- Description: The public healthcare sectors within developing nations face a lot of challenges because of constrained resources available to them. The South African public healthcare sector is no different. Although it serves the majority of the South African population, most of the financial resources are directed towards the private sector, which serves very few individuals when compared to the public healthcare sector. Apart from that, other challenges that the National Department of Health has to deal with include the lack of sufficiently trained healthcare employees who can work on the different levels of the public healthcare sector, as well as the burden of diseases such as HIV and Aids, tuberculosis and other chronic diseases. In order to improve service delivery, the National Department of Health is introducing Information and Communications Technology interventions that can increase efficiency and reduce costs, thereby improving the quality of service delivery. This research delivers an m-health application user experience framework to be proposed to the National Department of Health in South Africa, in order to assist in scaling up of m-health applications. The m-health applications that can benefit the South African population if scaled up successfully include those that can be used in remote data collection, treatment and compliance, accessing patients records, remote monitoring, communication and training for healthcare workers and applications that can be used for education and awareness. The study focused on three domains: the Human-Computer Interaction domain, public healthcare domain and Health Informatics domain. The proposed framework was realized by investigating mobile user experience components, mobile health requirements and the South African public healthcare domain components that contribute to the m-health user experience framework. This research was conducted through the interpretivist philosophy. Due to the exploratory nature of the study, an application of qualitative methodology was used. The conceptual theoretical framework was validated through a single case study approach by m-health user experience experts, who reside in South Africa. Data were analysed inductively. An m-health user experience framework was provided at the end of the study. An m-health user experience framework can assist the National Department of Health to look into design issues, address m-health requirements and put the domain needs in place, thus enabling the Department to successfully scale up implementations of m-health applications nationwide.

- Description: The human immunodeficiency virus/acquired immune-deficiency syndrome pandemic (HIV/AIDS) continues to increase in prevalence worldwide, particularly in South Africa. “AIDS, (Acquired Immune Deficiency Syndrome), has devastated the social and economic fabric of African societies, made orphans of a whole generation of children and become the epicentre of the HIV/AIDS pandemic” (Fassin, 2007: 76). Like the virus itself, the AIDS crisis mutates rapidly. Children who are AIDS orphans living in South Africa, as in other African countries, suffer from recurrent psychological trauma. It starts with the illness and death of their parents and is followed by cycles of poverty, malnutrition, stigma, exploitation, sickness and often sexual abuse. The figures concerning those affected, which are staggering, offer various predictions regarding the number of orphans left in the wake of the pandemic. Between 1990 and 2003, the number of orphaned children rose from fewer than 1 million to more than 12 million in sub-Saharan Africa (UNICEF, 2005: 68). South Africa is severely affected by the AIDS pandemic, with the largest number of HIV infections in the world, a total of 5.7 million (RSA, 2010: 10), affecting approximately 3.2 million women and 280 000 children aged from 0 to14 years. There is a significant variation in HIV prevalence per province, with the Eastern Cape (EC) reportedly having an average prevalence rate of 28 percent. Hence South Africa is likely to be the country with highest percentage of children orphaned by AIDS within its population. Orphanhood is a major consequence of the AIDS pandemic in South Africa with an estimated 2.2 million AIDS-orphaned children, 11,188 per 100,000 by 2015. Most children who are AIDS orphans living in township communities live predominantly uncared for and unsupported. Therefore the purpose of this research study was to develop strategies to provide care and support to children who are AIDS orphans living in township communities. To achieve the purpose of this research study, a qualitative, exploratory, descriptive and contextual design was used by the researcher to gain insight from health and social care practitioners who render care and support to children who are AIDS orphans living in these communities. The data from in-depth interviews with the health and social care practitoners was used by the researcher to develop strategies to provide care and support to children who are AIDS orphans living in township communities. The study comprised the following four phases: Phase One: During this phase, the researcher will present an overview of the current legislative framework policies at an international, national and provincial level, regarding the the rights of children living in South Africa. Phase Two: During this phase data from two research populations were collected and analysed. As the research process of this study dealt with two groups of participants, namely in group one health care practitioners and group two comprised social workers and psychologists, who work in the township communities to provide care and support to children who are AIDS orphans living in these communities. The researcher discussed each group separately in the discussion of phase two of the study. Phase Three: Comprised the themes identified in the data gathered from the transcribed in-depth interviews, the field notes as well as the reflective journals were cross-validated to ensure trustworthiness of the data which was then organised into a conceptual framework. The conceptual framework was used to clarify the relationships of the concepts and the themes identified during the research process and also used to link the gathered data to previously established models and theories (Schneider, 2004: 133). Phase Four: The last phase of the research design, focused on the development of the “Steps of progression strategies” to provide holistic care and support to children who are AIDS orphans living in township communities. This was done making use of the themes identified during data analysis and the literature sources used throughout this research process. The evaluation criteria of Chinn and Kramer (2008: 237-248) were used to evaluate the strategies. It is therefore concluded that the researcher succeeded in achieving the purpose of this study because strategies which were understandable, clear, simple, applicable and significant to nursing practice have been developed for use by the Department of Health and Department of Social Development as well as primary health and social care practitioners to provide holistic care and support to children who are AIDS orphans living in township communities in South Africa.

- Description: There is little understanding of the implementation efficiency and effectiveness of restoring plant invaded landscapes within operational contexts. South Africa's Working for Water (WfW) programme is arguably the most ambitious alien plant control programme in the world, yet little is known about its cost-effectiveness and the challenges it faces in linking poverty and environmental objectives. My first aim was to assess the cost-effectiveness of invasive plant removal, and the factors that underpin its effectiveness over large spatial and temporal scales. The second aim was to compare the accuracy of evidence-based findings with managers' experience-based beliefs, and to assess whether managers are willing to change their beliefs after being exposed to it. The third aim was to assess the costs and benefits of removal versus removal followed by active native re-vegetation. My final aim is to assess the challenges and lessons learnt by managers linking ecological restoration with poverty alleviation objectives, specifically within the public works model. My study area was focused primarily on two WfW river catchment projects in the western region of the Eastern Cape province. I adopted an interdisciplinary approach drawing from a range of methods such as observational studies, statistical modelling and interviews with managers. The key findings were that control efforts in the two catchment projects are largely inadequate owing to many sites being re-invaded and not enough resources being allocated to the catchments. It would take between 54 and 695 years to clear the respective catchments. In terms of cost-effectiveness, my results exceeded previous estimates by 1.5 to 8.6 times for each catchment project. After being exposed to the evidence-based findings, the managers did not change their beliefs when it came to forecasting the future effectiveness. I found that active native re-vegetation after removal of invasive plants is very costly and that priority should be given to understanding the effectiveness of the removal treatments on native species recovery. The managers cited significant challenges in effectively and efficiently meeting the programmes dual objectives. Based on a broader review of the public works literature I recommend WfW re-examine the type of public works they currently use.

- Description: The public higher education sector in South Africa is governed by an act of Parliament which defines the governance organs and mechanisms required within institutions and the sector. This doctoral thesis begins by contextualising ICT governance within the levels of systemic governance in South Africa that have relevance to this study. It then continues by advancing the understanding of the state of Information and Communication Technology (ICT) governance in public higher education institutions in South Africa by investigating the level of pervasiveness, the level of ICT process maturity and the alignment of ICT to the institutional processes within these institutions. Subsequently, this thesis results in the production of a contextually appropriate value framework that contributes towards an understanding of the need for the improvement of ICT governance. A survey of the extant literature on governance, ICT governance, and the frameworks and standards associated with governance indicates that the methodological approach currently favoured in ICT governance research is predominantly interpretive and based on surveys and interviews. This study leans towards a Design Science approach paradigm using surveys for the qualitative data collection process. The empirical research data was gathered from the highest ranking ICT officers at public higher education institutions in South Africa. This study makes contributions on a number of levels – from the first known measurement of ICT pervasiveness within the context of higher education, to the measurement of ICT process maturity and the measurement of the alignment of ICT and institutional processes, to the design of a value framework that breaks down governance into layers indicating which governance organs are appropriate at the relevant layers and which governance mechanisms can be used to address governance at each layer of governance. This study therefore makes a contribution to the ICT domain for development research by demonstrating praxis for the improvement of ICT governance implementation in the public higher education sector in South Africa. This study makes both method design and practical contributions to the ICT body of knowledge at various stages of the research process. These contributions include reflection on and analysis of the data gathering approaches that occur within the public higher education sector and in the complex environment of institutional autonomy and academic freedom; the use of Agency Theory to support the ‗governance intent‘ transfer between the layers of governance in the value framework and the synthesis of the improvement of ICT governance with value creation. The findings of this study further contribute to an understanding of the dynamics and interrelatedness of aspects of governance and management, while making a contribution to theoretical research by extending practical and empirical understanding of the ICT governance environment in the public higher education sector in South Africa.

- Description: In 1932, six Missionary Franciscan Friars from Bavaria came to South Africa at the invitation of Bishop Adalbero Michael Fleischer (1874-1963), a Mariannhill missionary in Natal. His vision was that they would establish a “Franciscan Family” there. Despite this invitation, the Friars were not welcomed by the Mariannhill community and went on to found the Mount Currie Prefecture (Diocese of Kokstad) in 1935. There the Franciscans became involved in the life of the ordinary people, who were otherwise abandoned by the government. Among other activities, they provided schools and gardens, and embarked on skills development projects for the youth. The Holy Cross Sisters - whose arrival in 1926 pre-dated that of the Friars - played a crucial role in these developments. From 1952, their educational work would be supplemented by the medical work of the Franciscan Missionaries of Mary Sisters. The arrival of Irish Franciscans (1946) and English Franciscans (1948) brought tensions and differing interpretations of the form that the vision should take. In addition, the Friars were men of their times and products of their particular backgrounds. For example, while some saw their African converts as equals, others were prejudiced against them. However, this thesis – the work of an “insider” to the faith and to the Order, argues that despite tensions and shortcomings, the settlement, and continuing presence, of the Franciscan Friars and Sisters in the Eastern Cape was an eventual realization of Bishop Fleischer’s vision, even though in a slightly different manner from the original ideal. In the early nineteen sixties a new and powerful spirit invaded the Catholic Church. The Second Vatican Council took place from 1962 to 1965. It may be argued that, from that time onward, the Church has never been the same again. In accordance with this call and invitation of the Second Vatican Council for quite some time it has been my earnest desire to get down into a more in-depth study of the history of the Franciscans in South Africa than that provided by the two existing works on this topic. It is my hope that this thesis will contribute to a soul-searching of the Franciscans as they continue to live and minister to the people of South Africa. This history will hopefully teach us to learn from and be inspired by the great and heroic deeds of our spiritual forebears – our Franciscan brothers and sisters – and also teach us to learn from their mistakes.

Coping with HIV and AIDS in marginal communities: a case study of Chivanhu Settlement in Nemanwa, Masvingo, Zimbabwe

- Description: This thesis seeks to understand and analyse HIV and AIDS and rural livelihoods in Zimbabwe with particular reference to an isolated and marginalised informal settlement called Chivanhu in Masvingo Province. The focus is specifically on questions around HIV susceptibility, AIDS vulnerability and household resilience. In this regard, it is important to recognise that HIV and AIDS cannot be lumped together as one medical or social condition. Rather, there is a progression from HIV infection to AIDS-related chronic illnesses to possible death, and livelihood strategies often alter along this HIV and AIDS time-line. Zimbabwe for over a decade now has gone through a series of economic and political crises which have impacted detrimentally on both urban and rural livelihoods, even for those households which are not directly affected by the HIV and AIDS pandemic. With the economy in free-fall, households have had to pursue a range of livelihood strategies in order to sustain themselves. These socio-economic conditions have in many ways facilitated susceptibility to HIV infection and vulnerability to AIDS. Many studies have examined this in relation to wellentrenched and stable communities in rural Zimbabwe. But the livelihood dynamics for such communities are significantly different to more unstable and informal settlements like Chivanhu, as thesis seeks to show. At the same time, the thesis offers a longitudinal study which is able to map the changes to the livelihoods of infected and affected households in Chivanhu. Though recognising the debilitating effects of the pandemic on these households, it also raises questions about the possible resilience of certain households despite great adversity. In doing so, it goes beyond the individual and household levels of analysis to consider the role of clusters (or groups of households) in responding to the impacts of HIV and AIDS. In this regard, particular emphasis is placed on gender and orphanhood. In the end, the thesis offers a nuanced analysis of the everyday complexities and challenges for affected households in a marginalised and informal rural community in Zimbabwe and thereby makes a contribution to re-theorising HIV and AIDS and rural livelihoods more broadly.

- Description: Different approaches to surface modification were investigated in this work on gold, glassy carbon, multi-walled carbon nanotube paper and on single-walled carbon nanotubes adsorbed on glassy carbon. These approaches include electrochemical grafting, electropolymerisation, click chemistry, axial ligation, adsorption and self-assembled monolayers. The modified surfaces were characterised using a variety of techniques; predominantly electrochemistry, scanning electrochemical microscopy and X-ray photoelectron spectroscopy. For the formation of self-assembled monolayers on gold, four new manganese(III) phthalocyanines (1a-d), octa-substituted at the peripheral position with pentylthio, decylthio, benzylthio, and phenylthio groups were synthesized and characterised. X-ray photoelectron spectroscopy was used to show the formation of a sulphur-gold bond. A number of approaches using 4-azidoaniline (2a) combined with azide-alkyne click chemistry and electrochemistry were also used to anchor ferrocene and pyridine moieties on to the carbon surfaces, including direct in situ diazotation and grafting, electropolymerisation, and the synthesis of the diazonium salt followed by grafting. Iron phthalocyanine was linked to the pyridine-clicked surfaces through axial ligation, where the strong axial bond formed by the interaction between the central metal and the lone pair of the nitrogen in the pyridine group resulted in stable modified electrodes. The potential of these surfaces for the detection of analytes such as thiocyanate, hydrazine and sulphite are briefly shown as well. This work also describes for the first time the possibility of performing local micro-electrochemical grafting of a gold substrate by 4-azidobenzenediazonium (2b) using scanning electrochemical microscopy in a single and simple one step approach, without complications from adsorption.

- Description: The importance of infrared (IR) technology (for detection in the 3-5 μm and 8-14 μm atmospheric windows) has spread from military applications to civilian applications since World War II. The commercial IR detector market in these wavelength ranges is dominated by mercury cadmium telluride (MCT) alloys. The use of these alloys has, however, been faced with technological difficulties. One of the materials that have been tipped to be suitable to replace MCT is InAs/InxGa1-xSb strained layer superlattices (SLS’s). Atmospheric pressure metal-organic vapour phase epitaxy (MOVPE) has been used to grow InAs/GaSb strained layer superlattices (SLS’s) at 510 °C in this study. This is a starting point towards the development of MOVPE InAs/InxGa1-xSb SLS’s using the same system. Before the SLS’s could be attempted, the growth parameters for GaSb were optimised. Growth parameters for InAs were taken from reports on previous studies conducted using the same reactor. Initially, trimethylgallium, a source that has been used extensively in the same growth system for the growth of GaSb and InxGa1-xSb was intended to be used for gallium species. The high growth rates yielded by this source were too large for the growth of SLS structures, however. Thus, triethylgallium (rarely used for atmospheric pressure MOVPE) was utilized. GaSb layers (between 1 and 2 μm thick) were grown at two different temperatures (550 °C and 510 °C) with a varying V/III ratio. A V/III ratio of 1.5 was found to be optimal at 550 °C. However, the low incorporation efficiency of indium into GaSb at this temperature was inadequate to obtain InxGa1-xSb with an indium mole fraction (x) of around 0.3, which had previously been reported to be optimal for the performance of InAs/InxGa1-xSb SLS’s, due to the maximum splitting of the valence mini bands for this composition. The growth temperature was thus lowered to 510 °C. This resulted in an increase in the optimum V/III ratio to 1.75 for GaSb and yielded much higher incorporation efficiencies of indium in InxGa1-xSb. However, this lower growth temperature also produced poorer surface morphologies for both the binary and ternary layers, due to the reduced surface diffusion of the adsorbed species. An interface control study during the growth of InAs/GaSb SLS’s was subsequently conducted, by investigating the influence of different gas switching sequences on the interface type and quality. It was noted that the growth of SLS’s without any growth interruptions at the interfaces leads to tensile strained SLS’s (GaAs-like interfaces) with a rather large lattice mismatch. A 5 second flow of TMSb over the InAs surface and a flow of H2 over GaSb surface yielded compressively strained SLS’s. Flowing TMIn for 1 second and following by a flow of TMSb for 4 seconds over the GaSb surface, while flowing H2 for 5 seconds over the InAs surface, resulted in SLS’s with GaAs-like interfacial layers and a reduced lattice mismatch. Temperature gradients across the surface of the susceptor led to SLS’s with different structural quality. High resolution x-ray diffraction (HRXRD) was used to determine the thicknesses as well as the type of interfacial layers. The physical parameters of the SLS’s obtained from simulating the HRXRD spectra were comparable to the parameters obtained from cross sectional transmission electron microscopy (XTEM) images. The thicknesses of the layers and the interface type played a major role in determining the cut-off wavelength of the SLS’s.

The screening and characterisation of compounds for modulators of heat shock protein (Hsp90) in a breast cancer cell model

- Description: Breast cancer is a leading cause of cancer death in Africa. Hsp90 has been identified as a target for anti-cancer treatments as its inhibition results in the disruption and ubiquitin–proteasome degradation of activated oncoproteins. Currently, there are no US Food and Drug Administration approved Hsp90 inhibitor drugs and existing Hsp90 inhibitors such as geldanamycin and novobiocin are hepatotoxic and display a low affinity for Hsp90, respectively. Therefore, there is a need for the development of Hsp90 inhibitors with improved inhibitory properties. In this study twelve natural compounds bearing a quinone nucleus were screened and characterised for the modulation of Hsp90. The compounds analysed formed three series; the sargaquinoic acid (SQA), naphthoquinone, and pyrroloiminoquinone alkaloid series. Certain compounds exhibited half maximal inhibitory concentrations of between 3.32 μM and 12.4 μM, while others showed no antiproliferative activity at concentrations of up to 500 μM in the MDA-MB-231 breast adenocarcinoma cell line. Immunofluorescence and Western analyses indicated that the modulation of Hsp90 and partner proteins by SQA was more similar to that of novobiocin. Isothermal titration calorimetry analyses suggested that SQA interacted with Hsp90β with a low affinity, and saturation-transfer difference nuclear magnetic resonance confirmed that this interaction with Hsp90β occurred through the methyl moiety bound to 1, 4 benzoquinone of SQA. Pulldown assays indicated SQA disrupted the association between Hsp90 and Hop dose-dependently, more similarly to novobiocin. Immunofluorescence and Western analyses performed on naphthoquinone and pyrroloiminoquinone alkaloid compounds indicated modulation of Hsp90 and Hsp90 partner proteins by the compounds. Naphthoquinone compounds were prioritised for analysis for binding to Hsp90β over the pyrroloiminoquinone alkaloid compounds. Lapachol interacted with Hsp90β with a low affinity however; this interaction was thought to be too weak to disrupt the association of Hsp90 and Hop. The remaining naphthoquinone compounds showed no interaction with Hsp90β, thus allowing the determination of a preliminary structure-activity relationship for these compounds. To the best of our knowledge, this is the first study to describe a systematic subcellular analysis of the effects of geldanamycin and novobiocin in comparison to sargaquinoic acid and compounds of the naphthoquinone and pyrroloquinoline scaffold on Hsp90 and its partner proteins. , Microsoft� Word 2010 , Adobe Acrobat 9.54 Paper Capture Plug-in
