An eco-physiological investigation of fisheries-induced evolution: comparing the resilience of larvae from exploited and unexploited commercial reef fish populations to projected ocean acidification

Marine organisms are being challenged by rapidly increasing temperatures, acidification, expansion of oxygen dead zones, and higher frequencies and magnitudes of extreme weather events. Exploited fish populations are also undergoing selective harvesting. Certain traits, such as large size, fast growth, and/or bold/active behaviours, are being actively targeted and removed from the population gene pool. This selective removal of individuals may compromise the capacity of fish populations to resist or recover from environmental disturbances and reduce their ability to adapt to a changing environment as many of these traits are heritable. As most marine fishes' embryonic and larval stages represent the period when individuals are most sensitive to environmental disturbances, they are a critical bottleneck to population persistence in the face of exploitation and climate change. This thesis aimed to quantify and compare the metabolic physiology, growth, and development of an exploited and endemic sparid, the roman seabream Chrysoblephus laticeps, during the early larval stages under 1) ocean acidification conditions expected by the year 2100 and 2) from populations experiencing dissimilar rates of exploitation. To quantify and compare the physiology of larvae, adult C. laticeps from an exploited population were captured and field-spawned. Fertilised eggs were placed into either control/present-day conditions (pH = 8.03, pCO2 ≈ 420 μatm) or high-pCO2/hypercapnic treatment conditions (pH = 7.63, pCO2 ≈ 1400 μatm). The metabolic physiology of individual larvae was determined using a novel rolling-regression technique on static respirometry data. Here, estimates of the minimum and maximum oxygen consumption rates (VO2) could be determined with high test-retest reliability. The very early developmental stages (yolk-sac stage) appeared resilient to high pCO2 conditions despite being exposed to treatment conditions throughout the embryonic stage. Preflexion larvae showed sensitivity to treatment conditions by exhibiting reduced metabolic and growth rates, consistent with metabolic depression, associated with environmental stress. However, by the onset of flexion, which coincides with gill development, acid-base regulation, and muscle differentiation, metabolic and growth rates of treatment larvae were significantly greater than that of controls. This suggests that acid-base regulation imposes a high cost to maintain internal pH homeostasis. Importantly, these elevated metabolic costs were likely mediated through increased feeding rates in experimental conditions where food was ad libitum. In natural conditions, where food availability may be varied, high pCO2 conditions could be associated with higher mortality rates. Based on evidence that protected/unexploited populations are more genetically diverse and are composed of individuals representing a greater range of metabolic phenotypes, offspring were collected from a protected population experiencing otherwise similar environmental conditions to the exploited population. Metabolic rates of control larvae were generally similar to those of the exploited population. However, minimum rates of VO2 were typically higher for larvae from the protected population at comparable life stages. Preflexion treatment larvae from the protected population did not appear to undergo a period of reduced metabolism or growth compared to their control counterparts. While metabolic rates at the onset of flexion were significantly higher for treatment larvae, this was not associated with growth differences. Growth over-compensation following periods of growth depression is often associated with deleterious effects, such as organ damage and body or developmental malformations. This suggests somewhat improved resilience to ocean acidification conditions. This thesis found evidence that larval C. laticeps are sensitive to ocean acidification conditions expected by 2100. When this stressor is combined with increasing thermal variability, changing current coastal regimes, and heterogeneous food availability, also expected to occur by 2100, ocean acidification may compromise the population persistence of this species. However, an energetics approach to stress-tolerance suggests that larvae from the protected population may inherently show greater resilience to climate change-related environmental stressors. Dual and targeted photodynamic therapy ablation of bacterial and cancer cells using phthalocyanines and porphyrins in the presence of carbon-based nanomaterials

- Description: Phthalocyanines (Pcs) and porphyrins bearing substituents that possess antibacterial/anticancer properties are used as photosensitizers (PS) for the first time in the work. For targeting specificity and improved photoactivity, the PSs were afterward functionalized with carbon nanomaterials such as graphene quantum dots (GQDs) and detonation nanodiamonds (DNDs) via covalent conjugation (amide or ester bonds) or by non-covalent conjugation (π-π stacking and electrostatic interactions). Furthermore, the PSs-DNDs nanoconjugates were conjugated to either chitosan-capped silver nanoparticles (CSAg) via amide bonds or to the bare silver nanoparticles (Ag NPs) using the silver- nitrogen affinity. The as-synthesized nanoconjugates were also fully characterized by spectroscopic and microscopic methods together with thermal analysis. The potential photocytotoxicity of the complexes alone and their nanoconjugates against S. aureus and/or E. coli planktonic and biofilm cultures has been evaluated in vitro. Compared to the non- quaternized PSs, the cationic analogs exhibited a higher photodynamic inactivation against the planktonic cells with log10 reduction values above 9 in the viable count using a concentration of ca. 1.25 μM following 30 min exposure to light (Light dose: 943 J/cm2 for Pcs and 250 mW/cm2 for porphyrins). Whereas, at a concentration of ca. 100 μM the cationic PSs showed complete eradication of biofilms upon 30 min exposure to light. As a result of conjugation to carbon-based nanomaterials and silver nanoparticles, the compounds proved to be more effective as they exhibited stronger antibacterial and anti-biofilm activities on the multi-drug resistant bacteria strains due to synergetic effect, compared to PSs alone. This suggests that the newly prepared nanohybrids (PS concentration ca. 100 μM) could be used as potential antimicrobial agents in the treatment of biofilm-related infections. The target nanoconjugates showed all the advantages of two different groups existing on a single entity. In light of the potential advantages of combined chemotherapy and photodynamic antimicrobial chemotherapy (PACT), this work reports for the first time the use of PACT-ciprofloxacin (CIP) dual therapy using selected indium quaternized PSs which showed higher photoactivity with complete eradication of both Gram-positive and Gram-negative bacteria biofilms at concentrations of 8 μM of PS versus 2 μg/mL of the antibiotic following 15 min irradiation time (light dose: 471 J/cm2 for Pcs and fluence: 250 mW/cm2 for porphyrins) on S. aureus. Whereas the total killing of E. coli was obtained when combining 8 or 16 μM of PS combined with 4 μg/mL of CIP. The combined treatment resulted in the complete eradication of the matured biofilms with the highest log10 reduction values of 7.05 and 7.20 on S. aureus and E. coli, respectively. Used as a model, positively charged dimethylamino-chalcone Pcs also exhibited interesting photodynamic therapy (PDT) activity against MCF-7 cancer cells giving IC50 values of 17.9 and 7.4 μM, respectively following 15 min irradiation. Additionally, the TD-B3LYP/LanL2DZ calculations were run on the dimethylaminophenyl- porphyrins to compare the singlet excitation energies of quaternized and non-quaternized porphyrins in vacuo. the study shows excellent agreement between time-dependent density- functional theory (TD-DFT) exciting energies and the experimental S1>S0 excitation energies. The small deviation observed between the calculated and experimental spectra arises from the solvent effect. - Description: My thesis is a collection of micro fiction, flash fiction, fairy tales, vignettes and short stories which explore the tension of being both an insider and an outsider. I have access to different cultures without belonging to any of them: as a child, my family moved from a South African Indian community to a formerly whites-only suburb; as an adult I have lived in South Korea, the United Arab Emirates and Sudan. My prose draws on my life experiences, family legends, neighbourhood gossip, news reports and historical events to question norms and ideas that I may have taken for granted had I been fully inside a single culture. In my thesis I frequently spell words phonetically to mimic how I hear or remember them. I also borrow words from languages I don’t speak. I want the languages I use and mix to corrupt each other, as Raymond Federman put it, in order to better express the voices and contexts of the communities I draw inspiration from. Kuzhali Manickavel’s Things We Found During the Autopsy showed me that culturally rich imagery can be used without interrupting narrative flow with explanations. I am also influenced by the poetic sense of rhythm and melody of Lydia Davis’s minimalist prose, and by Sandra Cisneros’s The House on Mango Street, where each concise short story stands alone but together creates a broad understanding of people and place. - Description: Although science education in Namibia receives much attention, learners’ performance is low in subjects such as Physical Sciences. The topic of Forces is among the topics in Physical Sciences where learners perform poorly. The provision of basic education in Namibia faces many challenges. One of these is that many teachers are not fully qualified for teaching the subjects they currently teach. Another (possibly related) challenge is that the majority of teachers in Namibian schools rely heavily on school textbooks when planning their lessons. In addition, learners use school textbooks as learning aids. The textbooks are developed and published in the private sector and based on the national curriculum statements. Quality of education has been amongst the major goals of education in Namibia. For quality assurance purposes, school science textbooks have to undergo a formal evaluation process. School textbooks, especially science textbooks, are multimodal. This means that they are designed with various modes, such as the textual and visual, often used in expressing scientific meanings. Literature reveals the textual and visual as individual modes having their own affordances, however, when integrated in school science textbooks contribute to strengthened meanings. No study published was found in Namibia or elsewhere that focused on analysing the intrasemiotic and intersemiotic sense relations of the textual and visual modes in Namibian school Physical Sciences textbooks. This study therefore aimed at contributing to filling this knowledge gap. The research is a qualitative case study and employed the interpretive paradigm. The selected Physical Sciences textbooks that constitute the data in this study were explored in depth via document analysis. Related textual and visual modes in the Physical Sciences textbooks were analysed in order to help answer the research questions of the study. Systemic Functional Linguistics was employed as the theoretical underpinning for this study. In this study, the sense relations were explored in terms of the sense relation themes which were coded and developed from common features of scientific discourse. The results reveal that scientific knowledge within the textual mode in the topic of Forces is communicated mostly through synonymy and repetition while within the visual mode it is mostly through antonymy, collocation, and synonymy. This contributed to meaning potential in the topic of Forces for the three Physical Sciences textbooks. The results further indicated that the overall cohesion within the textual and visual modes in the topic of Forces is achieved since repetition, which is the most direct form of lexical cohesion together with synonymy, collocation, and antonymy which are aspects of cohesion, occurred most frequently within these modes. This result further indicates that meaning potential in the topic of Forces in the three Physical Sciences textbooks were strengthened. While some sense relations occurred most frequently within individual modes, they were less frequent in other modes and vice versa. This indicates that different modes have different affordances. Finally, it was found that combining the textual and visual modes in the topic of Forces contributes to intersemiotic complementarity being achieved through the sense relations of collocation, hyponymy, and meronymy. , Thesis (MEd) -- Education, Education, 2021

The results further indicated that the overall cohesion within the textual and visual modes in the topic of Forces is achieved since repetition, which is the most direct form of lexical cohesion together with synonymy, collocation, and antonymy which are aspects of cohesion, occurred most frequently within these modes. This result further indicates that meaning potential in the topic of Forces in the three Physical Sciences textbooks were strengthened. While some sense relations occurred most frequently within individual modes, they were less frequent in other modes and vice versa. This indicates that different modes have different affordances. Finally, it was found that combining the textual and visual modes in the topic of Forces contributes to intersemiotic complementarity being achieved through the sense relations of collocation, hyponymy, and meronymy. , Thesis (MEd) -- Education, Education, 2021

The homodimeric cysteine main protease (Mpro) is crucial to the life cycle of the virus, as it cleaves the large polyproteins 1a and 1ab into matured, functional non-structural proteins. The Mpro exhibits high degrees of conservation in sequence, structure and specificity across coronavirus species, making it an ideal drug target. The Mpro substrate-binding profiles remain, despite the resolution of its recognition sequence and cleavage points (Leu-Gln↓(Ser/Ala/Gly)). In this study, a series of hexapeptide sequences containing the appropriate recognition sequence and cleavage points were generated and screened against the Mpro to study these binding profiles, and to further be the basis for efficiency-driven drug design. A multi-conformer hexapeptide substrate library comprising optimised 81000 models of 810 unique sequences was generated using RDKit within the context of python. Terminal capping with ACE and NMe was effected using SMILES and SMARTS matching. Multiple hexapeptides were complexed with chain B of crystallographic Mpro (PDS ID: 6XHM), following the validation of chain B for this purpose using AutoDock Vina at high levels of exhaustiveness (480). The resulting Vina scores ranged between -8.7 and -7.0 kcal.mol-1, and the reproducibility of best poses was validated through redocking. Ligand efficiency indices were calculated to identify substrate residues with high binding efficiency at their respective positions, revealing Val (P3), Ala (P1′); and Gly and Ala (P2′ and P3′) as leading efficient binders. Binding efficiencies were lowered by molecular weight. Substrate recognition was assessed by mapping of binding subsites, and Mpro specificity was evaluated through the resolution of intermolecular interaction at the binding interface. Molecular dynamics simulations for 20 ns were performed to assess the stability and behaviour of 132 Mpro systems complexed with KLQ*** substrates. Principal component analysis (PCA), was performed to assess II protein motions and conformational changes during the simulations. A strategy was formulated to classify and evaluate relations in the Mpro PCA motions, revealing four main clades of similarity. Similarity within a clade (Group 2) and dissimilarity between clades were confirmed. This study aimed to uncover the use of these learning support materials in the rural schools of Maputaland. The active learning framework, originally proposed and developed by O’Donoghue (2001), was used to analyse the materials. Collectively, the ACEP materials cover a range of active learning aspects; however alignment with the curriculum has resulted in an increased focus on experiments, accompanied by a loss of environmental content and a narrowing scope for active environmental learning. Workshop questionnaires and four school case studies revealed the patterns of practice of use of materials in schools. The stated use of materials by teachers is not fully realised in the actual classroom practice which centres on learning content and concept definitions. There is no culture of use of materials in the schools following the annual introduction of ACEP materials. It was also found that the marine and coastal knowledge holding power is outside the realm of the teachers’ practice and control. The findings of this study come at a time when there is uncertainty over the future of South African education and the curriculum. This research may inform the environmental education and coastal and marine education field as to their role in education and more specifically the development of learning support materials.

- Description: This qualitative study considers two bodies of theory, regenerative sustainability and social learning theory, within a tertiary education context. Universities offer unique opportunities, across both the formal and informal curriculum, that engage with the two-fold socioeconomic and environmental crises. The study explores both the promise of unlocking value in an ex-linear economy; and the healing and regenerative benefits in moving toward greener practices. Through an embedded case study at Rhodes University, in the Eastern Cape of South Africa, this research explores the value that is being unlocked in campus-based sustainability projects, when considered through a social learning lens (Lave and Wenger, 1991). Invitations to participate in this study were extended to project-leads of sustainability initiatives originating from Rhodes University campus. Six projects, which have been active on campus in recent years, were selected and, through semi-structured interviews, the insights of 12 participants were captured and analysed using the associated Value Creation Framework (VCF) developed by Wenger, Trayner and De Laat (Wenger, Trayner, de Laat, 2011) and later Wenger-Trayner and Wenger Trayner (2014, 2015, 2020). The VCF consists of value cycles that interconnect dynamically, including Orientating, Strategic, Enabling, Immediate, Potential, Applied, Realised and Reframed or Transformative value cycles. The study identified instances of all these value cycles, within and across the six embedded case studies (project). Drawing on social learning theorists the analysis further focused on instances of collaboration, agency and boundary crossing. Additional themes that emerged across the majority of the embedded study projects, were a convergence of socio and eco; a whole-systems perspective shared by project leads; the importance of the social commons; language as an enabler; and problem solving for system change. A recommendation is proposed to further unlock the unbounded value created through such projects by reframing the informal curriculum opportunities offered on university campuses as social learning spaces where students can practice agency. This combination of additive and subtractive manufacturing into a single system integrated the separate benefits of the two independent technologies. Mastercam was selected as the preferred CAD/CAM software package to generate toolpaths for the additive and subtractive processes. The Beckhoff HMI provided a user-friendly interface to interpret the generated G-code files, set the Work Coordinate System, and control user-defined parameters such as the feed rate. An in-depth statistical analysis of the developed platform's dimensional accuracy, repeatability, stability, and material wastage was performed. In addition, the effects of the independent adjustment of the subtractive manufacturing process parameters such as the spindle speed, feed rate, depth of cut and stepover distance were considered. Finally, a cycle time comparison was performed in producing a working prototype between the developed HM system and a commercially available 3D printer. Furthermore, the aim is to promote a sense of unity amongst these women as they enter a male-dominated field. South Africa’s skills shortages are widely regarded as key factors preventing the achievement of targeted economic growth rates. The decline and massive shortage in artisan trades has been attributed to the sharp decline of the apprenticeship system and the failure of substitute inter - ventions. By getting more women involved in the artisans’ field it will assist in gender equality and the current skills shortages. The treatise explores the issues and methods involved in the design of an Artisans Training Cen - tre for women. The research starts with an investigation into the nature of public space and civic facilities within a South African township context and how to create a more suitable environment for women, before exploring the development of an educational typology, while exploring the is - sues and processes involved in the design of a civic building in a township environment. The archi - tectural design is seen as an appropriate response to the identified architectural issues. Relevant precedents are chosen based on the nature of these issues and the context in which they are situated. The buildings are analysed in terms of these key issues and will thereby provide an understanding of the architectural identity of the civic and educational building archetype. An understanding of the architectural typology and an analysis of relevant precedents lead to the identification of a set of site selection criteria which can be used to select an appropriate site within the Motherwell precinct. The nature of Motherwell and the identified issues related to the selected site are then investigated in order to develop a set of informants and constraints. Specific elements which are unique to the selected site are then investigated individually while using relevant precedents to establish an appropriate response to the issues involved. In the context of the South African automotive industry, the volatility of the industry has seen many Original Equipment Manufacturers (OEM) and their suppliers shedding jobs. Furthermore, the replacement of the old automotive training board by the Sector of Education and Training (SETA) has created more complex challenges. Automotive companies are struggling to absorb the financial burden that comes with training skills transfer, due to the reduced workforce that is overloaded by work to cope in the current economic climate. The drivers of technical skills and knowledge transfer were investigated in a particular automotive company in Port Elizabeth, South Africa. A mail survey was directed to 168 technical employees in an automotive manufacturing plant. The survey tested the dependent variable (technical skills and knowledge transfer), and independent variables were its drivers (management of scarce skills, succession planning, trainee characteristics, training design and work environment). The survey had 50 questionnaire items. Statistical analysis was used to analyse the research questions through descriptive statistics, Pearson correlation and multiple regression analysis. Automotive companies are struggling to absorb the financial burden that comes with training skills transfer, due to the reduced workforce that is overloaded by work to cope in the current economic climate. The drivers of technical skills and knowledge transfer were investigated in a particular automotive company in Port Elizabeth, South Africa. A mail survey was directed to 168 technical employees in an automotive manufacturing plant. The survey tested the dependent variable (technical skills and knowledge transfer), and independent variables were its drivers (management of scarce skills, succession planning, trainee characteristics, training design and work environment). The survey had 50 questionnaire items. Statistical analysis was used to analyse the research questions through descriptive statistics, Pearson correlation and multiple regression analysis. The empirical results found that all of the independent variables showed positive correlations with the dependent variable, however succession planning, followed by work environment showed the most significant relationship with technical skills and knowledge transfer in an automotive organisation. , Thesis (MA) -- Faculty of Business and Economic science, 2022

The main aim of this study was to develop a model that uses machine learning techniques to recommend related research papers. The conceptual model was informed by literature on recommender systems in other domains. Furthermore, a literature survey on machine learning techniques helped to identify candidate techniques that could be used. The model comprises four phases. These phases are completed twice, the first time for learning from the data and the second time when a recommendation is sought. The four phases are: (1) identify and remove stopwords, (2) stemming the data, (3) identify the topics for the model, and (4) measuring similarity between documents. The model is implemented and demonstrated using a prototype to recommend research papers using a natural language processing approach. The prototype underwent three iterations. The first iteration focused on understanding the problem domain by exploring how recommender systems and related techniques work. The second iteration focused on pre-processing techniques, topic modeling and similarity measures of two probability distributions. The third iteration focused on refining the prototype, and documenting the lessons learned throughout the process. Practical lessons were learned while finalising the model and constructing the prototype. The treatise seeks to find a strategy for a contemporary, yet regional specific, intervention in a traditional context and attempts to fulfil the creation of a place of culture, identity, meaning and memory. The methodology used for this architectural treatise is qualitative. In order to establish an appropriate building type, Swellendam's history and contextual data were analysed, at the same time, selected literature and ideas were introduced to establish the project's value set. Methods and principles are extracted and reinterpreted by the design project. Among the various works of literatures/theories reviewed in this treatise is that of Jonathan Noble, Maurice Halbwach, and Kenneth Frampton, among others. This respective literature sets the basis of the project’s value set on place and identity, memory and meaning, and tectonics and materials. The town’s rich history, morphology and nature of physical context are selected as an appropriate site. Furthermore, the treatise explores museums and archives as building types. This is done through the comparison of precedents under the light of certain issues. From this, the building type’s activities and spatial, programmatical requirements are gathered. Amid this various information collected are a set of relevant principles. Therefore, a set of principles (approach to memory, meaning and identity; heritage and conservation; extracting from the existing/vernacular/context; and approach to light) is explored and precedents are used to represent and learn from principles. This design project goes through a series of iterations to fulfil the specific architectural concerns and to create architecture specific for the context (relation to place, culture, history and site). It is commonly assumed that SMPs employ the majority of professional accountants within the accounting field whose clients are typically small and medium-sized business entities (SMEs). SMEs account for most of the businesses around the world and play an important role in the development of economies as well as job creation. In a rapidly changing business environment, SMEs require a wider range of services and support from their professional accountants, thus, the needs and expectations of SMEs are constantly changing. Given the important contribution SMEs make to economic development, job creation, growth and innovation to communities globally, the success and survival rates of these businesses are of the utmost importance. Professional accountants of SMEs need to possess the relevant knowledge, skills, competencies and practical experience to provide professional business advisory services (non-accounting services) to create more value for the SME client. Research suggests that the SMEs decision to outsource business advisory services from their professional accountants is influenced by the relationship between the professional accountant and SME including the manner in which traditional compliance accounting services are provided (client services). In response, the objective of this study was to propose a graphical and narrative integrated training and client service framework to guide SME professional accountants. The framework provides guidance in respect of the knowledge, skills, competencies and practical experience a modern-day professional accountant should obtain during the various phases of learning and training whilst pursuing a career as a professional accountant. The framework further enhances the understanding of the traditional accounting services, client services and non-accounting services SME clients desire from their professional accountants as well as the role of professional accountants in providing or obtaining such services to ensure that the expectations of the client are met and, more importantly, contribute to the success of the SME. ABSTRACT vi An interpretivist research paradigm and a qualitative methodological approach were deemed most suitable for the current study. The interpretivist research paradigm approach enabled the researcher to understand the phenomena being studied by sharing the participants’ experiences in their own words thereby gaining a deeper understanding of the desires, services, skills and competencies they required or provided. The research population consisted of two population groups, firstly, professional accountants, accountants or bookkeepers who were either owners or were employed at SMPs and, secondly, SME owner-managers or the persons responsible for the accounting function employed at the SME. The research instrument used to guide the key-informant interviews in this research were two semi-structured interview schedules, one for each identified population group, which were divided into sections based on the themes and sub-themes. Based on key findings, the study proposes an integrated training and client service framework to guide SME professional accountants. The framework includes seven components, namely, formal higher education accounting programmes, workplace training, professional accountant, traditional accounting and client services, trust relationships, non-accounting services as well as networks of experts and other professionals. An important recommendation of the study is that professional accountants must effectively communicate and market their services to current as well as prospective SME clients to ensure that clients are aware of the potential services which can be provided or obtained from a trusted expert. The professional accountant could thus be regarded as a one-stop professional practice. This study focuses on compliance and monitoring for affirmative action for persons with disabilities in the South African workplace. Affirmative action is a transitory method of advancing equality. Constitutionally, it is acknowledged in South Africa that affirmative action in the form of substantive equality. This study explores whether or not employers utilize affirmative action in accordance with the Employment Equity Act. It also aims to evaluate how the Department of Employment and Labour and the courts implement affirmative action. The study considers the relevant literature available, statutes, and employment equality policies, as well as an evaluation of case law. In South Africa, individuals who have disabilities are one of the groups that the Employment Equity Act attempts to protect. People with disabilities are disadvantaged in the work arena, which has substantial psychological, social, and economic consequences. The government established the numerical target that employers must strive to achieve in the workplace. Consequently, it is essential to investigate further the obstacles that impede the employment of persons with disabilities including the physical accessibility of the environment and information, transportation, the attitudes of others (such as employers and co-employees), and the extra expenditures associated with managing a disability. The delivery, nature, and structure of an individual's education is one of the first determinants of socialization into society. There is a delicate relationship between poverty, disability, and social assistance programs that may have a substantial effect on the employment and job search of persons with disabilities. Interestingly, South Africa combines significant unemployment rates with effective social assistance programs. There are compelling economic, social, psychological, and political arguments for boosting the employment of those with disabilities. The policy of affirmative action is effective, but the study notes that the objectives have not yet been realized. The situation in South Africa is compounded by a variety of confusing circumstances, including substantial unemployment, strong social assistance, and disabled individuals with poor skill levels. The survey concluded that the majority of firms do not execute affirmative action appropriately and that enforcement is minimal. - Description: The Government of Zimbabwe is operating using the mantra: ‘Zimbabwe is open for business’. The notion of opening for business requires robust supporting economic infrastructure for enhanced productivity, in the form of reliable supply of electricity, accessible road/railway transport networks and availability of contemporary Information Communication Technology (ICT) infrastructure. The aim of the study was to develop a framework for making financing decisions for public infrastructure in Zimbabwe. The objectives of the study are to: determine the main sources of public infrastructure financing in Zimbabwe; establish innovative finance’s capacity to close the public infrastructure financing gap in Zimbabwe; assess the role played by public-sector accounting in attracting appropriate and efficient mechanisms to finance public infrastructure in Zimbabwe; and ultimately to develop and recommend a framework for selecting suitable and efficient mechanisms to finance public infrastructure in Zimbabwe. According to literature reviewed, public infrastructure is broadly financed by public sector entities using their own resources (internally financed) or through private sector investments and innovative financing instruments (externally financed). When infrastructure is internally financed, the study is theoretically guided by the Public Goods Theory and the Theory of Public Finance and Public Policy. When externally financed, the Risk Return and Pecking Order theories are important. There are many instruments used to finance public infrastructure and one project may be financed using one or more instruments. Therefore, considerations are given to the need for a framework that helps improve the efficiency of the financing decision. The study was designed as a multiple case study that focused on four sectors of economic infrastructure, that is, transport, energy (electricity), ICT as well as water and sanitation. The research used synchronous mixed methods to achieve the objectives of the study. Qualitative research methods addressed the following objectives: a) to determine the main sources of public infrastructure financing in Zimbabwe; b) to establish innovative finance’s capacity to close the public infrastructure financing gap in Zimbabwe; and c) to assess the role played by public-sector accounting in attracting appropriate and efficient mechanisms to finance public infrastructure in Zimbabwe. Whilst to develop and recommend a framework for selecting suitable and efficient mechanisms to finance public infrastructure in Zimbabwe the study combined qualitative and quantitative research methods. Qualitative data was collected through interviews conducted with officials and staff from government ministries, government departments, as well as parastatal enterprises dealing with the four infrastructure types. The same respondents were asked to complete a survey questionnaire used to address part of the objective that developed a framework for public infrastructure financing. Interview data were triangulated using secondary data extracted from reports and financial statements. Some of the secondary data was collected from the World Bank’s development indicators online repository. Qualitative data analysis was done using RQDA, an open-source computer-aided data analysis software. Findings from the study revealed that the main sources of finance for public infrastructure in Zimbabwe are the government through budget appropriations, and concessionary loans from the China Export-Import Bank. There was also finance obtained from multilateral financial institutions such as the Development Bank of Southern Africa and the African Export-Import Bank. The study revealed that there is currently very limited use of innovative financing instruments such as PPPs in financing public infrastructure in Zimbabwe, despite the country having legislation to support such financing arrangements. The innovations in financing observed in the study entail the use of conventional financing mechanisms in unconventional ways. However, there is scope for improving the financing of public infrastructure using innovative financing mechanisms and significantly mitigate the financing gap. Public sector accountants in Zimbabwe are mainly active in financial reporting, although the financial statements for most ministries, government departments and parastatal enterprises had qualified audit opinions from the Auditor General’s office. Public sector accountants are not active in financial management and cost and management accounting responsibilities. As a result, public sector accountants are not adding value to public money through offering advisory services in the efficient investment of public money, as well as financing public infrastructure assets using the most efficient financing mechanism. There is no uniformly applied framework when making financing decisions for public infrastructure in Zimbabwe. Therefore, a framework was developed and is recommended for use by this study. The developed framework entails eight steps that are interrelated and interconnected. Use of the proposed framework requires availability of data about infrastructure projects that have been done in the past. The study recommends that Zimbabwe should ensure a robust framework for protecting private sector investments, which can be achieved by ensuring policy consistency; creating and implementing a legal framework that protects private capital; and having economically viable infrastructure sectors, that are liberalised to allow private sector participation. The Government of Zimbabwe must take deliberate actions that ensure variety of financing options at the disposal of the public sector to lower costs of financing public infrastructure. It is also important to fully operationalise the legislation and policies designed to facilitate the participation of the private sector in financing public sector projects. - Description: The Government of Zimbabwe is operating using the mantra: ‘Zimbabwe is open for business’. The notion of opening for business requires robust supporting economic infrastructure for enhanced productivity, in the form of reliable supply of electricity, accessible road/railway transport networks and availability of contemporary Information Communication Technology (ICT) infrastructure. The aim of the study was to develop a framework for making financing decisions for public infrastructure in Zimbabwe. The objectives of the study are to: determine the main sources of public infrastructure financing in Zimbabwe; establish innovative finance’s capacity to close the public infrastructure financing gap in Zimbabwe; assess the role played by public-sector accounting in attracting appropriate and efficient mechanisms to finance public infrastructure in Zimbabwe; and ultimately to develop and recommend a framework for selecting suitable and efficient mechanisms to finance public infrastructure in Zimbabwe. According to literature reviewed, public infrastructure is broadly financed by public sector entities using their own resources (internally financed) or through private sector investments and innovative financing instruments (externally financed). When infrastructure is internally financed, the study is theoretically guided by the Public Goods Theory and the Theory of Public Finance and Public Policy. When externally financed, the Risk Return and Pecking Order theories are important. There are many instruments used to finance public infrastructure and one project may be financed using one or more instruments. Therefore, considerations are given to the need for a framework that helps improve the efficiency of the financing decision. The study was designed as a multiple case study that focused on four sectors of economic infrastructure, that is, transport, energy (electricity), ICT as well as water and sanitation. The research used synchronous mixed methods to achieve the objectives of the study. Qualitative research methods addressed the following objectives: a) to determine the main sources of public infrastructure financing in Zimbabwe; b) to establish innovative finance’s capacity to close the public infrastructure financing gap in Zimbabwe; and c) to assess the role played by public-sector accounting in attracting appropriate and efficient mechanisms to finance public infrastructure in Zimbabwe. Rapid developments in information technology have increased the number of smart appliances being used, leading to increased electricity usage. Devices and appliances in Smart Environments continue to consume electricity even when not in use, because of the standby function. The problems arise as the electricity consumption of the standby function accumulates to large amounts. Effective communication through visualisation of the electricity consumption in a Smart Environment provides a viable solution to reducing the consumption of electricity. This research aimed to design and developed a visualisation system that successfully communicates electricity consumption to the user using a variety of visualisation techniques. The Design Science Research Methodology was used to address the research questions and was used to iteratively design and develop an energy usage visualisation system. The visualisation system was created for the Smart Lab at the Nelson Mandela University's Department of Computing Sciences. A usability study was conducted to assess the usability and efficacy of the system. The system was found to be usable and effective in communicating power usage to potential customers, since the participants were able to complete the tasks in a short amount of time. An efficient approach for the synthesis of dolutegravir and its analogue exploiting flow chemistry

- Description: Africa has the highest Human Immunodeficiency Virus (HIV) prevalence in the world and has invested heavily in running its HIV programmes and conducting research of medical significance. Despite efforts in combating this disease the production, distribution, cost, and availability of antiretroviral (ARV) generics remains a major problem as they are imported from China and India. Exploring the recently emerged “enabling technique”, namely flow chemistry in the production of APIs has gained a lot of attention. This study was devoted on exploring the application of flow chemistry on the synthesis of a newly approved anti-HIV drug dolutegravir (DTG); an integrase inhibitor with a high genetic barrier to resistance with a daily dosing scheduled compared to other ARVs and its third-generation inhibitor analogue, cabotegravir. Chapter one covers a comprehensive background and literature review of the HIV epidemic, an introduction of antiretroviral therapy as well as detailed dolutegravir and cabotegravir synthesis. A brief introduction of continuous flow technology with its advantages and disadvantages is discussed in this chapter. The efficient seven-step continuous flow procedure afforded dolutegravir and cabotegravir in improved reaction times and yields compared to the traditional batch procedure was demonstrated. The significant advantage of this flow process includes the reduction of the overall reaction time from step one to step seven, from prolonged 68 hours in batch to 34 minutes. The overall yield of each reaction step improved dramatically upon flow optimization. The yields of the second step (selective ester hydrolysis), fourth step (cyclization) and fifth step (amidation) increased from 64 %, 40 % and 33 % in batch to 98 %, 71 % and 100 % yield by HPLC respectively. Other than improved yields and residence times which was aided advantages of microreactor technology including intensive mixing, the flow process is also much better because it is cost effective.
