- Description: In this study a presentation of the technology behind dye-sensitized solar cells, their design as well as the role of the different parts of the cell. The characterization of the cell is divided into four sections namely: the characterization of the paste required to make the TiO2 film and its optical properties using SEM-EDX and XRD analytical techniques; Analysis of the various absorptions of three Ru-based dyes using UV-Vis spectroscopy, Photoluminescence and Fourier Transform Infra-Red spectroscopy; the characterization and the analyses of the entire cell using Electrochemical Impedance Spectroscopy. The nine cells were prepared by examining RuL2(CN)2 , RuL2(NCS)2 or N3 dye and RuL2(NCS)2 TBA+ or N719 dye. [L = 2,2'-bipyridyl-4,4'-dicarboxylic acid ;TBA = tetra-butyl ammonium] were combined with three electrolytes namely: Z–150 , AN–50 and PN–50. The Iodolyte PN–50 is an iodide based low viscosity electrolyte with 50 mM of tri-iodide dissolved in a solvent called propionitrile (PN). The Iodolyte AN–50 is an iodide based low viscosity electrolyte with 50 mM of tri-iodide dissolved in a solvent called acetonitrile (AN). The Iodolyte Z–150 is an iodide based low viscosity electrolyte with 150 mM of tri-iodide dissolved in a solvent called 3-methoxypropionitrile (MPN) and with additives such an ionic liquid, malkylbenziimidazole and guanidine thiocyanate. A solar simulator was utilized with which the standard solar irradiation can be created in laboratory conditions. While incineration is the simplest way to recycle the used tyres it isn’t an efficient way to dispose of tyres. This research looks at developing a method for characterizing the New Reclamation Group (NRG) reclaim with the main focus on Hi-Res™ thermogravimetric analysis. Good quantification was possible using this technique. A trade off was established between resolution, sensitivity and time. While increasing the resolution allowed greater separation to be achieved the time for each experiment increased rapidly. Although kinetic models do exist for quantifying rubber components in vulcanized sample, they were not suitable for this study. When devulcanization causes significant molecular changes, such as with NRG reclaim, the decomposition profiles no longer match those of virgin materials. Formulations developed focused on mixing, rheometry, crosslink density and tensile properties. The NRG reclaim acted as a processing aid which lowered the maximum torque. This provides mixing safety as the temperature is decreased as a result of the lower torque. This effect was not seen with conventional reclaim. Rheometry tests indicated that the addition of the devulcanizates decreased the extent of cure. It was demonstrated that this could be linked to crosslink density. Testing of the 100%, 200%, 300% moduli correlated the crosslink density to the maximum torque. Although the addition of NRG reclaim reduced the tensile strength of the formulation, a link between crosslink density and ultimate tensile strength (UTS) couldn’t be made. The decrease in the UTS and increase in extension at break is possibly caused by an increase in low molecular weight material present in the formulations and decrease in crosslink density. This could possibly increase the mobility of polymer chains which could increases flexibility.

The characterization of DNAJC3: elucidating the function of the TPR domains

- Description: DNAJC3 is a novel member of the DNAJ family with two domains linked to co-chaperone functions, namely the tetratricopeptide repeat (TPR) and J domain. Out of the two domains, the TPR domains are the least characterized. Therefore, the aim of this study was to characterize and elucidate additional functions of DNAJC3 TPR domains through in silico, in vitro and ex vivo approaches. Through multiple sequence and structural alignment as well as electrostatic potential analysis, DNAJC3 TPR domain were found to be most similar to TPR-containing proteins with Hsp90 or Hsp70 independent functions. In vitro pull down assays illustrated that DNAJC3 TPR domains did not interact with either cytosolic Hsp90 and Hsp70 or Grp78 and Grp94 directly, however a potential indirect interaction with Grp94 and Hsp90 was observed in mammalian lysates, via pull down assays; suggesting the formation of a complex between the proteins mediated by a specific substrate. DNAJC3 TPR domains were found to bind indiscriminately to both native and heat denatured substrates in a dose dependent manner. DNAJC3 TPR domains bound to β-galactosidase with greater affinity than malate dehydrogenase (MDH), suggesting that DNAJC3 TPR domains might exhibit substrate specificity that has not been reported before. Preliminary ex vivo analysis of DNAJC3 in mammalian cells showed that induced stress conditions did not alter the cytosolic or endoplasmic reticulum (ER) localization, or levels of DNAJC3 protein, suggesting that the protein is not stress inducible. However, protein levels of DNAJC3 were dramatically reduced by Hsp90 inhibitor novobiocin at 500 μM. Transient knockdown DNAJC3 did not change the protein levels of either Grp78 or Grp94, but decreased the protein levels of Hsp70/Hsp90 organizing protein HOP. On the other hand, protein levels of DNAJC3 were increased in HOP depleted cells. In conclusion, this study was the first to experimentally demonstrate that DNAJC3 TPR domains do not interact directly with Hsp90, Hsp70, Grp78 or Grp94, and therefore DNAJC3 is unlikely to participate in traditional co-chaperone interactions with those proteins via its TPR domain. However, the J domain is known to interact with Grp78. The discovery that DNAJC3 TPR domains resemble that of TPR-containing proteins with functions independent of Hsp90 or Hsp70 suggests that DNAJC3 might link the Hsp70/Grp78 chaperone machinery to non co-chaperone related functions, which requires further analysis.

- Description: Malaria remains a public health problem and a high burden of disease, especially in developing countries. The unicellular protozoan malaria parasite of the genus Plasmodium infects about a quarter of a billion people annually, with an estimated 409 000 death cases. The majority of malaria cases occurred in Africa; hence, the region is regarded as endemic for malaria. Global efforts to eradicate the disease led to a decrease in morbidity and mortality rates. However, an enormous burden of malaria infection remains, and it cannot go unnoticed. Countries with limited resources are more affected by the disease, mainly on its public health and socio-economic development, due to many factors besides malaria itself, such as lack of access to adequate, affordable treatments and preventative regimes. Furthermore, the current antimalarial drugs are losing their efficacy because of parasite drug resistance. The emerged drug resistance has reduced the drug efficacy in clearing the parasite from the host system, causing prolonged illness and a higher risk of death. Therefore, the emerged antimalarial drug resistance has hindered the global efforts for malaria control and elimination and established an urgent need for new treatment strategies. When the resistance against classical antimalarial drugs emerged, the class of antifolate antimalarial medicines became the most common alternative. The antifolate antimalarial drugs target the malaria parasite de novo folate biosynthesis pathway by limiting folate derivates, which are essential for the parasite cell growth and survival. Yet again, the malaria parasite developed resistance against the available antifolate drugs, rendering the drugs ineffective in many cases. Given the previous success in targeting the malaria parasite de novo folate biosynthesis pathway, alternative enzymes within this pathway stand as good targets and can be explored to develop new antifolate drugs with novel mechanisms of action. The primary focus of this thesis is to contribute to the existing and growing knowledge of antimalarial drug discovery. The study aims to characterise the malaria parasite de novo folate synthesis pathway enzymes guanosine-5'-triphosphate (GTP) cyclohydrolase I (GCH1) and 6-pyruvoyl tetrahydropterin synthase (PTPS) as alternative drug targets for malaria treatment by using computational approaches. Further, discover new allosteric drug targeting sites within the two enzymes' 3D structures for future drug design and discovery. Sequence and structural analysis were carried out to characterise and pinpoint the two enzymes' unique sequence and structure-based features. From the analyses, key sequence and structure differences were identified between the malaria parasite enzymes relative to their human homolog; the identified sites can aid significantly in designing and developing new antimalarial antifolate drugs with good selectivity toward the parasites’ enzymes. GCH1 and PTPS contain a catalytically essential metal ion in their active site; therefore, force field parameters were needed to study their active sites accurately during all-atom molecular dynamic simulations (MD). The force field parameters were derived through quantum mechanics potential energy surface scans of the metals bonded terms and evaluated via all-atom MD simulations. Proteins structural dynamics is imperative for many biological processes; thus, it is essential to consider the structural dynamics of proteins whilst understanding their function. In this regard, the normal mode analysis (NMA) approach based on the elastic network model (ENM) was employed to study the intrinsic dynamics and conformations changes of GCH1 and PTPS enzymes. The NMA disclosed essential structural information about the protein’s intrinsic dynamics and mechanism of allosteric modulation of their binding properties, further highlighting regions that govern their conformational changes. The analysis also disclosed hotspot residues that are crucial for the proteins' fold stability and function. The NMA was further combined with sequence motif results and showed that conserved residues of GCH1 and PTPS were located within the identified key structural sites modulating the proteins' conformational rearrangement. The characterized structural features and hotspot residues were regarded as potential allosteric sites of important value for the design and development of allosteric drugs. Both GCH1 and PTPS enzymes have never been targeted before and can provide an excellent opportunity to overcome the antimalarial antifolate drug resistance problem. The data presented in this thesis contribute to the understanding of the sequence, structure, and global dynamics of both GCH1 and PTPS, further disclose potential allosteric drug targeting sites and unique structural features of both enzymes that can establish a solid starting point for drug design and development of new antimalarial drugs of a novel mechanism of actions. Lastly, the reported force field parameters will be of value for MD simulations for future in-silico drug discovery studies involving the two enzymes and other enzymes with the same Zn2+ binding motifs and coordination environments. The characterization of the interaction between Streptococcus pneumoniae PspC and Homo sapiens pIgR

- Description: Streptococcus pneumoniae is a commensal bacterium in the human nasopharyngeal tract and is known to cause severe respiratory related diseases in humans. The pneumococcal surface protein C (PspC) has been demonstrated to interact with human polymeric immunoglobulin receptor (pIgR) and its free form secretory component (SC). S. pneumoniae utilizes the pIgR recycling mechanism to bypass the nasopharyngeal epithelial barrier of the host through the PspC-pIgR interaction. Studies have shown the YPT motifs in PspC R domains to be involved in this interaction (Hammerschmidt et al., 2000). The exact amino acid sequences in pIgR/SC have not yet been elucidated but Elm et al., (2004), Lu et al., (2003) and Venables et al., (2013) have demonstrated that Ig-like domains 3 and 4 of SC are mutually critical in this interaction. Mutagenesis studies were conducted to give insight into regions of these domains responsible for this interaction. In order to select possible candidates for mutagenesis, homology modelling, protein-protein docking and multiple sequence alignments were performed. PspC-SC interaction is human specific (Hammerschmidt et al., 2000) and the conservation between known species was used to select amino acids located on highly variable loop regions of the SC molecule. Amino acids Ser257, Asp312 and Gln373 were suggested by previous studies and investigated in this study. Protein-protein docking was performed with Cluspro 2.0 and Haddock 2.2 webservers. The docking results, coupled with conservation information resulted in Arg304 also being selected for investigation. Additional candidates were identified for future studies. Point mutations of the selected amino acids were introduced with overlap PCR and confirmed by DNA sequencing. The SC-D3D4 proteins were expressed in vitro and refolded by an on-column method developed by Venables et al., (2013). The Biorad ProteOn™ XPR36 protein interaction array system at Rhodes University was utilized for measuring the effects of the SC-D3D4 mutations on affinity for PspC-R1R2. The affinities of expressed SC-D3D4 mutants for PspC-R1R2 were compared to the wild type (WT) SC-D3D4 in concentration dependent kinetic analyses. - Description: Streptococcus pneumoniae is a commensal bacterium in the human nasopharyngeal tract and is known to cause severe respiratory related diseases in humans. The pneumococcal surface protein C (PspC) has been demonstrated to interact with human polymeric immunoglobulin receptor (pIgR) and its free form secretory component (SC). S. pneumoniae utilizes the pIgR recycling mechanism to bypass the nasopharyngeal epithelial barrier of the host through the PspC-pIgR interaction. Studies have shown the YPT motifs in PspC R domains to be involved in this interaction (Hammerschmidt et al., 2000). The exact amino acid sequences in pIgR/SC have not yet been elucidated but Elm et al., (2004), Lu et al., (2003) and Venables et al., (2013) have demonstrated that Ig-like domains 3 and 4 of SC are mutually critical in this interaction. Mutagenesis studies were conducted to give insight into regions of these domains responsible for this interaction. In order to select possible candidates for mutagenesis, homology modelling, protein-protein docking and multiple sequence alignments were performed. PspC-SC interaction is human specific (Hammerschmidt et al., 2000) and the conservation between known species was used to select amino acids located on highly variable loop regions of the SC molecule. Amino acids Ser257, Asp312 and Gln373 were suggested by previous studies and investigated in this study. Protein-protein docking was performed with Cluspro 2.0 and Haddock 2.2 webservers. The docking results, coupled with conservation information resulted in Arg304 also being selected for investigation. Additional candidates were identified for future studies. Point mutations of the selected amino acids were introduced with overlap PCR and confirmed by DNA sequencing. The SC-D3D4 proteins were expressed in vitro and refolded by an on-column method developed by Venables et al., (2013). The Biorad ProteOn™ XPR36 protein interaction array system at Rhodes University was utilized for measuring the effects of the SC-D3D4 mutations on affinity for PspC-R1R2. The affinities of expressed SC-D3D4 mutants for PspC-R1R2 were compared to the wild type (WT) SC-D3D4 in concentration dependent kinetic analyses. The KD values for WT SC-D3D4 and Ser257, Arg304, Asp312 and Gln373 mutations were 1.4, 2.9, 4.1, 2.0, and 2.1 μM, respectively. Botswana is considered a stronghold for free roaming cheetahs in Africa, yet the country has had relatively limited research on its cheetahs, and information from the east of the country is lacking. Data on the current status of populations is thus required to make informed management decisions. My study provides estimates of population density, abundance, distribution and status for the demographically open cheetah population of the Northern Tuli Game Reserve (NOTUGRE) in Botswana. The effectiveness of two population monitoring methods, namely camera trapping and a photographic survey, were also investigated. Moreover, I report on the level of conflict between livestock farmers and predators on rural communal farmlands within and adjacent to NOTUGRE. Data were collected between May 2012 and November 2013. Results indicate a low population density of 0.61 ± 0.18 adult cheetahs per 100 km² and a minimum population size of 10 individuals (nine adults and one cub). Camera traps placed at cheetah scent-marking posts increased detection rates and provided ideal set up locations. This approach, together with Spatial Explicit Capture- Recapture (SECR) models, is recommended for future studies. The long-term studies that are required to better understand the status of cheetahs in Botswana do not exist. Thus, photographic surveys may provide an alternative method for providing baseline data on population numbers, distribution and demography. The third aspect of my study gathered information on levels of livestock loss and human tolerance of predators through the use of interviews (n = 80). Conflict with subsistence farmers is a concern as livestock depredation is relatively high (9.1% of total livestock owned) and farmers had an overall negative attitude towards conservation of large predators. My results suggest that human-predator conflict in this area is more complex than the direct financial loss from depredation. Hence, reducing depredation rates alone is unlikely to change farmer tolerance of wildlife on farmlands. Improved, responsible farm management, including self-responsibility for livestock rearing, and positive appreciation for wildlife are necessary. The NOTUGRE cheetah population requires further research to understand possible threats to the population. Furthermore, a better understanding of the connectivity between cheetahs of NOTUGRE, South Africa and Zimbabwe is required. The number of cheetahs within NOTUGRE is too small to sustain a viable population, hence conserving cheetahs outside of the protected area should be a priority for the conservation of the population. This can only be achieved through assistance and involvement from national authorities, local people and conservation organisations.

- Description: Water quality changes that occur during establishment and maintenance of nitrification in two identical recirculating aquaculture systems containing rainbow trout are described. The time taken for the nitrification process to become established was 40-45 days. Mortality of fish attributed to elevated nitrite concentrations were recorded during the initial conditioning phase of the systems. Un-ionised ammonia concentrations did not attain lethal levels during this period. Nitrate concentrations accumulated slowly throughout the study, while the pH and alkalinity of the water decreased with progressive nitrification. Levels of carbon dioxide, calcium, dissolved and suspended solids remained relatively stable until the carrying capacity of the systems was increased, upon which they increased rapidly and general water quality deteriorated. Permanently elevated concentrations of ammonia and nitrite served as an indication that the carrying capacity of the systems had been exceeded. The use of ozone as a water enhancement treatment in aquaculture systems during one- and six-hour applications was also considered in this study. Ozonation significantly reduced nitrite levels at · low concentrations (0,1 - 0,15 mg/l), although they returned to pre-treatment levels within a few hours of cessation of the treatment. The formation of an unstable, intermediate product that reforms as nitrite in the absence of ozone, rather than the complete oxidation of nitrite to nitrate, is proposed. Ozonation also resulted in decreased dissolved solids, and improved the clarity and odour of the water. Ozone had no effect on ammonia concentrations (at pH > 7,0), or on nitrate or calcium levels, and did not conclusively increase the redox potential of the water. Residual ozone concentrations up to 0,04 mg/l in a sixhour treatment had no harmful effects on rainbow trout, and there were no signs of gas-bubble disease arising from supersaturation of the water with oxygen. Activated carbon filters were effective at removing residual ozone from the water after treatment. Treatment of the water with oxygen alone had no effect on nitrite concentrations.

- Description: This thesis investigates the chemistry of 25 ascidian species collected from Algoa Bay, South Africa with a concerted focus on metal accumulation by these ascidians and the possible interaction of these metals with ascidian metabolites. Chapter 2 details the screening techniques employed to establish the presence of nitrogenous metabolites (1H- 15N HMBC), hyper-accumulated metal ions (ICP-MS) and potential metal ion/ ascidian metabolite complexes (LC-ICP-MS/ESI-MS). Unfortunately, exhaustive attempts to detect intact metal ion/ascidian metabolite complexes through the use of liquid chromatography with parallel inductively coupled plasma mass spectrometry/electrospray mass spectrometry (LC-ICPMS/ ESI-MS) were unsuccessful. However, the LC-ICP-MS/ESI-MS data obtained for the crude organic extracts of six of the Algoa Bay ascidian species, Distaplia skoogi, Aplidium monile, Aplidium sp., Didemnum sp., Leptoclindines sp. and Polycitor sp. enabled identification of a number of ten halogenated metabolites, namely the indoles 2.28-2.30, and the tyramine and tyrosine derivatives (2.31-2.33, 2.41, 2.43, 2.44 and 2.46), within the ascidian extracts. This study confirmed that LC-ICP-MS/ESI-MS is a powerful tool for the dereplication of halogenated metabolites in complex mixtures especially where these compounds are present in very small amounts. This study is also the first report of these compounds (eight of which are known) in African ascidians. Compounds 2.32 and 2.46 have not been reported before from a marine source. Compounds 2.28-2.30 and 2.33 were present in sufficient amounts in the respective ascidian extracts to allow their isolation and structure elucidation using standard spectroscopic techniques Chapter 3 explores the ability of ascidians to accumulate a wide range of metal ions at concentrations which are often orders of magnitude higher than those of the surrounding sea water. Inductively coupled plasma mass spectrometry (ICP-MS) was used to determine the total ion concentrations of 24 metals in 25 Algoa Bay ascidian species. To the best of our knowledge this is the largest and most extensive investigation of metal concentrations in a group of different ascidians occurring in the same area. Hypotheisizing that the metal ion concentrations for each ascidian specimen screened may represent a unique fingerprint for each specimen principal component analysis (PCA) was used in an attempt to establish whether there were spatial, temporal or phylogenetic relationships associated with the metal concentration fingerprints of the ascidians that formed part of this study. The PCA results showed that there were no statistically significant relationships between ascidian metal ion concentrations and either the collection year or the collection site of the ascidians. However, species from the family Didemnidae provided the clearest statistical evidence supporting a phylogenetic relationship between these ascidians and their hyperaccumulated metal ion profiles. Furthermore, these results suggested that ascidian species are indeed actively concentrating metal ions from the surrounding sea water and are not simply sinks for passively accumulated metal ions. Interestingly, the concentration of vanadium in the set of ascidians studied did not appear to correlate with any of the other metals accumulated by these ascidians suggesting that there is possibly a unique method employed for the accumulation of vanadium by ascidians. Chapter 4 investigated this possibility further after the nucleosides 4.10, 4.11, 4.13, 4.15, 4.17 and 4.40 were isolated from the vanadium accumulating ascidian Aplidium monile. Studies into the interactions between nucleosides and vanadyl are unfortunately rare and usually qualitative in nature with limited information provided about the stability or structures of the complexes formed. The vanadyl accumulating aplousobranch ascidians e.g. Aplidium monile dominated our study of Algoa Bay ascidians therefore providing us with the rationale to investigate the relatively little studied binding ability and stability of vandyl-nucleoside complexes. Potentiometric studies were conducted to determine the stability constants of complexes formed between the oxovanadium ion vanadyl (VO2+) and the commercially available nucleosides 4.10-4.14. The data afforded by this analysis clearly confirmed the complexity of the vanadyl/nucleoside complexation and suggested that guanosine (4.12) formed the most stable complex with oxovanadium ions. We were also able to establish a third protonation constant for the hydroxyl moiety in 4.12 with a logK 8.87 which has not been previously reported. Finally, Chapter 5 revisited the cytoxicity two Algoa Bay ascidians, Clavelina sp. and Atriolum marinense the extracts from which produced promising bioactivity results in previous studies against oesophageal cancer cells. The HP-20 fractionated extracts of Clavelina sp. and Atriolum marinense proved to be similalrly cytotoxic to breast cancer cells. With the exception for the 100% acetone(aq)fractions the NMR data for both species suggested that most active non polar fractions were dominated by what appeared to be structurally unremarkable fatty acid glycerides and as such were not pursued further. Purification of the 100% acetone(aq)fraction of A. marinense resulted in the isolation of a styrene trimer, 5.1, common to both ascidian extracts. The NMR simulation software WIN-DAISY was employed to confirm the structure of 5.1. Attempts to establish if 5.1 was an isolation artefact or a product of marine pollution were inconclusive

This study confirmed that LC-ICP-MS/ESI-MS is a powerful tool for the dereplication of halogenated metabolites in complex mixtures especially where these compounds are present in very small amounts. This study is also the first report of these compounds (eight of which are known) in African ascidians. Compounds 2.32 and 2.46 have not been reported before from a marine source. Compounds 2.28-2.30 and 2.33 were present in sufficient amounts in the respective ascidian extracts to allow their isolation and structure elucidation using standard spectroscopic techniques Chapter 3 explores the ability of ascidians to accumulate a wide range of metal ions at concentrations which are often orders of magnitude higher than those of the surrounding sea water. Inductively coupled plasma mass spectrometry (ICP-MS) was used to determine the total ion concentrations of 24 metals in 25 Algoa Bay ascidian species. To the best of our knowledge this is the largest and most extensive investigation of metal concentrations in a group of different ascidians occurring in the same area. Hypotheisizing that the metal ion concentrations for each ascidian specimen screened may represent a unique fingerprint for each specimen principal component analysis (PCA) was used in an attempt to establish whether there were spatial, temporal or phylogenetic relationships associated with the metal concentration fingerprints of the ascidians that formed part of this study. The PCA results showed that there were no statistically significant relationships between ascidian metal ion concentrations and either the collection year or the collection site of the ascidians. However, species from the family Didemnidae provided the clearest statistical evidence supporting a phylogenetic relationship between these ascidians and their hyperaccumulated metal ion profiles. Furthermore, these results suggested that ascidian species are indeed actively concentrating metal ions from the surrounding sea water and are not simply sinks for passively accumulated metal ions. Interestingly, the concentration of vanadium in the set of ascidians studied did not appear to correlate with any of the other metals accumulated by these ascidians suggesting that there is possibly a unique method employed for the accumulation of vanadium by ascidians. Chapter 4 investigated this possibility further after the nucleosides 4.10, 4.11, 4.13, 4.15, 4.17 and 4.40 were isolated from the vanadium accumulating ascidian Aplidium monile. Studies into the interactions between nucleosides and vanadyl are unfortunately rare and usually qualitative in nature with limited information provided about the stability or structures of the complexes formed. The vanadyl accumulating aplousobranch ascidians e.g. Aplidium monile dominated our study of Algoa Bay ascidians therefore providing us with the rationale to investigate the relatively little studied binding ability and stability of vandyl-nucleoside complexes. Potentiometric studies were conducted to determine the stability constants of complexes formed between the oxovanadium ion vanadyl (VO2+) and the commercially available nucleosides 4.10-4.14. The data afforded by this analysis clearly confirmed the complexity of the vanadyl/nucleoside complexation and suggested that guanosine (4.12) formed the most stable complex with oxovanadium ions. We were also able to establish a third protonation constant for the hydroxyl moiety in 4.12 with a logK 8.87 which has not been previously reported. Finally, Chapter 5 revisited the cytoxicity two Algoa Bay ascidians, Clavelina sp. and Atriolum marinense the extracts from which produced promising bioactivity results in previous studies against oesophageal cancer cells. The HP-20 fractionated extracts of Clavelina sp. and Atriolum marinense proved to be similalrly cytotoxic to breast cancer cells. With the exception for the 100% acetone(aq)fractions the NMR data for both species suggested that most active non polar fractions were dominated by what appeared to be structurally unremarkable fatty acid glycerides and as such were not pursued further. Purification of the 100% acetone(aq)fraction of A. marinense resulted in the isolation of a styrene trimer, 5.1, common to both ascidian extracts. The NMR simulation software WIN-DAISY was employed to confirm the structure of 5.1. Attempts to establish if 5.1 was an isolation artefact or a product of marine pollution were inconclusive

- Description: The high level of crime in South Africa raises the question about the failures of the criminal justice system on the one hand, and South Africa’s social policies on the other. Young people in South Africa can disproportionately be both victims and perpetrators of crime in the Republic of South Africa. The child accused in conflict with the law is dealt with in much the same way as their adult counterparts, as the criminal justice system was designed by adults for adults. South Africa became a signatory to the United Nations Convention on the Rights of the Child 19891(hereinafter referred to as UNCRC) on 16 June 1995. The UNCRC provides a backdrop to section 28 of the Constitution of the Republic of South Africa Act.2 Article 3(1) of the UNCRC provides as follows: “in all actions concerning children, whether undertaken by public or private social welfare institutions, courts of law, administrative authorities or legislative bodies, the best interests of the child shall be primary consideration.” South Africa is therefore according to article 40(3) of the UNCRC obliged to “establish laws, procedures, authorities and institutions specifically applicable to children in conflict with the law”.3 In terms of article 40(1) of the UNCRC “State Parties recognise the right of every child alleged as, accused of, or recognised as having infringed the penal law to be treated in a manner consistent with the promotion of the child’s sense of dignity and worth, which reinforces the child’s respect for the human rights and fundamental freedoms of others and which takes into account the child’s age and the desirability of promoting the child’s reintegration and the child’s assuming a constructive role in society.”4 1 Adopted by the General Assembly resolution 44/25 on 20 November 1989. 2 The Constitution of the Republic of South Africa Act, Act 108 of 1996. The primary focus of this study is on the impact that cash transfers have on recipients’ labour market participation. The focus is on these women, mainly to identify the causal effect of receiving a child support grant on their labour market participation. Location of focus was the New Brighton and Kwa Zakhele townships, since they have the highest number of social grant beneficiaries in Nelson Mandela Metro Municipality. An analysis of 101 questionnaires reveals that most of the grant recipients are either unemployed or underemployed. These beneficiaries do understand that the grant is meant to provide for the needs of the child concerned; however, because of their socioeconomic circumstances, the beneficiaries utilize the grant for other household purposes. Many of the respondents are actively job seeking but do not utilize the money for job seeking purposes; they mostly utilize it for household expenditure like groceries and electricity. The descriptive method was used to analyze the data and to find responses to the research questions and objectives. The results showed that the determinants of labour market participation by this group are multiple: for example, behavioral, loss of hope of ever finding a job, skills shortage or lack of work experience, lack of qualifications, and so on. Secondly, lack of government support for those who are keen to start self-generating projects leads to further despondency among those trying to find ways and means of generating income.

Do adults influence the information which children impart by the techniques they employ to obtain such information? Accepting the hypothesis that child witnesses are sources of information, the purpose of this study is to evaluate all aspects of the criminal justice system relating to child witnesses as well as available research on children to determine whether it is possible to obtain reliable and accurate information from children. Proposals will be formulated regarding the most successful methods of obtaining reliable and accurate information from children, and how these can be adapted for use by the legal system. The purpose of the study is to develop an approach to children in the legal system that will be fair to children while at the same time protecting the rights of the accused, and which will lead to a successful method of obtaining accurate information.

The choice of idols from a social psychological perspective

- Description: The study of adolescents’ idols has an over 100-year tradition. The meta-analysis of Teigen, Normann, Bjorkheim and Helland (2000) showed that idols, which are commonly understood as role models, changed over the last century which is attributed to changes in the social context. The present paper argues that Social Identity Theory (Tajfel & Turner, 1979, 1986) offers an appropriate theoretical framework to conceptualize social context by hypothesising a functional relationship between idols and identity management strategies moderated by the status position of the adolescent’s group s/he belongs to. The hypothesised functional relationship was tested in two studies with white and black adolescent South Africans. The results of the two studies supported our assumptions that the functional relationship between idols and identity management strategies is indeed moderated by status position. The results also indicate that Social Identity Theory seems to be an appropriate theoretical framework when social context is particularly conceptualised as social change.
