Electrospun nanofibers : an alternative sorbent material for solid phase extraction

- Description: The work described in the thesis seeks to lay a foundation for a better understanding of the use of electrospun nanofibers as a sorbent material. Three miniaturised electrospun nanofiber based solid phase extraction devices were fabricated. For the first two, 10 mg of electrospun polystyrene fibers were used as a sorbent bed for a micro column SPE device (8 mm bed height in a 200 μl pipette tip) and a disk (I) SPE device (5 mm 1 mm sorbent bed in a 1000 μl SPE barrel). While for the third, 4.6 mg of electrospun nylon nanofibers were used as a sorbent bed for a disk (II) SPE device, (sorbent bed consisting of 5 5 mm 350 μm stacked disks in a 500 μl SPE barrel). Corticosteroids were employed as model analytes for performance evaluation of the fabricated SPE devices. Quantitative recoveries (45.5-124.29 percent) were achieved for all SPE devices at a loading volume of 100 μl and analyte concentration of 500 ng ml-1. Three mathematical models; the Boltzmann, Weibull five parameter and the Sigmoid three parameter were employed to describe the break through profiles of each of the sorbent beds. The micro column SPE device exhibited a breakthrough volume of 1400 μl, and theoretical plates (7.98-9.1) while disk (I) SPE device exhibited 400-500 μl and 1.39-2.82 respectively. Disk (II) SPE device exhibited a breakthrough volume of 200 μl and theoretical plates 0.38-1.15. It was proposed that the formats of future electrospun nanofiber sorbent based SPE devices will be guided by mechanical strength of the polymer. The study classified electrospun polymer fibers into two as polystyrene type (relatively low mechanical strength) and nylon type (relatively high mechanical strength).

The notion of scientific literacy in South Africa has emerged largely due to the government’s recognition of the role that science and technology plays in economic growth, employment creation, social redress and social development. However, in light of South Africa’s learner performance on international and national assessments such as TIMMS (2003) and PIRLS (2006), as well as the problems of teaching and learning in a second language, there appears to be a primary and pressing need to develop learners’ fundamental sense of scientific literacy (Norris & Phillips, 2003). Expanding learners’ ability to read, write and communicate in science may provide the necessary framework for engaging learners in the critical principles and foundations of the scientific endeavour (Hand, Prain, & Yore, 2001). As such, this study focuses on equipping and training grade six and seven science teachers to develop scientifically literate learners via professional development workshops with a strategy that supports reading, writing, talking and conducting (‘doing’) science through scientific investigations. The typology of triangulation and the mixed method research approach was supported by a fully mixed, concurrent, and equal status design (Leech & Onwuegbuzi, 2007). Quantitative data were collected from the baseline and post-intervention testing of learners’ problem solving skills, as well as their literacy skills in English and isiXhosa. Qualitative measures were generated through classroom observations, teacher interviews and learners’ science notebooks. The study was conducted in two different milieus in the Eastern Cape, South Africa. The first setting, in the rural area of Tyumie Valley near the Hogsback Mountains, was comprised of a sample of grade six and seven (multi-grade classrooms) teachers (n=7) and learners (n=168) from five experimental schools and two comparison schools. The second setting, in the urban townships area east of Port Elizabeth, was comprised of a sample of grade six teachers (n=8) and learners (n=675) from six experimental schools and two comparison schools. Mean differences between the experimental and the comparison groups were computed for the Raven’s Standard Progressive Matrices (RSPM) and the literacy tests, and the data generated were treated with an Analysis of Covariance (ANCOVA). The data suggest that the scientific literacy strategy improved the experimental learners’ problem solving skills. Both experimental groups demonstrated greater gains than that of the comparison schools. However, statistically significant improvements were only detected in Port Elizabeth. Improvements in learners’ literacy skills in isiXhosa and English varied according to each milieu. While the teachers initially identified challenges to learners’ reading and writing abilities, the analysis of learners’ science notebooks suggested that they used writings to support their investigations. Some teachers cited difficulties with certain aspects of the model, such as problems with developing an investigable question and argumentation, yet overall, teachers found the strategy useful for developing learners’ language skills, as well for strengthening their pedagogical practices in science. Teachers’ gradual improvements in the use of the model suggest that they were able to use the scientific literacy strategy to support the cognitive and linguistic development of second-language learners.

Ecological assessment and biogeography of coastal vegetation and flora in southern Mozambique

- Description: This study considered aspects of the vegetation of the coastal zone of southern Mozambique, an area with a rich biodiversity. The vegetation and flora were assessed in their current state including the associated anthropogenic pressures. The environmental conditions that determined the distribution of the vegetation were evaluated. A hierarchical classification, description and ecological interpretation of the vegetation and flora of the coastal zone in southern Mozambique are presented. Analysis was based on 242 sample quadrats that were distributed in a stratified manner throughout the study area. The vegetation could be grouped into six distinct types of vegetation: Dune Forest, Coastal Forest, Coastal Grassland, Coastal Miombo, Coastal Savanna and Coastal Woodland. Species diversity was high for the all the vegetation types, particularly in the woody elements. A total of 673 species in 410 genera was recorded. They represented 104 families with Fabaceae and Rubiaceae being the most common. Of these, 6.6% were endemic or near-endemic to the coastal zone of southern Mozambique with Coastal Forest being the habitat with the greatest endemism. All soils from different vegetation types had poor agricultural potential. Soil properties were the strongest defining environmental feature separating the vegetation types: e.g. pH was high in Dune Forest compared to the others. Results from Maxent modelling suggest that the distribution of endemic species is influenced by a combination of climatic and non-climatic variables. Soil type, temperature annual range and precipitation of the driest month were the most important predictor variables. Overlaying the potential distributions of the seven selected species indicated two areas of abundance of endemic species – these should be given attention for conservation. Endemic species are not well protected in southern Mozambique – their sampled and potential habitats are largely outside protected areas. Hence, additional reserves should be created to improve their protection. Most endemic and near-endemics species were found in the south, from Ponta de Ouro (Matutuine, south of Maputo Province) to Manhica district (north of the Maputo Province) forming part of the Maputaland Centre of Endemism. A second concentration of endemism was found in the Inhambane Province, specifically the Inhassoro and Vilanculos districts. This is proposed to be an Important Plants Area (IPA). Because most endemics and near-endemics are found in the Coastal Forest, their main threat is harvesting for charcoal production, although none of the endemic species are specifically targeted for charcoal production. In Inhambane they are also threatened by the tourism industry, agriculture and settlements. The impact of the habitat destruction on endemic species is expected to cause severe declines in the near future. The tourism industry and harvesting of trees for charcoal production and over-frequent fires are the main drivers of vegetation loss in this region. Shifting agriculture, harvesting for firewood and construction materials, cattle grazing (at a minor scale) are also impacts, but these were only observed in a small area of Maputo Province. Although the study was done in coastal zone of southern Mozambique, effective management of whole coastal zone of the country will be required to maintain a functioning and diverse ecosystem. Priorities for management are to ensure that forests are protected, in particular, Dune Forest. Actions are required to minimise degradation of coastal vegetation. Further research on Coastal Forest restoration should be planned, as field observations in this study confirm that forest in the coastal zone of southern Mozambique has resilience.

- Description: This study has involved the design, synthesis and evaluation of novel HIV-1 enzyme inhibitors accessed by synthetic elaboration of Baylis-Hillman adducts. Several series of complex coumarin-AZT and cinnamate ester-AZT conjugates have been prepared, in high yields, by exploiting the click reaction between appropriate Baylis-Hillman derived precursors and azidothymidine (AZT), all of which have been fully characterised using spectroscopic techniques. These conjugates, designed as potential dual-action HIV-1 inhibitors, were tested against the appropriate HIV-1 enzymes, i.e. HIV-1 reverse transcriptase and protease or HIV-1 reverse transcriptase and integrase. A number of the ligands have exhibited % inhibition levels and IC50 values comparable to drugs in clinical use, permitting their identification as lead compounds for the development of novel dual-action inhibitors. In silico docking of selected ligands into the active sites of the respective enzymes has provided useful insight into binding conformations and potential hydrogen-bonding interactions with active-site amino acid residues. A series of furocoumarin carboxamide derivatives have been synthesised in four steps starting from resorcinol and these compounds have also been tested for HIV-1 integrase inhibition activity. The structures of unexpected products isolated from Aza-Baylis-Hillman reactions of N-tosylaldimines have been elucidated by spectroscopic analysis, and confirmed by single crystal X-ray analysis. A mechanism for what appears to be an unprecedented transformation has been proposed. Microwave-assisted SeO₂ oxidation of Baylis-Hillman-derived 3-methylcoumarins has provided convenient and efficient access to coumarin-3-carbaldehydes, and a pilot study has revealed the potential of these coumarin-3-carbaldehydes as scaffolds for the construction of tricyclic compounds. The HCl-catalysed reaction of tert-butyl acrylate derived Baylis-Hillman adducts has been shown to afford 3-(chloromethyl)coumarins and α-(chloromethyl)cinnamic acids, the Zstereochemistry of the latter being established by X-ray crystallography. ¹H NMR-based experimental kinetic and DFT-level theoretical studies have been undertaken to establish the reaction sequence and other mechanistic details. Base-catalysed cyclisation on the other hand, has been shown to afford 2H-chromene rather than coumarin derivatives.

- Description: This study focused on community development education and practice for the democratic and transformative South context. Social and community development are relatively new approaches to social welfare and community development is emerging as a discipline and profession to contribute to the goal of developmental social welfare. Community work, albeit to a minimal extent and not community development was viewed as a method of social work during apartheid. With community development being prioritized as an intervention strategy for poverty reduction by the national government, the gap in community development education needs to be addressed. The goal of the study was to develop an undergraduate curriculum framework for community development education that would produce a new generation of ethical community development professionals to contribute to the goal of developmental social welfare. The study focused on the integrated approach to community development education and practice; with education of the student as a whole human being as critical for community development in a transformative South Africa. To achieve its goal, the study identified three themes that were critical to the research: a conceptual framework, clarifying key terms; a curriculum framework for community development education; and qualities and ethics for a new generation of development practitioners. The qualitative study was based in the Eastern Cape Province, South Africa and because of the lack of a referential framework for community development education, the researcher looked towards learning from India which has over fifty years of experience in social and community development education and practice. The state of Kerala established a social development approach in 1957 and was chosen as a learning experience. The state’s Kudumbashree Mission which aims at poverty alleviation through the empowerment of women was chosen as a case study. The integral and values based education system of Sri Sathya Sai University in the state of Andhra Pradesh and the community development education and practice of Loyola College in the state of Kerala were examined. The research design was exploratory, conceptual, descriptive and developmental in nature. The qualitative methodology best suited the nature of this study and a combination of methods to obtain data from a variety of sources across national boundaries was employed. The participants interviewed in both countries were purposively selected for their involvement in social and community development directly as target groups, practitioners, managers and policy makers. The research produced a philosophical and theoretical framework that is unitary and integral and aligned with the current social development policy to guide community development education and practice. The unitary and integral framework encompasses the individual within the family and community context and locates the various dimensions of development such as the social, physical, cultural, psychological, spiritual, political and economical dimensions within a unitary platform. The framework is dominated by the holistic and humanistic philosophies without negating the rational and pragmatic perspectives. In conclusion, the study conceptualizes community development as a natural process; the integration of individual and collective consciousness and an intervention. The outcome of the study is a recommended curriculum framework for community development education that would be of relevance to the Eastern Cape Province in particular and for the South African context generally.

This is close to the yield predicted by several empirical models and the fishery now appears to be entering a phase where increases in effort will not be matched by increased yields. The mechanisms for controlling effort in the fishery appear to be inadequate and particular concern was expressed about the lack of co-ordination between the Zimbabwean and Zambian fishery authorities in planning the development of this shared resource.

Ecology of key cerithioidean gastropods in the mangroves of the iSimangaliso Wetland Park, KwaZulu-Natal South Africa

- Description: Gastropods are one of the most diverse species groups in mangrove habitats, however, many of their specific roles in relation to ecological patterns and processes are currently largely unknown. The overall aim of this research project was to provide basic ecological information for key gastropod species from subtropical mangroves within a protected area. South African mangroves cover relatively small areas and are restricted to estuaries, these habitats therefore present unique opportunities and challenges to the species that occur in them. Three gastropod species, Terebralia palustris, Cerithidea decollata, and Melanoides tuberculata, all occur at their natural southernmost range limit within South Africa and were selected based on their prominence and occurrence in mangrove habitats of the iSimangaliso Wetland Park, a UNESCO World Heritage Site. Trophic linkages and resource partitioning, resource utilization rates, and ecological resilience were investigated respectively using: 1) a stable isotope (δ15N and δ13C) approach; 2) an experimental approach to quantify feeding dynamics (ingestion rate, consumption/digestion efficiency and grazing impact); and 3) a mixed-effects modelling approach to relate population responses to environmental variables. The diet of T. palustris was seasonally variable and a number of sources were incorporated by different sized snails, but their grazing impact on microphytobenthos was not significant. The results also indicated an ontogenetic shift in the dietary niche for T. palustris through robust partitioning of resources between different size classes. The diets of C. decollata and M. tuberculata were dominated by different primary resources as a function of where they occurred in the mangroves. Melanoides tuberculata consumed a wide variety of primary resources, a typical trait of an opportunistic generalist species. The ingestion rate of M. tuberculata was not dependent on the availability of microphytobenthos, and was highest when conditions were oligotrophic. The resilience of C. decollata was related to the tree-climbing behaviour of this species and its occurrence was best explained by sediment conductivity. These responses were considered in conjunction to what has previously been reported on the resilience of the mangrove trees. The results of this research project have provided new basic ecological information for all three gastropod species in this data-deficient subtropical region. This information can potentially be used in comparative studies for these species in other regions or in broader scale ecological studies. Terebralia palustris has recently experienced a range contraction along the South African coastline. This research project has shown that the diet of this species is highly variable and that food limitation and competition for resources should be considered as potential drivers of the local decline. Cerithidea decollata has in contrast expanded its distributional range in this region. This research project has shown that this species has a generalist diet and exhibits traits in relation to tolerance that are expected to have facilitated its expansion into temperate saltmarsh habitats that occur in dynamic estuaries. Melanoides tuberculata is a globally invasive species, and as South African populations are within its native range, ecological information from this region is valuable as it can be used to investigate the potential ecological effects following introduction into new habitats beyond the native range. Biological drivers have a significant impact on mangrove ecosystem functioning, particularly in relation to recycling and the retention of organic carbon generated through primary productivity. Understanding the ecological linkages that maintain ecological functioning and stability is therefore an important step towards conserving and sustainably managing threatened ecosystems such as mangrove forests.

The growth and reproduction of Patella granularis (Mollusca : patellogastropoda) on the south-east coast of South Africa

- Description: Aspects of the biology and ecology of Patella granularis were investigated along a 130km stretch of the south-east coast of South Africa. Distribution, biomass, density and population structure were investigated at seven localities. In addition, a more detailed study of the growth rate and reproductive biology of populations inhabiting three different substrata (aeolianite, quartzitic sandstone and mussel shells) was conducted. The genetic relationships between these three populations was also examined, as was the foraging behaviour of the limpets inhabiting an aeolianite and a quartzitic sandstone shore. Finally, differences in food availability on the different substrata were studied. On the south-east coast, P. granularis has a wide intertidal distribution, occurring from the upper Balanoid zone through to the Cochlear zone, where it is a common inhabitant of mussel shells. The mean shell length of P. granularis was found to decrease down the shore. The largest limpets (46.6 mm shell length) were found on an offshore island in Algoa Bay. At most localities investigated, the sex ratio deviated from a 1:1 ratio with more males than females being recorded on five shores. Both limpet density and biomass were lower on the south-east coast when compared to data published for west coast populations. On the south-east coast, both density and dry biomass were highest in the lower Balanoid zone. Allozyme electrophoresis indicated that P. granularis inhabiting aeolianite, quartzitic sandstone and mussel shells are all part of a single population. Extremely high genetic identity values (0.998), low levels of heterozygosities (0.035 - 0.061), low levels of polymorphisms (25% - 31%) and low FST values (0.021) all suggest that the three populations of P. granularis form a common breeding group, despite the high levels of phenotypic plasticity observed. On all shores, P. granularis was found to grow allometrically, increasing in shell height more rapidly than shell length. Estimation of the growth rate (determined by the Von Bertalanffy growth model) of P. granularis suggested that limpets inhabiting the mussel shells grew more slowly, and attained a smaller maximum size, than those inhabiting both the aeolianite and the quartzite (K = 0.25, 0.32 and 0.33 respectively; 27.12 mm, 31.89 mm and 32.96 mm respectively). Previous work has shown that west coast P. granularis grow more quickly (K = 0.7) and reach a greater size (. 40 mm). Translocation of limpets among sites suggested that limpet size in the mussel beds was spatially constrained. Shell microgrowth bands were deposited tidally, but could not be used for aging limpets due to shell erosion. Limpets from the aeolianite had the greatest reproductive fitness, producing more eggs (.366 000/limpet) than those inhabiting quartzite (.119 500/limpet) or mussel shell limpets (.85 800/limpet). Aeolianite limpets also spawned throughout the year, whereas those from the quartzite and mussel shells spawned twice a year (once in winter and once in summer) although a great deal of interannual variability was observed. The onset of sexual maturity occurred at a similar age in all limpets (1 - 2 years) and is probably genetically entrenched. P. granularis inhabiting both an aeolianite and a quartzitic shore were active during nocturnal low tides. All limpets returned to a home scar after foraging. Whilst foraging, limpets inhabiting the aeolianite shore moved shorter distances (.17 cm) at a slower rate than those from quartzite (.30 cm). Limpets that were translocated from one substratum to the other initially moved similar distances to the source group, but after a maximum period of one week, moved distances that mirrored those moved by the resident limpets. Neither season nor tidal phase influenced the distances foraged. No directionality in foraging was found. Wear of radula teeth, particularly the pluricuspid tooth, was greater in limpets from the quartzite. It is hypothesised that the observed differences in life-history parameters and foraging behaviour of limpets both within the south-east coast and between the west and south-east coasts are related to food abundance. Chlorophyll-a, and hence microalgal biomass, was consistently higher on aeolianite (.2.5 times) than on both quartzite and mussel shells. Estimates of chlorophyll-a were higher (although not significantly) in winter. Previous studies determined that primary productivity is also higher along the west coast. Finally, the lack of evidence for migration of limpets from the low-shore to high-shore in south-east coast P. granularis is discussed. It is suggested that this species settles opportunistically within its physiological tolerances and responds morphologically to localised environmental conditions.

A management model for human settlements: a case study of Nigeria and South Africa

- Description: Persistent depreciating human settlements is a global challenge, particularly in developing countries and this challenge contributes to the continuously increasing housing gap, despite several novel policies and programmes. Although there is a dearth of research-based literature on human settlements management, studies show that property management, Facility Management, housing management and urban management impact the sustainability of all these built environment milieus which form part of the human settlements’ sub-components. This research aimed at altering the afore-said challenge by exploring the management of human settlements in Nigeria and in South Africa. Philosophically, the research is pragmatic, and it rests on positivist deductive reasoning. The methods used for data collection include a review of literature, collection of archival records, and empirical studies of purposively selected professionals and stakeholders involved in human settlements’ management in Nigeria and in South Africa. The findings indicate that existing human settlements in the case studies are “good” and “acceptable” with the majority adopting an outsourced method of management. Furthermore, the lack of maintenance records, the deficiency in policy monitoring and the training of management and maintenance personnel were the three highest ranking factors indicated to be affecting human settlements management. Moreover, an inhabitant’s income, the time available for management, professional expertise and technology employed for management are recognised as the critical sustainability sub-factors affecting human settlement management. The factor analysis established ownership, human resource and capital, social, legal, technology, economic, physical and environmental factors as vital and very significant in achieving sustainable management of human settlements. The study recommends the professionalisation of the field of human settlements as well as a formulation of management policy among others. The study developed a management model for human settlements, and it advocates that human settlements managers, owners and organisations should apply the model in this study, to improve their human settlement management practices, so as to achieve sustainability, and in essence, Sustainable Development Goal 11. It is hoped that the suggested management model will influence human settlement policy development and review, the designing of human settlement management programmes including the framework for their ongoing monitoring and evaluation. The study furthermore contributes to the development of housing and human settlement curricula and evolving human settlement research in both developed and developing countries.

- Description: Persistent depreciating human settlements is a global challenge, particularly in developing countries and this challenge contributes to the continuously increasing housing gap, despite several novel policies and programmes. Although there is a dearth of research-based literature on human settlements management, studies show that property management, Facility Management, housing management and urban management impact the sustainability of all these built environment milieus which form part of the human settlements’ sub-components. This research aimed at altering the afore-said challenge by exploring the management of human settlements in Nigeria and in South Africa. Philosophically, the research is pragmatic, and it rests on positivist deductive reasoning. The methods used for data collection include a review of literature, collection of archival records, and empirical studies of purposively selected professionals and stakeholders involved in human settlements’ management in Nigeria and in South Africa. The findings indicate that existing human settlements in the case studies are “good” and “acceptable” with the majority adopting an outsourced method of management. Furthermore, the lack of maintenance records, the deficiency in policy monitoring and the training of management and maintenance personnel were the three highest ranking factors indicated to be affecting human settlements management. Moreover, an inhabitant’s income, the time available for management, professional expertise and technology employed for management are recognised as the critical sustainability sub-factors affecting human settlement management. The factor analysis established ownership, human resource and capital, social, legal, technology, economic, physical and environmental factors as vital and very significant in achieving sustainable management of human settlements. Theoretical perspectives drawn from identity, hermeneutics and anthropology of extraordinary experience were used as analytical lens in the study. The study revealed that identity within the Catholic Charismatic Renewal was constructed principally in relation to the Holy Spirit. A spiritual experience or encounter was central in such identity construction, and the subjective and intersubjective identity of the members revolved around the two key concepts of ‘spirit-centred identity’ and ‘empowerment’. The study indicated that the members of the movement constructed their identity in ways that demonstrated their loyalty to the Church and their status as an officially recognised Catholic movement. In addition, their identity was also constructed in relation to the functions that they perceived themselves to perform within the Catholic Church. Although the members of the movement considered their charismatic and catholic identities to be compatible, they understood themselves to be different from other Catholics. The study also showed that while music was considered to be important within the movement, the context and content of its ritual music were overwhelmingly Pentecostal, and music that projected a catholic identity was relegated to the background and given expression mainly in the realm of individual religious practice. The study suggests that identity construction within the Catholic Charismatic Renewal is multiple, ongoing, flexible, and sometimes contradictory and paradoxical. Through its identity construction, the members of the movement demonstrate the heterogeneity of the Catholic space and seek to extend the boundaries of Catholicism.

- Description: Access to, control and ownership of land and water, amongst other natural resources in Zimbabwe, shape and affect rural lives, livelihoods, social relations and social organisation. Rural poverty has been entrenched and exacerbated by, amongst other factors, highly restricted access to these scarce resources. Historically, Zimbabwe’s rural areas (such as communal areas, smallholder irrigation schemes and resettlement areas) have existed as sites of struggles where contestations and negotiations over access to, control or ownership of these resources have taken place. Resultantly, multifaceted and dynamic social relations have been weaved and contested social spaces carved out. In rural Zimbabwe, contestations have tended to be complex, nuanced and intricate, working themselves out in different ways across time and space. In their heightened and more visible state, they have been characterised by violent physical expressions which, in the history of the country, involved two wars of liberation, the First Chimurenga (1896-1897) and the Second Chimurenga (1960s to 1980). The most recent violent manifestation was through nation-wide land invasions, politically christened the Third Chimurenga, which peaked in 2000 and continued sporadically to this day. Few studies on smallholder irrigation schemes in Zimbabwe have focused on understanding how contestations for access to scarce land and water resources are framed and negotiated at the local level. Cognisant of this lacuna, this thesis uses social constructionism in examining, as a case study, Mushandike Smallholder Irrigation Scheme in Masvingo Province in order to understand and analyse how land and water politics occur at the local level. The study deploys a qualitative research methodology approach in examining local water and land politics, which involved original irrigation beneficiaries and more recent land invaders. Findings of the thesis indicate that land and water shortages have increased considerably in the past two decades at the irrigation scheme due to the influx of land invaders into the scheme. This influx has had a negative impact on agricultural production and other livelihood strategies. Both scheme members and land invaders lay claim to land and water at Mushandike. These claims are intricately constructed and contested, and they are linked to broader issues such as partisan party-politics, policy developments, and tradition, origin, indigeneity and belonging. Though the struggles over land and water at Mushandike are firmly rooted in the concrete conditions of existence and experiences of beneficiaries and land invaders, external actors such as political leaders, state bureaucrats and traditional chiefs tend to complicate and intensify the contestations.

Due to the cosmopolitan nature of cyanobacteria, detection of BMAA in cyanobacteria has caused concerns about human exposure risk. This study was therefore based on the hypothesis that BMAA poses a health risk to humans either by direct ingestion or by indirect exposure to BMAA from a cyanobacterial source via a freshwater food chain. A validated gas chromatography-mass spectrometry (GC-MS) BMAA analysis method and a confirmatory liquid chromatography-mass spectrometry (LC-MS) method, with improved sensitivity, were developed in addition to a LC-MS/MS method for analyte confirmation. These methods were used to quantify BMAA in South African cyanobacteria, isolated from various potable water reservoirs. The majority of the isolates tested, contained BMAA. Possible human exposure by direct consumption of BMAA released from cyanobacterial blooms was investigated by the development of a robust solid phase extraction (SPE) method used for BMAA concentration and quantification in raw and treated tap water. Despite the use of the SPE method that facilitated the concentration of BMAA from large quantities of water, no free dissolved BMAA was detected in raw or processed fresh water. The fate of exogenous BMAA was therefore investigated firstly by evaluating the efficacy of standard water treatment processes employed in South Africa and secondly by investigating the possibility of BMAA bioaccumulation and biomagnification in aquatic food chains. Standard water treatment processes proved highly efficient at removing free dissolved BMAA, explaining the absence of BMAA in treated tap water. However, the cause of the BMAA absence in raw potable water remained unknown. Uptake of BMAA by model aquatic organisms was investigated in controlled experiments. BMAA uptake was documented in both Ceratophyllum demersum and Daphnia magna, however, BMAA-protein association and biomagnification were not observed in D. magna. BMAA had an inhibitory effect on the oxidative stress enzyme acitivties of both organisms tested (as well as human S9 extracts), resulting in accumulation of detrimental reactive oxygen species (ROS) in the cells. Exposure of crop plants to BMAA in controlled experiments resulted in BMAA uptake, protein association, and subsequent inhibition of the antioxidative enzyme activities. However, BMAA was detected in neither free nor protein-associated form in natural crop plants irrigated with known BMAA-containing bloom water. Post-mortem liver samples of Clarias gariepinus (Catfish) and Crocodylus niloticus (Crocodile), from a natural fresh water ecosystem that experienced frequent cyanobacterial blooms, contained both free and protein-associated BMAA. Higher BMAA concentrations were found in crocodile liver samples compared to fish liver samples, strongly suggesting biomagnification from one trophic level to the next. A Strategy for good IT Governance in South African Municipalities

The implementation of IT relies on the adoption of sound IT governance practices, whose outcome is to ensure that municipalities derive value from the use of IT and minimise the risk associated with it. However, the implementation of IT governance has proved to be challenging in most municipalities. This research sought to devise a strategic approach to the effective and viable implementation of IT governance in municipalities in South Africa. The research was carried out by adopting a design science research approach, which is increasingly being used for studies in the IT discipline as it assists in solving problems through the design of innovative artefacts, in this case a strategy for the implementation of IT governance. To address the objectives, the research was carried out through case studies of South African municipalities which enabled a detailed investigation of phenomena in its natural context. The data was collected by means of mixed methods consisting of a literature review, a survey and semi-structured interviews. A literature review connected to the research problem was done in order to identify the present state of the issues and how they have been confronted. A survey was sent out to municipalities, institutions and individuals with a mandate to support municipalities, while semi-structured interviews were conducted in five municipalities, government departments and regulatory bodies. A thematic analysis of the data collected was performed which led to a diagnosis of the issues. These include inadequate IT infrastructure; lack of coordinated external support; inadequate IT skills; lack of strong and committed IT leadership; poor IT controls; and fragmented planning. Guiding policies and a set of coherent actions for solving the issues were developed. This culminated in the devising of a strategy for the coordinated implementation of IT governance in municipalities. The primary contribution of this study is the formulation of a strategy for the sound implementation of IT governance in South African municipalities. The research contends that by identifying the strategic issues facing municipalities and implementing the proposed actions, most of which fall outside the municipalities, a new contribution to knowledge has been made. Accordingly, the strategy facilitates the development of plans that address the root causes of the poor implementation of IT in municipalities.

- Description: This study analysed the current inter-jurisdictional rules in the South African Value-Added Tax (VAT) system, identified shortcomings, and proposed legislative amendments or additions to address these shortcomings. The research was conducted within an interpretative post positivism paradigm, applied a qualitative research methodology, and a doctrinal research method. A detailed review of the literature was conducted to establish the theoretical basis of a good tax system and the theory underpinning indirect and consumption taxation. The literature review also included an in-depth analysis of the South African VAT system and its treatment of resident and non-resident businesses with a South African physical or economic reach, and its treatment of local and cross-border transactions, including imported services. The literature review also considered the international VAT treatment of these transactions. To obtain a wider range of expert opinions regarding shortcomings in inter-jurisdictional rules in the South African VAT system, data was collected through structured interviews with South African and global VAT and indirect tax experts, using a questionnaire that was specifically designed for this purpose. This study proposed amendments and additions to the VAT Act, dealing with the VAT registration of non-resident suppliers; addressing various issues relating to the interjurisdictional VAT rate; proposing measures in connection with imported services; and legislating the intention of the legislator to tax final utilisation or consumption. The study finally recommended the introduction of a general place of supply rule linked to residency; specific place of supply rules for electronic, broadcasting, and telecommunication services; and zero rating provisions for electronic, broadcasting, and telecommunication services provided to non-resident suppliers by resident suppliers for services initiated outside South Africa.

The study also noted that the observed heritage education practices are oriented more towards addressing issues related to marginalisation and alienation of indigenous cultures and practices, than enhancing learners’ agency to manage and utilise local heritage resources in a more sustainable ways. Based on these findings the study recommends re-positioning heritage education within the framework of Education for Sustainable Development (ESD). ESD acknowledges both issues of social justice and the dialectical interplay between nature and culture; as such, it may allow for representation and use of indigenous heritage constructs in ways that expand current political orientations to include sustainability as an additional objective of heritage education. Given that little research focusing on heritage education has been undertaken within southern Africa, the findings of this study provide a basis upon which future research may emerge.

It explores the link between language learning and teaching as well as the teaching strategies used within the classroom. In particular, the study seeks to analyse how isiXhosa is taught at three different levels of instruction, namely at Home Language (HL), First Additional Language (FAL) and Second Additional Language (SAL) levels. Qualitative methods were used, and the study took the form of interpretive case studies within the respective schools. The purpose of using multiple case studies was to investigate the reality within the three sites selected. The three schools had three extreme settings, in the sense that one of the schools is a rich isiXhosa environment and the other two schools are English environments. The tools used for data gathering were interviews, classroom observations, and an analysis of documents from the Department of Basic Education. Data was then presented and analysed in Chapter 4 and 5 against the backdrop of an extensive literature review in Chapter 2 as well as a detailed methodological approach as outlined in Chapter 3. One of the findings of this research indicates that in two of the schools the teaching culture is largely from a western perspective, whereas in one of the schools the learners are primarily isiXhosa speaking and teachers use a different linguistic approach to imparting knowledge. In the private and ex-Model C school it was found that a lack of exposure to isiXhosa is the primary cause of language problems for L2 learners. Secondly the L1 is not appropriately maintained or promoted in the school environment because it is presumed that learners are sufficiently exposed to their L1 at home. The research found therefore that in this particular schooling environment there is an inconsistency between the curricula that is taught in relation to the linguistic abilities of the learners, many of whom are mother tongue speakers of isiXhosa. Furthermore and more generally, it was found that teachers are still not well informed concerning South Africa’s Language-in-Education-Policy and there is a need for more inservice training that will focus on the nature of additional language acquisition in order to address the challenges of teaching these languages. The thesis concludes that extensive work needs to be done in order to reposition the teaching of isiXhosa at all three levels, but particularly at FAL level. This research shows that there is a disjuncture between the proposed curriculum/learning outcomes and the standards or levels achieved by the learners, more especially at FAL and SAL where oral proficiency in isiXhosa remains a challenge. Specific recommendations are contained in the final chapter of the thesis which also makes reference to the draft policy of the Ministry of Basic Education regarding the incremental introduction of the teaching of African languages from 2014 onwards. This thesis also makes takes as a point of departure the importance of multilingualism in a multicultural society such as South Africa where language is suggested as a strong factor in the fostering of social cohesion. It is for this reason that the thesis argues that the expert teaching of African languages, in this case isiXhosa, at both mother tongue and second language levels is of fundamental importance to the future of South African society.

Isolation and characterization of genes encoding heat shock protein 70s (hsp 70s) from two species of the coelacanth, Latimeria chalumnae and Latimeria menadoensis

- Description: The extant coelacanths have a close resemblance to the coelacanth fossil records dating back to 230mya. Like their predecessors, the extant coelacanths inhabit rocky caves at a depth of 100-300m below sea level. In the Comoros, the water temperature at these depths is estimated to fluctuate between 14-20°C. High-level adaptation to these environment and lack of competition are thought to have led to the morphological uniformity and slow change throughout the history of the coelacanths. Under stress conditions, proteins unfold or misfold leading to the formation of aggregates. Molecular chaperones facilitate the correct folding of other proteins so that they can attain a stable tertiary structure. In addition, molecular chaperones aid the refolding of denatured proteins and the degradation of terminally misfolded protein after cellular stress. Heat shock proteins form one of the major classes of molecular chaperones. Here we show that, despite high-level adaptation to a unique habitat and slow change, the genome of the coelacanth encodes the major and highly conserved molecular chaperone, Hsp70. Latimeria menadoensis and Latimeria chalumnae contain intronless hsp70 genes encoding Hsp70 proteins archetypal of known Hsp70s. Based on the coelacanth codon usage, we have shown that bacterial protein expression systems, particularly Escherichia coli, may not be appropriate for the overproduction of coelacanth Hsp70s and coelacanth proteins in general. Also interesting, was the discovery that like the rat Hsc70, the L. menadoensis Hsp70 could not reverse thermal sensitivity in a temperate sensitive E. coli DnaK mutant strain, BB2362. We also report the successful isolation of a 1.2 kb region of L. menadoensis hsp70 upstream regulatory region. This region contain three putative heat shock elements, a TATA- box and two CAAT-boxes. This regulatory region resembled the Xenopus, mouse, and particularly tilapia hsp70 promoters, all of which have been shown to drive the expression of reporter genes in a heat dependent manner. Taken together, this data is the first to strongly suggest an inducible Hsp70-base cytoprotection mechanism in the coelacanth. It further provides basis to formulate testable predictions about the regulation, structure and function of Hsp70s in the living fossil, Latimeria.
