- Description: The voltammetric behavior of vitamin B1 in alkaline medium at carbon paste electrode (CPE) is discussed. A method based on cyclic voltammetry for the determination of vitamin B1 at carbon paste electrode modified with manganese phthalocyanine (MnPc-CPE) is presented. In pH 10 buffer, the modified electrode showed catalytic activity towards the oxidation of vitamin B1 by considerably increasing the oxidation current of one of the observed waves compared to the oxidation current obtained at an unmodified electrode. The modified electrode was stable towards the determination of vitamin B1 compared to the unmodified electrode that is completely poisoned after a few scans. A linear relationship between the oxidation current and the concentration of vitamin B1 in solution was observed. The detection limit for vitamin B1 at the modified electrode was of the order of 10−5 mol dm−3. Commercial vitamin B1 tablets were analyzed using the modified electrodes.

Therefore, this demonstrates that they are potentially suitable photosensitive agents for PACT use. The TD-B3LYP/LanL2DZ calculations were performed to evaluate the singlet excitation energies of quaternized and non-quaternized porphyrins in vacuo. Our study shows excellent agreement between time-dependent density-functional theory (TD-DFT) excited energies and experimental S1 > S0 excitation energies. The small deviation observed between the calculated and experimental spectra arises from the solvent effect. The excitation energies observed in these UV-visible spectra mostly originated from electron promotion between the highest occupied molecular orbital (HOMO) for the less intense band and the HOMO−1 for the most intense band of the ground states to the lower unoccupied molecular orbital (LUMO) of the excited states.

- Description: The nanophotosensitizers based on acetophenoxy tetrasubstituted metallophthalocyanines (MPc) and detonation nanodiamonds (DNDs) were successfully formed and their photophysicochemical properties were determined. The zinc(II)Pc and indium(III)Pc complexes along with their nanoconjugates were found to have high singlet oxygen quantum yields (0.72 − 0.84) associated with the heavy central metal effect. The ability of the functional groups present on the DNDs to bind to the bacteria cell and the improved solubility of the nanoconjugates due to DNDs resulted in effective photodynamic antimicrobial therapy (PACT) activity against S. aureus planktonic cells, with the highest log reduction of 9.72 ± 0.02 for the conjugate of InPc conjugate with DNDs after 30 min irradiation. Compared to the non-quaternized phthalocyanines 3 and 4, the cationic phthalocyanines 3a and 4a exhibit a higher photodynamic inactivation against the planktonic cells with log reduction values above 9 at a concentration of 1.25 µM. This was attributed to the positive charge which enhances cellular uptake. More interestingly, 3a and 4a show a higher photodynamic inactivation (less than 3% of S. aureus survived) on their biofilm counterparts thanks to their stronger affinity to these cells. 3a and 4a Pcs also exhibited interesting PDT activity against MCF-7 cancer cells giving IC50 values of 17.9 and 7.4 μM, respectively following 15 min irradiation. The obtained results in this work show that the positively charged phthalocyanines 3a and 4a are potential antibacterial photosensitizers that show some selectivity toward the Gram-positive and Gram-negative bacteria as well as MCF-7 breasts cancer cells.

Applying PACT followed by the antibiotic treatment led to a remarkable reduction in the biofilm cell survival of two antibiotic-resistant bacterial strains, S. aureus (Gram-positive) and E. coli (Gram-nenative). Treating both bacteria strains with PACT alone showed no significant activity at 32 µM with 15 min irradiation, while CIP alone exhibited a minimum biofilm inhibition concentration (MBIC) at 4 and 8 µg/mL on S. aureus and E. coli, respectively following 24 h incubation. The combined treatment resulted in the complete eradication of the matured biofilms with high log10 reduction values of 7.05 and 7.20 on S. aureus and E. coli, respectively, at low concentrations. It was found that 15 min PACT irradiation of 8 µM of complexes (1 and 2) combined with 2 µg/mL of CIP have a 100% reduction of the resistant S. aureus biofilms. Whereas the total killing of E. coli was obtained when combining 8 µM of complex 1 and 16 µM of complex 2 both combined with 4 µg/mL of CIP.

The as-formed nano-photosensitizers were fully characterized by spectroscopic methods and their photophysicochemical properties were investigated as well. Photodynamic antimicrobial chemotherapy was performed on the planktonic cells of S. aureus strain. And the results show that these nano assemblies were able to completely inhibit the metabolic activity of the resistant bacteria strain S. aureus with a 10.26 log reduction in the viable count. Again, asymmetrical Pc showed higher photocatalytic activity compared to the symmetrical complex with higher kobs and fast initial rates for the former. The photocatalysis obeyed the Langmuir-Hinshelwood kinetic model. The target conjugates showed all the advantages of two different groups existing on a single entity.

A potential problem associated with constant application of viral biopesticides is the differing susceptibility to the virus observed between different geographic populations of the insect host. This could be related to a number of factors, including biological performance and fitness of the target insect population. This study compared a variety of phenotypic traits between geographically distinct T. leucotreta populations collected from the Addo, Marble Hall, Citrusdal and Nelspruit regions of South Africa, and reared under laboratory conditions for several generations. Traits including pupal mass, female fecundity, egg hatch, pupal survival, adult eclosion and developmental time were used as parameters to measure biological performance and fitness. Insects from the Citrusdal region of the Western Cape exhibited significantly lower pupal mass, female fecundity, egg hatch, pupal survival, adult eclosion and the longest duration in larval and pupal development compared to the other colonies investigated. This is the first study to report differences in the performance of laboratory reared T. leucotreta from different geographic locations, and the findings may have important implications for the application of viral biopesticides for the control of this pest in South Africa.

Single restriction endonuclease (REN) analysis of viral genomic DNA extracted from purified occlusion bodies showed that isolates differed in their DNA profiles. Partial sequencing of granulin and egt genes from the different isolates and multiple alignments of nucleotide sequences revealed the presence of single nucleotide polymorphisms (SNPs), some of which resulted in amino acid substitutions in the protein sequence. Based on these findings as well as comparisons with other documented CrleGV isolates, we propose two phylogenetic groups for CrleGV-SA isolates recovered in this study.

- Description: The synthesis and photophysicochemical properties of mono- and tetra-pyridyloxy substituted indium(III) phthalocyanines (InPcs) are presented in this study. General trends are described for quantum yields of fluorescence, triplet, singlet oxygen and photodegradation as well as lifetimes of fluorescence and triplet state of these compounds. The complexes exhibited high singlet oxygen quantum yields (ФΔ) ranging from 0.44 to 0.66 in DMF, and from 0.44 to 0.69 in DMSO and ФΔ = 0.31 for the quartenized tetra substituted InPc which is soluble in water. The triplet quantum yields (ФT) ranged from 0.77 to 0.95 in DMF and from 0.77 to 0.94) in DMSO. The tetra substituted photosensitizers do not differ in their inactivation of bacteria with over 8 log reduction of viable bacteria when compared with the mono substituted photosensitizer which could only manage a 1 log reduction.

The QDs and MNPs were linked to the Pcs via an amide bond and by chemisorption unto AuNP surface. There is a general decrease in fluorescence quantum yields of the Pcs in the presence of all the nanoparticles. There is an increase triplet quantum yields for Pcs in the presence of AuNPs and QDs, but not in the presence of MNPs. AuNPs conjugates irrespective of the central atoms have the highest singlet oxygen quantum yield and are more photo-stable than all the other conjugates. MPcs are less photostable in the presence of MNPs.

Low fluorescence quantum yields (ΦF) of 0.01 and 0.02 were observed for PbTepyPc and PbTpyPc, respectively. The singlet oxygen quantum yields (ΦΔ) were 0.60 and 0.68, for PbTepyPc and PbTpyPc, respectively using DPBF as a quencher in DMF. Singlet oxygen production of the embedded sensitizers in electrospun fiber were quantified using ADMA and were found to be ΦΔ = 0.41 and ΦΔ = 0.21 for PbTepyPc and PbTpyPc, respectively. Photodynamic inactivation of Escherichia coli (E. coli) with the quaternized photosensitizer at 5 μM, totally inactivated the E. coli (with log CFU = 10 decrease). Only 0.4 log CFU decrease was obtained with PbTpyPc. The embedded non-quaternized photosensitizer (PbTpyPc) was less active on the gram negative bacteria but the quaternized photosensitizer (PbTepyPc) was effective towards inactivation of E. coli.
