Augmentative releases of Dactylopius austrinus De Lotto (Dactylopiidae; Hemiptera) for biological control of Opuntia aurantiaca Lindley (Cactaceae), in South Africa

- Description: Opuntia aurantiaca Lindely (Cactaceae) is an invasive alien cactus which has detrimental effects on agroecosystems and indigenous biodiversity in South Africa. Dense infestations over large areas reduce grazing capacity and indigenous biodiversity. Despite the release of a biological control agent, the cochineal insect Dactylopius austrinus De Lotto (Dactylopiidae), the weed is still considered a major problem in many parts of the country. Biological control has relied heavily on classical biological control, with little augmentative biological control implemented. This study investigated the outcome of mass-rearing and augmentative releases of D. austrinus for the control of O. aurantiaca. Augmentative releases are thought to improve the level of control by increasing agent densities in the field and thus increasing the level of damage inflicted to the plants. All data were collected with the intention to optimize release strategies so that the maximum benefit from the biological control agent could be achieved. An impact study was conducted using potted plants in a greenhouse to quantify the efficacy of multiple releases of the agent on the target weed. All three of the release treatments showed consistently higher proportion of cochineal than the controls, as well as the insect exclusion treatments, and these differences were statistically significant. The number of cladodes per plant increased significantly for the insect exclusion and control treatment over the period of the study, whilst all three release treatments decreased steadily over the same period. This study indicated that the agent is damaging to O. aurantiaca and that a single release event was beneficial but that multiple releases did not result in greater levels of control. A post-release evaluation was carried out to quantify the impact of releases of D. austrinus on O. aurantiaca in the field. Plots where the agent was excluded were compared with those where the agent was left at natural field densities and three treatments where agent populations were augmented to varying degrees through releases. The percentage of cochineal infested cladodes for all treatments decreased over time from the initiation of the experiment in October 2017 until the end of the experiment in October 2018. Opuntia aurantiaca densities also decreased over time for all treatments. The insect exclusion treatment had the greatest number of plants for the duration of the study, but this was not significantly different from other treatments. Dactylopius austrinus was damaging to O. aurantiaca, but climatic conditions in the field limited the efficacy of releases. Although O. aurantiaca density decreased during the experiment, it was evident that the reduced number of plants was not due to augmentation of the cochineal populations from the releases that were conducted. The experiment was conducted over a very dry period, when cochineal was particularly effective, so although augmentative releases did not improve the level of control, the natural population of cochineal was high and very damaging to O. aurantiaca over the course of the experiment. Releasing during wet periods, when the agent is less effective, could augment agent populations at a time when natural populations would be low, and hence improve levels of control further. Although this study was limited to a short period of two years, the results of this study suggest that the number of releases is less important than the timing of releases. Releasing immediately after periods of high rainfall is likely to be beneficial, while releasing during dry periods, or during winter when temperatures are low, is less effective. Dactylopius austrinus populations should be constantly monitored so that releases can be conducted when cochineal populations are low and the climatic conditions are correct. If the timing of release events is appropriate, then the over level of control of O. aurantiaca using D. austrinus could be improved.

Causes of road projects’ delays: a case of Blantyre

- Description: The Construction Industry (CI) is recognised as a formidable sector that has the potential of facilitating the growth and development of a nation’s economy by providing the necessary infrastructure that is required to spur the growth of other industries. In many countries, such as Malawi, the city councils (CCs) also influence the implementation and management of such infrastructure as mandated by the Malawi Local Government Act (1998). Successful projects should be implemented in such a manner that completion of the projects is within the golden triangle of cost, time and quality. This implies that projects are completed within the planned budget, stipulated time and specified quality. City Councils in Malawi, however, face various challenges in their implementation of infrastructure projects leading to delayed project completion, resulting in extended project implementation periods. This study aimed at finding the causes of road projects construction delays in Blantyre City Council, (BCC) which is one of the four CCs in Malawi, the second largest and the commercial capital. The study followed a qualitative research method using a case study approach. This study combined in-depth, face-to-face interviews of identified stakeholders with general routine work observations and project document gathering to collect the primary data while secondary data was obtained from books, research journal articles, government documents, unpublished theses, some international organisation reports, general internet information and other academic related sources of relevant information. The participants included council representatives (council service committee chairperson, secretariat management staff), contractors’ representatives and consultants’ representatives. The participants were chosen using a non-probability sampling method of purposive and snowball selection, targeting their involvement in the previous and/or the current project implementation cycles. The study also involved visiting some of the ongoing and completed projects and collecting project photographs. The findings from the case study revealed challenges the council faced due to a shortage of engineers in conducting detailed proposed projects surveys resulting in incomplete project scope definition (detailed designs) on some projects, before the procurement of contractors as the major cause of project construction delays. Other factors that were identified were: service providers delaying the removal of existing public utility infrastructure from project sites, inclement weather and the client delays in issuing instructions to the contractors during the implementation of the projects. Another factor was the shortage of construction equipment and construction materials by some contractors. Based on the research findings, the main recommendation is to increase the number of technical employees, especially engineers and other key technical staff such as quantity surveyors by improving the employees’ conditions of service. Improving the service conditions will attract and retain the right quality and quantity of technical employees to undertake the management of the construction projects. Alternatively, all positions for professionals who are deemed as scarce or hard to maintain could be placed on a contract basis, renewable after a period of two to three years. This would be able to attract those professionals who are ready to offer their expertise for a certain period but may not necessarily want to work until the government mandatory retirement. This will be in addition to the engagement of external project consultants to assist in project management, which the council started implementing in 2017. As for the delay in the removal of utility services infrastructure on proposed project sites, the client needs to commence with the process early in the planning and procurement process before handing over the project sites to the contractors. General contractor delay causes can be minimised by raising the selection criteria of contractors to flush out as well as black-listing incompetent contractors.

- Description: Previous studies conducted on wool production in the communal areas of the Eastern Cape have concluded future research should focus on whether the observed differences in average wool price received between shearing sheds are due to differences in clip preparation (i.e. classing) or differences in the quality of the wool being produced (i.e. genetics).The objective of this study was to establish whether there are indeed geographical differences in the average wool price received by communal wool producers in the Eastern Cape and, if so, whether this difference in earnings is attributable to differences in the quality of wool produced and/or clip preparation. The current research explored eight years of wool production records of communal wool farmers (2009/10 to 2016/17) obtained from BKB, the largest wool broker in South Africa. Statistical analyses included descriptive statistics, analysis of variance, parametric tests (i.e. t-test) and multiple regression analysis. The conclusions drawn from this study are: that there are indeed statistically significant differences in the average wool price received between the different geographic areas of the Eastern Cape communal regions, and that the primary determining factor is wool quality, while clip preparation also plays an important but secondary role. The study identified specific geographic areas where developmental interventions are most critical, with specific emphasis on improving the genetic composition of the wool sheep flocks. Specific strategies are recommended, namely:Removing inferior-quality rams and replacing them with superior-quality rams to prevent any inferior nondescript rams from breeding and producing progeny; Replacing the inferior-quality nondescript ewes in specific areas with superior-quality ewes, and Improving the survival rate of lambs and thereby contributing towards expediting the rate of genetic improvement of the communal wool sheep flocks.

The hepatoprotective capacity of selected natural products from South Africa

- Description: Drug-induced liver injury (DILI) is recognized as a significant clinical problem, which may account for up to 50% of all cases of acute liver failure. DILI is initiated by the bioactivation of parent drug molecules to produce chemically reactive metabolites. These reactive intermediates induce mitochondrial dysfunction and oxidative stress leading to glutathione (GSH) depletion and damage to cellular proteins, lipids and nucleic acids, which eventually culminates in necrotic cell death. The principal objective of this study is to establish an in vitro screening platform to identify potential hepatoprotective natural products (plants and mushrooms) from South Africa (SA). Aqueous plant extracts (Cyclopia intermedia, Opuntia ficus indica and Kigelia africana), and aqueous and ethanolic macrofungal extracts (Ganoderma lucidum, Russula capensis, Pleurotus ostreatus and Lenzites elegans) were prepared and screened against HepG2 and VERO cells to assess their safety using Hoechst 33342-PI dual labelling. A drug-induced hepatotoxic model was established, using the dietary supplement menadione (vitamin K3). Hoechst 33342- PI, -CellROX® Orange and -TMRE dual labelling was used for necrosis, oxidative stress and mitochondrial membrane potential depolarization (ΔΨm) detection, respectively. The accuracy of the hepatoprotection model was confirmed through HepG2 cellbased assays (Hoechst 33342- PI, -CellROX® Orange and -TMRE dual labelling) that measured the protective effects of natural products against the menadione-induced toxicity, anti-oxidant assays (DPPH, NO, ORAC, CAPe and FRAP) that measured their anti-oxidant potential and enzyme assays (βglucuronidase, carboxylesterase and CYP450 isoform 3A4) that measured their effects on drug metabolism. Silymarin was used as a positive control for each assay. Menadione displayed significant cell death, increased oxidative stress and decreased ΔΨm at an elevated concentration of 100 μM; confirming the hepatotoxicity model, where necroptosis was suspected to be menadione’s cell death mode. Only ethanolic G. lucidum was cytotoxic. All three aqueous plant extracts demonstrated strong anti-oxidant capacities out of all the tested extracts; where C. intermedia displayed the most promising DPPH, NO, ORAC, CAPe and FRAP activity, followed by aqueous G. lucidum. Aqueous plant and ethanolic macrofungal extracts (C. intermedia, O. ficus indica, K. africana, and ethanolic P. ostreatus, R. capensis) displayed decreased menadione-induced ROS production and protected against menadione-induced ΔΨm depolarization, posing them and aqueous G. lucidum potential therapeutic interventions for DILI. Ethanolic L. elegans demonstrated the highest enzyme inhibition for each assay and presented genotoxicity, ruling it out as a therapeutic strategy against DILI. Together these assays addressed several aspects relating to DILI and hepatoprotection, and served as a good starting point in evaluating the therapeutic value of natural products from South Africa.

Identifying methane emissions with isotopic and hydrochemical clues to their origin across selected areas of the Karoo Basin, South Africa

- Description: Currently, only sparse data exists on the methane emissions from the Main Karoo Basin, South Africa, where the potential discovery of vast quantities of unconventional natural gas from the black shales of the Whitehill Formation has sparked great interest in the prospect of hydraulic fracturing. In this study, a new infield methane quantification instrument (Picarro G2201-i) is used to identify freely emitted methane and more importantly collect stable carbon isotope ratios (δ13C-(CH4)g) that is used in obtaining information regarding the genetic origin and thermal maturity of the methane. Hydrochemical information (TOC, δ13C-TOC, 3H, δ18O, δ2H, and anions) were also scrutinised to assist in the determination of the origin of methane across the Karoo Basin. Twenty-one sites were investigated across four provinces in the Main Karoo Basin. Six of these sites are thermal springs, three are shallow boreholes (<100 m in depth), and twelve were Soekor (The Southern Oil Exploration Corporation)/deep boreholes (>1500 m). Of these 21 sites, 17 were suitable for either methane analysis or groundwater sampling, with four of the Soekor boreholes unsuitable. The presence of methane in groundwater and being freely emitted seems to be a common occurrence above the Main Karoo basin and of the 17 sites investigated, 14 had freely emitted methane emission. All but one of these sites had δ13C-(CH4)g signatures greater than -50 ‰, indicating a thermogenic origin. Combining the results obtained from the Picarro instrument with those compiled by Talma & Esterhuyse (2015), a higher resolution distribution map was created. The δ13C-CH4 signatures show patterning with a decreasing trend from the southern Karoo Basin to the north, which corresponds to the general decrease in thermal maturity of the Ecca shales (Whitehill Formation) northward across the Karoo Basin. The δ13C-(CH4)d results from a case study conducted by Eymold et al. (2018) differ significantly with the data collected in this study that included several of the same sampling locations. This is explained by a two phase partitioning (gas + water) that leads to the thermogenic endmember of methane being released in its free state (analysed by Picarro G2201-i) and microbial methane that is formed in situ remains dissolved in the water (analysed by Eymold et al. 2018). Soekor and deep sites; SA 1/66, KA 1/66, and KWV-1 that have direct pathways for methane migration from the Whitehill are deemed the best proxies to resolve the thermogenic endmember of methane, with δ13C-(CH4)g signatures of -26.32‰, 31.66‰, and -34.57‰, respectively. The hydrochemistry results suggests that that free methane emissions do not necessarily have to be associated with saline Cl- waters, as multiple sites have CH4 emissions with low salinities (Cl < 50 mg/L) and that methane in its free state can migrate to the surface due to buoyancy. The results also indicate that dolerite intrusions act as conduits for upward migration of groundwater from depth, but that the deep groundwater signatures proposed by Murray et al., (2015) are related to their different migration pathways and water-rock interactions rather than being representative of the deep formation waters. Using an initial assessment δ13C-CH4, TOC concentration [TOC] and the tritium (3H) values, where water samples that have 3H ≤ 1 TU, detectable TOC and δ13C-CH4 signatures > -50‰ could indicate hydraulic connectivity between the shallow aquifer and an organic/CH4 rich sedimentary layer, which may or may not be from the Whitehill Formation. However, this method for determining aquifer connectivity requires further investigations in the Karoo Basin context. The results obtained in this study add to the limited isotopic data of methane across the Karoo Basin and demonstrates the effectiveness of an infield identification of methane emissions using the Picarro G2201-i.

Pharmacological Evaluation and Medicinal Potential of Vachellia Karroo (Hayne) Banfi & Galasso Pods traditionally used to treat Sexually Transmitted Diseases in the Eastern Cape, South Africa.

- Description: Vachellia karroo is widely used in folk medicine in the Eastern Cape Province South Africa, however, the pods are usually discarded as waste. The current study was conducted to evaluate the phytochemical content and the antioxidant activity of V. karroo pods. The pods were extracted using acetone, distilled water, hexane and methanol. The total phenol, flavonoid, proanthocyanidin, alkaloid and saponin contents of the various extracts were determined spectrometrically and antioxidant activity was evaluated using inhibition of 2,2-diphenyl-1- picrylhydrazyl (DPPH), 2,2´-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid (ABTS), nitric oxide (NO) radicals, while total antioxidant capacity (TAC) was estimated by the phosphomolybdenum assay. V. karro pods had significant amounts of total phenols, flavonoids and proanthocynadin, although, proanthocynadin was not detected in the aqueous and hexane extracts. Methanol and acetone extracts showed higher phenolic, proanthocynadin and flavonoids contents (52.47 ± 6.82 and 29.31 ± 1.49), (334.8±85.1 and 231.22 ±1.80),(288±4.26 & 208.2±17.7) compared to the aqueous and hexane extracts. The alkaloid and saponin contents were 26.67 and 12.85 % respectively. The IC50 values of the methanol extract for DPPH, ABTS and total antioxidant capacity (TAC) were 0.345, 0.017 and 0.116 mg/mL, respectively. Methanol extract of V. karroo pod showed higher polyphenolic content of all the extracts analysed with corresponding strong free radical scavenging potential. These underutilized pods could serve as a new source of antioxidant compounds which could help in combating various ailments. The great antioxidant activity displayed by V. karroo pods extract supports the therapeutic use of this plant in traditional medicine and are attributed to the phytochemical content.The current information suggests that extracts from Vachellia karroo pods might be a ix cheap potential source of natural antioxidants that could be of great importance for the treatment of free radical related diseases. Furthermore, it makes a case for the utilization of the pods instead of discarding them as waste materials. The evaluation of antimicrobial potential of Vachellia karroo extracts was carried out using agar dilution assay against 8 bacterial strains-4 gram-positive [ Enterococcus faecalis (ATCC 29212), Staphylococcus aureus (OK), Bacillus subtilis KZN, Bacillus cereus, and Streptococcus pyogenes] and 4 gram-negative strains[ Vibrio cholera, Klebsiella pneumonia (ATCC 4352), Pseudomonas aeruginosa (ATCC 19582), Salmonella typhi (OK) ]. Six fungal isolates[ Trichophyton mucoides ATCC 201382, Candida albicans (ATCC 10231), Candida glabatra, Penicillium chrysogenum, Aspergillus fumigatus, Penicillum aurantiogriseum] were usedor antifungal assessment. The methanol extract exhibited broad-spectrum activity, with Gram-positive strains being more sensitive than Gram-negative strains. The extracts also showed great inhibition against Candida albicans the fungal isolate responsible for causing candidiasis in women. Since the pods showed promising antimicrobial activity, they could serve as a cheap source for the treatment and management of these sexually transmitted infections. The brine shrimp toxicity test revealed successful hatching of the cysts was in the order: Aqueous extract> methanol extract> hexane extract> acetone extract. The hatching of nauplii was in a concentration dependent fashion, with hatching success decreasing with increase in concentration of extracts. Lethality of extracts determined based on Meyerʼs index of toxicity, showed that the acetone and hexane extracts of V. karroo were moderately toxic. The results of this study indicated that aqueous and methanolic extracts of Vachellia karroo pods were not toxic, therefore supporting its traditional therapeutic usage. This implies that rather than discard x the pods as is waste as it being currently done, Vachellia karroo pods could be salvaged and processed along with the leaves, thus reducing environmental pollution. Proximate parameters (moisture, ash, crude fibre, crude fat, proteins, and carbohydrate) were evaluated using ALASA methods, and elemental analysis by ICP-OES technique. Nutritional analysis showed that V. karroo pods had low content of crude fat and high content of crude fibre, ash, crude protein, and carbohydrate sufficient to meet the recommended dietary allowances. The pods were rich in major minerals Ca, K, P and Mg, with sufficient amount of trace elements Na, Fe, Zn, and Cu. The outcome of this study suggests that Vachellia karroo pods have good nutritional potentials to support or complement the recommended dietary allowance and could be a cheap source of essential nutrients that could help in ameliorating most nutritional challenges as well as contribute remarkably to the amount of nutrient intake in human and animal diets.

Zoning the Southwestern Indian Ocean to mitigate impacts from ocean-based hydrocarbon exploration and production on sea turtles

- Description: The conflict between sea turtles and the numerous socio-economic developments in the Southwestern Indian Ocean (SWIO) are set to intensify as countries are looking to develop their ocean-based economies. The Hydrocarbon Exploration and Production (HEP) industry is of particular importance, since many of the SWIO governments view it as catalyst for development. This has raised concerns about potentially significant environmental impacts from the HEP industry, to sea turtles and their habitats, based on international examples where sea turtles have been severely negatively impacted upon, like the Deepwater Horizon spill in the Gulf of Mexico. Given that the four sea turtles species in the SWIO are listed on the IUCN Red List of threatened species, the aim of this study was to derive priority areas for sea turtles in the face of HEP, that could be used in an ocean zoning strategy for sustainable economic development of HEP in the SWIO region. To achieve this, the study spatially represented the main life-history stages of sea turtles, i.e. the breeding, migrating and foraging areas of Caretta caretta (loggerhead turtles), Dermochelys coriacea (leatherback turtles), Chelonia mydas (green turtles) and Eretmochelys imbricata (hawksbill turtles), within a telemetry derived distribution for each species. This spatial representation was used to quantify the extent of Marine Protected Areas (MPAs) conserving sea turtles in the SWIO, which revealed that sea turtle breeding areas were well represented in MPAs, including C. caretta (~40 %), C. mydas (~53 %), E. imbricata (~59 %) and D. coriacea (~22 %), the latter being least protected by MPAs during breeding, possibly due to a far greater extent of their internesting areas than the other three species. MPA coverage of breeding areas could be positively correlated to the increasing population trends of C. caretta and C. mydas in the SWIO, and therefore the assumption was made that increasing population trends of sea turtles are in part related to MPA protection of their breeding areas. In addition, the potential impacts on sea turtles from existing and proposed HEP developments were assessed and mapped by using a novel, species-specific rating index. The results revealed the extensive nature of potential water pollution impacts on sea turtles, constituting 16 of the top 28 most significant impacts from HEP on sea turtles. Other significant impacts on sea turtles associated with the HEP industry, included habitat destruction, light pollution and noise pollution. Importantly, this study found that ~70 % of all potential HEP impacts (irrespective of significance) on adult nesting sea turtles could be avoided if seasonal sea turtle movement during critical life stages are included as species-specific HEP mitigation measures. The data and maps on the main life-history stages of sea turtles, and the potential cumulative impacts from the HEP industry, were used in a Systematic Conservation Planning process, to derive a concept ocean zoning. As final outcome of this study, the concept ocean zoning highlighted areas where increased protection to sea turtles, and management of the conflict between sea turtles and the HEP industry, will be required if the HEP industry is to develop in a sustainable manner in the SWIO.

Effect of single-walled carbon nanotubes on various properties of reclaimed rubber/natural rubber blends

- Description: Mechanical and thermal processes are the preferred methods used to reclaim rubber from waste tyres. However, these methods produce reclaimed rubber (RR) with poor quality and lowered properties compared to virgin rubber. As a result, RR is commonly blended with virgin natural rubber (NR) in order to enhance its quality and properties to widen its application scope. The use of RR in the production of new rubber products is however limited to only about 1-10%. This is because the amounts of RR above this percentage negatively affect the properties of the resultant RR/NR blends. Researchers have used various conventional and nonconventional fillers such as carbon black and maize stalk fibres, respectively, in an attempt to enhance the properties of RR/NR blends. One main disadvantage with the use of these fillers is the need for high quantities to achieve desired properties. Several researchers have therefore focused on developing other reinforcing fillers. The fillers that have recently attracted a lot of attention of the present researchers include single-walled carbon nanotubes (SWCNTs), which are very small tubes solely made up of carbon molecules. This is because SWCNTs offer many impressive properties, and even small quantity of these nanofillers is sufficient to achieve desired properties due to their small size, large surface area and high aspect ratio. The main challenge with the use of SWCNTs however is getting them to disperse well in the rubber matrix. Therefore, their surface is either modified and then mixed with rubber or a dispersant is used to disperse them in the rubber matrix, forming a masterbatch. The common nanomaterial masterbatch product for applications in rubber is the TUBALLTM RUBBER (TR), which is a masterbatch product consisting of 1 wt. % TUBALL™ SWCNTs dispersed, using 1.5 wt.% sulfonol as a dispersant, in 97.5 wt.% NR (from latex Taytex FA). This product is a versatile masterbatch designed to enhance the mechanical, chemical, electrical and thermal properties of rubbers since it contains SWCNTs. However, the performance of this product has not been explored in the cases where recycled/reclaimed rubber is used. The main aim of this project was therefore to explore the effect of SWCNTs (via using TR) in the formulations of RR/virgin NR blends. This project was divided into two parts; (i) optimisation of SWCNTs based on tensile properties of specific formulations of compounds. These compounds were prepared by mixing various amounts of TR and NR using a Banbury internal mixer and two-roll mill. (ii) the effect of the addition of SWCNTs from the optimum SWCNTs/NR masterbatch compound on curing, mechanical, aging, dynamic mechanical and thermal properties of RR/NR blends was studied. Curing properties explored include minimum torque (ML), maximum torque (MH), scorch time (ts0.5), curing time (t90) and cure rate. Mechanical properties studied are tensile strength, modulus at 100%, 200%, 300% and 500% elongation, elongation at break, hardness and resilience. Dynamic mechanical properties include stiffness, storage modulus (E'), loss modulus (E''), loss tangent delta (tanδ) and glass transition temperature (Tg). Thermal degradation (stability) was studied by means of thermogravimetric analysis (TGA) for analysing the effect of SWCNTs inclusion on RR/NR blends. The optimum SWCNTs/NR masterbatch compound contained 0.1 wt. % of SWCNTs. The results obtained for various properties indicate that SWCNTs from the optimum SWCNTs/NR masterbatch compound enhanced the curing, mechanical, aging and dynamic mechanical properties of RR/NR blends with lower RR loading. Thermal properties of RR/NR blends were found to be unaffected by the addition of SWCNTs. However, owing to their impressive properties, the use of SWCNTs as nanofillers in RR/NR blends have the greatest potential for extending the applications of RR/NR blends.

- Description: This study investigates the effect of two fresh seaweeds, a formulated diet and different feeding regimes on the growth (weight, diameter and height) and gonad quality (gonadosomatic index, colour, texture, firmness and gonad maturity) of the collector sea urchin, Tripneustes gratilla under farm conditions for eighteen weeks. The gonad enhancement study was divided into two sections. The first section investigated the effect of feeds on the somatic growth and gonad quality for 12 weeks. The feeds investigated during this phase of the study were fresh Ulva (U); a 50:50 mixture of fresh Ulva and Gracilaria (UG); fresh Gracilaria (G) and a formulated diet, which contained 200g of dried Ulva per kg (designated as 20U). The second section or phase of this study started for a period of 6 weeks and investigated the effect of a change of diet on urchins previously fed the various feeds from section 1. The feeding regimes used were (U-20U) i.e urchins previously fed with Ulva were subsequently fed 20U diet, similarly, (UG-20U); (G-20U) and (20U- 20U). For the first section of the experiment, no differences were observed in the growth (weight, diameter and height) of urchins fed both fresh diets and the formulated diet. However, urchins fed the formulated diet produced gonad weight of 50.72 ± 5.4g which was significantly higher (P<0.001) than those fed fresh diets. The gonads of urchins fed the Gracilaria diet were significantly darker (P= 0.023) in colour compared with those examined from urchins fed the formulated diet, but were not significantly different from any of the other fresh diets tested (Ulva and Ulva mixed with Gracilaria). Gonad from each of the feed treatments were similar in terms of the redness (a*) and yellowness (b*) as well as the texture and firmness. The gonad maturities of urchins fed the various diets were not significantly different from each other. By the end of the second section/phase of the study (6 week period following the diet change), somatic growth (weight, height and diameter) of urchins in all treatment groups did not differ significantly from each other (ANOVA, P= 0.784; P= 0.988; P= 0.28 respectively). Gonad weight of urchins in treatment groups UG-20U and G-20U became similar to those fed the 20U diet at the end of the trial (week18). However, the gonad weight of urchins in the 20U treatment group remained significantly greater than urchins in the U-20U group at the end of the trial (Kruskal-Wallis ANOVA, P= 0.002). Gonad colour of urchins in the G-20U treatment became significantly lighter (ANOVA, P= 0.029) than those in the U-20U group after the change of diet. However, gonad lightness (L*) of urchins in treatment groups U-20U and G–20U did not differ significantly from those in the UG-20U and 20U treatment groups. All feed regimes were similar in terms of the gonad‟s redness (a*) and yellowness (b*), texture and firmness as well as gonad maturity at the end of the trial. This gonad enhancement study on wild collected adult T. gratilla has shown that somatic growth (urchin weight, test diameter and height) does not differ between the dietary treatments (fresh seaweeds and a formulated diet) tested in this study. The formulated feed (20U diet) does however enhance gonad growth of T. gratilla under farm conditions, supporting previous laboratory trials conducted on this species fed similar diets, indicating that sea urchins gonad enhancement can be cultured under farm conditions in South Africa.

- Description: This study presents some of the findings of a long-term comparative study at the Nelson Mandela University George campus which started in 2014. This research compares the differences in crop yields and soil fertility under conventional and organic farming systems. Although previous research that was conducted will be presented briefly, the main focus of this dissertation is to report on the research results generated during the 2016/17 and 2017/18 cropping seasons. The baseline study (indicator measurements) was done in 2014 and investigated soil fertility before the different farming systems were implemented. The rapid increase in human population threatens food security, especially in developing countries. Climate change together with the degradation of soil and other natural resources due to high-input industrial farming, provide further challenges in terms of agriculture and future food security. Agricultural production in South Africa faces three obstacles: water scarcity, infertile soils and poor institutional support. Therefore, regenerative and sustainable agriculture practices such as organic farming, are increasingly being promoted as an alternative to conventional farming systems in southern Africa. What is the best way of farming? This is the question every farmer is confronted with. Some say organic whilst others say conventional, but how do we determine this? What is the role of crop rotation in pest management, soil fertility and crop yield? So we set up the research to compare the two farming systems so that decisions can be based on sound scientific findings. Results showed a significant increase in the case of the organic yield in season 2016/17 for cabbage and in both seasons (2016/17 & 2017/18) for cowpea and sweet potato, proving that organic farming yields can increase over time. Soil fertility changes from the baseline year (2014) till the end of season 2017/18 were compared. Overall organic farming systems showed very promising results with higher levels of soil pH followed by a significant increase in important soil nutrients (P, K & C) compared to conventional systems. Crop rotation also had positive effects.

- Description: The applications of machine learning have created an opportunity to deal with complex problems currently encountered in radio astronomy data processing. Calibration is one of the most important data processing steps required to produce high dynamic range images. This process involves the determination of calibration parameters, both instrumental and astronomical, to correct the collected data. Typically, astronomers use a package such as Common Astronomy Software Applications (CASA) to compute the gain solutions based on regular observations of a known calibrator source. In this work we present applications of machine learning to first generation calibration (1GC), using the KAT-7 telescope environmental and pointing sensor data recorded during observations. Applying machine learning to 1GC, as opposed to calculating the gain solutions in CASA, has shown evidence of reducing computation, as well as accurately predict the 1GC gain solutions representing the behaviour of the antenna during an observation. These methods are computationally less expensive, however they have not fully learned to generalise in predicting accurate 1GC solutions by looking at environmental and pointing sensors. We call this multi-output regression model ZCal, which is based on random forest, decision trees, extremely randomized trees and K-nearest neighbor algorithms. The prediction error obtained during the testing of our model on testing data is ≈ 0.01 < rmse < 0.09 for gain amplitude per antenna, and 0.2 rad < rmse <0.5 rad for gain phase. This shows that the instrumental parameters used to train our model more strongly correlate with gain amplitude effects than phase.

Nutritional and pharmacological profile of moringa oleifera lam. Leaves cultivated in the South African ecotype

- Description: Moringa oleifera Lam. is native to the Indian ecotype, which has become naturalized in many tropic and subtropic regions worldwide, thus a great variation in the bioactive compounds of the plant is recorded. The introduction of Moringa oleifera to South Africa occurred in the turn of the twentieth century with the aim of combating malnourishment, aiding health and fostering economic development. Therefore, this study was conducted with the aim of evaluating and authenticating the nutritional parameters, essential oil and bioactive components found in the various solvent extracts of Moringa oleifera leaves cultivated in the South African ecotype. It also elucidated some of the plants medicinal properties by investigating the antioxidant and antibacterial properties of the plant. The essential oil of the leaves was extracted using two methods (solvent free microwave extraction and hydrodistillation) and analysed using Gas Chromotography/Mass Spectrometer. The safety profile of the leaves was investigated using brine shrimp model. The results from the study revealed that Moringa oleifera leaves cultivated in South Africa were rich in crude protein levels (28.72%) and carbohydrate (28.84%). Among the minerals tested for, the leaves contained high content of calcium (1603.33 mg/100 g), potassium (1690 mg/100 g), zinc (13.03 mg/100 g) and iron (21.13 mg/100 g). The vitamin E content had the highest concentration (89.43 mg/100 g) among the vitamins evaluated. The leaves of Moringa oleifera had low quantities of antinutrients in relation to minerals, thus ensuring nutrients availability. This study identified 15 fatty acids in the dried leaves of Moringa. The highest fatty acid detected were polyunsaturated fatty acids, linoleic acid (56.36) and alpha-linolenic acid (687.58), 7 of the fatty acids were saturated fatty acids, though relatively low besides stearic acid. 15 amino acids were present, 7 of which were essential amino acids, namely; threonine, tyrosine, methionine, valine, phenylalanine, isoleucine and leucine. vi Among the phytochemicals evaluated, the aqueous extract gave the highest total phenolic content (21.01 ± 2.315 mg GAE/g), the acetone extract gave the highest proanthocyanidin (15.33 3.5 mg CE/g) and flavonoid contents (25.04±3.28 mg QE/g), while saponin and alkaloid contents were 27.73 ± 9.97% and 7.32 ± 2.73 % respectively. All the extracts exhibited high radical scavenging power against the 2,2- diphenyl-1-picrylhydrazyl radical with an IC50 less than 0.025 mg/mL. Aqueous extract gave the highest capacity based on ABTS and TAC assays with IC50 of 0.074 mg/ mL and 0.055 mg/mL respectively. Ethanol extract exhibited the least antioxidant capacity among the extracts, as well as the least antimicrobial activity on both Gram −ve and Gram +ve bacteria with all its minimum inhibitory concentration values greater than 5 mg/mL. The acetone extract exhibited the strongest antimicrobial activity among the extracts. A total of 8 and 24 chemical compounds were found from the solvent-free microwave extracted and hydrodistilled oils, respectively. The major components found from hydrodistillation were 2-Hexanal (16.37%), Nonanal (6.09%), 5 9-undecadien-2-one 6, 10 dimethyl- (e) - (8.01%), trans-beta-Ionone (8.02%) and octadecane (9.67%). For the solvent free microwave extracted oil, Benzene acetaldehyde (5.68%), Octadecane, Pentacosane and Eicosane which were 11.03%, were the major components. Toxicity evaluation revealed that mortality was time dependent. As the exposure period increased, mortality kept increasing. The hatching success of the cysts in acetone, aqueous and ethanol extracts were 35.7, 33.6 and 35.2% respectively. Acetone extract showed the most prominent hatching success. Hatchability and lethality were in a concentration dependent fashion. Among the extracts, acetone extract exhibited the highest lethality (12.3%) to the nauplii, followed by ethanol (10.9%), then aqueous (5.6%) extracts. All the results obtained from this study suggest that Moringa has the potential of eradicating malnutrition and aid health. It contains the necessary nutritional and bioactive compounds and the leaves proved not to be toxic at the levels (0.0625 – 1 mg/mL) tested.

- Description: Large industrial and residential developments near towns and along highways associated with public policies have transformed the pattern of development over the recent years, creating a new urbanisation phenomenon; urban sprawl. Indiscriminate population densities, discontinuous and fragmented settlements, largely define urban sprawl. The progression of urban sprawl can be described by transformation in pattern over time, like proportional increase in built-up surface to population leading to rapid urban spatial expansion. Stemming an understanding from the processes, causes and patterns of urban sprawl, the consequences of sprawl on land and vegetation can be analysed. Environmental impacts to both the rural and urban population emanate from such instances, creating an excessive liability to the government. With attention to this and recognising the lack of discussion on the matter, the research deliberates some of the impacts observed in Buffalo City Metropolitan Municipality, Eastern Cape, South Africa. The study makes use of Geographic Information Systems and Remote Sensing with the assistance of landscape metrics. The influence of urban sprawl in this municipality has revealed impacts on vegetation, green areas and land in general. The results disclose that urban sprawl is a multidimensional phenomenon that is better explained using various methods (indices). Buffalo City Metropolitan municipality is located in Eastern Cape amidst the thicket ecosystem, the municipality has grown and expanded over the recent past. The study spread over an 18-year period from 1994- 2012. Based on field surveys and SPOT imagery, built-up areas of BCMM was extracted for different periods. Data used for the study are census data for BCMM, 1994, 2000, 2006 and 2012 SPOT images, images of BCMM acquired from Google earth 2018. The rate of transformation of the area was calculated and it was higher compared to that of population growth. Based on this data urban growth are analysed with the assistance of landscape metrics that include Shannon entropy. The outcomes confirm that this metropolitan municipality has experienced sprawl and sprawl has done so at cumulative rate.

A crowdsensing method for water resource monitoring in smart communities

- Description: Crowdsensing aims to empower a large group of individuals to collect large amounts of data using their mobile devices, with the goal of sharing the collected data. Existing crowdsensing studies do not consider all the activities and methods of the crowdsensing process and the key success factors related to the process. Nor do they investigate the profile and behaviour of potential participants. The aim of this study was to design a crowdsensing method for water resource monitoring in smart communities. This study opted for an exploratory study using the Engaged Scholarship approach, which allows the study of complex real-world problems based on the different perspectives of key stakeholders. The proposed Crowdsensing Method considers the social, technical and programme design components. The study proposes a programme design for the Crowdsensing Methodwhich is crowdsensing ReferenceFrameworkthat includes Crowdsensing Processwith key success factors and guidelines that should be considered in each phase of the process. The method also uses the Theory of Planned Behaviour (TPB) to investigate citizens’intention to participate in crowdsensing for water resource monitoring and explores their attitudes, norms and perceived behavioural control on these intentions. Understanding the profiles of potential participants can assist with designing crowdsensing systems with appropriate incentive mechanisms to achieve adequate user participation and good service quality. A survey was conducted to validate the theoretical TB model in a real-world context. Regression and correlation analyses demonstrated that the attitudes, norms and perceived behavioural control can be used to predict participants’ intention to participate in crowdsensing for water resource monitoring. The survey results assisted with the development of an Incentive Mechanism as part of the Crowdsensing Method. This mechanism incorporates recruitment and incentive policies, as well as guidelines derived from the literature review and extant system analysis. The policies, called the OverSensepolicies, provide guidance for recruitment and rewarding of participants using the popular Stackelberg technique. The policies were evaluated using simulation experiments with a data set provided by the case study, the Nelson Mandela Bay Municipality. The results of the simulation experiments illustrated that the OverSenserecruitmentpolicycan reduce the computing resources required for the recruitment of participants and that the recruitment policy performs better than random or naïve recruitment policies. The proposed Crowdsensing Method was evaluated using an ecosystem of success factors for mobile-based interventions identified in the literature and the Crowdsensing Method adhered to a majority (90%) of the success factors. This study also contributes information systems design theory by proposing several sets of guidelines for crowdsensing projects and the development of crowdsensing systems. This study fulfils an identified need to study the applicability of crowdsensing for water resource monitoring and explores how a crowdsensing method can create a smart community.

The cultivation of Gracilaria gracilis in the Swartkops estuary, South Africa

- Description: Gracilaria gracilis (Stackhouse) M. Steentoft, L. M. Irvine & W. F. Farnham is a species of red macroalgae which occurs naturally within the Swartkops Estuary in South Africa. Bunches of the species were inserted into the lay of ropes, which were then either suspended in the water column at the Tiger Bay Marina, or staked at the surface of the sediment in the mudflats where the initial material was harvested from. The first site was used for the suspended growth trial, while the second site was for the bottom grown trial. Both of the growth trials ran for the same time period, which was approximately three weeks per season. This study examined three facets of the G. gracilis material that was grown. The growth rate and biomass were recorded in order to examine the potential for the growth of the species on longlines within the estuary. Secondly, the crude nutritional content of the species was determined in order to assess its potential for use as an abalone feed, specifically the dry and ashed weights, and the protein, carbohydrate and lipid contents. Lastly, the cultivated biomass was examined in terms of its agar gel yield and properties, which included the gel strength and the gel –melting and –forming temperatures. The growth rates for the cultivated G. gracilis were poor, and the largest positive growth rate of 1.17 ± 3.2% was found in spring, for the bottom grown trial. This season also saw the only increase in biomass above that which was seeded onto the line, and the final average weight after 22 days was 354.28 ± 68.04 g. All other growth rates were negative, and biomass declined in both growth trials across all the seasons. The suspended growth trials showed more extreme declines, and were heavily contaminated with epiphytic material, while the bottom grown trial had more gradual decrease, and it is likely that conditions associated with the culture systems were not ideal. The largest percentage of protein was found in autumn, where the bottom grown trial had a value of 88 ± 2.32%. For all the seasons, the lipid content had an average percentage of 1.80 ± 0.75% for the bottom grown trial, while the suspended trial was very similar, and had a value of 1.74 ± 0.58%. The nutritional composition of the cultivated biomass was not found to be a good abalone feed in comparison to other studies in land-based aquaculture facilities. Agar characteristics extracted in the present study showed the greatest potential for use as a valuable economic resource. The native agar extracted from the cultivated material was very close to the standards set by United States Pharmacopeia for all the characteristics, and has the potential to possibly be improved further by using an alkali treatment.

Syntheses and photophysico-chemical properties of phthalocyanines in the presence of silica nanoparticles

- Description: This thesis reports on the syntheses and characterizationof symmetrical (charged and neutral), asymmetrical (neutral) metallophthalocyanines (MPcs) and subphthalocyanines (SubPcs). The charged and neutral Pcs were physically doped onto silica nanoparticles (SiNPs). The asymmetrical MPc was also chemically linked to SiNPs. Spectroscopic and microscopic techniques were used to confirm the formation of SiNPs-MPc conjugates. The photophysics and photochemistry of the MPcs were assessed when alone and in conjugates (with SiNPs). The studies showed no significant changes in fluorescence quantum yields (ϕF) and fluorescence lifetimes (ϕF) of MPcs following doping except for 2-SiNPs (2 = Zn tetraaminophenoxyphthalocyanines) and 6-SiNPs (doped) (6 = Zn tris[(4-(pyridine-4-ylthio)2-thio-4-methylthiazol-5yl) acetic acid phthalocyanine) where there was a decrease in the ϕF value. Also for 1-SiNPs (1 = unsubstituted ZnPc) there was an elongation in τF which could be due to the protection offered by SiNPs. Both charged/neutral MPcs displayed high triplet quantum yields (ϕT) and singlet quantum yields (ϕΔ) following doping except for 2-SiNPs where there was a decrease in the latter. For 1-SiNPs there was an increase in ϕT but a decrease inϕΔ .There wasa decrease in ϕT and an increase in ϕΔfor4-SiNPs (4 = Zn tetrasulfophenoxyphthalocyanine), the decrease in ϕT could be due to the orientation of theMPc in SiNPs. An increase in both ϕT and ϕΔ for 6-SiNPs (linked) compared to 6-SiNPs (doped) was observed. Complex 5 (5 = Zn tetra-kis-(dodecylmercapto) phthalocyanine) showed a low ϕΔ value.

- Description: The increase in the use of social media has led many researchers and companies to investigate the potential uses of the data that is generated by these social media platforms. This research study investigates how the use of sentiment variables, obtained from the social media platform Twitter, can be used to augment housing transfer data in order to develop a predictive model. The Design Science Research (DSR) methodology was followed, guided by a Social Media Framework. Experimentation was required within the Design Cycle of the DSR methodology, which lead to the adoption of the Experimental Research methodology within this cycle. An initial literature review identified regression models for property price prediction. Through experimentation, Gradient Boosting regression was identified as an optimal regression model for this purpose. Thereafter a review of sentiment analysis models was conducted which resulted in the proposal of a CNN-LSTM model for the classification of Tweets. Initial experimentation conducted with this proposed model resulted in an obtained accuracy comparable to the top performing sentiment analysis models identified. A dataset obtained through SemEval, a series of evaluations of computational semantic analysis systems, was used for this phase. For the final experimentation, The CNN-LSTM model was used to obtain sentiment variables from Tweets that were collected from the Western Cape Province in 2017. This property dataset was augmented with the sentiment variables, after which experimentation was conducted by applying Gradient Boosting regression. The augmentation was done in two ways, either based on suburb pertaining to the property, or to the month in which the property was transferred. The results indicate that a model for Property Price Prediction Utilising Sentiment Analysis demonstrates a small improvement when suburb-based sentiment, obtained from Tweets with a minimum threshold per suburb, is utilised. An important finding was the fact that, when geo-coordinates are removed from the dataset, the sentiment variables replace them in the regression results, producing the same level as accuracy as when the coordinates are included.

- Description: Eight new oxovanadium(IV) dithiocarbamate complexes, formed by the interaction of vanadyl sulfate pentahydrate and as-synthesized dithiocarbamate ligands, potassium N-morpholine-4- dithiocarbamate ligand (L1), potassium N-piperidinedithiocarbamate (L2), potassium N-(4- methoxyphenyl)dithiocarbamate ligand (L3), potassium N-diallyldithiocarbamate ligand (L4), potassium N-phenyldithiocarbamate ligand (L5), potassium N-dibenzylminedithiocarbamate ligand (L6), potassium N-(4-methylphenyl)dithiocarbamate ligand (L7), N-(1Himidazoledithiocarbamate) fluoride ligand (L8) in aqueous ethanol are described. The resulting complexes were elemental analyses, molar conductance, infrared, electronic, and nuclear magnetic resonance spectral studies. The spectral studies confirmed the formation of the ligands and complexes. Four complexes [VO(L1)2], [VO(L3)2], [VO(L6)2], and [VO(L8)2] derived from potassium N-morpholine-4-dithiocarbamate ligand (L1), potassium N-(4- methoxyphenyl)dithiocarbamate ligand (L3), potassium N-dibenzylminedithiocarbamate ligand (L6), and N-(1H-imidazoledithiocarbamate) fluoride ligand (L8) were characterized using cyclic voltammetry, to study their electrochemical behavior. Square pyramid oxovanadium(IV) complexes were proposed with the metal ion coordinated to two bidentate dithiocarbamate ligands through sulphur to complete its square pyramidal geometry. The anticancer screening of four oxovanadium(IV) dithiocarbamate complexes [VO(L1)2], [VO(L3)2], [VO(L6)2], and [VO(L8)2] are described.

Critical success factors of South African construction companies

- Description: The aim of the study was to identify the driving forces of the South African construction industry for registered cidb Grade 9 companies. The next objective was to identify the critical success factors (CSF) that are required to be used by the forward-looking and pro-active cidb Grade 9 companies. The most important critical success factors were then highlighted to form a core group of CSFs for construction industry. From this core group of CSFs an attempt was then made to identify business strategies that can assist companies in gaining a competitive advantage in the construction industry of South Africa. The research was based on the problem statement from which the sub-questions and hypothesis were derived. The primary data was collected through a mixed method of quantitative and qualitative research. A ontological philosophy was used for the research to effectively collaborate with the mixed method of research used. The approach of the research was a mix of both inductive and deductive, as it pursued to identify the forces and factors dominating the construction industry. The study subsequently determined what business strategies best suit these results. The research questionnaire was distributed between senior managers of grade 9 cidb construction companies and found that the most eminent driving forces of the industry are political and government decision making, as well as economic conditions. The CSFs showed a clear indication of what the respondents perceived as the most important factors. Although there have been many advances in technology and processes over the years, the most important CSF for contractors are still the processes of bidding and tendering and the factors relating to these. The qualitative research portion of the study was an investigation on what business strategies were best related to the key driving forces and most important CSFs. Interviews were held with directors of cidb grade 9 construction companies. The themes that were identified from the analyses of the qualitative data were that business strategies relevant to differentiation, diversification, cost leadership or hybrid were seen as the most effective for construction companies. The bidding and tendering process is not merely the same process that it used to be. Frequent reviews of what the industry is going through have also become a major necessity. The research was limited to companies registered as grade 9 with the Construction Industry Development Board. The research is seen as valued as the construction industry is highly important towards the development of the economy of a country and the impact that it has on communities.

Factors influencing survivorship of Portulacaria afra cuttings in restoration of degraded lands

- Description: Heavy over-browsing, over-grazing, and over-trampling has caused the degradation of vast areas of subtropical thicket, including parts of the Greater Addo Elephant National Park (GAENP). Severe degradation has resulted in the loss of vegetation cover and the deterioration of soils and soil functioning. The South African Department of Environmental Affairs (DEA), and specifically the Natural Resource Management Programme (NRM), have invested in restoration initiatives such as the Subtropical Thicket Restoration Programme (STRP), which has aimed to demonstrate the logistical and practical feasibility of restoring these degraded thickets by planting en masse Portulacaria afra (spekboom) cuttings. However, the planted P. afra cuttings often succumb to various abiotic and biotic stresses. This project aimed to identify various factors which may influence the survival of P. afra cuttings, including i) soil properties, ii) water (rainfall, infiltration, run-off, etc.), iii) browsing by mega-herbivores, iv) topographical features such as slope and aspect, or v) plant species or communities. Biodiversity and carbon baselines were carried out at 50 plots within the Main Camp, Darlington and Kabouga sections of the GAENP, providing pre-planting vegetation and soil data. The en masse planting of P. afra occurred following the completion of the baselines. Following a minimum rest period of three months after planting, survivorship counts were carried out at all plots. Infiltration rate had a significant positive relationship with planted P. afra survivorship across all of the sites. Positive trends between increases in litter and root biomass and planted P. afra cuttings survivorship in Kabouga were also evident. Magnesium, water-holding capacity and increasing soil clay percentage were found to have a significant negative relationship with planted P. afra survivorship in Kabouga. Aluminium, magnesium, and boron all had significant positive relationships with planted P. afra cuttings survivorship in Addo Main Camp. No soil variables had any significant relationship with survivorship of planted P. afra cuttings at Darlington. The presence of elephants (Loxidonta africana) had a significant negative relationship with planted P. afra survivorship. Plant communities in more ‘moderately’ degraded states, characterized by the retention of canopy dominant species, woody vegetation cover, limited bareground and higher cover of litter had higher survivorship of planted P. afra cuttings than those in more severely degraded states. The grass Panicum maximum, and forb Atriplex semibaccata were associated with positive relationships with planted P. afra survivorship across all sites. Future research should aim to develop matrices which plot likely survivorship percentages with parameters of measurable variables, such as infiltration, severity of soil capping, browsing densities, chemical indicators, and degradation severity, to identify suitable biophysical planting conditions for P. afra cuttings. Further research should assess whether the presence of elephants, in high densities, can reduce the economic feasibility of the en masse plantings of P. afra cuttings in attempts to restore degraded landscapes.
