A total of 432 1day old unsexed broiler chicks (Aviane 48) were randomly allocated to four dietary treatments (TRTS) in 72 cages. There were 18 cages per treatment and each cage allocated 6 chicks. Water and feed was provided at ad libitum. The feeding phases were, prestarter (0-7 Days), starter (8-18 Days), grower (19-28 Days), finisher (29-35 Days). The four TRTS contained graded levels of MOLM at 1000g/ton, 750g/ton, 500g/ton, and 0g/ton (control), respectively. The birds were slaughtered at 35 days of age. Breast muscles were sampled for meat, ultimate pH (pHu ), colour, drip loss over a 7 days shelf-life test. After each day’s test sub-samples were dipped in liquid nitrogen and kept at -180 C for thiobarbituric acid reactive substances determination. On Day1 and Day 7 extra sub-samples were also kept at -180 C for fatty acids analysis.The TRTS had no effect on average feed intake (AFI), feed conversion efficiency (FCE), and on average daily gain (ADG). Slaughter weight (SW), carcass weight (CW), dressing percentage and gizzard weight (GW) values were similar in all TRTS. Liver weight (LW), heart weight (HW), and gastro-intestinal fat (GIF) differed in all the TRTS, with treatment 2 having the highest value of HW (28.3±2.55), and LW (44.2±1.60) was the highest on treatment 4 . The pH values in all TRTS were constant from Day1 to Day5, reached peak on Day6, and then declined on Day7. Meat from broilers given treatment 1 with MOLM (1000g/ton) had the highest lightness (L*) values. The redness (a*) values were the highest in meat from treatment 2 (750g/ton MOLM). Treatments had no effect on yellowness (b*) values and on drip loss of the breasts. During storage L* values were high from Day1 to Day5 and decreased from Day6 to Day7. Drip loss increased with storage time as expected. Treatment 4 (control) had the highest proportions of poly-unsaturated fatty acids (PUFA) (30.3±1.87). Treatment 1 (1000g/ton) had the highest proportion of saturated fatty acids (SFA) (60.9±1.87). Treatment 1 (1000g/ton) had the highest proportion of SFA (60.9±4.30). Treatment 2 (750g/ton) had the highest n-6/n-3 ratio than other TRTS. Days had no effect (P>0.05) on PUFA, SFA, and n-6/n-3 ratio. Treatment 1 had a highest amount of malondialdehyde (MDA), treatment 4 had no effect (P>0.05) on MDA . Storage time had an effect (P<0.05) on MDA levels, except for on Day1 and Day7. Day2 had the highest amount of MDA (0.7±0.08). The use of MOLM as an additive in broiler diets reduced lipid oxidation in meat, and maintained the quality of the broiler meat during storage. It also did not have any adverse effects on the growth performance of broilers. Therefore, it has the potential to be used as an additive in broiler diets.

- Description: This study was carried out to determine alternative approaches used by resource-limited farmers to enhance fertility and in the treatment of reproduction ailments of goats in Mount Frere area, Eastern Cape Province of South Africa. Furthermore, information on farmer perceptions pertaining to these practices was also gathered. A questionnaire survey was conducted to determine materials used by resource-limited farmers to enhance fertility and treat reproductive ailments. The survey revealed 10 plant species that were commonly used for reproductive health in goats. Elephantorrhizza elephantina (37.1%) and Rhoicissus tomentosa (25.7%) were the most frequently cited plant species. Elephantorrhiza elephantina and R. tomentosa recorded the highest Fidelity Level (FL) values of 92.86% and 75% respectively, for their use in the treatment of reproductive ailments and these were the plants on which further studies were conducted. The study also revealed that farmers had different perceptions on the effectiveness of the traditional medicines. An in-vivo assessment was done to determine efficacy of E. elephantina and R. tomentosa using serum estradiol and progesterone profiles. The highest (P<0.05) serum estradiol and progesterone concentration was observed in animals administered with aqueous extracts of E. elephantina and R. tomentosa mixture while the least (P<0.05) concentrations were observed in the negative control group. Does administered with a mixture of aqueous E. elephantina and R. tomentosa extract had the highest (P<0.05) body weight (65 kg) as well as body condition scores, while the negative control which were administered with distilled water had the least body weights (54 kg) and body condition scores. Does that were administered with a mixture of E.elephantina and R. tomentosa were all pregnant with twins (100%), confirmed by a doppler ultrasound scan.

- Description: This study was done in order to assess the incidence of Staphylococcus species from milk of cows with subclinical mastitis and their antimicrobial sensitivity in some selected antibiotics and Usnea barbata lichen extracts. The study was conducted in two different commercial dairy farms. Staphylococcus species isolates were identified using several biochemical tests which included Gram’s staining test, catalase test and oxidase test. A commercial API® staph kit (bioMerieux, France) was used to confirm the bacterial organisms to their species level. The antimicrobial sensitivity of individual species was determined according to the Clinical Laboratory Standard Institute (CLSI) for the selected antibiotics. Agar well diffusion method and the broth micro-dilution technique were used to determine the sensitivity of Staphylococcus species in U. barbata extracts. A total of 467 milk samples were screened for bacterial identification from the two farms. Fifteen different Staphylococcus species isolates were identified from all milk samples that were examined. The most frequently isolated species included Staphylococcus xylosus (54.34%), Staphylococcus hominis (24.78%), Staphylococcus aureus (16.38%), Staphylococcus saprophyticus (16.12%) and Staphylococcus haemolyticus (11.63%). Most Staphylococcus species were resistant to Penicillin (75.35%), Nalidixic acid (72.55%) and Ampicillin (63%). Furthermore, the bacterial sensitivity evaluation of U. barbata lichen extracted with methanol and ethyl-acetate against selected Staphylococcus species isolates showed 92.31% and 53.85% susceptibility, respectively. The minimum inhibitory concentration (MIC) of the methanol and ethyl-acetate extracts ranged between 0.0390 to 10 mg/ml. There was a relatively high incidence of Staphylococcus species identified in milk of cows with subclinical mastitis from both farms. Conversely, Staphylococcus species isolates were resistant to antibiotics (mostly penicillin and ampicillin) commonly used in the farms. Furthermore, the study investigated the antimicrobial sensitivity of U. barbata extract in-vitro which may validate its use in traditional medicine for treatment of cows with mastitis.

- Description: The objective of the study was to determine sheep growth performance, fleece weights and wool quality characteristics in the sweet and sourveld communal rangelands of the Eastern Cape Province of South Africa. Data were obtained in 80 ewes aged two to three years, selected in four ecologically different communities. Two communities (Roxeni and Tyabane) were in a sweetveld area and the other two communities (Luzi I and Luzi II) were in a sourveld area. The ewes from Tyabane were nondescript, whereas ewes from Roxeni, Luzi I and Luzi II were crossbred between Merinos and non-descript sheep. Selected ewes were monitored for live weight, body condition scoring and fleece weight during shearing. Wool samples were collected from each ewe and were analysed for fibre diameter, comfort factor, clean yield, wool crimps, staple length, staple strength and the Position of break. Sheep from Roxeni were heavier (P <0.05) (39.9 ± 1.1kg) than sheep from Tyabane (29.8 ± 1.1kg). Sheep from Tyabane had the lightest (P <0.05) fleece weight (1.0 ± 1.1kg) and sheep from Roxeni had the heaviest (3.1 ± 1.1kg). There were no significant differences (P ≥0.05) between body condition scores of sheep in Roxeni and the Luzi communities. The live weights of the two-year old ewes were similar (P ≥0.05) to the three-year old ewes’ live weight. However, the three year old ewes had higher (P <0.05) body condition scores than the two year old ewes. Two year old ewes also had lower (P <0.05) fleece weight than the three years old ewes. Wool from Roxeni, Luzi I and Luzi II had lower fibre diameter (P <0.05) than wool from Tyabane. Roxeni sheep had the highest clean yield (P <0.05) of wool, followed by Luzi I and Luzi II; Tyabane had lowest clean yield percentage of wool. Wool from sheep grazing on sweetveld had a higher (P <0.05) clean yield percentage (74.3 ± 0.7%) than those grazing on sourveld (71.7 ± 0.7%). There were higher staple mid-breaks (69.5 ± 5.2) and lower base-breaks (30.5 ± 5.2) in sweetveld (P <0.05) than in sourveld, 7.5 ± 5.2 and 92.5 ± 5.2 for mid-breaks and lower base-breaks, respectively. It was concluded that sheep raised in the sweetveld and bred with purebred Merino rams had higher performance than the ones reared in the sourveld. The young upgraded ewes had a higher wool quality than the older ewes raised on communal rangelands.

- Description: Feeding behaviour of Xhosa lop-eared, Nguni and Nguni × Boer goat genotypes kept on rangelands of the False Thornveld By Archibold Garikayi Bakare The study was conducted at the University of Fort Hare farm to assess the feeding behaviour of three goat genotypes: Xhosa lop-eared (XLE), Nguni (NGN) and Nguni × Boer crossbred (NBC) genotypes. The time spent on different activities (grazing, browsing and other activities) on rangelands was recorded using stop watches. The micro-histological faecal analysis technique was used to identify and quantify the plant species that were consumed by the goats across the four seasons. Time spent browsing was high in post-rainy season for XLE (55.94 %) compared to NGN (52.97 %) and crossbred NBC (45.95 %) goats. The high browsing activity was also noted for XLE in hot-wet and cool-dry season (P < 0.05). The NBC goats on the other hand; devoted most of their time grazing compared to XLE and NGN goats across seasons (P < 0.05). Generally, time of day had no effect (P > 0.05) on foraging activities among the goat breeds. Grewia occidentalis, Panicum maximum and Diospyros lycioides were the most preferred plant species by all genotypes across seasons in the paddock. It was concluded that crossbreds spent more time grazing as compared to XLE, which are more of browsers. It could, therefore, be recommended that XLE complement with grazers (cattle and sheep) for efficient management of feed resources in rangelands.

- Description: Feeding behaviour of Xhosa lop-eared, Nguni and Nguni × Boer goat genotypes kept on rangelands of the False Thornveld By Archibold Garikayi Bakare The study was conducted at the University of Fort Hare farm to assess the feeding behaviour of three goat genotypes: Xhosa lop-eared (XLE), Nguni (NGN) and Nguni × Boer crossbred (NBC) genotypes. The time spent on different activities (grazing, browsing and other activities) on rangelands was recorded using stop watches. The micro-histological faecal analysis technique was used to identify and quantify the plant species that were consumed by the goats across the four seasons. Time spent browsing was high in post-rainy season for XLE (55.94 %) compared to NGN (52.97 %) and crossbred NBC (45.95 %) goats. The high browsing activity was also noted for XLE in hot-wet and cool-dry season (P < 0.05). The NBC goats on the other hand; devoted most of their time grazing compared to XLE and NGN goats across seasons (P < 0.05). Generally, time of day had no effect (P > 0.05) on foraging activities among the goat breeds. Semi-scavenging (1.3 ± 0.05kg) carcasses were heavier (P<0.05) than full scavenging ones (1.0 ± 0.06kg). The ultimate pH (24h) (pHu) of meat from full scavenging chickens (6.0 ± 0.03) was higher (P<0.05) than that of meat from the semi-scavenging chickens (5.7 ± 0.04). Meat from the full scavenging chickens had higher L* values (60 ± 1.2) than meat from the semi-scavenging chickens (47 ± 0.8). The a* value of meat of full scavenging chickens was significantly (P<0.05) less (4 ± 0.4) than that of meat from semi-scavenging chickens (15 ± 1.3). There was a significant positive (0.60) correlation between pHu and L* of meat from village chickens across rearing systems. The semi-scavenging system meat had better sensory attributes than the meat from the full scavenging system. Female consumers scored the typical flavour of village chicken meat across rearing systems higher (P< 0.05) than male consumers. The Shona consumers scored the meat higher (P< 0.05) than the Xhosa, Zulu, Ndebele and other tribes for initial juiciness, first bite impression and muscle fibre and tenderness of the chicken across rearing systems. - Description: The objective of the study was to evaluate cow reproductive performance in the sweetveld and sourveld communal grazing areas of the Eastern Cape Province, South Africa. In the first experiment, farmer perceptions were obtained using participatory rural appraisals. Farmers ranked lack of fences, tick-borne diseases, poor animal condition during winter and poor breeding practices, respectively as major constraints limiting cattle production in the Eastern Cape. Cattle, sheep and goats, in that order, were ranked as the most important livestock species and were mainly kept for meat, cash and ceremonies, respectively. The non-descript cattle breed was the most common breed found in the smallholder areas. Most farmers preferred Nguni breed because of its adaptive attributes. In the second experiment, structured questionnaires were administered, between June and August 2006, to a total of 551 farmers from 10 communities of the Eastern Cape. There was a significant association (P<0.05) between the use of pregnancy diagnoses and community. About 87 and 77 % of the interviewed farmers did not respond on the extent of pregnancy and calving rates in their herds. A higher proportion of farmers from Hekele (51%) and from Upper Mnxe (45.3%) communities reported low number of bulls as a major constraint to cow reproductive performance. Body condition and ovarian activity were measured in the sweet and sour veld types. Body condition score of animals was measured from March iii 2007 until January 2008 and ovarian activity of cows was performed by a veterinarian through rectal palpation in June, August and October 2007 and January 2008. From March to July, there was a marked decline in body condition on both veld types. In the sweetveld, body condition improved from September until January, whereas in the sourveld the improvement in body condition started in October. The cows in both veld types conceived throughout the year. Most cows in the sweetveld were cycling in January and August (P<0.05) whereas in the sourveld there was no distinct period when the animals were cycling. Overall, there were no differences in the proportion of cows that were cycling between the sour and sweet veldts (P>0.05). There were more cows cycling in sourveld in October than in the sweetveld. Reproductive performance of cows in communal areas could, therefore, be determined by levels and quality of nutrition. - Description: The broad objective of the study was to determine the socio-economic importance, health and welfare aspects of donkeys used for carting in a peri-urban area. A survey was conducted among 71 donkey owners in Joza, a peri-urban area near Grahamstown in the Eastern Cape Province (ECP) of South Africa to investigate their perceptions on socio-economic importance, health and welfare aspects of donkeys used for transport. Two hundred and seventy-one donkeys used for carting were assessed through direct observation of health and behaviour parameters to determine their welfare status. Results revealed that donkeys are indeed of immense value to people, being used daily (54.93%) for income generation activities and at least once per month (7.08%) for domestic chores. Most of the respondents (32%) earned R 600.00 per week while 11% earned at least R 200.00 per week from carting using donkeys. Donkeys were used for carting of goods (90.14%) more than for manure (16.9%). Gender and age of owners significantly (P < 0.05) influence perceptions on source of income while education level influenced their perceptions on carting firewood, goods and water. Results on management practices and health problems encountered by working donkeys showed that most of the owners managed their animals poorly. Most donkeys were being beaten (74.65%) during work, made to travel long distances (43.66%), worked long hours (52.11%) without adequate water (59.10%) and feed supplements (83.10%). Wounds (95.97%), coughing (75.65%) and lameness (64.79%) were some of the health problems encountered by donkeys. Significant relationships (P ˂ 0.05) were observed between owners’ perceptions on management practices and health problems encountered by donkeys. Among the observed animals, 61% were thin and mostly apathetic (26.2%) than medium and fat animals. Responses to observer approaches were significantly associated with sex and body condition score (BCS), with stallions in good body condition being more aggressive. Donkeys having a BCS of 3-5 avoided chin contact either by kicking out or moving away. Results further showed that most of the donkeys were suffering from external injuries, with the hindquarters (39.85%), shoulder (32.10%) and spine (21.77%) being the mostly affected areas. The prevalence of wounds on tail-base, belly, flank and neck were seen in less than 13% of the animals. The prevalence of wounds was influenced by age and BCS of the animals. Young donkeys with good BCS were less affected than old donkeys. Lameness, poor coat condition, external parasites, abnormal mucous membrane and dental problems were observed in less than 30% of the animals. It was concluded that donkeys play a pivotal role economically and socially to the livelihoods of people in a peri-urban area (Joza) in terms of income generation and transportation. However, these animals are experiencing multiple health and welfare problems. Therefore, more access to veterinary services, training on donkey use and management, health and welfare promotion programs are of paramount importance in solving the problem of poor health and welfare in donkeys.

- Description: Surveys were conducted to compare the seasonal tick prevalence and loads, and sero-prevalence of tick-borne diseases (TBD) in Nguni and non-descript cattle on the sweet and sour communal rangelands of the Eastern Cape Province. The tick species observed on both rangeland types were Rhipicephalus appendiculatus (71.0 %), Rhipicephalus (Boophilus) species (29.2 %) and Rhipicephalus evertsi evertsi (40.2 %). Hyalomma species (19.0 %) occurred only on the sour rangeland. Tick loads were higher (P < 0.05) in the hot-wet season than in the cool-dry season. Cattle in the sweet rangeland had significantly lower (P < 0.05) tick loads than those in the sour rangeland. Rhipicephalus appendiculatus loads were lower (P < 0.05) in the indigenous Nguni than non-descript cattle in the hot-wet and post-rainy season. Hyalomma species were also significantly lower (P < 0.05) in the Nguni than non-descript cattle in all the seasons. Three TBDs were observed, namely Babesia bovis (44.6 %), Babesia bigemina (45.9 %) and Anaplasma marginale (25.6 %). All the animals were sero-negative for Ehrlichia ruminantium. Nguni cattle had lower (P < 0.05) sero-prevalence for A. marginale in the cool-dry season and B. bigemina in the cool-dry and hot-wet seasons. Cattle in the sweet rangeland had significantly lower sero-prevalence of B. bovis and B. bigemina. Infection with B. bovis and A. marginale decreased (P < 0.05) the packed cell volume. Nguni cattle were recommended for use in the integrated control of ticks and TBD in the communal areas of South Africa as they were better able to cope with tick and TBD infestations than non-descript breeds.

Hyalomma species (19.0 %) occurred only on the sour rangeland. Tick loads were higher (P < 0.05) in the hot-wet season than in the cool-dry season. Cattle in the sweet rangeland had significantly lower (P < 0.05) tick loads than those in the sour rangeland. Rhipicephalus appendiculatus loads were lower (P < 0.05) in the indigenous Nguni than non-descript cattle in the hot-wet and post-rainy season. Hyalomma species were also significantly lower (P < 0.05) in the Nguni than non-descript cattle in all the seasons. Three TBDs were observed, namely Babesia bovis (44.6 %), Babesia bigemina (45.9 %) and Anaplasma marginale (25.6 %). All the animals were sero-negative for Ehrlichia ruminantium. Nguni cattle had lower (P < 0.05) sero-prevalence for A. marginale in the cool-dry season and B. bigemina in the cool-dry and hot-wet seasons. Cattle in the sweet rangeland had significantly lower sero-prevalence of B. bovis and B. bigemina. Infection with B. bovis and A. marginale decreased (P < 0.05) the packed cell volume. Nguni cattle were recommended for use in the integrated control of ticks and TBD in the communal areas of South Africa as they were better able to cope with tick and TBD infestations than non-descript breeds.

Meat quality attributes were determined from the cattle slaughtered for traditional ceremonies and from cattle slaughtered at the abattoirs. Data for questionnaires was summarized as frequencies for each ceremony and statistical differences were analysed using chi square test (X2). The effect of Age, breed and sex on beef pH, colour, meat tenderness, temperature and cooking loss of cattle slaughtered during traditional ceremonies, at low, medium and high throughput abattoirs were analyzed using General Linear Model (PROG GLM) of SAS (2003). There was a significant age effect (P < 0.05) on beef tenderness and cooking loss of cattle slaughtered for traditional ceremonies. Age of cattle that have milk teeth to 1 year and age of 6 years to 8 years had an effect on the beef tenderness (25.55 ± 8.05) and cooking loss (48.00 ± 4.26) of cattle slaughtered for traditional ceremonies in Table 4.1. There was no significant sex effect on beef tenderness and cooking loss of cattle slaughtered for traditional ceremonies. Non-descript and Angus cattle slaughtered at low throughput abattoir had an effect on the colour of the beef in Table 4.2. The lightness (27.49 ± 2.53) and yellowness (21.16 ± 0.79) of the beef shows that there was a significant breed effect on cattle that are slaughtered at Low throughput abattoir, however redness of the meat was not affected by breed after slaughter. Cattle that are slaughtered at high throughput abattoir between ages of 6 to 8 years and also cattle that are more than 8 years in Table 4.4 had an effect on cooking loss (p < 0.05). Age between 6 to 8 years had an effect on the beef tenderness from cattle that are slaughtered at medium throughput abattoir. It was concluded that the meat tenderness of the young cattle with milk teeth slaughtered for traditional ceremonies was affected. On the other hand the meat tenderness and cooking loss of beef from the cattle slaughtered from low, medium and high throughput abattoir were affected by sex and breed mostly.

- Description: Fleas are one of the major external parasites affecting free range chickens, causing irritation, tissue damage, blood loss and toxicosis which in turn affect quality and quantity of meat and egg production. There are many commercial insecticides available that are effective against fleas. These commercial insecticides have, however become expensive to most resourcelimited farmers and therefore unaffordable, causing farmers to seek low cost alternatives. This study was conducted to document, determine the existence of external parasites in freerange chickens and validate the alternative remedies used in controlling free-range chicken fleas by resource-limited farmers. A questionnaire survey was used to collect data on external parasites of free-range chickens and ethno-veterinary control remedies used by resourcelimited farmers to control these parasites at Amatola basin, in Eastern Cape, South Africa. Resource-limited farmers perceived that mites (79.6%), fleas (64.5%), lice (10.8%) and ticks (6.5%) were problematic parasites of chickens. To control these parasites, resource-limited farmers commonly use ash (28%) madubula (13% carbolic acid) (26.7%), Jeyes fluid (13% carbolic acid) (10%), paraffin (8.4%), used engine oil (2.8%), plants Tagetes minuta, Clutia pulchella, Calpurnia aurea (5.2%) and a few (4.2%) used commercial drugs Karbadust (Carbarly 5%) and mercaptothion 5%. About 7.5% of the respondents used neither of the remedies. To confirm the farmers’ perception on problematic external parasites of free-range chickens a diagnostic survey was conducted. Fifty free-range chickens were randomly selected and examined for external parasites. About 96% of the free-range chickens examined harboured at least one species of external parasites. Fleas (Echidnophaga gallinacea) (50.7%) were the most prevalent followed by lice, Menopon gallinae (12.4%); Menacanthus stramineus (5.3%) and Knemidocoptes mutans (0.57%). Given that Jeyes fluid, used engine oil, paraffin, C. aurea, C. pulchella and T. minuta were mainly used in controlling these parasites, it was important to determine the potential dermal irritant effects of these plants. A dermal irritant effect test was, therefore, conducted using a rat model. Forty eight rats, with parts of the backs shaven, were used to screen the materials for irritation. All the materials tested did not cause any visible skin irritation on rats (p>0.05). Regarding that ethnoveterinary materials were non irritant on rat skin in vitro repellency and contact bio-assay models were used to assess the insecticidal properties of Tagetes minuta, Calpurnia aurea, Clutia pulchella, used engine oil, paraffin and Jeyes fluid on fleas. For the in vitro bioassay T. minuta and Jeyes fluid at 100% concentration demonstrated a repellency level of 76 and 83%, respectively. Tagetes minuta was the most effective among plant materials (p<0.05). Clutia pulchella, C. aurea, used engine oil and paraffin showed insignificant repellency (p>0.05). For the contact bio-assay, C. pulchella, C. aurea and T. minuta at a concentration of 100% resulted in flea mortality of 83.5, 73.3 and 42.5%, respectively. The efficacy of Clutia pulchella compared well with that of Karbadust which had a mean mortality of 97.5%. Paraffin, used engine oil, and Jeyes fluid (19.2%) caused higher flea mortality of more than 82%. In the in vivo study 60 free-range chickens were artificially infested with fleas and test materials were topically applied on infested sites. Test materials exhibited varying flea load reduction efficacy. Used engine oil and Jeyes fluid at 76.8% concentration had a reduction efficacy of 100 and 96% after 3 days post application of test materials. These were not significantly different to that of the positive control Kabadust (carbaryl 5%). The plants C. pulchella and C aurea at 100% concentration had an efficacy of 92 and 77%, respectively. The commonly used remedies by resource-limited farmers to control fleas vary in efficacy. Some of the materials are as effective as the commercial insecticides hence they have a potential to be exploited as insecticides. Further investigations on plant compounds with insecticidal properties and their toxicity, need to be conducted before the plants are recommended for use.

- Description: The study was conducted to determine the effect of pre-slaughter stress of pigs on the levels of cortisol, creatine kinase (CK) and their subsequent relationship with pork quality. From our knowledge, there is paucity of information on transportation, lairage duration and sex effects on saliva cortisol and its relation with serum cortisol, urine cortisol and other meat quality traits. Stage one of the study assessed the effect of sex and time to slaughter (transport and lairage duration) on the levels of cortisol and CK in crossbred pigs. The second stage of the study evaluated the effect of sex and lairage duration on pork quality. The trials comprised of 60 22-weeks old commercial crossbred pigs [(30 boars vs. 30 gilts) with an average body weight of 76.9kg for gilts and 78.3kg for boars. Thirty pigs (15 Boars vs. 15 Gilts) were slaughtered on arrival after a 120km road journey and the other 30, after travelling the same road and distance, were slaughtered after having been in the lairage for 20 hours. Saliva samples were obtained during three time periods: between 07:00-10:00 on the day before the journey, on arrival at the abattoir and after lairage. Levels of cortisol in serum and urine and CK in serum samples collected at slaughter were determined. The samples (saliva and serum) were centrifuged at 20oC for 10 min at 3550 x g and stored at -20oC in separate vials until analysis. Pork quality was assessed using muscle pH, colour, thawing and cooking losses and Warner-Bratzler Shear Force. Correlations between cortisol levels in saliva, serum and urine and meat quality were assessed. The effect of sex and time to slaughter on saliva cortisol was significant. Statistical analysis showed significant interaction of sex by time to slaughter on serum cortisol. It was only the effect of sex that demonstrated higher (P <0.05) serum creatine kinase levels in gilts. Likewise, urine cortisol was influenced (P <0.05) only by time to slaughter. Saliva cortisol after transportation (r = 0.52) was correlated (P <0.05) with urine cortisol. The study suggests that there are greater responses of the Hypothalamic-pituitary-adrenal axis following time to slaughter (transport and lairage duration) between gilts and boars. This can be used in measuring cortisol in saliva, serum and urine, and serum creatine kinase to determine stress levels in pigs. Besides the lightness (L*) colour value which was significantly lower (P <0.05) in meat from gilts than from boars, the results of this study showed no interaction or individual effect (sex or lairage duration) on pH and other colour measurements. Meat cooking loss was affected (P <0.05) by gender x lairage interaction. Effect of lairage duration (as an individual effect) on the measured meat quality traits was not significant (P <0.05). Correlation analysis showed significant (P <0.05) negative relation between baseline saliva cortisol (r = -0.40) and the pork L* value. Saliva cortisol after lairage duration (r = -0.38 and r = 0.38) was correlated (P <0.05) with pH45 and pork a* value, respectively. Serum cortisol (r = -0.35) was negatively correlated (P <0.05) with pH45. Moreover, there were negative correlations (P <0.05) between saliva cortisol after transportation (r = -0.35), saliva cortisol after lairage duration (r = -0.44), serum cortisol (r = -0.40) and meat cooking loss. The study suggests that cortisol in saliva and serum can be used to determine effect of time to slaughter (transport and lairage duration) on meat quality traits from gilts and boars. - Description: The study was conducted to determine the effect of pre-slaughter stress of pigs on the levels of cortisol, creatine kinase (CK) and their subsequent relationship with pork quality. From our knowledge, there is paucity of information on transportation, lairage duration and sex effects on saliva cortisol and its relation with serum cortisol, urine cortisol and other meat quality traits. Stage one of the study assessed the effect of sex and time to slaughter (transport and lairage duration) on the levels of cortisol and CK in crossbred pigs. The second stage of the study evaluated the effect of sex and lairage duration on pork quality. The trials comprised of 60 22-weeks old commercial crossbred pigs [(30 boars vs. 30 gilts) with an average body weight of 76.9kg for gilts and 78.3kg for boars. Thirty pigs (15 Boars vs. 15 Gilts) were slaughtered on arrival after a 120km road journey and the other 30, after travelling the same road and distance, were slaughtered after having been in the lairage for 20 hours. Saliva samples were obtained during three time periods: between 07:00-10:00 on the day before the journey, on arrival at the abattoir and after lairage. Levels of cortisol in serum and urine and CK in serum samples collected at slaughter were determined. The samples (saliva and serum) were centrifuged at 20oC for 10 min at 3550 x g and stored at -20oC in separate vials until analysis. Pork quality was assessed using muscle pH, colour, thawing and cooking losses and Warner-Bratzler Shear Force. Correlations between cortisol levels in saliva, serum and urine and meat quality were assessed. The effect of sex and time to slaughter on saliva cortisol was significant. Statistical analysis showed significant interaction of sex by time to slaughter on serum cortisol. It was only the effect of sex that demonstrated higher (P <0.05) serum creatine kinase levels in gilts. Likewise, urine cortisol was influenced (P <0.05) only by time to slaughter. Saliva cortisol after transportation (r = 0.52) was correlated (P <0.05) with urine cortisol. The study suggests that there are greater responses of the Hypothalamic-pituitary-adrenal axis following time to slaughter (transport and lairage duration) between gilts and boars. This can be used in measuring cortisol in saliva, serum and urine, and serum creatine kinase to determine stress levels in pigs. Besides the lightness (L*) colour value which was significantly lower (P <0.05) in meat from gilts than from boars, the results of this study showed no interaction or individual effect (sex or lairage duration) on pH and other colour measurements. Meat cooking loss was affected (P <0.05) by gender x lairage interaction. Effect of lairage duration (as an individual effect) on the measured meat quality traits was not significant (P <0.05). Correlation analysis showed significant (P <0.05) negative relation between baseline saliva cortisol (r = -0.40) and the pork L* value. Saliva cortisol after lairage duration (r = -0.38 and r = 0.38) was correlated (P <0.05) with pH45 and pork a* value, respectively. Serum cortisol (r = -0.35) was negatively correlated (P <0.05) with pH45. Moreover, there were negative correlations (P <0.05) between saliva cortisol after transportation (r = -0.35), saliva cortisol after lairage duration (r = -0.44), serum cortisol (r = -0.40) and meat cooking loss. The study suggests that cortisol in saliva and serum can be used to determine effect of time to slaughter (transport and lairage duration) on meat quality traits from gilts and boars. Superovulation of donors involved insertion of a controlled internal drug release device (CIDR) and two injections of FSH daily 12 hours apart for 4 days on a decreasing dosage. Fresh Nguni semen was collected from proven Nguni stud bulls and assessed by computer aided sperm analysis (CASA) before artificial insemination (AI). The doses of AI were prepared and conducted twice, 12 hours apart on synchronized and superovulated Nguni cows or heifers. Embryos were flushed 7 days after AI using a non-surgical technique. Embryos were immediately evaluated under stereo microscope and classified according IETS standard codes (C1, C1- and C2). All transferrable embryos were vitrified. Two pilot study sites were chosen in Eastern Cape – Great kei; (n= 5) and Limpopo - Vuvha; (n=5) provinces for Embryo transfer. Each recipient cow was implanted with one frozen-thawed embryo. For oestrous synchronization, cows (Braman, Bonsmara and Nguni type) were selected in different villages, regardless of parity, age, breed and body weight following pregnancy diagnosis. Cows were grouped according to breed type and body condition scores (BCS) on a scale of 1-5. Group 1 had BCS of ≤ 2.5 in KwaZulu-Natal (n=81) and Limpopo n=71), Group 2 had BCS of ≥ 3 in KwaZulu-Natal (n=79) and Limpopo (n=100) cows. Cows were synchronized by inserting the controlled intravaginal drug release (CIDR) and removed on Day 8, followed by administration of prostaglandin. The white heat mount detectors (HMD) were placed on the individual cow’s tail head as an indicator for oestrous response if colour changed to red and inseminated twice at 12 hours interval. Pregnancy diagnosis was performed by an ultra sound scanner and rectal palpation 90 days after TAI in embryos and semen recipient cows. There was no significant difference on the superovulatory response rate between Nguni cows (40%) and heifers (40%). There was a significant difference on the ovary reaction (number of corpus luteum) of cows (11.33±1.41) and heifers (4.00±0.57). There were no significant differences observed on the embryo quality between Nguni cows (2.5±1.00 and 1.25±0.59) and heifers (0.83±0.41 and 1.00±0.36) for excellent (C1) and good (C1-). However, cows had more numbers of unfertilized ova (5.5±1.05 and 1.75±0.47) and degenerate embryos (3.66±1.00 and 1.25±0.39) than heifers. Village cows responded to oestrous synchronization successfully in KZN (100%) and Limpopo (99%) regardless of body conditions and breed type. The lowest pregnancy rate was recorded in Brahman and Bonsmara type cows with BCS of ≤ 2.5 regardless of Province. Interestingly, Nguni type cows with same body condition of ≤ 2.5 had higher average pregnancy rate of 59.5% in Limpopo and 53.5% in KZN. However, cows with BCS of ≥3 had better pregnancy rate regardless of cow breed type, and province. In conclusion, only 40% of both Nguni cows and heifers responded to superovulation. However, Nguni cows had better ovaries reaction compared to heifers. The quality of embryos recovered was similar for both Nguni cows and heifers. Moreover, there was a positive correlation between total sperm motility and fertilization rate bull 1 (93.7%) inseminated the cows (67.5%) and bull 2 (83.5%) inseminated the heifers (53.5%). Higher pregnancy rate (60%) was recorded in Limpopo compared to Eastern Cape (0%). Interestingly, more than 99% of village cows responded to synchronization and inseminated with frozen-thawed semen successfully. Village Nguni type cows were not affected by body condition scoring as they had higher and similar pregnancy rate as those that had body condition of ≥ 3. It is suggested that it is not advisable to breed synchronized Brahman and Bonsmara type cows with the body condition of ≤ 2.5 except in Nguni cow type as more than 57% average pregnancy rate was achieved.

Effect of dietary inclusion of Moringa oleifera Lam leaf meal on feed conversion efficiency, meat quality, fatty acid composition, shelf life and consumer health-related perceptions of pork

- Description: The objective of the study was to determine the feed conversion efficiency (FCE), carcass characteristics, physico-chemical quality, fatty acid (FA) composition and shelf life of pork from pigs fed diets containing either 0% (T1), 2.5% (T2), 5% (T3) or 7.5% (T4) Moringa oleifera leaf meal (MOLM). Consumer health-related perceptions on pork and fatty acids were also investigated. Twenty four crossbred Large White x Landrace pigs of both sexes at 18 weeks of age and initially weighing 71.6 kg on average were housed individually and had ad libitum access to one of the four dietary treatments for a period of six weeks. Average daily feed intake (ADFI) and average daily gain (ADG) and feed conversion ratios (FCR) were calculated as an indication of FCE and pigs were slaughtered at an average live weight of 99.6 kg. Carcass traits such as back fat thickness (BFT), carcass temperatures and pH readings taken 45 minutes and 24 hours (pH45 and pHu) post mortem were recorded. M. longissimus thoracis et lumborum (LTL) samples were taken from each carcass for the determination of lightness (L*), redness (a*), yellowness (b*), thawing loss percentage (TL%), cooking loss percentage (CL%), Warner Bratzler Shear Force (WBSF), shelf life and FA composition. Furthermore a survey was conducted amongst 80 University of Fort Hare Students to determine their health-related perceptions on pork and fatty acids as well as their pork consumption frequency using questionnaires. The FCE of pigs fed on 0-5% MOLM (T1, T2 and T3) diets did not differ significantly, but the FCE of pigs fed 7.5% MOLM (T4) was significantly (p<0.05) reduced. No significant relationship was reported between inclusion of MOLM carcass characteristics and physico-chemical pork quality. There was however a significant improvement (p<0.05) in the shelf life of the pork from MOLM fed pigs in terms of colour and odour during 10 days of refrigerated storage (at 3±1°C). There was a significant (p<0.05) reduction in the total intramuscular fat (IMF) content and the saturated fatty acid (SFA) C18:0 (stearic acid) content and an overall non-significant (p>0.05) increase in the poly unsaturated fatty acid (PUFA) content of pork from MOLM fed pigs. The possible reason for this is that the feeding of MOLM was commenced when the pigs were at an advanced age and weight. The survey revealed that the majority of interviewed students consume fresh (39.7%) and processed (32.4%) pork two to three times a week, perceive pork to be generally healthy (70%) and the second most healthy meat type (39.5%). While the majority (55.4%) were aware of the health implications of FAs they did not know which FA classes pose more of a health risk (51.3%). In conclusion2.5% and 5% of MOLM in finisher pig feed did not negatively affect FCE, carcass characteristics or physico-chemical meat quality; significantly improved pork shelf life and reduced total IMF and SFA content of pork but 7.5% MOLM negatively affected FCE; and the majority of students perceive pork to be the second most healthy type of meat and are generally aware of the health implications of FAs.

- Description: Characterization of village chicken phealth and management practices in the Amatola Basin of the Eastern Cape Province, South Africa By Njongenhle M. B. NYONI The majority of rural households in South Africa own village chickens which contribute significantly to their livelihoods. However, limited research has been conducted to characterize, understand and develop village chicken production in South Africa. This hinders the designing and implementation of village chicken-based developmental programmes that will benefit farmers in rural areas. Thus, it is imperative to initiate studies to describe village chicken production. This study was conducted to characterize village chicken production in Amatola Basin of the Eastern Cape Province of South Africa. Baseline information was gathered using a questionnaire survey of 81 identified from 7 out of 13 villages using snowball sampling technique. Consequently, 20 households were selected to participate in a monitoring study (from July 2010 to June 2011) through purposive sampling. Most (60%) chicken flocks were owned by women and mainly raised to meet household food requirements. Some (28.4%) farmers occasionally sold cocks and hens to neighbours, at an average price of R50 (USD7.55) per bird, as a way of culling their flocks and generating income. However, village chickens were mainly kept for food security rather than for their terminal benefit of cash. Although, all chicken flocks were produced under the scavenging feeding system, most (96%) flocks were provided with supplementary feed and drinking water. A majority (93.8%) of households also provided some form of shelter for their chickens. The mean cock to hen to chick ratio was 1:5:15. On average, iii each household owned 17 (±2 S.E.M.) chickens, and hens had 3.3 clutches per annum. Chicks hatched and chickens received as gifts represented the flock entries, while exits included mortality, consumption, sales and gifts or donations, in that order. Generally, both exits and entries were more pronounced in the summer season than in other seasons. Most (81.5%) farmers experienced chicken losses due to predation and health related problems. Parasitism ranked high amongst the village chicken health challenges. During monitoring, the endoprasites identified included nematodes, cestodes and protozoa. Coccidia were the most prevalent endoparasites. The chickens were also infested with a variety of ectoparasites namely: mites, lice, fleas and ticks. The sticktight flea Echidnophaga gallinacean was the most prevalent species. Although ectoparasite infestations were most pronounced in summer, almost all chickens harbored one or more of these parasites throughout the study period. The prevalence and intensity of parasite infestations were significantly (P<0.05) higher in the summer season compared to the other seasons. This was followed by the autumn, spring and winter seasons, respectively. The majority (77.5%) of farmers resorted to alternative remedies, mostly medicinal plants, for treating diseases and controlling parasites. Medicinal plants were used because they were locally available, cheap and perceived to be effective. Therefore, village chicken production in the rural Eastern Cape Province is characterised by several challenges which counter the significant role chickens play in the livelihood of rural households.

However, limited research has been conducted to characterize, understand and develop village chicken production in South Africa. This hinders the designing and implementation of village chicken-based developmental programmes that will benefit farmers in rural areas. Thus, it is imperative to initiate studies to describe village chicken production. This study was conducted to characterize village chicken production in Amatola Basin of the Eastern Cape Province of South Africa. Baseline information was gathered using a questionnaire survey of 81 identified from 7 out of 13 villages using snowball sampling technique. Consequently, 20 households were selected to participate in a monitoring study (from July 2010 to June 2011) through purposive sampling. Most (60%) chicken flocks were owned by women and mainly raised to meet household food requirements. Some (28.4%) farmers occasionally sold cocks and hens to neighbours, at an average price of R50 (USD7.55) per bird, as a way of culling their flocks and generating income. However, village chickens were mainly kept for food security rather than for their terminal benefit of cash. Although, all chicken flocks were produced under the scavenging feeding system, most (96%) flocks were provided with supplementary feed and drinking water. A majority (93.8%) of households also provided some form of shelter for their chickens. The mean cock to hen to chick ratio was 1:5:15. On average, iii each household owned 17 (±2 S.E.M.) chickens, and hens had 3.3 clutches per annum. Chicks hatched and chickens received as gifts represented the flock entries, while exits included mortality, consumption, sales and gifts or donations, in that order. Generally, both exits and entries were more pronounced in the summer season than in other seasons. Most (81.5%) farmers experienced chicken losses due to predation and health related problems. Parasitism ranked high amongst the village chicken health challenges. During monitoring, the endoprasites identified included nematodes, cestodes and protozoa. Coccidia were the most prevalent endoparasites. The chickens were also infested with a variety of ectoparasites namely: mites, lice, fleas and ticks. The sticktight flea Echidnophaga gallinacean was the most prevalent species. Although ectoparasite infestations were most pronounced in summer, almost all chickens harbored one or more of these parasites throughout the study period. The prevalence and intensity of parasite infestations were significantly (P<0.05) higher in the summer season compared to the other seasons. This was followed by the autumn, spring and winter seasons, respectively. The majority (77.5%) of farmers resorted to alternative remedies, mostly medicinal plants, for treating diseases and controlling parasites. Medicinal plants were used because they were locally available, cheap and perceived to be effective. Therefore, village chicken production in the rural Eastern Cape Province is characterised by several challenges which counter the significant role chickens play in the livelihood of rural households.

Gall sickness, heart water and footrot caused most of the sheep mortalities. Dohne Merinos were the common genotypes in the two villages. Total entrances for each flock were higher (p < 0.05) in hot-dry season and hot-wet season than in other seasons. Most of the entrances were lambs and were born in hot-dry season (September) and cool-dry season (June) for larger flocks (10.90 ± 3.02) and for small flocks (3.65 ± 3.02). High lamb mortalities were experienced in the post-rainy (April) and hot-wet (December) season. There was a significant interaction between season and flock size. Most of the sales occurred in the hot-wet season. Ecological area had significant effect on sheep production potential (p < 0.05) in both flock classes. The average sheep production efficiency (SPE) value for Gaga and Sompondo were 0.50 ± 0.116 and 0.50 ± 0.096 respectively. The SPE for large flock was higher (p < 0.05) by season and flock size. - Description: The objective of the study was to determine growth performance, physico-chemical attributes, oxidative stability and sensory quality of pork from pigs fed one of three dietary treatments, eac containing 0%, 2.5% or 5% (T1, T2 and T3, respectively) Moringa oleifera leaf meal (MOLM). Dietary treatments were formulated to be isonitrogenous and isoenergetic for weaner (6 – 8 weeks) and grower (9 – 13 weeks) phases. Twelve Large White (LW) and 12 Kolbroek (KB) male pigs at 6 weeks of age, initially weighing an average 10 kg, were randomly allocated to one of the dietary treatments, each with four replicates, in individual pens. Feed and water were offered ad libitum. The Longissimus thoracis et lumborum (LTL) muscle of each carcass (n = 24) was sampled for meat quality and fatty acid analyses. Breed differences in measured parameters were observed. In pigs receiving 5% MOLM, the LW had significantly the highest ADFI (P<0.05) than KB pigs. In pigs receiving 2.5% and 5% MOLM, backfat thickness was significantly highest (P<0.05) compared to those receiving 0% MOML. Dietary inclusion of MOLM had no significant effects (P>0.05) on the physico-chemical quality of pork from LW pigs; although it significantly (P<0.05) increased a* and reduced WBSF values in pork from KB pigs. Pork from LW pigs receiving 5% MOLM had significantly increased (P<0.05) n-3 content of the subcutaneous tissue of LW pigs compared to KB, and also when compread to other treatment groups. In both breeds, the composition of PUFA: SFA and n-6: n-3 in the subcutaneous adipose tissue was significantly (P<0.05) lower in T2 and T3 than in T1. The n-3 levels for pork muscle from LW pigs receiving 2.5% and 5% MOLM, and from KB pigs receiving 5% MOLM, were significantly (P<0.05) lower. Inclusions of MOLM significantly (P<0.05) reduced the n-6: n-3 fatty acids in pork from both LW pigs and KB pigs. Consumer scores on sensory attributes (aroma intensity, initial impression of juiciness, first bite, sustained impression of juiciness, muscle fibre and overall tenderness and overall flavor intensity) significantly (P<0.05) increased as the level of MOLM inclusion in the diet increased; and higher scores were observed in fried meat than in boiled for most sensory attributes. It may be concluded that inclusion of MOLM in pig diets up to 5% improved the ADFI, a*, tenderness of pork without adversely affecting the FCR and other physico-chemical quality attributes; and resulted in desired increase in levels of n-3 and reduced the n-6: n-3 fatty acid ratio.

Glutathione (GSH), catalase (CAT) and superoxide dismutase (SOD) and lipid peroxidation (thiobarbituric acid reactive substances, TBARS) were taken as parameters to evaluate in vivo AA of the meat. For the second experiment, twelve male broilers were used in four groups of dietary supplementation. Three broilers were allocated per group. After slaughter, the breast muscle was sampled for total phenols, flavonoids, proanthocyanidins, TPC, ABTS, GSH, CAT, SOD, lipid peroxidation, ultimate pH (pH24), colour (L* (lightness), a* (redness), b* (yellowness)), WB-shear force and cooking loss. In both chapters Moringa leaves exhibited the highest AA. Meat from goats supplement with Moringa exhibited the highest AA. Meat from broilers supplemented with a mixture of Moringa leaves, broiler finisher and crushed maize, had the highest (P < 0.05) pH24 and L* values. Moringa can therefore be used to preserve meat without changing the quality of meat.
