The larval ecology and the oceanographic conditions in the area suggest a good potential for the enhancement of roman stocks outside the reserve through larval dispersal. A high rate of illegal fishing just inside the seaward boundary of the MPA could severely compromise its function.We suggest that a change of the seaward boundary of the reserve to coincide with a latitudinal line could increase its function as a harvest refuge for resident reef fishes such as roman, facilitate voluntary compliance and monitoring and prosecution of illegal fishing without a significant negative impact on the commercial linefishing fleet in the area. Simple adjustments such as the one proposed here could be attempted at a number of South African MPAs as they would be beneficial to achieve fishery and conservation goals alike.

- Description: Judge Elna Revelas’s decision in the case of Mohamed v Mohamed 1 may be described as one of those run-of-the-mill applications in terms of rule 43 which are routinely heard on motion court days in any one of our high courts across the country. This case note suggests that her decision belies such a description. Instead, this note suggests that her decision marks a move away from existing jurisprudence on Muslim marriages in a way which may undermine, rather than promote, the recognition and respect for the marriage institutions of different religious systems and beliefs. I tentatively suggest that by granting the rule 43 application the court may have effectively imposed civil marriage obligations on a religious marriage even though the parties had not concluded a marriage in terms of the Marriage Act.2 As such, the decision has potentially radical consequences for parties in Muslim marriages and highlights the complex issues that courts have had to face in the last two decades without any guiding legislation. In order to understand the judgment properly, its context has to be considered. This context includes (1) the numerous judgments extending protection to women in Muslim marriages in the last two decades against the backdrop of the coming into effect of the Constitution of the Republic of South Africa, 1996; and (2) the application by the Women’s Legal Centre Trust (hereafter “WLCT”) to the Constitutional Court to force the President and Parliament to enact legislation to recognise and protect Muslim marriages.

Nine putative callose synthase genes were identified in a data base search, of which eight were expressed in the leaves, but with similar level of expression in control and aphid-infested tissue. Four out of 12 examined β-1,3-glucanases were expressed in the leaves, and three of them were up-regulated in aphid-infested tissue. They were all more strongly induced by RWA than BCA. The results suggest that callose accumulation may be partly responsible for the symptoms resulting from RWA feeding and that a callose-inducing signal may be transported in the phloem. Furthermore it is concluded that the absence of callose deposition in BCA-infested leaves is not due to a stronger induction of callose-degrading β-1,3-glucanases in this tissue, as compared to RWA-infested leaves.

We have shown here by site directed mutagenesis and size exclusion chromatography for the first time that the TPR1 and TPR2B domains of Hop independently dimerized, and that the dimerization of TPR2B was not dependent on its predicted two-carboxylate clamp residues. - Description: The relationships between the spatial distribution of gully erosion and topographic thresholds in the form of slope angle, position and configuration, as well as land use change in the form of abandoned lands were examined in several affected catchments of the Eastern Cape Province, South Africa. Land use and permanent gullies were mapped, digitized from orthophoto maps in Arc/info 3.5.2 GIS and converted to shapefiles using ArcView 3.2 GIS. Relationships between the mapped phenomena and topographic variables were sought using a Digital Elevation Model (DEM) in Idrisi Kilimanjaro GIS. A comparison between areas with a high potential for gullying and actual gully erosion was made using the Stream Power Index (SPI) as a surrogate for critical flow shear stress. Field surveys were also conducted to assess the present condition of the gullied sites as well as to validate DEM derivations. Seventy five percent of the gullied area was noted to lie on abandoned lands. A predominance of gullying in concave bottom lands was also identified. The SPI values highlighted a distinct preferential topographic zone for gully location. A conceptual model depicting the interaction between land use and topographic parameters to induce gully erosion was developed. This should assist local authorities to develop a policy regarding management of abandoned lands.

The protein sacsin, which is mutated in the early-onset neurodegenerative disease autosomal recessive spastic ataxia of Charlevoix-Saguenay, is a node in this interactome. Here, we have established the neuronal expression of sacsin and functionally characterized domains of the 4579 amino acid protein. Sacsin is most highly expressed in large neurons, particularly within brain motor systems, including cerebellar Purkinje cells. Its subcellular localization in SH-SY5Y neuroblastoma cells was predominantly cytoplasmic with a mitochondrial component. We identified a putative ubiquitin-like (UbL) domain at the N-terminus of sacsin and demonstrated an interaction with the proteasome. Furthermore, sacsin contains a predicted J-domain, the defining feature of DnaJ/Hsp40 proteins. Using a bacterial complementation assay, the sacsin J-domain was demonstrated to be functional. The presence of both UbL and J-domains in sacsin suggests that it may integrate the ubiquitin–proteasome system and Hsp70 function to a specific cellular role. The Hsp70 chaperone machinery is an important component of the cellular response towards aggregation prone mutant proteins that are associated with neurodegenerative diseases. We therefore investigated the effects of siRNA-mediated sacsin knockdown on polyglutamine-expanded ataxin-1. Importantly, SACS siRNA did not affect cell viability with GFP-ataxin-1[30Q], but enhanced the toxicity of GFP-ataxin- 1[82Q], suggesting that sacsin is protective against mutant ataxin-1. Thus, sacsin is an ataxia protein and a regulator of the Hsp70 chaperone machinery that is implicated in the processing of other ataxialinked proteins.
