- Description: There is a lack of research studies in South Africa that explore young men’s lived experiences of being fathered. The aim of this study focused on exploring how Black South African men construct and understand their own lived experiences of being fathered by their biological father. This research study is situated within a social constructionism methodology and theoretical framework. Data were collected using semi-structured interviews with young men between the ages of 18-24. Data were analysed using thematic analysis as a way of analysing participants’ stories. Three central themes with subthemes emerged. The main themes regarded the father behaviour, fatherly roles and participants’ constructions of fatherhood. The study argues that fatherhood is a social construct based on participants’ lived experiences. - Description: Extensive reading, otherwise known as “reading for pleasure”, “recreational reading” or “voluntary reading”, appears to be absent in the Namibian society, especially within public educational institutions. With the reformed Education System, the Namibian Curriculum for Basic Education makes room for a reading period in a 7-day cycle, from grade 1 to 11, in which all learners are expected to engage in a sustained silent reading (SSR) of interesting material of their choice. Nevertheless, such a period seems to be insufficient for engaging learners in an enjoyable reading experience, which could possibly create a positive attitude towards reading as well as enhance their reading skills of comprehension and reading fluency, possibly resulting in a habit of individual SSR. This research intended to explore an out-of-school, extensive reading program, over the period of one year, in the hope of cultivating Grade 8 learners’ positive attitude toward reading, in a selected secondary school in the township of Katutura, Windhoek, Khomas Region, Namibia. This one-year study, in which eight Grade 8 learners participated, focused on English only reading, as it is the Language of Learning and Teaching (LoLT). Operating within the interpretivist paradigm, the research adopted a qualitative approach, using a single case study to explore how participants make meaning of their experiences and attitudes toward ER. Qualitative data was collected through pre- and post-questionnaires and document analysis of learners’ reading journals as well as the researcher’s reflective journal. The research site was purposively and conveniently selected. The study drew on Krashen’s, input and affective filter hypotheses as the driving theory of this study, which according to some scholars, is the main second language acquisition theory applied in ER research. - Description: Studies in learner English proficiency in Namibia have shown that a considerable number of learners in primary and high schools have low proficiency. Naturally, this will be problematic for both learners and teachers who operate under a policy that dictates that from Grade 4 upwards, English is the medium of instruction in government schools. Literature has shown a general bias towards English as a medium of instruction from both the Namibian government and Namibian parents, despite the broader level of low English proficiency among learners. Research reviewed within this study has outlined the multiple benefits of code-switching for both learners and teachers. In fact, a recent study on the perceptions of code-switching in classrooms among Namibian learners indicated a positive attitude towards code-switching as a learning tool. Grade 8 Technical Drawing classes tend to be challenging due to the complexity and novelty of some of the concepts and terminologies used when teaching the subject. This makes it particularly difficult for learners whose first language is not English. Learners struggle to understand and make meaning of the concepts and objects used in Orthographic Drawing. Code-switching has been seen as a beneficial pedagogical tool. The study was therefore conducted to explore the Grade 8 teachers’ code-switching when teaching Orthographic Drawing in the Khomas educational region, Namibia. The study used the Matrix Language Frame Model by Myers-Scotton as the theoretical framework which focuses on two crucial language aspects of participating in code-switching and the matrix and embedded language, which guided this study. The qualitative approach employed a case study design which was used to gain first-hand experience in the Technical Drawing classrooms. Semi-structured interviews and an observation checklist were the research instruments that were used to generate data on the use of code-switching in Orthographic Drawing. Two schools, two Technical Drawing classes with 40 learners and two teachers in the Khomas region, Namibia were conveniently and purposively sampled. The data were analysed using coding, descriptive statistics and thematic data analysis. The primary and secondary results indicate that code-switching occurrence has constructive effects. - Description: The Namibian examiners’ reports have repeatedly reported that Biology is generally poorly performed in most schools and the topic on food tests has been identified as one of the problematic areas. My assumption is that this could be due in part to the lack of inadequate hands-on practical activities that are conducted in most schools in Namibia. In my view, for students to understand food tests and its associated concepts, they need to do hands-on practical activities. It is against this background that in this study I explored enablers and/or constraints when Grade 10 Biology teachers mediate learning of food tests using hands-on practical activities. This study is underpinned by an interpretive paradigm, within which a qualitative case study approach was employed. For my baseline data, I used a questionnaire and I also interviewed two Grade 10 Biology teachers using semi-structured interviews. Afterwards, the two Grade 10 Biology teachers were observed while teaching the topic of food tests. The lessons were videotaped and thereafter I conducted a stimulated recall interview watching the videos with each of these teachers. Further, we also discussed and reflected as a group on the mediation of learning on food tests. I used Shulman’s Pedagogical Content Knowledge (PCK) as my theoretical framework. Within PCK, I used the five Topic-Specific Pedagogical Knowledge (TSPCK) components by Mavhunga and Rollnick as my analytical framework. A thematic approach to data analysis was employed to come up with categories, sub-themes and themes. The findings revealed that Biology teachers do not have a dedicated laboratory for conducting Biology practicals and would instead conduct them in a common laboratory that is employed for both Physical Science and Biology or in their classrooms. There are inadequate resources to complement hands-on activities and teachers lack the capacity, skills and necessary knowledge needed to deliver practical work. The two participants did not bother to do enough practical work and instead taught Biology as a very theoretical topic, which disadvantaged learners for Paper 3, which serves as an alternative to course work. The study also revealed that teachers failed to conduct Biology practicals. They claimed to be doing so, but there were few practicals/experiments being carried out in those schools. The participants stated that the issue of time to conduct practical experiments for food tests was not sufficient. They complained that the processes were exhausting and tiresome and left them drained as they ran around trying to implement the practical experiments. Furthermore, a large number of learners results in overcrowded Biology classes and makes it difficult for the teachers to control them, which affected the teachers’ ability to perform practical experiments. The participants said that the high number of learners makes it difficult for them to accommodate all students in the laboratories, and that the laboratories themselves do not accommodate many learners. The study, thus, recommends that there should be continuous professional development programmes in schools to assist Biology teachers who have challenges in doing hands-on practical activities. This topic has proven to be one of the most difficult for Namibian learners as reflected by their poor responses to questions in the Grade 11 and 12 Namibian Senior Secondary Certificate ‘O’ examinations. Teaching abstract concepts such as redox reactions requires higher cognitive thinking skills and thus presents a challenge. Against this background, this study sought to explore how teachers mediate learning of redox reactions in their classrooms. The focus was on conceptual understanding through linking content to the context of the learners. The study was underpinned by an interpretivist paradigm, within which a qualitative case study approach was adopted. Two Grade 11 Chemistry teachers from the Ohangwena and Oshana regions in Namibia were the research participants. I used semi-structured interviews, lesson observations and stimulated recall interviews to collect data. I used Shulman’s Pedagogical Content Knowledge as a theoretical framework in this study and the data sets were analysed using the Topic-Specific Pedagogical Content Knowledge components. - Description: The Namibian Science curriculum clearly states that learning of science should be promoted through using inquiry-based approaches. However, it does not state how teachers should go about promoting inquiry-based approaches in their classrooms, especially in under-resourced rural schools. This is exacerbated in part by the fact that there is inadequate or lack of professional development for science teachers which focus in particular on promotion of inquiry-based approaches. As a result, science teachers tend to ignore inquiry-based approaches in their classrooms. It is against this background that my study sought to explore how the use of a ‘mini-ecosystem’ enables and/or constrains grade 5 learners from an under-resourced rural school to make sense of scientific inquiry. The study is underpinned by an interpretive paradigm. Within the interpretive paradigm, a qualitative case study approach, using the Predict-Explain-Explore-Observe-Explain (PEEOE) framework was adopted. This case study was carried out in an under-resourced rural Namibian school and the participants were grade 5 Natural Science and Health Education learners. I also invited a teacher from the school to be my critical friend and a participant observer. Data were generated using the Views About Scientific Inquiry (VASI) questionnaire, observations, focus group interviews and learners’ reflections. Vygotsky’s socio-cultural theory was my theoretical framework, and within this theory, I used mediation of learning, social interactions, the zone of proximal development and self-regulation as lenses to analyse my data. A thematic approach to data analysis was adopted. That is, qualitative data were analysed inductively to come up with sub-themes and thereafter common sub-themes were combined to form themes. The findings of the study revealed that the observation of mini-ecosystems enabled learners to interact and participate with each other in their respective groups. Moreover, learners were able to identify some scientific concepts such as evaporation, condensation, water cycle and rainfall. - Description: The Namibian Science curriculum clearly states that learning of science should be promoted through using inquiry-based approaches. However, it does not state how teachers should go about promoting inquiry-based approaches in their classrooms, especially in under-resourced rural schools. This is exacerbated in part by the fact that there is inadequate or lack of professional development for science teachers which focus in particular on promotion of inquiry-based approaches. As a result, science teachers tend to ignore inquiry-based approaches in their classrooms. It is against this background that my study sought to explore how the use of a ‘mini-ecosystem’ enables and/or constrains grade 5 learners from an under-resourced rural school to make sense of scientific inquiry. The study is underpinned by an interpretive paradigm. Within the interpretive paradigm, a qualitative case study approach, using the Predict-Explain-Explore-Observe-Explain (PEEOE) framework was adopted. This case study was carried out in an under-resourced rural Namibian school and the participants were grade 5 Natural Science and Health Education learners. I also invited a teacher from the school to be my critical friend and a participant observer. Data were generated using the Views About Scientific Inquiry (VASI) questionnaire, observations, focus group interviews and learners’ reflections. Vygotsky’s socio-cultural theory was my theoretical framework, and within this theory, I used mediation of learning, social interactions, the zone of proximal development and self-regulation as lenses to analyse my data. A thematic approach to data analysis was adopted. That is, qualitative data were analysed inductively to come up with sub-themes and thereafter common sub-themes were combined to form themes. The findings of the study revealed that the observation of mini-ecosystems enabled learners to interact and participate with each other in their respective groups. Moreover, learners were able to identify some scientific concepts such as evaporation, condensation, water cycle and rainfall. These findings are in contrast with the fact that they seemed to struggle to answer the VASI questionnaire that was conducted prior to observation. School students consistently demonstrate lack of understanding of stoichiometry-related concepts and skills such as the particulate nature of matter and related mathematical problem-solving. The practical implication for curriculum developers involves increasing the instances of integration of stoichiometry concepts at stronger levels of semantic gravity, for a more even semantic gravity range of integration types. Natural Sciences and Physical Sciences teachers also need to source and include real-life examples for explanations and applications of stoichiometry concepts when teaching, in light of current curriculum documents focusing more on abstract forms of integration. - Description: This study observes the literacy engagement of a group of learners enrolled in Grades 8–10 in Nombulelo High School, a poorly-resourced school in the city of Makhanda in the Eastern Cape, South Africa. The learners participated in a book club hosted and run by St Andrew’s College, a privileged independent school, as a community engagement initiative. The idea of extending literacy engagement and engagement with written texts beyond textbooks used in schools is critical for learners with ambitions to enter higher education. Studying at a university requires a lot of reading, and if reading has not been taken up as a practice that involves more than ‘text consulting’ (Geisler, 1994) students will be unlikely to read the number of texts required of them. Studies (see Geisler, 1994 for an overview) have shown how the literacy of the university is very different to school based literacies. The assumption behind the study on which this thesis reports is that engagement with fictional texts might promote reading and bring about understandings of this activity as enjoyable and not a task only associated with schooling. The study is underpinned by a critical realist philosophy which allowed for the identification of structures and mechanisms that led to the emergence of literacy events in learners’ lives and to their experiences and observations of those events. The study was guided by the following questions: How do learners from a poorly-resourced high school engage around fictional texts in the context of a book club? What enables or constrains this engagement? The study was impacted by the Covid-19 pandemic in that lockdown requirements meant that learners from St Andrew’s College could not participate in the book club as much as anticipated initially as they had been forced to return home to pursue online learning. Learners from Nombuelo High School were, however, granted access to College premises, where they met in the school library following strict Covid protocols. The study draws on in-depth interviews, observations and document analysis of five learners from Nombulelo High School who participated in the book club, as well as on book reviews they wrote for the book club website. The critical realist analysis allows for the identification of mechanisms in learners’ homes and communities that enable literacies, including those that are screen-based such as using a computer, mobile phones and other technologies. This study found evidence of challenges regarding school based texts, reading fictional texts and viewing it as an enrichment of the school project. Because of children were African the emergence of communal practices and story telling is woven throughout the results section. Thus, this study explored how primary school mathematics teachers use Kahoot as a formative assessment tool. The study adopted an exploratory case study underpinned by the interpretive paradigm. It was conducted in a primary school in the Shiselweni Region in Swaziland. The participants were ten mathematics teachers teaching different grades at the school. Data was generated using observations, workshops, structured questionnaires, journal reflections and focus group interviews. The theoretical and analytical framework that underpinned this study was a combination of Davis' (1989) Technology Acceptance Model (TAM) and Mishra & Koehler's (2006) Technological Pedagogical Content Knowledge (TPACK) framework. The study results showed that the perceptions and attitudes of primary school mathematics teachers towards using Kahoot generally appear to be positive. The study also revealed that all the participants found Kahoot helpful and perceived it as easy to use. The study found that the provision of technical support for teachers; and financial support for schools to implement technology use in the classroom were enabling factors for using Kahoot as a formative assessment technology. Constraining factors were lack of high-speed internet connectivity; and lack of basic technological knowledge for using Kahoot in formative assessment. The study concluded that Kahoot increases learner motivation, engagement, attention, enjoyment, and knowledge retention. This study recommended the integration of Kahoot gamification into the mathematics curriculum.The study recommends that there is a need for schools to initiate the presence of technologies in mathematics classrooms and encourage teachers to integrate technologies assessment and teaching. To explore this importance, senior phase mathematics teachers are encouraged to adopt gamification technology such as Kahoot! in assessment. The study adopted a case study research design with a interpretivist research paradigm, as this method allows detailed exploration and understanding of a specific context through a close connection with the participants. Data was collected using semi-structured questionnaires, focus group interviews, non-participant observations, and reflective journals. The research involved ten purposively selected senior-phase mathematics teachers from two schools in the Sarah Baartman District, Eastern Cape Province, South Africa. The study drew upon Vygoysky's Sociocultural Theory for its theoretical framework, while the Technological, Pedagogical, and Content Knowledge was used for analysis. The data analysis pursued an inductive approach. Adopting a thematic analysis included coding participant responses, organizing codes into concepts and categories, and developing a theoretical framework from the resultant categories. A four-phase process was involved: coding, conceptualizing, categorizing, and theorizing, was employed. This process and using NVivo software for data organization and management helped identify patterns, themes, and significant concerns surrounding the study's core subject. The findings from the study were as follows: Firstly, it was revealed that most teachers have a positive perception and attitude towards using Kahoot! for formative assessment, believing that it enhances teaching and learning quality. However, they showed scepticism due to insufficient Information and Communication Technologies knowledge and training. Second, teachers' pedagogical and technological experiences were predominantly enjoyable. Kahoot! was accepted as beneficial for promoting collaboration, interaction, and immediate feedback. Third, the study identified enabling and constraining factors in using Kahoot! for formative assessment. The enabling factors include Kahoot!, which was convenient and accessible, promoted immediate feedback and fostered collaboration and interaction. The constraining factors include Information and Communication Technologies and network interference, which posed limitations. The research concluded that Kahoot! as a platform for formative assessment indeed scaffold mathematical understanding. The study recommends increasing teachers' access to educational technology, incorporating continuous professional development programs, providing subject-specific training, and encouraging teachers to adapt to emerging technologies. - Description: On 19 May 2018 Ashwin Willemse, a coloured Springbok rugby player walked off the SuperSport set, a DSTV sports program, during a live broadcast. He accused his white co-hosts Nick Mallet and Naas Botha (two white apartheid-era Springboks) of patronising him (SABC News, 2018; Eyewitness News, 2018). Responses to the Willemse incident reflected racial divisions in the country. It appeared that many white South Africans criticised Willemse’s behaviour, while many black South Africans stated that they could identify with his response to the subtle racisms experienced in everyday life, and which he claimed to have experienced on the SuperSport program (SABC News, 2018; Eyewitness News, 2018). The racialised media outcry led Professor Jonathan Jansen, to ask: “why do two groups of people, staring at the same event ‘see’ two different realities” (Maleka, 2018: 4)? This thesis probes possible answers to Jonathan Jansen’s question regarding the Ashwin Willemse walkout. The thesis explores and analyses the diverse racialised responses to the ‘Willemse walkout’ through race theories and ‘coloniality of being’. Qualitative research methods were used in the form of a reception analysis involving two racially homogenous (black and white) focus group interviews, two in-depth individual interviews with black research participants, three in-depth individual interviews with white research participants, and a sample of tweets from black and white commentators who had opinions about the ‘Willemse walkout’. The thesis concludes that most black research participants supported Willemse’s actions and most of the white people in the group disagreed with Willemse’s behaviour. - Description: On 19 May 2018 Ashwin Willemse, a coloured Springbok rugby player walked off the SuperSport set, a DSTV sports program, during a live broadcast. He accused his white co-hosts Nick Mallet and Naas Botha (two white apartheid-era Springboks) of patronising him (SABC News, 2018; Eyewitness News, 2018). Responses to the Willemse incident reflected racial divisions in the country. It appeared that many white South Africans criticised Willemse’s behaviour, while many black South Africans stated that they could identify with his response to the subtle racisms experienced in everyday life, and which he claimed to have experienced on the SuperSport program (SABC News, 2018; Eyewitness News, 2018). The racialised media outcry led Professor Jonathan Jansen, to ask: “why do two groups of people, staring at the same event ‘see’ two different realities” (Maleka, 2018: 4)? This thesis probes possible answers to Jonathan Jansen’s question regarding the Ashwin Willemse walkout. The thesis explores and analyses the diverse racialised responses to the ‘Willemse walkout’ through race theories and ‘coloniality of being’. Qualitative research methods were used in the form of a reception analysis involving two racially homogenous (black and white) focus group interviews, two in-depth individual interviews with black research participants, three in-depth individual interviews with white research participants, and a sample of tweets from black and white commentators who had opinions about the ‘Willemse walkout’. The thesis concludes that most black research participants supported Willemse’s actions and most of the white people in the group disagreed with Willemse’s behaviour. Understanding students' entrepreneurship is critical as entrepreneurship is touted as a reasonable response to alleviate the high unemployment in South Africa. The study explored how students became entrepreneurs (or how their entrepreneurship intentions were formed) and the factors that enhanced (enablers and barriers) and inhibited them. The significance of living with and after COVID-19, youth and graduate unemployment alleviation places entrepreneurship development by institutions of higher learning, governments and private sector in the centre of economic recovery. The establishment of the Entrepreneurship Development in Higher Education (EDHE) in 2016, highlights the commitment of the South African government, for Higher Education Institutions (HEI) to engage more in entrepreneurship and do more research to understand how students become entrepreneurs and unravel the challenges. Understanding students' entrepreneurship intentions can facilitate a supportive environment for students entrepreneurship. With “very little research about student entrepreneurship at this university” being the guiding research problem, the aim was to gain insight into the entrepreneurship intention of university students by exploring factors that influence students to become entrepreneurs. The study employed Ajzen’s Theory of Planned Behaviour to explore the entrepreneurial intentions of university students in terms of the underlying attitudes, subjective norms, and perceived behavioural control. The students who participated in the study fell into three categories of entrepreneurial intention: (i) Intending to start a business, (ii) starting a business, and (ii) those who were already operating a business. All the participants, irrespective of their stage of entrepreneurial intention, demonstrated a positive attitude and self-efficacy. All participants showed self-efficacy by identifying barriers and taking positive steps to overcome them. The main barriers were access to funding, capital, training, business opportunities with the university and lack of interaction with established entrepreneurs. The results demonstrated the close interplay of attitudes, social norms and perceived behavioural control in students’ entrepreneurial intentions. Social recognition of entrepreneurship, social capital, education and role models contributed to positive attitudes, and social norms influenced students’ entrepreneurial intentions. A supportive environment was found to be critical for nurturing entrepreneurship intentions. The study recommends a supportive climate cultivated by Rhodes University to promote entrepreneurship. - Description: Challenges to learners’ meaning-making in the topic of electricity and magnetism contributes to underperformance in grade 10 Physical Science in Namibian schools. Teacher talk in content-based classrooms not only contributes to learners’ language development (Gibbons, 2003), but also facilitates meaning-making and cumulative knowledge-building (Halliday, 1999). However, it is possible that there are differences between the classrooms talk of novice and experienced Namibian Physical Science teachers. An understanding of differences between experienced and novice Namibian science teacher talk could inform teacher training and professional development and potentially help improve learners’ meaning-making in topics such as electricity and magnetism. However, no study could be found in the Namibian context which explored whether novice and experienced teacher talk afforded similar semantic and lexical access to meaning-making. This research gap provided a strong rationale for undertaking the study reported in this thesis. The study sought to investigate the extent to which novice and experienced grade 10 Namibian Physical Science teachers’ classroom talk provides semantic and lexical access to learners for the topic of electricity and magnetism. The research is informed by Legitimation Code Theory (LCT) and Systemic Functional Linguistics (SFL). Research has shown that the two theories are complementary and can be used to characterize teachers’ pedagogical practices (Maton, 2014). A quantitative case study methodology was followed in this study with LCT’s semantic density and SFL’s lexical density as analytical tools to analyze the two teachers’ classroom talk. The results from a t-test for semantic density show that there is a significant difference in the means for “semantic density waving” criterion (t-value of -2.331; p-value for 2 tail test at 95% level of 0.0040) and for the “linking downward escalators or single references” criterion (t-value of 4.649; p-value for 2 tail test at 95% level of 0.001) of teacher talk by the teachers. The overall results for semantic density indicate that the experienced teacher affords better epistemological access through semantic waves than the novice teacher, whereas the novice teacher affords better epistemological access through semantic range and semantic flow. In terms of lexical density, both teachers afforded similar access, characterized by their talk veering towards the level of information density associated with written text. The study makes a methodological contribution to science education research through its characterization of novice and experienced teachers’ talk in terms of semantic waves and lexical density. This study also provides empirical insight into the differences between language use by novice and experienced Namibian Physical Science teachers, which can inform the work of educational institutions, advisory and inspectorate services in the ministry of education, and school managers. - Description: This is a case study conducted in the Methodist Church of Southern Africa (MCSA), Grahamstown, in the Eastern Cape. Against the backdrop of identifying and conceptualising ethically oriented leadership competencies, the purpose of this study was to explore service as a leadership competency. The literature reviewed in this research underscored the importance of effective and ethical leadership, but critiqued the Westernised, individualistic orientation of the conceptualisation of service as a leadership competency. Based on a review of the literature, this study proposes that when African leaders are in leadership four components of Ubuntu are involved when they exercise service as a leadership competency. These components are collective consideration, collective compassion, community server and Ubuntu humility. The main aim of this study is to investigate if leaders display these four components when exercising service towards their followers, and if so, how, and why they do so. A conceptual framework of components of Ubuntu, which culminated in research themes and propositions, were produced from the literature chapter. A qualitative research method was adopted in this study in which eight participants were selected through convenience sampling for this case study. One minister and seven Vice-Presidents of various organisations in the Methodist Church were interviewed. Semi-structured interviews were conducted for data collection and a deductive thematic analysis approach was employed to analyse data. Also, the critical incident technique (CIT) was utilised to structure interview questions, which is best fitting to solicit complex and comprehensive data from the interviewees. In its findings, the study confirmed the presence of the four components of Ubuntu, but several delimitations and limitations are acknowledged. Nevertheless, the study has made some contribution to understanding the display of service as a leadership competency in an African context. - Description: Endo-xylanases (shortly called xylanases) are a group of glycoside hydrolase enzymes that target β-D-1,4-linkages in the xylan backbone, leading to the production of xylooligosaccharides (XOS) of varying degree of polymerization (DP). Xylan is an indigestible non-starch polysaccharide present in monogastric animal feeds which in high amounts leads to increased digesta viscosity, slow movement of digesta in the intestines, malabsorption of nutrients among other challenges. The aim of this study was to investigate the effect of xylanase 2A (Xyn2A) from Trichoderma viride on broiler chicken feeds, particularly the hydrolysis of the xylan content, reduction of feed viscosity and the effect of produced XOS on eliciting the growth of gut associated probiotic bacteria. Xyn2AE was successfully induced with 0.8 mM isopropyl β-D-1-thiogalactopyranoside (IPTG) and produced in Escherichia coli BL21 (DE3) and Xyn2AC was expressed in tobacco mosaic plants. For the purification of Xyn2AE, an immobilized metal affinity chromatography (IMAC) column and diafiltration using a 3kDa cut-off Amicon filter membranes were used. Xyn2AE and Xyn2AC showed a xylanase active band at a relative weight of 21 kDa. Both enzymes showed high specificity towards soluble wheat arabinoxylan (WAX), with specific activities of 7.61 U/mg for Xyn2AE and 536.5 U/mg for Xyn2AC. Xyn2A kinetic parameters (Vmax and Km) were determined by Michaelis-Menten plots on soluble and insoluble WAX. The Vmax and Km values of Xyn2AC were 1003.01 U/mg and 9.25 mg/mL, 302.89 U/mg and 13.54 mg/mL, respectively. The Vmax and Km values of Xyn2AE for soluble and insoluble WAX were 20.45 U/mg and 12.95 mg/mL, and 8.31 U/mg and 13.15 mg/mL. Xyn2A enzymes displayed optimum activity at pH and temperature parameters of 5.0 and 50°C, respectively, and stability in temperatures ranging between 50 and 80°C and pH 4.0-9.0. Broiler chicken feeds were hydrolysed using Xyn2AE over a 24 h period and analysed using the dinitrosalicylic (DNS) assay, thin layer chromatography (TLC), viscometry and visualized using scanning electron microscope (SEM). The results showed a release of release of XOS xylotriose, xylopentose and xylohexose; enzyme’s ability to decrease the viscosity of the feeds and punched holes of feed surface, which was indicative of xylanase action. XOS produced during hydrolysis was used to study prebiotic effect on selected few bacteria and released short chain fatty acids (SCFAs) were measured. Additionally, SCFAs formation was detected in the presence of XOS as a carbon source for S. thermophilus and L. bulgaricus, whereas B. subtilis formed fewer organic acids in the presence of XOS. The results obtained from this study demonstrated that the supplementation of Xyn2A on broiler feeds has ii a positive effect in decreasing feed viscosity. For the purification of Xyn2AE, an immobilized metal affinity chromatography (IMAC) column and diafiltration using a 3kDa cut-off Amicon filter membranes were used. Xyn2AE and Xyn2AC showed a xylanase active band at a relative weight of 21 kDa. Both enzymes showed high specificity towards soluble wheat arabinoxylan (WAX), with specific activities of 7.61 U/mg for Xyn2AE and 536.5 U/mg for Xyn2AC. Xyn2A kinetic parameters (Vmax and Km) were determined by Michaelis-Menten plots on soluble and insoluble WAX. The Vmax and Km values of Xyn2AC were 1003.01 U/mg and 9.25 mg/mL, 302.89 U/mg and 13.54 mg/mL, respectively. The Vmax and Km values of Xyn2AE for soluble and insoluble WAX were 20.45 U/mg and 12.95 mg/mL, and 8.31 U/mg and 13.15 mg/mL. Xyn2A enzymes displayed optimum activity at pH and temperature parameters of 5.0 and 50°C, respectively, and stability in temperatures ranging between 50 and 80°C and pH 4.0-9.0. Broiler chicken feeds were hydrolysed using Xyn2AE over a 24 h period and analysed using the dinitrosalicylic (DNS) assay, thin layer chromatography (TLC), viscometry and visualized using scanning electron microscope (SEM). The results showed a release of release of XOS xylotriose, xylopentose and xylohexose; enzyme’s ability to decrease the viscosity of the feeds and punched holes of feed surface, which was indicative of xylanase action. XOS produced during hydrolysis was used to study prebiotic effect on selected few bacteria and released short chain fatty acids (SCFAs) were measured. Additionally, SCFAs formation was detected in the presence of XOS as a carbon source for S. thermophilus and L. bulgaricus, whereas B. subtilis formed fewer organic acids in the presence of XOS. The results obtained from this study demonstrated that the supplementation of Xyn2A on broiler feeds has ii a positive effect in decreasing feed viscosity. - Description: Ciprofloxacin (CPH) is a broad-spectrum antibiotic used to treat bone, joint, and skin infections. It is commercially available as an extended-release tablet and as a cream dosage form. CPH is a bactericidal active pharmaceutical ingredient (API) of the fluoroquinolone drug class. It inhibits deoxyribonucleic acid (DNA) replication by inhibiting bacterial DNA topoisomerase and DNA gyrase enzymes. Common adverse effects include nausea, vomiting, unusual fatigue, pale skin, and may increase the risk of tendinitis, which could be a major concern. CPH is, according to the Biopharmaceutics Classification System (BCS), classified as a BCS class IV drug exhibiting low oral bioavailability, low solubility, and intestinal permeability. CPH was chosen as a good candidate for the study because of its stability in solutions, its low molecular weight (331.4 g/mol), and its moderate lipophilicity (log P = 0.28) [16]. The use of conventional ear drops in the ear is effective, avoids hepatic first metabolism and extensive protein binding and may reduce adverse effects as a low dose may be used to achieve a therapeutic effect. However, conventional ear drops and oral antibiotics have a long onset of action and have to be taken/applied in short intervals. For convenience and assurance of a long residence time in the ear, CPH may be delivered by using a niosomal formulation, a liquid at room temperature, to allow administration into the ear without the need to constantly apply the ear drops for long periods of time. A simple, rapid, precise, accurate, reproducible, and specific reversed-phase high-performance liquid chromatography (RP-HPLC) method using ultraviolet (UV) detection for the quantitation of CPH was developed and optimized using a central composite design (CCD). The method was validated using International Conference on Harmonisation (ICH) guidelines and was found to be linear, precise, accurate, and specific for the analysis of CPH. Since the method is specific, it was used to quantify CPH in commercial and experimental formulations and monitor CPH released during in-vitro release testing. The compatibility of CPH and potential excipients was investigated during preformulation studies using Fourier transform infrared spectroscopy (FT-IR) and differential scanning calorimetry (DSC) to identify and select suitable excipients for use during formulation development activities. No apparent interactions were evident between CPH, and the excipients tested. The probe sonication method was used to manufacture CPH loaded niosomes using different surfactants/surfactant combinations, and a combination of Tween® 80: sodium lauryl sulfate (SLS) was found to be the best composition in terms of both entrapment efficiency and Zeta potential. The limits for the independent input variables used for the manufacture included amplitude, sonication time, and amount of cholesterol were determined. Design of experiments (DOE) was used to design the study. The input variables investigated included amplitude, amount of cholesterol, and sonication time. The output or responses monitored included Zeta potential, vesicle size, polydispersity index (PDI), and entrapment efficiency. Non-ionic surfactant systems are predominantly stabilized by steric stabilization, and there is only a minor electrostatic element from adsorbed hydroxyl ions. With the inclusion of SLS it is to be expected that Zeta potential will be a contributing factor. DOE using Box-Behnken design (BBD) and response surface methodology (RSM) in addition to Artificial Neural Networks (ANN) were used for the optimization of the formulation. The optimized formulation had a composition of 1 g cholesterol, 1 g of Tween® 80, 1 g of SLS and was prepared at an amplitude of 11.294 % with a sonication time of 3.304 minutes. The formulation exhibited zero-order release kinetics and had an average pH of 7.45. The formulation was stored at 4 ℃ and 25 ℃ and was assessed for vesicle size, entrapment efficiency, Zeta potential, colour, lamellarity, and PDI every 7 days for 4 weeks. The lead formulation stored at 4 ℃ was more stable than the formulation at 25 ℃ in terms of entrapment efficiency, PDI and vesicle size during the 4-week period. CPH loaded niosomes for transtympanic delivery in the treatment of otitis media were developed and optimized.
