Evaluation of the final effluents of some wastewater treatment plants in Amathole and Chris Hani District Municipality of the Eastern Cape Province as sources of vibrio pathogens in the aquatic environment

- Description: Certain areas in the world still depend on the receiving water bodies as sources of domestic water and for recreational purposes. The discharge of poor quality effluents from wastewater treatment plants can impact negatively on these water bodies, as they can act as vehicles for pathogens to the environment, posing a threat to humans if such water is used without precaution. Vibrio species are amongst those pathogens that can survive wastewater treatment processes, ending up in the environment, hence the aim of this study was to evaluate the final effluents of some wastewater treatment plants as sources of vibrio pathogens. Five wastewater treatment plants (WWTP) located in Amathole and Chris Hani District Municipalities in the Eastern Cape were used in this study. Samples were collected monthly from September 2012 – August 2013 and analysed using the standard membrane filtration technique. Yellow and green colonies on TCBS agar were enumerated as presumptive Vibrio species and expressed as CFU/100ml for each plant. Colonies were later picked based on their phenotypic characteristics, sub-cultured on fresh TCBS agar to ascertain purity. These presumptive isolates were then subjected to Gram staining and Oxidase test. Gram negative and Oxidase positive isolates were selected for further confirmation using Polymerised Chain Reaction (PCR). PCR was also employed for characterisation of Vibrio into three species viz V. parahaemolyticus, V. fluvialis and V. vulnificus. Antibiogram profile of the characterised species was then determined together with the presence of relevant antibiotic resistance genes Vibrio densities for the twelve month period ranged between 0 - 1.48×104 CFU/100ml with two of the plants located in East bank and Queenstown characterized by extremely high counts and one plant( Reeston) with very low counts.Three hundred presumptive Vibrio isolates were screened for identity confirmation. Of these, the dominating species found was V. fluvialis (28.6%) followed by V. vulnificus (28%) and the least was found to be V. parahaemolyticus (11.6%). The remaining unidentified 31.6% were suspected to belong to other Vibrio species not covered within the scope of this study. All the confirmed isolates i.e., V. parahaemolyticus, V. vulnificus and V. fluvialis were susceptible to imipenem, gentamicin and meropenem and resistant to only tetracycline. Between 60-100% of the V. parahaemolyticus isolates, 7.1% to 100 % V. vulnificus isolates and 2.5 to 100 % V. fluvialis showed resistances to polymixin B, sulfamethazole, erythromycin, penicillin G, chloramphenicol, trimethroprim and trimethroprim & sulfamethazole. Antibiotic Resistance Genes that were assessed included dfRA, SXT, floR and Sul2 varying in proportion with each species showing diversity in the Vibrio community. The dfR A gene was detected in all the V. parahaemolyticus isolates while floR gene was not detected in any of the isolates belonging to the three species. The distribution of sul2 gene cut across the species being 1% (1) in V. fluvialis, 3% (1) in V. parahaemolyticus and 4% (3) in V. vulnificus. The SXT gene was only determined in V. parahaemolyticus. It is clear that the final effluents of the selected plants are reservoirs for Vibrio pathogens as well as antibiotic resistance genes in the environment. The isolation of Vibrio from WWTP shows that this pathogen is in circulation in some pockets of the population. Therefore, wastewater treatment plants need to be properly monitored to ensure that they comply with set guidelines.

Electing high-order modes in solid state laser resonators

- Description: The first chapter considered the fundamental processes of laser operation: photon absorption, spontaneous and stimulated emissions. These processes are considered when designing a laser gain medium. A four-level laser scheme was also illustrated. Then, the basic components and operating principle of a simple laser system was presented using a diode end-pumped Nd:YAG solid state laser resonator. The second chapter considered laser light as light rays propagating in the resonator and extensively discussed the oscillating field in the laser resonators. It examined the characteristics of the fundamental Gaussian mode and the same theory was applied to higher-order modes. Chapter three started with an introduction to beam shaping and proceeded to present a review of some intra-cavity beam shaping techniques, the use of; graded phase mirrors, difractive elements { binary phase elements and spiral phase elements. Also, a brief discussion was given on the concept of conventional holography and digital holography. The phase-only spatial light modulator (SLM) was presented, which by default is used to perform (only) phase modulation of optical fields and how it can be use to perform amplitude modulation also. Finally, a detailed discussion of the digital laser which uses the intracavity SLM as a mode selection element was presented, since it was the technique used in the experiment. The elegance of dynamic on-demand mode selection that required only a change of the grey-scale hologram on the SLM was one quality that was exploited in using the digital laser. The next two chapters presented the experiments and results. The concept of the digital laser was first used in the experiment in chapter four, to assemble a stable diode endpumped Nd:YAG solid state laser resonator. Basically, the cavity was of hemispherical configuration using an intra-cavity SLM (virtual concave mirror) as a back re ector and a at mirror output coupler. A virtual concave mirror was achieve on the SLM by using phase modulation to generate the hologram of a lens, which when displayed on the SLM made it to mimic a concave mirror. Then the next phase was using symmetric Laguerre-Gaussian mode function, of zero azimuthal order to generate digital holograms that correspond to amplitude absorbing concentric rings. These holograms, combined with the hologram that iv mimics a concave mirror were used on the SLM to perform high-order Laguerre-Gaussian modes selection in the cavity. The fifth chapter presented the results of the mode selection and considered the purity of the beam at the output coupler by comparing measured modal properties with the theoretical prediction. The outcome confirmed that the modes were of high purity and quality which further implied that the cavity was indeed selecting single pure high-order modes. The results also demonstrated that forcing the cavity to oscillate at higher-order modes (p = 3) extracted 74% more power from the gain medium compared to the fundamental mode (p = 0), but this extra power is only accessible beyond a critical pump input power of 38.8 W. Laser brightness describes the potential of a laser beam to achieve high intensities while still maintaining a large Rayleigh range. It is a property that is dependent on beam power and its quality factor. To achieve high brightness one needs to generate a beam that extracts maximum power from the gain with good beam quality. Building on the experiments demonstrated in this study, one can make the correct choices of output coupler's re ectivity, the laser gain medium's length and doping concentration and the pump mode overlap for a particular mode to further enhance energy extraction from the cavity, and then using well known extra-cavity techniques to improve the output beams quality factor by transforming the high-order mode back to the fundamental mode. This will electively achieve higher laser brightness.

An assessment of financial accountability on social grants at the South African Social Security Agency (SASSA), 2011-2012

- Description: This research assesses the nature and extent of financial accountability on social grants at the South African Social Security Agency (SASSA). With respect to public funds, strategies and mechanisms, including the effectiveness of these strategies, were analysed to ensure whether SASSA was accountable to both the people and the government. Rabrenovic (2009:22) describes financial accountability as an obligation of the government to assure the citizens that money is spent in the best possible and most effective way. The government has to provide answers and justification for its action and regularly inform the public on how it spends the public funds. The extent to which the public-private partnership model adopted by SASSA for social grant management has enhanced financial accountability and yielded the desired outcome has also been tested. Some of the findings were that the problems facing SASSA were the financial constraints that entailed reduction in new programmes, halting infrastructures and the acquisition and filling only of funded posts from the Auditor General of South Africa (AGSA) and the high costs of administering social grants. This study used the qualitative research approach. The researcher used official document reviews because of the sensitivity of the research problem and opted for a desktop research. The documentary sources include SASSA’s annual reports for the period 2011/12. The recommendations were that people should be trained in financial and performance skills and in terms of accountability the accounting officers should account for lack of transgressions and poor performance.
