Towards the modeling of indigenous poultry production in the Eastern Cape Province, South Africa: characterization and extension evaluation for poverty reduction

Various initiatives, policies and programs have been enacted by the government with attempting to reduce poverty at the national and provincial levels; poverty is yet to be abated. The target of the Millennium Development Goal 1 (MDG1) of halving poverty by the year 2015 notwithstanding, poverty remains a threat to quality livelihoods. Agriculture has continued to be one of the pillars of government efforts to address poverty. However, in livestock production interventions, efforts of government had largely been on cattle, sheep, goats, pigs and exotic poultry with little attention being given to indigenous poultry production (IPP). This study explores the option of using IPP to address rural poverty by capitalizing on its minimal inputs while recognizing its prevalence in a variety of households. The study addresses the characterization of the IPP from the perspectives of housing, feeding, healthcare management, breeding and marketing options.Skill competencies of the indigenous poultry farmers (IPFs) and the Agricultural Development Technicians (ADTs) were examined. This study reviewed poultry models in different parts of the world with special attention to Africa. The human resource development program of the department of rural development and agrarian reform was discussed. The study employed the use of multi-methods approach, the quantitative and qualitative research methodologies. Descriptive statistical analysis, frequent count, percentage, means, standard deviation, chisquare, and principal component analysis was used in the quantitative data analysis while the “open social system” was used for the qualitative methodology. Findings revealed that IPFs face some challenges among which are, the poor housing that exposed the birds to inclement weather, predator attacks and stock theft, high mortality of chicks after hatching and expensive feed for the flock. The IPFs showed competencies in nine skill items that included ability to identify chicken predators (x=3.92) and high yielding chickens (x=3.79); control of predators(x=3.77); methods of using ethno veterinary drugs to treat chicken diseases and pests (x=3.72), and identify signs of diseases (x=3.69), among others. However, the ADTs did not show any competency in any of the 32 skills items. The principal component analysis with Varimax rotation was performed to ascertain the dimensionality of the measures. Six factors with eigen value of >1, which accounted for 77.317% were extracted, with each factor loading ranging from 0.523 to 0.93. Factor loading after rotation that emerged on the same component was described; as brooding, shelter and care of the chicks; predators and healthcare; hygiene and litter management; feeds and feeding stuff; and record keeping and marketing. The findings on the human resource development revealed that staff meetings were the most common method of capacity development. This was followed by in-service-training, formal study, workshop, and on the job training. The farmers’ field school and study tours were sparingly used. However, respondents were in favor of on-site training, staff meetings, formal study and in-service training. The findings on the appropriate model suggest a theoretical indigenous poultry production model (IPPM) for the Eastern Cape Province (ECP). The “open social system” was used to develop a framework for an indigenous poultry cooperative society (to be known as Abafuyi Benkukhu Zemveli [ABZ]). This was meant to create a binding force for capacity development, a strong economic foundation through equity contributions, creation of marketing channels, and the development of a concept of ‘‘our own'' product in the market. Lastly, the study offers options for a training program that would accommodate the new initiatives, with a strong capacity development training approach. In this context, the study advocates for institutional support for the IPFs and the incorporation of indigenous poultry in the curriculum of the agricultural training programs at higher education institutions in South Africa.

Validating the use of Hippobromus pauciflorus (L.f) Radlk for the treatment of eye infections in the Eastern Cape South Africa

- Description: An ethnobotanical survey of plants used for the treatment of eye infections was carried out in the Eastern Cape Province of South Africa. The study revealed that 12 plant species are commonly used with Hippobromus pauciflorus (L.f) Radlk being the most frequent. The crude extracts of the leaves, stem bark and roots of the plant were investigated for antimicrobial activity against 10 bacterial and four fungal strains. The methanol extracts of the plant parts were the most active and showed appreciable activity against Gram-positive and Gram-negative bacteria. The methanol extracts of the leaves and stem bark inhibited the growth of fungi with activities ranging from 78.70 percent to 100 percent on Aspergillus niger and Penicillium notatum. The acetone extracts of the leaves and stem bark were active against A. niger (51.76 percent) and P. notatum (77.22 percent). The aqueous extract of Hippobromus pauciflorus leaves at 50, 100 and 200 mg/kg body weight doses were evaluated for anti-inflammatory, analgesic and antipyretic activities in male Wistar rats. Anti-inflammatory activity was studied by using carrageenan-and histamine induced oedema right hind paw volume while the analgesic effect was evaluated using formalin-induced pain and tail flick nociception response. The brewer’s yeast-induced pyrexia model was used for the antipyretic investigation. The extract at all the doses used significantly inhibited both the carrageenan- and histamine-induced inflammation in a manner that was not dose dependent. The extract reduced the formalin-induced pain licking as well as prolonged the reaction time in the tail flick-induced pain. The effects of the aqueous extract of the plant leaves at 50, 100 and 200 mg/kg body weight doses were investigated for 14 days on some biochemical parameters of male Wistar rats. The extract at all the doses tested did not significantly alter the levels of white blood cells, red blood cells, mean corpuscular volume, platelets, neutrophils, monocytes, lymphocytes and large unstained cells. While the levels of haemoglobin, packed cell volume and basophils increased at specific doses, those of mean corpuscular haemoglobin, mean corpuscular haemoglobin concentration and eosinophils decreased. In vitro antiviral activities of the plant against herpes simplex virus type 1(HSV-1) and coxsakie virus B6 were investigated. Cytotoxicity was evaluated by MTT assay in Vero cells. At concentrations ranging from 165-270/μg/ml there was significant antiviral activity against HSV-1, but toxicity was also noted. There was no antiviral activity against coxsakie virus B6. Bioactivity-guided fractionation of the leaves of H. pauciflorus yielded three known compounds. From the ethyl acetate fraction, two compounds were isolated (epicatechin and -sitosterol ) and from the n- hexane fractions, one compound (lupeol) was isolated. They were isolated and identified using various techniques. The antimicrobial, anti-inflammatory, analgesic and anti-pyretic activity of these compounds have been reported in literature. The structure and distribution of foliar appendages on the leaves of H. pauciflorus were examined by scanning electron microscope. The leaves have long unicellular nonglandular trichomes which were distributed over the mid rib and densely populated at the edges of the adaxial and abaxial surfaces. In general, the experiments and tests conducted in this study appear to have justified the use of Hippobromus pauciflorus for the treatment of eye infections and make a substantial contribution to the knowledge base of the use of herbal medicine for the treatment of the eye infections.

- Description: Abstract This thesis will present an investigation into the variability of the maximum height of the ionospheric F2 layer, hmF2, with hour, season and latitude over the South African region. The dependence of hmF2 on solar and magnetic activity is also investigated. Data from three South African stations, namely Madimbo (22.4 S, 26.5 E), Grahamstown (33.3 S, 26.5 E) and Louisvale (28.5 S, 21.2 E) were used in this study. Initial results indicate that hmF2 shows a larger variability around midnight than during daytime for all the seasons. Monthly median values for hmF2 were used in all cases to illustrate the variability, and the International Reference Ionosphere (IRI) model has been used to investigate hmF2 predictability over South Africa. This research represents the initial steps towards a predictive model for the hmF2 parameter, with the long term aim of developing a new global hmF2 predictive model for the IRI. It is believed that this work will contribute signi cantly towards this aim through the understanding of the hmF2 parameter over a region that has not previously been investigated.

Variation in the essential oil composition of Calendula Officinalis L

- Description: Variations in the yield, chemical composition, antibacterial, and antioxidant properties of the essential oils of Rosmarinus officinalis L. cultivated in Alice, Eastern Cape of South Africa over a period of 12 months using the solvent-free microwave extraction and traditional hydrodistillation methods were evaluated. The GC-MS analyses of the essential oils revealed the presence of 33 compounds with 1,8-cineole, a-pinene, camphor, verbenone, bornyl acetate and camphene constituting about 80 percent of the oils throughout the period of investigation, with the solvent-free microwave extraction method generally yielding more of the major components than the hydrodistillation method. Each of the major components of the oils varied in quantity and quality of yield at different periods of the year. The method of extraction and time of harvest are of importance to the quantity and quality of essential oil of Rosmarinus officinalis. Higher amounts of oxygenated monoterpenes such as borneol, camphor, terpene- 4-ol, linalool, a-terpeneol were present in the oil of SFME in comparison with HD. However, HD oil contained more monoterpene hydrocarbons such as a-pinene, camphene, β-pinene, myrcene, a-phellanderene, 1,8-cineole, trans- β-ocimene, γ-teprinene, and cis-sabinene hydrate than SFME extracted oil. Accumulation of monoterpene alcohols and ketones was observed during maturation process of Rosmarinus leaves. Quantitative evaluation of antibacterial activity, minimum inhibitory concentration values were determined using a serial microplate dilution method. The essential oils obtained using both methods of extraction were active against all the bacteria tested at a concentration of 10 mg mL-1. The minimum inhibitory concentrations for the SFME extracted oils ranged between 0.23 and 1.88 mg mL-1, while those of the HD extracted oils varied between 0.94 and 7.5 mg mL-1, thus suggesting that the oil obtained by solvent free microwave extraction was more active against bacteria than the oil obtained through hydrodistillation. The antioxidant and free radical scavenging activity of the obtained oils were tested by means of 1,1-diphenyl-2-picrylhydrazyl radical (DPPH+) assay and β- carotene bleaching test. In the DPPH+ assay, while the free radical scavenging activity of the oil obtained by SFME method showed percentage inhibitions of between 48.8 percent and 67 percent, the HD derived oil showed inhibitions of between 52.2 percent and 65.30 percent at concentrations of 0.33, 0.50 and 1.0 mg mL-1, respectively. In the β-carotene bleaching assay, the percentage inhibition increased with increasing concentration of both oils with a higher antioxidant activity of the oil obtained through the SFME than the HD method. Thin layer chromatography (TLC) was used to analyze the chemical composition of the extracts using three eluent solvent systems of varying polarities i. e. CEF, BEA and EMW and sprayed with vanillin-sulfuric acid. The chemical composition of the different extracts was similar with the exception of methanol and water extracts which had only one or two visible compounds after treating with vanillin-spray reagent. To evaluate the number of antibacterial compounds present in the fractions, bioautography was used against two most important nosocomial microorganisms. S. aureus (Gram positive) and E. coli (Gram negative). Nearly all the crude serial extraction fractions contained compounds that inhibited the growth of E. coli. The hexane extract had the most lines of inhibition followed by ethyl acetate. Bioassay-guided fractionation against E. coli was used to isolate antibacterial compounds. The largest number of antibacterial compounds occurred in the hexane fraction. Furthermore we tried to complete the characterization by extracting and studying other biologically important plant metabolites such as phenolic compounds to evaluate the antioxidant capacity of Rosmarinus extracts

The antioxidant and free radical scavenging activity of the obtained oils were tested by means of 1,1-diphenyl-2-picrylhydrazyl radical (DPPH+) assay and β- carotene bleaching test. In the DPPH+ assay, while the free radical scavenging activity of the oil obtained by SFME method showed percentage inhibitions of between 48.8 percent and 67 percent, the HD derived oil showed inhibitions of between 52.2 percent and 65.30 percent at concentrations of 0.33, 0.50 and 1.0 mg mL-1, respectively. In the β-carotene bleaching assay, the percentage inhibition increased with increasing concentration of both oils with a higher antioxidant activity of the oil obtained through the SFME than the HD method. Thin layer chromatography (TLC) was used to analyze the chemical composition of the extracts using three eluent solvent systems of varying polarities i. e. CEF, BEA and EMW and sprayed with vanillin-sulfuric acid. The chemical composition of the different extracts was similar with the exception of methanol and water extracts which had only one or two visible compounds after treating with vanillin-spray reagent. To evaluate the number of antibacterial compounds present in the fractions, bioautography was used against two most important nosocomial microorganisms. S. aureus (Gram positive) and E. coli (Gram negative). Nearly all the crude serial extraction fractions contained compounds that inhibited the growth of E. coli. The hexane extract had the most lines of inhibition followed by ethyl acetate. Bioassay-guided fractionation against E. coli was used to isolate antibacterial compounds. The largest number of antibacterial compounds occurred in the hexane fraction. Furthermore we tried to complete the characterization by extracting and studying other biologically important plant metabolites such as phenolic compounds to evaluate the antioxidant capacity of Rosmarinus extracts

Six study sites, two each from communal grazing, commercial farming and game reserves were selected. Two watering points were selected in each study site. Two 500 m transects were laid from the selected watering points. Each transect was divided into sub-transects at 25 m, 50m, 100m, 200m, 300m and 500m from water points. A 100 m2 plot was marked in each sub-transect to record grass biomas, species composition, structure and distribution of woody vegetation and physical and chemical soil properties. Data were collected for two seasons 2012/13 (winter and summer). Thirty and 41 grass and woody species respectively were identified in all study areas. The most common and dominating grass species include Cynodon dactylon, Digitaria eriantha, Eragrostis obtusa, Setaria sphacelata and Sporobolus fimbriatus. Cynodon dactylon and S. sphacelata occurred more abundantly (p < 0.05) in the game reserves than in the other land management catergories. All the grass species had similar (p > 0.05) abundance along distance gradient from water points. Grass dry matter (GDM) showed significant differences (p < 0.05) between and within land management systems. However, GDM was not significantly affected by season, location of water point within each farm or reserve and distance along water points. Acacia karoo, Coddia rudis and Ehretia rigida were the most dominant woody species. Tree equivalent (TE) density of all encroaching woody plants combined was significantly (p < 0.05) higher on the communal area (1732 TE ha-1) than the commercial ranches (1136 TE ha-1) and game reserves (857 TE ha-1), but with no marked variations along distance from water points under all the land management systems. The electric conductivity (EC) was significantly (P < 0.01) higher in game reserves than in communal grazing areas and ranches. Soil organic matter percentage showed greatest and lowest values in the game reserves and commercial respectively. Soil pH and bulk density did not vary but soil organic matter (SOM), EC, bulk density and soil compaction were significant different with no increasing or decreasing trends. Soil properties were affected by herbivore pressure and trampling around water points with inconsistence magnitude and direction. In conclusion, grass species composition and GDM did not respond to distance from water points because either grazing gradient was absent or the length of transects was not enough to explain the absence or presence of gradients. High proportion of A. karoo and high densities of seedlings and saplings would seem as very good indicators of the woody vegetation changes in the different land management systems and distance from water points. The soil quality indicators around the water-points showed that livestock and game affected soil parameters.

- Description: Knowledge of water use, through water foot printing (WF) in smallholder agriculture crop production is the key to the global fight against poverty, achievement of food security and sustainability within the world’s rural community. Water footprint of a crop can be defined as the volume of fresh water used to produce a certain crop in all the steps in the production line. This study, therefore aimed at contributing towards improvements in rural livelihoods by raising awareness of the increased productive use of green, blue and grey water in smallholder agriculture in South Africa. This was done through determination of water footprints of five vegetable crops, i.e. potatoes (Solanum tuberosum), tomatoes (Solanum lycopersicum), dry beans (Phaseolus vulgaris), cabbage (Brassica oleracea spp) and spinach (Spinacia oleracea) in the 2000-2013 period. Quantification of water footprints has been done worldwide but, in South Africa (SA) focus has mostly been on the industrial and domestic sector. Water footprint assessment framework, was used to estimate the full impact of vegetable production on water resources at Zanyokwe, Thabina and Tugela Ferry irrigation schemes as case studies. The CROPWAT@ model was used to calculate crop evapotranspiration, differentiating green and blue water. Local climatic data were obtained from SA weather services, while the crop and soil parameters were obtained from the FAO data base. Nitrogen was considered the main pollutant hence its use in the grey water footprint calculation. Generally, Thabina irrigation scheme had the highest water footprint, followed by Tugela Ferry irrigation scheme whilst Zanyokwe irrigation scheme had the lowest. Green beans had the highest water footprint at all the three irrigation schemes with Thabina irrigation scheme having the highest (3535.1 m3/ton). For Tugela Ferry irrigation scheme, the calculated WF was 2753 m3/ton whilst the lowest was observed at ZIS i.e. 2407.6 m3/ton. Cabbage had the lowest water footprint. The highest water footprint for growing cabbage was 254.5 m3/ton in TFIS, 223.1 m3/ton in TIS and the lowest was 217.8 m3/ton in ZIS. The differences observed in the WF of a crop at each scheme maybe attributed to the differences management, weather and environmental characteristics, in the three locations. Moreover, the needs for ET are related to soil type and plant growth, and primarily depend on crop development and climatic factors which are closely related to climatic demands. The grey water footprint was calculated using the recommended fertilizer application rates for all the three sites. Green beans had the highest WFgrey i.e. 373 m3/ton and the lowest was cabbage with 37 m3/ton. Potato, spinach and tomatoes had 156 m3/ton, 214 m3/ton and 132 m3/ton, respectively. Grey water footprint in this study was higher as compared to other studies, possibly because of the high rates of nitrogen fertilizers used in the calculations and the low yields farmers get. Compared with estimates from other studies, the water footprints of vegetable production within smallholder irrigation schemes was relatively high. There is therefore, a need to focus on crop management and tillage practices that will help in increasing yield while minimizing water usage.
