- Description: Drug metabolism is a specialised subset of xenobiotic metabolism, pertaining to the breakdown and elimination of pharmaceutical drugs. The enzymes involved in these pathways are the cytochrome P450 family of isozymes. Metabolism is an important factor in determining the pharmacological effects of drugs. The main aim of this study was to develop a system whereby the major metabolites of drugs can be produced in vitro. An in vitro system was developed and optimised using commercially prepared microsomes from rat liver and coumarin (by monitoring its conversion to 7-hydroxycoumarin) as a model. The optimum running conditions for the incubations were 50 μM coumarin, 50 μg protein/ml microsomes, 1 mM NADP⁺, 5 mM G6P and 1U/ml G6PDH incubated for 30 minutes at 38℃. The HPLC method for the detection of coumarin and 7-hydroxycoumarin was also validated with respect to linearity, reproducibility, precision, accuracy and lower limits of detection and quantification. The system developed was then tested using microsomes prepared from fresh bovine liver on these ten drugs of interest in doping control in horse racing: diazepam, nordiazepam, oxazepam, promazine, acepromazine, chlorpromazine, morphine, codeine, etoricoxib and lumiracoxib. The bovine liver microsomes were prepared using differential centrifugation and had activity on a par with the commercial preparations. This in vitro system metabolised the drugs and produced both phase I and II metabolites, similar to those observed in humans and horses in vivo. For example, the major metabolites of the benzodiazepine drug, diazepam, nordiazepam, temazepam and oxazepam as well as the glucuronidated phase II products were all found after incubations with the bovine liver microsomes. The metabolism of the drugs was also investigated in silico using the computational procedure, MetaSite. MetaSite was able to successfully predict known metabolites for most of the drugs studied. Differences were observed from the in vitro incubations and this is most likely due to MetaSite using only human cytochrome P450s for analysis.

- Description: This thesis looks at language as the medium of all our ideas and sentiments. The thesis represents a position statement regarding the development of isiZulu as an academic language. The field of Economics is used to merely illustrate and support the points that are being made in this work. It is argued that each language is viewed as the means of expression of the cultural heritage of its people, and it remains a reflection of cultural groups who speak that particular language. It is a fact that indigenous African Languages have been, for obvious reasons, blatantly understudied during the apartheid years in South Africa (Rudwick 2004). Languages have market value and the desirability of English as the most important global language today has an effect in most Black learners in South Africa. The study examined the development of indigenous African languages, isiZulu in particular, in Zululand University where ninety percent of students and lecturers are isiZulu first language speakers, and came to a conclusion that there is an urgent need for Black South African students to learn academic subjects through their mother tongues. This would help them in thorough understanding and interpretation of analytic text presented in a foreign language such as English. The study advocates that isiZulu, which is the mother-tongue of many students at the University of Zululand, should be a language for upward mobility to these students, but as the situation stands, isiZulu becomes a neglected or marginalised language since it is not the normal medium of instruction and it is not used for effective communication in their education system.

- Description: The search for the cure for cancer is currently a multi-billion dollar industry and the search for the elusive “magic bullet”, i.e. the perfect cancer drug that would interact therapeutically with cancerous tissues while having a minimal effect on healthy cells, is the topic of many research studies in the world today. A large number of novel drugs or drug complexes and conjugates are being synthesized and subjected to rigorous evaluation in the race to find the perfect cure. ECDG (Ethylene diCysteine DeoxyGlucose) seems to have promising cancer targeting ability. Even though this compound has been described in a few publications, we could not find any reference to the current use of ECDG in oncology clinics, either as a therapeutic agent, or as a diagnostic tool for imaging purposes. It was also not possible to purchase pure ECDG anywhere in the world. This prompted us to further investigate ECDG as a possible candidate for cancer targeting research, either as an imaging agent for cancer diagnosis or complexed with an anti-cancer agent for therapeutic purposes. Detailed investigations done in our laboratory can be divided into the following categories: - Development of a synthetic method for ECDG on a multigram scale ; - Purification of prepared ECDG not using the described dialysis method that only allows the purification of small quantities of ECDG (mg scale) ; Detailed investigation of the chemistry involved in the preparation of pure ECDG and its metal complexes ; - Investigation of the stability of ECDG and its metal complexes that is essential data required for any pharmaceutical agent ; - Preparation of ECDG complexes for use as a diagnostic tool, i.e. complexation with 99mTc ; Investigation of the bio distribution of ECDG-ReO complexes ; - Preparation of an ECDG kit as a diagnostic tool for use in oncology clinics. The development of novel aromatic ligands having similar characteristics compared to ECDG, containing an N2S2 chromophore as donor atoms, to further investigate their targeting capabilities, have also been investigated. All intermediates and final compounds were characterized mainly by ESI MS, in some cases IR and NMR whenever available. Successful preparation and purification of ECDG ands its metal complexes was achieved and extensively characterized and evaluated. Efforts directed towards the development of ECDG at NECSA, South Africa, were also rewarded with significant success. Furthermore, significant development regarding the synthesis of two novel compounds with ECDG-like characteristics was also completed.

Assuming that these actions are permissible, at what point will the judge's dialogue with counsel or line of questioning go beyond permissible limits? These are the questions with which this article grapples. Based on an analysis of the Constitutional Court decisions in State v Basson (2) 2007 (1) SACR 566 (CC) and Bernert v ABSA Bank Ltd 2011 (3) SA 92 (CC), and several Supreme Court of Appeal and other Commonwealth decisions, the article explores the circumstances in which the recusal of judges has been sought, or judicial decisions have been challenged on appeal on the basis of an allegation that there have been violations of the principle of fair hearing as enshrined in the Constitution. The article draws on the 'apprehension of bias' jurisprudence to establish the utility of the presumption of impartiality and the hybrid test of double-reasonableness in contexts where a judge's conduct is in question. The article concludes that the dialogue between the bench and bar is a useful component of adjudication in our adversarial system and should be limited by the rules of impartiality only in very exceptional circumstances.

- Description: This thesis reports on an analysis of the discursive construction of female and male physical identity in children’s literature and explicitly combines corpus linguistic methods with a critical discourse approach. Based on three novels from each of the Chronicles of Narnia and the Harry Potter series, it shows clear gendering of body parts, not only in terms of the purely quantitative preferences for certain body parts to be associated with one or other gender, but in terms of discourse prosody, or the uses to which the body parts are put. Human body parts in these series are mostly used in the following four ways, all of which show differences in realisation in terms of gender: · to describe individuals, physically, in order to distinguish one from the other; · to convey emotion, unintentionally as well as consciously; · for physical interaction between people and · for interaction with the world more broadly: responses to danger and agency, i.e. the ability to act on the world and the nature of what is achieved. The use of body parts by characters to express emotion and act agentively on the world is revealed to be strongly gendered in the two series. I characterise the most prominent patterns in terms of the bodily products blood, sweat and tears, of which the last is strongly connected to female characters, who are generally associated with emotion. The other two, referring to active participation in fighting and injury, as well as agency, are almost exclusively reserved for males, with female characters rendered unable to act on the physical world as a result of overwhelming feelings. The females’ response to danger suggests stereotyped discourses of inequality which see women and girls as requiring protection and being physically incapable. Thus gender is still a particularly salient aspect in these widely-read examples of children’s literature, despite plots which appear to be fairly positive towards women. The strength of the inclusion of a corpus approach in this study lies in its capacity to reveal objective, and often fairly covert, trends in language use. These in turn enrich the critical analysis of discourses in these influential texts, which facilitates social change through linguistic analysis.

The disempowering faces of the flexible firm: a case study of Gaborone Private Hospital

- Description: The purpose of the study was to investigate the ways in which the practice of t the Flexible Firm Model (FFM) disempowered the non-citizen professional nurses at Gaborone Private Hospital (GPH) in Gaborone, Botswana. GPH is a member of the Life Health care group of private hospitals based in South Africa. This study sought to probe the manner in which the practice of FFM influenced the employment relationship for non-citizen professional nurses. It also sought to examine the impact of the nature of the employment relationship on career development and representation and participation. Both qualitative and quantitative research methodologies were applied. Questionnaires were used to collect quantitative data. Thirty-five questionnaires were issued out to professional nursing staff and thirty-three responses were received. The response rate was ninety-four percent. The qualitative research data consisted of twelve in-depth interviews with professional nurses, the human resources manager , a professional nurse agency owner and a private clinic co-owner. Document analysis in the form of labour legislation, employee guides and employment contracts was used to validate data collected from the in-depth interviews. The results from the questionnaire revealed that an overwhelming majority of the professional nurses were Non-Citizens' dependent contractors. Although just under half of the participants held two or more previous contracts with GPH, an overwhelming majority had never been promoted. In addition, none of the participants were affiliated to the Botswana Nurses Association (BNA), the local equivalency of a professional nursing trade union. The in-depth interviews disclosed a sense of helplessness at their perceived disempowerment within the employment relationship, a poor career development and weak representation and participation. This study concluded that disempowerment at GPH was manifested through worker representation and participation and in effective skill upgrade. These forms of disempowerment were made worse by the global professional nurse shortage and maladministration of avail able human resources. Effective use of human resources and continued education could be employed to circumvent the adverse results of disempowerment.

This study sought to probe the manner in which the practice of FFM influenced the employment relationship for non-citizen professional nurses. It also sought to examine the impact of the nature of the employment relationship on career development and representation and participation. Both qualitative and quantitative research methodologies were applied. Questionnaires were used to collect quantitative data. Thirty-five questionnaires were issued out to professional nursing staff and thirty-three responses were received. The response rate was ninety-four percent. The qualitative research data consisted of twelve in-depth interviews with professional nurses, the human resources manager , a professional nurse agency owner and a private clinic co-owner. Document analysis in the form of labour legislation, employee guides and employment contracts was used to validate data collected from the in-depth interviews. The results from the questionnaire revealed that an overwhelming majority of the professional nurses were Non-Citizens' dependent contractors. Although just under half of the participants held two or more previous contracts with GPH, an overwhelming majority had never been promoted. In addition, none of the participants were affiliated to the Botswana Nurses Association (BNA), the local equivalency of a professional nursing trade union. The in-depth interviews disclosed a sense of helplessness at their perceived disempowerment within the employment relationship, a poor career development and weak representation and participation. This study concluded that disempowerment at GPH was manifested through worker representation and participation and in effective skill upgrade. These forms of disempowerment were made worse by the global professional nurse shortage and maladministration of avail able human resources. Effective use of human resources and continued education could be employed to circumvent the adverse results of disempowerment.

This was done to make a preliminary assessment of the conservation status of the rivers of this region. Aquatic insects from the orders Ephemeroptera, Odonata, Plecoptera, Megaloptera, Trichoptera and the dipteran family Simuliidae were collected using techniques to maximize the number of taxa found. The insects collected were identified to species level where possible. Water physicochemical parameters were recorded at all sites for each sampling trip to characterize these rivers and to establish a set of baseline data for future comparisons. These parameters included measurements made on site and analysis of the concentrations of all the major ions in water samples in the laboratory. Multivariate analyses including Principle Components Analysis and Canonical Correspondence Analysis were used to reveal patterns within the water physicochemistry and species distributional data. Results include the identification of 123 species from 70 genera and 30 families. A total of 31 species were found to be undescribed, of which 17 were not previously collected and are thus completely new to science. In addition, four of these species could not be placed into any known genus. The analysis of water physicochemistry showed a clear distinction between rivers of Table Mountain sandstone and Bokkeveld shale origin. Downstream effects of anthropogenic influences were discernable too. Distinct macroinvertebrate assemblages were found based upon stream order and water chemistry composition. pH proved to be the most important driver of invertebrate assemblage composition. The high levels of endemism of the macroinvertebrates found within the upper reaches of these rivers and their degree of ecological specialization make these systems a priority for the conservation of aquatic biodiversity on a national and global scale.

- Description: Plasmodium falciparum 1–deoxy–D–xylulose–5 phosphatereductoisomerase (PfDXR) plays a role in isoprenoid biosynthesis in the malaria parasite and is absent in the human host, making this parasite enzyme an attractive target for antimalarial drug design. To characterize PfDXR, it is necessary to produce large quantities of the enzyme in a soluble and functional form. However, the over–production of malarial proteins in prokaryotic host systems often results in the formation of truncated proteins or insoluble protein aggregates. A heterologous expression system was developed for the production of active PfDXR using codon harmonization and tight control of expression in the presence of lac repressor. Yields of up to 2 mg/l of enzyme were reported using the optimised expression system, which is 8 to 10– fold greater than previously reported yields. The kinetic parameters Km, Vmax and kcat were determined for PfDXR; values reported in this study were consistent with those reported in the literature for other DXR enzymes. A three–dimensional model of the malarial drug target protein PfDXR was generated, and validated using structure–checking programs and protein docking studies. Structural and functional features unique to PfDXR were identified using the model and comparative sequence analyses with apicomplexan and non–apicomplexan DXR proteins. Residues Val44 and Asn45, essential for NADPH binding; and catalytic hatch residues Lys224 and Lys226, which are unique to the species of Plasmodium, were mutated to resemble those of E. coli DXR. Interestingly,these mutations resulted in significant reductions in substrate affinity, when compared to the unmutated PfDXR. Mutant enzymes PfDXR(VN43,44AG) and PfDXR(KK224,226NS) also demonstrated a decreased ability to turnover substrate by 4–fold and 2–fold respectively. This study indicates a difference in the role of the catalytic hatch of PfDXR with regards to the way in which it captures substrates. The study also highlights subtle differences in cofactor binding to PfDXR, compared with the well characterized EcDXR enzyme. The validated PfDXR model was also used to develop a novel efficient in silico screening method for potential tool compounds for use in the rational design of novel DXR inhibitors. Following in silico screening of 46 potential DXR inhibitors, a two–tiered in vitro screening approach was undertaken. DXR inhibition was assessed for the 46 novel compounds using an NADPH– ependant DXP enzyme inhibition assay and antimalarial potential was assessed using P.falciparum–infected erythrocyte growth assays. Select compounds were tested in human cells in order to determine whether they were toxic to the host. From the parallel in silico and in vitro drug screening, it was evident that only a single compound demonstrated reasonable potential binding to DXR (determined using in silico docking), inhibited DXR in vitro and inhibited P. falciparum growth, without being toxic to human cells. Its potential as a lead compound in antimalarial drug development is therefore feasible. Two outcomes were evident from this work. Firstly, analogues of known antimalarial natural products can be screened against malaria, which may then lead towards the rational design of novel compounds that are effective against a specific antimalarial drug target enzyme, such as PfDXR. Secondly, the rational design of novel compounds against a specific antimalarial drug target enzyme can be untaken by adopting a coupled in silico and in vitro approach to drug discovery.

South Africa is the home of the largest number of people living with HIV/AIDS in the world. Historically, South Africa is one the countries that had a very disorderly past and this history is relevant to the explosive spread of HIV/AIDS in the region. The first cases of HIV were diagnosed in 1982 and that is when the first death from this disease was recorded. In 2004 over 5 million people out of a total 46 million South Africans were HIV positive, giving a total prevalence rate of 11 percent. HIV/AIDS has affected societies in many ways. Individuals, households and enterprises have all been affected by the pandemic. For individuals, incomes are lost as a result of HIV/AIDS. When individuals fall sick due to the disease, they loose their jobs and incomes. In most cases, these individuals are breadwinners in their households. The available money is then spent on medical services at the expense of other household investments. For example, the loss of a breadwinner inevitably affects rural households investments on farm inputs such as fertiliser, seeds etc. The result is low agricultural output and thus poverty. Dependent members of the households suffer immensely from the loss of income. Those at school are likely to drop out. When children drop out of school, they are more likely to be unemployed or underemployed. This makes them more vulnerable to being poor and other vices, such as crime or even HIV/AIDS. Poverty is another social and health issue that has a two-way relationship with HIV/Aids. Poverty increases individuals’ vulnerability to HIV/AIDS especially for females. Through its impact on productivity and loss of life of the economically productive members of the society, the disease condemns many to poverty. - Description: Maize is the staple food for most South Africans. This implies that any damage to the maize crop will affect food security of many South Africans. Although Eastern Cape Province is not a traditionally maize producing area, smallholder farmers in the province produce it mostly for subsistence purposes and some sell the surplus on the local market or use it to secure other good through barter trading. In South Africa, insect-resistant Bt maize/yieldgard has been used commercially for approximately 10 years now. Available impact studies on Bt maize reveal that, this technology is beneficial not only to farmers but consumers of maize products as well. Welfare gains as well as positive effects for human health are realised by both groups. Due to the costs and effectiveness associated with traditional and conventional maize stem borer control methods, Bt technology has the potential to be part of the solution. This thesis has attempted to investigate the economic viability of planting Bt maize seeds under smallholder farming conditions and identify factors as well as perceptions relating to attributes of Bt maize and to analyze the relationships between those perceptions and choices regarding use of Bt technology. Data was collected from 90 households who were selected using purposive sampling through the use of the snowball method. To collect data, a questionnaire was administered through face-to-face interviews. Gross margin analysis revealed that Bt maize is a more profitable option as compared to conventional maize seeds. Furthermore, econometric analyses, through use of the binomial regression model revealed that perceptions could be used to distinguish between users and non-users of Bt maize seed in the Eastern Cape Province. Results of inferential analysis indicate that the statistically significant variables at 5% level are gene erosion, quality and nutrition of products and food labels for Bt maize products perceptions. On the other hand, low expenses, seed market availability and farmers’ knowledge perceptions were significant at 10%. These findings suggest that an adjustment in each one of the significant variables can significantly influence the probability of Bt maize adoption. In view of the research findings, several policy proposals are suggested to support policy formulation. To collect data, a questionnaire was administered through face-to-face interviews. Gross margin analysis revealed that Bt maize is a more profitable option as compared to conventional maize seeds. Furthermore, econometric analyses, through use of the binomial regression model revealed that perceptions could be used to distinguish between users and non-users of Bt maize seed in the Eastern Cape Province. Results of inferential analysis indicate that the statistically significant variables at 5% level are gene erosion, quality and nutrition of products and food labels for Bt maize products perceptions. On the other hand, low expenses, seed market availability and farmers’ knowledge perceptions were significant at 10%. These findings suggest that an adjustment in each one of the significant variables can significantly influence the probability of Bt maize adoption. In view of the research findings, several policy proposals are suggested to support policy formulation. This is an absolute and should always be exercised under certain controlled conditions, as stipulated by the Labour Relations Act 66 of 1995. Convention 87 of the International Labour Organising (ILO) recognises the right of trade unions, as an organisation of workers set up to further and defend their interest (Article 10), to formulate their programs and organise their activities (Article 3); this means that unions have the right to negotiate with employers and to express their views on economic and social issues affecting the occupational interest of their members. This constitutes the position that the right to strike is one of the legitimate and indeed essential means available to workers for furthering and defending their occupational interest. Balanced against the right of every teacher to strike is the right of everyone to have a basic education as set out in section 29 of the Constitution. In terms of section 29(1)(a) everyone has a right, enforceable against the state, to basic education. This creates a strong positive right. Aspects of the right to education are found in human rights treaties and declarations. This right to education is contained in article 26 of the Universal Declaration of Human Rights (1948) which states that “everyone has the right to education”. The International Covenant of Economic, Social and Cultural v Rights of 1966 covers the right to education comprehensively, especially article 13 and 14. In 1989 the Convention on the Rights of the Child further confirmed this right. It is significant to note that in 2007 the Constitutional Court elevated the “best interest” principle to a right. - Description: Mussels are important ecological engineers on intertidal rocks where they create habitat that contributes substantially to overall biodiversity. They provide secondary substratum for other free-living, infaunal or epifaunal organisms, and increase the surface area for settlement by densely packing together into complex multilayered beds. The introduction of the alien invasive mussel Mytilus galloprovincialis has extended the upper limit of mussels on the south coast of South Africa, potentially increasing habitat for associated fauna. The aim of this study was to describe the structure of mussel beds, the general biodiversity associated with multi- and monolayered mussel beds of indigenous Perna perna and alien M. galloprovincialis, and to determine the relationship between mussels and epibiotic barnacles. This was done to determine the community structure of associated macrofauna and the role of mussels as biological facilitators. Samples were collected in Plettenberg Bay, South Africa, where M. galloprovincialis dominates the high mussel zone and P. perna the low zone. Three 15 X 15 cm quadrats were scraped off the rock in the high and low zones, and in the mid zone where the two mussel species co-exist. The samples were collected on 3 occasions. In the laboratory mussel-size was measured and sediment trapped within the samples was separated through 75 μm, 1 mm and 5 mm mesh. The macrofauna was sorted from the 1 mm and 5 mm sieves and identified to species level where possible. The epibiotic relationship between mussels and barnacles was assessed by measuring the prevalence and intensity of barnacle infestation and the condition index of infested mussels. Multivariate analysis was used on the mean abundance data of the species for each treatment (Hierarchical clustering, multidimensional scaling, analysis of similarity and similarity of percentages) and ANOVA was used for most of the statistical analyses. Overall, the results showed that tidal height influences the species composition and abundance of associated fauna. While mussel bed layering influenced the accumulation of sediments; it had no significant effect on the associated fauna. Time of collection also had a strong effect. While there was an overlap of species among samples from January, May and March, the principal species contributing to similarity among the March samples were not found in the other two months. The outcomes of this study showed that low shore mussel beds not only supported a higher abundance and diversity of species, but were also the most structurally complex. Although the condition index of mussels did not correlate to the percentage cover of barnacle epibionts, it was also evident that low shore mussels had the highest prevalence. The levels of barnacle infestation (intensity) for each mussel species were highest where it was common and lowest where it was least abundant. This is viewed as a natural artefact of the distribution patterns of P. perna and M. galloprovincialis across the shore. Mussels are more efficient as facilitators on the low mussel zone than the high mussel zone possibly because they provide habitats that are more effective in protecting the associated macrofauna from the effects of competition and predation, than they are at eliminating the effects of physical stress on the high shore. Although mussels create less stressful habitats and protect organisms from the physical stress of the high shore, there are clear limitations in their ability to provide ideal habitats. The biological associations in an ecosystem can be made weak or strong depending on the external abiotic factors and the adaptability of the affected organisms.

- Description: The use of livestock guarding dogs (LGDs) to mitigate farmer-predator conflict in Namibia was evaluated. As farmer-predator conflict has two sides, LGDs were evaluated in terms of livestock production and conservation. The main objectives in terms of livestock production were to document: 1) the perceived ability of LGDs to reduce livestock losses in a cost-effective manner; 2) the farmers’ satisfaction with LGD performance; and 3) factors influencing LGD behaviour. The main objectives in terms of conservation were to record: 1) predator killing by farmers relative to LGD introduction; 2) direct impacts of LGDs on target (damage-causing) species; and 3) the impact of LGDs on non-target species. This evaluation was conducted on LGDs bred by the Cheetah Conservation Fund (CCF) and placed on farms in Namibia. The data were collected during face-to-face interviews with farmers using LGDs. Historical data from the CCF programme were used in conjunction with a complete survey of the farmers in the CCF LGD programme during 2009-2010. In terms of livestock production, 91 percent of the LGDs (n = 65) eliminated or reduced livestock losses. Subsequently, 73 percent of the farmers perceived their LGDs as economically beneficial, although a cost-benefit analysis showed that only 59 percent of the LGDs were cost-effective. Farmers were generally satisfied with the performance of their LGDs. However, farmer satisfaction was more closely linked to good LGD behaviour than the perceived reduction in livestock losses. The most commonly-reported LGD behavioural problems (n = 195) were staying at home rather than accompanying the livestock (21 percent) and chasing wildlife (19 percent). LGD staying home behaviour was linked to a lack of care on subsistence farms, as high quality dog food was not consistently provided. Care for LGDs declined with LGD age on subsistence, but not commercial, farms. In terms of conservation, predator-killing farmers killed fewer individuals in the year since LGD introduction than previously; this result was only significant for black-backed jackal Canis mesomelas. However, 37 LGDs killed jackals, nine killed baboons Papio ursinus, three killed caracals Caracal caracal and one killed a cheetah Acinonyx jubatus (n = 83). Farmers and LGDs combined killed significantly more jackals in the survey year than the same farmers (n = 36) killed before LGD introduction. Conversely, five farmers killed 3.2 ± 2.01 cheetahs each in the year before LGD introduction, whereas LGDs and these farmers combined killed only 0.2 ± 0.2 cheetahs per farm in the survey year. Only 16 LGDs (n = 83) killed non-target species. The high LGD success rate in terms of livestock production was facilitated by livestock husbandry practices in the study area. In terms of conservation, LGDs were more beneficial for apex predators than for mesopredators and had a minor impact on non-target species.

The study applied quantitative and qualitative approaches for biographical and socio economic information; and for in-depth understanding and verification respectively. Data collection was through structured interviews and survey questionnaires. The findings reveal that there are random, short-term poverty alleviation initiatives taking place in Nompumelelo informal settlement area. Projects that could have been self-sustaining collapsed due to lack of skills and training. The study indicates that the level of unemployment is high at 68 percent.

Productivity growth is viewed by many executives and business leaders as the key element in maintaining competitiveness in the global economy. The success of the South African manufacturing and its global competitiveness lie in its productivity. By focusing on promoting productivity, South African manufacturers will establish a source of sustained competitive gains, while labour will establish a sustainable source of income growth. The Eberspacher South Africa Port Elizabeth (ESA) is not in any way immune to the level of competition that impacts all businesses in South Africa. ESA not only competeswith other local catalytic converter companies in terms of a high level of productivity, but also with other manufacturing plants within the Eberspacher Group globally. In Eberspacher all exhaust manufacturing plants are constantly benchmarked, comparing productivity levels, quality levels, cost and delivery. This data is used not only to determine goals, but also forms a key determinant as to where any new business is placed. The internal competition encourages all manufacturing plants within Eberspacher to continously improve plant productivity within group. A company is competitive if it is able to produce products that are either cheaper or better than those of other firms. Most of ESA customers are internal final assembly plants from Europe, USA, India and China. ESA therefore constantly have to ensure that they are competitive in all aspects of business, including productivity. In ESA, the overall productivity performance of the manufacturing plant is of utmost importance to ensure the ability to supply all the customers with cost competitive products, at the right time and with zero defects. Productivity growth is viewed by many executives and business leaders as the key element in maintaining competitiveness in the global economy. The success of the South African manufacturing and its global competitiveness lie in its productivity. By focusing on promoting productivity, South African manufacturers will establish a source of sustained competitive gains, while labour will establish a sustainable source of income growth. The Eberspacher South Africa Port Elizabeth (ESA) is not in any way immune to the level of competition that impacts all businesses in South Africa. ESA not only competeswith other local catalytic converter companies in terms of a high level of productivity, but also with other manufacturing plants within the Eberspacher Group globally. In Eberspacher all exhaust manufacturing plants are constantly benchmarked, comparing productivity levels, quality levels, cost and delivery. This data is used not only to determine goals, but also forms a key determinant as to where any new business is placed. The internal competition encourages all manufacturing plants within Eberspacher to continously improve plant productivity within group. A company is competitive if it is able to produce products that are either cheaper or better than those of other firms. Most of ESA customers are internal final assembly plants from Europe, USA, India and China. ESA therefore constantly have to ensure that they are competitive in all aspects of business, including productivity. In ESA, the overall productivity performance of the manufacturing plant is of utmost importance to ensure the ability to supply all the customers with cost competitive products, at the right time and with zero defects. The PA-SWCNT and MTAPc modified electrodes were applied as hybrids for electrochemical sensing of H2O2. The step-by-step fabrication of the electrode was followed using electrochemistry, impedance spectroscopy, scanning electron microscopy and Raman spectroscopy and all these techniques confirmed the fabrication and the immobilization of PA-SWCNT, MnTAPc and CoTAPc onto gold surfaces. The apparent electron transfer constant (kapp) showed that the carbon nanotubes and metallo-phthalocyanines hybrids possess good electron transfer properties compared to the bare, pre-grafted and the MTAPc modified gold electrode surfaces without PA-SWCNT. The electrochemical sensing of hydrogen peroxide was successful with PA-SWCNT–MTAPc hybrid systems showing higher electrocatalytic currents compared to the other electrodes. The analytical parameters obtained using chronoamperometry gave good linearity at H2O2 concentrations ranging from 1.0 to 30.0 μmol L−1. The values for the limit of detection (LoD) were found to be of the orders of 10−7 M using the 3δ for all the electrodes. The PA-SWCNT–MTAPc modified SPAuEs were much more sensitive compared to PA–MTAPc modified SPAuEs.

- Description: The Republic of South Africa emerged as 'a product of a historical moment' and also as a reaction against imperialism, colonialism, racial discrimination and domination over the majority of black South Africans by the white minority. The democratic dispensation came into being also for the first time in its life in memorial for a long turbulent history followed by the general democratic elections held on 27th April 1994. The 1994 first general election liberated South Africa from the apartheid system and its subsequent primary objective was “… to transform South Africa into a non-racial and democratic society”. The new democratic government now looks politically different from the racist regime because the current government since 1994 has been, and to date still is, a truly and broadly representative of the South African citizens and also a transparent one, whereas the defunct apartheid government was characterized primarily by, among other things, the violation of human rights, denying black South Africans of any rights of basic services, no rights of owning property or land, no freedom of association and speeches and firmly practised discrimination which was detrimental to the majority of the black population groups in this country. - Description: Herbal remedies are commonly used in developing countries for the treatment of various diseases, including constipation. The rationale for utilizing medicinal plants for the treatment of diseases rested largely on the belief that they are safe and free of side effects. However, there is limited scientific evidence on the safety and efficacy of these herbal medicines to back up their continued therapeutic application. Aloe ferox Mill. (Aspodelaceae), known as Cape aloe, locally called ikhala is a medicinal plant used by the people of the Eastern Cape Province for the treatment of gastrointestinal problems and constipation. The plant is a perennial shrub with thick succulent leaves bearing brown thorns on the margin and bright orange flowers arranged in oval lanceolate. It occurs in all weather in bush veld, road side, gardens and undisturbed places. According to the ethnomedicinal information, A. ferox is used as purgative. This research project was therefore designed to evaluate its ability in the treatment of constipation and to investigate its possible toxicological property. At the beginning of this programme, a survey of plants used for the treatment of constipation in Nkonkobe Municipality of the Eastern Cape Province was carried out using a questionnaire, which was administered to herbalists, traditional healers and rural dwellers. The study revealed 10 plant species from 8 families that are used for the treatment of constipation in the province. Four plants, Aloe ferox Mill, Boophone distischa L.f Herb, Alepidea amatybica Eckl and Artemisia afra Jacq, were repeatedly mentioned. Based on the frequency of usage, perceived efficacy and availability to the rural dwellers and the traditional healers, Aloe ferox was the most commonly used of the plants for the treatment of constipation. The plant was thus, chosen for the study. The invivo laxative effect of the aqueous extract of Aloe ferox in the treatment of loperamide-induced constipation in Wistar rats was investigated at varying concentrations. The leaf extract at all the dosages investigated (50, 100 and 200 mg/kg body weight) improved intestinal motility, increased fecal volume and normalized body weight in the constipated rats. This was an indication of its laxative properties. However, the laxative property of the herb at 200 mg/kg body weight of the extract showed best efficacy and compares favourably well with senokot, a standard laxative drug. These findings have therefore, lent scientific credence to the folkloric use of the herb by the people of the Eastern Cape of South Africa as a laxative agent. Toxicological evaluation of aqueous leaf extract of Aloe ferox in loperamide-induced constipation was studied at 50, 100, and 200 mg/kg body weight. The oral administration of the extracts did not show any significant effect on the liver and kidney body weight ratios as well as the kidney and liver function indices. The extracts, at all the dosages investigated, did not alter the levels of creatinine, uric acid, urea, calcium and potassium ions. Similarly, the levels of total protein, albumin, bilirubin and gamma glutamyl transferase (GGT) were not significantly different from the control. The plant extract appreciably normalized the elevated activities of alkaline phosphatase (ALP), alanine transaminase (ALT) and aspartate transaminase (AST) in the untreated constipated rats following treatment with the extract. The extract did not show a significant effect on the hematological parameters except for the increase in the lymphocyte count in the untreated constipated rats, which was attenuated after administering the herb. ThThe available evidence in this study suggests that A. ferox may be safe as an oral remedy for constipation. Generally, the effect of the extract compared favourably well with senokot, a recommended drug for the treatment of constipation. The antioxidant activities against 1, 1 diphenyl- 2 picrylhydrazl (DPPH), 2,2’ – azinobis [3- ethylbenzothiazoline -6- sulfonic acid] diammonium salt (ABTS), hydrogen peroxide (H2O2), Nitric oxide (NO), lipid peroxidation and the ferric reducing agents were investigated spectrophotometrically. Alkaloids, saponins, tannins, total phenols, flavonoids, flavonols and proanthocyanidin were also determined to assess their effects on the antioxidants activity of this plant. The phytochemical content of the ethanol and acetone extracts were consistently high compared to other solvents extracts. The level of tannins was not significant (P > 0.05) as compared with other solvent extracts. The free radical scavenging activity of the extracts was high even at lower concentrations (0.025 mg/ml) except in DPPH and lipid peroxidation. The ferric reducing potential of the extracts was concentration dependent and significantly different from Vitamin C and butylated hydroxytoluene (BHT) that were used as standard drugs. The present study showed a high level of scavenging activity of the leaf extracts of Aloe ferox in all the solvent extracts. Both ethanol and methanolic extract showed potent antioxidant activities than acetone and aqueous extracts. The study indicated that the leaf extracts of Aloe ferox might be a valuable source of natural antioxidant for both medicine and food industries. A. ferox leaf consists of the gel, latex and mesophyll layer; however, the main active constituents of the latex and the leaf exudate of Aloe ferox are anthraquinones which are believed to be responsible for the laxative property. The laxative compound in Aloe ferox leaf extract was isolated and characterized by extracting the plant material in methanol and extract suspended in distilled water. Partitioning was done with n-hexane, ethyl acetate and butanol respectively and was co-spotted with the over-the-counter (OTC) laxative drugs. This led to the successive column chromatography and thin layer chromatography (TLC) of the most active ethyl acetate fraction on silica gel with benzene/ethanol/ammonia hydroxide (BEA: 90:10:1), ethyl acetate/methanol/water (EMW: 40:5.4:5) and chloroform/ethyl acetate/formic acid (CEF: 50:40:10) as the mobile phase. The successive chromatograph and TLC afforded two compounds of Rf 0.420 (blue) and 0.831 (yellow) with the over-the-counter (OTC) drugs. These compounds were not totally elucidated due to their small quantity and instability. However, hydroxyl (OH) and carboxyl groups (COOH) was established as common to the extracted compounds, which might be responsible for the biological activity recorded for the plant extract.
