- Description: The purpose of IHL is to protect civilians and provide obligations that parties to the conflict are to adhere to. These obligations in case of a non-international armed conflict emanate from Common Article 3, Additional Protocol II and customary international humanitarian law. The reason for the imposition of these obligations on the parties to the conflict is for the need to protect the civilian population against the effects of hostilities which mostly are women and children. As the conflict in Somalia has gone on for too long, IHL plays an important role in protecting civilians. As the Geneva Conventions regulate armed conflicts together with its Additional Protocols they set out the requirements for the treatment of those not taking part in hostilities. In direct contravention of the Geneva Conventions by the parties to the conflict, the persons not taking part in hostilities have been the victims of attacks by armed groups. Even though armed groups have obligations, breaches have continued to occur resulting in impunity and perpetrators of violence have gone unpunished and there is need to close the accountability gap in respect of holding armed groups accountable in Somalia. The aim of this research is to assess how armed groups in Somalia can be held accountable for the atrocities they have committed. In an attempt to close the gap the researcher discusses the obligations that armed groups have, and when these obligations are breached many result in criminal accountability in respect of war crimes. It is also stated in the research that a State has an obligation to prosecute those in breach of IHL obligations. For prosecution to be possible in Somalia there is need for the Federal government of Somalia to adopt new legislation to enforce the justice system in the attempt to hold armed groups accountable and where possible to also utilise available courts as it is difficult and expensive to establish a tribunal. To end impunity armed groups are to be held accountable.

- Description: In order to meet the different public needs and requirements of contemporary urban society, a better understanding of public perceptions, appreciation and knowledge of the assorted values derived from urban trees is beneficial for improved urban greening. Thus, this study carried out an assessment of urban residents’ knowledge and appreciation of the benefits of trees and the role they play in enhancing the quality of life in urban systems. The study primarily focused on the intangible benefits (also referred to as cultural and regulating services) of urban trees, because most research to date in South Africa on trees and the benefits derived from them is biased towards the provisioning services of trees in rural areas. The significance and contribution of the intangible benefits of trees in urban systems is often underestimated and not seriously taken into account by urban planners, policy makers and other key decision makers. The study was conducted in three different residential settlement types (formal townships, RDP settlements and informal settlements) in two towns in the Limpopo Province in South Africa, namely Bela-Bela and Tzaneen. Findings revealed that the intangible benefits of trees are seen to play a significant role in improving both residents’ quality of life and the environmental conditions in cities, including the preservation of biodiversity. Findings also showed that urban residents had strong spiritual connections with trees and placed considerable cultural and historical importance on specific tree species and individual trees. Urban residents in both towns had moderate knowledge of the intangible benefits of trees based on the knowledge score that was created for the purposes of this study. Fifty seven percent of the residents had ‘reasonable’ knowledge of some of the intangible benefits of trees, while 43% had low levels of knowledge. That said, the majority of residents demonstrated high appreciation (98%) of the intangible benefits of trees (especially social and cultural values), with few residents not recognising these, suggesting that knowledge does not necessarily lead to appreciation. Age and level of education were found to influence knowledge of the intangible benefits of trees. A large proportion (86%) of residents who possessed tertiary level qualifications had more knowledge of the intangible benefits of trees as compared to residents with no formal education. Findings also revealed that younger respondents (60%) had more knowledge of the intangible benefits of trees. Factors that influenced appreciation of the intangible benefits of trees included links to crime, peoples’ value systems (their pro-environmental and altruistic values) and residential settlement type. Sixty five percent of residents in both towns felt that urban street trees and trees in public spaces attract criminals and promote crime, while 67% of RDP and 96% of informal township residents did not use or appreciate the social and recreational benefits of trees mainly because of the absence or long distances to public parks. A Willingness To Pay (WTP) survey revealed that a high proportion of residents in Bela-Bela (86%) were WTP a small annual fee in order to sustain greening initiatives in their communities compared to the residents in Tzaneen (53%). In terms of the planting and retention of trees, it was found that 300 households in both study towns had planted and retained a total of 1 615 trees in spite of the various factors that negatively influenced planting and retention of trees such as residency time and tenure security. The majority (66%) of informal township residents said tenure security was an important factor to consider when making tree planting decisions, while this was not an issue for formal township and RDP residents. Policy implications and recommendations are presented to help municipalities and urban planners improve and develop effective policies and programmes that will enable implementation of sustainable urban greening programmes

Determinants of household savings in South Africa: an econometric approach

In South Africa saving levels have been declining, particularly household savings. This has been a major cause of concern as low savings hinder economic growth. In light of this, the study explored the determinants of household savings in South Africa over the period 1990 to 2011 using quarterly data. Based on the review of the theoretical and empirical literature, particular attention was paid to the effects of age dependency ratio, the level of household income, inflation and real interest rate on household savings. Apart from informal graphical test, the study employed the Augmented Dickey-Fuller and Phillips Perron unit root tests to test for stationarity in the time series. To identify the long-run and short-run dynamics among the variables, the study used the Johansen co-integration and the Error Correction Mechanism. Results of the study indicated that age dependency ratio, inflation and real interest rate have a positive impact on household savings whilst income has a negative long run relationship with household savings. In addition, the findings revealed that income, inflation and real interest rate play a major role in determining household savings whereas age dependency ratio is insignificant. The study recommends that the government should employ a countercyclical fiscal policy to avoid the development of excessive current account deficits during periods of more rapid economic growth, rising investment and falling saving.

The crisis emerged in the 1990s with the introduction of a structural adjustment programme and deepened further with the fast track land reform programme initiated by the Zimbabwean government in the year 2000. The crisis has involved a down-sizing of the Zimbabwean economy and a massive rise in the rate of unemployment in the formal economy. Consequently urban life became increasingly unbearable for poor blacks and informal economic activities blossomed and started to make a significant contribution to household income and livelihoods. The role of women in the informal economy was particularly pronounced. Theoretically, the thesis is underpinned by the sustainable livelihoods framework. In examining the vulnerability context of the Magaba women traders and the institutional interventions which complicate the lives and livelihoods of these traders, I identify and unpack their diverse livelihood activities and strategies and the resources (or assets) they deploy in constructing urban livelihoods. Though their livelihood portfolios complement any earnings from formal employment by household members and though they contribute to their household’s sustenance, there are a number of daily challenges which they face in their trading activities and which they seek to counteract through a range of often ingenious coping mechanisms. The thesis is important for a number of reasons. It fills an important empirical gap in the study of Magaba market specifically, it brings to the fore the gendered character of the informal trading activities in urban Zimbabwe, and it deploys the livelihoods framework in a manner which is sensitive to both structure and agency.

- Description: HIV/AIDS has been an obstacle to socio-economic development and a major cause of loss of human life. It has also caused vast inequities and frustration to the public health sector. One of the significant efforts made by the public health sector to combat the epidemic is the implementation of a mandatory HIV/AIDS testing policy to scale-up HIV treatment. This dissertation examines the impact of this policy on the human rights of people infected with and affected by HIV/AIDS. Coercive government policies aimed at controlling the AIDS pandemic often infringe on the rights of individuals known to be or suspected of living with HIV/AIDS and this decreases the effectiveness of public health measures. The research methodology involved the study of written literature and a comparative literature study of the law and practice obtaining in South Africa, Uganda and Canada. It revealed that voluntary testing is effective and suitable in South Africa. This dissertation aimed to show that any public health approach that aims to achieve a comprehensive prevention strategy must be consistent with respect for human rights as enshrined in regional and international human rights law. Public health and human rights should, therefore, not be regarded as opposing forces; rather they should be seen as a unified system of protection of human welfare under the Bill of Rights and the Constitution. The solution to the crisis lies not only in testing every single person but also requires a shift of focus to more pressing issues that include gender equality, stigma and discrimination; prioritizing human rights, institutional capacity and resources; and an end to extreme poverty. A human rights-based approach to HIV/AIDS testing, such as the Voluntary Counselling and Testing (VCT) is recommended. Therefore, failure to adhere to the core principles of testing - which are informed consent, counselling and confidentiality of the test result - will only hinder the global fight against HIV/AIDS. The rights of those affected by HIV/AIDS need to be protected in order to address public health imperatives. This can be done through the use of the law as an instrument of social change as well as education and awareness. Governance and management of urban trees and green spaces in South Africa: ensuring benefits to local people and the environment

- Description: In the face of rapid urbanization and global climate change, urban trees and green spaces (UTGS) can contribute to the welfare of people and the urban environment. Urban trees and green spaces can assist to address urbanization challenges related to environmental degradation. While functions of UTGS have been well documented in the developed world, they have not yet received full attention in much of sub-Saharan Africa. Consequently, UTGS are under threat from urban development and fragmentation. Notably, the problems associated with UTGS also fall into the governance realm and indications are that poor governance and management of UTGS can negatively influence the potential benefits of UTGS to local people and the environment. This formed a basis for this research. The main objective of the study was to determine the current governance and management approaches to UTGS in South Africa. Through document search and review, the study determined the governance institutions influencing UTGS at national level and at provincial level (in the Limpopo and Eastern Cape Provinces). Face-to-face and online survey methods were used to determine the extent to which 28 local municipalities had adopted planned, systematic and integrated management of UTGS. The snowball approach was used to determine the key actors involved in UTGS activities and interviews were conducted to establish the roles and capabilities of these actors. A total of 540 household interviews were conducted to determine the institutional factors influencing local peoples’ ability to access, plant and use UTGS. The findings of the study showed that UTGS have not been adequately covered in existing governance institutions and practice at national and provincial levels. Local government municipalities were not managing their UTGS in a planned or systematic manner due to constraining factors such as insufficient funds, insufficient personnel, lack of equipment and lack of political support. Only 7.1 % of the surveyed municipalities had an urban tree management plan and an estimate of the urban tree stock; 32.1 % had tree policies; 28.6 % had tree bylaws; 21.4 % had tree planting schedules; 10.7 % had tree maintenance schedules and only 3.6 % had tree inspection schedules. Key actors involved in UTGS activities differed among levels of government. Most of the actors, however, either planted trees or provided tree seedlings to municipalities and the local people. Tenure security was a key institutional factor affecting peoples’ ability to plant, use or even remove trees from their residential plots. The same applied to trees in the streets and public parks. Whereas most respondents did not require permission to plant (79.8 %) or remove (75 %) trees on their residential plots, a majority of them required permission to plant and remove trees from streets (over 70 %) and public parks (over 80 %). However, with regard to planting and removing urban trees in public open spaces, 54% of the respondents indicated that permission was not required suggesting a lack of clarity among local residents on the issue. Overall, the findings of this study indicate that there is no political recognition and support for UTGS at almost all levels of government. This has resulted in the lack of incorporation of UTGS in urban planning and development and has caused UTGS to receive limited funding to permit planned and systematic management. Given the current rates of urbanization and urban development, the lowly prioritised UTGS are vulnerable to exploitation. To conserve UTGS and promote their potential contribution to local people and the environment, UTGS must be recognized and placed on political and development agendas. There is a need to develop national guidelines for UTGS management, assess the extent of the urban forest resource in local municipalities, clearly define the roles and capabilities of different actors, integrate UTGS in the urban planning and development system, and most of all seek to involve the local people in overall management and governance of UTGS.

- Description: In the face of rapid urbanization and global climate change, urban trees and green spaces (UTGS) can contribute to the welfare of people and the urban environment. Urban trees and green spaces can assist to address urbanization challenges related to environmental degradation. While functions of UTGS have been well documented in the developed world, they have not yet received full attention in much of sub-Saharan Africa. Consequently, UTGS are under threat from urban development and fragmentation. Notably, the problems associated with UTGS also fall into the governance realm and indications are that poor governance and management of UTGS can negatively influence the potential benefits of UTGS to local people and the environment. This formed a basis for this research. The main objective of the study was to determine the current governance and management approaches to UTGS in South Africa. Through document search and review, the study determined the governance institutions influencing UTGS at national level and at provincial level (in the Limpopo and Eastern Cape Provinces). Face-to-face and online survey methods were used to determine the extent to which 28 local municipalities had adopted planned, systematic and integrated management of UTGS. The snowball approach was used to determine the key actors involved in UTGS activities and interviews were conducted to establish the roles and capabilities of these actors. A total of 540 household interviews were conducted to determine the institutional factors influencing local peoples’ ability to access, plant and use UTGS. The findings of the study showed that UTGS have not been adequately covered in existing governance institutions and practice at national and provincial levels. Local government municipalities were not managing their UTGS in a planned or systematic manner due to constraining factors such as insufficient funds, insufficient personnel, lack of equipment and lack of political support. Only 7.1 % of the surveyed municipalities had an urban tree management plan and an estimate of the urban tree stock; 32.1 % had tree policies; 28.6 % had tree bylaws; 21.4 % had tree planting schedules; 10.7 % had tree maintenance schedules and only 3.6 % had tree inspection schedules. Key actors involved in UTGS activities differed among levels of government. It was found that the ALSI resembles series that follow random walk hypothesis with strong evidence of RWH indicated in the conducted forecasting tests which showed vast variance between forecasted values and actual indicating little or no forecasting strength in the series. To further validate the findings in this research, the variance ratio test was conducted under heteroscedasticity and it also strongly corroborated that the existence of a random walk process cannot be rejected in the JSE. It was concluded that since the returns follow the random walk hypothesis, it can be said that JSE is efficient in the weak form level of the EMH and therefore opportunities of making excess returns based on out- performing the market is ruled out and is merely a game of chance. In other words, it will be of no use to choose stocks based on information about recent trends in stock prices.

MST programmes have been seen to improve the performance of athletes. This particular research study focuses on the development of an anxiety and arousal control MST programme designed specifically for, and tailored exclusively to, the needs of four archers from the Rhodes Archery Club in Grahamstown, Eastern Cape, South Africa. The development of the programme was guided by the Organisational Development Process model as the research methodology design. Data from two quantitative measures (CSAI-2 and Sport Grid-R), a focus group, and an individual interview were integrated with currently existing mental skills literature and theory to devise this particular MST programme. The results of the assessment phase showed that individual archers have different experiences in how anxiety and arousal affect their performances. The results also showed that archers have different zones in which they feel their performance is good. These zones depend on their subjective interpretations of anxiety and arousal. The results also showed that other factors that include personality differences, level of experience and level of expertise also influence how anxiety and arousal affect performance. The archers had previous exposure to MST programmes although they had not received systematic training in mental skills training. The programme was developed in the form of MST workshops that would run over a three week period. As part of the programme the archers would be given a mental skills manual to aid in the mental skills training. It is recommended in future that more research in MST programmes be done as a way of improving the performance of athletes in South Africa

Method: Interviews were used to explore the experiences and challenges of SLWHA. Results: The study shows that support groups are pivotal in the wellbeing of SLWHA in meeting diverse needs such as sense fo belonging, emotional, psychological, information and knowledge,socio-economic as well as health and physical needs. Support group members experience relatively similar living experiences, dynamic as their various lives are, they are able to share their emotions and problems arising from their HIV statuses, thereby lessening their burdens. Some negative experiences were observed, like issues of confidentiality, depression and stress. Conclusion: University support groups can be improved through structured programs that includes various stakeholders like health professionals (wellness centre), university management, and others including academics within the university. Also, expanding their borders via the use of social networks, and expanding their activities to include music, sports, vegetable gardening, group visitations, skills training, and other useful projects shall help to improve lives of victims and fight against the infection of other community members.

There is great concern that the poor will be unable to adapt to the impacts of climate variability and change while HIV/AIDS will exacerbate the impacts of such stressors and deepen the insecurities of many communities already affected by this disease. Studies that consider the interlinked effects of climate change and HIV/AIDS along with other multiple stressors are increasingly needed. This study, located in two rural communities in the Eastern Cape Province of South Africa, namely Lesseyton and Willowvale, assessed the responses of vulnerable households to the linked shocks and stressors of climate change and HIV/AIDS. This involved assessing, through household surveys, life history interviews and Participatory Learning and Action (PLA), the way in which multiple stressors interacted and affected vulnerable households, the way in which these households responded to and coped with such shocks and stressors, and the barriers which prevented them from coping and adapting effectively. Unemployment emerged as the dominant stress amongst households. The lack of development and having too few opportunities for employment has limited vulnerable households from being able to invest in assets, such as education or farming equipment. This, in combination with the impacts of increased food and water insecurity from recent drought, has created an extremely vulnerable environment for these households. They rely largely on two important safety-nets, namely social capital and the use of natural and cultivated resources; however the latter has been limited due to the impacts of water scarcity and an inability to farm. It was evident that there was little planned long-term adaptation amongst households and from government. Maladaptive short-term coping strategies, such numerous household members depending on one social grant and transactional sex, were too often relied upon, and although they may have helped relieve the stress of shocks momentarily, they did not provide for the long-term well-being of individuals and households. Poor communication and capacity between the different levels of government and between the government (especially at the local level) and the two rural communities has created an environment full of uncertainty and lacking in advocacy. Local government needs increased human, informational, and financial capacity and a clear delegation of responsibilities amongst the different departments in order for the two communities to benefit from the implementation of support strategies. There is also a great need for educational programmes and capacity development within the two rural communities, particularly based on improved coping and longer-term adaptation strategies in response to climate change in order for households to better prepare themselves for the future.

- Description: Professional nurses working in an intensive care unit (ICU) are faced with the death of critically ill patients frequently. Modern day medicine and technology have made it possible for advanced life-sustaining measures to be implemented on patients who, without medical intervention, would otherwise not have survived. The question is raised: is modern technology preserving life and prolonging the dying process, or is it in the best interest of the patient for treatment to be withdrawn? Nurses, caring for these patients and their families, are practicing at the bedside of these dying patients and are thus often faced with end-of-life issues, particularly withdrawal of treatment. The primary functions of critical care nurses are toward their patients. Physicians are responsible for making decisions regarding withdrawal of treatment. However, the nurses in the ICU are responsible for implementing the decisions made; sometimes contradicting what they believe in. The experience of end-of-life issues, namely withdrawal of treatment, is a cause of distress for the professional nurse. Little research has been done on how the ICU nurses deal with end-of-life issues and what support structures are required to assist nurses in dealing with end-of-life issues (Hov, Hedelin & Athlin, 2006:204) The objectives of the study were to explore and describe the professional nurses‟ lived experiences of end-of-life issues in the intensive care unit. The study aimed to make recommendations regarding support strategies to assist professional nurses in dealing with end-of-life issues in the intensive care unit. The researcher has selected a qualitative research approach with an explorative, descriptive and contextual design in order to conduct the study. Data was collected by means of semi-structured interviews. Data was analysed using the steps as illustrated by Tesch‟s method. Ethical principles were maintained throughout the research study. The findings of this study are to be presented in a journal publication.

- Description: Professional nurses working in an intensive care unit (ICU) are faced with the death of critically ill patients frequently. Modern day medicine and technology have made it possible for advanced life-sustaining measures to be implemented on patients who, without medical intervention, would otherwise not have survived. The question is raised: is modern technology preserving life and prolonging the dying process, or is it in the best interest of the patient for treatment to be withdrawn? Nurses, caring for these patients and their families, are practicing at the bedside of these dying patients and are thus often faced with end-of-life issues, particularly withdrawal of treatment. The primary functions of critical care nurses are toward their patients. Physicians are responsible for making decisions regarding withdrawal of treatment. However, the nurses in the ICU are responsible for implementing the decisions made; sometimes contradicting what they believe in. The experience of end-of-life issues, namely withdrawal of treatment, is a cause of distress for the professional nurse. Little research has been done on how the ICU nurses deal with end-of-life issues and what support structures are required to assist nurses in dealing with end-of-life issues (Hov, Hedelin & Athlin, 2006:204) The objectives of the study were to explore and describe the professional nurses‟ lived experiences of end-of-life issues in the intensive care unit. The study aimed to make recommendations regarding support strategies to assist professional nurses in dealing with end-of-life issues in the intensive care unit. The researcher has selected a qualitative research approach with an explorative, descriptive and contextual design in order to conduct the study. Data was collected by means of semi-structured interviews. Data was analysed using the steps as illustrated by Tesch‟s method. Ethical principles were maintained throughout the research study. The findings of this study are to be presented in a journal publication.

The best understood aspects of Hop so far indicate that residues in the concave surfaces of the three tetratricopeptide repeat (TPR) domains in the protein bind selectively to the C-terminal motifs of Hsp70 and Hsp90. Recent research suggests that P. falciparum Hop (PfHop), PfHsp90 and PfHsp70 do interact and form complex in the P. falciparum trophozooite and are overexpressed in this infective stage. However, there has been almost no computational research on malarial Hop protein in complex with other malarial Hsps.The current work has focussed on several aspects of the in-silico characterisation of PfHop, including an in-depth multiple sequence alignment and phylogenetic analysis of the protein; which showed that Hop is very well conserved across a wide range of available phyla (four Kingdoms, 60 species). Homology modelling was employed to predict several protein structures for these interactions in P. falciparum, as well as predict structures of the relevant TPR domains of Human Hop (HsHop) in complex with its own Hsp90 and Hsp70 C-terminal peptide partners for comparison. Protein complex interaction analyses indicate that concave TPR sites bound to the C-terminal motifs of partner proteins are very similar in both species, due to the excellent conservation of the TPR domain’s “double carboxylate binding clamp”. Motif analysis was combined with phylogenetic trees and structure mapping in novel ways to attain more information on the evolutionary conservation of important structural and functional sites on Hop. Alternative sites of interaction between Hop TPR2 and Hsp90’s M and C domains are distinctly less well conserved between the two species, but still important to complex formation, making this a likely interaction site for selective drug targeting. Binding and interaction energies for all modelled complexes have been calculated; indicating that all HsHop TPR domains have higher affinities for their respective C-terminal partners than do their P. falciparum counterparts. An alternate motif corresponding to the C-terminal motif of PfHsp70-x (exported to the infected erythrocyte cytosol) in complex with both human and malarial TPR1 and TPR2B domains was analysed, and these studies suggest that the human TPR domains have a higher affinity for this motif than do the respective PfHop TPR domains. This may indicate potential for a cross species protein interaction to take place, as PfHop is not transported to the human erythrocyte cytosol.

The modified surfaces were characterised using a variety of techniques; predominantly electrochemistry, scanning electrochemical microscopy and X-ray photoelectron spectroscopy. For the formation of self-assembled monolayers on gold, four new manganese(III) phthalocyanines (1a-d), octa-substituted at the peripheral position with pentylthio, decylthio, benzylthio, and phenylthio groups were synthesized and characterised. X-ray photoelectron spectroscopy was used to show the formation of a sulphur-gold bond. A number of approaches using 4-azidoaniline (2a) combined with azide-alkyne click chemistry and electrochemistry were also used to anchor ferrocene and pyridine moieties on to the carbon surfaces, including direct in situ diazotation and grafting, electropolymerisation, and the synthesis of the diazonium salt followed by grafting. Iron phthalocyanine was linked to the pyridine-clicked surfaces through axial ligation, where the strong axial bond formed by the interaction between the central metal and the lone pair of the nitrogen in the pyridine group resulted in stable modified electrodes. The potential of these surfaces for the detection of analytes such as thiocyanate, hydrazine and sulphite are briefly shown as well. This work also describes for the first time the possibility of performing local micro-electrochemical grafting of a gold substrate by 4-azidobenzenediazonium (2b) using scanning electrochemical microscopy in a single and simple one step approach, without complications from adsorption.

- Description: It has been argued that assessing learner performance is considered to be one of the most important things a teacher can do to promote learners’ learning. The Mathematics and Physical Science guidelines on assessment make a distinction between ‘assessment of learning’ and ‘assessment for learning’. Both processes are seen to be complementary aspects of continuous assessment (CASS) practices which constitute 25% of the final year learner’s mark in the National Senior Certificate. How teachers understand these processes and how they achieve them during their CASS practices were the focus of the study. This was a qualitative study in which two senior secondary schools in the East London District in the Eastern Cape participated. The respondents were four grade 12 Physical Science and Mathematics teachers and two heads of department (HoDs). Data were collected through semi-structured interviews and document analysis. There were five main findings. First, it was found that CASS assessment practices were skewed towards fulfilling accountability requirements of the Department of Education. Teachers focused on the Programme of Assessment (PoA) tasks and ignored other forms of assessment. Second, it appeared the use of the term ‘informal’ in the Department policy documents with regard to ‘assessment for learning’ encouraged teachers to treat informal assessment tasks as unimportant, not to be marked and recorded. In some cases assessment tasks were used to ‘occupy’ learners when teachers did not feel like teaching, so that learners would not be idle and disruptive in class. Third, some teachers did not set their own assessment tasks; they took previous papers and did cut and paste. Such a practice seemed to preclude assessment tasks that are tailor-made for particular learners. This practice also may undermine the alignment of learning outcomes and assessment standards. Fourth, moderation of CASS tasks was sometimes ignored and in other times done not strictly according to guidelines. These findings cast doubt on the validity and reliability of CASS marks. Finally, in most assessment for learning tasks, learners were not given feedback by teachers. This appears to undermine one of the most fundamental uses of assessment, which is feedback on performance that guides learning. It can be concluded that the CASS assessment practices did not reflect a balance between ‘assessment of learning’ and ‘assessment for learning’. This state of affairs seemed to be encouraged by the fact that district departmental support systems were too technicist and appeared to encourage teachers to do assessment to fulfil accountability purposes rather than for learners’ learning. It is recommended that research on assessment practices based on probability sampling for which results can be generalised to the target population should be carried out. It is further recommended that courses on assessment should be run for teachers in order to renew and/or give them assessment skills that will enable them to achieve a balance between assessment for learning and assessment of learning.

- Description: The heat shock proteins of ~ 70 kDa (Hsp70s) are a conserved group of molecular chaperones important in maintaining the protein homeostasis in cells, carrying out functions including refolding of misfolded or unfolded proteins. Hsp70s function in conjunction with a number of other proteins including Hsp40 cochaperones. Central to the regulation Hsp70 activity is the Hsp70 ATPase cycle, involving ATP hydrolysis by Hsp70, and stimulation of this ATP hydrolysis by Hsp40. PfHsp70-1, the major cytosolic Hsp70 in the malaria parasite, Plasmodium falciparum, and PfHsp70-x, a novel malarial Hsp70 recently found to be exported to the host cell cytosol during the erythrocytic stages of the P. falciparum lifecycle, are both thought to play important roles in the malaria parasite’s survival and virulence, and thus represent novel antimalarial targets. Modulation of the function of these proteins by small molecules could thus lead to the development of antimalarials with novel targets and mechanisms. In the present study, malarial Hsp70s (PfHsp70-1 and PfHsp70-x), human Hsp70 (HSPA1A), malarial Hsp40 (PfHsp40) and human Hsp40 (Hsj1a) were recombinantly produced in Escherichia coli. In a characterisation of the chaperone activity of recombinant PfHsp70-x, the protein was found to have a basal ATPase activity (15.7 nmol ATP/min/mg protein) comparable to that previously described for PfHsp70-1, and an aggregation suppression activity significantly higher than that of PfHsp70-1. In vitro assays were used to screen five compounds of interest (lapachol, bromo-β-lapachona and malonganenones A, B and C) belonging to two compound classes (1,4 naphthoquinones and prenylated alkaloids) for modulatory effects on PfHsp70-1, PfHsp70-x and HsHsp70. A wide range of effects by compounds on the chaperone activities of Hsp70s was observed, including differential effects by compounds on different Hsp70s despite high conservation (≥ 70 % sequence identity) between the Hsp70s. The five compounds were shown to interact with all three Hsp70s in in vitro binding studies. Differential modulation by compounds was observed between the Hsj1a-stimulated ATPase activities of different Hsp70s, suggestive of not only a high degree of specificity of compounds to chaperone systems, but also distinct interactions between different Hsp70s and Hjs1a. The effects of compounds on the survival of P. falciparum parasites as well as mammalian cells was assessed. Bromo-β-lapachona was found to have broad effects across all systems, modulating the chaperone activities of all three Hsp70s, and showing significant toxicity toward both P. falciparum parasites and mammalian cells in culture. Malonganenone A was found to modulate only the malarial Hsp70s, not human Hsp70, showing significant toxicity toward malarial parasites (IC₅₀ ~ 0.8 μM), and comparatively low toxicity toward mammalian cells, representing therefore a novel starting point for a new class of antimalarials potentially targeting a new antimalarial drug target, Hsp70.

- Description: The heat shock proteins of ~ 70 kDa (Hsp70s) are a conserved group of molecular chaperones important in maintaining the protein homeostasis in cells, carrying out functions including refolding of misfolded or unfolded proteins. Hsp70s function in conjunction with a number of other proteins including Hsp40 cochaperones. Central to the regulation Hsp70 activity is the Hsp70 ATPase cycle, involving ATP hydrolysis by Hsp70, and stimulation of this ATP hydrolysis by Hsp40. PfHsp70-1, the major cytosolic Hsp70 in the malaria parasite, Plasmodium falciparum, and PfHsp70-x, a novel malarial Hsp70 recently found to be exported to the host cell cytosol during the erythrocytic stages of the P. falciparum lifecycle, are both thought to play important roles in the malaria parasite’s survival and virulence, and thus represent novel antimalarial targets. Modulation of the function of these proteins by small molecules could thus lead to the development of antimalarials with novel targets and mechanisms. In the present study, malarial Hsp70s (PfHsp70-1 and PfHsp70-x), human Hsp70 (HSPA1A), malarial Hsp40 (PfHsp40) and human Hsp40 (Hsj1a) were recombinantly produced in Escherichia coli. In a characterisation of the chaperone activity of recombinant PfHsp70-x, the protein was found to have a basal ATPase activity (15.7 nmol ATP/min/mg protein) comparable to that previously described for PfHsp70-1, and an aggregation suppression activity significantly higher than that of PfHsp70-1. In vitro assays were used to screen five compounds of interest (lapachol, bromo-β-lapachona and malonganenones A, B and C) belonging to two compound classes (1,4 naphthoquinones and prenylated alkaloids) for modulatory effects on PfHsp70-1, PfHsp70-x and HsHsp70. A wide range of effects by compounds on the chaperone activities of Hsp70s was observed, including differential effects by compounds on different Hsp70s despite high conservation (≥ 70 % sequence identity) between the Hsp70s. The five compounds were shown to interact with all three Hsp70s in in vitro binding studies. Differential modulation by compounds was observed between the Hsj1a-stimulated ATPase activities of different Hsp70s, suggestive of not only a high degree of specificity of compounds to chaperone systems, but also distinct interactions between different Hsp70s and Hjs1a. The effects of compounds on the survival of P. falciparum parasites as well as mammalian cells was assessed. Bromo-β-lapachona was found to have broad effects across all systems, modulating the chaperone activities of all three Hsp70s, and showing significant toxicity toward both P. falciparum parasites and mammalian cells in culture. Malonganenone A was found to modulate only the malarial Hsp70s, not human Hsp70, showing significant toxicity toward malarial parasites (IC₅₀ ~ 0.8 μM), and comparatively low toxicity toward mammalian cells, representing therefore a novel starting point for a new class of antimalarials potentially targeting a new antimalarial drug target, Hsp70.

Inkcubeko Nendalo: a bio-cultural diversity schools education project in South Africa and its implications for inclusive Indigenous Knowledge Systems (IKS) Sustainability.

Emerging research reveals the increasing pressure on this biodiversity with many wild resources continuing to be utilised for livelihood purposes even within urban environments. The Rio conventions, particularly the CBD, call for an integrated approach to conservation that incorporates local environmental knowledge and practices. In a bid to market itself as globally competitive, South Africa’s Curriculum 2005 (C 2005) is primarily focused on Western-based scientific knowledge, which sidelines the contribution of indigenous knowledge systems (IKS) and ignores the holistic nature of indigenous worldviews. The Inkcubeko Nendalo programme is designed to revitalise cultural identity, showing children the value of local indigenous knowledge and cultural environmental values.
