Photodynamic antimicrobial chemotherapy activities of porphyrin- and phthalocyanine-platinum nanoparticle conjugates

- Description: This work reports on the conjugation of differently shaped Pt nanoparticles (PtNPs) with ClGa(III) 5,10,15,20-tetrakis-(4-carboxyphenyl) porphyrin (1) as well as chloro - (5,10,15,20-tetrakis (4- (4- carboxy phenycarbonoimidoyl) phenyl) porphyrinato) gallium(III) (2) The work also reports on platination of dihydroxosilicon octacarboxyphthalocyanine (OH)₂SiOCPc (3) to give dihydroxosilicontris(diaquaplatinum)octacarboxyphthalocyanine (OH)₂SiOCPc(Pt)₃ (4). The resulting conjugates were used for photodynamic antimicrobial chemotherapy against S. aureus, E. coli and C. albicans. The degree of photo-inactivation is dependent on concentration of the conjugates, light dose (fluence) and illumination time. The log reduction obtained for 1 when conjugated to cubic PtNPs was 4.64 log (which indicate 99.99 percent of the bacteria have been killed), which is much higher than 3.94 log unit for 1-hexagonal PtNPs and 3.31 log units for 1-unshaped PtNPs. Complex 2 conjugated to hexagonal PtNPs showed 18 nm red shift in the Soret band when compared to 2 alone. Complex 2 and 2-hexagonal PtNPs as well showed promising photodynamic antimicrobial chemotherapy (PACT) activity against S. aureus, E. coli and C. albicans in solution where the log reduction obtained was 4.92, 3.76, and 3.95 respectively for 2-hexagonal PtNPs. The singlet oxygen quantum yields obtained were higher at 0.56 for 2-hexagonl PtNPs in DMF while that of 2 was 0.52 in the same solvent. This resulted in improved PACT activity for 2-hexagonal PtNPs compared to 2. Complex 4 showed slight blue shifting of the absorption spectrum when compared to complex 3 The antimicrobial activity of 4 were promising as the highest log reduction value was observed when compared to the porphyrin conjugates.

It focuses on instilling Ubuntu values of sharing and giving that are of critical significance in teaching and learning. The context of the thesis is located broadly within socio-economic rights and specifically on education rights. In South Africa where the state has not adequately met the educational obligations for the economically and socially less privileged citizens, the emergence of educational NGOs that focus on providing education to the poor is of vital importance. Although other NGOs that confront the state are important in pushing the state to deliver especially on school infrastructure, teacher deployment and other educational challenges, Gadra Education model ensures academic success for the learner. Lessons can be drawn from Gadra Education which can be potentially useful to state schools and other NGOs that seek to advance education rights to disadvantaged communities.

- Description: This research deals with achieving and maintaining price stability in Sub-Saharan Africa (SSA) through the practice of monetary-policy transparency (MPT). On the one hand, MPT refers to a monetary strategy whereby the central bank is insulated from political influence and made accountable to society through disclosure of its policies, procedures, economic models, data and forecasts, operations and political practices (such as objectives, personnel independence, and the like). On the other hand, price stability refers to achieving and maintaining low and stable levels of inflation conducive for long-term planning and poverty alleviation. The primary objective of this research was to investigate MPT in SSA as it represents a powerful means whereby economic agents’ expectations may be coordinated and managed by the central bank to achieve its societal, objective function of low inflation. The empirical evidence shows that, first, a dependent central bank is more likely to slip into hyperinflation. Second, a SADC (2008) model central bank law is not independent enough to be used as a benchmark for any central bank or as a charter for a regional central bank. Third, the degree of central bank independence in SSA is relatively lower than that in industrialised economies. Fourth, the determinants of MPT in SSA are trade openness, and financial depth that are important factors influencing policy-makers to adopt monetary-policy transparency. Fifth, MPT is associated with a decline in the inflation rate. Sixth, MPT had no significant effect on economic output, whilst trade openness was positively associated with real GDP.

- Description: Purpose – Empirical work on the career development processes of vulnerable groups such as refugees is beginning to receive currency within the academic literature. However, little is known about the intersection of factors such as gender and structural constraints on such processes especially within developing countries such as South Africa. The purpose of this study was to explore the career development processes of women refugees in South Africa. Design/methodology/ approach – A qualitative research approach through narrative inquiry was used. Snowball sampling was employed to select the 20 women refugees who took part in this research. The study location was the city of Port Elizabeth located in the Eastern Cape Province. The three levels of meaning-making incorporated in previous narrative research was utilised to analyse the data. Findings Results show that women refugees are more concerned with a short-term desire to survive and acquire basic commodities than a long-term focus to advance and develop their careers. This is mainly compounded by constraints personal to the individual and those in the environment. Finally, the results illustrate that career development processes of women refugees are affected by the inextricably intertwined factors associated with their gender, immigration status as well as person-environmental factors. Practical implications – The research suggests strategies that policy makers in South Africa and other developing nations that are hosting refugees can use to assist women not just their career development but also their lived experiences. Originality/value – This study extends and advances literature on the career development issues of vulnerable groups such as women refugees especially in developing nations such as South Africa.

- Description: The central argument of this inter-disciplinary study is that IsiXhosa written poetry of 1912 – 2012 is a terrain of the struggle between the contending dominant ideologies of Segregation, Apartheid and Charterism (post-Apartheid); and the subordinate/ subaltern ideologies of Africanism, Charterism (pre-democracy), Pan- Africanism, Black Consciousness Movement and other post Apartheid ideologies. The study highlights the mutual relationship between the text and the context by focussing on the ideological contest which manifests itself in both form and structure (i.e. aesthetic ideology) and the content (i.e. authorial ideology) of the poetry of different epochs between 1912 and 2012. The study is located within the framework of Antonio Gramsci’s theory of cultural construction. Gramsci postulates that ideology and culture play a significant role in the process of asserting hegemony. Important concepts that constitute Gramsci’s theory of praxis are: ideology, culture, hegemony, organic intellectuals and both ideological and repressive state apparatuses. The first chapter presents the problem, the objectives, the methodology, and the scope of the study. The second chapter presents Gramsci’s theory of cultural construction and the work of scholars who developed his theory further. The tool that is employed for analysis and interpretation of textual significations of IsiXhosa written poetry is the revolutionary aesthetics, which is proposed by Udenta. The third chapter analyses and interprets literature of the epoch of 1912-1934 and exposes the contest between Segregation and Africanism ideologies. The fourth chapter contextualises and analyses the literature of 1934 – 1948, the second phase of contestation between Segregation and Africanism. The fifth chapter deals with literature of the first and second halves of the Apartheid epoch (1948 - 1973). The Apartheid ideology contested with the Africanist ideology which transformed into the Charterism ideology in 1955. In 1960 Pan-Africanism ideology and in 1969 Black Consciousness Movement ideologies entered the contest. The sixth chapter examines literature of the period 1973 – 1994 which is the second phase of the Apartheid epoch that ends with the “glasnost” period of 1990 - 1994. The seventh chapter studies literature of the democracy period of 1994 – 2012. The eighth chapter is the summary and general conclusion. - Description: Single Point Diamond Turning (SPDT) is a technology widely applied for the fabrication of contact lenses. One of the limiting factors in polymer machining is wear of the diamond tool due to electrostatic discharge resulting in poor surface quality of the machined products. The research work presented in this dissertation highlights the electrostatic properties of contact lenses during machining operations and the effects these properties have on the surface quality of the work piece materials. Two contact lens samples were experimented on, Definitive 74 (Silicone Hydrogel) and Tyro 97 (Rigid Gas Permeable). The electrostatic surface potentials (ESPs) were measured during turning operations using an electrostatic voltmeter and the surface roughness measurements were taken using a surface profilometer. Response Surface Methodology (RSM) techniques were employed to create predictive models for both surface roughness and ESPs with respect to the cutting speed, feed rate and depth of cut. Predictive surface roughness models were successfully generated for both materials and the cutting speed and feed rate were identified as the parameters with most effect on surface roughness. In addition, an electrostatic model was successfully generated for the Definitive 74 contact lens material which cited the cutting speed and feed rate as the most effective parameters on the material’s electrostatic behaviour. However, no relationship was evident between the machining parameters and electrostatic behaviour of Tyro 97.

Appraising the appraisal framework: evidence from Grahamstown property advertisements

- Description: This thesis considers how interpersonal meaning choices in property advertisements are best reflected in a context of constraint in Grahamstown, South Africa, using and appraising Martin & White’s (2005) APPRAISAL Framework. The study is comparative in two senses since the data is analysed using Martin & White’s (2005) APPRAISAL Framework and a revision to it, and property advertisements from two competing estate agencies are analysed: Remax Frontier Properties and Pam Golding Properties. An initial strict application of Martin & White’s (2005) framework is followed by a second, based on analyst difficulties and framework limitations experienced in the first analysis, as well as those experienced by other researchers. Revisions to the original framework include added ‘local’, context-driven features and sub-categories: Exclusivity and Convenience in Attitude, a Scale of Intensity in Graduation, and a category ‘Invite’ in Engagement. These enable a richer, more detailed account of the alignment strategies and interpersonal micro-politics at play in the property advertisements than is possible using the original framework. Findings from the analyses reveal four facts of note. Firstly, that while the original Martin & White (2005) APPRAISAL Framework captures a general level of interpersonal meaning in the data, it does so more fully when it includes contextual and contextually-driven categories that are informed by local knowledge. Secondly, two levels of meaning are expressed in the data. One is aimed at an ‘external’ audience; the other, truer, fuller and more contentious, is aimed at what appears to be the intended audience only. Thirdly, and relatedly, specific contextual and cultural knowledge is needed by the intended audience to access the intended meaning. - Description: This thesis considers how interpersonal meaning choices in property advertisements are best reflected in a context of constraint in Grahamstown, South Africa, using and appraising Martin & White’s (2005) APPRAISAL Framework. The study is comparative in two senses since the data is analysed using Martin & White’s (2005) APPRAISAL Framework and a revision to it, and property advertisements from two competing estate agencies are analysed: Remax Frontier Properties and Pam Golding Properties. An initial strict application of Martin & White’s (2005) framework is followed by a second, based on analyst difficulties and framework limitations experienced in the first analysis, as well as those experienced by other researchers. Revisions to the original framework include added ‘local’, context-driven features and sub-categories: Exclusivity and Convenience in Attitude, a Scale of Intensity in Graduation, and a category ‘Invite’ in Engagement. These enable a richer, more detailed account of the alignment strategies and interpersonal micro-politics at play in the property advertisements than is possible using the original framework. Findings from the analyses reveal four facts of note. Firstly, that while the original Martin & White (2005) APPRAISAL Framework captures a general level of interpersonal meaning in the data, it does so more fully when it includes contextual and contextually-driven categories that are informed by local knowledge. Secondly, two levels of meaning are expressed in the data. One is aimed at an ‘external’ audience; the other, truer, fuller and more contentious, is aimed at what appears to be the intended audience only. Thirdly, and relatedly, specific contextual and cultural knowledge is needed by the intended audience to access the intended meaning. Teachers collaboratively and actively engaged in a range of activities that relate to classroom practices. Teacher learning was also enabled when teachers engaged in maths overlapping communities of practice, shared classroom experiences in friendly ways with fellow NICLE teachers and engaged with NICLE presenters who mutually respected and regarded them as professionals. Such affordances were said to enable teachers to engage learners in maths classes and improve their understanding of specific primary maths concepts. On the other hand teachers felt challenged by the travelling distance, limited time and also raised the tension of how to scale-up maths professional development initiatives to include schools from their community. The study makes a theoretical contribution by illustrating how Bernstein’s pedagogic identity model and its elaboration by Tyler (1999) provides analytical tools to interrogate macro educational changes and connect these to the micro processes and teacher identities.

The South Africa Government proposed Responsible Tourism as the guiding principle for sustainable tourism development (Goodwin, 2011: 138). The problem is compounded by shortcomings identified in the nature of the research conducted about tourism and communities and the methodologies used. Tourism-Community relationships Theory was applied as the conceptual framework of the study. This theory advocates a holistic approach when conducting research about the impacts of tourism, encompassing community perspectives, attitudes, reactions and considerations for the future development and control of tourism (Pearce, Moscardo & Ross, 1996: 2). The insights gained contributed an emic understanding of the keyinformants’ perspectives imparted in their own language. This study is located in the Interpretevist paradigm, underpinned by Constructivist philosophy. Meaning is created through the reality of key-informants, their perspectives and the interaction of individuals in a community (Cresswell, 2014: 8). The Instrumental Case Study Approach (Stake, 1995) contributed an etic perspective in terms of the researcher’s interpretations. The single, qualitative case was applied to gain deeper insights about the phenomenon of Responsible tourism. The case investigated was Stormsriver Adventures (SRA), an eco-adventure tourism business in the Tsitsikamma tourism destination in the Eastern Cape region of South Africa. SRA is reputed to be business best practice in its implementation of Responsible Tourism. Data triangulation was established through multiple methods of data collection, namely in-depth interviews with key-informants, documentation sourced from them and researcher observation. Data analysis methods, namely direct interpretation, categorical aggregation, pattern seeking and naturalistic generalisations (Stake, 1995), were used. The member checking process assisted with establishing data triangulation as keyinformants corroborated or dispelled the researcher’s interpretation of data. Findings were structured according to within-case themes, namely SRA’s perspective of its Responsible Tourism practices, the community’s perspective of SRA’s Responsible Tourism practices and perspectives about the future development and control of tourism. The study contributes new theoretical insights through applying the inductive approach to selected findings. Findings indicate that the perspectives of the community are divided. SRA keyinformants conveyed positive economic, social and environmental impacts of SRA’s Responsible Tourism practices. Key-informants not directly linked to SRA raised negative economic impacts and questioned the accuracy of SRA’s marketing messages. Within-case conclusions are that it is not the responsibility of one tourism business that practices Responsible Tourism to achieve sustainability within the tourism destination. To this end it is recommended that SRA and the community find common ground and that the misunderstandings that exist, be addressed. The study concludes that in order to achieve successful Responsible Tourism- Community Relationships, a holistically and integrated approach is required. To this end the recommendations made provide an implementation framework within the context of a tourism destination. The study adds value to the field knowledge about the implementation of Responsible Tourism by the private sector in South Africa and also contributes new knowledge to the field of tourism in terms of the particular methodology used.

This view has been questioned in the last century. The size of the ‘invasion’, and indeed its very existence, have come into doubt. However, this doubting school of thought does not seem to take into account all of the evidence. An interdisciplinary, nuanced approach has been taken in this thesis. Firstly, the question of Germanic raiding has been examined, with reference to the Saxon Shore defences. It is argued that these defences, in their geographical context, point to the likelihood of raiding. Then the written sources have been re-examined, as well as physical artefacts. In addition to geography, literature and archaeology (the disciplines which are most commonly used when the coming of the Angles and Saxons is investigated), linguistic and genetic data have been examined. The fields of linguistics and genetics, which have not often both been taken into consideration with previous approaches, add a number of valuable insights. This nuanced approach yields a picture of events that rules out the ‘traditional view’ in some ways, such as the idea that the Saxons exterminated the Britons altogether, but corroborates it in other ways. There was an invasion of a kind (of Angles – not Saxons), who came in comparatively small numbers, but found in Britain a society already mixed and comprising Celtic and Germanic-speaking peoples: a society implied by Caesar and Tacitus and corroborated by linguistic and genetic data.

- Description: This research report documented in this thesis focused mainly on the participation of grade 9 girls during practical work in the topic of ‘Electricity’ in Physical Science lessons at Mwatya (pseudonym) Junior Secondary School in Ohangwena region of Namibia. The study was triggered by the lack of participation of girls during practical work. Underpinned by an interpretive paradigm, the study took the form of a qualitative case study and my unit of analysis was girls’ participation during practical work on electricity in particular. The perceptions and experiences of girls about practical work in Physical Science were investigated using the administered questionnaires. The participation of girls during practical work was further explored using interviews (semi-structured and focus group interviews) and observations of selected girls. A thematic approach to data analysis was adopted. The qualitative data from the audio and video transcripts were analysed according to the categories developed and themes that emerged from the analysis process. The categories developed for presenting the data are: teaching methods preferred by girls; importance and roles of practical activities; things that girls enjoy during practical activities; problems experienced by girls during practical activities; preference for doing practical work in groups or as an individual task; if practical work given in groups should the sex of learners be considered or not; activities preferred by girls when they are in single sex and in mixed sex groups; factors that enable girls to participate; and factors that prevent girls from participating. The study found that girls’ perceptions about practical work affect their participation during practical work in the topic of Electricity. - Description: This research report documented in this thesis focused mainly on the participation of grade 9 girls during practical work in the topic of ‘Electricity’ in Physical Science lessons at Mwatya (pseudonym) Junior Secondary School in Ohangwena region of Namibia. The study was triggered by the lack of participation of girls during practical work. Underpinned by an interpretive paradigm, the study took the form of a qualitative case study and my unit of analysis was girls’ participation during practical work on electricity in particular. The perceptions and experiences of girls about practical work in Physical Science were investigated using the administered questionnaires. The participation of girls during practical work was further explored using interviews (semi-structured and focus group interviews) and observations of selected girls. - Description: In democratic South Africa, policies place much emphasis on the need for transformational leadership. This challenges school leaders to ensure that their practice is in line with the democratic and inclusive values espoused therein. This thesis is an account of my journey of learning about educational leadership and how I attempted to influence transformation at my own school. The development of my living theory of educational management is grounded in my desire to make a positive change to the quality of teaching and learning at my school, by embodying and exemplifying such values in my leadership. My learning as an educational leader comprises my living theory on improving my educational leadership within a socially challenged context. I explain the context and problems experienced at my school and provide evidence of the need to move from the hierarchic, autocratic form of leadership, still prevailing at many South African schools. I adopted the theoretical framework of servant leadership to enable me to develop a more contextually sensitive and visionary style of leadership through critical reflection on my own practice. My stimulus for this journey of learning stemmed from the perceived contradiction between my espoused beliefs about leadership and my actual practice. My own autocratic leadership style was one of the main barriers that prevented teachers from attaining autonomy and taking on leadership roles within the school. My leadership style was more in alignment with the values of accountability, discipline and efficiency than those of care, trust and the development of the potential of others. This interrogation of my ontological values informed my subsequent interventions to improve my practice. Following an action research design, I investigated the quality of my leadership to determine which areas I needed to improve, took action to improve these and evaluated the change against the values inherent in the notion of servant-leadership. I embarked on a journey that helped me to shift my practice from being based on previously held authoritarian professional values towards values that underpin a more transformational leadership, such as care and trust. My journey of learning was guided by the tenets of self-study action research, which required critical self-reflection and holding myself accountable for my own actions The practical knowledge I gained through this self-reflection on my practice enabled me to make professional judgements, which then became conceptual knowledge in the form of a living theory generated by my research. This was made possible through a continuous process of data generation to extract evidence to test the validity of the claims to knowledge I made. Multiple sources of data (written, graphic and multimedia) were used to better understand the scope of happenings throughout the research and to monitor my practice over time. I explain how I used my improved understanding of leadership to promote collegiality for building quality relationships to promote teacher leadership for school improvement and how I subjected these claims to social and personal validation procedures. The significance of this study is that it contributes to new forms of practice and theory in terms of showing how a values-based approach to school leadership can influence positive change in teacher practice. While this study is a narrative of my practice, it is also a narrative of theorising about how my colleagues and I have come to know and how our thinking has changed about our work and ourselves. Although I had to indicate a cut-off point in this action research enquiry, the knowledge gained will continue to develop and influence my practice in the future and hopefully will be judged as useful by others in positions of leadership. The thesis is thus an original contribution to educational knowledge in the field of self-study action research. Assessment of the effects of stream channel incision on soil water levels, soil morphology and vegetation in a wetland in the Hogsback area, Eastern Cape, South Africa

- Description: Wetland degradation in the form of channel incisioning can significantly alter the hydrological functioning of a wetland. In this study in a small headwater wetland in the Hogsback area, Eastern Cape Province, the impact of channel incisioning on soil water levels and soil morphology was examined. A good correlation (R2 = 0.89) exists between the depth of channel incisioning and average water table depths in most of the 21 installed piezometers. In localised cases the upslope supply of water was in equilibrium with drainage from the piezometers. Although all the studied soils show hydromorphic characteristics, those continuously saturated close to the surface exhibit redox accumulations in oxygen supplying macropores, whereas gleyic colour patterns occur deeper in soils where the water table has been lowered by channel incision. The nature and occurrence of different hydromorphic soil indicators observed confirm the contribution of soil morphology as a valuable indicator of long-term average soil water conditions. The study evaluates the effects of hydrological processes on plant species composition and distribution in a Hogsback wetland. Twenty-four circular plots with a radius of 2 m were established between March and August 2013. Within each sample plot, the habitat information and species present were recorded including Braun-Blanquet cover-abundance value for each and every species present in the plot. Plant species were identified in the field and the unknown plant species were collected, pressed, oven-dried and identified at the National Herbarium, South African Biodiversity Institute (SANBI), Pretoria and Schonland Herbarium, Rhodes University, Grahamstown. A total of 39 taxa belonging to 19 families and 34 genera were recorded from the Hogsback wetland. Of the documented taxa, three were exotic and three endemic to South Africa, indicating diversity and dynamic nature of Hogsback wetland flora. Plant families with the highest number of species were: Poaceae (11 species), Asteraceae (5 species), Onagraceae and Cyperaceae with 3 taxa each. The low number of exotic plant species in Hogsback wetland (three taxa in total) indicates limited anthropogenic influence. Unique species recorded in Hogsback wetlands are three species that are endemic to South Africa, namely, Alchemilla capensis, Helichrysum rosum and Lysimachia nutans. Five main floristic associations were identified from the Hierarchical Cluster Analysis whilst Detrenched .Correspondence Analysis (DCA) identified four groups. The Canonical Correspondence Analysis (CCA) indicates that edaphic factors, particularly in an area covered with water, erosion category, organic matter content and water table depth were the most important environmental variables measured accounting for the vegetation pattern present in the Hogsback wetland.

- Description: Critical Infrastructure is often overlooked from an Information Security perspective as being of high importance to protect which may result in Critical Infrastructure being at risk to Cyber related attacks with potential dire consequences. Furthermore, what is considered Critical Infrastructure is often a complex discussion, with varying opinions across audiences. Traditional Critical Infrastructure included power stations, water, sewage pump stations, gas pipe lines, power grids and a new entrant, the “internet of things”. This list is not complete and a constant challenge exists in identifying Critical Infrastructure and its interdependencies. The purpose of this research is to highlight the importance of protecting Critical Infrastructure as well as proposing a high level framework aiding in the identification and securing of Critical Infrastructure. To achieve this, key case studies involving Cyber crime and Cyber warfare, as well as the identification of attack vectors and impact on against Critical Infrastructure (as applicable to Critical Infrastructure where possible), were identified and discussed. Furthermore industry related material was researched as to identify key controls that would aid in protecting Critical Infrastructure. The identification of initiatives that countries were pursuing, that would aid in the protection of Critical Infrastructure, were identified and discussed. Research was conducted into the various standards, frameworks and methodologies available to aid in the identification, remediation and ultimately the protection of Critical Infrastructure. A key output of the research was the development of a hybrid approach to identifying Critical Infrastructure, associated vulnerabilities and an approach for remediation with specific metrics (based on the research performed). The conclusion based on the research is that there is often a need and a requirement to identify and protect Critical Infrastructure however this is usually initiated or driven by non-owners of Critical Infrastructure (Governments, governing bodies, standards bodies and security consultants). Furthermore where there are active initiative by owners very often the suggested approaches are very high level in nature with little direct guidance available for very immature environments.

- Description: Critical Infrastructure is often overlooked from an Information Security perspective as being of high importance to protect which may result in Critical Infrastructure being at risk to Cyber related attacks with potential dire consequences. Furthermore, what is considered Critical Infrastructure is often a complex discussion, with varying opinions across audiences. Traditional Critical Infrastructure included power stations, water, sewage pump stations, gas pipe lines, power grids and a new entrant, the “internet of things”. This list is not complete and a constant challenge exists in identifying Critical Infrastructure and its interdependencies. The purpose of this research is to highlight the importance of protecting Critical Infrastructure as well as proposing a high level framework aiding in the identification and securing of Critical Infrastructure. To achieve this, key case studies involving Cyber crime and Cyber warfare, as well as the identification of attack vectors and impact on against Critical Infrastructure (as applicable to Critical Infrastructure where possible), were identified and discussed.
