- Description: CD23, the low affinity IgE receptor, is expressed by various cell types and has numerous functions depending on the form of the protein, its interaction with various ligands and the type of cell involved. CD23 is pivotal in the regulation of IgE, with the soluble form involved in up-regulation, while the membrane bound form is involved in the down-regulation. It is clear why it is believed to be a central molecule in allergic responses, and a therapeutic target for the treatment of allergic disease. In this study a recombinant form of the entire extracellular domain of the protein, exCD23, was produced by PCR cloning and expressed in E. coli. His•Tag™s were introduced onto the C-terminus and N-terminus, respectively, in order to simplify the purification procedure. After renaturation and purification, the recombinant exCD23 bound IgE, indicating its activity. From the IgE binding studies it was established that the position of the tag did not influence the binding. GST•Tagged™ exCD23 was also produced in an attempt to increase the solubility of the recombinant protein, but this proved unsuccessful. Butyrate differentiated EoL-1 cells were treated with the Nterminal His•Tagged™ exCD23, and the protein appeared to suppress the secretion of the constitutively expressed cytokines, especially IL-8 and IFN- , when compared to untreated cells. In addition, treatment of the EoL-1 cells with exCD23 had a significant proliferative effect, but could not induce differentiation of this cell line into mature eosinophilic-like cells.

- Description: This study looks at the impact of elephant feeding on the Xeric Succulent Thicket component of Eastern Cape Subtropical Thicket (ECST) in Addo Elephant National Park (AENP). Observations of elephant feeding were carried out and vegetation transects were surveyed for impact of elephant feeding. The results indicated that the Nyati elephants spent the majority of their time grazing (nearly 90%), particularly the cow-young herds, and especially when the herd gathered in larger numbers. Browsing events were concentrated on Acacia karroo (81%) and there was no significant difference between the sexes in their preference for this species. Despite being subjected to most of the browsing, the majority of A. karroo trees were undamaged and the effect of elephants was generally light. It appears unlikely that, three years after re-introduction to Nyati, the elephants have had an effect on community structure of the vegetation. Surveys were conducted on stands of the alien invasive weed prickly pear Opuntia ficus-indica, and it was recorded that elephants in Nyati have had a dramatic effect on prickly pear, utilising all adult plants assessed and destroying 70% of them. This level of destruction in such a short period of time suggests that prickly pear is a highly favoured species. The results from the present study suggest that elephants can play a role in the control of prickly pear. Results are discussed in terms of elephants as both megaherbivores and keystone species, and as agents of intermediate disturbance.

Rehabilitation of the Orange River Mouth Salt Marsh : seed, wind and sediment characteristics

- Description: The Orange River is an important source of freshwater and like many other wetlands in semi-arid regions, supports various social (Spurgeon, 1998), economic (Spurgeon, 1998; Bornman et al., 2005) and ecological functions. The saltmarsh at the Orange River Mouth has become degraded over time following numerous anthropogenic impacts. As a result the Transboundary RAMSAR site was placed on the Montreux record emphasising the importance for rehabilitation. The potential of the marsh for natural rehabilitation was assessed through three physical factors which were considered to have the most influence on the saltmarsh i.e. 1) sediment 2) water 3) wind. Three sampling areas were chosen to investigate the sediment characteristics of the ORM saltmarsh and the suitability for seed germination and adult survival. Site A was representative of the general marsh area, Site B was thought to have favourable sediment conditions for saltmarsh growth because of the large numbers of seedlings and Site C was prone to inundation by wind blown sediment. The sites were sampled in 2005 (dry conditions) and in 2006 after high rainfall and river flooding. Electrical conductivity (EC) of the sediment throughout the marsh was hypersaline in many instances above the tolerance range for S. pillansii (> 80 mS.cm-1) The freshwater event in 2006 lowered salinity significantly in two of the three sites. Differences in sediment characteristics were also compared for three habitats i.e. driftlines, open sites and under vegetation. Driftlines (C. coronopifolia = 872 seedlings m-2; S. pillansii = 1296 seedlings m-2) and the microhabitat associated with adult plants (C. coronopifolia = 803 seedlings m-2; S. pillansii = 721 seedlings m-2) created favourable conditions for seedling growth, however open unvegetated (C. coronopifolia = 56 seedlings m-2; S. pillansii = 49 seedlings m-2) areas had significantly lower seedling density. Due to the marsh currently being in a desertified state this study aimed to establish whether the remaining vegetation could produce enough seed to revegetate the marsh. Laboratory studies indicated that seeds of both species germinated best in freshwater (0 psu). The germination of S. pillansii seeds was 40 percent at 0 psu compared to 5 percent at 35 psu. After storage under hypersaline conditions (35 psu) C. coronopifolia showed 100 percent seed germination when returned to freshwater whereas storage at 70 psu decreased the viability of S. pillansii seeds. The plants are producing adequate seed that will allow for regrowth and rehabilitation if sediment and groundwater characteristics are suitable for seed germination, seedling growth and adult survival. However the increase in bare areas at the Orange River mouth as a result of salt marsh dieback has increased the available sediment source. The wind blown sediment has covered large areas of the remaining adult salt marsh vegetation, particularly in the northern corner at Site C, causing further die-back.

- Description: Geographic Information System technology today allows for the rapid analysis of vast amounts of spatial and non-spatial data. The power of a GIS can only be effected with the rapid collection of accurate input data. This is particularly true in the case of the South African National Working for Water (WFW) Programme where large volumes of spatial data on alien vegetation infestations are captured throughout the country. Alien vegetation clearing contracts cannot be generated, for WFW, without this data, so that the accurate capture of such data is crucial to the success of the programme. Mapping Invasive Alien Plant (IAP) data within WFW is a perennial problem (Coetzee, pers com, 2002), because not enough mapping is being done to meet the annual requirements of the programme in the various provinces. This is re-iterated by Richardson, 2004, who states that there is a shortage of accurate data on IAP abundance in South Africa. Therefore there is a need to investigate alternate methods of data capture; such as remote sensing, whilst working within the existing WFW data capture standards. The aim of this research was to investigate the use of Landsat ETM imagery as a data capture source for mapping alien vegetation for the WFW Programme in terms of their approved mapping methods, for both automated and manual classification techniques. The automated and manual classification results were compared to control data captured by differential Global Positioning Systems (DGPS). The research tested the various methods of data capture using Landsat ETM images over a range of study sites of varying complexity: a simple grassland area, a medium complexity grassy fynbos site and a complicated indigenous forest site. An important component of the research was to develop a mapping (classification) Ranking System based upon variables identified by WFW as fundamental in data capture decision making: spatial and positional accuracy, time constraints and cost constraints for three typical alien invaded areas. The mapping Ranking System compared the results of the various mapping methods for each factor for the study sites against each other. This provided an indication of which mapping method is the most efficient or suitable for a particular area.

Hydrogenases from sulphate reducing bacteria and their role in the bioremediation of textile effluent

- Description: The continuing industrial development has led to a corresponding increase in the amount of waste water generation leading to a consequential decline in levels and quality of the natural water in the ecosystem. Textile industries consume over 7 x 10[superscript 5] tons of dyes annually and use up to 1 litre of water per kg of dye processed and are third largest polluters in the world, the problem being aggravated by the inefficiencies of the dye houses. An abundance of physio-chemical methods are in use world wide, however, there is increasing concern as to their impact in effectively treating textile effluents as they introduce secondary pollutants during the ‘remediation’ process which are quite costly to run, maintain and clean up. Research on biological treatment has offered simple and cost effective ways of bioremediating textile effluents. While aerobic treatment of textile dyes and their effluents has been reported, its major draw back is commercial up-scaling and as such anaerobic systems have been investigated and shown to degrade azo dyes, which form the bulk of the dyes used world wide. However, the mechanisms involved in the bioremediation of these dyes are poorly understood. The aims of this study were to identify and investigate the role of enzymes produced by sulphate reducing bacteria (SRB) in bioremediating textile dye and their effluents. Sulphate reducing bacteria were used in this study because they are tolerant to harsh environmental conditions and inhibit the proliferance of pathogenic micro-organisms. The appearance of clear zones in agar plates containing azo dye concentrations ranging from 10 – 100 mgl[superscript -1] showed the ability of SRB to decolourize dyes under anaerobic conditions. Assays of enzymes previously reported to decolourise azo dyes were not successful, but led to the identification of hydrogenase enzyme being produced by SRB. The enzyme was found to be localised in the membrane and cytoplasm. A surface response method was used to optimize the extraction of the enzyme from the bacterial cells resulting in approximately 3 fold increase in hydrogenase activity. Maximum hydrogenase activity was found to occur after six days in the absence of dyes but was found to occur after one day in the presence of azo dyes. A decline in hydrogenase activity thereafter, suggested inhibition of enzymatic activity by the putative aromatic amines produced after azo cleavage. Purification of the hydrogenase by freeze drying, poly ethylene glycol, and Sephacryl – 200 size exclusion- ion exchange chromatography revealed the enzyme to have a molecular weight of 38.5 kDa when analyzed by a 12 % SDS-PAGE. Characterisation of the enzyme revealed optimal activity at a pH of 7.5 and temperature of 40 °C while it exhibited a poor thermal stability with a half-life of 32 minutes. The kinetic parameters V[subscript max] and K[subscript m] were 21.18 U ml[superscript -1} and 4.57 mM respectively. Application of the cell free extract on commercial dyes was not successful, and only whole SRB cells resulted in decolourisation of the dyes. Consequently trials on the industrial dyes and effluents were carried out with whole cells. Decolourisation rates of up to 96 % were achieved for the commercial dyes and up to 93 % for the industrial dyes over a period of 10 days.

Assays of enzymes previously reported to decolourise azo dyes were not successful, but led to the identification of hydrogenase enzyme being produced by SRB. The enzyme was found to be localised in the membrane and cytoplasm. A surface response method was used to optimize the extraction of the enzyme from the bacterial cells resulting in approximately 3 fold increase in hydrogenase activity. Maximum hydrogenase activity was found to occur after six days in the absence of dyes but was found to occur after one day in the presence of azo dyes. A decline in hydrogenase activity thereafter, suggested inhibition of enzymatic activity by the putative aromatic amines produced after azo cleavage. Purification of the hydrogenase by freeze drying, poly ethylene glycol, and Sephacryl – 200 size exclusion- ion exchange chromatography revealed the enzyme to have a molecular weight of 38.5 kDa when analyzed by a 12 % SDS-PAGE. Characterisation of the enzyme revealed optimal activity at a pH of 7.5 and temperature of 40 °C while it exhibited a poor thermal stability with a half-life of 32 minutes. The kinetic parameters V[subscript max] and K[subscript m] were 21.18 U ml[superscript -1} and 4.57 mM respectively. Application of the cell free extract on commercial dyes was not successful, and only whole SRB cells resulted in decolourisation of the dyes. Consequently trials on the industrial dyes and effluents were carried out with whole cells. Decolourisation rates of up to 96 % were achieved for the commercial dyes and up to 93 % for the industrial dyes over a period of 10 days.

The results show that over the 40 years since the enclosure was built, changes have occurred which have resulted in a significant difference in the abundance of Increaser 2 and Decreaser grass species between the inside and the outside of the enclosure. Increaser 2 and Decreaser categories are the result of a grass species classification depending on whether the species thrives or is depressed by heavy grazing. The difference in grass species composition and structure between the inside and the outside of the enclosure indicates that the grazing animals within the KNP have influenced the grass composition in a way that favours the dominant animals. This has resulted in a declining roan antelope population - one of the species that is considered as a 'rare antelope'. Many artificial waterpoints (boreholes and dams) have also been closed throughout the KNP in the hope of resulting in a change in vegetation structure and composition in favour of the roan. Veld condition assessment data for 87 boreholes throughout the Park was analyzed to determine whether the veld in the vicinity is beginning to change towards a more Decreaser dominated sward which would favour the roan. The results were analyzed for the different regions of the Park; and they indicate that changes are becoming evident; however, the results are not particularly conclusive, yet. The majority of the boreholes were closed between 1994 and 1998 which means that not a lot of data were available to be analyzed. A similar study conducted in another 10 years time might reveal more meaningful results. However the results are moving in the direction hoped for by the management of the KNP. The results show that the grass composition has a higher proportion of Decreaser grasses since the closure of the waterpoints, and the grass biomass around these areas has also improved. The results were analyzed on an individual basis; and then on a regional basis as the minimal data meant that the individual analyses did not provide any significant results. A third study was then done on the impact that the rapidly increasing elephant population on the vegetation within the Riparian zone along three rivers in the Far Northern region of the KNP. The riparian zone is an important part of the landscape, in terms of providing food for many animals as well as shade. The elephant population has increased substantially since the termination of the culling program and this means that the feeding requirements of the population has increased which could result in severe damage upon the vegetation, as elephants can be very destructive feeders. The results show surprising differences between the three years of data that were analyzed; however the results indicate that the elephants are targeting specific height ranges of trees when feeding; however they do not seem to consistently target specific tree species. This is positive for the diversity of the Riparian zone as this region is very important both ecologically and aesthetically for the tourists who visit the Park.

- Description: Aquatic toxicity tests are increasingly being used in water resource management worldwide, and currently in South Africa, policy and legislation are being drafted to reflect this international trend. While standard toxicity test methods and test organisms are being considered to develop and set water quality guidelines and effluent discharge limits, it is not clear whether guidelines and discharge limits set using these standard test organisms will be sufficient to protect South Africa’s scarce water resources. As part of ongoing research to investigate the use of indigenous riverine organisms as toxicity test organisms a number of potential species have been identified, including the freshwater shrimp Caridina nilotica. For much of the history of aquatic toxicological data the bulk of the data has been generated by acute toxicity testing, based on short exposures and using mortality as the response end point. There are relatively few chronic, longterm tests with sub-lethal endpoints. However, it was recognized that information about longer exposure durations and non lethal response endpoints was needed, instead of mortality. Chronic tests can provide a more environmentally realistic measure of chemical toxicity than acute toxicity tests. Caridina nilotica has been identified as a potential standard toxicity test organism, as it is widely distributed, easy to find and it occurs in flowing waters. It is an indigenous species which can be easily cultured and maintained in the laboratory and is also ecologically important. Both adults and juveniles have been used successfully in acute toxicity tests at the Institute for Water Research (Rhodes University) and the ability to rear the organisms under laboratory conditions has allowed the development of chronic toxicity tests using C. nilotica. Chronic early life stage tests include continuous exposure of the early life stages, which are presumed to be the most sensitive for aquatic organisms. This study reports on the embryonic development of C. nilotica at the culture temperature of 24⁰C. Morphological developmental stages were monitored and measured and 7 developmental stages were identified. Based on the measurements of the features that were identified, toxicity tests using the reference chemicals sodium chloride (NaCl), sodium sulphate (Na₂SO₄) and cadmium chloride (CdCl₂) were undertaken to test the suitability of C. nilotica embryonic development for chronic toxicity tests for use in water resource management. The length, width, length:width ratios and area of the features decreased in size when exposed to the chemicals. The Lowest Observed Effect Concentration (LOEC) values were 2000mg/L for Na₂SO₄, 3000mg/L for NaCl and 0.31mg/L for CdCl₂. The No Observed Effect Concentration (NOEC) values were 1000mg/L for Na₂SO₄, 2000mg/L for NaCl and <0.31mg/L for CdCl₂. Further research on the teratogenic effects of single chemicals and industrial effluent on developing C. nilotica embryos needs to be undertaken in order to evaluate the described test protocol for use in water resource management.

Mammals, such as aardvarks (Orycteropus afer, Pallas 1766), excavate burrows in order to avoid predators and climatic extremes and are termed “ecosystem engineers” as they physically modify their environment and in doing so create new habitats and alter the availability of resources to other species. In this study I assessed the microhabitat conditions (maximum and minimum temperature, relative humidity and seed abundance) of aardvark burrows in relation to paired control sites. In addition, I evaluated the use of aardvark burrows by other vertebrate and invertebrate species and investigated the impact of aardvark burrow mounds on landscape scale floristic diversity. Maximum temperatures were significantly lower (p < 0.05) and minimum temperatures and midday humidity were significantly higher (p < 0.05) inside the burrows at the three study sites, Kwandwe Private Game Reserve (Kwandwe), Mountain Zebra National Park (MZNP) and Tswalu Kalahari Reserve (Tswalu). There were no significant differences between the concentration of seeds, the average numbers of unique individual small mammals, trap success or small mammal species richness recorded inside the burrows compared to outside (p > 0.05). At all three sites, small mammal species diversity was higher in the burrows but this result was also not significant (p > 0.05 for all). Trap success and the number of individuals captured was higher at Tswalu than the other two sites (p < 0.05 for both). The different methods used in this study revealed a total of 25 mammal, seven bird, one amphibian and six reptile species utilising aardvark burrows. There were significant differences in insect community assemblages between the burrows and open control areas at Kwandwe and Tswalu (p < 0.05 for both) but not at MZNP (p > 0.05). The parasitic guild was more prominent inside the burrows than outside but their abundance was not as high as anticipated, possibly due to the placement of traps closer to the burrow entrances than the sleeping chambers. The complex structure of the burrows prevented the placement of traps in close proximity to the sleeping chambers. As expected, the amount of bare earth was significantly higher on active and recently abandoned burrow mounds compared to the old burrow mounds and reference plots at all three sites (p < 0.05 for all), with the exception of the active burrows at Tswalu. Overall, the different plot types were characterised by significantly different plant communities during all the seasons at MZNP, during three of the seasons at Kwandwe and only during winter at Tswalu. The total species richness recorded on the reference plots was higher than on the burrow mounds at all three sites. However, species diversity on the reference plots was not significantly higher than the burrows at any of the sites (p > 0.05 for all sites). Although the results were not significant, the overall species diversity at a site level was greater than the reference patches at Kwandwe and Tswalu (p > 0.05 for both). Aardvarks fulfil the criteria of a significant ecosystem engineer and their presence in arid and semi-arid environments is likely to be critical to the survival of other individual organisms and species, particularly when alternative burrowing animals are either absent or restricted in their activities. Thus, aardvark populations should be considered a conservation priority in arid and semi-arid ecosystems.

- Description: This thesis presents a history of the landscape of Nqabara, an administrative area in a rural and coastal area of the Eastern Cape Province, South Africa. In the process of investigating landscape history, the inquiry engages with a range of data sources from diverging discursive contexts, including data from ethnographic fieldwork, from the consultation of archival documents and historical reports as well as from comparative historic and ethnographic research, necessitating a critical consideration of the epistemological contexts of data production and the dialogue between researcher and data. Furthermore, in its aim to move beyond historical description towards explanation, the study interrogates the dualist ontological conceptualisations of nature and culture, society and ecology, object and meaning upon which are built three dominant conceptual frameworks concerned with human-environment relationships: social-ecological systems theory, transdisciplinary landscape studies and political ecology. Drawing primarily upon the works of James Gibson, Anthony Giddens and Tim Ingold, an ontological foundation is developed to guide the enquiry and move towards an alternative understanding of the relationship of people’s livelihoods with respect to the landscape in which it is lived, which I call here the praxisembodiment perspective. This ontology takes the situated patterns of action of a situated agent-in-its-environment as its point of departure and proceeds to develop a framework explaining how relations among the patterns of action of different agents-in-their-environment, emerge in structures that simultaneously enable and constrain future action. The foundation is thereby provided for a monist understanding of how landscape and social structure emerge simultaneously from the complex intersection of patterns of actions and interactions of agents in their environment. This framework calls for an understanding of time, space and scale, not as independent variables influencing process and action, but as emergent properties arising from the patterns of actions of situated agents. Finally the alternative ontology is applied to the history of landscape and livelihoods of people of Nqabara. It is concluded that an appropriate understanding and explanation of the critical transformations in the landscape as well as in social institutions, should be sought through analysis of the complex ways in which patterns of action of multiple spatial and temporal rhythms and between multiple agents in an environment, intersect and resonate.

Synthesis and characterization of symmetrical and unsymmetrical ferrocenyl ligands for use in the preparation of Redox Active Ruthenium Alkylidene Complexes

A change in the standard redox potential of ferrocene (465 mV), to more positive potentials in vinylferrocene 1 (478 mV) and 4-phenylvinylferrocene 3 (499 mV), showed how manipulation of a redox potential can be effected on the ferrocenyl moiety by just using conjugation effects. A shift by +13 mV is observed in 1 and this potential more than doubled in 3 (+34 mV). Ferrocenylderived ruthenium alkylidene complexes were also prepared in a cross metathesis of 1 and 3 with Grubbs’ 1 (676.5 mV) to give complexes Ferrocenylidenebis( tricyclohexylphosphine)dichlororuthenium 14, 4-ferrocenylphenylidene-bis (tricyclohexylphosphine)dichlororuthenium 15 respectively. The extent of the electronic communication between the ferrocenyl group and the ruthenium centre was then estimated by looking at the positive or negative redox potential shifts of 14 and 15 as a result of 1 and 3. A large positive potential shift by 180 mV in 14 indicated that there was a strong electronic communication between the two metal centres, while the smaller, yet significant positive potential shift by 89.5 mV in 15 showed 3 to have a lesser effect on the ruthenium centre. Compounds 14 and 15 were tested in a Ring Closing Metathesis (RCM) of diethyldiallylmalonate showed enhanced reactivity.

- Description: The role of antioxidants in the prevention of neurodegenerative diseases has been well documented. The use of synthetic antioxidants has decreased due to the ssociation of these compounds with certain cancers. Thus, the search for novel natural antioxidants has gained much focus in research. Most common methods of determining antioxidant capacity are the radical generated assays and biological assays such as lipid peroxidation and the nitroblue tetrazolium assay. Electrochemical methods have been proposed for the determination of bio-active compounds such as antioxidants. The electrochemical methods of cyclic voltammetry and square wave voltammetry were evaluated for the determination of antioxidant capacity initially examining known antioxidants and then using plant extracts of Sutherlandia frutescens as a case study. The antioxidant properties determined by electrochemical methods were validated utilising the non-biological methods of the DPPH, TEAC, ferrozine and FC assay and biological pharmacological methods. The results indicated that Sutherlandia frutescens contains potent antioxidant compounds that are able to reduce lipid peroxidation. The electrochemical techniques of square wave voltammetry and cyclic voltammetry were applied for the screening of a large number of extracts of various algae for the detection of antioxidant compounds. The results indicated that electrochemistry can be used as a preliminary method for the rapid screening of a large number of crude samples for antioxidant compounds. Electrochemical methods were also evaluated as a method for guiding the isolation and purification of antioxidant metabolites in Sargassum elegans. Solvent partitioning and fractionation of the marine alga allowed for the purification of antioxidant compounds. At each step of purification electrochemical methods were utilized to determine which fractions contained the more potent antioxidant compounds and thus guide further purification. The purified antioxidant compounds were elucidated using NMR to determine the structure of the antioxidant compounds.

The design and assessment of an integrated municipal waste beneficiation facility : towards improved sewage sludge management in developing countries

- Description: This research aimed to firstly confirm the hypothesis that the current management of sewage sludge generated during the treatment of wastewater in the Eastern Cape Province of South Africa is poor and, as such, this material poses a threat to local environmental and human health. Secondly, through a rational design approach, the current research also aimed to design and assess the suitability of a novel integrated process incorporating appropriate technologies for improved sewage sludge management. The lack of an appropriate tool for the assessment of the integrated process necessitated the development of an appropriate technology assessment (TA) methodology based on environmental, technical, social and economic criteria. A combination of informal participatory methods such as semi-structured interviews and formal research methods including questionnaires, risk assessment exercises and laboratory analyses were used. Based on the above it was estimated that 116 tons dry sludge were generated in the province per day and that the concentration of heavy metals present in the sludge was generally within the limits for reuse on agricultural land. Furthermore the sludge from all sample sewage treatment works (STWs) was found to be free of any detectable pathogens. Despite the above, a preliminary risk assessment and chemical analysis revealed that existing sludge management practices at sample STWs posed a threat to the environment, particularly to water resources and exacerbated the problems associated with the discharge of poorly treated municipal wastewater. The root causes of the poor sludge management were considered to be a lack of non-regulatory incentives and financial resources. Highly integrated ecologically engineered systems were thought to offer a solution to the dual problem of poor sludge management and municipal effluent treatment, while providing necessary economic incentives. To facilitate the design of a system appropriate to local conditions, it was necessary to first develop a rational design methodology, which incorporated a detailed TA step. The result of the design process was an Integrated Waste Beneficiation Facility (IWBF) that incorporated a number of process units comprised of appropriate technologies including composting, vermicomposting, algal ponding technology and aquaculture. A detailed TA indicated that the benefits of the proposed IWBF would, at the majority of sample sites, outweigh the potential negative impacts and it was thus recommended that investigations should continue on pilot-scale facilities. Furthermore, although the proposed TA based on four sustainability criteria categories was thought to provide a more accurate assessment of the true sustainability of a technology, the acquisition of information was problematic highlighting the need to re-think current TA methodologies and to address associated constraints allowing the tool to be used and fully comprehensive.

- Description: Acquired Immunodeficiency Syndrome (AIDS) is a human disease caused by the human immunodeficiency virus type 1 (HIV-1) and it is one of the biggest social, economic and health challenges in the world. The Joint United Nations Programme on HIV/AIDS (UNAIDS) and World Health Organization (WHO) estimated that between 33.4 to 46.0 million people around the world were living with HIV/AIDS in December 2005 and the highest estimates are in the Sub-Saharan Africa (around 25 million). In more developed countries a combined antiretroviral therapy called highly active antiretroviral therapy (HAART) is used, which results in reduced progression to AIDS in most patients. Despite the beneficial effects of HAART, significant side effects are experienced by treated patients. In addition, most infected people live in countries where the treatment is very expensive or, in many cases, not available at all. These people therefore rely on medicinal plants for health care. In this study, aqueous extracts from Sutherlandia frutescens and Lobostemon trigonus were screened for potential anti-HIV activities in a series of in vitro enzymatic assays, including reverse transcriptase, HIV-1 protease and glycohydrolases. Two extracts of Sutherlandia leaves (SFL-1 and SFL-2) were prepared that inhibited HIV reverse transcriptase and a Lobostemon leaf extract (LTL) was shown to also inhibit this enzyme. All extracts were assayed at 1.25mg/ml. Tannin content was determined for all active extracts using a tannic acid assay. SFL-1 and SFL-2 were found to contain about 6 percent and 7 percent tannins, respectively, and LTL contained 31% tannins by weight. Tannins were removed using polyamide columns and three fractions were collected for each. The extracts were also fractionated with Sephadex G-25, Amberlite IR 120 and Dowex 1-X8 as size exclusion, cation exchange and anion exchange, respectively. Extracts were also fractionated by preparative thin layer chromatography where two compounds were separated from S. frutescens extract with high activity against reverse transcriptase while showing insignificant inhibition towards other enzymes tested. SFL-BFW-10 and SFL-WEF-7 inhibited reverse transcriptase by almost 100 percent and the IC50 values calculated for these compounds were 0.34 and 0.23mg/ml, respectively. Cytotoxicity of these compounds was evaluated on Chang liver cells and peripheral blood mononuclear cells (PBMCs). None of these compounds showed any significant inhibition of cell proliferation. The purity of these compounds could not be confirmed because there was insufficient material to use in the techniques required to show purity and identification. Therefore, TLC was used to determine the nature of these compounds. SFL-BFW-10 was identified as an organic acid and SFL-WEF-7 was identified as flavonoid.

The extracts were also fractionated with Sephadex G-25, Amberlite IR 120 and Dowex 1-X8 as size exclusion, cation exchange and anion exchange, respectively. Extracts were also fractionated by preparative thin layer chromatography where two compounds were separated from S. frutescens extract with high activity against reverse transcriptase while showing insignificant inhibition towards other enzymes tested. SFL-BFW-10 and SFL-WEF-7 inhibited reverse transcriptase by almost 100 percent and the IC50 values calculated for these compounds were 0.34 and 0.23mg/ml, respectively. Cytotoxicity of these compounds was evaluated on Chang liver cells and peripheral blood mononuclear cells (PBMCs). None of these compounds showed any significant inhibition of cell proliferation. The purity of these compounds could not be confirmed because there was insufficient material to use in the techniques required to show purity and identification. Therefore, TLC was used to determine the nature of these compounds. SFL-BFW-10 was identified as an organic acid and SFL-WEF-7 was identified as flavonoid.

The focus of the study was then shifted toward thiuram disulfide accelerated sulfur vulcanisation, with emphasis being placed on a rate comparison. Three groups of accelerators were investigated, namely the aromatic, linear aliphatic and cyclic aliphatic thiuram disulfide adducts. The analysis methods that were employed were conventional rubber (cis-1,4-polyisoprene) techniques coupled to model (squalene) compound investigations. The data that was collected consisted of rheometrical torque vs. time data in the rubber system while the data obtained in the model compound study consisted of sulfur and accelerator concentration data as determined by means of high performance liquid chromatography (HPLC). The aromatic accelerators were synthesised in our labs by means of an addition reaction between the aromatic amine and CS2 in basic medium and subsequent oxidation with K3Fe(CN)6, all in a 1:1 molar ratio. The reaction yield was low due to the instability of the dithiocarbamate intermediates and a sluggish oxidation reaction. In the rate constant determination a first order mathematical approach was used for the rubber system as crosslinking is considered to roughly obey first order kinetics. The model compound data was also found to more accurately fit the first order rate law, with an initial slopes method also being applied to the system to determine secondary rate constants and relative rates for the system. The determination of vulcanisation rate constants in the cis-1,4-polyisoprene system was a success, while the rate data determined by means of the squalene model was more related to the rate of accelerator and sulfur consumption as opposed to the rate of crosslinking as is the case with the rubber rate data. The sulfur first order rate data mirrored the rate data derived from the rubber system more closely than the corresponding accelerator rate data, the relative rate data determined by means of initial slopes method, proved that the homolytic cleavage of thiuram disulfides and the subsequent formation of accelerator polysulfides were not limiting steps. This is seen in the similar relative rate data derived from both the raw sulfur and accelerator data in systems that exhibit vastly different vulcanisation rates. Squalene was deemed a suitable model for the cis-1,4-polyisoprene system, although one should consider the extent of charring and solution effects in the individual systems to account for possible incongruities that may be observed between the rubber and simulated system. The lack of agreement between the rubber and model compound rate constant data lies in the fact that the rate of crosslinking is not simplistically related to the rate at which accelerator and sulfur is consumed, this being especially true for the rate at which the accelerator is consumed. - Description: Apart from the concentrations of chromite in layers within the Great Dyke and other ultramafic complexes, chromite also occurs as interstitial grains throughout the olivine-bearing rock-types. These olivine-bearing rocks include no rites, gabbros, dunites and pyroxenites. Chromite concentration in these rocks varies from 0.48 to 3.09 per cent of the rock, usually in the form of chromite (Ahrens, 1965; Worst, 1960). A small fraction of this chromite settled to form chromitite layers whilst the remainder is retained within the rock mass as finely disseminated chromite and chromite interstitial to olivine. This retained chromite is much finer grained than layer chromite and is the primary source of eluvial chromite (Cotterill, 1981). During weathering of the serpentine rock and transportation by rainwater, the heavier chromite and magnetite grains are re-deposited along watercourses and vleis or valleys as the speed of the water is retarded sufficiently for the heavier particles to settle. The lighter serpentine material is removed and the chromite concentration in the soil is increased, thus resulting in eluvial chromite (Keech et ai, 1961; Worst, 1960; Prendergast, 1978). The concentration of chromite particles in soil can be up to 15 (or more) Cr₂O₃ %, resulting in economic and exploitable deposits, located primarily along the Great Dyke fiacks. A preliminary evaluation of the eluvials indicate that the Great Dyke could be host to up to 10 million tonnes of potential chromite concentrates which could be processed from such eluvial concentrates. These chromite-rich soils can be mined more cheaply than the traditional seams mining and processed into chromite concentrates through simple mechanical processing techniques of spirals, jigs and heavy media separators. The resultant chromite concentrates are of high quality and can be used to manufacture chromite ore briquettes, which are an alternative to lumpy chromite smelter feed. The main challenges to eluvial mining are the inevitable environmental degradation and coming up with methods that could possibly mitigate against such environmental damage. The distribution of these eluvials over vast plains as thin soil horizons, necessitate use of mobile concentrator plants and hence establishment of extensive infrastructure. These challenges, however, are not insurmountable and test mining and previous production runs have proved profitable. The eluvials are also associated with some lateritic nickel concentrations. The nickel occurs in close association with some oxide such as goethite and garnierite and is associated with iron-manganiferous soil pisolites. The analyses of these pisolites indicate high nickel grades of generally above 1.00 %Ni. Such high nickel-content of Great Dyke laterites warrant, further investigations.

Ultimately we were able to produce a modularized, easily extend-able program which effectively makes use of Monte Carlo simulation techniques to price lookback, Asian and barrier exotic options. Theories of variance reduction were validated except in cases where we used control variates in combination with the other variance reduction techniques in which case we observed increased variance. Again, the main aim of this half thesis was to produce a C++ program which would produce stable pricings of exotic options.
