobranchus woodii were more dominant on flood tides, whilst larvae of marine-spawned species, such as Sardinops sagax, were dominant on the ebb tide. Light trap catches yielded a different composition in terms of development stage / size and species, when compared to towed net studies. Towed plankton nets were again used to study the shear fronts that characterise the mouth region of the canalised Kowie Estuary. It was hypothesised that the convergence zone of the front provides feeding opportunities for ichthyoplankton. Ichthyoplankton, zooplankton and phytoplankton was collected from within and immediately outside of the convergence zone during frontal conditions. Species specific distribution trends emerged from this study. Postflexion larvae and early juvenile stages of the estuary dependent Mugilidae were only present in the foam line of the convergence zones. Some known predators of ichthyoplankton were also present in slightly higher numbers in the convergence zone (isopods) while other predators such as mysid shrimps, chaetognaths and cnidarians were more abundant out of the convergence zone. It appears that these zones may provide access to temporary food patches in the lower estuary but will be accompanied by a trade-off with increased isopod predators. The Kowie Estuary is a heavily impacted system and lack of adequate freshwater supply; artificial channelling and pollution ultimately have a negative impact of the success of this system as a nursery area. Rehabilitation of marginal areas in the lower estuary and marina is suggested as remedial action to re-establish the integrity of the nursery function this system could offer.

Structure and functioning of fish assemblages in two South African estuaries, with emphasis on the presence and absence of aquatic macrophyte beds

- Description: Temporarily open/closed estuaries (TOCEs) are the dominant estuary type in South Africa. These systems are often characterized by extensive beds of submerged macrophytes, which form important foraging and shelter habitats for fishes, especially for estuary-dependent fish species such as the Cape stumpnose Rhabdosargus holubi and Cape moony Monodactylus falciformis that are commonly associated with them. A loss of submerged macrophytes from an estuary has been shown to affect the fish community as well as reducing overall system productivity. The TOC East Kleinemonde Estuary, situated in the warm-temperate biogeographic region of South Africa has been subject to an ongoing long-term fish monitoring project since 1995. During the period 1995 to 2002, this estuary contained large beds of the submerged macrophytes Ruppia cirrhosa and Potamogeton pectinatus. However, subsequent to a major flood event in 2003 these macrophytes have been largely absent from this system. The effect of the loss of submerged macrophytes on the East Kleinemonde fish assemblage was investigated through an analysis of seine and gill net catch data. Seine net catches for a 12 year period, encompassing six years of macrophyte presence and six years of macrophyte senescence, revealed changes in the relative abundance of certain fish species. Vegetation-associated species such as R. holubi and M. falciformis decreased in abundance whereas sediment-associated species, especially members of the family Mugilidae, increased in abundance following loss of the macrophytes in this estuary. The critically endangered pipefish Syngnathus watermeyeri was only recorded in catches during years in which macrophyte beds were present. In addition to the analysis of catch data, the importance of macrophytes as a primary energy source for selected estuarine fishes was explored through the analysis of carbon and nitrogen stable isotopes. Prior to conducting these analyses, common methodological practices to address the presence of carbonates and lipids within isotope samples were evaluated. A subset of samples were either acid washed to remove carbonates, or lipids were removed according to the method of Bligh and Dyer (1959) as both of these compounds have been shown to affect stable carbon and nitrogen isotope ratios. The suitability of the lipid normalization models of Fry (2002) and Post et al. (2007) for samples of estuarine fish muscle were also tested. Based on this evaluation both models are suitable for use with estuarine fish muscle tissue, however since neither carbonate nor lipid content of any of the samples used in this study was high all samples were left untreated in the following analysis. Carbon isotope ratios from a wide range of fish species collected from the East Kleinemonde Estuary during the macrophyte-senescent phase were compared with individuals of the same species from the neighbouring West Kleinemonde Estuary (where extensive beds of R. cirrhosa and P. pectinatus were present) and revealed the influence of submerged macrophyte material in the diet of fishes in the latter system. However, it was apparent that these plants are not directly consumed but rather contribute to the detrital pool that forms a food source for most invertebrate and some fish species. The most significant source of carbon for East Kleinemonde fishes during the macrophyte senescent phase appeared to have a more depleted origin; probably from benthic or pelagic microalgae. In conclusion, while the importance of macrophyte beds as shelter and foraging habitats for estuarine fishes are well documented, their role in terms of the structuring and functioning of fish assemblages in TOCEs remains somewhat uncertain. The findings of this study were possibly masked by the resilience of vegetation-associated species to the loss of this habitat, as well as by life history characteristics of species such as R. holubi that allow their numerical dominance despite habitat change. Nonetheless, macrophyte senescence in the East Kleinemonde Estuary resulted in the loss of at least one species and the reduced abundance of vegetation-associated species, probably reflective of reduced food resources and/or increased vulnerability to predation. As a result, beds of submerged macrophytes are an important habitat within TOCEs.

Carbon isotope ratios from a wide range of fish species collected from the East Kleinemonde Estuary during the macrophyte-senescent phase were compared with individuals of the same species from the neighbouring West Kleinemonde Estuary (where extensive beds of R. cirrhosa and P. pectinatus were present) and revealed the influence of submerged macrophyte material in the diet of fishes in the latter system. However, it was apparent that these plants are not directly consumed but rather contribute to the detrital pool that forms a food source for most invertebrate and some fish species. The most significant source of carbon for East Kleinemonde fishes during the macrophyte senescent phase appeared to have a more depleted origin; probably from benthic or pelagic microalgae. In conclusion, while the importance of macrophyte beds as shelter and foraging habitats for estuarine fishes are well documented, their role in terms of the structuring and functioning of fish assemblages in TOCEs remains somewhat uncertain. The findings of this study were possibly masked by the resilience of vegetation-associated species to the loss of this habitat, as well as by life history characteristics of species such as R. holubi that allow their numerical dominance despite habitat change. Nonetheless, macrophyte senescence in the East Kleinemonde Estuary resulted in the loss of at least one species and the reduced abundance of vegetation-associated species, probably reflective of reduced food resources and/or increased vulnerability to predation. As a result, beds of submerged macrophytes are an important habitat within TOCEs.

The anticancer activity of Cyathula prostrata on two malignant cell lines

- Description: Plants have always been a source of medicine and are still being used by traditional healers in the rural part of Africa, Asia and India to treat a range of illnesses including cancer. The in vitro anticancer activity of an 80 percent ethanol extract of Cyathula prostrata, an annual branching shrub used by traditional healers in Nigeria to treat cancer was investigated. No previous studies have outlined the possible pathways and mechanisms used by cancer cells when treated with C. prostrata. Dose response analysis was performed to determine the effective cytotoxic concentrations of C. prostrata on HeLa (cervical cancer cell line) and U937 (myelo-monocytic cell line). The IC50 values were 100.8 μg/ml and 64.4 μg/ml for HeLa and U937 cells, respectively. All further experiments were performed using 125 μg/ml C. prostrata extract and 50 μM cisplatin as positive control. With the use of the fluorescent DNA binding dye propidium iodide, the induction of tumour cell death by C. prostrata extract has been linked to cell cycle arrest in the G1 phase at 24 and 48 h. In both cell lines, more than 80 percent of the C. prostrata treated cells were found in the G1 phase after 48 hours of treatment. The annexin V-FITC/PI assay revealed an increase in the percentage apoptotic cells from 4.9 percent to 53.1 percent at 24 h and 8.3 percent to 50.3 percent at 48 h. Since apoptosis induction can occur via a number of different pathways, distinct features were used as markers to investigate the mode of action of this C. prostrata extract. Markers such as activated caspase-8, p21 and cyt-c, were investigated with the aid of fluorescently labelled (FITC) antibodies with analysis using flow cytometry. No change in p21 levels was observed in response to treatment with the extract for up to 48 h. Cell cycle arrest in G1 was therefore not induced by this cyclin-CDK inhibitor. Increase in caspase-8 activation was observed in response to treatment with the extract with no cyt-c release from the mitochondria. The lack of cyt-c release was due to no change in mitochondrial membrane potential, which was investigated with the aid of fluorescent mitochondrial dyes and flow cytometric techniques. Caspase-8 activation is unique to the extrinsic apoptotic pathway. The results from this study therefore show that C. prostrata extract induces apoptosis via the extrinsic pathway and that this activation in independent of the mitochondria. The levels of hTERT, the catalytic subunit of telomerase, were investigated as an additional molecular target for C. prostrata. This was also investigated using FITC labelled antibodies and flow cytometry. A decrease in hTERT levels was observed following C. prostrata treatment. - Description: Austroglanis gilli and A. barnardi are endemic to the Clanwilliam-Olifants System in the Western Cape, South Africa. The populations of each of these species are considered to be threatened by various anthropogenic activities, which include inappropriate agricultural practice and impacts of alien invasive fish species. The purpose of this thesis was to assess the life-history of these two endangered species in order to contribute to understanding their biology. Such information is vital for the development of strategies for their conservation. Marginal zone and marginal increment analyses from sectioned lapilliar otoliths of both A. gilli and A. barnardi showed a unimodal peak, suggesting a single annulus formation. The oldest specimens of A. gilli and A. barnardi were 12+ and 14+ years, respectively. The growth of A. gilli and A. barnardi was relatively slow and was best described by the von Bertalanffy growth curve as: L, = 131.56(1 - exp(- 0.27(t - 1.18 ))) for male and L, = 113.86(1 - exp(- 0.43(1- 0.74))) for female A. gilli from the Rondegat River. In the Noordhoeks River, growth was L, = 99.67(1 - exp(- 0.53(t - 0.35))) for male and L, = 96.60(1 - exp(- 0.64(t - 0.11 ))) for female A. gilli, and L, =71.02(1-exp(-0.26(1-3.07))) and L, = 69.50(1-exp(-0.36(1 - 1.88))) for male and female A. barnardi, respectively. The average natural mortality for the combined sexes was estimated using catch curve analysis at 0.37 ± 0.12 per year for A. gilli from the Rondegat River, and at 0.71 ± 0.05 and 0.39 ± 0.04 per year for A. gilli and A. barnardi from the Noordhoeks River, respectively. For A. gilli in the Rondegat River, the first maturity was estimated at 3.3 years (97.3 mm SL) for males and 3.1 years (94.3 mm SL) for females. In the Noordhoeks River, the first maturity for A. gilli was estimated at 2.0 years (71.9 mm SL) for males and 1.7 (66.4 mm SL) for females, and for A. barnardi at 2.0 (55.0 nun SL) for males and 2.9 years (58.9 mm SL) for females. Maturity corresponded closely to the asymptotic sizes from the von Bertalanffy curves, suggesting a shift in energy use from somatic growth to gonad development. Macroscopic assessment of the state of gonads, the Ganado-somatic index and histological examinations revealed that both A. gilli and A. barnardi have a single spawning season. The presence of oocytes in different stages of development in each of the ovaries of mature females collected between November and January suggested asynchronous, iteroperous serial spawning. The resorption of yolk was observed from ovaries collected between February and March and no vitellogenic oocyte was visible from ovaries collected between April and August. A similar trend was observed for males, with spermatozoa filling the lumen between November and January and some residual spermatozoa being present in the lumens soon after the breeding season. Spermatocytes and spermatids were dominant in the testes until October. An Index of Relative Importance revealed that A. gilli feeds predominantly on the benthic macroinvertebrate larvae of Ephemeroptera (particularly Baetidae), Diptera (particularly Chironomidae and Simuliidae) and Trichoptera. Austroglanis barnardi feeds predominantly on dipteran larvae (particularly Chironornidae and Simuliidae). Chi square contingency tables showed a significant difference in the dominant prey items of A. gilli and A. barnardi in the Noordhoeks River (X₂= 53.79, d.f. = 4, p > 0.001) and A. gilli between Rondegat and Noordhoeks rivers (x₂ = 34.74, d.f. = 4, p > 0.001). The Spearman's rank correlation test showed no shifts in the diet of A. barnardi from Noordhoeks River and A. gilli from Rondegat River with a change in size and season (p>O.OS). However, there was a shift in the diet of A. gilli from the Noordhoeks River which could suggest a distinct patchiness of benthic macroinvertebrates between the riffle feeding areas used by juveniles and other biotopes used by adults. The occurrence, in stomach contents, of other prey items from a wide variety of taxa and the presence of allochthonous material from the terrestrial environment could suggest an opportunistic feeding guild for both Austroglanis species. The life-history traits of A. gilli and A. barnardi, which are charaterized by slow growth, long life span and low relative fecundity, indicate that both species are relatively precocial and K-selected. The population of a precocial species is relatively stable and if population numbers were to be greatly reduced, they would require a long time to rebuild. An urgent conservation intervention is therefore recommended for Austroglanis spp. so as to maintain the diversity of populations within these species. The creation of protected river reserves and raising public conservation awareness may minimise activities that result in altered river hydrology and the destruction of complex benthic habitats.

The botanical importance and health of the Bushmans estuary, Eastern Cape, South Africa

- Description: The Bushmans Estuary is one of the few permanently open estuaries in the Eastern Cape that is characterized by large intertidal salt marshes. Freshwater inflow to the estuary has decreased as a result of abstraction by more than 30 weirs and farm dams in the catchment. The mean annual run-off is naturally low (38 x 106 m3 y-1) and thus abstraction and reduction of freshwater inflow to the estuary is expected to cause a number of changes. The aims of this study were to determine the current health/status of the estuary based on the macrophytes and microalgae and identify monitoring indicators for the East London Department of Water Affairs, River Health Programme. Changes in the estuary over time were determined from available historical data which were compared with present data. This analysis showed that under normal average conditions freshwater inflow to the estuary is very low, less that 0.02 m3 s-1 most of the time. Under these conditions the estuary is in a homogenous marine state. Vertical and horizontal salinity gradients only form when high rainfall and run-off occurs (> 5 m3.s-1). Salinity gradients from 30.1 PSU at the mouth to 2.2 PSU in the upper reaches were measured in 2006 after a high flow event. However the estuary quickly reverted back to its homogenous condition within weeks after this flood. This study showed that freshwater inflow increased nutrient input to the estuary. Total oxidised nitrogen (TOxN) and soluble reactive phosphorus (SRP) concentrations were higher in August 2006, after the flood, than during the other low flow sampling sessions. TOxN decreased from a mean concentration of 21.6 μM in 2006 to 1.93 μM in February 2009. SRP decreased from 55.3 μM to 0.2 μM respectively. With the increased nutrient availability, the response in the estuary was an increase in phytoplankton biomass. After the 2006 floods the average water column chlorophyll-a was 9.0 μg l-1, while in the low freshwater inflow years it ranged from 2.1 to 4.8 μg l-1. The composition of the phytoplankton community was always dominated by flagellates and then diatoms, with higher cell numbers in the nutrient-enriched 2006 period. Although the water column nutrient data indicated that the estuary was oligotrophic, benthic microalgal biomass (11.9-16.1 μg.g-1) in the intertidal zone was comparable with nutrient rich estuaries. Benthic species indicative of polluted conditions were found (Nitzschia frustulum, Navicula gregaria, Navicula cryptotenelloides). These benthic species were found at the sites where wastewater / sewage seepage had occurred. Benthic diatom species also indicated freshwater inflow. During the high flow period in 2006 the dominant diatoms were fresh to brackish species that were strongly associated with the high concentrations of TOxN and SRP (Tryblionella constricta, Diploneis smithii, Hippodonta cf. gremainii, and Navicula species). During the freshwater limited period of 2008 and 2009 the benthic diatom species shifted to a group responding to the high salinity, ammonium and silicate concentrations. The species in this group were Nitzschia flexa, Navicula tenneloides, Diploneis elliptica, Amphora subacutiuscula and Nitzschia coarctata. Ordination results showed that the epiphytic diatom species responded to different environmental variables in the different years. Most of the species in 2008/2009 were associated with high salinity, temperature, dissolved oxygen, ammonium and silicate concentrations while the response was towards TOxN and SRP in 2006. The dominant species were Cocconeis placentula v euglyphyta in 2006; Nitzschia frustulum in 2008; and Synedra spp in 2009. The average biomass of the epiphytes was significantly lower in May 2008 than in both August 2006 and February 2009; 88.0 + 17.7 mg.m-2, 1.7 + 0.8 mg.m-2, and 61.8 + 14.4 mg.m-2 respectively. GIS mapping of past and present aerial photographs showed that submerged macrophyte (Zostera capensis) cover in 1966 and 1973 was less than that mapped for 2004. Salt marsh also increased its cover over time, from 86.9 ha in 1966 to 126 ha in 2004, colonizing what were bare sandy areas. Long-term monitoring of the health of the Bushmans Estuary should focus on salinity (as an indicator of inflow or deprivation of freshwater), benthic diatom identification and macrophyte distribution and composition (for the detection of pollution input), and bathymetric surveys (for shallowing of the estuary due to sedimentation).

- Description: The Bushmans Estuary is one of the few permanently open estuaries in the Eastern Cape that is characterized by large intertidal salt marshes. Freshwater inflow to the estuary has decreased as a result of abstraction by more than 30 weirs and farm dams in the catchment. The mean annual run-off is naturally low (38 x 106 m3 y-1) and thus abstraction and reduction of freshwater inflow to the estuary is expected to cause a number of changes. The aims of this study were to determine the current health/status of the estuary based on the macrophytes and microalgae and identify monitoring indicators for the East London Department of Water Affairs, River Health Programme. Changes in the estuary over time were determined from available historical data which were compared with present data. This analysis showed that under normal average conditions freshwater inflow to the estuary is very low, less that 0.02 m3 s-1 most of the time. Under these conditions the estuary is in a homogenous marine state. Vertical and horizontal salinity gradients only form when high rainfall and run-off occurs (> 5 m3.s-1). Salinity gradients from 30.1 PSU at the mouth to 2.2 PSU in the upper reaches were measured in 2006 after a high flow event. However the estuary quickly reverted back to its homogenous condition within weeks after this flood. This study showed that freshwater inflow increased nutrient input to the estuary. Total oxidised nitrogen (TOxN) and soluble reactive phosphorus (SRP) concentrations were higher in August 2006, after the flood, than during the other low flow sampling sessions. TOxN decreased from a mean concentration of 21.6 μM in 2006 to 1.93 μM in February 2009. SRP decreased from 55.3 μM to 0.2 μM respectively. With the increased nutrient availability, the response in the estuary was an increase in phytoplankton biomass. After the 2006 floods the average water column chlorophyll-a was 9.0 μg l-1, while in the low freshwater inflow years it ranged from 2.1 to 4.8 μg l-1. The composition of the phytoplankton community was always dominated by flagellates and then diatoms, with higher cell numbers in the nutrient-enriched 2006 period. Although the water column nutrient data indicated that the estuary was oligotrophic, benthic microalgal biomass (11.9-16.1 μg.g-1) in the intertidal zone was comparable with nutrient rich estuaries. Benthic species indicative of polluted conditions were found (Nitzschia frustulum, Navicula gregaria, Navicula cryptotenelloides). These benthic species were found at the sites where wastewater / sewage seepage had occurred. Benthic diatom species also indicated freshwater inflow. During the high flow period in 2006 the dominant diatoms were fresh to brackish species that were strongly associated with the high concentrations of TOxN and SRP (Tryblionella constricta, Diploneis smithii, Hippodonta cf. gremainii, and Navicula species). During the freshwater limited period of 2008 and 2009 the benthic diatom species shifted to a group responding to the high salinity, ammonium and silicate concentrations. The species in this group were Nitzschia flexa, Navicula tenneloides, Diploneis elliptica, Amphora subacutiuscula and Nitzschia coarctata. Ordination results showed that the epiphytic diatom species responded to different environmental variables in the different years. A total of 8174 larval and early juvenile fishes were caught, representing 12 families and 23 taxa. The Clupeidae, Gobiidae and Blenniidae were the dominant fish families. Common species included Gilchristella aestuaria, Caffrogobius gilchristi, Omobranchus woodi, Liza dumerilii, Glossogobius callidus and Myxus capensis. Estuarine resident species (Category I) predominantly in the preflexion developmental stage, dominated the system. A total of 19 zooplankton taxa were recorded. Copepoda dominated the zooplankton community. Dominant species included Pseudodiaptomus hessei, Acartia longipatella, Halicyclops sp., Mesopodopsis wooldridgei, and the larvae of Paratylodiplax edwardsii and Hymenosoma orbiculare. Mean larval fish density showed similar trends seasonally, spatially and across salinity zones, with mean zooplankton density in the Sundays Estuary. Gut content analysis of five larval fish species: Gilchristella aestuaria, Pomadasys commersonnii, Monodactylus falciformis, Myxus capensis and Rhabdosargus holubi, revealed species specific diet and prey selection. Although larval fish diet contained a variety of prey items, guts were dominated by P. hessei, chironomid larvae, Corophium triaenonyx, copepod eggs and insect larvae. Physico-chemical drivers and the interactions between these two plankton communities provide information that enables a more holistic view of the dynamics occurring in the Sundays Estuary planktonic ecosystem.

The E. coli RNA degradosome, a large multi-component protein complex which originates through specific protein interactions, has been referred to as the “RNA decay machine” and is responsible for mRNA turnover. The degradosome functions to process RNA and its key components have been identified. The scaffold protein is RNase E and it tethers the degradosome to the cytoplasmic membrane. Polynucleotide phosphorylase (PNPase), ATP-dependent RNA helicase (RhlB helicase) and the glycolytic enzyme enolase associate with RNase E to form the degradosome. Polyphosphate kinase associates with the degradosome in substoichiometric amounts, as do the molecular chaperones DnaK and GroEL. The role of DnaK as well as that of enolase in the RNA degradosome is unknown. Very limited research has been conducted on the components of the RNA degradosome under conditions of stress. The aim of this study was to understand the role played by enolase in the assembly of the degradosome under conditions of stress, as well as investigating the protein levels of molecular chaperones under these conditions. The RNA degradosome was successfully purified through its scaffold protein using nickel-affinity chromatography. In vivo studies were performed to investigate the protein levels of DnaK and GroEL present in the degradosome under conditions of heat stress, and whether GroEL could functionally replace DnaK in the degradosome. To investigate the recruitment of enolase to the degradosome under heat stress, a subcellular fractionation was performed to determine the localization of enolase upon heat shock in vivo. The elevated temperature resulted in an increased concentration of enolase in the membrane fraction. To determine whether there is an interaction between enolase and DnaK, enolase activity assays were conducted in vitro. The effect of DnaK on enolase activity was measured upon quantifying DnaK and adding it to the enolase assays. For the first time it was observed that the activity of enolase increased with the addition of substoichiometric amounts of DnaK. This indicates that DnaK may be interacting with the RNA degradosome via enolase.
